4. Platycodon grandiflorum saponin D enhances the sensitivity of human rectal cancer cell strain SW1463/Oxa with drug resistance to oxaliplatin

Abstract

Objective To investigate the enhancing effect of platycodon grandiflorum saponin D on the sensitivity of human rectal cancer drug-resistant cell line SW1463 to oxaliplatin (Oxa). Methods Human rectal cancer cell line SW1463 was collected to establish a stable oxaliplatin-resistant cell strain SW1463/Oxa. The constructed cell strain SW1463/Oxa was randomly divided into platycodon grandiflorum saponin D group, Oxa group, platycodon saponin D+Oxa group and control group. The cell proliferation curve was detected by MTT. Deoxyribonucleic acid methyltransferase 3a (DNMT3a), phosphorylated histone γ-H2AX, DNA double strand repair protein RAD51 and signal transducer and activator of transcription 3(STAT3) messenger RNA (mRNA) expression were detected by RT-qPCR. DNMT3a, γ-H2AX, RAD51, STAT3 protein expression and phosphorylated STAT3(p-STAT3) level were detected by Western blot. Results Human rectal cancer drug resistant cell strain SW1463/Oxa was successfully constructed. Compared with the control group and Oxa group, the cell proliferation, DNMT3a, RAD51, STAT3 mRNA and protein expression and p-STAT3 level of platycodon saponin D group and platycodon saponin D+Oxa group decreased(P＜0.05), and the expression of γ-H2AX mRNA and protein increased(P＜0.05). Conclusions Platycodon grandiflorum saponin D may enhance the chemo-sensitivity of human rectal cancer drug-resistant cell strain SW1463/Oxa to Oxa, and the potential mechanism is inhibition of expression of DNMT3a, RAD51, STAT3 and promotion of expression of γ-H2AX.

7. Timed and choreographical multiparty session types theory

Abstract

Multiparty session types (MPSTs) are the formalism for describing protocols among multiple participants from a global point of view. They are used to check type-safety and deadlock freedom for programs. In real-time distributed systems, however, protocol descriptions often include: time-related specifications, such as deadline and timeout, by imposing time restrictions on interactions to guarantee the quality of service; and parallel computation for improving efficiency. The original theory of multiparty session types lacks the ability for expressing time and parallel actions in the global description. Therefore, we propose the theory of timed choreographical session types to solve the problem. To express time-related specifications, we associate time constraints and clock resets to asynchronous message passing in global types and local types. We define the labelled transition systems of timed global and local types, and prove that soundness and completeness are preserved through projection. We add a delay primitive to session calculus to achieve a minimum extension of asynchronous π- calculus and prove theorems of type preservation and subject reduction. We also formalise the theorem of time error-freedom and properties of feasibility and wait- freedom to guarantee that no deadlock is caused by time error. Additionally, we show that timed global session types enjoy progress property if the global types without time constraints and resets are verified to progress. In the theory of choreographical session types, for the purpose of precise thread management, explicit fork/merge operations for parallel composition and choice/join for branching are defined in both global and local session types and session calculus. To ensure well-behaved communications, we formalise the definition of well-formedness for global types and present sufficient conditions for verifying well-formedness. Based on choreographical session calculus, we define the operational semantics and typing system for processes and prove the theorems of subject reduction and type safety. When we model scenarios of concurrent real-time systems, it is natural to combine the formalisms of timed session types and choreographical session types. Then we add time assertions to labelled message passing operations in choreographical global and local types, and delay primitive in session calculus. Due to the orthogonality of time and parallel composition, we easily prove the theorems of subject reduction, and time error-freedom within the timed choreographical context. Due to introduced complexity in global types, we present the progress enablement and time progress property to guarantee that processes will progress.

31. Effects of soil drought and nitrogen deposition on BVOC emissions and their O 3 and SOA formation for Pinus thunbergii.

Abstract

Soil drought and nitrogen (N) deposition can influence the biogenic volatile organic compound (BVOC) emissions and thereby their ozone (O 3 ) and secondary organic aerosol (SOA) formation. This study addressed their single and combined effects on BVOC emissions of Pinus thunbergii by laboratory simulation experiments. The results showed that light drought (LD, 50% soil volumetric water content (VWC)) stimulated isoprene, monoterpene, sesquiterpene, and total BVOC emissions, while moderate drought (MD, 30% and 40% VWC) and severe drought (SD, 10% and 20% VWC) inhibited their emissions (except for sesquiterpene in 20% VWC). N deposition decreased other VOC emissions and increased isoprene and sesquiterpene emissions. Total BVOCs and monoterpene were stimulated in low N deposition (LN, 2 g N/(m 2 ·yr)) and inhibited in moderate (MN, 5 g N/(m 2 ·yr)) and high N deposition (HN, 10 g N/(m 2 ·yr)). Under combined treatment of soil drought and N deposition, total BVOC, monoterpene, and other VOC emissions were inhibited, sesquiterpene had no significant change, and isoprene emission was inhibited in MD combined treatment but promoted in SD. The O 3 formation potential (OFP) and SOA formation potential (SOAP) from the changed BVOC emissions were calculated, OFP and SOAP of BVOC emissions and their compositions varied significantly among the treatments.
