Black soybean hull, as main by-production in black bean processing, has significant biological activity. In this study, the polysaccharide from black soybean hull (BSPS-H) was obtained by enzymatic hydrolysis, the yield was 5.3%, and its chemical composition and structural composition were further studied. The results shown that BSPS-H was mainly composed of Mannose, Galactose, and Glucose (molar ratio = 48.1: 29.7: 15.8), and might be existed in the form of pyran ring in α/β-linkage type. The anti-oxidant activity shown that BSPS-H had a certain capacity of anti-oxidation in a concentration-dependent manner. Among them, it has the best scavenging effect on DPPH free radicals. When the polysaccharide concentration was 10 mg/ml, the scavenging rate reached 94.37%. The results of anti-fatigue effect indicated that BSPS-H could prolong the duration of exhaustive swimming in mice, increased the contents of liver glycogen, reduced the content of blood urea nitrogen (BUN), enhanced the activity of lactate dehydrogenase (LDH) and reduced the accumulation of lactic acid (LA). This study provides a fundamental basis for the development and use of BSPS-H in medicine and functional foods.

Deep mining of resources has destroyed the stress balance of original rock, formed water gushing channel, and caused the outburst of confined water, thus causing disaster. At present, “Ground Controlled Directional Bedding Branch Drilling (GCD-D)” grouting has achieved remarkable results in controlling water disasters. However, “what is the scientific design principle of GCD-D?” This key scientific question has not yet been answered by systematic science. According to the regional engineering geology and hydrogeological conditions of North China-type coalfields, this study uses the methods of analogy, theoretical analysis, and model analysis to demonstrate and analyze and obtains six basic principles for the selection of the target layer for the layout of “GCD-D.” The design principle of “GCD-D” was put forward for the first time. The results can provide a scientific theoretical basis for the control of confined water disaster in deep mining of North China coalfields.

The significance of biodiversity research is to understand the structure and function of the community, and then to protect and monitor the community. The metric of biodiversity is the base of biodiversity conservation. Species richness and evenness are the most common descriptors of biodiversity. Whether it is diversity information measure, probability measure or geometric measure, they all express the combination of species richness and evenness in different ways. This study presents a new biologically meaningful measure of species diversity, which evaluates species richness and evenness independently, designated as DRE. The novelty of our method is to use “absolute discrepancy” to express the dissimilarity between the observed community and the uniform distribution community with the same species composition and same abundance of each species, and then measure the species evenness. The logarithmic transformation of the species number is used to measure species richness with values ranging between 0 and 1. We test the performance of this measure using simulated data and observations of natural and planted forests in different climatic zones. The results showed that the new diversity index (DRE) has superior statistical qualities compared with the traditional indices. Especially, in extremely uneven communities, the new measure describes the causes of diversity changes than the traditional DRE. In addition, DRE is more sensitive to the abundance changes of rare species in the simulated community, and the interpretation of the results is more intuitive and meaningful. It is an improved method to evaluate the species diversity of any ecosystem.

The shallow-layer resources in the Huanghebei Coalfield have been depleted, and the deep coal seam of the Taiyuan Formation (lower coal group) is the main continuous resource in mining at this stage. With the annual increase in mining depth, the exploitation of lower coal groups is being met with the influence of high ground stress, high water pressure, high temperature, and strong mining disturbances, as a result of which the threat of water inrush is particularly serious. Based on the grouting data of the coalface floor aquifer, this paper proposes an evaluation method for the control of water rushing into the coalface floor. By comparing the test data with mine electrical exploration data through ArcGIS, the results show that the water pressure threshold based on ArcGIS is twice the water pressure of the grouting reconstruction layer as the optimal solution. The research results can provide a reference for the prevention and control of water inrush in the lower coal group of Huanghebei Coalfield.

Three β-glucosidases from Bifidobacterium adolescentis ATCC15703, namely, BaBgl1A, BaBgl3A, and BaBgl3B, were overexpressed in Escherichia coli. The recombinant β-glucosidases were sufficiently purified using Ni2+ affinity chromatography, and BaBgl1A exhibited the best purification efficiency with a purification factor of 2.3-fold and specific activity of 71.2 U/mg. Three recombinant β-glucosidases acted on p-nitrophenyl-β-glucopyranoside (pNPβGlc) at around pH 7.0 and 30–50°C. The results of the substrate specificity assay suggested that BaBgl1A acted exclusively as β-1,2-glucosidase, while BaBgl3A and BaBgl3B acted mostly as β-1,3-glucosidase and β-1,4-glucosidase, respectively. The substrate specificity of the three recombinant enzymes was further studied using the ginsenosides Rb1 and Rd as substrates. The results of thin-layer chromatography and high-performance liquid chromatography analyses showed that BaBgl1A exhibited the highest bioconversion ability on Rb1 and Rd, where it hydrolyzed the outer C-3 glucose moieties of Rb1 and Rd into the rare ginsenosides Gypenoside XVII and F2; BaBgl3A exhibited medium bioconversion ability on Rb1, where it hydrolyzed both the outer C-3 and C-20 glucose moieties of Rb1 into Gyp XVII and Rd; and BaBgl3B was not active on Rb1 and Rd. These β-glucosidases will act as new biocatalytic tools for transforming ginsenosides and preparing active glycosides and aglycone.

Abstract Background Water shortage can limit plant growth, which can be ameliorated by arbuscular mycorrhizal (AM) symbiosis through physiological and metabolic regulations. Deciphering which physiological and metabolic processes are central for AM-mediated regulations is essential for applications of mycorrhizal biotechnology in dryland agriculture. Methodology In this study, the influence of AM symbiosis on growth performance, photosynthesis, and organ accumulation of key C and N metabolites were assessed by growing maize (Mo17, Lancaster Sure Crop) seedlings inoculated with or without AM fungus (Rhizophagus irregularis Schenck & Smith BGC AH01) under different water regimes in greenhouse. Results Drought stress reduced shoot growth, while AM symbiosis significantly improved growth performances, with significant changes of photochemical processes and organ concentration of the key metabolites. AM symbiosis increased root levels of the metabolites in ornithine cycle and unsaturation of fatty acids regardless of water conditions. Root putrescine (Put) concentration was higher in AM than non-inoculated (NM) plants under well-watered conditions; the conversion of Put via diamine oxidase to γ-aminobutyric acid (GABA) occurred in roots of AM plants under drought stress. Leaf concentration of Put, the tricarboxylic acids, and soluble sugars significantly increased in AM plants under drought stress, showing higher values compared to that of NM plants. Moreover, photosystem II efficiency and chlorophyll concentration were higher in AM than NM plants regardless of water status. Conclusion Fatty acid- and ornithine cycle-related metabolites along with soluble sugars, Put, and GABA were the key metabolites of AM-mediated regulations in response to drought stress.

Abstract By studying the Riemann problem for the Aw–Rascle traffic model with different pressure laws, which is the coupling of two one-dimensional hyperbolic systems, we investigate the resonance phenomena. The main difficulty arises from the possible resonance behavior which may result in multiple solutions. We discover a new and interesting phenomenon showing that there exist infinitely many solutions for some certain initial data, which is quite different compared to earlier studies for the isentropic model of a fluid flow in a nozzle with variable cross-section and the shallow water equations with discontinuous topography. In order to overcome this difficulty, we impose the so-called TV-condition to obtain the uniqueness of solution to the Riemann problem.

Deep, highly pressurized aquifers seriously threaten the exploitation of resources of North China coalfield. At present, there is no particular guidance system to evaluate the prevention and control effect of water inrush disaster in deep coal seam mining. Therefore, this research focuses on the evaluation of the method for confined water disaster prevention and control effect of deep coal seam mining. Based upon various methods, such as effective thickness and water resistance capacity of aquifuge, water inrush coefficient, GIS (Geographic Information System) analysis of grouting data, dense drilling verification, and professional analysis system based on integrated sequence evaluation, the evaluation of the method for confined water disaster prevention and control effect of deep coal seam mining was put forward for the first time. The proposed method was called the “sequence verification evaluation (SVE)” method. The prevention and control of the effects of Ordovician water inrush disaster in deep mining of Huanghebei coalfield was evaluated using the proposed SVE method. The results show that the SVE method can effectively reflect the prevention and control of the effects of water inrush from confined water and provide scientific basis for the safe production of deep coal seam mining.

Heat shock proteins (HSPs) are a large class of highly conserved chaperons, which play important roles in response to elevated temperature and other environmental stressors. In the present study, 5 HSP90 genes and 17 HSP70 genes were systematically characterized in spotted seabass (Lateolabrax maculatus). The evolutionary footprint of HSP genes was revealed via the analysis of phylogeny, chromosome location, and gene copy numbers. In addition, the gene structure features and the putative distribution of heat shock elements (HSEs) and hypoxia response elements (HREs) in the promoter regions were analyzed. The protein-protein interaction (PPI) network analyses results indicated the potential transcriptional regulation between the heat shock factor 1 (HSF1) and HSPs and a wide range of interactions among HSPs. Furthermore, quantitative (q)PCR was performed to detect the expression profiles of HSP90 and HSP70 genes in gill, liver, and muscle tissues after heat stress, meanwhile, the expression patterns in gills under alkalinity and hypoxia stresses were determined by analyzing RNA-Seq datasets. Results showed that after heat stress, most of the examined HSP genes were significantly upregulated in a tissue-specific and time-dependent manners, and hsp90aa1.1, hsp90aa1.2, hsp70.1, and hsp70.2 were the most intense responsive genes in all three tissues. In response to alkalinity stress, 11 out of 13 significantly regulated HSP genes exhibited suppressed expression patterns. Alternatively, among the 12 hypoxia-responsive-expressed HSP genes, 7 genes showed induced expressions, while hsp90aa1.2, hsp70.1, and hsp70.2 had more significant upregulated changes after hypoxic challenge. Our findings provide the essential basis for further functional studies of HSP genes in response to abiotic stresses in spotted seabass.

Various environmental stresses strongly influence plant development. Among these stresses is drought, which is a serious threat that can reduce agricultural productivity and obstruct plant growth. Although the mechanism of plants in response to drought has been studied extensively, the adaptive strategies of Amygdalus mira (Koehne) Yü et Lu grown in drought and rewatered habitats remain undefined. Amygdalus mira from the Tibetan Plateau has outstanding nutritional and medicinal values and can thrive in extreme drought. In this study, the physiological and proteomic responses in leaves of A. mira were investigated during drought and recovery period. The changes in plant growth, photosynthesis, enzymes, and non-enzymatic antioxidant under drought and rewatering were also analyzed in leaves. Compared with controls, A. mira showed stronger adaptive and resistant characteristics to drought. In addition, the proteomic technique was also used to study drought tolerance mechanisms in A. mira leaves. Differentially expressed proteins were identified using mass spectrometry. Accordingly, 103 proteins involved in 10 functional categories: cytoskeleton dynamics, energy metabolism, carbohydrate metabolism, photosynthesis, transcription and translation, transport, stress and defense, molecular chaperones, other materials metabolism, and unknown function were identified. These results showed that an increase of stress-defense-related proteins in leaves after drought treatment contributed to coping with drought. Importantly, A. mira developed an adaptive mechanism to scavenge reactive oxygen species (ROS), including enhancing antioxidant enzyme activities and non-enzymatic antioxidant contents, reducing energy, and adjusting the efficiency of gas exchanges. These results may help to understand the acclimation of A. mira to drought.

Abstract Background The broad-leaved Korean pine mixed forest is an important and typical component of a global temperate forest. Soil microbes are the main driver of biogeochemical cycling in this forest ecosystem and have complex interactions with carbon (C) and nitrogen (N) components in the soil. Results We investigated the vertical soil microbial community structure in a primary Korean pine-broadleaved mixed forest in Changbai Mountain (from 699 to 1177 m) and analyzed the relationship between the microbial community and both C and N components in the soil. Abstract Background China has a long tradition of managing planted forests. Different species of Populus, Eucalyptus, Larix, Cunninghamia and Pinus are planted to satisfy the local demand for wood products and provide ecological services at the same time. Evidence of the greater resilience of natural forests provides the motivation to develop asymmetric planting patterns, which is the focus of this study. We present a new method for designing plantation patterns that follow those observed in natural ecosystems and to maintain some regularity for operational convenience. Methods Based on the uniform angle index, we analyzed the spatial structure of six natural forests in different regions of China. The uniform angle index describes the degree of spatial uniformity of the n nearest neighbors of a given reference tree. Accordingly, we identified all possible patterns of a neighborhood group within a regular planting pattern and developed a method to optimize planting point arrangements that contain some randomness as well as a minimum degree of regularity. Results (1) There are 13 types of structural units in a regular planting, including seven random units, five even units and one cluster unit; (2) Five near-natural arrangements are presented with a minimum proportion of 50% of random units. These five arrangements represent a combination of regularity for operational convenience and asymmetry. Conclusions The new planting patterns developed in this study are expected to increase the asymmetric competition and resilience of these important ecosystems. Some experimental plantings, based on our findings, have already been established, e.g., in Pinus tabulaeformis plantations in Tianshui, Gansu Province, and in a Populus deltoides plantation in Fangshan near Beijing.

Abstract Low-dimensional semiconductors exhibit remarkable performances in many device applications because of their unique physical, electrical, and optical properties. In this paper, we report a novel and facile method to synthesize In2S3 quantum dots (QDs) at atmospheric pressure and room temperature conditions. This involves the reaction of sodium sulfide with indium chloride and using sodium dodecyl sulfate (SDS) as a surfactant to produce In2S3 QDs with excellent crystal quality. The properties of the as-prepared In2S3 QDs were investigated and photodetectors based on the QDs were also fabricated to study the use of the material in optoelectronic applications. The results show that the detectivity of the device stabilizes at ~ 1013 Jones at room temperature under 365 nm ultraviolet light irradiation at reverse bias voltage.

Abstract GDP-l-fucose, the key substrate for fucosyloligosaccharide biosynthesis, has been synthesized via a de novo pathway in bacteria. In the present study, genes for GDP-l-fucose biosynthesis were cloned into the expression vector pET-28a (+) to construct five E. coli strains, with recombinant enzymes being purified by using Ni–NTA chromatography. Following optimization of the 3-step reaction, Glk, ManB and ManC were added to the reaction mixture, after which Gmd and WcaG were added to overcome feedback inhibition from the end-product to produce GDP-l-fucose at 178.6 mg/l, with a yield of 14.1%. Our studies provide the basis for using cell-free enzyme production of GDP-l-fucose.

Abstract This paper focuses on a degenerate boundary value problem arising from the study of the two-dimensional Riemann problem to the nonlinear wave system. In order to deal with the parabolic degeneracy, we introduce a partial hodograph transformation to transform the nonlinear wave system into a new system, which displays a clear regularity–singularity structure. The local existence of classical solutions for the new system is established in a weighted metric space. Returning the solution to the original variables, we obtain the existence of classical solutions to the degenerate boundary value problem for the nonlinear wave system.

Understanding the diversity and complexity of stand structure is important for managing the biodiversity of forest ecosystem, and stand structural diversity is essential for evaluating forest management activities. Based on the relationship of adjacent trees, a quantitative method of stand structure diversity is proposed to express the heterogeneity of stand structure in tree species, distribution pattern, species separation and size differentiation. In this study, we defined the diversity of structural unit types and derived a new index of forest structural diversity (SD′) employing the additivity principle of the Shannon–Weiner index. The efficiency of the index was verified by applying the new measure to sixteen field survey samples at different locations. The mountain rainforest in Hainan had the highest forest structural diversity, followed by broad-leaved Korean pine forests in Jiaohe (2), Jiaohe (1) and an oak-broadleaved mixed natural forest at Xiaolongshan (2). The SD′ values of plantations and pure natural forest were lower. The simulated data of different thinning methods and the intensity of broad-leaved Korean pine forests show that the new measure can reflect forest management changes on stand structure diversity. The value of SD′ compared with no treatments and the differences were greater as thinning intensity increased. The SD′ index provides minimum and maximum values for different structural unit types in forests to achieve a unified comparative basis for calculating forest structure diversity. It has the characteristics of the general diversity index and can well express the diversity of tree species, distribution pattern and size differentiation simultaneously. The SD′ index can not only calculate the structural diversity of mixed species forests but can also be used to calculate the structural diversity of pure forests. It can also be used to evaluate the change in stand structure diversity after management interventions.

Nitrogen fertilization is common for poplar trees to improve growth and productivity. The utilization of N by poplar largely depends on fertilizer application patterns; however, the underlying regulatory hubs are not fully understood. In this study, N utilization and potentially physiological regulations of two poplar clones (XQH and BC5) were assessed through two related experiments (i: five levels of N supply and ii: conventional and exponential N additions). Poplar growth (leaf area) and N utilization significantly increased under fertilized compared to unfertilized conditions, whereas photosynthetic N utilization efficiency significantly decreased under low N supplies. Growth characteristics were better in the XQH than in the BC5 clone under the same N supplies, indicating higher N utilization efficiency. Leaf absorbed light energy, and thermal dissipation fraction was significantly different for XQH clone between conventional and exponential N additions. Leaf concentrations of putrescine (Put) and acetylated Put were significantly higher in exponential than in conventional N addition. Photorespiration significantly increased in leaves of XQH clone under exponential compared to conventional N addition. Our results indicate that an interaction of the clone and N supply pattern significantly occurs in poplar growth; leaf expansion and the storage N allocations are the central hubs in the regulation of poplar N utilization.

Abstract Negative conspecific density dependence is one of the principal mechanisms affecting plant performance and community spatial patterns. Although many studies identified the prevalence of density dependent effects in various vegetation types by analyzing conspecific spatial dispersal patterns (spatial patterning) of forest trees, interactions between individuals and heterospecific neighboring trees caused by density-dependent effects are often neglected. The effects of negative density dependence lead us to expect that neighbourhood species segregation would increase with increasing tree size and that larger trees would be surrounded by more heterospecific neighbours than would smaller trees. We studied four mapped 1-Ha plots on Changbaishan Mountain in North-eastern China and used marked point pattern analysis to explore whether trees of different sizes exhibited differences in neighbourhood species segregation; we also determined whether larger trees were more likely to have heterospecific neighbours than smaller trees were. Our results show that bigger trees generally have higher species mingling levels. Neighborhood species segregation ranged from lower than expected levels to random or nearly random patterns at small scales as tree size classes increased under heterogeneous Poisson null model tests. This study provides some evidence in support of negative density dependent effects in temperate forests.

Abstract The discovery and creation of biocatalysts for plant biomass conversion are essential for industrial demand and scientific research of the plant cell wall. α-1,2 and α-1,3-l-arabinofuranosidases are debranching enzymes that catalyzing hydrolytic release of α-l-arabinofuranosyl residues in plant cell wall. Gene database analyses shows that GH62 family only contains specific α-l-arabinofuranosidases that play an important role in the degradation and structure of the plant cell wall. At present, there are only 22 enzymes in this group has been characterized. In this study, we cloned a novel α-1,3-arabinofuranosidase gene (poabf62a) belonging to glycoside hydrolase family 62 from Penicillium oxalicum sp. 68 and expressed it in Pichia pastoris. The molecular mass of recombinant PoAbf62A was estimated to be 32.9 kDa. Using p-nitrophenyl-α-l-arabinofuranoside (pNPαAbf) as substrate, purified PoAbf62A exhibited an optimal pH of 4.5 and temperature of 35 °C. Results of methylation and 13C NMR analyses showed that PoAbf62A was exclusively α-1,3-arabinofuranosidase, specific for cleavage of α-1,3-arabinofuranosyl residues, and with the absence of activity towards α-1,2-arabinofuranose and α-1,5-arabinofuranose. Therefore, PoAbf62A exhibits high activity on sugar beet arabinan and wheat arabinoxylan, because their branched side chain are decorated with α-1,3-arabinofuranose. On the other hand, there is a lack of activity with linear-α-l-1,5-arabinan and xylan that only contained α-l-1,5-arabinofuranose or β-1,4-xylose. The α-1,3-arabinofuranosidase activity identified here provides a new biocatalytic tool to degrade hemicellulose and analyze the structure of plant cell walls.

The NAC (NAM, ATAF and CUC) gene family plays a crucial role in the transcriptional regulation of various biological processes and has been identified and characterized in multiple plant species. However, genome-wide identification of this gene family has not been implemented in Juglans mandshurica, and specific functions of these genes in the development of fruits remain unknown. In this study, we performed genome-wide identification and functional analysis of the NAC gene family during fruit development and identified a total of 114 JmNAC genes in the J. mandshurica genome. Chromosomal location analysis revealed that JmNAC genes were unevenly distributed in 16 chromosomes; the highest numbers were found in chromosomes 2 and 4. Furthermore, according to the homologues of JmNAC genes in Arabidopsis thaliana, a phylogenetic tree was constructed, and the results demonstrated 114 JmNAC genes, which were divided into eight subgroups. Four JmNAC gene pairs were identified as the result of tandem duplicates. Tissue-specific analysis of JmNAC genes during different developmental stages revealed that 39 and 25 JmNAC genes exhibited upregulation during the mature stage in walnut exocarp and embryos, indicating that they may serve key functions in fruit development. Furthermore, 12 upregulated JmNAC genes were common in fruit ripening stage in walnut exocarp and embryos, which demonstrated that these genes were positively correlated with fruit development in J. mandshurica. This study provides new insights into the regulatory functions of JmNAC genes during fruit development in J. mandshurica, thereby improving the understanding of characteristics and evolution of the JmNAC gene family.

This article focuses on a one-dimensional nonlinear wave equation which is the Euler-Lagrange equation of a variational principle whose Lagrangian density involves linear terms and zero term as well as quadratic terms in derivatives of the field. We establish the global existence of weak solutions to its Cauchy problem by the method of energy-dependent coordinates which allows us to rewrite the equation as a semilinear system and resolve all singularities by introducing a new set of variables related to the energy.

Chokecherry (Padus virginiana L.) is an important landscaping tree with high ornamental value because of its colorful purplish-red leaves (PRL). The quantifications of anthocyanins and the mechanisms of leaf color change in this species remain unknown. The potential biosynthetic and regulatory mechanisms and the accumulation patterns of anthocyanins in P. virginiana that determine three leaf colors were investigated by combined analysis of the transcriptome and the metabolome. The difference of chlorophyll, carotenoid and anthocyanin content correlated with the formation of P. virginiana leaf color. Using enrichment and correlation network analysis, we found that anthocyanin accumulation differed in different colored leaves and that the accumulation of malvidin 3-O-glucoside (violet) and pelargonidin 3-O-glucoside (orange-red) significantly correlated with the leaf color change from green to purple-red. The flavonoid biosynthesis genes (PAL, CHS and CHI) and their transcriptional regulators (MYB, HD-Zip and bHLH) exhibited specific increased expression during the purple-red periods. Two genes encoding enzymes in the anthocyanin biosynthetic pathway, UDP glucose-flavonoid 3-O-glucosyl-transferase (UFGT) and anthocyanidin 3-O-glucosyltransferase (BZ1), seem to be critical for suppressing the formation of the aforesaid anthocyanins. In PRL, the expression of the genes encoding for UGFT and BZ1 enzymes was substantially higher than in leaves of other colors and may be related with the purple-red color change. These results may facilitate genetic modification or selection for further improvement in ornamental qualities of P. virginiana.

Patterns of tree distribution are an important attribute of forest structure and directly affect the health and stability of forest ecosystems. This paper studied the effects of forest management on the forest distribution pattern of Larix kaempferi plantations with an aim to improve the quality of the spatial structure of plantations by providing scientific grounds for near-nature distribution pattern adjustment. We set up 15 long-term positioning and monitoring plots of 20 m × 20 m of L. kaempferi plantations in Xiaolongshan, Gansu Province, China, based on the Clark and Evans index, and applied the structure analysis method of the neighboring tree relationship to determine the standard angle of the uniform angle index of regularly distributed plantation forests. The changes in forest distribution patterns were compared before and after randomized management (R1 (dumbbell-shaped random unit), R2 (torch-shaped random unit) and R1:R2 = 1:2 models) and underlayer tending and unmanaged control (CK). The results showed that (1) Under different management modes, the distribution patterns obtained by the Clark and Evans index and the mean values of the uniform angle index were entirely the same; all stand patterns were evenly distributed. (2) Regardless of randomized management or underlayer tending, the number of trees in the even distribution was the largest, accounting for more than 40%. (3) The mean values of the uniform angle index of most stands increased obviously after randomized management, among which the R1 and R2 stands showed an upward trend, while the R1:R2 = 1:2 stands showed a slight decrease. However, the mean values of the uniform angle index of the stands with underlayer management showed a decreasing trend. (4) The proportion of random trees in most stands increased significantly after randomized management, increasing by 1%~19% compared with that before adjustment, while the proportion of random trees in the underlayer tending treatment decreased by 2%~10%. Our results suggest that (1) The method based on the modified uniform angle index standard angle could effectively improve the accuracy of the distribution pattern of individual trees in plantation forests and further improve the accuracy of the overall stand pattern quantitatively. (2) Randomized management significantly increased the proportion of random trees in the stands, which accelerated the evolution of the forest distribution pattern of the L. kaempferi plantation to a random distribution.

Internet search changed the way we store and recall information and possibly altered our brain functions. Previous studies suggested that Internet search facilitates the information-acquisition process. However, this process may cause individuals to lose the ability to store and recollect specific contents. Despite the numerous behavioral studies conducted in this field, little is known about the neural mechanisms underlying Internet searches. The present study explores potential brain activity changes induced by Internet search. The whole paradigm includes three phases, namely, pre-resting state fMRI (rs-fMRI) scan, 6-day Internet search training, and post rs-fMRI scan. We detected the functional integrations induced by Internet search training by comparing post- with pre-scan. Regional homogeneity (ReHo) and functional connectivity (FC) were used to detect intra- and interregional synchronized activity in 42 university students. Compared with pre-scan, post-scan showed decreased ReHo in the temporal gyrus, the middle frontal gyrus, and the postcentral gyrus. Further seed-based FC analysis showed that the temporal gyrus exhibited decreased FC in the parahippocampal cortex and the temporal gyrus after training. Based on the features of current task and functions exhibited by these brain regions, results indicate that short-term Internet search training changed the brain regional activities involved in memory retrieval. In general, this study provides evidence that supports the idea that Internet search can affect our brain functions.

The Chinese pine (Pinus tabulaeformis Carr.) is an ecologically and economically important evergreen coniferous tree which dominates warm temperate forests throughout northern China. We established two permanent plots within the Chinese pine forest in the Jiulong Mountains, Beijing, China. To understand the structural characteristics and dynamics of these plots, we analyzed the spatial structural characteristics within nearest-neighbor relationships using the bivariate distributions of the stand spatial structural parameters: uniform angle index, W; mingling index, M; dominance index, U; and crowding index, C. Results revealed that most trees in the forest were randomly distributed. The predominant individuals and randomly arranged trees were in very dense areas and surrounded by the same species. In addition, both plots exhibited a uniform size differentiation pattern. The two plots differed mainly in the level of species mixture and dominance. The majority of reference trees in the pure Chinese pine forest (plot 1) exhibited poor species mingling and low dominance, whereas trees in the mixed Chinese pine forest (plot 2) were evenly distributed in each mingling class and most trees were of intermediate dominance. The study results are useful for optimizing forest management activities in the studied stands, promoting tree growth, regeneration and habitat diversity, and improving forest quality at a fine scale.

Internet gaming disorder (IGD), defined as the persistent use of online games with ignorance of adverse consequences, has increasingly raised widespread public concerns. This study aimed at elucidating the precise mechanisms underlying IGD by comparing intertemporal decision-making process between 18 IGD participants and 21 matched healthy controls (HCs). Both behavioral and fMRI data were recorded from a delay discounting task. At the behavioral level, the IGD showed a higher discount rate k than HC; and in IGD group, both the reaction time (delay − immediate) and the discount rate k were significantly positively correlated with the severity of IGD. At the neural level, the IGD exhibited reduced brain activations in the dorsolateral prefrontal cortex and bilateral inferior frontal gyrus compared to HC during performing delay trials relative to immediate ones. Taken together, the results suggested that IGD showed deficits in making decisions and tended to pursuit immediate satisfaction. The underlying mechanism arises from the deficient ability in evaluating between delayed reward and immediate satisfaction, and the impaired ability in impulse inhibition, which may be associated with the dysfunction of the prefrontal activation. These might be the reason why IGD continue playing online games in spite of facing severe negative consequences.

A better understanding of soil fungal communities is very useful in revealing the effects of an agroforestry system and would also help us to understand the fungi-mediated effects of agricultural practices on the processes of soil nutrient cycling and crop productivity. Compared to conventional monoculture farming, agroforestry systems have obvious advantages in improving land use efficiency and maintaining soil physicochemical properties, reducing losses of water, soil material, organic matter, and nutrients, as well as ensuring the stability of yields. In this study, we attempted to investigate the impact of a mulberry/alfalfa intercropping system on the soil physicochemical properties and the rhizosphere fungal characteristics (such as the diversity and structure of the fungal community), and to analyze possible correlations among the planting pattern, the soil physicochemical factors, and the fungal community structure. In the intercropping and monoculture systems, we determined the soil physicochemical properties using chemical analysis and the fungal community structure with MiSeq sequencing of the fungal ITS1 region. The results showed that intercropping significantly improved the soil physicochemical properties of alfalfa (total nitrogen, alkaline hydrolysable nitrogen, available potassium, and total carbon contents). Sequencing results showed that the dominant taxonomic groups were Ascomycota, Basidiomycota, and Mucoromycota. Intercropping increased the fungal richness of mulberry and alfalfa rhizosphere soils and improved the fungal diversity of mulberry. The diversity and structure of the fungal community were predominantly influenced by both the planting pattern and soil environmental factors (total nitrogen, total phosphate, and total carbon). Variance partitioning analysis showed that the planting pattern explained 25.9% of the variation of the fungal community structure, and soil environmental factors explained 63.1% of the variation. Planting patterns and soil physicochemical properties conjointly resulted in changes of the soil fungal community structure in proportion.

One of the aims of sustainable forest management is to preserve the diversity and resilience of ecosystems. Unfortunately, changes in the soil microbial communities after forest management remain unclear. We analyzed and compared the soil microbial community of a natural Quercus aliena var. acuteserrata forest after four years of four different management methods using high-throughput sequencing technology. The forest management methods were close-to-nature management (CNFM), structure-based forest management (SBFM), secondary forest comprehensive silviculture (SFCS) and unmanaged control (CK). The results showed that: (1) the soil microbial community diversity indices were not significantly different among the different management methods. (2) The relative abundance of Proteobacteria in the SBFM treatment was lower than in the CK treatment, while the relative abundance of Acidobacteria in the SBFM was significantly higher than that in the CK treatment. The relative abundance of Ascomycota was highest in the CNFM treatment, and that of Basidiomycota was lowest in the CNFM treatment. However, the relative abundance of dominant bacterial and fungal phyla was not significantly different in CK and SFCS. (3) Redundancy analysis (RDA) showed that the soil organic matter (SOM), total nitrogen (TN), and available nitrogen (AN) significantly correlated with the bacterial communities, and the available potassium (AK) was the only soil nutrient, which significantly correlated with the composition of the fungal communities. The short-term SBFM treatment altered microbial bacterial community compositions, which may be attributed to the phyla present (e.g., Proteobacteria and Acidobacteria), and the short-term CNFM treatment altered microbial fungal community compositions, which may be attributed to the phyla present (e.g., Ascomycota and Basidiomycota). Furthermore, soil nutrients could affect the dominant soil microbial communities, and its influence was greater on the bacterial community than on the fungal community.

Comprehensive evaluation of forest state is the precondition and critical step for forest management. To solve the problem that the radar plot and unit circle only focus on the value of each the evaluation index, this paper proposes a novel method for comprehensively and simultaneously evaluating the functionality and inhomogeneity of forest state based on the modified unit circle method. We evaluated the forest state of the Quercus aliena BL. var. acuteserrata Maxim. ex Wenz. broad-leaved mixed forest in the Xiaolong Mountains Forest Area of Gansu Province and the Pinus koraiensis Sieb. et Zucc. broad-leaved mixed forest in Jilin Province in China. According to the principle of comprehensive, scientific and operability, 10 evaluation indices on forest structure and vitality were selected to construct the evaluation indicator system. Each index was normalized based on the assignment method and ensured to be strictly positive based on reciprocal transformation method. The areas and arc length of the closed graph, formed by connecting every two adjacent indicators, in the radar plot and unit circle were extracted. Based on the isoperimetric theorem (isoperimetric inequality), a comprehensive evaluation model was constructed. Compared with radar chart and unit circle method, each index in the newly proposed unit circle method is represented by an independent sector region, reflecting the contribution of the index to the overall evaluation result. Each index has the same relative importance weight, contributing to the estimation the relative sizes of each aspect of forest state. The unique area and arc length of the closed graph help summarize the overall performance with a global score. The expression effect of improved unit circle has been enhanced, and as an English proverb put it, “A picture is worth a thousand words.„ The new proposed method simultaneously evaluates the functionality and inhomogeneity of the forest state and it is a powerful tool for the diagnosis of forest state problems and the decision-making of forest management.

Paenibacillus polymyxa exhibits remarkable hemicellulolytic activity. In the present study, 13 hemicellulose-degrading enzymes were identified from the secreted proteome of P. polymyxa KF-1 by liquid chromatography-tandem mass spectrometry analysis. α-L-arabinofuranosidase is an important member of hemicellulose-degrading enzymes. A novel α-L-arabinofuranosidase (PpAbf51b), belonging to glycoside hydrolase family 51, was identified from P. polymyxa. Recombinant PpAbf51b was produced in Escherichia coli BL21 (DE3) and was found to be a tetramer using gel filtration chromatography. PpAbf51b hydrolyzed neutral arabinose-containing polysaccharides, including sugar beet arabinan, linear-1,5-α-L-arabinan, and wheat arabinoxylan, with L-arabinose as the main product. The products from hydrolysis indicate that PpAbf51b functions as an exo-α-L-arabinofuranosidase. Combining PpAbf51b and Trichoderma longibrachiatum endo-1,4-xylanase produced significant synergistic effects for the degradation of wheat arabinoxylan. The α-L-arabinofuranosidase identified from the secretome of P. polymyxa KF-1 is potentially suitable for application in biotechnological industries.

Aim of the study: Neighborhood-based stand spatial structure parameters can quantify and characterize forest spatial structure effectively. How these neighborhood-based structure parameters are influenced by the selection of different numbers of nearest-neighbor trees is unclear, and there is some disagreement in the literature regarding the appropriate number of nearest-neighbor trees to sample around reference trees. Understanding how to efficiently characterize forest structure is critical for forest management.Area of study: Multi-species uneven-aged forests of Northern ChinaMaterial and methods: We simulated stands with different spatial structural characteristics and systematically compared their structure parameters when two to eight neighboring trees were selected.Main results: Results showed that values of uniform angle index calculated in the same stand were different with different sizes of structure unit. When tree species and sizes were completely randomly interspersed, different numbers of neighbors had little influence on mingling and dominance indices. Changes of mingling or dominance indices caused by different numbers of neighbors occurred when the tree species or size classes were not randomly interspersed and their changing characteristics can be detected according to the spatial arrangement patterns of tree species and sizes.Research highlights: The number of neighboring trees selected for analyzing stand spatial structure parameters should be fixed. We proposed that the four-tree structure unit is the best compromise between sampling accuracy and costs for practical forest management.

OBJECTIVE: This study modeled win and lose trials in a simple gambling task to examine the effect of entire win-lose situations (WIN, LOSS, or TIE) on single win/lose trials and related neural underpinnings. METHODS: The behavior responses and brain activities of 17 participants were recorded by an MRI scanner while they performed a gambling task. Different conditions were compared to determine the effect of the task on the behavior and brain activity of the participants. Correlations between brain activity and behavior were calculated to support the imaging results. RESULTS: In win trials, LOSS caused less intense posterior cingulate activity than TIE. In lose trials, LOSS caused more intense activity in the right superior temporal gyrus, bilateral superior frontal gyrus, bilateral anterior cingulate, bilateral insula cortex, and left orbitofrontal cortex than WIN and TIE. CONCLUSIONS: The experiences of the participants in win trials showed great similarity among different win-lose situations. However, the brain activity and behavior responses of the participants in lose trials indicated that they experienced stronger negative emotion in LOSS. The participants also showed an increased desire to win in LOSS than in WIN or TIE conditions.

PP2C-type phosphatases play roles in signal transduction pathways related to abiotic stress. The cyanobacterial PP2C-type phosphatase tPphA specifically dephosphorylates the PII protein, which is a key regulator in cyanobacteria adapting to nitrogen-deficient environments. Previous studies have shown that residue His39 of tPphA is critical for the enzyme’s recognition of the PII protein; however, the manner in which this residue determines tPphA substrate specificity is unknown. Here, we solved the crystal structure of H39A, a tPphA variant. The structure revealed that the mutation of residue His39 to alanine changes the conformation and the flexibility of the loop in which residue His39 is located, and these changes affect the substrate specificity of tPphA. Moreover, previous studies have assumed that the FLAP subdomain and the third metal (M3) of tPphA could mutually influence each other to regulate PP2C catalytic activity and substrate specificity. However, despite the variable conformations adopted by the FLAP subdomain, the position of M3 was consistent in the tPphA structure. These results indicate that the FLAP subdomain does not influence M3 and vice versa. In addition, a small screen of tPphA inhibitors was performed. Sanguinarine and Ni2+ were found to be the most effective inhibitors among the assayed chemicals. Finally, the dimeric form of tPphA was stabilized by cross-linkers and still exhibited catalytic activity towards p-nitrophenyl phosphate.

In this paper, we establish the global existence of weak solutions to the initial-boundary value and initial value problems for two classes of nonlinear wave equations which are the Euler-Lagrange equation of a variational principle. We use the method of energy-dependent coordinates to rewrite these equations as semilinear systems and resolve all singularities by introducing a new set of dependent and independent variables. The global weak solutions can be constructed by expressing the solutions of these semilinear systems in terms of the original variables. [ABSTRACT FROM AUTHOR]
