Deciphering the physical origin of transient crustal deformation is a difficult challenge in the field of continuous borehole observations of crustal movement because the relevant influencing factors of the phenomenon are extremely multifarious. In the Shanxi Rift, squall lines are a frequently occurring mesoscale convective system. Consequently, strain transients are always observed by the borehole dilatometer network within the Shanxi Rift; however, a fundamental understanding of the transient phenomena induced by squall lines remains elusive. To address this challenge, we adopted a second-order Butterworth band-pass filters (0.5–2 h) to retrieve the borehole volumetric deformations excited by a "dry" squall line from September 21, 2017, that occurred in the Shanxi Rift. The elastic loading of Ricker-wavelet-shaped atmospheric waves induced by squall line, which can cause the main signature of transient deformation to adopt a highly similar shape; this pressure-induced deformation process can last approximately 148–164 min and has a maximum magnitude of approximately 19 nstrain. This work of signal processing clearly identifies the crustal transient induced by a squall line and provides new insights into the origin of some short-lasting crustal deformations in the Shanxi Rift and elsewhere. IL-6 plays an important role in oogenesis in humans. However, at the preimplantation stage, IL-6 production and the role in embryo development remain unclear. In this study, IL-6 concentrations in single-embryo media were analyzed. In addition, the association between IL-6 production and blastocyst formation was investigated. Single-embryo culture media from 194 embryos were collected on day 6 after fertilization and divided into four groups according to the developmental stage of the corresponding embryo, as follows: cleavage stage group, morula-early blastocyst group, unavailable full blastocyst group, and available full blastocyst group. IL-6 concentrations were significantly lower in the cleavage stage group than in the morula-early blastocyst group (p = 0.009), in the unavailable full blastocyst group (p = 0.003), and in the available full blastocyst group (p < 0.001). Logistic regression analysis showed that IL-6 concentration in single-embryo medium was significantly associated with blastocyst formation (odds ratios β1 = 1.876, 95% CI 1.433 to 2.644, p < 0.0001). A novel point-of-care testing (POCT) method for the determination of proteases was developed for the first time using a designed disposable capillary-fill device based on the cleavage of electrogenerated chemiluminescence (ECL)-label-tagged peptide probes and enabling elimination of the light-shielding from the magnetic beads (MBs). As a proof-of-principle, prostate-specific antigen (PSA) was taken as a model analyte, and streptavidin-coated magnetic beads bound with ruthenium-complex-tagged specific peptide (biotin-HSSKLQK) were utilized as MB ECL probes. The capillary-fill device was designed to be divided into a reaction zone and detection zone. In the reaction zone, the bio-cleavage reaction between the PSA analyte with the peptide on the surface of the MB ECL probes occurred, while in the detection zone, ECL emission was produced by a screen-printed carbon electrode, Ag/AgCl reference electrode and carbon counter electrode. When the analyte PSA was introduced into the suspension of MB ECL probes in the reaction zone of the device, biocleavage of the peptide occurred, and the cleaved Ru1 part was released from the surface of the MB ECL probes. The capillary-filled device was tilted 90°, and with the aid of gravity, the solution containing the released Ru1 part flowed to the surface of the working electrode in the detection region of the device, while the MB ECL probes were fixed in the reaction zone by an external magnet. PSA can be determined by the ECL emission from the released Ru1 part in the presence of the co-reactant tri-n-propylamine at the detection zone. Under the optimal conditions, the developed ECL method showed a low detection limit of 0.12 ng mL−1 for PSA. This work demonstrates that the developed ECL biosensing approach can eliminate the MB light-shielding effect and quantify proteases with high sensitivity and selectivity, which could be easily extended to POCT-based ECL biosensing for other proteases. The existing building stock faces the challenge of low energy efficiency and requires renovation and upgrading to meet society′s goals of carbon reduction and sustainable development. This study presents an optimization framework utilizing genetic algorithms to develop robust retrofit plans that balance the need for improved energy efficiency, cost-effectiveness considerations for householders, and uncertainties regarding climate conditions. A case study of an aged residential building in a hot and humid region of China is used to demonstrate the proposed method. The optimization results show a potential energy demand reduction of 81.5%. However, due to the relatively long time required to realize economic benefits from high investments, short-term optimization tends to favor solutions with high energy demand and low primary costs. To effectively reduce carbon emissions, it is recommended to consider the long-term economic benefits of retrofits and prioritize solutions with high energy efficiency. However, it is important to acknowledge that the expensive nature of retrofit investments may pose barriers to residents. Introduction: This study identified the trajectories of organized youth sports over 9 years in youths aged 9-18 years and examined whether the trajectories predicted physical activity, sedentary behavior, and obesity in midlife.Methods: Self-reported organized youth sports trajectories were identified for participants between 1980 and 1989 (N=3,474). Accelerometer-derived physical activity was quantified for participants (n=1,349) in 2018-2020. Sociodemographic, physical activity, and TV viewing data were collected through questionnaires either at baselines or follow-up. Adult BMI was calculated to clarify obesity. Associations of organized youth sports trajectories with adult physical activity, sedentary behavior, and obesity were evaluated using mixture models, which were stratified by sex and conducted in 2022.Results: Three organized youth sports trajectories were identified for boys and girls (sustained high-sports participation, 12.0%/7.5%; sustained moderate-sports participation, 14.0%/13.3%; and low-sports/nonparticipation, 74.0%/79.2%). Boys sustaining both moderate- and high-sports participation had higher levels of adult self-reported physical activity (β=0.59, p=0.007; β=0.69, p<0.001) than low-sports/nonparticipating boys. Girls sustaining both moderate- and high-sports participation accumulated more total physical activity (β=113.4, p=0.009; β=144.3, p=0.002), moderate-to-vigorous physical activity (β=7.86, p=0.016; β=14.01, p<0.001), step counts (β=1,020, p=0.003; β=1,045, p=0.005), and self-reported physical activity (β=0.79, p<0001; β=0.63, p=0.003) in midlife than their low-sports/nonparticipating counterparts. Girls sustaining moderate-sports participation accumulated more light-intensity physical activity (β=19.79, p=0.012) and less sedentary time (β= -27.65, p=0.002), and those sustaining high-sports participation had lower obesity prevalence (OR=0.41, p=0.009) 40 years later than low-sports/nonparticipating girls.Conclusions: Sustained participation in organized youth sports is independently predictive of physical activity patterns, sedentary time, and obesity in midlife, especially in girls, thus contributing to the development of a healthy and active lifestyle across the life course. Aberrant activation of eIF4E contributes to gastric cancer growth and resistance. MAPK‐interacting kinases (MNKs) regulate eIF4E phosphorylation and activity in tumor but not normal cells and are potentially safe targets for the treatment of various cancers. Our work reveals that tomivosertib, a potent and highly selective dual MNK1/2 inhibitor, preferentially sensitizes gastric cancer to chemotherapy via suppressing MNK‐eIF4E‐β‐catenin. We firstly demonstrate that tomivosertib displays higher efficacy than other MNK inhibitors in inhibiting gastric cancer cells. In addition, tomivosertib significantly augments the inhibitory effects of 5‐FU and paclitaxel but not everolimus, suggesting that tomivosertib preferentially sensitizes gastric cancer to chemotherapy. We next show that eIF4E overexpression and phosphorylation coordinately regulate β‐catenin signaling in gastric cancer. Rescue studies confirm that tomivosertib inhibits gastric cancer via targeting MNK‐ eIF4E‐β‐catenin. Finally, we demonstrate that the in vitro functional and mechanism observations are translatable to in vivo gastric cancer model in mice. Tomivosertib is now in Phase 2 clinical trials. This model integrates deep learning algorithms with an attention mechanism, and innovatively incorporating structural images of COFs and the statistical features derived from these images as model inputs. Results reveal that the COFNet can satisfactorily predict the specific surface area of COFs. It significantly outperforms the publicly available Zeo++ software. Background: CXCL1 belongs to a member of the ELR + CXC chemokine subgroups that also known as GRO-alpha. It has been recognized that several types of human cancers constitutively express CXCL1, which may serve as a crucial mediator involved in cancer development and metastasis via an autocrine and/or paracrine fashion. However, the expression pattern and clinical significance of CXCL1 in human uterine cervix cancer (UCC), as well as its roles and mechanisms in UCC tumor biology remains entirely unclear.Methods: The expression and clinical significance of CXCL1 in UCC tissues was explored using immunohistochemistry and bioinformatics analyses. The expression and effects of CXCL1 in HeLa UCC cells were assessed using ELISA, CCK-8 and transwell assays. Western blotting experiments were performed to evaluate the potential mechanism of CXCL1 on malignant behaviors of HeLa UCC cells.Results: The current study demonstrated that CXCL1 was expressed in HeLa UCC cells, PHM1-41 human immortalized cervical stromal cells, as well as cervical tissues, with UCC tissues having an evidently high level of CXCL1. This high level of CXCL1 in cancer tissues was notably related to poor clinical stages and worse survival probability, rather than tumor infiltration and patient age. In addition, CXCL1 expression was extremely correlated with CCL20, CXCL8 and CXCL3 cancer-associated chemokines expression. In vitro, the growth and migration abilities of HeLa cells were significantly enhanced in the presence of exogenous CXCL1. Gain-function assay revealed that CXCL1 overexpression significantly promoted growth and migration response in HeLa cells in both autocrine and paracrine manners. Finally, we found that CXCL1 overexpression in HeLa cells influenced the expression of ERK signal-related genes, and HeLa cell malignant behaviors derived from CXCL1 overexpression were further interrupted in the presence of the ERK1/2 blocker.Conclusion: Our findings demonstrate the potential roles of CXCL1 as a promoter and a novel understanding of the functional relationship between CXCL1 and the ERK signaling pathway in UCC. Existing studies have proved that the incorporation of polypropylene fiber can effectively improve the impact resistance of concrete. In this paper, the split Hopkinson pressure bar and digital pulse shaping technology are used to further study the dynamic compression performance and constitutive relationship of concrete in the cold regions under the deteriorating effect of freeze-thaw cycle, and obtain the dynamic increase factor. Studies have shown that polypropylene fiber-reinforced high-strength concrete has a strain rate enhancement effect. The number of freeze-thaw cycle times at low strain rates has an obvious effect on concrete deterioration, and the effect is weaker at higher strain rates. By comparing three typical dynamic strength growth factor models with the test results, after revising the model, the dynamic increase factor equations under different freeze-thaw circulation times were fitted. To explore the dynamic response and damage evolution of coal under multiple high-pressure air blasting (HPAB), simulated coal specimens were used in the HPAB experiments, and the variation laws of stress field, vibration field, damage field, and cumulative fracture failure process in specimens were analyzed from a macro point of view. A scanning electron microscope (SEM) and mercury intrusion porosimetry (MIP) were used to observe the pore distribution near the blasthole of the specimen and analyze the variation law of pore structure parameters under multiple HPAB from the microscopic point of view. The test results show that (1) the stress wave generated by HPAB has a great impact on the near zone. After multiple HPAB, the damage value at the place 50 mm away from the blasthole increases by 3.91 times compared with the one shock from HPAB, and the strain peak and vibration velocity are reduced by 17.86% and 63.05%, respectively. With the increase of distance, the internal damages of the specimens in the middle and far zones are mainly driven by the stress wave and the high-pressure air, and the strain peak, vibration velocity, and damage degree gradually decrease. (2) With the increase of shock times in HPAB tests, the stress wave attenuation index decreases at first and then increases, and the damages degree of the middle and far zones increase slowly in the first few shocks and then increase rapidly. The site coefficient (k) shows an overall decreasing trend, whereas the attenuation coefficient (α) tends to increase. (3) The multiple HPABs have a great impact on the pore structure of the specimens. Compared with unshocked specimen, the cumulative mercury injection and pore volume increased by 152.04% and 135.05%, respectively. The number of connecting pores with large pore diameter is significantly increased. The multiple HPAB can effectively improve the pore and fracture structure in the specimens and form a relatively developed fracture network channel. High-pressure air blasting (HPAB) is one type of physical blasting technique that enhances the extraction rate of coalbed methane by impacting the coal mass with high-pressure gas to create cracks within it. First, based on the physical and mechanical parameters of the simulated coal rock mass, the RHT constitutive model of the coal rock mass was established, and its parameters were determined. Then, the laws of crack propagation and stress wave decay in coal induced by high-pressure air blasting were revealed by comparing the effect with that of equivalent explosive blasting. Next, the HPAB experiment was simulated to explore the coal crack propagation law under in-situ stress conditions. Finally, the HPAB experiment was carried out and the results of this experiment were compared with the numerical simulation results. The results indicate that the crack propagation induced by high-pressure air blasting is considered as two major stages, i.e., the crack initiation and crack propagation stage induced by the stress wave and the crack stable propagation stage induced by the duration high-pressure gas. In the case of equal energy, the peak stress wave of high-pressure gas is smaller, decays more slowly and has a longer action time, compared to explosive blasting. Therefore, the number of initial random cracks in coal mass induced by high-pressure air blasting is less, and the range of crack propagation induced by high-pressure air blasting is larger. When λ = 0 (λ is the ratio of the horizontal in-situ stress to the vertical in-situ stress), the in-situ stress in the coal seam can promote the propagation of vertical cracks but inhibit the propagation of horizontal cracks. The comparison of ibuprofen with acetaminophen for blood pressure (BP) in preeclampsia remains controversial. We conduct a systematic review and meta-analysis to compare the impact of ibuprofen versus acetaminophen on BP for preeclampsia. We search PubMed, Embase, Web of Science, EBSCO, and Cochrane Library databases through October 2019 for randomized controlled trials (RCTs) assessing the effect of ibuprofen versus acetaminophen on BP for preeclampsia. This meta-analysis is performed using the random-effect model. Four RCTs are included in the meta-analysis. Overall in preeclampsia patients, ibuprofen and acetaminophen show similar systolic BP (SBP) (standard mean difference [SMD] = 0.04; 95% CI = −0.26–0.34; p=.81), diastolic BP (DBP) (SMD = 0.15; 95% CI = −0.18–0.48; p = 0.38), mean BP (MAP) (SMD = 0.02; 95% CI = −0.29–0.33; p =.91), severe range BP (SMD = −0.10; 95% CI = −0.40–0.19; p =.50), severe hypertension (SMD = 1.18; 95% CI = 0.85–1.62; p =.32), and satisfaction level (SMD = 1.2; 95% CI = 0.95–1.53; p =.13). With the proposal of China's dual-carbon goal, the recovery of industrial waste heat (IWH) for district heating has gained increasing attention. Recently, some scholars have proposed that combined heat and water (CHW) systems are more economical compared with separate heat and water supplies. Beijing lacks desalinated water (DW) and clean low-carbon heat, whereas Tangshan in Hebei Province is rich in IWH. By combining CHW with seasonal thermal energy storage, a case of long-distance transport of hot DW from Tangshan to Beijing was planned, and the costs, energy consumption, and feasibility were analyzed. As a result, under the optimal heat source temperature and effect number of 70 °C and 10 for seawater desalination, the heat from various IWH sources of the three costal steel plants is recovered maximally to produce about 35000 t/h of 95 °C DW. Then, the hot DW is transported to Beijing with a long-distance heat loss ratio controlled below 3 %. Annually, 271.2 million tons of DW and 91.5 million GJ of heat is supplied. The introduction of IWH can save 2.68 billion Nm3 of natural gas per year and the discounted payback period is about 10 years, which has obvious energy-saving, environmental, and economic benefits. • Industrial waste heat is recovered for combined heat and water supply. • Annual utilization of waste heat from three iron and steel plants is maximized. • Thermal economic analysis of combined heat and water system is carried. • Economic long-distance transport radius reaches 200 km. • Seasonal thermal storage plays a dual role in heat storage and peak shaving. Purpose: The longitudinal influence of parental leisure‐time physical activity (LTPA) on their offspring's LTPA is poorly understood. This study examined the longitudinal associations between parental LTPA and offspring's LTPA at two‐time intervals. Method: Child (offspring) participants (N = 3596) were enrolled from the Cardiovascular Risk in Young Finns Study in 1980. Their LTPA was self‐rated through nine phases from baseline to 2018 and categorized by year into youth (1980‒1986) and adult (1992‒2018) LTPA. Parental LTPA was assessed with a single self‐reported question at three phases from 1980 to 1986. Latent growth curve modeling stratified by gender was fitted to estimate the potential pathways between parental LTPA and offspring's youth and adult LTPA. Results: Higher initial levels of paternal and maternal LTPA were independently associated with greater initial levels of youth and adult LTPA of offspring in both genders, respectively, except maternal LTPA, which did not associate with male offspring's adult LTPA. The initial levels of paternal LTPA were directly related to changes in male offspring's youth LTPA after adjusting for age, residential place, paternal education and occupation, having siblings, and offspring's body mass index. Conclusion: Our study demonstrates that the initial levels of parental LTPA are directly linked to the initial levels of offspring's LTPA during youth and adulthood, while changes in parental LTPA are unrelated to changes in offspring's youth and adult LTPA for either gender over time. Earth's water resources are critical for supporting livelihoods and food security but are being increasingly overexploited to support global agriculture. Diversifying cropping systems could potentially resolve unsustainable water use but trade‐offs with other aspects of sustainability and food security have not yet been assessed. We performed a detailed analysis of 31 different field crop rotations conducted during 1990–2019 in the North China Plain, to assess the potential impact of crop diversification on actual evapotranspiration (ETa), changes in regional groundwater table, grain yield, economic output, and water use efficiency (WUE) and to identify configurations that can achieve co‐benefits across multiple dimensions. We found that a combination of lowering the cropping index (i.e., harvest frequency), incorporating fallow periods, and introducing higher‐value crops into the currently dominant winter wheat–summer maize double cropping system can reduce growing season ETa by as much as 31%, mitigate groundwater decline by 19% or more, and increased economic output and economic WUE by more than 11% and 3%, respectively. We also found that multiple diversified wheat‐maize–based rotations—all with rotation lengths greater than 2 years—achieve co‐benefits across all evaluated dimensions. This study provides new empirical evidence of the opportunities for diversified crop rotations to balance the multiple objectives of food production, sustainable groundwater use, and farmer profitability. Controlled blasting technology is an effective way to enhance coalbed methane recovery, but the theoretical research has fallen behind engineering practice seriously. Therefore, in this work, based on the damage and fracture mechanics theory, the coal damage process and mechanism under the multiple actions of the blasting stress wave (BSW) were analyzed firstly. Then, the simulation experiment of coal damage accumulation was designed and carried out; during the experiment, the ultrasonic velocity of the samples was measured and the damage values were calculated. Under the multiple actions of BSW, the initial radial cracks around the blast-hole wall or in the area near the explosion source are formed under the action of tangential tension of BSW and the internal pressure of the blast-hole, and under the combined actions of stress wave, gas, and in situ stress, the primary cracks in the middle and far area of the explosion source expand and form the mesoscopic cracks because of the damage accumulation at the crack tip. And then the mesoscopic cracks grow and intersect to form a macroscopic crack. Finally, the macroscopic cracks expand and intersect with the initial radial cracks, the samples break suddenly under the load far below their fracture toughness because of the damage accumulation effects. A very challenging but meaningful task in geodynamic research is determining how to clearly disentangle and quantify the frequency-dependent barometric responses of borehole dilatometers. However, we still lack a fundamental understanding of how the Chinese TJ-2 borehole dilatometer network has responded to barometric pressure at different frequencies since the late 1990s. For this reason, we systematically analyze the transfer functions of volumetric strain changes associated with barometric pressure at 21 stations. The results demonstrate that the barometric responses for most sites are generally flat and near-flat in the low- (0.1–0.5 cpd) and high-frequency (8–30 cpd) bands, respectively, while drastically fluctuating in the intermediate-frequency band (0.5–8 cpd) due to the tidal influences. Overall, many sites exhibit strongly frequency-dependent behaviors over the whole frequency range (0.1–30 cpd), which are likely controlled mainly by the complex properties of sensor-grout-formation systems. In light of both the low- and high-frequency barometric responses of dilatometers, whether the surrounding formations are linear elastic medium can be verified. The mechanical properties of three stations, with surrounding formations that are thought to be linear elastic structures, can therefore be roughly but quantitatively estimated by utilizing barometric responses, which are shown to be consistent with the parameters tested by rock mechanics. Altogether, this work not only provides a useful method for frequency-dependent pressure correction, but also offers profound insights to advance sensor-grout-formation modeling. Coal separation based on computer vision has attracted substantial attention in recent years. However, developing reliable object detection models relies on large-scale annotated dataset, which in industrial practice is time-consuming and labor-intensive to obtain. In this paper, we propose a novel data augmentation model called dual attention deep convolutional generative adversarial network (DADCGAN) to expand dataset scale and improve object detection. For the first time, the proposed DADCGAN, which adopts DCGAN as its foundation architecture, introduces efficient channel attention and external attention mechanisms to capture essential feature information from the channel and spatial dimensions of images, respectively. Moreover, spectral normalization and two time-scale update rule strategies are incorporated to stabilize the training process. The implementation of our proposed data augmentation strategy includes two steps. First, traditional pixel transformation is used to expand an original small dataset. Then, our GAN-based data augmentation is executed to further expand the dataset by generating synthetic images. Experimental results show that our DADCGAN model achieves the lowest FID value, decreasing the FID by 21.30–71.96 % compared to other baseline GAN models, showcasing its ability to produce more realistic coal-gangue images. Finally, the data augmentation strategies are applied to the YOLOv4 model, enhancing the mAP by 9.26 %, highlighting its significance in enhancing coal-gangue object detection. Image style transfer is a significant challenge in computer vision, where the goal is to transfer the style of a reference image to a source image while preserving its background and feature information. Despite previous progress in this area, many studies have overlooked the issues of local details and complexity, leading to unrealistic generated images and slow model inference speeds. This paper proposes an efficient image style transfer method based on cross-fusion attention (CFA), inspired by generative adversarial networks (GAN) and a new joint loss. Our novel CFA encodes local contextual information between low-level and high-level features, models remote dependencies of features, and embeds CFA into the encoder of our proposed model to increase the diversity of small features and focus on more prominent feature parts. Furthermore, we use frequency domain loss to improve image reconstruction quality and a joint loss to train the network. Our experiments on multiple datasets demonstrate that our proposed method offers significant advantages in terms of inference speed, background information preservation, and local details, making it a promising approach to image style transfer. To verify the in vitro results, we used a xenograft model to validate the capability of miR‐188 in tumorigenesis. Overexpression of miR‐188 reduced viability and metastasis of cancer stem cells. Similar results were reproduced in vivo, where overexpression of miR‐188 retarded tumour growth in mice. We also identified MDK as a target of miR‐188, and overexpression of MDK was found in lung cancer samples. Overexpressed MDK promoted the malignant behaviours of lung cancer stem cells. In addition, the Hippo pathway was found to be inactivated in lung cancer tissues, presenting as increased levels of YAP and TAZ. Suppression of the Hippo pathway also enhanced lung cancer stem cell activity and promoted the growth of xenograft tumours. To sum up, our results reveal that miR‐188 inhibits the malignant behaviours of lung cancer stem cells and the growth of xenograft tumours. Novel antitubercular agents are urgently needed to combat the emergence of global drug resistance to human tuberculosis. Mycobacterial membrane protein Large 3 (MmpL3) is a promising drug target because its activity is essential and required for cell-wall biosynthesis. Several classes of MmpL3 inhibitors have been developed against Mycobacterium tuberculosis (Mtb) with potent anti-tuberculosis activity. These include the drug candidate SQ109, which has progressed to phase IIb/III clinical trials. Here, we have determined the crystal structures of MmpL3 in complex with NITD-349 and SPIRO. Both inhibitors bind deep in the central channel of transmembrane domain and cause conformational changes to the protein. The amide nitrogen and indole nitrogen of NITD-349 and the piperidine nitrogen of SPIRO interact and clamp Asp645. Structural analysis of the two structures reveals that these inhibitors target the proton relay pathway to block the activity of MmpL3. The findings presented here enrich our understanding of the binding modes of MmpL3 inhibitors and provide directions to enable further rational drug design targeting MmpL3. Zinc finger protein 703 (ZNF703) is a new member of the zinc finger protein family of transcription factors that plays an important role during embryogenesis in metazoans. The overexpression of ZNF703 contributes to tumorigenesis and progression of a number of malignancies by activating the Akt/mammalian target of rapamycin (mTOR) signaling pathway. This pathway is activated in medullary thyroid cancer (MTC), but its mechanism of action is not yet fully understood. The aim of the present study was to examine the role of ZNF703 and its association with Akt/mTOR activation in MTC. The present study used the phosphorylation of Akt1 protein at serine 473 (pAkt473) as an indicator of signaling activation. Immunohistochemistry (IHC) staining and western blot analyses were performed in order to examine the expression of ZNF703 in 34 cases of MTC and 12 cases of corresponding normal thyroid tissues. ZNF703 expression in MTC was significantly higher compared with the corresponding normal thyroid tissues (P<0.05). Furthermore, expression of ZNF703 was associated with tumor size, lymph node metastasis and advanced stage of disease. IHC also demonstrated that the level of ZNF703 was positively correlated with p-Akt473 in the 34 cases of MTC. The human MTC cell line TT was selected for further investigation as TT cells exhibit Akt/mTOR activation. The biological effects of silencing ZNF703 in TT cells on proliferation and apoptosis, both in vitro and in vivo were investigated in the present study. ZNF703 silencing inhibited the proliferation of TT cells in vitro and inhibited xenograft tumor growth in vivo. These effects were accompanied by the substantial decrease of pAkt473 and the induction of p53 protein. To study the freeze-thaw damage characteristics of concrete, the uniaxial compressive tests of concrete under different number of freeze-thaw cycles were conducted, and the damage variable of freeze-thaw was obtained. The test results showed that the stress was a function of strain and freeze-thaw damage variable, and it can describe the degradation of concrete strength. Meanwhile, the equation for the stress-strain curved surface about strain and freeze-thaw damage variable was also proposed in this paper. The derivative function of the stress-strain curved surface equation with respect to strain presented the change of elastic modulus with the increase of freeze-thaw cycle number. Nitrogen, phosphorus co-doped carbon dot has been synthesized via a hydrothermal method and it gives rise to bright green emission at 504 nm and the excitation-dependent behavior is monitored. Such nanoplatform displays rapid and highly selective detection to nitrite ions in water. Carbon nanomaterials have aroused considerable interests due to excellent optical performances, but most of achieved carbon dots possess blue luminescence under UV-light irradiations. Herein, we report the preparation of phosphorus and nitrogen co-doped carbon quantum dots (PNCQDs) via hydrothermal method by employment of 1,3-phenylenediamine, citric acid and H 3 PO 4 as precursors. The particle size of the water-soluble PNCQDs is in the range between 4 and 6 nm and the average diameter is determined to be 4.75 nm. The resulted quantum dots give rise to bright green emission at 504 nm and the excitation-dependent behavior has been found. Moreover, it demonstrates luminescence stability toward a broad pH range from 1 to 10. Fluorometric titration studies reveal that close interactions occur between carbon dots and nitrite ions and a switched off effect has been achieved. • We designed a blasting-vibration damage accumulation experiment, during the experiment, the ultrasonic wave velocity was monitored, and the vibration damage value of the specimens were calculated. • We analyzed the change process of the ultrasonic wave velocity and the accumulation process of vibration damage after the samples were subjected to different vibration loads. • The strength and durability tests of the concrete were carried out to analyze the influence of the effect of damage accumulation from blasting vibration on the uniaxial compressive strength and the durability (impermeability, carbonization resistance, and resistance to chloride ion penetration) of young concrete. • Based on the experiment results, we put forward a blasting-vibration safety standard for young concrete based on the damage accumulation effect, in order to ensure the strength and durability of the concrete, the blasting-vibration velocity should be controlled within 1.5 cm/s. In blasting engineering practice, the accumulation of blasting-vibration damage is inevitable. The objective of this study is to put forward a blasting-vibration safety standard for young concrete based on the effects of damage accumulation. The ultrasonic wave velocity change process and the vibration damage accumulation process of the young concrete were investigated based on the blasting-vibration damage accumulation experiment. Furthermore, the influence of the damage accumulation effect on the compressive strength and durability of concrete were studied. Young concrete subjected to multiple vibration loads, when the vibration velocity is less than 1.0 cm/s, the change process of the ultrasonic wave velocity can be divided into three stages, the damage deterioration effect of vibration do not appear, and the vibration can improve the compressive strength and the durability of the concrete. However, when the vibration velocity is greater than 1.5 cm/s, the change process of the ultrasonic wave velocity goes through four stages, the damage deterioration effects of vibration begin to appear in the second stage and appear macroscopically in the third stage. If the vibration velocity reaches 3.5 cm/s, the damage deterioration effects appear macroscopically from the first stage, the compressive strength and durability of concrete decrease, and the degree of reduction increases with the increasing of vibration velocity. The experimental results indicate that the velocity of blasting ground vibration of 1.5 cm/s should be used as the blasting-vibration safety standard for young concrete based on the damage accumulation effect. Oleanane-type ginsenosides are a class of compounds with remarkable pharmacological activities. However, the lack of effective preparation methods for specific rare ginsenosides has hindered the exploration of their pharmacological properties. In this study, a novel glycoside hydrolase PlGH3 was cloned from Paenibacillus lactis 154 and heterologous expressed in Escherichia coli. Sequence analysis revealed that PlGH3 consists of 749 amino acids with a molecular weight of 89.5 kDa, exhibiting the characteristic features of the glycoside hydrolase 3 family. The enzymatic characterization results of PlGH3 showed that the optimal reaction pH and temperature was 8 and 50 °C by using p-nitrophenyl-β-d-glucopyranoside as a substrate, respectively. The Km and kcat values towards ginsenoside Ro were 79.59 ± 3.42 µM and 18.52 s−1, respectively. PlGH3 exhibits a highly specific activity on hydrolyzing the 28-O-β-d-glucopyranosyl ester bond of oleanane-type saponins. The mechanism of hydrolysis specificity was then presumably elucidated through molecular docking. Eventually, four kinds of rare oleanane-type ginsenosides (calenduloside E, pseudoginsenoside RP1, zingibroside R1, and tarasaponin VI) were successfully prepared by biotransforming total saponins extracted from Panax japonicus. This study contributes to understanding the mechanism of enzymatic hydrolysis of the GH3 family and provides a practical route for the preparation of rare oleanane-type ginsenosides through biotransformation. Abstract The successful invasion of alligator weed (Alternanthera philoxeroides) into both aquatic and dry terrestrial environments has been attributed to its phenotypic plasticity and high rates of growth and vegetative propagation. Here, we evaluated whether the anatomical structures of alligator weed make it well adapted to colonize the aquatic-terrestrial transition zone. Anatomical and histochemical analysis revealed an impermeable apoplastic barrier that consists of a cuticle and an endodermis of the adventitious root and stem in both aquatic and terrestrial environment; stem and root possess a periderm in the terrestrial environment; and the root has a lignified cortex and hypodermis in the aquatic environment. A supernumerary cambia produces secondary vascular bundles, parenchyma and adventitious buds. Dilated parenchyma cells exist in the aged terrestrial roots. Aerenchyma tissue and pith cavities in the roots and stems produce air spaces, and the aquatic roots possess large aerenchyma compared with intercellular aerenchyma of terrestrial roots. In summary, phenotypic plasticity of anatomical characters, such as impermeable apoplastic barriers, air spaces, and reproductive supernumerary cambia, may be evidence that alligator weed is well adapted to the aquatic–terrestrial transition zone. The propagation and attenuation rule of blasting vibration wave parameters is the most important foundation of blasting vibration prediction and control. In this work, we pay more attention to the influence of the damage accumulation effect on the propagation and attenuation rule of vibration wave parameters. A blasting damage accumulation experiment was carried out, the ultrasonic wave velocity of the specimens was measured, and the damage value was calculated during the experiment. The blasting vibration wave was monitored on the surface of the specimens, and its energy was calculated by using the sym8 wavelet basis function. The experimental results showed that with the increase in the number of blasts, the damage continues to increase; however, the vibration velocity and the main frequency decrease continuously, the unfocused vibration wave energy in the zone near to the blasting source is rapidly concentrated in the low-frequency band (frequency bands 1 to 3), and the energy is further concentrated in the low-frequency band in the intermediate zone and zone far from the blasting source. There is a distortion process in which the vibration velocity and the main frequency increase slightly and the energy of the blasting vibration wave converges to the high-frequency band (the 5th band) before the sudden unstable fracture failure of the specimens. The experimental results indicate that the prediction and evaluation of blasting vibration should consider the variation rule of blasting vibration wave parameters synthetically based on the cumulative damage effect, and it is not safe to use only one fixed vibration control standard for the whole blasting operation. Purpose: The long‐term effects of sociodemographic and health characteristics on television viewing (TV) time changes have not been identified in adulthood. We aimed to examine the modifiable and non‐modifiable determinants of changes in TV‐time in young adults over 10 years. Methods: Participants (N = 2929) aged 24‐39 years were recruited between 2001 and 2011 from the Cardiovascular Risk in Young Finns Study. Data were collected using questionnaires and a medical examination. The determinants of changes in TV‐time were estimated using latent growth modeling for men and women separately. Results: For men, inverse associations with initial levels of TV‐time were observed for students becoming employed and already has children, and direct associations were observed for both those who stayed a smoker and those who stayed overweight/obese. Increasing attention to health habits was inversely associated with a slope of TV‐time, whereas age and becoming unemployed were positively associated with the slope of TV‐time. For women, inverse associations with the levels of TV‐time were found for age, staying in non‐manual work, and paying consistently high and increasing attention to health habits, and direct associations were found for staying unemployed, smoking and overweight/obese, and becoming employed, single and non‐smoking. Increasing physical activity, becoming employed, motherhood, and normal weight were inversely associated with the slope of TV‐time, whereas age and staying in non‐manual work were positively associated with the slope of TV‐time. Conclusions: This suggests several gender‐specific determinants of changes in TV‐time that can help identify potential targets for interventions to prevent excessive TV‐time in adulthood. Accurate prediction of carbon price effectively ensures the stability of the carbon trading market and reduces carbon emissions. However, making accurate prediction is challenging because the carbon price is highly nonlinear and nonstationary due to complex influential factors. Thus, we propose a multifactorial hybrid forecasting framework, ET-MVMD-LSTM, to integrate three advanced algorithms for a reliable multi-step ahead prediction of the carbon price. First, extremely randomized tree (ET) is used to determine the optimal input variables for the modeling to follow. Then, multivariate variational mode decomposition (MVMD) is executed to simultaneously decompose the screened input variables into relatively regular sub-modes, which reflect characteristics at different scales. Subsequently, long short-term memory (LSTM) with a stable forecasting ability is employed to model each mode individually to effectively extract the long-term trend and short-term fluctuation features. The final forecast is reconstructed by the ensemble of the predictions of all sub-modes. Plasma spray physical vapor deposition (PS-PVD) is a new promising technology for processing thermal barrier coatings (TBCs). In this study, the microstructural evolution of PS-PVD yttria-stabilized zirconia (YSZ) TBCs at 1150 °C was investigated by scanning electron microscopy, electron backscattered diffraction, and impedance spectroscopy. Bridging is observed between the secondary columns of the coatings during the first 8 h of heat treatment, while sintering necks are observed between the quasi-columns over the heat-treatment interval of 12–20 h. As the heat-treatment time increases to 100 h, the nanogaps between the secondary columns disappear and a large number of shrinkage pores are observed in the columns, leading to a typical cellular structure. Besides, the average grain size of the YSZ coatings increases from 0.20 (as-deposited) to 0.55 µm (after 50 h of heat treatment). The resistance of the YSZ grains increases while that of the YSZ grain boundaries decreases because of grain coarsening. During the first 4 h of heat treatment, the resistance of the thermally grown oxide (TGO) scale increases because of its thickening. An electrogenerated chemiluminescence (ECL) biosensor array for the individual and simultaneous detection of multiple biomarkers has been developed at first time on the basis of the aptamer-based biosensor incorporating a versatile signal probe. As principle-of-proof, the acute myocardial infarction (AMI) biomarkers, myoglobin(Myo), cardiac troponin I(cTnI) and cardiac troponin T(cTnT) were chosen as model analytes. The biosensor array was fabricated by self-assembling thiolated specific ssDNA aptamers on the surface of gold electrodes, respectively. After each target analyte was bound to the capture probes and then to each corresponding biotinylated antibody, and finally to the versatile ECL signal probe, ECL signals were recorded using PMT or CCD as a detector. For PMT model, the developed method showed extremely low detection limits for 0.30 ng/mL cTnT, 31 pg/mL Myo, 0.79 pg/mL cTnI. For ECL image model, a biosensor array containing three target biosensors and a control biosensor was found to be highly sensitive, no cross-talking, and accurate towards simultaneous detection of the AMI biomarkers. An understanding of biomass yield and quality response to N supply is essential to breeding and cultivation of sorghum ( Sorghum bicolor (L.) Moench) for the production of bioethanol or forage. A two – year field experiment was conducted in 2013 and 2014 on semiarid marginal land in Inner Mongolia (39°10′N, 109°53′E) to determine the effects of N fertilizer rate (0, 60, 120, and 240 kg ha −1 ) on biomass yield and quality of sweet sorghum (GT-8) and biomass sorghum (GN-11), and on soil conditions. Results indicated that (1) biomass yield, leaf/stem ratio, and crude protein (CP), protein yield, theoretical ethanol yield (TEY), and soil net NO 3 − -N accumulation of both sorghum varieties in 2013 and 2014 significantly increased with the increasing N fertilizer rate from 0 to 240 kg ha −1 , whereas dry matter ratio and relative feed value (RFV) varied inversely with the N application rates. (2) Compared to biomass sorghum (GN-11), sweet sorghum (GT-8) demonstrated significant enhancement of forage quality components including CP, dry matter digestibility (DMD), dry matter intake (DMI), total digestible nutrients (TDN), net energy for lactation (NE L ), and RFV, with stems exhibiting higher forage quality than leaves. Biomass sorghum (GN-11) exhibited better performance for bioenergy production, with an average biomass yield of 7.9 t ha −1 and TEY of 3046.7 L ha −1 averaged across all N rates. (3) Annual average amount of net NO 3 − -N accumulation in 0–90 cm layers of two years significantly increased when fertilizer N exceeded 120 kg N ha −1 . In short, N fertilizer of 60 kg ha −1 is recommended to sweet sorghum for the forage production and 120 kg N ha −1 to biomass sorghum for bioenergy feedstock sustainability, respectively, meanwhile to prevent the soil degradation on semiarid marginal lands. The weak interlayer interaction and strong intralayer bonding in two-dimensional (2D) lamellar materials give rise to ultra-low friction properties when the contacted interface between two crystalline is incommensurate, known as structural superlubricity. The origin of friction in a two-dimensional heterojunction is complicated by the interplay among normal forces, edge effects and twist angle (θ). In this article, molecular dynamics (MD) has been adopted to theoretically investigate the friction characteristics in graphene/MoS 2 layered heterojunction. The results demonstrate that friction of graphene/MoS 2 interface at θ = 0° is approximately 100 times larger than the one at θ = 30°. Unexpectedly, whether graphene slider experiences in-plane vibrational motion perpendicular to the sliding path depends on the initial configuration and relative twist angle. To quantitatively understand the origin of friction, the friction contributions from different edge atoms and internal atoms have been investigated. It is found that friction of the edge perpendicular to the sliding direction is much larger than that of the other edges. An electrochemiluminescence (ECL) bioassay with high sensitivity and anti-fouling ability was developed for determination of matrix metalloproteinase 9 (MMP-9) secreted from living cells under external stimulation. A peptide with sequence of CLGRMGLPGK and a new cyclometalated iridium(III) complex bearing carboxyl group, (pq)2Ir(dcbpy) (pq = 2-phenylquinoline, dcbpy = 2,2′-bipyridyl-4,4′-dicarboxyli acid, abbreviated as Ir) were employed as molecular recognition substrate and ECL emitter, respectively. The peptide was labelled with the Ir to form Ir-peptide as ECL probe. Ir-peptide was self-assembled onto Nafion and gold nanoparticles (AuNPs) modified glassy carbon electrode (AuNPs/Nafion/GCE) and then both of 6-mercapto-1-hexanol (MCH) and zwitterionic peptide as blocking reagents were co-assembled on Ir-peptide/AuNPs/Nafion/GCE to form an anti-fouling ECL peptide-based biosensor. MMP-9 can be quantified in the range 1.0–50 ng·mL−1 with a detection limit of 0.50 ng·mL−1 based on the decreased ECL intensity. Relative standard derivation was 2.3% for six fabricated anti-fouling ECL peptide-based biosensors after reaction with 50 ng·mL−1 MMP-9. The anti-fouling ECL peptide-based biosensor can be used to monitor MMP-9 secreted from living cells under external stimulation. 96.0%–108.0% of recoveries were obtained in 60-diluted cell culture media. This study demonstrates that the ECL biosensor by the combination of iridium(III) complex-based sensitive ECL method and the anti-fouling interface provides a promising way for the determination of MMP-9 in biological sample, which is viable in clinical diagnosis and point-of-care test of protease. Based on CT and MRI images acquired from normal pressure hydrocephalus (NPH) patients, using machine learning methods, we aim to establish a multimodal and high-performance automatic ventricle segmentation method to achieve an efficient and accurate automatic measurement of the ventricular volume. First, we extract the brain CT and MRI images of 143 definite NPH patients. Second, we manually label the ventricular volume (VV) and intracranial volume (ICV). Then, we use the machine learning method to extract features and establish automatic ventricle segmentation model. Finally, we verify the reliability of the model and achieved automatic measurement of VV and ICV. In CT images, the Dice similarity coefficient (DSC), intraclass correlation coefficient (ICC), Pearson correlation, and Bland–Altman analysis of the automatic and manual segmentation result of the VV were 0.95, 0.99, 0.99, and 4.2 ± 2.6, respectively. The results of ICV were 0.96, 0.99, 0.99, and 6.0 ± 3.8, respectively. The whole process takes 3.4 ± 0.3 s. In MRI images, the DSC, ICC, Pearson correlation, and Bland–Altman analysis of the automatic and manual segmentation result of the VV were 0.94, 0.99, 0.99, and 2.0 ± 0.6, respectively. The results of ICV were 0.93, 0.99, 0.99, and 7.9 ± 3.8, respectively. The whole process took 1.9 ± 0.1 s. We have established a multimodal and high-performance automatic ventricle segmentation method to achieve efficient and accurate automatic measurement of the ventricular volume of NPH patients. Energy sorghum tolerates adverse climatic and edaphic conditions and has great potential as biofuel feedstock in marginal land. This study investigates the potential energy sorghum biomass production and uptake of nitrogen (N), phosphorus (P), and potassium (K) on a sandy loam marginal land in a semi-arid region, in order to define optimum N fertilizer rate to produce the highest biomass yield with minimal nutrient elimination. Five N rate treatments (0, 60, 120, 180, and 240 kg ha) and two sorghum varieties (sweet type Guotian-8 (GT-8) and biomass type Guoneng-11 (GN-11)) were used. Yield increment was observed as N level increased, but the standout treatment appeared to be N rate of 60 kg ha which significantly increased biomass yield vs. controls by 68.8% in 2014 and 64.1% in 2015. Biomass yield exhibited non-significant differences between N rate treatments from 60 to 240 kg ha, although the highest biomass yield (9.2-11.9 t ha) was observed in the 120 kg N ha treatment. Nutrient analysis showed that N, P, and K accumulation in aboveground plants increased with N rate increase, ranging between 32.2 and 119.1, 7.9 and 19.2, and 22.1 and 94.0 kg ha, respectively, for the highest N rate of 240 kg ha. Substantial amounts of N were extracted from the soil in control and 60 kg N ha treatments, despite the low fertility and organic matter content of the soil. Moreover, nitrogen (N) use efficiency (NUE) was maximized at lower N rates. A decline in physiological N use efficiency (PNUE) resulted in decreased agronomic N use efficiency (ANUE) at higher N rates. Hence, it is concluded that N fertilizer rate between 60 and 120 kg ha would be the optimal N requirement to facilitate sustainable production of energy sorghum on a sandy wasteland. Purpose: We examined associations between childhood temperamental activity, physical activity (PA), and television (TV) viewing over a 30-year period. Method: The participants (1220 boys and 1237 girls) were aged 3, 6, 9, and 12 years in 1980 and were followed until 2011. Temperamental activity was evaluated by participants' mothers at baseline. The PA was assessed based on maternal ratings of the child from ages 3 to 6 and via self-report age from the age of 9 across all measurements. TV viewing was assessed using self-reports taken from 2001 to 2011. The associations between temperamental activity and the level and change of PA and TV viewing were determined using linear growth modeling stratified by gender and age group. Results: High temperamental activity assessed from ages 9 to 12 was associated with high levels of childhood PA in both genders, but with a steeper decline in PA levels during the first 9 years of follow-up in boys. High temperamental activity assessed from ages 3 to 6 was associated with the decline of PA from childhood to youth in girls. High childhood temperamental activity was associated with decreased levels of PA in adulthood in men, but not in women. The associations between childhood temperamental activity and TV viewing during adulthood seemed to be positive but not consistently significant in all age and gender groups. Conclusion: High temperamental activity may contribute to the development of a physically inactive lifestyle. The combined heat and water (CHW) system has been studied by many scholars since it was proposed, and a project has been implemented in Shandong, China in 2021. However, there have been few studies on optimization and comparisons of cases on the heat source side of CHW. Taking a coal-fired thermal power plant as an example, through the reasonable matching of steam flows and analysis of thermal economic parameters, the effect number and heat source steam temperature of a multi-effect distillation (MED) + multi-stage flash (MSF) combined heat and water production (CHWP) system were optimized. The optimal MED heat source temperature was found to be 70 °C, and the optimal MED and MSF effect numbers were 10 and 5, respectively. Finally, the waste heat in the power plant was utilized as much as possible throughout the year. However, the prediction of PM 2.5 and PM 10 concentrations has great uncertainty and instability because of the dynamics of atmospheric flows, making it difficult for a single model to efficiently extract the spatial–temporal dependences. This paper reports a robust forecasting system to achieve accurate multi-step ahead forecasting of PM 2.5 and PM 10 concentrations. First, correlation analysis is adopted to screen the spatial information on pollution and meteorology that may facilitate the prediction of concentrations in a target city. Then, a spatial–temporal attention mechanism is used to assign weights to original inputs from both space and time dimensions to enhance the essential information. Subsequently, the residual-based convolutional neural network with feature extraction capabilities is employed to model the refined inputs. Finally, five accuracy metrics and two additional statistical tests are applied to comprehensively assess the performance of the proposed forecasting system. In addition, experimental studies of three major cities in the Yangtze River Delta urban agglomeration region indicate that the forecasting system outperforms various prevalent baseline models in terms of accuracy and stability. Quantitatively, the proposed STA-ResCNN model reduces root mean square error by 5.595 %-15.247 % and 6.827 %-16.906 % for the average of 1–4 h ahead predictions in three major cities of PM 2.5 and PM 10 , respectively, compared to baseline models. The applicability and generalization of the proposed forecasting system are further verified by the extended applications in the other 23 cities in the entire region. ContextEucommiae Cortex and Radix Dipsaci, occurring in a ratio of 1:1 in Du-Zhong-Wan (DZW), a Chinese herbal medicine, is available as a water extract followed by ethanol precipitation for the treatment of osteoporosis, fractures and menopausal syndrome. ObjectiveThis study investigates the protective effects of DZW in ovariectomy (OVX)-induced bone loss in a rat osteopenia model. Materials and methodsSixty Sprague–Dawley rats were randomly divided into the sham-operated group (SHAM) and five OVX subgroups: OVX with vehicle (OVX), 17β-estradiol (E2) and with three graded doses of DZW. Daily oral administration of the different samples started on the fifth week and lasted for 12 weeks, respectively. The body weight, uterus wet weight, serum biochemical parameters, bone mineral density (BMD), bone biomechanical properties, bone microarchitecture and immunohistochemistry were examined. ResultsCompared with the SHAM group, the DZW treatment significantly reversed the osteoporotic changes in OVX rats. The DZW-H group showed that serum tartrate-resistant acid phosphatase 5b (TRACP-5b) levels reduced by 152.25% (p < 0.01) and osteocalein (OCN) levels dose dependently increased by 118.43% (p < 0.01) as compared with the OVX group. Compared with the OVX group, the DZW at different three dosages of DZW evidently increased the right femur BMD by 112.43, 114.56 and 116.45%, and dramatically promoted bone quality and bone strength (p < 0.05). Further, immunohistochemical evaluation also showed that DZW administration increased ER expression in uteri (p < 0.01). ConclusionsDZW exhibits an anti-osteoporotic effect, probably mediated via phyto-estrogenic effects. It might be a potential herbal alternative for the management of postmenopausal osteoporosis. A novel capacitor mismatch calibration method is presented to compensate the capacitor mismatches in a successive approximation register (SAR) ADC. The method features a new weight‐balancing split capacitive DAC which allows automatic extraction and calibration of the capacitor mismatches by itself. As a result, the ADC's resolution can be substantially improved with minimum area and power penalty. The calibration method was verified and implemented in a 14 bit, 10 kS/s SAR ADC, fabricated in a 0.13 µm standard CMOS process. The measured signal‐to‐noise‐plus‐distortion and spurious‐free dynamic range are 72.7 and 86 dB after calibration with 13.6 and 21.8 dB improvements, respectively. New anti-tubercular agents are urgently needed to address the emerging threat of drug resistance to human tuberculosis. Here, we have used structure-assisted methods to develop compounds that target mycobacterial membrane protein large 3 (MmpL3). MmpL3 is essential for the transport of mycolic acids, an important cell-wall component of mycobacteria. We prepared compounds that potently inhibit the growth of Mycobacterium tuberculosis (Mtb) and other mycobacteria in cell culture. The cryoelectron microscopy (cryo-EM) structure of mycobacterial MmpL3 in complex with one of these compounds (ST004) was determined using lipid nanodiscs at an overall resolution of 3.36 Å. The structure reveals the binding mode of ST004 to MmpL3, with the S4 and S5 subsites of the inhibitor-binding pocket in the proton translocation channel playing vital roles. We investigated the association of parental physical activity (PA) trajectories with offspring's youth and adult PA. Self-reported PA data were extracted from the Young Finns Study with three follow-ups for parents between 1980 and 1986 and nine follow-ups for their offspring in youth between 1980 and 2011 (aged 9-39 years, n = 2402) and in adulthood in 2018. Accelerometer-derived PA was quantified in 2018-2020 (aged 43-58 years, n = 1134). Data were analyzed using mixture models and conducted in 2022. We identified three trajectories for fathers and mothers (high-stable activity, 20.2%/16.6%; moderate-stable activity, 50.5%/49.6%; and low-stable activity, 29.4%/33.7%) and four for youth male and female offspring (persistently active, 13.4%/5.1%; increasingly active, 32.1%/43.1%; decreasingly active, 14.4%/12.6%; and persistently low-active, 40.1%/39.1%). Compared to low-stable active parents, high-stable active fathers had a higher probability of having their sons and daughters classified as persistently active, increasingly active, and decreasingly active in youth (Brange = 0.50-1.79, all p < 0.008), while high- and moderate-stable active mothers had significantly increased likelihood of having their daughters classified as persistently active and decreasingly active in youth (Brange = 0.63-1.16, all p < 0.009). Fathers' and mothers' high-stable activity was associated with higher self-reported PA of adult offspring than parental low-stable activity. Persistently active and increasingly active offspring in youth accumulated more adult total PA, moderate-to-vigorous PA, step counts, and self-reported PA than persistently low-active ones (all p < 0.036). Parental persistent PA, particularly paternal persistent PA, predicts offspring's PA concurrently and prospectively. Increasing and maintaining PA in youth predicts higher PA levels in midlife. We consider a live streaming platform connecting streamers and users. Streamers perform for users in a virtual chat room on the platform. Users watch streamers' performances and purchase virtual gifts for their favorite streamers. There are two types of streamers: non-contracted ones who stream independently and contracted ones who sign with and work for the platform. Based on observed industrial practice, the platform provides non-contracted streamers with a straight commission plan and contracted streamers with one of three alternative compensation plans, namely, the basic compensation plan (B-plan), salary-based incentive plan (SI-plan), and commission-based incentive plan (CI-plan). The platform first decides its compensation plans for two types of streamers, then the streamers decide their type and effort level. We explore and compare the streamers' optimal decisions under different compensation plans. We interestingly find that the SI/CI bonus does not always motivate streamers to perform better. The SI bonus is only effective for the streamers whose ability is neither low nor high while the CI bonus can effectively motivate the majority of streamers except those having extremely low ability or some low-intermediate-ability streamers. In addition, the platform can screen out low-ability streamers and attract high-ability streamers to sign with it by properly designing its compensation plans. Finally, we examine managerial insights for the platform. Iris tectorum Maxim, a well-known herb medicine, is commonly used for treatment of inflammation, cough, and pharyngitis for a long time in China. Tectoridin, main active ingredient of Iris tectorum Maxim, is often used for its quality control. This study was aimed to analyze the pharmacokinetic profile of tectorigenin (the metabolite of tectoridin) after oral administration of I. tectorum Maxim extract, and to compare the pharmacokinetic characterization of tectorigenin after oral administration of I. tectorum Maxim extract (ITME) and pure tectoridin (PT) in rats. In addition, a simple, reliable and sensitive UPLC–MS/MS method was developed for determination of tectorigenin in rat plasma, using kaempferol as internal standard. The processed samples were separated on a Poroshell 120 SB-C 18 column and detected by positive electrospray ionization in multiple reaction monitoring (MRM) mode. The method validation results indicated that the established method was simple, specific and reliable. The pharmacokinetic results showed that the plasma concentration of tectorigenin in ITME group was much higher than that of the PT group ( p < 0.01). Moreover, compared to PT group, t 1/2 value and AUC 0–∞ value were also notably increased in ITME group ( p < 0.01). In conclusion, potential interaction exists between those chemical components in ITME, and the co-existing components in ITME could notably promote the absorption of tectoridin in rats, however, the exact compound(s) which enhance the absorption of tectoridin should be investigated in future study.
