Pregnant women with chronic hepatitis B (CHB) are a priority population for hepatitis B care. Identification of HBV status prior to pregnancy would facilitate timely maternal interventions and perinatal care. In our study, we aimed to study the epidemiology of CHB among women of childbearing age (WoCBA, 18–49 years) in Alberta, Canada. We retrospectively analysed Alberta Analytics databases to study CHB epidemiology, natural history and care linkage among WoCBA in Alberta, between April 2012 and March 2021. A Poisson regression was conducted to estimate incidence of newly identified CHB cases and prevalence trends, whereas predictors of care linkage were determined using logistic regression. Age/sex‐adjusted incidence of newly identified CHB among WoCBA between 2015 and 2020 was 36.2/100,000 person/years, highest among individuals aged 30–39 years. Incidence of newly identified CHB decreased from 52.6 to 18.2/100,000 between 2015 and 2020, but prevalence increased from 131.7 to 248.6/100,000 in the same period. Newly identified CHB incident cases (n = 2124) had lower survival rates than age/sex‐matched Canadians, with a standardized mortality ratio of 5.7 (95% CI 2.6–11.0). Increasing age (years) at diagnosis (HR, 1.2; 95% CI 1.1–1.3) was independently associated with mortality. Comorbid hepatocellular carcinoma, anti‐HBV treatment and year of diagnosis were not significantly associated with mortality. Of the 1927 women with 2436 hepatitis B surface antigen‐positive pregnancies from 2012 to 2020, only 27.6% had recommended HBV assessment during pregnancy. Of those women meeting criteria for antiviral therapy to prevent mother‐to‐child transmission (MTCT), only 66.4% received treatment. Interferons (IFNs) are signalling proteins primarily involved in initiating innate immune responses against pathogens and promoting the maturation of immune cells. Interferon Regulatory Factor 7 (IRF7) plays a pivotal role in the IFNs signalling pathway. The activation process of IRF7 is incited by exogenous or abnormal nucleic acids, which is followed by the identification via pattern recognition receptors (PRRs) and the ensuing signalling cascades. Upon activation, IRF7 modulates the expression of both IFNs and inflammatory gene regulation. As a multifunctional transcription factor, IRF7 is mainly expressed in immune cells, yet its presence is also detected in keratinocytes, fibroblasts, and various dermal cell types. In these cells, IRF7 is critical for skin immunity, inflammation, and fibrosis. IRF7 dysregulation may lead to autoimmune and inflammatory skin conditions, including systemic scleroderma (SSc), systemic lupus erythematosus (SLE), Atopic dermatitis (AD) and Psoriasis. This comprehensive review aims to extensively elucidate the role of IRF7 and its signalling pathways in immune cells and keratinocytes, highlighting its significance in skin‐related and connective tissue diseases. Abstract\nSTATEMENT OF NOVELTYPlants play a crucial role as a removal pathway in constructed wetlands, demonstrating the ability to absorb and tolerate antibiotics from wastewater. However, the specific contribution of plants in this regard has not yet to be sufficiently established. To gain a more comprehensive insight into the associated processes, we selected three common wetland plant species, Canna indica L. (C. indica), Cyperus alternifolius L. (C. alternifolius), and Thalia dealbata Fraser (T. dealbata), to evaluate their capacity for uptake, accumulation, and physiological response in the removal of sulfamethoxazole (SMX) at varying initial concentrations (10, 30, 100, and 300 µg/L) under hydroponic conditions. The results showed that SMX removal was more efficient at lower concentrations (10 and 30 µg/L) than at higher concentrations (100 and 300 µg/L). Moreover, plant systems were found to consistently outperform unplanted systems in SMX removal. Among the assessed species, C. indica was identified as being relatively effective in the removal of SMX, whereas the performance of C. alternifolius was notably less pronounced. A positive correlation was observed between the concentration of SMX in the plant tissues and that in the external aqueous medium. However, plant tissue residues contributed only a minor fraction to the overall removal of SMX. Wetland plants absorb SMX through their roots, and we accordingly detected significantly higher concentrations in submerged plant tissues. Furthermore, we also detected reductions in net photosynthetic rates indicative of potential phytotoxicity, which is associated with the accumulation of antibiotic in the shoot tissues. Accumulation of SMX in the roots and rhizomes was also found to be associated with the development of shorter roots, with this effect becoming more pronounced with an increase in the concentration of exogenous SMX. However, despite these adverse effects, plants can detoxify antibiotics via the glutathione pathway. Of the assessed plant species, C. indica was identified as the most SMX tolerant, as indicated by Km and Vmax values, with C. alternifolius being the least tolerant. Our findings in this study reveal the potential value of wetland plants in the sequestration of antibiotics and provide evidence for the underlying mechanisms of action. These findings could make an important contribution to the implementation of phytoremediation in antibiotic-contaminated water.Three wetland plants with fibrous root systems, namely Canna indica, Cyperus alterniflius, and Thalia dealbata, were selected to investigate the removal efficiencies of sulfamethoxazole in the hydroponic system by different emergent plants, quantify the contribution of uptake and accumulation for sulfamethoxazole in plant tissues, and assess the physiological responses of plants and their effect on the removal of sulfamethoxazole. This article discusses the clinical characteristics of rosacea in perimenopausal women. Rosacea is a chronic inflammatory skin disease that can be classified into four subtypes. Perimenopause is a stage in the aging process characterized by the cessation of ovarian function and a decline in estrogen levels. The incidence of rosacea rises in middle-aged women, especially those aged 40 to 60, and tends to be more severe in patients aged 45 and older. Hormonal imbalance is associated with the occurrence of rosacea, suggesting that perimenopause and hormonal changes may be underlying mechanisms. The study found that perimenopausal women with rosacea had a significantly affected quality of life, with high levels of anxiety and depression. The severity of rosacea symptoms was negatively correlated with estradiol levels, suggesting that estrogen may play a protective role in maintaining optimal skin physiology. [Display omitted] Titanium (Ti) was an excellent medical metal material, but the lack of good antibacterial activity confined its further practical application. To solve this dilemma, a coating containing graphene oxide (GO) and copper (Cu) was prepared on the surface of Ti sheet (Ti/APS/GO/Cu). First, physical sterilization could be carried out through the sharp-edged sheet structure of GO. Second, the oxygen-containing functional group on the surface of GO and the released Cu2+ would generate reactive oxygen species for chemical sterilization. The synergistic effect of GO and Cu substantially enhanced the in vitro and in vivo antibacterial property of Ti sheet, thereby reducing bacterial-related inflammation. Quantitatively, the antibacterial rate of Ti/APS/GO/Cu against E. coli or S. aureus reached over 99%. Besides, Ti/APS/GO/Cu showed excellent biocompatibility and no toxicity to cell. To effectively treat the pollutant in sewage, a new-type three-dimensional aerogel based on graphene oxide and waste paper was fabricated via simple mixing and freeze-drying processes. Therein, waste paper was activated in advance, and polyvinyl alcohol served as physical crosslinking agent. The results showed that the obtained aerogel with porous three-dimensional network structure exhibited good thermal stability. When it acted as adsorbent for treating organic dye (methylene blue) and antibiotic (ciprofloxacin), the adsorption process was explored. In single system, the pseudo-first-order and pseudo-second-order kinetic models were used to discuss the detail about the aerogel adsorbing pollutant. Moreover, the Langmuir and Freundlich adsorption models were applied to describe the equilibrium isotherms and calculate the isotherm constants. Thermodynamic results stated that the adsorption of pollutant onto aerogel was feasible and spontaneous. In binary system, the adsorption capacity of methylene blue by aerogel increased, while the adsorption capacity of ciprofloxacin by aerogel declined. The pseudo-second-order kinetics was consistent with the adsorption of pollutant by aerogel in such binary system. Background: Aging remains a common influencing factor for many diseases. Previous studies have shown that age is significantly associated with rosacea among female cases and that the incidence of rosacea increases with age. However, previous studies did not specifically analyze the clinical characteristics of different age groups. Objective: This study aimed to analyze and compare the clinical characteristics of female patients of rosacea among different age groups. Methods: We conducted a retrospective study of 840 female rosacea subjects and compared cutaneous features, aggravating factors, systemic diseases, and psychological states across age groups. The patients were divided into three groups according to their age at diagnosis: ≤30 years，31–44 years，and ≥45 years. Results: In our study, the mean age of subjects was 35.9 ± 10.23 years. The common symptoms included telangiectasia (82.6%), persistent erythema (82.0%), burning/stinging sensation (89.3%), dry sensation (74.0%), and pruritis (41.9%). Hot temperature (89.9%), emotional changes (67.3%), spicy food (55.6%), and sun exposure (50.7%) were the common aggravating factors. Some patients had comorbidities of systemic disorders (20.4%). Of the patients, 48.8% presented with anxiety and 35.2% with depression. The clinical characteristics were found to be significantly different among the different age groups. Middle‐aged and older patients (≥45 years) were more likely to have more serious persistent erythema and telangiectasia. And these patients were relatively less affected by some of the influencing factors and had more systemic diseases of the digestive system, endocrine metabolic system, and cardiovascular system (p < 0.05). Conclusion: We revealed the impact of age on the characteristics of rosacea, which indicated that the clinical features of rosacea are more complex and more difficult to treat in females over the age of 45. Purpose: Until now, the microbiome of the nasal cavity and its contribution to nasal mucosal disease has remained poorly understood. The advent of cultivation-free molecular methods makes it possible to characterize the total microbiome of the nasal cavity. We sought to assess the microbial diversity and composition of the middle meatus in allergic rhinitis (AR) patients, chronic rhinosinusitis patients without polyps (CRSsNP) and a control population to determine the microbiota associated with the pathogenesis of AR and CRSsNP. Methods: Microbial characterization was determined using 16S rRNA gene sequencing of 122 nasal swabs collected from patients with AR (n = 52) and CRSsNP (n = 37), and from healthy controls (n = 33). Results: There was no difference in nasal microbiome richness and diversity among the three groups, and the dominant phyla were similar among three groups including Firmicutes, Proteobacteria, Actinobacteria and Bacteroidetes. However, Spirochaetae abundance was significantly higher in AR than in the control group after FDR correction (FDR p = 0.021). At the genus level, although there was no statistical significance after FDR correction, there was a trend that Pseudomonas and Peptostreptococcaceae abundance were higher in AR than in controls (p = 0.005, p = 0.005) and CRSsNP (p = 0.023, p = 0.034); Lactobacillus abundance was lower in AR than in controls (p = 0.021); Moraxella abundance was lower in CRSsNP than in controls (p = 0.006); Haemophilus abundance was higher in CRSsNP than in AR (p = 0.003) but lower in AR than in controls (p = 0.018). Background: Little is known regarding the role of the microbiome of the paranasal sinuses and its contribution to sinus mucosal health and disease. Consequently, we examined the microbiome of chronic rhinosinusitis patients with polyps (CRSwNP) and a control population to provide new insights into the microbiota associated with the pathogenesis of CRSwNP. Methods: Fifty‐nine CRSwNP patients and 27 controls were enrolled in the study. The bacterial communities of the middle meatus were detected using 16S ribosomal RNA (rRNA)‒targeted Illumina MiSeq sequencing after microbial DNA was extracted from swabs. Results: Although there was no difference in diversity between the 2 groups, richness was lower in the CRSwNP group than in the control group (p = 0.03). At the phylum level, Firmicutes, Proteobacteria, Actinobacteria, and Bacteroidetes were predominant in both groups; however, the relative abundance was different, with the proportions of Actinobacteria (predominantly Corynebacterium) and Dolosigranulum being significantly higher in the control group than in the CRSwNP group. Conclusion: These results support the theory of microbial dysbiosis as the pathogenesis of CRSwNP. Purpose: Substantial gaps exist between clinical practice and evidence-based cancer care, potentially leading to adverse clinical outcomes and decreased quality of life for cancer patients. This study aimed to evaluate the usefulness of clinical pathways as a tool for improving quality of cancer care, using breast, colon, and rectal cancer pathways as demonstrations. Methods: Newly diagnosed patients with invasive breast, colon, and rectal cancer were enrolled as pre-pathway groups, while patients with the same diagnoses treated according to clinical pathways were recruited for post-pathway groups. Results: Compliance with preoperative core biopsy or fine-needle aspiration, utilization of sentinel lymph node biopsy, and proportion of patients whose tumor hormone receptor status was stated in pathology report were significantly increased after implementation of clinical pathway for breast cancer. For colon cancer, compliance with two care processes was significantly improved: surgical resection with anastomosis and resection of at least 12 lymph nodes. Regarding rectal cancer, there was a significant increase in compliance with preoperative evaluation of depth of tumor invasion, total mesorectal excision treatment of middle- or low-position rectal cancer, and proportion of patients who had undergone rectal cancer surgery whose pathology report included margin status. Moreover, total length of hospital stay was decreased remarkably for all three cancer types, and postoperative complications remained unchanged following implementation of the clinical pathways. Conclusions: Clinical pathways can improve compliance with standard care by implementing evidence-based quality indicators in daily practice, which could serve as a useful tool for narrowing the gap between clinical practice and evidence-based care. An efficient chemoenzymatic process for the synthesis of azoxybenzenes was developed. A peracid was generated in situ by Novozym 435, and then a range of anilines were oxidized by the produced peracid to afford azoxybenzenes in yields ranging from 63.1 to 94.1 %. The MADS-box transcription factors are known to be involved in several aspects of plant growth and development, especially in floral organ specification. However, little is known in eggplant. Here, 120 eggplant MADS-box genes were identified and categorized into type II (MIKCC and MIKC*) and type I (Mα, Mβ, and Mγ) subfamilies based on phylogenetic relationships. The exon number in type II SmMADS-box genes was greater than that in type I SmMADS-box genes, and the K-box domain was unique to type II MADS-box TFs. Gene duplication analysis revealed that segmental duplications were the sole contributor to the expansion of type II genes. Cis-elements of MYB binding sites related to flavonoid biosynthesis were identified in three SmMADS-box promoters. Flower tissue-specific expression profiles showed that 46, 44, 38, and 40 MADS-box genes were expressed in the stamens, stigmas, petals, and pedicels, respectively. In the flowers of SmMYB113-overexpression transgenic plants, the expression levels of 3 SmMADS-box genes were co-regulated in different tissues with the same pattern. Correlation and protein interaction predictive analysis revealed six SmMADS-box genes that might be involved in the SmMYB113-regulated anthocyanin biosynthesis pathway. Introduction: Recent advancements in androgenetic alopecia (AGA) treatment have highlighted the efficacy of botulinum toxin (BoNT). However, inconsistencies in injection sites and depths warrant attention. It remains unclear which injection strategy is most beneficial for patients. Methods: This split-scalp randomized controlled trial divided each enrolled participant's scalp along the midline: one side was randomized to receive intramuscular BoNT injections in the surrounding muscles, while the other side received intradermal BoNT injections directly into the balding areas. This study evaluated the impact of treatment on hair density and diameter through trichoscopic examinations conducted at baseline and 12 weeks post treatment. Additionally, assessments of pain and overall safety were integrated into the study protocol. Results: Twenty-nine patients completed the study, with significant improvements in hair density observed in the intramuscular injection group compared to the intradermal group (p < 0.001). Both groups exhibited increases in hair diameter, but no significant difference was found between the two methods (p = 0.433). Pain evaluation revealed that intradermal injections in balding areas were more painful than intramuscular injections (p = 0.036), with no severe adverse reactions reported except for a single case of alopecia areata following injection. Conclusion: Our research revealed that both BoNT injection strategies enhanced hair diameter 12 weeks post-treatment, with intramuscular injections significantly improving hair density more effectively. Despite the promising outcomes, the variability in patient responses underscores the necessity for personalized approaches and further research to refine injection protocols for optimized efficacy and safety. Purpose: The role of the microbiome in the paranasal sinuses and its contribution to sinus mucosal health and disease remains poorly understood. Consequently, we examined the nasal microbiome of chronic rhinosinusitis patients with polyps (CRSwNP), chronic sinusitis without nasal polyps (CRSsNP) and a control population, associated with IL-5 of nasal polyp tissues and postoperative follow-up of CRSwNP patients, in search of nasal microbial community characteristics related to pathogenesis and prognosis of CRSwNP, providing a new perspective for further understanding of the disease. Methods: The middle meatus secretions of 77 CRSwNP, 36 CRSsNP and 34 non-CRS subjects were collected. The bacterial microbiome composition was detected using high-throughput sequencing technology based on 16S rRNA, and the differences in the nasal microbial diversity among the three groups were compared. At the same time, nasal polyp tissues were collected to detect the expression of IL-5 and analyse its relationship with the structural characteristics of nasal microbial colonies. Postoperative follow-up of patients with CRSwNP was conducted for 1 year to record the recurrence of nasal polyps and analyse the correlation between the recurrence of nasal polyps and IL-5 as well as the characteristics of nasal microbial diversity. Results: The results showed that the average Sobs index (579.31) of the non-CRS group was significantly higher than that of the CRSwNP group (387.31, P = 0.03). PCoA analysis showed that the microbial distribution in the three groups was mostly similar, with only a few unique to each group. At the phylum level, Actinobacteria and Chlamydia in the non-CRS group were significantly higher than those in the CRSwNP and CRSsNP groups. At the genus level, Corynebacterium and Dolosigranulum in the non-CRS group were significantly higher than those in the CRSwNP and CRSsNP groups. Twenty-five CRSwNP patients had nasal polyps that were IL-5 positive, accounting for 32.47%, and the relative abundance of Enterobacter was 6.37% ± 5.92%, which was significantly higher than 0.58% ± 0.11% in the IL-5 negative group. No significant difference was found after correction (p = 0.026, FDR p > 0.05). One year after surgery, 77 patients with CRSwNP who underwent surgery were successfully followed up, and 12 patients with CRSwNP relapsed, with a recurrence rate of 15.6%. Total nasal symptom scores (TNSS) were significantly higher in the recurrent group than in the nonrecurrent group (P = 0.000). No differences in microbial diversity were found between the CRSwNP populations in the recurrent group and the nonrecurrent group at both the phylum and genus levels. For the nonrecurrent CRSwNP group, the relative abundance of Actinobacteria (PDR P = 0.012) and Corynebacterium (PDR P = 0.003) was higher than that before surgery, and the relative abundance of Bacteroidetes (PDR P = 0.040) was lower than that before surgery. However, for the recurrence CRSwNP group, there was no significant difference in the nasal microbiome between postoperation and preoperation. Conclusion: In conclusion, microbial dysbiosis in the nasal cavity is associated with the pathogenesis of CRSwNP. In Southwest China, the inflammatory pattern of nasal polyps is not dominated by eosinophilic infiltration of Th2-type inflammation. The recurrence of nasal polyps after ESS may be potentially related to the decrease in protective bacteria and the increase in pathogenic bacteria, and the improvement of postoperative bacterial disorder is correlated with the nonrecurrence of CRSwNP. Electric fatigue behaviors of the poled lanthanum-doped lead zirconate titanate (PLZT) ceramics were investigated using a home-made electric loading apparatus in conjunction with a conventional X-ray diffractometer. The XRD data were measured under actions of the applied direct current electric fields on the as-received poled PLZT specimens being experienced various cycles (10N) of alternating current (AC) electric fields. Experimental data showed that in addition to apparent degradation in remnant polarization, asymmetries in hysteresis loops and fraction of 90° domain switching curves were observed. It was found that an offset electrical field (ΔEN) was induced in the poled specimen by polarization with its direction against the direction of the polarization. Furthermore, the magnitude of ΔEN and the degree of asymmetry (δN) decreased as the number of AC electric fatigue cycles (10N) varied from 100 to 106. In situ XRD data suggested that the poled specimen exhibited different abilities to reorient a-domains into c-domains when the applied electric fields were opposite. To understand the inflammatory microenvironment and microbiome factors for prognosis of chronic rhinosinusitis with polyps (CRSwNP), we explored the difference in characteristics of the microbiome of the nasal sinuses and inflammatory cytokines between recurrent and non-recurrent groups. We collected nasal secretions and polyp tissue from 77 CRSwNP patients. Then, we extracted microbial DNA from cotton swabs, performed high-throughput sequencing based on 16S rRNA to detect bacterial community composition, and analyzed cytokines such as IL-5, IL-8, IL-17a, IL-17e, IL-18, IL-27 and INF-gamma from polyp tissue using Luminex. The eosinophil and neutrophil cells in the peripheral blood and polyp tissue were counted. Postoperative follow-up of patients with CRSwNP for 1 year was conducted to record the recurrence of nasal polyps and analyze the correlation between the recurrence of nasal polyps and the characteristics of inflammatory cytokines, inflammatory cell count and nasal microbial diversity. After 1 year of follow-up, there were 12 recurrent patients, including 5 males and 7 females. Postoperative recurrence of nasal polyps was not significantly correlated with age, sex, asthma, allergic rhinitis or other allergic diseases in CRSwNP patients. In terms of the total nasal symptom score, the recurrent group was significantly higher than the non-recurrent group. In nasal polyp tissues, eosinophils (40.83/HP) and neutrophils (30.83/HP) in patients with CRSwNP in the recurrent group were significantly higher than those in the non-recurrent group (13.72/HP), and neutrophils (18.5/HP) were also significantly higher in the recurrent group than the non-recurrent group. The expression levels of IFN-, IL-17A, IL-17E and IL-18 were significantly higher in the recurrent group than in the non-recurrent group, and the positive rates were not different. In Southwest China, Enterobacteria and anaerobic bacteria may be correlated with the inflammatory pattern expression of nasal polyps. The neutrophil-mediated inflammatory response plays an important role in patients with CRSwNP in Southwest China and is correlated with nasal polyp recurrence. Recurrence of nasal polyps after endoscopic sinus surgery may be potentially associated with a reduced abundance of protective microorganisms and an increased number of pathogenic microorganisms. The results showed that the layer could set free massive Cu2+ to strive against E. coli and S. aureus for self-defense when exposed to a slightly acidic condition. By contrary, a little Cu2+ was released in the physiological situation, which could avoid damage to the normal cells and showed excellent in vitro pH-dependent antibiosis. Besides, in vivo experiment confirmed that Ti-Cu@SF could work as an antibacterial material to kill S. aureus keenly and display negligible toxicity in vivo. Cold stimulation and exercise are two independent stimulus variables, each yielding unique impacts on human health. However, in many cases, individuals often encounter both factors simultaneously, particularly in colder regions where outdoor physical activity is prevalent. Despite numerous studies exploring the effects of cold exposure and exercise independently, only a limited number of comprehensive reviews have evaluated their combined influence on health. This review aims to bridge this gap by meticulously examining the potential benefits and detriments of both cold stimulation and exercise on human health. Specifically, the review focuses on their impacts on cardiovascular, respiratory, and immune function, as well as the favorable effects of moderate exercise on the human body. While evidence suggests that cold exposure can enhance health outcomes in certain disease contexts, it also poses risks, particularly for vulnerable populations. Furthermore, while moderate exercise is generally beneficial, engaging in physical activity in cold weather conditions can increase the risk of hypothermia, frostbite, and other cold-related injuries. Consequently, there is an urgent need to develop safe and effective guidelines for exercising in cold weather, particularly for individuals living in colder regions. Flower abscission is an important developmental process that can significantly reduce the yield of horticultural plants. We previously reported that SmMYB113 is a key transcription factor promoting anthocyanin biosynthesis and improve fruit quality. However, the overexpression of SmMYB113 in eggplant increased flower drop rate and reduced fruit yield. Here, we elucidate the regulatory mechanisms of SmMYB113 on flower abscission in eggplant. RNA-seq analysis indicated that the regulation of flower abscission by SmMYB113 was associated with altered expression of genes related to ethylene biosynthesis and signal transduction, including ethylene biosynthetic genes SmACS1 , SmACS8 and SmACO4. Then, the ethylene content in flowers and the function of ethephon (ETH, which promotes fruit ripening) and 1-Methylcyclopropene (1-MCP, which acts as an ethylene perception inhibitor) were analyzed, which revealed that SmMYB113 directly regulates ethylene-dependent flower abscission. Yeast one-hybrid and dual-luciferase assays revealed that SmMYB113 could directly bind to the promoters of SmACS1 , SmACS8 , and SmACO4 to activate their expression. Through construction of a yeast two-hybrid (Y2H) screening library, the protein SmERF38 was found to interact with SmMYB113, and verified by Y2H, bimolecular fluorescence complementation (BiFC), and luciferase complementation assay. Furthermore, dual-luciferase assays showed that SmERF38 enhanced the role of SmMYB113 on the promoters of SmACS1. Auxin response factors (ARFs) are important plant transcription factors that are differentially expressed in response to auxin and various abiotic stresses. ARFs play important roles in mediating plant growth and stress responses; however, these factors have not been studied in eggplants. In this study, genome–wide identification and the functional analysis of the ARF gene family in eggplants (Solanum melongena L.) were performed. A total of 20 ARF (SmARF) genes were identified and phylogenetically classified into three groups. Our analysis revealed four functional domains and 10 motifs in these proteins. Subcellular localization showed that the SmARFs localized in the nucleus. To investigate the biological functions of the SmARFs under 2,4–D and salt stress treatments, quantitative real–time RT–PCR (qRT–PCR) was conducted. Most SmARF genes exhibited changes in expression in response to 2,4–D treatments in the flowers, especially SmARF4 and 7B. All SmARF genes quickly responded to salt stress, except SmARF17 and 19 in leaves, SmARF1A and 7B in roots, and SmARF2A, SmARF7B, and SmARF16B in stems. N-carbamylglutamate (NCG) has been shown to enhance performance in neonatal piglets. However, few studies have demonstrated the effect of NCG on the intestinal mucosal barrier. This study was conducted to determine the effects of dietary NCG supplementation on intestinal mucosal immunity in neonatal piglets after an Escherichia coli (E. coli) challenge. New-born piglets (4 d old) were assigned randomly to one of four treatments (n = 7), including (I) sham challenge, (II) sham challenge +50 mg/kg NCG, (III) E. coli challenge, and (IV) E. coli challenge +50 mg/kg NCG. On d 8, pigs in the E. coli challenge groups (III and IV) were orally challenged with 5 mL of E. coli K88 (108 CFU/mL), whereas pigs in the sham challenge groups (I and II) were orally dosed with an equal volume of water. On d 13, all piglets were sacrificed, and samples were collected and examined. The results show that average daily gain in the E. coli challenged piglets (III and IV) was decreased (PE.coli<0.05). However, it tended to be higher in the NCG treated piglets (II and IV). Ileum secretory IgA, as well as IFN-γ, IL-2, IL-4 and IL-10 in ileal homogenates, were increased in E. coli challenged piglets (III and IV). Similarly, ileum SIgA and IL-10 levels, and CD4+ percentage in NCG treated piglets (II and IV) were higher than no-NCG treated piglets (PNCG<0.05). However, the IL-2 level was only decreased in the piglets of E. coli challenge + NCG group (IV) compared with E. coli challenge group (III) (P<0.05). No change in the IL-2 level of the sham challenged piglets (III) was observed. In conclusion, dietary NCG supplementation has some beneficial effects on intestinal mucosal immunity in E. coli challenged piglets, which might be associated with stimulated lymphocyte proliferation and cytokine synthesis. Abstract: Both H2O2 and NO are involved in multiple physiological responses in plants. Metallothionein (MT) can bind heavy metal ions and reduce metal toxicity. Copper toxicity has become a major problem with increasing agricultural and environmental pollution. Here, we investigated the possible roles of ROS, NO and metallothionein in tomato plant responses to copper toxicity. We found that Cu2+ stress caused the rapid release of H2O2 and chlorotic leaves, and it stunted root growth and development. Cu treatment also caused an increase in NOS enzyme activity and NO release in roots and leaves. Application of the NO donor SNP efficiently alleviated the copper toxicity effect, as shown by increases in chlorophyll content and the biomass of fresh/dry leaves. SNP treatment also induced the transcription and increased activities of anti-oxidant enzymes, including catalase, peroxidase, superoxide dismutase and ascorbate peroxidase, and led to reduced H2O2 accumulation in the leaves. Special inhibitors or scavengers of NO synthesis diminished the ameliorating effect of NO on copper toxicity. NO application induced MT transcription and accumulation in leaves. Furthermore, the antisense-MT transgenic tomato was more sensitive to copper stress, and this effect could not be efficiently reversed by NO treatment. From these data, we propose that NO induces tomato tolerance to copper toxicity through antioxidant enzyme activity and metallothionein accumulation, and that metallothionein acts downstream of NO signaling. Background: The red (R) and blue (B) light wavelengths are known to influence many plant physiological processes during growth and development, particularly photosynthesis. To understand how R and B light influences plant photomorphogenesis and photosynthesis, we investigated changes in leaf anatomy, chlorophyll fluorescence and photosynthetic parameters, and ribulose-1, 5-bisphosphate carboxylase/oxygenase (Rubisco) and Calvin cycle-related enzymes expression and their activities in sweet pepper (Capsicum annuum L.) seedlings exposed to four light qualities: monochromatic white (W, control), R, B and mixed R and B (RB) light with the same photosynthetic photon flux density (PPFD) of 300 μmol/m2·s. Results: The results revealed that seedlings grown under R light had lower biomass accumulation, CO2 assimilation and photosystem II (PSII) electron transportation compared to plants grown under other treatments. These changes are probably due to inactivation of the photosystem (PS). Biomass accumulation and CO2 assimilation were significantly enriched in B- and RB-grown plants, especially the latter treatment. Their leaves were also thicker, and photosynthetic electron transport capacity, as well as the photosynthetic rate were enhanced. The up-regulation of the expression and activities of Rubisco, fructose-1, 6-bisphosphatase (FBPase) and glyceraldehyde-phosphate dehydrogenase (GAPDH), which involved in the Calvin cycle and are probably the main enzymatic factors contributing to RuBP (ribulose-1, 5-bisphosphate) synthesis, were also increased. Background: Plant phospholipase D (PLD), which can hydrolyze membrane phospholipids to produce phosphatidic acid (PA), a secondary signaling molecule, has been proposed to function in diverse plant stress responses. Both PLD and PA play key roles in plant growth, development, and cellular processes. PLD was suggested to mediate the regulation of stomatal movements by abscisic acid (ABA) as a response to water deficit. In this research, we characterized the roles of the cucumber phospholipase D alpha gene (CsPLDα, GenBank accession number EF363796) in the growth and tolerance of transgenic tobacco (Nicotiana tabacum) to drought stress. Results: The CsPLDα overexpression in tobacco lines correlated with the ABA synthesis and metabolism, regulated the rapid stomatal closure in drought stress, and reduced the water loss. The NtNCED1 expression levels in the transgenic lines and wild type (WT) were sharply up-regulated after 16 days of drought stress compared with those before treatment, and the expression level in the transgenic lines was significantly higher than that in the WT. The NtAOG expression level evidently improved after 8 and 16 days compared with that at 0 day of treatment and was significantly lower in the transgenic lines than in the WT. The ABA content in the transgenic lines was significantly higher than that in the WT. The CsPLDα overexpression could increase the osmolyte content and reduce the ion leakage. The proline, soluble sugar, and soluble protein contents significantly increased. By contrast, the electrolytic leakage and malondialdehyde accumulation in leaves significantly decreased. The shoot and root fresh and dry weights of the overexpression lines significantly increased. These results indicated that a significant correlation between CsPLDα overexpression and improved resistance to water deficit. Conclusions: The plants with overexpressed CsPLDα exhibited lower water loss, higher leaf relative water content, and heavier fresh and dry matter accumulation than the WT. Light qualities are thought to affect many plant physiological processes during growth and development. To investigate how light qualities influence tomato ( Solanum lycopersicum ) seedlings, the present study evaluated the effects of different light qualities generated by light emitting diodes (LEDs) with the same photosynthetic photon flux density (PPFD), including monochromic light red (657 nm, R), blue (457 nm, B), purple (417 nm, P) or white (W), combination of R and B lights (R:B = 1:1, 1R1B and R:B = 3:1, 3R1B) on the net photosynthetic rate (Pn), growth rate, carbohydrate content and sucrose-metabolizing enzyme activities. The results showed that relative to W, the seedling plant height and stem diameter were significantly promoted by combination of R and B lights and monochromic R light. However, the level of root growth was lower under R and P light, and the seedling growth and Pn were significantly suppressed under the latter. Additionally, R light significantly increased the contents of fructose and glucose, and combination of R and B lights significantly enhanced total carbohydrate, starch and sucrose accumulation, especially for 3R1B treatment. Activity of sucrose synthase (SS) was promoted under the different treatments and reached its highest value under 3R1B, which appeared to be a major contributor to the significantly higher content of starch under this treatment. Furthermore, R, B and P light reduced activity of sucrose phosphate synthase (SPS). Activities of acid invertase (AI) and neutral invertase (NI) were significantly increased by R light, but were markedly reduced under P. The results presented here indicated that monochromic R and combination lights 3R1B, could regulate the plant morphology and photosynthesis by the effects on the metabolism of carbohydrate into fructose, glucose, sucrose and starch, mainly through the enhanced activities of AI and NI under the former treatment, while SS and SPS in the latter treatment, respectively. Background: Talaromyces marneffei is an opportunistic dimorphic fungus prevalent in Southeast Asia. We previously demonstrated that Mp1p is an immunogenic surface and secretory mannoprotein of T. marneffei. Since Mp1p is a surface protein that can generate protective immunity, we hypothesized that Mp1p and/or its homologs are virulence factors. Methodology/Principal Findings: We examined the pathogenic roles of Mp1p and its homologs in a mouse model. All mice died 21 and 30 days after challenge with wild-type T. marneffei PM1 and MP1 complemented mutant respectively. None of the mice died 60 days after challenge with MP1 knockout mutant (P<0.0001). Seventy percent of mice died 60 days after challenge with MP1 knockdown mutant (P<0.0001). All mice died after challenge with MPLP1 to MPLP13 knockdown mutants, suggesting that only Mp1p plays a significant role in virulence. The mean fungal loads of PM1 and MP1 complemented mutant in the liver, lung, kidney and spleen were significantly higher than those of the MP1 knockout mutant. Similarly, the mean load of PM1 in the liver, lung and spleen were significantly higher than that of the MP1 knockdown mutant. Histopathological studies showed an abundance of yeast in the kidney, spleen, liver and lung with more marked hepatic and splenic necrosis in mice challenged with PM1 compared to MP1 knockout and MP1 knockdown mutants. Likewise, a higher abundance of yeast was observed in the liver and spleen of mice challenged with MP1 complemented mutant compared to MP1 knockout mutant. PM1 and MP1 complemented mutant survived significantly better than MP1 knockout mutant in macrophages at 48 hours (P<0.01) post-infection. The mean fungal counts of Pichia pastoris GS115-MP1 in the liver (P<0.001) and spleen (P<0.05) of mice were significantly higher than those of GS115 at 24 hours post-challenge. • The quality of tomatoes was predicted by hyperspectral imaging and chemometrics. • 12 quality indexes of three varieties of tomato were measured in the study. • A new comprehensive quality index (CQI) was proposed through factor analysis. • The MLR model was established to predict the comprehensive quality of tomatoes. The quality of tomatoes is usually predicted by measuring a single index, rather than a comprehensive index. To find a comprehensive index, visible and near infrared (Vis-NIR) hyperspectral imaging was used for capturing the images of three varieties of tomatoes, and twelve quality indexes were measured as the reference standards. The changing trends and correlations of different indexes were analyzed, and comprehensive quality index (CQI) was proposed through factor analysis. The characteristic wavelengths were selected by successive projection algorithm (SPA) based on the hyperspectral data, which was used to establish three regression models for CQI prediction. The result indicated that MLR achieved good performance with R V 2 = 0.87, RMSEV = 1.33 and RPD = 2.58. After that, spatial distribution map was generated to visualize the CQI in tomato fruit. This study indicated that the comprehensive quality of tomatoes can be predicted non-destructively based on hyperspectral imaging and chemometrics, determining the optimal harvesting period. Plant phospholipase D (PLD), which can hydrolyze membrane phospholipids to produce phosphatidic acid (PA), a secondary signaling molecule, has been proposed to function in diverse plant stress responses. In this research, a qRT-PCR analysis indicated that the expression of a cucumber phospholipase D alpha gene ( CsPLDα ) was induced by salt and drought stresses in the roots and leaves. To further study the roles of CsPLDα in regulating plant tolerance to salt, polyethylene glycol (PEG) and abscisic acid (ABA) stresses, transgenic tobacco plants constitutively overexpressing CsPLDα were produced. A qRT-PCR analysis showed that the CsPLDα transcript levels were high in transgenic tobacco lines, whereas no expression was found in wild type (WT) tobacco, indicating that CsPLDα was successfully transferred into the tobacco genome and overexpressed. Under normal conditions for 30 d, seeds of transgenic lines germinated neatly, and the seedlings were robust and bigger than WT plants. When treated with different concentrations of NaCl, PEG and ABA, germination rates and seedling sizes of the transgenic lines were significantly greater than WT. In addition, the germination times for transgenic lines were also remarkably shorter. Further studies indicated that transgenic lines had longer primary roots and more biomass accumulation than WT plants. The water loss in transgenic lines was also much lower than in WT. These findings suggest that the CsPLDα overexpression positively regulates plant tolerance to hyperosmotic stresses, and that CsPLDα is involved in the ABA regulation of stomatal closure and the alleviation of ABA inhibition on seed germination and seedling growth. The present study was designed to determine the interactive effect of exogenous melatonin and nitric oxide (NO) on sodic alkaline stress mitigation in tomato seedlings. It was observed that exogenous melatonin treatment elevated NO levels in alkaline–stressed tomato roots. However, exogenous NO had little effects on melatonin levels. Importantly, melatonin–induced NO generation was accompanied by increased tolerance to alkaline stress. Chemical scavenging of NO reduced melatonin–induced alkaline stress tolerance and defense genes’ expression. However, inhibition of melatonin biosynthesis had a little effect on NO-induced alkaline stress tolerance. These results strongly suggest that NO, acting as a downstream signal, is involved in the melatonin–induced tomato tolerance to alkaline stress. Arbutin, as a simple polyphenol, can be extracted from almost all the natural plants and has various important bioactivities. Regioselective acylation of arbutin can improve its cell membrane penetration and enhance its biological activity. The regioselective acylation of arbutin catalyzed by lipase from Candida sp. (CSL, expressed from Canadia sp. 99–125) has been successfully conducted in non-aqueous media. Under the optimum conditions (arbutin, 0.73 mmol; vinyl acetate, 29.71 mmol; organic solvent, 20 ml tetrahydrofuran; initial water activity, 0.63; temperature, 40 °C; enzyme dosage, 100 mg enzyme powder in this reaction system), the highest enzyme activity (3.71 ± 0.13 μmol/h/mg) could be obtained. The conversion could reach 91.42 ± 2.43% after 24 h and the regioselectivity of the CSL-catalyzed acylation was highly specific at the C-6′ position in the glucose moiety of arbutin (>99%). Seventy-two, suckling piglets, obtained from 9 litters standardized to 8 piglets, were assigned to 1 of 3 treatments (n = 24) to compare short-term, early administration with intermittent, longer-term administration of Lactobacillus reuteri I5007. The treatments were a control (given a placebo of 0.1% peptone water from day 1 to 5) or treatments in which 1.7 × 1010 CFU L. reuteri was administrated either daily for 4 days starting on day 1 or every 4th day from day 1 to 17. Five piglets per treatment were killed at 3 time points (day 7, 14 and 21). Denaturing Gradient Electrophoresis of ileal digesta revealed an increase in the presence of L. reuteri I5007 and Clostridium lentocellum (on day 14 and 21) in the every 4th-day treatment and Actinobacillus porcinus (on day 7 and 14) in both L. reuteri treatments, while reducing the abundance of E. coli on day 21 in the every 4th-day treatment. Real-time qPCR of ileal digesta showed an increase in Bifidobacterium spp. on day 14 for both L. reuteri I5007 treatments. An increase in the concentration of lactic acid and a lower pH was observed in the first 4-day treatment on day 7 and the every 4th day treatment on day 14. The relative abundance of mRNA for TGF-β was increased while that for IFN-γ was decreased in the mesenteric lymph nodes of piglets treated with L. reuteri every 4th day. In conclusion, early intervention with L. reuteri increases the presence of beneficial bacteria and decreases the presence of undesirable microbes in the lower gastrointestinal tract. The changes appear to be mediated by altering the intestinal pH through lactic acid production resulting in favorable bacterial species colonization. S-adenosyl- l-methionine ( SAM) synthetase is the key enzyme involved in the biosynthesis of SAM, which serves as a common precursor for polyamines ( PAs) and ethylene. A SAM synthetase cDNA ( SlSAMS1) was introduced into the tomato genome using the Agrobacterium tumefaciens transformation method. Transgenic plants overexpressing SlSAMS1 exhibited a significant increase in tolerance to alkali stress and maintained nutrient balance, higher photosynthetic capacity and lower oxidative stress compared with WT lines. Both in vivo and in vitro experiments indicated that the function of SlSAMS1 mainly depended on the accumulation of Spd and Spm in the transgenic lines. A grafting experiment showed that rootstocks from Sl SAMS1-overexpressing plants provided a stronger root system, increased PAs accumulation, essential elements absorption, and decreased Na+ absorption in the scions under alkali stress. As a result, fruit set and yield were significantly enhanced. To our knowledge, this is the first report to provide evidence that Sl SAMS1 positively regulates tomato tolerance to alkali stress and plays a major role in modulating polyamine metabolism, resulting in maintainability of nutrient and ROS balance. Abstract: Nitric oxide (NO) and polyamines (PAs) are two kinds of important signal in mediating plant tolerance to abiotic stress. In this study, we observed that both NO and PAs decreased alkaline stress in tomato plants, which may be a result of their role in regulating nutrient balance and reactive oxygen species (ROS), thereby protecting the photosynthetic system from damage. Further investigation indicated that NO and PAs induced accumulation of each other. Furthermore, the function of PAs could be removed by a NO scavenger, cPTIO. On the other hand, application of MGBG, a PA synthesis inhibitor, did little to abolish the function of NO. To further elucidate the mechanism by which NO and PAs alleviate alkaline stress, the expression of several genes associated with abiotic stress was analyzed by qRT-PCR. NO and PAs significantly upregulated ion transporters such as the plasma membrane Na+/H+ antiporter (SlSOS1), vacuolar Na+/H+ exchanger (SlNHX1 and SlNHX2), and Na+ transporter and signal components including ROS, MAPK, and Ca2+ signal pathways, as well as several transcription factors. The light frequency of red (R) and blue (B) plays a crucial role within plant growth cycle. Whereas, the relationship between the metabolic pathways involved in photosynthesis and carbohydrate accumulation in tomato seedling and hormone regulation still needs further investigation. This research comprehensively analyzed changes in leaf anatomy, stomatal traits, pigment content, gas exchange parameters, chlorophyll fluorescence, Rubisco and Calvin cycle-related enzyme expression and activities, as well as endogenous hormones biosynthesis in tomato (Solanum lycopersicum L.) seedlings, which radiated by four light qualities, including monochromatic white light (W), B, R and mixed light of R and B (RB). The findings showed the biomass accumulation as well as CO 2 assimilation were greatly increased in seedlings under B and especially under RB light. Their leaves were also thicker, and stomatal conductance, levels of pigment, photosynthetic electron transport capacity, and the photosynthetic rate were enhanced. In addition, enzyme expression level and activity related to the Calvin cycle were up-regulated. Moreover, RB-induced growth was accompanied by a significant inhibition in gibberellin (GA) concentration as well as an increment in auxin (indole-3-acetic acid (IAA)) in stem and root. Overall, photosynthesis of RB-treated seedlings was positively regulated by leaf anatomy and photosynthetic electron transportation as well as maintaining the activation states of the Calvin cycle, and then inducing changes in carbohydrate accumulation. Abstract: Alkali stress is a major factor that limits crop yield, and nitric oxide (NO) is involved in the regulation of plants tolerant to abiotic stress. In the present study, sodium nitroprusside (SNP), a NO donor, reversed the chlorosis of cucumber leaves caused by alkali stress. Physiological analysis indicated that application of SNP protected mesophyll cell ultrastructure from damage by alkali stress. SNP increased nutrient element utilization, pigment content, photosynthetic capacity and accumulation of organic acids. In this study, Solexa sequencing was used to investigate the effect of SNP on expression of genes involved in cucumber response to alkali stress. About 5.9 million (M) and 5.8M 21-nt cDNA tags were sequenced from the cDNA library of the alkali treatment and SNP treatment, respectively. When annotated, a total of 10,271 genes for the alkali stress treatment from the Solexa sequencing tags and 10,288 genes for SNP treatment were identified. We detected 901 differentially expressed genes in two samples, of which 437 and 464 of them were up- or down-regulated by SNP under alkali stress, respectively. The expression levels of 11 differentially expressed genes were confirmed by real-time RT-PCR. The trends observed agreed well with the Solexa expression profiles, although the degree of change was diverse in amplitude. Gene ontology analysis revealed that differentially expressed genes were mainly involved in response to abiotic stress, cellular metabolic process, photosynthesis, transmembrane transportation and organelle development, which were in accordance with physiological results. Effect of nitrate (NO3 −) on lipid peroxidation, activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and ascorbate peroxidase (APX), soluble sugar, and proline accumulation were investigated in cucumber [Cucumis sativus L. cv. ‘Xintaimici’ (salt-tolerant) and ‘Shennongchunwu’ (salt-sensitive)] seedlings treated with 14 (control) and 140 mM NO3 − for seven days. The results showed that excess NO3 − increased malondialdehyde (MDA) content and electrolyte leakage in cucumber roots of both cultivars, and the degree of increase was higher in ‘Shennongchunwu’. The activities of SOD, CAT, POD, and APX increased in the roots of ‘Xintaimici’, but decreased in roots of ‘Shennongchunwu’, with respect to the control plants. Compared to the control, the soluble sugar and proline content increased in the cucumber roots of both cultivars, especially in ‘Xintaimici’. These results suggested that ‘Xintaimici’ tolerance to NO3 − stress may partly be due to higher activities of antioxidant enzymes and accumulation of osmotic regulation substances. Dysregulation of Axl receptor tyrosine kinases is implicated in the pathogenesis of several sensitive and refractory cancers. In this study we showed that Axl expression was upregulated in drug-resistant cancer cells, as compared to those in parental cells. Downregulation of Axl expression by RNAi led to a decrease of K562/ADR and MCF-7/ADR cells invasion, proliferation in soft agar, and increased cells chemosensitivity in vitro. In nude mice bearing xenografts, downregulation of Axl in MDR cells enhanced the anticancer activity of intraperitoneally applied chemotherapeutic drugs. Abstract: Phospholipase D (PLD; EC3.1.4.4) is thought to have a crucial role in plant signal transduction. Here, a full-length cDNA clone encoding PLD was isolated from cucumber (Cucumis sativus L.) by RT-PCR and RACE. The 2756-bp full-length cDNA sequence of cucumber PLD (CsPLD) contains an open reading frame of 2427bp encoding 808 amino acids. The deduced amino acid sequence showed high sequence similarities with PLDs from other plant species. According to the phylogenetic analysis, CsPLD belongs to PLDα group and contains two characteristic HKD motifs, one IYIENQFF motif and one C2 domain. The tertiary structure of the CsPLD protein was also predicted and found to be composed mainly of random coils. In both the salt-tolerant ‘Xintaimici’ and salt-sensitive ‘Shennongchunwu’ cultivars, CsPLD expression differed among roots, stems, leaves, flowers and mature fruit; expression was highest in leaves and lowest in fruit. The transcript level of CsPLD in salt-tolerant cucumber was higher than that in the salt-sensitive cucumber plants after 182mmolL−1 nitrate treatment. CsPLD expression in both Xintaimici and Shennongchunwu increased with increasing nitrate concentration, although CsPLD mRNA expression in roots of Shennongchunwu peaked at 140mmolL−1 nitrate and then decreased. In addition, CsPLD expression was induced in leaves of both cultivars by various treatments, including abscisic acid, salicylic acid, NaCl, drought and low temperature; however, there was no significant induction in its expression following treatment for 30min with either H2O2 or high temperature. In roots, there was a significant increase in CsPLD expression following all treatments in both cultivars, except in the roots of Xintaimici in response to H2O2. These results suggest that, in cucumber, PLD functions under conditions of excess nitrate and other abiotic stresses. [Copyright &y& Elsevier]
