In this paper, natural language processing technology is used to establish coupled Hidden Markov Models for speech formalization and features of the two channels. The adaptive weight selection method is used to recognize speech under different signal-to-noise ratios in the recognition stage. The audio-visual recognition system is applied to actual graffiti art education research, and the feasibility and development direction of audio-visual graffiti in the classroom are studied through questionnaires. It can be found that most teachers and students believe that the concept of audiovisual graffiti painting is helpful or more helpful in the cultivation and development of students’ subconscious mind. Most teachers and students believed audiovisual doodling was beneficial or advantageous for developing students’ subconscious. In the acceptance test and learning effect analysis of the audiovisual graffiti course, the mean values of the four dimensions of perceived usefulness (41.87), perceived ease of use (18.11), willingness to use (31.45), and perceived experience of the process (28.31) are basically around the maximum value, which indicates that the audiovisual technology is well accepted and that the mean value of the experimental group’s performance score (83.68) is significantly higher than that of the control group. Control group. The audiovisual course has been found to enable learners to concentrate on the learning situation faster and master the teaching essential contents more thoroughly.

Based on the current situation of student cultivation mode and student management in colleges and universities, this paper puts forward six basic assumptions on student cultivation and management, and combines the assumptions to obtain the game payment matrix of student cultivation mode and student management. The dynamic equations of the cultivation and management process are reproduced by using mathematical calculus to derive the game payment matrix. Under the constraints, the game evolution model equilibrium point is tested for stability, the game evolution state is derived, and the game evolution model is completed to optimize the innovation of college student training mode and student management mechanism. Numerical simulation analysis is performed on the evolutionary game of college student cultivation and management based on the parameter settings of the evolutionary game model. The results show that the evolutionary game model will converge to a stable strategy point when the ratio of managers (teachers) and students’ initial participation in cultivation and management activities is set to (0.05, 0.95), (0.05, 0.05), (0.95, 0.05) or (0.95, 0.95). This study has a beneficial impact on cultivating exceptional talents, and it also provides a solid resource for society to deliver top-notch talent.

Objective To design and synthesize 4 phenoxyacetic acid compounds, characterize their inhibitory potencies on human glutathione-S-transferase Mu (glutathione-S-transferase Mu, GSTM) by different methods, and investigate the structure-activity relationship between phenoxyacetic acid compounds and GSTM. Methods Four phenoxyacetic acid compounds were designed and synthesized, and they are ethyl 2-(4-acryloylphenoxy)acetate (Ⅰa) and N, N'-(butane-1, 4-diyl)-bis-(4-acryloyl phenoxyacetic amide) (Ⅱa) containing α, β-unsaturated ketone structure, and ethyl 2-(4-propionylphenoxy)acetate (Ⅰb), N, N'-(butane-1, 4-diyl)-bis-(4-propionyl phenoxyacetic amide) (Ⅱb) not containing α, β-unsaturated ketone structure. They were characterized by the half-maximal inhibitory concentration (IC50) and apparent inhibition constant (Ki) using initial velocity method, and the dissociation constants (Kd) for GSTM utilizing fluorescence analysis. Results Ⅰa and Ⅱa contained α, β-unsaturated ketone structure that undergoes electrophilic addition reaction with GSH. After incubation with enzyme and GSH, Ⅰa and Ⅱa generated products in situ, with a IC50 value of 67.00±6.00 and 0.10±0.02 μmol/L for GSTM respectively (n=2). Ⅰb and Ⅱb without α, β -unsaturated ketone structures had no inhibitory potencies for GSTM. The Ki of Ⅰa against GSH was 19.9±1.6 μmol/L, and the Ki against CDNB was 9.1±0.7 μmol/L (n=3). The Ki of Ⅱa against GSH was 0.063±0.005 μmol/L, and against CDNB was 0.079±0.006 μmol/L (n=3). The Ki of Ⅱa was lower than that ofⅠa (P < 0.05). The Kd of Ⅰa, Ⅰb, Ⅱa, and Ⅱb measured by fluorescence analysis were 41.5±1.8, 38.9±1.7, 18.0±0.9, and 19.5±1.0 μmol/L, respectively (n=2). The Kd between Ⅰa and Ⅰb, and between Ⅱa and Ⅱb had no significant differences(P>0.05). The Kd of Ⅱa was lower than that of Ⅰa, and the Kd of Ⅱb was lower than that of Ⅰb (P < 0.05). Conclusion α, β-unsaturated ketone structure is the essential functional group of phenoxyacetic acid compounds for inhibition on GSTM. Comparing with monovalent Ⅰa and Ⅰb, divalent Ⅱa and Ⅱb have higher affinity for GSTM. The product of Ⅱa has inhibitory potency significantly higher than that of Ⅰa.

Objective To compare the immobilization effect of 2 kinds of hydrophilic magnetic beads with Ni2+-nitrilotriacetic acid (Ni2+-NTA) group or carboxyl group and surface modified by ampholytic ions on Mycobacterium tuberculosis peptide deformylase (MtPDF), and to determine the target enzyme immobilization scheme appliable to screening MtPDF ligand mixture library using magnetic separation combined with high performance liquid chromatography-mass spectrometry (HPLC-MS). Methods The plasmids pET-28a(+): def with N-terminal 6×His tagged MtPDF was successfully constructed and transfected into E.coli BL21(DE3) for MtPDF recombinant expression. The products were collected and purified using nickel-ion chelating affinity chromatography column, and then identified with the enzymatic properties. Then the obtained MtPDF was coupled to magnetic beads with Ni2+-NTA (MtPDF-Ni2+-NTA) or carboxyl group (MtPDF-carboxy), respectively. The immobilization capacity, enzyme activity, enzyme stability before and after immobilization, and the response to the inhibitor actinonin were compared between the 2 schemes. Results The immobilization capacity of MtPDF-Ni2+-NTA was 99.7±4.5 mg/g (n=3). When the ratio of amount of enzyme to magnetic beads was 1 ∶15, the retention activity of the immobilized MtPDF was (107.2±13.9)% (n=3) of the free enzyme, and decreased to 43% after 3 h under shaking at 0 ℃ and pH value of 7.5. By contrast, the immobilization capacity of MtPDF-carboxy was 36.9±2.6 mg/g (n=3), and the retention activity of the immobilized MtPDF reached to (102.6±15.4)% (n=3) when the ratio of amount of enzyme to magnetic beads was 1 ∶60, consistent with that on the Ni2+-NTA magnetic beads, with negligible changes within 8 h under shaking at 0 ℃ and pH 7.5. The response of MtPDF-carboxy to the inhibition of actinonin had no significant difference from that of free MtPDF (P>0.05). Conclusion MtPDF-carboxy magnetic beads present stable retention activity, and has similar inhibitory response as that of free MtPDF, and thus is more suitable for screening of mixture library to discover high affinity ligands for MtPDF.
