Residual δ-ferrite is ubiquitous in M50NiL bearing steel and seriously deteriorates impact toughness. In the present work, the formation mechanism of δ-ferrite in M50NiL steel and the influence of element segregation on its formation process were investigated. The residual δ-ferrite was proved to form directly from the final liquid phase at the end of solidification due to serious segregation of Mo and V elements. Calculation results indicate that the segregation of Mo and V elements expanded the “L + δ'' two-phase region and stabilized δ-ferrite. Due to the high concentration of Mo and V, large-sized V-rich MC and Mo-rich M2C carbides would precipitate in δ-ferrite at 900–1200 °C if the cooling rate is slow. When kept at 1200 °C for no more than 30 min, Mo and V elements derived from the decomposition of large-sized carbides are preferentially diffused into δ-ferrite, enhancing the stability of δ-ferrite. To achieve the effective elimination of δ-ferrite in M50NiL steel, it is necessary to keep the ingot at 1200 °C for at least 20 h.

Abstract Untethered miniature soft robots have significant application potentials in biomedical and industrial fields due to their space accessibility and safe human interaction. However, the lack of selective and forceful actuation is still challenging in revolutionizing and unleashing their versatility. Here, we propose a focused ultrasound-controlled phase transition strategy for achieving millimeter-level spatially selective actuation and Newton-level force of soft robots, which harnesses ultrasound-induced heating to trigger the phase transition inside the robot, enabling powerful actuation through inflation. The millimeter-level spatial resolution empowers single robot to perform multiple tasks according to specific requirements. As a concept-of-demonstration, we designed soft robot for liquid cargo delivery and biopsy robot for tissue acquisition and patching. Additionally, an autonomous control system is integrated with ultrasound imaging to enable automatic acoustic field alignment and control. The proposed method advances the spatiotemporal response capability of untethered miniature soft robots, holding promise for broadening their versatility and adaptability.

A new type of lightweight high aluminum bearing steel had been developed based on the widely used high carbon chromium bearing steel. Via multi-scale experimental characterizations and theoretical calculations, here we mainly focused on the evolution of microstructure including precipitates under different quenching temperatures. Accordingly, the mechanical properties, mechanisms and fracture features of the new high-Al steel after tempering were characterized in detail. As austenitizing temperature rose, matrix of steel varied from multiphase structure of martensite, retained austenite (RA) and ferrite to the full martensite and RA. RA was found in all four samples with the austenitizing temperature from 850 °C to 1000 °C. In terms of precipitates, a transformation from M3C to M7C3 were observed. As a phase newly introduced by Al, AlFe3C (κ carbides) would dissolve into matrix at 900 °C. Eventually, the yield strength of the researched steel could reach 1430 MPa with an unnotched impact energy of 35 J. Theoretical results showed that high strength of researched steel was mainly caused by solid solution effect of carbon and high density of dislocation. In terms of toughness and fracture observation, high impact energy was attributed to the highest density of grain boundary, and also, κ carbides were found at the origin of secondary crack. This study highlighted the significant effect of Al addition and quenching process based on the traditional bearing steel, which showed a novel method and idea to reduce significantly the weight of bearings.

Abstract Background Apoptosis is involved (directly and indirectly) in several physiological processes including tissue remodeling during the development, the turnover of immune cells, and a defense against harmful stimuli. The disordered apoptotic process participates in the pathogenesis of various diseases, such as neoplasms, and chronic inflammatory or systemic autoimmune diseases, which are associated with its inadequate regulation. Caspases are vital components of the apoptotic pathway that are involved in developmental and immune processes. However, genome-wide identification and functional analysis of caspase have not been conducted in Mytilus coruscus, which is an economically important bivalve. Results Here, 47 caspase genes were identified from the genomes of M. coruscus, and the expansion of caspase-2/9 and caspase-3/6/7 genes were observed. Tandem duplication acts as an essential driver of gene expansion. The expanded caspase genes were highly diverse in terms of sequence, domain structure, and spatiotemporal expression profiles, suggesting their functional differentiation. The high expression of the expanded caspase genes at the pediveliger larvae stage and the result of apoptosis location in the velum suggest that the apoptosis mediated by them plays a critical role in the metamorphosis of M. coruscus larvae. In gill, caspase genes respond differently to the challenge of different strains, and most caspase-2/9 and caspase-3/6/7 genes were induced by copper stress, whereas caspase-8/10 genes were suppressed. Additionally, most caspase genes were upregulated in the mantle under ocean acidification which could weaken the biomineralization capacity of the mantle tissue. Conclusions These results provide a comprehensive overview of the evolution and function of the caspase family and enhanced the understanding of the biological function of caspases in M. coruscus larval development and response to biotic and abiotic challenges.

This study delves into Ulcerative colitis (UC), a persistent gastrointestinal disorder marked by inflammation and ulcers, significantly elevating colorectal cancer risk. The emergence of single-cell RNA sequencing (scRNA-seq) technology has opened new avenues for dissecting the intricate cellular dynamics and molecular mechanisms at play in UC pathology. By analyzing scRNA-seq data from individuals with UC, our study has revealed a consistent enhancement of inflammatory response pathways throughout the course of the disease, alongside detailing the characteristics of endothelial cell damage within colitis environments. A noteworthy finding is the downregulation of Phospholysine Phosphohistidine Inorganic Pyrophosphate Phosphatase (LHPP), which exhibited a inversely correlate with STAT3 expression levels. The markedly reduced expression of LHPP in both the tissues and plasma of UC patients positions LHPP as a compelling target for therapeutic intervention. Our findings highlight the pivotal role LHPP could play in moderating inflammation, spotlighting its potential as a crucial molecular target in the quest to understand and treat UC.

Bacterial biofilms are widely found in nature, and they can be a major factor in many local chronic infections inside the human body. To date, effective elimination of biofilms is intractable for conventional antibiotic therapies due to the existence of bacteria‐protecting extracellular polymeric substances. For biofilm eradication in tiny tubular structures, magnetic hydrogel micromachines (MHM) are designed that are capable of performing mechanical disruption of biofilms while releasing antibacterial agents in a controllable fashion. Two modes of motion controlled by external magnetic fields, namely planar rotation and wobbling modes, have been investigated to drive the micromachines to interact and destruct biofilms. In addition, the presented micromachines are composed of thermosensitive PNIPAm hydrogel that can load with the antibacterial agent (H2O2) and release them upon heating. The Fe3O4 nanoparticles embedded in the hydrogel matrix act as catalysts for the Fenton reaction, generating bacteria‐killing free radicals. The synergetic biofilm elimination effect has been experimentally demonstrated in well plates and small tubes. The presented MHMs may provide a new design to treat biofilm‐related infections in narrow and confined spaces.

Plant height and leaf area index (LAI) are crucial growth indicators that reflect the growth status of tomatoes in greenhouses, enabling accurate determinations to effectively estimate crop transpiration and formulate irrigation strategies for reducing agricultural water waste. There is a need for the increased application of related models to simulate tomato growth indices in the traditional greenhouse production in China. This study proposes a nondestructive, real-time monitoring and simulation device for measuring tomato plant height and leaf area index. The weight of aboveground tomatoes was obtained by suspending tomato plants on dynamometers, while the total weight of stem and leaf organs was determined using a distribution coefficient simulation model. The R2 value between the measurements from the electronic scale and those from the aboveground fresh weight device for tomatoes was 0.937, with an RMSE value of 0.05 kg. The monitoring device did not affect the average tomato growth during operation. The device will not affect the growth of tomatoes during monitoring. A multiple linear regression was used to compare the measured and simulated values of the plant height and leaf area index of various types of greenhouse tomatoes cultivated in different greenhouse types. The average R2 value for simulating plant height was 0.817 with an RMSE of 10.81 cm. The average R2 value for the leaf area index was 0.854, with an RMSE of 0.55 m2·m−2. The simulated values for plant height and leaf area index closely matched the measured values, indicating that the model has high accuracy and applicability in traditional Chinese greenhouses (solar greenhouses and insulated plastic greenhouses). However, further optimization is required for commercially produced, continuous plastic greenhouses equipped with greenhouse environmental control equipment.

This study constructs a power switching device open-circuit fault diagnosis model for a three-level neutral point clamped inverter based on the multi-scale shuffled convolutional neural network (MSSCNN) and extracts and classifies the fault information contained in the output current of inverters. The model employs depthwise separable convolution and channel shuffle techniques to improve diagnostic accuracy and reduce model complexity. The experimental results show that the new model has lower model complexity, better noise resistance and higher average diagnostic accuracy compared with fault diagnosis models based on CNN, ResNet, ShuffleNet V2 and Mobilenet V3 networks.

In cold regions, the low irrigation water temperature is an important factor of low-temperature stress for greenhouse crops. In this paper, an irrigation water-heating system (IWHS) is proposed to increase the water temperature by utilizing the excess heat in the solar greenhouse. The heat-collection capacity of the system was analyzed by screening the IWHS process parameters in a Chinese solar greenhouse, and a warm-water irrigation experiment for lettuce was conducted. The results demonstrated that the water temperature increased with the increase in wind speed, and the increase in daily average water temperature reached the maximum value of 8.6 °C at 4.5 m/s wind speed. When the heat exchanger was installed at a height of 3.0 m, the collector capacity increased by 17.8% and 6.0% compared with the heating capacity at 0 m and 1.5 m, respectively, and the operation termination water temperature was 22.0–32.2 °C and its coefficient of performance (COP) was optimal. Surface darkening of the heat exchanger did not affect the heat-collection capacity of the system. Using the IWHS effectively improved the temperature of lettuce irrigation water in the Chinese solar greenhouse. The increased frequency of warm-water irrigation significantly promoted lettuce growth and increased the average yield per plant by 15.9%. Therefore, IWHS effectively increased the irrigation water temperature in a Chinese solar greenhouse in winter. Improving the system would enhance its economic and application value.

The magnetic field has unique advantages in manipulating miniature robots working inside the human body, such as high transparency to biological tissue and good controllability for field generation. Generally, the actuated magnetic robot can be classified into two categories: tethered devices like intravascular microcatheters and untethered devices like helical swimmers. Among these, the tethered devices have a long history and good clinical application prospects, considering their high‐dose delivery and easy removal after the procedure. As an evolution of traditional continuum medical devices, the integration with magnetic actuation provides them with better scalability and improved dexterity. Although rapidly developed in the last two decades, the field of tethered magnetic robots requires further advancements in terms of design, fabrication, modeling, and control, especially for clinical applications. Herein, the recent progress of magnetically actuated continuum medical robots is focused on, intending to offer readers a comprehensive survey of the state‐of‐the‐art technologies and an information collection for future system design.

Peer conflicts commonly happen in children’s daily interaction, and strategies they employed to deal with the conflicts have an impact on peer conflict resolution. It has been illustrated that children’s understanding of emotion plays an important role in social communication. However, there is little research focusing on the relation between emotional comprehension and peer conflict resolution strategies. In this study, 90 children of 3 to 6 finished the Test of Emotional Comprehension, and their preschool teachers were required to complete the Conflict Resolution Strategy Questionnaire, which scored each child’s conflict resolution strategies. The results showed that: (a) the preference of conflict resolution strategies differed in age, and girls tended to adopt positive strategies; (b) children’s emotional comprehension grew with age; and (c) children’s conflict resolution strategies and emotional comprehension were closely related. Children’s emotional comprehension can positively predict the overall conflict resolution strategies and negatively predicted negative strategies, whereas mental emotional comprehension can positively predict positive strategies. The factors affecting children’s emotional comprehension and conflict resolution strategies, and their relationship were discussed in depth.

Hot-compression bonding with subsequent soaking treatment is employed to manufacture large high-speed homogeneous steels in the current study. The effects of the deformation temperature (1050, 1100, and 1150 °C), strain (10%, 30%, and 50%), and holding time (1 and 12 h) on the interfacial microstructure and tensile strength of joints were investigated. The results indicated that complete bonding occurred at 1050 °C/50%, 1100 °C/30%, 1100 °C/50%, 1150 °C/30%, and 1150 °C/50%, and the tensile strength of the obtained joints was comparable to that of the base material. In other cases, interfacial voids were formed along the straight interface, which deteriorated the bonding strength. These voids were eliminated by a post-holding treatment at 1150 °C for 12 h, promoting the transformation of a straight interface to bulged grain boundaries. This resulted in the complete healing of the interface, except for samples bonded at 1050 °C/10%. The bulging of the original bonding interface due to the plastic deformation and atomic diffusion was the main mechanism for interfacial healing. Stable interfacial oxide nanoparticles were formed under a high vacuum and did not affect the recovery of joint strength. To validate the practicality of this method, an integrated high-speed steel with a homogeneous tensile strength was manufactured by bonding four pieces of blanks.

Glutathione peroxidase 8 (GPX8), located in the endoplasmic reticulum, is associated with poor prognosis in several cancers. However, the expression and functions of GPX8 in cancers remain unclear. The purpose of this study was to explore the expression and functions of GPX8 in glioblastoma (GBM). We obtained expression data of GPX8 by accessing the TCGA, CGGA, GEPIA, and TIMER2.0 databases and validated them using western blot and immunohistochemistry. The Kaplan–Meier overall survival curve and Cox regression model were used to evaluate the prognostic value of GPX8 in glioma patients. Gene ontology (GO) and function enrichment analysis were used to investigate the potential function of GPX8 in GBM. Correlation analysis was used to clarify the role of GPX8 in proneural–mesenchymal transition (PMT). We studied the correlation between GPX8 expression and GBM immune infiltration by accessing cBioPortal and TIMER2.0 databases. Here, we demonstrated that GPX8 was significantly upregulated in GBM, and was associated with IDH-wildtype and mesenchymal subtype with poor prognosis. Survival analysis results indicated that GPX8 is an independent prognostic factor for overall survival (OS) in all WHO-grade glioma patients. Through the functional studies, we found that high expression of GPX8 correlated with mesenchymal signature and negatively correlated with proneural signature, indicating that GPX8 might promote PMT in GBM. Finally, based on correlation analysis, we found that the expression of GPX8 was associated with immune infiltration and the IL1/MYD88/IRAK/NF-κB pathway in GBM. Our results show that GPX8 is a key factor affecting the prognosis of GBM patients, and its targeting has the potential to provide a novel therapeutic approach.

Tyrosinase is a type-3 copper protein with six conserved histidine residues within the copper-binding sites. It participates in mollusk nacre formation. Here, we identified nacreous-layer-specific tyrosinases (NLSTyr) from Pinctada fucata martensii (PmTyr-4 and PmTyr-6), as well as their homologs in Pinctada maxima (PmaxTyr and PmaxTyr4) and Pinctada margaritifera (PmarTyr and PmarTyr-4), which encoded tyrosinases without the six conserved histidine residues within the copper-binding sites. PmTyr-4 and PmTyr-6 mRNAs were spatially concentrated in the mantle central and pearl sac, which are the organs responsible for nacre formation. During shell regeneration and pearl formation, PmTyr-4 and PmTyr-6 were also significantly highly expressed in the mantle and pearl sac. RNA interference showed that PmTyr-4 participated in nacreous-layer formation. The recombinant protein of PmTyr-4 (rPmTyr-4) inhibited the calcium carbonate precipitation rate. Correspondingly, calcium carbonate crystallization assay showed that the aragonite crystals of the rPmTyr-4 group were smaller than those of the control group. Moreover, the calcite and aragonite morphologies of the rPmTyr-4 group were modified compared with the control group. These results suggested that NLSTyr in pearl oyster inhibited calcium carbonate precipitation and affected crystal morphologies during nacre formation. Our findings provided new insights into the evolution and function gain of tyrosinase in Mollusk.

ObjectivesThis is a comparative cohort study aiming to evaluate the mortality risk factors for patients with nosocomial meningitis (NM) induced by multidrug-resistant Enterobacteriaceae (MDRE) in China. The clinical features and therapies of patients and the resistance mechanisms of MDRE pathogens were also assessed.MethodsMDRE-NM patients from two neurosurgical centers in China from 2014 to 2019 were included in this study. Clinical features were extracted from the medical record databases of the two centers. The molecular mechanisms underlying the microbiological resistance mechanisms of each MDRE pathogen were determined, Kaplan–Meier survival analysis was conducted, and multivariable analyses were performed using a Cox proportional hazard model.ResultsNinety MDRE-NM patients were included in this study. Klebsiella pneumoniae accounted for the highest proportion of causative pathogens (46/90, 51.1%), and 40 causative pathogens (44.4%) were meropenem-resistant. blaKPC (27/40, 67.5%) was the predominant carbapenem resistance gene. Multivariate Cox analysis showed that external ventricular drainage (EVD) [hazard ratio (HR) = 2.524, 95% confidence interval (CI) = 1.101–5.787, p = 0.029] and a Glasgow Coma Scale (GCS) score ≤;8 (HR = 4.033, 95% CI = 1.526–10.645, p = 0.005) were mortality risk factors for patients with MDRE-NM. A total of 90.0%, 94.4%, and 97.8% of MDRE-NM patients received antibiotic prophylaxis (AP), antibiotic empirical therapy (AET), and antibiotic definitive therapy (ADT), respectively.ConclusionsNM caused by MDRE is an important sign of the failure of neurosurgery. MDRE possesses multiple drug resistance genotypes, and EVD and a GCS score ≤;8 are independent mortality risk factors for patients with MDRE-NM, which deserve the attention of microbiologists and neurosurgical clinicians.

Abstract Background To determine the phenotype, molecular characterisation and risk factors of postoperative meningitis induced by Extended-spectrum β-lactamase (ESBL)-producing Enterobacteriaceae (EPE) in China. Methods We performed a multi-centre comparative cohort study of postoperative meningitis patients infected with Enterobacteriaceae in 4 neurosurgical centres in China from January 2014 to December 2019. Phenotype and molecular characteristics of the isolates were reviewed and tested, and independent risk factors of the EPE meningitis were evaluated by binary logistic regression. Results In total, 220 Enterobacteriaceae include 78 EPE were available in this study. 85.6% (67/78) ESBL-related genes were tested, and bla SHV (14.9%) and bla SHV + bla TEM + bla CTX-M-9 (20.9%) were found to be the most frequent mono and combined ESBL-related genes harboured by Enterobacteriaceae. On binary logistic analysis, craniotomy (OR. 2.583, 95% C.I. 1.274–5.235, P = 0.008) and malignancy (OR. 2.406, 95% C.I. 1.299–4.456, P = 0.005) were the associated independent risk factors to meningitis induced by EPE. Conclusions To the best of our knowledge, this is the largest series focusing on risk factors of EPE meningitis which has been conducted in China. Craniotomy and malignancy were independent risk factors for EPE meningitis. The risk factors identified may be further utilized in clinical practice and research to avoid and reduce the mortality in future.

Abstract Background Nicotinic acetylcholine receptors (nAChRs) are among the oldest and most conserved transmembrane receptors involved in signal transduction. Despite the prevalence and significance of cholinergic signaling, the diversity and evolution of nAChRs are not fully understood. Result By comparative genomic analysis, we found massive expansions of nAChR genes in molluscs and some other lophotrochozoans. The expansion is particularly pronounced in stationary bivalve molluscs with simple nervous systems, with the number of nAChR genes ranging from 99 to 217 in five bivalves, compared with 10 to 29 in five ecdysozoans and vertebrates. The expanded molluscan nAChR genes tend to be intronless and in tandem arrays due to retroposition followed by tandem duplication. Phylogenetic analysis revealed diverse nAChR families in the common ancestor of bilaterians, which subsequently experienced lineage-specific expansions or contractions. The expanded molluscan nAChR genes are highly diverse in sequence, domain structure, temporal and spatial expression profiles, implying diversified functions. Some molluscan nAChR genes are expressed in early development before the development of the nervous system, while others are involved in immune and stress responses. Conclusion The massive expansion and diversification of nAChR genes in bivalve molluscs may be a compensation for reduced nervous systems as part of adaptation to stationary life under dynamic environments, while in vertebrates a subset of specialized nAChRs are retained to work with advanced nervous systems. The unprecedented diversity identified in molluscs broadens our view on the evolution and function of nAChRs that are critical to animal physiology and human health.

The polyamine putrescine (Put) is a ubiquitous small cationic amine. It plays an essential role in controlling the innate immune response. However, little is known about its function in mollusks. In this study, the Put content was observed to increase in the serum of pearl oyster Pinctada fucata martensii after 6 and 24 h of lipopolysaccharide (LPS) stimulation. Activities of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) increased, and nitric oxide synthase was downregulated in the Put group (i.e., combined treatment with Put and LPS) compared with that in the LPS group (i.e., combined treatment with phosphate-buffered saline and LPS). Furthermore, activities of alkaline phosphatase and acid phosphatase were inhibited after 6 h of LPS stimulation. The expression levels of the nuclear factor kappa B, IκB kinase, Janus kinase, and signal transducer and activator of transcription proteins genes were all significantly suppressed at 12 and 24 h in the Put group. Pseudomonas aeruginosa and Bacillus subtilis grew better after being incubated with the serum from the Put group than that from the LPS group. Additionally, the Put treatment remarkably inhibited the autophagy of hemocytes mediated by the AMP-activated protein kinase-mammalian target of rapamycin-Beclin-1 pathway. This study demonstrated that Put can effectively inhibit the inflammatory response induced by LPS in pearl oysters. These results provide useful information for further exploration of the immunoregulatory functions of polyamines in bivalves and contribute to the development of immunosuppressive agents.

The exoskeleton outside the soft body in shelled Mollusca defends against physical damage and parasite invasion and frequently undergoes biological and abiotic disturbance in natural and farmed environments, causing shell damage and environmental exposure, thereby threatening survival. To investigate the immune response and biocalcification after shell damage in pearl oyster Pinctada fucata martensii, a semi-sessile bivalve with a well-developed shell structure, we constructed a shell damage repair model and detected the transcriptome of mantle edge. In the first stage (4–12 h post-shell notching) the suppression of the secretion of the shell matrix protein (SMP) and activation of immune signaling pathways suggested the functional transformation of the mantle from biomineralization to immune defense. In the second stage (12–24 h), the enhancement of protein synthesis and processing, protein export-related functions, and energy production-related pathways indicated that the mantle prepared materials and energy for repairing the damaged shell and tissues. In the third stage, the upregulation of 68 SMPs involved in the shell formation indicated the reopening of shell fabrication and the repair of mantle tissue cells from immune defense to biomineralization. Furthermore, some SMPs burdened immune response molecules and the controller of biomineralization, which indicated the crosslinking between immune response and biomineralization. Our research outlines three stages an individual suffers during shell damage and provides insights into immune response and biocalcification crosslinking in pearl oyster and improves understanding of how the mantle tissue can quickly respond to environmental threats.

Macrosegregation is the key issue in the solidification field. Oxygen and its inclusions play the important role in driving the melt flow and the resulting macrosegregation in steel. Here, to reveal the inducing mechanism and quantitative model of oxygen content in real industrial steel ingots, we demonstrate for the first time that there exists the critical oxygen content in triggering the formation of channel-type segregation, the most undesirable macrosegregation type in steel. Our multiscale simulations from density functional theory calculations to multiphase/multicomponent macromodel, clarify the quantitative conditions initializing channel-type segregation and reveal two typical growth modes via oxide flotation. The oxygen content model and criterion to induce the channel onset is built accordingly, which are validated by the numerous full ingot dissections and experimental characterizations including the in situ electrolysis of inclusions, X-ray microtomography, scanning electron microscope, large-scale measurement system of inclusions and chemical analysis. With oxygen controlled below this critical value of 0.0008 wt%, channel disappears. This study quantitatively uncovers the novel role of oxygen in steel, changes the traditional sole concept of cleanliness, and highlights an innovative and controlling-effective route to fabricate homogenized steel.

The immune response after transplantation during pearl production is a major factor that cause nucleus rejection and death of the host pearl oysters. To find out the crucial metabolites involved in the immune response after transplantation, we performed metabolomic analysis of the serum before and after transplantation from pearl oyster Pinctada fucata martensii. The analysis detected a total of 201 metabolites, which included many amino acids. Among the 201 metabolites, 35 were revealed as significantly diff ;erential metabolites (SDMs), such as acetylcholine (ACh) and d-glutamine. Pathway analysis indicated that the identified SDMs were involved in six pathways, such as sphingolipid metabolism and d-glutamine and d-glutamate metabolism. Transcriptomic analysis showed 19 and 18 nAChRs were induced by 2–3000-fold through the pre-conditioning treatment or transplantation surgery, respectively. Dual-Luciferase analysis demonstrated that overexpression of nAChRs downregulated the luciferase activity of the pNF-κB-Luc reporter, and stimulation of nAChRs could inhibit the LPS-induced up-regulation of NF-κb, IKK and IL-17, suggesting that the upregulated ACh after transplantation may function through nAChRs by inhibiting NF-κb signal pathway. These findings could help elucidate the corresponding metabolites after transplantation, thereby providing useful information in regulating the immune response to improve the survival rate of pearl oyster after transplantation.

A high-temperature environment is one of the most important factors limiting the growth of crops in Chinese solar greenhouses during summer. To reduce the substrate temperature of summer plant cultivation in a Chinese solar greenhouse, we proposed a water-circulating tomato-root zone-substrate-cooling system (WCTRZSCS). The system used water as the circulating medium, a chiller as the cooling source, and polyethylene raised temperature resistance (PE-RT) pipes laid in the substrate as the cooling component. The greenhouse was divided into test area TS1 (one PE-RT pipe), TS2 (two PE-RT pipes), and a control area CK (no PE-RT pipe) for the root-zone substrate-cooling test. The results demonstrated that (1) in the summer, WCTRZSCS can effectively reduce the substrate temperature, and (2) WCTRZSCS improves the temperature conditions for tomato vegetative growth. There were significant differences in plant height, stem diameter, dry weight, fresh weight, leaf area, net photosynthetic rate, total root length, and total root projection area between tomatoes in the test and control areas (p < 0.05). The TS1 and TS2 growth rates were 60.2% and 81.2% higher than CK, respectively, and the light-utilization efficiency was 56.3% and 81.3% higher than CK. (3) The system’s cooling energy consumption per unit ground area was 35.2~67.5 W·m−2, and the coefficient of performance (COP) was 5.3~8.7. Hence, WCTRZSCS can effectively reduce the substrate temperature in the root zone, but the profit by tomato cannot offset the cost of using WCTRZSCS. Through the optimization of and improvement in the system, its economy may be further improved, and it is expected to be applied in practical production.

The thermophysical properties of a working-fluid play an important role in the process of stirring-heating. The heating process of stirring is accompanied by two processes: the friction between the solid mechanism and the working-fluid and the viscous dissipation of the working liquid. Traditionally, the sensible heat of water-based working-fluids is low, while that of oil-based working-fluids is higher, but the load capacity is relatively low. In order to find a balance between the two, an optimal stirring working-fluid should be selected. In this study, an experimental method was used to study the heating process of 30 kinds of working-fluids. The numerical evaluation model of the effects of thermophysical properties on the comprehensive evaluation index of heat (CEIH) was established by multiple linear regression methods, and a computational fluid dynamics (CFD) tool was used to analyze the heat generation and flow field of different working-fluids in the stirring-heating device. The results show that viscous dissipation is the most important way of stirring-heating. CFD can completely replace the experiment to study the heating effect of stirring. The thermophysical properties of the working-fluid affect the upper circulation and the overall velocity of the double circulation flow. The experimental results and regression model analysis show that specific heat capacity has the greatest effect on the heating effect, but density will also play a positive role in the stirring-heating. Water-based salt solutions such as KCl can achieve a better heating effect, and oil-based working-fluids are not always the best choice.

In applications where motor inversion is forbidden, it is important to detect the initial rotor position of the motor. For this reason, based on coupling injection of high-frequency signal, a novel method of initial rotor position detection for brushless DC motors (BLDCM) is proposed in this paper. Firstly, the proposed method detects the relationship between three-phase winding inductances by injecting the high-frequency detection signal into motor windings in a coupling way, and the initial rotor position is determined into two sectors with 180 degrees electric angle difference. Then, the polarity of the permanent magnet rotor is determined by applying two opposite voltage vectors to motor windings, so that the initial rotor position is determined into a unique sector, and the positioning accuracy is 30 degrees electric angle. The proposed method significantly reduces the amplitude of the detection signal while increases its frequency by the way of coupling injection, thus reducing the response current and electromagnetic torque generated by the high-frequency signal and reducing the possibility of rotor inversion. Finally, the effectiveness of the proposed method is verified by experimental results.

The internal air temperature of Chinese solar greenhouse (CSG) has the problem of uneven spatial and temporal distribution. To determine temperature distribution at different locations, we designed a greenhouse temperature real-time monitoring system based on virtual local area network (VLAN) and estimate, including interpolation estimation module, data acquisition, and transmission module. The temperature data were obtained from 24 sensors, and the Ordinary Kriging algorithm estimated the temperature distribution of the whole plane according to the data. The results showed that the real-time temperature distribution monitoring method established was fast and robust. In addition, data validity rate for VLAN technology deployed for data transmission was 2.64% higher than that of cellular network technology. The following results are obtained by interpolation estimation of temperature data using gaussian model. The average relative error (ARE) of estimate, mean absolute error (MAE), root mean square error (RMSE), and determination coefficient (R2) were −0.12 °C, 0.42 °C, 0.56 °C, and 0.9964, respectively. After simple optimization of the number of sensors, the following conclusions are drawn. When the number of sensors were decreased to 12~16, MAE, RMSE, and R2 were 0.40~0.60 °C, 0.60~0.80 °C, and >0.99, respectively. Furthermore, temperature distribution in the greenhouse varied in the east–west and north–south directions and had strong regularity. The calculation speed of estimate interpolation algorithm was 50~150 ms, and greenhouse Temperature Distribution Real-time Monitoring System (TDRMS) realized simultaneous acquisition, processing, and fast estimate.

The accurate estimation of rotor position and speed before flying restart is of great significance to improve the operation reliability of permanent magnet synchronous motor systems. The traditional multizero vector short-circuit method can improve the estimation accuracy of speed and rotor position, but the increased number of short-circuits reduces the electromagnetic torque response speed after the power supply recovers. In order to accurately estimate the initial speed and rotor position before the flying restart and effectively improve the electromagnetic torque response speed, a shoot-through zero vector short-circuit method based on quasi-Z-source inverter (qZSI) is proposed. This method breaks the limitation of regulating DC link voltage under the normal operation of the motor in the conventional methods, and puts forward a new idea of advancing the regulation of the DC link voltage to the stage of abnormal operation before the motor restarts. By designing the insertion mode of the mixed vectors and analyzing the action time of each vector before the flying restart, the accurate estimation of position and speed is realized and, meanwhile, the boost of the qZSI’s DC link voltage is achieved, thus giving the sensorless flying restart method a faster torque response speed for the PMSM system driven by qZSIs.

Abstract Background Congenital cataract is the most frequent cause of blindness during infancy or early childhood. To date, more than 40 loci associated with congenital cataract have been identified, including at least 26 genes on different chromosomes associated with inherited cataract. This present study aimed to identify the genetic mutation in a six-generation Chinese family affected with congenital cataract. Methods A detailed six-generation Chinese cataract family history and clinical data of the family members were recorded. A total of 27 family members, including 14 affected and 13 unaffected individuals were recruited. Whole exome sequencing was performed to determine the disease-causing mutation. Sanger sequencing was used to confirm the results. Results A known missense mutation, c. 139G > A (p. D47N), in Cx50 was identified. This mutation co-segregated with all affected individuals and was not observed in the unaffected family members or in 100 unrelated controls. The homology modeling showed that the structure of the mutant protein was different with that wild-type Cx50. Conclusions The missense mutation c.139G > A in GJA8 gene is associated with autosomal dominant congenital cataract in a six-generation Chinese family. The result of this present study provides further evidence that the p. D47N mutation in CX50 is a hot-spot mutation.

Channel segregation (CS) is the most typical defect during solidification of NbTi alloy. Based on numerical simulation and experimental characterizations, we deeply elucidated its characteristics, formation mechanism, effecting factor and prediction criterion. According to acid etching, industrial X-ray transmission imaging, 3D X-ray microtomography and chemical analysis, it was found that in a casing ingot, by He cooling, finer grain size, weaker segregation and slighter CS can be obtained compared with air-cooled ingot. The simulation results of macrosegregation show that CS is caused by the strong natural convection in the mushy zone triggered by the thermo-solutal gradient. Its formation can be divided into two stages including channel initiation and growth. In addition, due to the stronger cooling effect of the He treatment, the interdendritic flow velocity becomes smaller, consequently lowering the positive segregation and CS and improving the global homogenization of the final ingot. Finally, to predict the formation of CS, the Rayleigh number model was proposed and its critical value was found to be 15 in NbTi alloy for the first time. When it is lower than the threshold, CS disappears. It provides an effective tool to evaluate and optimize the solidification parameters to fabricate the homogenized NbTi ingot in engineering practice.

The influence of hot top design on feeder channel segregates (F-CS) and centerline shrinkage porosities (C-SP) were investigated both experimentally and numerically. Two 100-ton 30Cr2Ni4MoV steel ingots with different insulating hot tops were longitudinally sectioned. The experimental results showed few channel segregates but severe shrinkage porosities appeared in the ingot with poorly insulated hot top, while it was the opposite case after the improved hot topping practice. By employing the finite element numerical simulation, the critical condition for the formation of F-CS in 30Cr2Ni4MoV steel was verified to be R2.1G ≤ 1.0 × 10−5 °C mm1.1 s−2.1. Through coupling with the published C-SP criterion (GR−0.5 ≤ 2.5 °C mm−1.5 s0.5), it was found out that the increase of hot-top height and preheating temperature would aggravate F-CS while alleviate C-SP contrarily. Hence, to comprehensively control those two defects, the optimum hot-top height and preheating temperature for 100-ton ingot were suggested to be 700 mm and 600 °C, respectively. Ultimately, the ratio of the solidification time for the whole ingot to the ingot body (tf/tb) was proposed as a novel criterion for hot top design. A novel hot-work die steel, named 5Cr5Mo2, was designed to obtain superior thermal stability. The proposed alloy is evaluated in terms of its hardness, microstructure, and tempering kinetics. Compared with the commonly used H13 steel, the softening resistance of the designed steel is superior. Based on SEM and transmission electron microscopy (TEM) observations, a higher abundance of fine molybdenum carbides precipitate in 5Cr5Mo2 steel. Strikingly, the coarseness rate of the carbides is also relatively low during the tempering treatment. Moreover, owing to their pinning effect on dislocation slip, the dislocation density of the 5Cr5Mo2 steel decreases more slowly than that of the H13 steel. Furthermore, a mathematical softening model was successfully deduced and verified by analyzing the tempering kinetics. This model can be used to predict the hardness evolution of the die steels during the service period at high temperature.

Different austenitizing temperatures were used to obtain medium-carbon low-alloy (MCLA) martensitic steels with different lath martensite microstructures. The hierarchical microstructures of lath martensite were investigated by optical microscopy (OM), electron backscattering diffraction (EBSD), and transmission electron microscopy (TEM). The results show that with increasing the austenitizing temperature, the prior austenite grain size and block size increased, while the lath width decreased. Further, the yield strength and tensile strength increased due to the enhancement of the grain boundary strengthening. The fitting results reveal that only the relationship between lath width and strength followed the Hall−Petch formula of. Hence, we propose that lath width acts as the effective grain size (EGS) of strength in MCLA steel. In addition, the carbon content had a significant effect on the EGS of martensitic strength. In steels with lower carbon content, block size acted as the EGS, while, in steels with higher carbon content, the EGS changed to lath width. The effect of the Cottrell atmosphere around boundaries may be responsible for this change.

PURPOSE:This study aims to explore gene expression signatures and serum biomarkers to predict intrinsic chemoresistance in epithelial ovarian cancer (EOC). PATIENTS AND METHODS:Gene expression profiling data of 322 high-grade EOC cases between 2009 and 2010 in The Cancer Genome Atlas project (TCGA) were used to develop and validate gene expression signatures that could discriminate different responses to first-line platinum/paclitaxel-based treatments. A gene regulation network was then built to further identify hub genes responsible for differential gene expression between the complete response (CR) group and the progressive disease (PD) group. Further, to find more robust serum biomarkers for clinical application, we integrated our gene signatures and gene signatures reported previously to identify secretory protein-encoding genes by searching the DAVID database. In the end, gene-drug interaction network was constructed by searching Comparative Toxicogenomics Database (CTD) and literature. RESULTS:A 349-gene predictive model and an 18-gene model independent of key clinical features with high accuracy were developed for prediction of chemoresistance in EOC. Among them, ten important hub genes and six critical signaling pathways were identified to have important implications in chemotherapeutic response. Further, ten potential serum biomarkers were identified for predicting chemoresistance in EOC. Finally, we suggested some drugs for individualized treatment. CONCLUSION:We have developed the predictive models and serum biomarkers for platinum/paclitaxel response and established the new approach to discover potential serum biomarkers from gene expression profiles. The potential drugs that target hub genes are also suggested.
