Microglia/macrophage activation plays an essential role in Ischemic stroke (IS). Nuclear receptor corepressor 1 (NCoR1) has been identified as a vital regulator in macrophages. The present study aims to explore the functions of macrophage NCoR1 in IS. Macrophage NCoR1 knockout (MNKO) mice and littermate control mice were subjected to middle cerebral artery occlusion (MCAO). Our data showed that macrophage NCoR1 deficiency significantly reduced the infarct size and infarct volume as well as brain edema after MCAO. Additionally, MNKO induced less microglia/macrophage infiltration and activation, neuroinflammation, apoptosis of neuronal cells, and BBB disruption in brains after IS. Mechanistic studies revealed that NCoR1 interacted with LXRβ in microglia and MNKO impaired the activation of the Nuclear factor-κB signaling pathway in brains after IS. Our data demonstrated that macrophage NCoR1 deficiency inhibited microglia/macrophage activation and protected against IS. Targeting NCoR1 in microglia/macrophage may be a potential approach for IS treatment.

Four molecular electrocatalysts based on cobalt complexes, CoT(X)PP (X = H (1), OH (2), CN (3), COOH (4)), were prepared from meso-tetra-p-X-phenylporphin (H2T(X)PP, X = H, OH, CN, COOH) by reaction with cobalt acetate to be used for electrolytic proton or water reduction. The electrochemical properties and the corresponding catalytic activities of these four catalysts were investigated by cyclic voltammetry. Controlled potential electrolysis with gas chromatography analysis confirmed that the turn-over frequencies (TOF) per mol of catalyst per hour were 42.4, 38.6, 55.5, and 70.1 mol H2 at an overpotential of 941.6 mV (in DMF) in the acetic acid solution containing catalyst. In neutral buffered aqueous solution (pH 7.0), these four molecular catalysts had TOF per mol of catalyst per hour of 352.53, 313.7, 473.4, and 714.6 mol H2, respectively, with an overpotential of 837.6 mV, indicating that complex 4 had better activity than complexes 1, 2, and 3. The Faraday efficiencies of complexes 1–4 were 99.1, 99.6, 100.4, and 99.0% at 72 h of consecutive reduction on a glassy carbon electrode, respectively. These results indicate that the electronic properties of the ligands play a crucial role in determining the catalytic activity of the cobalt complex and are consistent with the phenomenon that the catalytic activity of the benzene porphyrins is significantly increased in the presence of electron-withdrawing groups, and the CoT(COOH)PP is the most active catalyst.

Alterations of protein glycosylation are closely related with pathophysiological regulation. Due to the structural macro- and microheterogeneity, low stoichiometry, and low ionization efficiency of glycopeptides, high-performance tools to enrich glycopeptides, especially the negatively charged and labile sialoglycopeptides, are essential to enhance the identification of the underexplored glycoproteome. Here, we present the first implementation of zwitterionic hydrophilic interaction chromatography with the exposed choline group (ZIC-cHILIC) in StageTip for simultaneous enrichment and fractionation of intact glycopeptides. In a model study using lung cancer cells, early elution by a high percentage of acetonitrile prominently prefilters nonglycopeptides, facilitating high enrichment specificity for glycopeptides (92-96%) and sialoglycopeptides (77-89%) in the subsequent hydrophilic fractions. The stepwise elution shows a high glycopeptide fractionation efficiency by a 80% in triplicate analysis) as well as superior coverage of 4.6- to 12.0-fold and 2.1- to 35.6-fold more glycopeptides and sialoglycopeptides compared to conventional TiO2 and ZIC-HILIC, respectively. To the best of our knowledge, the result with 2742 sialoglycopeptides among 7367 unique glycopeptides and 166 glycans from 2434 N-glycosites of 1118 glycoproteins (Byonic score > 100) provides one of the deepest glycoproteomic profiles in single-cell type. Without the immunoprecipitation step, the large-scale glycoproteomic atlas also reveals site-specific glycosylation of many druggable receptor proteins, such as EGFR, MET, ERBB2, ERBB3, AXL, and IGF1R. The demonstrated high enrichment specificity and identification depth show that stepwise ZIC-cHILIC is an efficient method to explore the under-represented sialoglycoproteome.

The vector graphics with gradient mesh can be attributed to their compactness and scalability; however, they tend to fall short when it comes to real-time editing due to a lack of real-time rasterization and an efficient editing tool for image details. In this paper, we encode global manipulation geometries and local image details within a hybrid vector structure, using parametric patches and detailed features for localized and parallelized thin-plate spline interpolation in order to achieve good compressibility, interactive expressibility, and editability. The proposed system then automatically extracts an optimal set of detailed color features while considering the compression ratio of the image as well as reconstruction error and its characteristics applicable to the preservation of structural and irregular saliency of the image. The proposed real-time vector representation makes it possible to construct an interactive editing system for detail-maintained image magnification and color editing as well as material replacement in cross mapping, without maintaining spatial and temporal consistency while editing in a raster space. Experiments demonstrate that our representation method is superior to several state-of-the-art methods and as good as JPEG, while providing real-time editability and preserving structural and irregular saliency information.

Background Glucocorticoids (GCs) overuse is associated with decreased bone mass and osseous vasculature destruction, leading to severe osteoporosis. Platelet lysates (PL) as a pool of growth factors (GFs) were widely used in local bone repair by its potent pro-regeneration and pro-angiogenesis. However, it is still seldom applied for treating systemic osteopathia due to the lack of a suitable delivery strategy. The non-targeted distribution of GFs might cause tumorigenesis in other organs. Results In this study, PL-derived exosomes (PL-exo) were isolated to enrich the platelet-derived GFs, followed by conjugating with alendronate (ALN) grafted PEGylated phospholipid (DSPE-PEG-ALN) to establish a bone-targeting PL-exo (PL-exo-ALN). The in vitro hydroxyapatite binding affinity and in vivo bone targeting aggregation of PL-exo were significantly enhanced after ALN modification. Besides directly modulating the osteogenic and angiogenic differentiation of bone marrow mesenchymal stem cells (BMSCs) and endothelial progenitor cells (EPCs), respectively, PL-exo-ALN also facilitate their coupling under GCs’ stimulation. Additionally, intravenous injection of PL-exo-ALN could successfully rescue GCs induced osteoporosis (GIOP) in vivo. Conclusions PL-exo-ALN may be utilized as a novel nanoplatform for precise infusion of GFs to bone sites and exerts promising therapeutic potential for GIOP. Graphical Abstract

Intervertebral disc degeneration (IVDD) is a highly prevalent musculoskeletal disorder characterized by a local inflammatory response associated with the IL-1β/NLRP3 inflammasome positive feedback loop. Rice bran-derived gamma-oryzanol (Ory) as a sterol ferulate has attracted much attention due to its powerful anti-inflammatory, hypoglycemic and hypolipidemic health effects. As a clinical pharmaceutical for autonomic disorders, Ory's role in musculoskeletal degenerative disease remains unknown.This study aims to validate the role of Ory in IVDD and explore the potential mechanism.Establishing the in vitro and in vivo IVDD models to detect the protective effect and molecular mechanism of Ory.The anti-ECM degradation, antioxidant and anti-NLRP3 inflammasome activation effects of Ory on IL-1β-stimulated nucleus pulposus (NP) cells were assessed by immunoblotting and immunofluorescence, etc. MRI, S-O staining and immunohistochemistry were performed to estimate the effects of Ory administration on acupuncture-mediated IVDD in rats at imaging and histological levels.Ory treatment inhibited IL-1β-mediated ECM degradation, oxidative stress and NLRP3 inflammasome activation in NP cells. By interfering with NF-κB signaling and ROS overproduction, Ory interrupted IL-1β/NLRP3-inflammasome positive cycle. In vivo experiments showed that Ory delayed acupuncture-mediated IVDD development.Our results support the potential application of Ory as a therapeutic compound for IVDD.

In this study, a hierarchical mesoporous VO2/graphene@NiS2 hybrid aerogel is synthesized through the sol–gel and hydrothermal growth approaches. The resultant VO2/graphene@NiS2 hybrid aerogel electrode offers a high specific capacitance up to 1280.0 F g–1 (142.22 mAh g–1) at a current density of 1 A g–1 and exhibits an outstanding cycling performance. The distinguished electrochemical performance of the VO2/graphene@NiS2 electrode is attributable to the synergistic effect among VO2 particles, graphene networks, and NiS2 nanoflakes, which not only increases the specific surface area and conductivity but also gives extra pseudocapacitance to the VO2/graphene@NiS2 hybrid aerogel electrode. With the advancement of technology, the data generated in our lives is getting faster and faster, and the amount of data that various applications need to process becomes extremely huge. Therefore, we need to put more effort into analyzing data and extracting valuable information. Cloud computing used to be a good technology to solve a large number of data analysis problems. However, in the era of the popularity of the Internet of Things (IoT), transmitting sensing data back to the cloud for centralized data analysis will consume a lot of wireless communication and network transmission costs. To solve the above problems, edge computing has become a promising solution. In this paper, we propose a new algorithm for processing probabilistic skyline queries over uncertain data streams in an edge computing environment. We use the concept of a second skyline set to filter data that is unlikely to be the result of the skyline. Besides, the edge server only sends the information needed to update the global analysis results on the cloud server, which will greatly reduce the amount of data transmitted over the network. Most of the previously reported cases of intra-abdominal lipoma torsion originated in the mesentery or omentum. However, an antimesenteric lipoma of the ileum with torsion has not been reported before. Case Report: A 67-year-old man presented to the emergency department with acute abdominal pain. Contrast-enhanced computed tomography (CECT) of the abdomen and pelvis only showed a giant fat-containing, soft-tissue, intra-abdominal tumor, suspected to be a lipoma. Laparotomy was performed, and the presence of torsion of the antimesenteric lipoma of the ileum was confirmed. Beside tumor resection, en bloc segmental resection of the ileum with end-to-end anastomosis was performed to avoid bowel stricture and obtain tumor-free margins. Conclusions: CECT is the modality of choice to detect an intra-abdominal lipoma. Urgent surgical intervention should be considered if the symptoms persist and torsion cannot be excluded. If simple excision is not adequate because of poor accessibility of the tumor stalk, en bloc segmental resection with end-to-end anastomosis should be considered.

The Lu2SiO5 dried gel was prepared by the sol-gel reaction followed by a 24h drying at 90°C.The thermogravimetry–differential scanning calorimetry (TG-DSC) analysis curves of the dried gel demonstrates mass and enthalpy changes as temperature raised.The obtained dried gel was calcined at 900°C,1000°C,1100°Cand1200°C each for 2h to prepare polycrystalline Eu3+-dropped Lu2SiO5 phosphor.The phase composition and crystal structure was identified with an X-ray diffractometer (XRD).The dried gel was crystallized into A-type LSO phase at 1000°C, and transferred to B-type LSO phase when temperature was raised above 1050°C.The obtained LSO grain size is 200-300nm revealed in Field-emission scanning electron microscopy (FE-SEM) images.The photoluminescence spectrum of the obtained 2mol% Eu3+-dropped Lu2SiO5 phosphor showed a broad excitation band around 263nm and a sharp emission peak at 613nm.

Background: Amyloid-β deposition and accumulation of autophagic vacuoles are pathologic features of Alzheimer’s disease (AD). Dysregulation of the endosomal–autophagic–lysosomal (EAL) pathway, which impairs amyloid precursor protein processing, is one of the earliest changes in AD. However, the precise role of EAL pathway in neurodegeneration remains unclear. This study aimed to investigate the role of EAL pathway in AD and further study the mechanism of EAL dysfunction. Methods: We used 3-, 7-, and 12-month-old APPswe/PSEN1dE9 (APP/PS1) mice to model different stages of AD with age- and gender-matched wild-type littermates as controls (4–7 mice per group) and detected the changes of EAL markers, endosomal organizers Rab5 and Rab7, autophagosome marker LC3B, and lysosomal proteins Lamp1/2 in cortex and hippocampus by immunohistochemistry and Western blotting analysis. To further explore the mechanism of EAL dysregulation in AD, components of the class III phosphatidylinositol 3-kinase (PI3KC3) complex, activators of Rab7 (Beclin1 and UVRAG), and the negative regulator of Rab7 (Rubicon) were also measured in this two brain regions. Results: In 7-month-old APP/PS1 brain that amyloid beta initiated to accumulate intracellularly, EAL pathway, and related PI3KC3 members, UVRAG and Beclin1 were upregulated both in cortex and hippocampus (all P< 0.05). By the age of 12 months old, when abundant amyloid plaques formed, EAL markers, UVRAG, and Beclin1 were also upregulated in the cortex (all P< 0.05). However, Rab7 was decreased significantly (P = 0.0447), accompanied by a reduction of its activating PI3KC complex component Beclin1 (P = 0.0215) and enhancement of its inhibiting component Rubicon (P = 0.0055) in the hippocampus. Conclusions: Our study implies that EAL pathway, represented as Rab7 and its PI3KC3 regulators’ expressions, showed temporal and spatial variation in brains at different stages of AD. It provides new insights into the role of EAL pathway in pathogenesis and indicates potential therapeutic targets in neurodegenerative diseases. A four-layer functionally gradient material (FGM) of Cu/Ti3SiC2 was fabricated by hot pressing (HP) in vacuum. Oxidation behavior of Cu/Ti3SiC2 FGM was also investigated. The results show that the oxidation dynamics of Cu/Ti3SiC2 FGM at 600~800 °C for 30 h in air followed parabolic rate law. The hardness was increased with increasing content of Ti3SiC2. Thermal shock resistance was well owing to the effect of thermal stresses mitigated with FGM.

The diversity and community distribution of culturable bacteria, actinomycetes, fungi in semiarid desert steppe in northwestern part of China were studied by using dilution plate count, clone isolation and 16S rDNA sequence analysis methods. The results showed that the numbers of culturable bacteria, actinomycetes and fungi of desert steppe were 1.23×106 cfu·g-1, 0.19×106 cfu·g-1 and 0.18×106 cfu·g-1, respectively, compared with those of the cultivated land, whose numbers were 3.03×106 cfu·g-1, 0.53×106 cfu·g-1 and 0.05×106 cfu·g-1 respectively. The numbers of culturable bacteria and actinomycetes in the desert steppe soil were lower than those of the cultivated land, while the fungal number in desert steppe soil was higher than that of the cultivated land. A total of 14 bacterial strains were isolated from the desert steppe soil. The strains belonged to the following three phyla：γ-Proteobacteria（Psychrobacter）, Actinobacteria（Kytococcus）, and Firmicutes（Bacillus, Lysinibacillus, Solibacillus, Aerococcus）. Among which Bacillus and Psychrobacter were the predominant genus. Nineteen bacterial strains were isolated from the cultivated land and were further affiliated to four phyla：ɑ-Proteobacteri（Rhizobium, Sinorhizobium）, γ-Proteobacteria（Pseudomonas）, Firmicutes（Bacillus） and Actinobacteria（Microbacterium, Arthrobacter, Micrococcus, Kocuria）. Actinobacteria was the predominant phylum（occupied 57.9%）. A total of 8 actinomycetes strains were isolated from the desert steppe soil and were identified to belong to three phylogenetic groups：Atreptomyces, Micromonspora, Intrasporangium. And Atreptomyces, Micromonspora were the dominant communities in cultivated land. The fungi strains from the desert steppe were identified and designated as Alternaria and Cladosporium. But Penicillium, Alternaria, Aspergillus, Mucor and Coniothecium were isolated from cultivated land. The results indicated that the microbial diversities of the desert steppe and cultivated land were abundant and different. The microbial numbers and community distributions changed at different depths in the same area. The culturable microbial communities of the cultivated land were more abundant than those of the desert steppe soils.

The localization of object position in space is one of the most important visual abilities in humans. Motion-induced position shift is a perceptual illusion in which the position of a moving object is perceived to be shifted in the direction of motion. In this study, we wanted to explore whether and how Alzheimer’s disease (AD) affects this illusion. We recruited a group of patients with early AD and a group of age-matched healthy controls. In our experiments, two drifting Gabor patches moving in opposite directions were presented and participants were asked to report whether the upper Gabor appeared rightwards or leftwards of the lower one. We measured the psychometric functions, of which the point of subjective alignment was taken as the magnitude of motion-induced position shift. We compared the position shift across the two groups at three different retinal eccentricities. We found that position shifts were systematically smaller in the AD group as comparing to the elderly control group. Our data demonstrated that AD patients were less prone to motion-induced position shift. The results add to the existing knowledge of perceptual deficits in AD patients. We suggest that motion induced position shift may be effective as a new behavioral indicator for AD identification.

Ti/Al/TiC mixed powders were used as the raw materials for the fabrication of functionally graded material (FGM) in situ synthesis by spark plasma sintering. The sample was analyzed by SEM combined with Metallographic microscope and Energy Disperse Spectroscopy. The results show that dense Ti2AlC/TiC functionally graded material (FGM) was produced by spark plasma sintering. Micro-hardness tester is used to character the distribution of hardness on the material .The results show that the Vickers hardness of every layer increases with TiC content and temperature increasing.

Two Co(II)-based coordination polymers: [Co2(L)2(bib)2]n (1) and {[Co2(L)2(bimb)2]·2H2O·DMA)}n (DMA = N,N-dimethylacetamide) (2) using 4,4′-bis(thiomethylene) dibenzoic acid (H2L) as the first ligand, 1,4-bis(imidazol)-butane (bib) and 1,4-bis(imidazol-1-ylmethyl)benzene) (bimb) as the second ligands, were synthesized under the condition of solvothermal and characterized by X-ray single-crystal diffraction, elemental analysis, IR spectra and thermogravimetric analysis (TGA). X-ray single-crystal diffraction analysis exhibits that 1 and 2 belong to the triclinic crystal system, space group P-1. The central Co(II) ions exhibit a regular spatial tetrahedral structure which are coordinated by two O atoms from two L2− ligands and two N atoms from two N-donor ligands. Furthermore, the photocatalytic degradation properties and mechanism of 1 and 2 for methylene blue (MB) were investigated. The results show that the band gap (Eg) of 1 and 2 is 3.78 eV and 3.82 eV, and the equipotential point (pHpzc) of 1 is 4.25. Both 1 and 2 show excellent photocatalytic degradation for MB. •OH is the main active substance causing MB degradation. The photocatalytic degradation for MB of 1 and 2 is in line with the first order kinetics equation.

The size of TiC grains are about 3-6 μm. With TiC content in the layers increasing, Ti2AlC grains gradually decrease. The lathy Ti2AlC grains of the FGM have a length of about 10 μm and a width of about 4 μm. The interface of each layer has a good combination with a certain thickness transition layer.Microhardness tester is used to analyse the hardness of material.The results show that the Vickers hardness of every layer increases with TiC content increasing. The thermal shock performance test found that material has good thermal shock performance.

A very simple synthesis method of AlN-SiC solid solution powder, i.e. self-combustion of Al, Si3N4 and C mixture in the air only with assistance of high-energy mechanical milling, was proposed in the present work. The effect of compositions on the combustion behavior and the product was studied and the results showed that the amount of Al in the starting powder was the key factor affecting the combustion behavior, the phase assemblages and the morphology of products.

The epithelial–mesenchymal transition (EMT) plays a crucial role in pancreatic ductal adenocarcinoma (PDAC) development and progression. TWIST activated by intra-tumoral hypoxia functions to promote the EMT. We hypothesized that TWIST and the downstream gene pathway could mediate PDAC progression under hypoxia. Therefore, 90 PDAC tissue specimens were immunostained for TWIST and other proteins. Pancreatic cancer cell lines were used for in vitro experiments and nude mice were used to confirm the in vivo data. Expression of TWIST and HIF-1α proteins was significantly upregulated, whereas expression of E-cadherin and p16 was down-regulated in PDAC tissues compared to that of non-tumor tissues and in tumor tissues obtained from patients with tumor involving splenic artery than those without splenic artery involvement. Up-regulated TWIST in tumor tissues were associated with worse prognosis in PDAC patients. The in vitro data showed that HIF-1α-induced TWIST overexpression promoted tumor cell growth and EMT under a hypoxic condition via TWIST interaction with Ring1B and EZH2. In vivo data showed that TWIST overexpression or a hypoxic condition induce xenograft growth, abdominal metastasis and low mouse survival, whereas knockdown of either Ring1B or EZH2 expression suppressed tumor xenograft growth and metastasis and prolonged survival of nude mice. TWIST was the key player in promotion of pancreatic cancer development and metastasis under a hypoxic condition through interaction with Ring1B and EZH2 to regulate expression of E-cadherin and p16 proteins in pancreatic cancer cells.

Copolymer emulsions containing fluorine were prepared by emulsion polymerization with MMA, BA and G06B using SDBS as emulsifier. The copolymer was characterized by FTIR and DSC. The size and distribution of the latex particle in emulsion were obtained by particle size analyzer. The stability of emulsion was observed and the solvent resistance of latex films was investigated. Surface properties of the copolymer film were investigated by contact angle measurement. The results showed that adding fluorine group to copolymer could increase hydrophobic property of the latex film and decrease the surface free energy of copolymer film. Glass fiber will have excellent properties when combining with the copolymer emulsion.

Treadmill Measurement of Maximal Aerobic Capacity in Untrained Students Aged 9-18 Year

Europium-doped nanocrystalline ZrO2: Eu3+ powders with different doped concentrations were prepared by emulsion processing method, with different doped concentration and calcination temperature on the material grain size, structure, size and luminescent properties. The results show that: Eu3+ has a stabilizing effect on tetragonal phase of ZrO2. Emission spectra indicates that the main emission peak in the 593m and 608m, “Blue Shift “will occur in 5D0 → 7F2 transition of the emission spectra of sample with the sintering temperature at 1000°C. Samples with the sintering temperature soaring, spectral emission intensity also increases.

The cyclic oxidation behavior of Ti2AlN and Ti2AlN/TiN composites were investigated at 800°C, 900°C and 1000°C. The phase composition, thickness of oxidation layer and element content of samples were investigated by XRD and SEM coupled with EDS. The cyclic oxidation behaviors generally follow a parabolic rate law, with good thermal shock resistance. The antioxidation property of Ti2AlN/TiN composite is better than Ti2AlN. Oxidation process of single-phase Ti2AlN is composed by the outward diffusion of Al3+ and Ti4+, and inward diffusion of oxygen. Oxidation process of Ti2AlN/TiN composite consists of two parts: firstly, the oxidation of TiN, the oxidation temperature is 500°C; the second step is the oxidation of Ti2AlN, carried out in the 800-1000 °C.
