Search

NASA has launched the decade-long Arctic-Boreal Vulnerability Experiment (ABoVE). While the initial phases focus on field and airborne data collection, early integration with modeling activities is important to benefit future modeling syntheses. We compiled feedback from ecosystem modeling teams on key data needs, which encompass carbon biogeochemistry, vegetation, permafrost, hydrology, and disturbance dynamics. A suite of variables was identified as part of this activity with a critical requirement that they are collected concurrently and representatively over space and time. Individual projects in ABoVE may not capture all these needs, and thus there is both demand and opportunity for the augmentation of field observations, and synthesis of the observations that are collected, to ensure that science questions and integrated modeling activities are successfully implemented.

There was no significant difference in NK cell (285.58± 182.84 vs 528.92± 208.17) and B cell (186.66± 86.62 vs 334.17± 130.46). The proportion of naive CD4+ T cell (3.75± 0.50 vs 8.54± 1.61) in T lymphocyte subsets in patients with BPH was significantly reduced (P< 0.05). There was no significant difference in memory CD4+ T cell (87.9± 6.37 vs 92.63± 5.94), CD8+CD28+ T cell (60.52± 13.86 vs 64.32± 12.78), CD8+CDDR+ T cell (36.58± 12.87 vs 31.92± 8.54) and CD8+CD38+ T cell (2.1± 1.90 vs 2.55± 2.01).Conclusion: Immune dysfunction raised the risk of viral infection, inflammatory stimulation, and tumor induction in prostate cells, leading to hyperplasia, and immune non-response was potentially a key factor in the transformation of BPH into prostate cancer.Keywords: benign prostatic hyperplasia, lymphocyte subsets, pathogenesis, flow cytometry

Climate and terrestrial biosphere models consider nitrogen an important factor in limiting plant carbon uptake, while operational environmental models view nitrogen as the leading pollutant causing eutrophication in water bodies. The community Noah land surface model with multi-parameterization options (Noah-MP) is unique in that it is the next-generation land surface model for the Weather Research and Forecasting meteorological model and for the operational weather/climate models in the National Centers for Environmental Prediction. In this study, we add a capability to Noah-MP to simulate nitrogen dynamics by coupling the Fixation and Uptake of Nitrogen (FUN) plant model and the Soil and Water Assessment Tool (SWAT) soil nitrogen dynamics. This model development incorporates FUN's state-of-the-art concept of carbon cost theory and SWAT's strength in representing the impacts of agricultural management on the nitrogen cycle. Parameterizations for direct root and mycorrhizal-associated nitrogen uptake, leaf retranslocation, and symbiotic biological nitrogen fixation are employed from FUN, while parameterizations for nitrogen mineralization, nitrification, immobilization, volatilization, atmospheric deposition, and leaching are based on SWAT. The coupled model is then evaluated at the Kellogg Biological Station-a Long Term Ecological Research site within the US Corn Belt. Results show that the model performs well in capturing the major nitrogen state/flux variables (e.g., soil nitrate and nitrate leaching). Furthermore, the addition of nitrogen dynamics improves the modeling of net primary productivity and evapotranspiration. The model improvement is expected to advance the capability of Noah-MP to simultaneously predict weather and water quality in fully coupled Earth system models.

This study presents a new algorithm for parallel computation of river flow that builds on recent work demonstrating the relative independence of distant river reaches in the update step of the Muskingum method. The algorithm is designed to achieve enhanced fixed-size parallel speedup and uses a mathematical approximation applied at the boundaries of large subbasins. In order to use such an algorithm, a balanced domain decomposition method that differs from the traditional classifications of river reaches and subbasins and based on network topology is developed. An application of the algorithm and domain decomposition method to the Mississippi River Basin results in an eightfold decrease in computing time with 16 computing cores which is unprecedented for Muskingum-type algorithms applied in classic parallel-computing paradigms having a one-to-one relationship between cores and subbasins. An estimated 300 km between upstream and downstream reaches of subbasins guarantees the applicability of the algorithm in our study and motivates further investigation of domain decomposition methods.

The Project for the Intercomparison of Land-Surface Parameterization Schemes (PILPS) aims to improve understanding and modeling of land surface processes. PILPS phase 2(d) uses a set of meteorological and hydrological data spanning 18 yr (1966-83) from a grassland catchment at the Valdai water-balance research site in Russia. A suite of stand-alone simulations is performed by 21 land surface schemes (LSSs) to explore the LSSs' sensitivity to downward longwave radiative forcing, timescales of simulated hydrologic variability, and biases resulting from single-year simulations that use recursive spinup. These simulations are the first in PILPS to investigate the performance of LSSs at a site with a well-defined seasonal snow cover and frozen soil. Considerable model scatter for the control simulations exists. However, nearly all the LSS scatter in simulated root-zone soil moisture is contained within the spatial variability observed inside the catchment. In addition, all models show a considerable sensitivity to longwave forcing for the simulation of the snowpack, which during the spring melt affects runoff, meltwater infiltration, and subsequent evapotranspiration. A greater sensitivity of the ablation, compared to the accumulation, of the winter snowpaek to the choice of snow parameterization is found. Sensitivity simulations starting at prescribed conditions with no spinup demonstrate that the treatment of frozen soil (moisture) processes can affect the long-term variability of the models. The single-year recursive runs show large biases, compared to the corresponding year of the control run, that can persist through the entire year and underscore the importance of performing multiyear simulations.

Using atmospheric forcing data generated from a general circulation climate model, sixteen land surface schemes participating in the Project for the Intercomparison of Land-surface Parametrization Schemes (PILPS) were run off-line to equilibrium using forcing data from a GCM representative of a tropical forest and a mid-latitude grassland grid point. The values for each land surface parameter (roughness length, minimum stomatal resistance, soil depth etc.) were provided. Results were quality controlled and analyzed, focusing on the scatter simulated amongst the models. There were large differences in how the models' partitioned available energy between sensible and latent heat. Annually averaged, simulations for the tropical forest ranged by 79 W m-2 for the sensible heat flux and 80 W m-2 for the latent heat flux. For the grassland, simulations ranged by 34 W m-2 for the sensible heat flux and 27 W m-2 for the latent heat flux. Similarly large differences were found for simulated runoff and soil moisture and at the monthly time scale. The models' simulation of annually averaged effective radiative temperature varied with a range, between all the models, of 1.4 K for tropical forest and 2.2 K for the grassland. The simulation of latent and sensible heat fluxes by a standard 'bucket' models was anomalous although this could be corrected by an additional resistance term. These results imply that the current land surface models do not agree on the land surface climate when the atmospheric forcing and surface parameters are prescribed. The nature of the experimental design, it being offline and with artificial forcing, generally precludes judgements concerning the relative quality of any specific model. Although these results were produced de-coupled from a host model, they do cast doubt on the reliability of land surface schemes. It is therefore a priority to resolve the disparity in the simulations, understand the reasons behind the scatter and to determine whether this lack of agreement in decoupled tests is reproduced in coupled experiments.

Attempts to model surface-atmosphere interactions with greater physical realism have resulted in complex land surface schemes (LSS) with large numbers of parameters. A companion paper describes a multicriteria calibration procedure for extracting plot-scale estimates of the preferred ranges of these parameters from the various observational data sets that are now available. A complementary procedure is presented in this paper that provides an objective determination of the multicriteria sensitivity of the modeled variables to the parameters, thereby allowing the number of calibration parameters and hence the computational effort to be reduced. Two case studies are reported for the BATS model using data sets of typical quality but very different location and climatological regime (ARM-CART and Tucson). The sensitivity results were found to be consistent with the physical properties of the different environments, thereby supporting the reasonableness of the model formulation. Further, when the insensitive parameters are omitted from the calibration process, there is little degradation in the quality of the model description and little change in the preferred range of the remaining parameters. The potential for using the various observational data sets that are now available to extract plot-scale estimates for the parameters of a complex LSS via advanced parameter estimation methods developed for hydrological models is explored in this paper. Results are reported for two case studies using data sets of typical quality but very different location and climatological regime (ARM-CART and Tucson). The traditional single-criterion methods were found to be of limited value. However, a multicriteria approach was found to be effective in constraining the parameter estimates into physically plausible ranges when observations on at least one appropriate heat flux and one properly selected state variable are available. The water-balance components of 16 Soil-Vegetation Atmospheric Transfer (SVAT) schemes were evaluated by comparing predicted and observed streamflow, predicted evapotranspiration and evapotranspiration inferred from an atmospheric moisture budget analysis, and soil moisture storage changes for a seven-year period (1980-1986) using data from the Red-Arkansas River basins of the Southern Great Plains of the USA. The evaluations support the following suggestions: (a) The mean annual runoff of all models follows, at least generally, the strong climatic East-West gradient of precipitation, although most models predict too much runoff in the dry part of the basin. (b) The mean monthly storage change tends to be underestimated, even though all models capture reasonably well the seasonality of the evapotranspiration. (c) The wide range of conceptualizations used for generation of surface and subsurface runoff strongly affect runoff generation on seasonal, and shorter, time scales. Model responses to summer precipitation ranged from almost no summer runoff (one model) to the (more common) situation of persistent overprediction of summer runoff, especially in the driest part of the basin. (d) All models tended to underpredict evapotranspiration in summer and overpredict in winter. (e) Model-derived mean seasonal cycles of changes in soil moisture storage are qualitatively similar to those inferred from observations, but most models do not predict the decrease in April soil moisture storage and the increase in October that is inferred from observations.

The energy components of sixteen Soil-Vegetation Atmospheric Transfer (SVAT) schemes were analyzed and intercompared using 10 years of surface meteorological and radiative forcing data from the Red-Arkansas River basin in the Southern Great Plains of the United States. Comparisons of simulated surface energy fluxes among models showed that the net radiation and surface temperature generally had the best agreement among the schemes. On an average (annual and monthly) basis, the estimated latent heat fluxes agreed (to within approximate estimation errors) with the latent heat fluxes derived from a radiosonde-based atmospheric budget method for slightly more than half of the schemes. The sensible heat fluxes had larger differences among the schemes than did the latent heat fluxes, and the model-simulated ground heat fluxes had large variations among the schemes. The spatial patterns of the model-computed net radiation and surface temperature were generally similar among the schemes, and appear reasonable and consistent with observations of related variables, such as surface air temperature. The spatial mean patterns of latent and sensible heat fluxes were less similar than for net radiation, and the spatial patterns of the ground heat flux vary greatly among the 16 schemes. Generally, there is less similarity among the models in the temporal (interannual) variability of surface fluxes and temperature than there is in the mean fields, even for schemes with similar mean fields.

Sixteen land-surface schemes participating in the project for the Intercomparison of Land-surface Schemes (PILPS) Phase 2(c) were run using 10 years (1979-1988) of forcing data for the Red-Arkansas River basins in the Southern Great Plains region of the United States. Forcing data (precipitation, incoming radiation and surface meteorology) and land-surface characteristics (soil and vegetation parameters) were provided to each of the participating schemes. Two groups of runs are presented. (1) Calibration-validation runs, using data from six small catchments distributed across the modeling domain. These runs were designed to test the ability of the schemes to transfer information about model parameters to other catchments and to the computational grid boxes. (2) Base-runs, using data for 1979-1988, designed to evaluate the ability of the schemes to reproduce measured energy and water fluxes over multiple seasonal cycles across a climatically diverse, continental-scale basin. All schemes completed the base-runs but five schemes chose not to calibrate. Observational data (from 1980-1986) including daily river flows and monthly basin total evaporation estimated through an atmospheric budget analysis, were used to evaluate model performance. In general, the results are consistent with earlier PILPS experiments in terms of differences among models in predicted water and energy fluxes. The mean annual net radiation varied between 80 and 105 W m-2 (excluding one model). The mean annual Bowen ratio varied from 0.52 to 1.73 (also excluding one model) as compared to the data-estimated value of 0.92. The run-off ratios varied from a low of 0.02 to a high of 0.41, as compared to an observed value of 0.15. In general, those schemes that did not calibrate performed worse, not only on the validation catchments, but also at the scale of the entire modeling domain. This suggests that further PILPS experiments on the value of calibration need to be carried out.

Because of the large water storage capacity of soil relative to the atmosphere, changes in soil moisture storage can significantly affect the regional atmospheric budgets of water and energy on monthly, seasonal and longer time scales. Therefore proper modeling of soil liquid water processes is essential to a correct representation of the climate system. This study focuses on the class of summary models of liquid water fluxes in the vadose, or unsaturated zone of the soil, which are applicable to global or regional climate modeling studies. Fourteen such models are represented in this intercomparison study. Observational data from the HAPEX experiment provide validation. Because only limited observational data were available to constrain these models during their development and validation, the models have evolved very diverse treatments of the relevant processes: the basic Darcian (soil internal) and Hortonian (surface liquid flow) processes, as well as the boundary conditions of baseflow drainage and lateral interflow. The annual total local runoff is systematically underestimated by all but one of the participant models. This is one of the few significant biases between the consensus of participant models and the observations. The modeled runoff, averaged over the 14 models, differs from the budget estimate from observations by about 40%. During the period of runoff generation (late winter and early spring) the average model fails to deplete the soil water store as rapidly as is observed, a result consistent with the underprediction of runoff. One cannot rule out insufficient characterization of the field site soils as a primary cause of these discrepancies. Results suggest that model sources of the discrepancy are about equally likely to be related to the prediction of bare soil evaporation (discussed elsewhere in this issue) as they are to the parameterization of runoff and drainage processes.

Timosaponin B-II (TB-II), a major steroidal saponin constituent in Anemarrhena asphodeloides Bunge, exhibits various activities, including anti-inflammatory and hypoglycemic functions. However, the anti-DN effects and potential mechanism(s) of TB-II have not been previously reported.Purpose: To investigate the effect of TB-II on DN in alloxan-induced diabetic mice.Methods: TB-II was isolated and purified from A. asphodeloides Bunge using macroporous adsorption resin and preparative high-performance liquid chromatography. The effect of TB-II on DN was evaluated in alloxan-induced diabetic mice using an assay kit and immunohistochemical determination in vivo. The expression of mammalian target of rapamycin (mTOR), thioredoxin-interacting protein (TXNIP), and nuclear transcription factor-κB (NF-κB) signaling pathways was also measured using Western blot analysis.Results: TB-II significantly decreased the blood glucose levels and ameliorated renal histopathological injury in alloxan-induced diabetic mice. In addition, TB-II remarkably decreased the levels of renal function biochemical factors, such as kidney index, blood urea nitrogen, serum creatinine, urinary uric acid, urine creatinine, and urine protein, and it reduced lipid metabolism levels of total cholesterol and triglycerides and the levels of inflammatory cytokines interleukin-6 and tumor necrosis factor-α in alloxan-induced mice. Furthermore, TB-II inhibited the expression of mTOR, TXNIP, and NF-κB.Conclusion: The results revealed that TB-II plays an important role in DN via TXNIP, mTOR, and NF-κB signaling pathways. All samples were measured at different time intervals by high performance liquid chromatography.Results: The results showed that the effective permeability coefficient (Peff) and apparent permeability coefficient of ECH were 0.83×10-6–3.23×10-6 cm/s and 2.99×10-6–9.86×10-6 cm/s, respectively. Urbanization has accelerated dramatically across the world over the past decades. Urban influence on surface temperatures is now being considered as a correction term in climatological datasets. Although prior research has investigated urban influences on precipitation for specific cities or selected thunderstorm cases, a comprehensive examination of urban precipitation anomalies on a global scale remains limited. This research is a global analysis of urban precipitation anomalies for over one thousand cities worldwide. We find that more than 60% of the global cities and their downwind regions are receiving more precipitation than the surrounding rural areas. Moreover, the magnitude of these urban wet islands has nearly doubled in the past 20 y. Urban precipitation anomalies exhibit variations across different continents and climates, with cities in Africa, for example, exhibiting the largest urban annual and extreme precipitation anomalies. Cities are more prone to substantial urban precipitation anomalies under warm and humid climates compared to cold and dry climates. Cities with larger populations, pronounced urban heat island effects, and higher aerosol loads also show noticeable precipitation enhancements. This research maps global urban rainfall hotspots, establishing a foundation for the consideration of urban rainfall corrections in climatology datasets. We utilized city-scale simulations to quantitatively compare the diverse urban overheating mitigation strategies, specifically tied to social vulnerability and their cooling efficacies during heatwaves. We enhanced the Weather Research and Forecasting model to encompass the urban tree effect and calculate the Universal Thermal Climate Index for assessing thermal comfort. Taking Houston, Texas, and United States as an example, the study reveals that equitably mitigating urban overheat is achievable by considering the city's demographic composition and physical structure. The study results show that while urban trees may yield less cooling impact (0.27 K of Universal Thermal Climate Index in daytime) relative to cool roofs (0.30 K), the urban trees strategy can emerge as an effective approach for enhancing community resilience in heat stress-related outcomes. Social vulnerability-based heat mitigation was reviewed as vulnerability-weighted daily cumulative heat stress change. The results underscore: (i) importance of considering the community resilience when evaluating heat mitigation impact and (ii) the need to assess planting spaces for urban trees, rooftop areas, and neighborhood vulnerability when designing community-oriented urban overheating mitigation strategies., (© The Author(s) 2024. Background: As a genus within the Clavicipitaceae, Metarhizium exhibits rich morphological and ecological diversity, with a wide distribution and a variety of hosts. Currently, sixty-eight species of Metarhizium have been described., New Information: A new species of Metarhizium , M.puerense (Hong Yu bis), was described in Pu'er City, Yunnan Province, south-western China. Based on morphological characteristics and multilocus phylogenetic analyses, Metarhiziumpuerense was confirmed to be phylogenetically related to M.album , but was clearly separated and formed a distinct branch. In contrast, the host of Metarhiziumalbum was plants and leafhoppers and that lepidopteran larvae were the host of M.puerense . Heteromeric pore-forming proteins often contain recognition patterns or stereospecific selection filters. However, the construction of heteromeric pore-forming proteins for single-molecule sensing is challenging due to the uncontrollability of producing position isomers and difficulties in purification of regio-defined products. To overcome these preparation obstacles, we present an in situ strategy involving single-molecule chemical modification of a heptameric pore-forming protein to build a stereo- and regio-specific heteromeric nanopore (hetero-nanopore) with a subunit stoichiometric ratio of 3:4. The steric hindrance inherent in the homo-nanopore of K238C aerolysin directs the stereo- and regio-selective modification of maleimide derivatives. Our method utilizes real-time ionic current recording to facilitate controlled voltage manipulation for stoichiometric modification and position-based side-isomer removal. Single-molecule experiments and all-atom molecular dynamics simulations revealed that the hetero-nanopore features an asymmetric stereo- and regio-defined residue structure. The hetero-nanopore produced was characterized by mass spectrometry and single-particle cryogenic electron microscopy. In a proof-of-concept single-molecule sensing experiment, the hetero-nanopore exhibited 95% accuracy for label-free discrimination of four peptide stereoisomers with single-amino-acid structural and chiral differences in the mixtures. We introduce University of Texas - GLObal Building heights for Urban Studies (UT-GLOBUS), a dataset providing building heights and urban canopy parameters (UCPs) for more than 1200 city or locales worldwide. UT-GLOBUS combines open-source spaceborne altimetry (ICESat-2 and GEDI) and coarse-resolution urban canopy elevation data with a machine-learning model to estimate building-level information. Validation using LiDAR data from six U.S. cities showed UT-GLOBUS-derived building heights had a root mean squared error (RMSE) of 9.1 meters. Validation of mean building heights within 1-km 2 grid cells, including data from Hamburg and Sydney, resulted in an RMSE of 7.8 meters. Testing the UCPs in the urban Weather Research and Forecasting (WRF-Urban) model resulted in a significant improvement (55% in RMSE) in intra-urban air temperature representation compared to the existing table-based local climate zone approach in Houston, TX. Additionally, we demonstrated the dataset's utility for simulating heat mitigation strategies and building energy consumption using WRF-Urban, with test cases in Chicago, IL, and Austin, TX. Street-scale mean radiant temperature simulations using the SOlar and LongWave Environmental Irradiance Geometry (SOLWEIG) model, incorporating UT-GLOBUS and LiDAR-derived building heights, confirmed the dataset's effectiveness in modeling human thermal comfort in Baltimore, MD (daytime RMSE = 2.85°C). Objective: Bladder outlet obstruction (BOO) results in significant fibrosis in the chronic stage and elevated bladder pressure. Piezo1 is a type of mechanosensitive (MS) channel that directly responds to mechanical stimuli. To identify new targets for intervention in the treatment of BOO-induced fibrosis, this study investigated the impact of high hydrostatic pressure (HHP) on Piezo1 activity and the progression of bladder fibrosis., Methods: Immunofluorescence staining was conducted to assess the protein abundance of Piezo1 in fibroblasts from obstructed rat bladders. Bladder fibroblasts were cultured under normal atmospheric conditions (0 cmH 2 O) or exposed to HHP (50 cmH 2 O or 100 cmH 2 O). Agonists or inhibitors of Piezo1, YAP1, and ROCK1 were used to determine the underlying mechanism., Results: The Piezo1 protein levels in fibroblasts from the obstructed bladder exhibited an elevation compared to the control group. HHP significantly promoted the expression of various pro-fibrotic factors and induced proliferation of fibroblasts. Additionally, the protein expression levels of Piezo1, YAP1, ROCK1 were elevated, and calcium influx was increased as the pressure increased. These effects were attenuated by the Piezo1 inhibitor Dooku1. The Piezo1 activator Yoda1 induced the expression of pro-fibrotic factors and the proliferation of fibroblasts, and elevated the protein levels of YAP1 and ROCK1 under normal atmospheric conditions in vitro. Background: TBK1 positively regulates the growth factor-mediated mTOR signaling pathway by phosphorylating mTOR. However, it remains unclear how the TBK1-mTOR signaling pathway is regulated. Considering that STING not only interacts with TBK1 but also with MARCH1, we speculated that MARCH1 might regulate the mTOR signaling pathway by targeting TBK1. The aim of this study was to determine whether MARCH1 regulates the mTOR signaling pathway by targeting TBK1., Methods: The co-immunoprecipitation (Co-IP) assay was used to verify the interaction between MARCH1 with STING or TBK1. The ubiquitination of STING or TBK1 was analyzed using denatured co-immunoprecipitation. The level of proteins detected in the co-immunoprecipitation or denatured co-immunoprecipitation samples were determined by Western blotting. Stable knocked-down cells were constructed by infecting lentivirus bearing the related shRNA sequences. Scratch wound healing and clonogenic cell survival assays were used to detect the migration and proliferation of breast cancer cells., Results: We showed that MARCH1 played an important role in growth factor-induced the TBK1- mTOR signaling pathway. MARCH1 overexpression attenuated the growth factor-induced activation of mTOR signaling pathway, whereas its deficiency resulted in the opposite effect. Mechanistically, MARCH1 interacted with and promoted the K63-linked ubiquitination of TBK1. This ubiquitination of TBK1 then attenuated its interaction with mTOR, thereby inhibiting the growth factor-induced mTOR signaling pathway. Importantly, faster proliferation induced by MARCH1 deficiency was weakened by mTOR, STING, or TBK1 inhibition., Conclusion: MARCH1 suppressed growth factors mediated the mTOR signaling pathway by targeting the STING-TBK1-mTOR axis., (© 2024. The global diversity of fungi has been estimated between 2 to 11 million species, of which only about 155 000 have been named. Most fungi are invisible to the unaided eye, but they represent a major component of biodiversity on our planet, and play essential ecological roles, supporting life as we know it. Although approximately 20 000 fungal genera are presently recognised, the ecology of most remains undetermined. Despite all this diversity, the mycological community actively researches some fungal genera more commonly than others. This poses an interesting question: why have some fungal genera impacted mycology and related fields more than others? To address this issue, we conducted a bibliometric analysis to identify the top 100 most cited fungal genera. A thorough database search of the Web of Science, Google Scholar, and PubMed was performed to establish which genera are most cited. The most cited 10 genera are Saccharomyces , Candida , Aspergillus , Fusarium , Penicillium , Trichoderma , Botrytis , Pichia , Cryptococcus and Alternaria . Case studies are presented for the 100 most cited genera with general background, notes on their ecology and economic significance and important research advances. This paper provides a historic overview of scientific research of these genera and the prospect for further research. Aim: Hyperhomocysteine has been recognized as an independent risk factor of multiple diseases, including several eye diseases. In this study, we aim to investigate whether increased homocysteine (Hcy) is related to cataracts, and to explore whether dysregulation of mTOR-mediated autophagy and connexin expression are underlying mechanisms., Method: We first developed a method of liquid chromatography tandem mass spectrometry to accurately measure serum concentrations of Hcy in 287 cataract patients and 334 healthy controls. Next, we treated human lens epithelial (HLC-B3) cells with Hcy at different concentrations and durations, and then analyzed expression of autophagy-related markers and connexins, as well as phosphorylated mTOR (p-mTOR) in these cells by Western blotting. Formation of autophagic vacuoles and intracellular Ca 2+ in the Hcy-treated cells were observed by fluorescence microscopy. Further, we performed a rescue experiment in the Hcy-treated HLC-B3 cells by pre-incubation with rapamycin, an mTOR inhibitor., Results: The serum levels of Hcy in patients with cataracts were significantly increased compared to those in healthy controls. In cultured HLC-B3 cells, expression of autophagy related markers (LC3B and Beclin1) and connexins (Cx43 and Cx50) was inhibited by Hcy treatment in a dose- and duration-dependent manner. Accumulation of Ca 2+ in the Hcy-treated lens epithelial cells was observed as a consequence of reduced connexin expression. Meanwhile, expression of p-mTOR increased, representing up-regulation of the mTOR pathway. Soil fungi are pivotal in alpine and arctic ecosystems that are vulnerable to climate changes. Previous studies have shown broad connections between soil fungi in the arctic and alpine regions, but most of these studies are mainly from Europe and North America, with more sporadic studies from East Asia. Currently, little is known about the biogeographic relationships between soil fungi in alpine meadows of southwestern China (AMSC) and other regions of the world. In addition, the regional-scale spatial patterns of fungal communities in the AMSC, as well as their driving factors and ecological processes, are also poorly understood. In this study, we collected roots and surrounding soils of two dominant ectomycorrhizal plants, Bistorta vivipara and B. macrophylla from the AMSC, and performed bioinformatic and statistical analyses based on high-throughput sequencing of ITS2 amplicons. We found that: (1) fungi from the AMSC were closely related with those from boreal forests and tundra, and saprotrophic fungi had higher dispersal potential than ectomycorrhizal fungi; (2) community compositions exhibited clear divergences among geographic regions and between root and soil samples; (3) climate was the predominant factor driving regional-scale spatial patterns but had less explanatory power for saprotrophic and total fungi from roots than those from soils; (4) homogeneous selection and drift were the key ecological processes governing community assembly, but in communities of saprotrophic and total fungi from soil samples, drift contributed less and its role was partially replaced by dispersal limitation. This study aims to report three new species of Conoideocrella and Moelleriella from Yunnan Province, Southwestern China. Species of Conoideocrella and Moelleriella parasitize scale insects (Coccidae and Lecaniidae, Hemiptera) and whiteflies (Aleyrodidae, Hemiptera). Based on the phylogenetic analyses of the three-gene nrLSU, tef-1α , and rpb1 , it showed one new record species ( Conoideocrella tenuis ) and one new species ( Conoideocrella fenshuilingensis sp. nov.) in the genus Conoideocrella , and two new species, i.e., Moelleriella longzhuensis sp. nov. and Moelleriella jinuoana sp. nov. in the genus Moelleriella . The three new species were each clustered into separate clades that distinguished themselves from one another. All of them were distinguishable from their allied species based on their morphology. Morphological descriptions, illustrations, and comparisons of the allied taxa of the four species are provided in the present paper. In addition, calculations of intraspecific and interspecific genetic distances were performed for Moelleriella and Conoideocrella .

Main Conclusion: The resurrection plant Boea hygrometrica selectively recruits and assembles drought-specific microbial communities across the plant-soil compartments, which may benefit plant growth and fitness under extreme drought conditions. Plant-associated microbes are essential for facilitating plant growth and fitness under drought stress. The resurrection plant Boea hygrometrica in natural habitats with seasonal rainfall can survive rapid desiccation, yet their interaction with microbiomes under drought conditions remains unexplored. This study examined the bacterial and fungal microbiome structure and drought response across plant-soil compartments of B. hygrometrica by high-throughput amplicon sequencing of 16S rRNA gene and internal transcribed spacer. Our results demonstrated that the diversity, composition, and functional profile of the microbial community varied considerably across the plant-soil compartments and were strongly affected by drought stress. Bacterial and fungal diversity was significantly reduced from soil to endosphere and belowground to aboveground compartments. The compartment-specific enrichment of the dominant bacteria phylum Cyanobacteriota and genus Methylorubrum in leaf endosphere, genera Pseudonocardia in rhizosphere soil and Actinoplanes in root endosphere, and fungal phylum Ascomycota in the aboveground compartments and genera Knufia in root endosphere and Cladosporium in leaf endosphere composed part of the core microbiota with corresponding enrichment of beneficial functions for plant growth and fitness. Moreover, the recruitment of dominant microbial genera Sphingosinicella and Plectosphaerella, Ceratobasidiaceae mycorrhizal fungi, and numerous plant growth-promoting bacteria involving nutrient supply and auxin regulation was observed in desiccated B. hygrometrica plants. Our results suggest that the stable assembled drought-specific microbial community of B. hygrometrica may contribute to plant survival under extreme environments and provide valuable microbial resources for the microbe-mediated drought tolerance enhancement in crops., (© 2024. Background: The carcinogenic transcription factor c-Myc is the most aggressive oncogene, which drive malignant transformation and dissemination of triple-negative breast cancer (TNBC). Recruitment of many cofactors, especially WDR5, a protein that nucleates H3K4me chromatin modifying complexes, play a pivotal role in regulating c-Myc-dependent gene transcription, a critical process for c-Myc signaling to function in a variety of biological and pathological contexts. For this reason, interrupting the interaction between c-Myc and the transcription cofactor WDR5 may become the most promising new strategy for treating c-Myc driven TNBC., Methods: Immunoprecipitation and mass spectrometry (IP-MS) is used to screen proteins that bind c-Myc/WDR5 interactions. The interaction of METTL3 with c-Myc/WDR5 in breast cancer tissues and TNBC cells was detected by Co-IP and immunofluorescence. Subsequently, we further analyzed the influence of METTL3 expression on c-Myc/WDR5 protein expression and its interaction stability by Western blot and Co-IP. The correlation between METTL3 and c-Myc pathway was analyzed by ChIP-seq sequencing and METTL3 knockdown transcriptome data. The effect of METTL3 expression on c-Myc transcriptional activity was detected by ChIP-qPCR and Dual Luciferase Reporter. At the same time, the overexpression vector METTL3-MUT (m6A) was constructed, which mutated the methyltransferase active site (Aa395-398, DPPW/APPA), and further explored whether the interaction between METTL3 and c-Myc/WDR5 was independent of methyltransferase activity. In addition, we also detected the changes of METTL3 expression on TNBC's sensitivity to small molecule inhibitors such as JQ1 and OICR9429 by CCK8, Transwell and clonal formation assays. Finally, we further verified our conclusions in spontaneous tumor formation mouse MMTV-PyMT and nude mouse orthotopic transplantation tumor models., Results: METTL3 was found to bind mainly to c-Myc/WDR5 protein in the nucleus. It enhances the stability of c-Myc/WDR5 interaction through its methyltransferase independent mechanism, thereby enhancing the transcriptional activity of c-Myc on downstream glucose metabolism genes. Notably, the study also confirmed that METTL3 can directly participate in the transcription of glucose metabolism genes as a transcription factor, and knockdown METTL3 enhances the drug sensitivity of breast cancer cells to small molecule inhibitors JQ1 and OICR9429. The study was further confirmed by spontaneous tumor formation mouse MMTV-PyMT and nude mouse orthotopic transplantation tumor models., Conclusion: METTL3 binds to the c-Myc/WDR5 protein complex and promotes glycolysis, which plays a powerful role in promoting TNBC progression. The aim of this study was to investigate the effects of solid-state fermented cottonseed meal (FCSM) inclusion levels on the growth performance, serum biochemical parameters and hepatic lipid metabolism in geese from 28 to 70 d of age. A total of 288 twenty-eight-d-old male geese were randomly divided into 4 treatments with FCSM levels of 0, 5, 15 and 25% including 0, 22.74, 67.33, 111.27 mg FG/kg diet, respectively. Each treatment contained 6 replicates and 12 birds per replicate. Treatments of FCSM inclusions from 0 to 25% had no effect on growth rate and feed intake in geese during d 28 to 70. The F/G ratio was increased (P < 0.05) in geese fed the diet with 25% FCSM compared with birds fed the diet with 0% FCSM. Treatment with 25% FCSM levels had no effect on the contents of TC, TG, HDL-C, LDL-C, but increased (P < 0.05) AST and ALT activities in serum of geese at d 70. Treatment with 25% FCSM increased the contents of FG, HDL-C, TC, C18:2n6, C20:4n6 and PUFA and decreased (P < 0.05) the contents of NEFA, SFA, MUFA in liver compared with treatment of 0% FCSM inclusion. Additionally, treatment with 25% FCSM decreased (P < 0.05) the PPARα, AMPK, and LXR mRNA expression related to lipid deposition, and increased (P < 0.05) PPARγ and ACC mRNA expression related to lipolysis in liver compared with birds fed the diet with 0% FCSM. Overall, treatment with 0 to 15% FCSM (<=67.33 mg FG/kg diet) had no adverse effects on the growth performance and lipid metabolism of geese. However, treatment fed 25% FCSM (111.27 mg FG/kg diet) decreased feed efficiency and promoted hepatic lipid deposition associated with the alteration of related gene expression in geese at 28 to 70 d of age., (Copyright © 2024. The pursuit of high-performance electronic devices has driven the research focus toward 2D semiconductors with high electron mobility and suitable band gaps. Previous studies have demonstrated that quasi-2D Bi 2 O 2 Se (BOSe) has remarkable physical properties and is a promising candidate for further exploration. Building upon this foundation, the present work introduces a novel concept for achieving nonvolatile and reversible control of BOSe's electronic properties. The approach involves the epitaxial integration of a ferroelectric PbZr 0.2 Ti 0.8 O 3 (PZT) layer to modify BOSe's band alignment. Within the BOSe/PZT heteroepitaxy, through two opposite ferroelectric polarization states of the PZT layer, we can tune the Fermi level in the BOSe layer. Consequently, this controlled modulation of the electronic structure provides a pathway to manipulate the electrical properties of the BOSe layer and the corresponding devices.

General anesthesia is widely applied in clinical practice. However, the precise mechanism of loss of consciousness induced by general anesthetics remains unknown. Here, we measured the dynamics of five neurotransmitters, including γ-aminobutyric acid, glutamate, norepinephrine, acetylcholine, and dopamine, in the medial prefrontal cortex and primary visual cortex of C57BL/6 mice through in vivo fiber photometry and genetically encoded neurotransmitter sensors under anesthesia to reveal the mechanism of general anesthesia from a neurotransmitter perspective. Results revealed that the concentrations of γ-aminobutyric acid, glutamate, norepinephrine, and acetylcholine increased in the cortex during propofol-induced loss of consciousness. Dopamine levels did not change following the hypnotic dose of propofol but increased significantly following surgical doses of propofol anesthesia. Notably, the concentrations of the five neurotransmitters generally decreased during sevoflurane-induced loss of consciousness. Furthermore, the neurotransmitter dynamic networks were not synchronized in the non-anesthesia groups but were highly synchronized in the anesthetic groups. These findings suggest that neurotransmitter dynamic network synchronization may cause anesthetic-induced loss of consciousness.

We present an encoding scheme of a single logical qubit with single-sided quantum dot (QD)-cavity systems, which is immune to the collective decoherence. By adjusting the Purcell factor to satisfy the balanced reflection condition, the detrimental effects of unbalanced reflection between the coupled and uncoupled QD-cavity systems can be effectively suppressed. Furthermore, the fidelity of each step can be increased to unity regardless of the strong coupling regime and the weak coupling regime of cavity quantum electrodynamics (QED) with the assistance of waveform correctors. The scheme requires QD-cavity systems and simple linear optical elements, which can be implemented with the currently experimental techniques.
