Objective: To construct models of high-risk human papillomavirus (HPV) infection with precancerous lesions or cervical cancer and explore the immune function. Methods: Using CRISPR/Cas9, the expression vector HPV16-E6-E7-Rosa26 was microinjected into fertilized eggs of C57BL/6 N mice using homologous recombination, and the F0 generation was obtained for reproduction. Then, the formation of precancerous lesions was promoted via intramuscular injection of estradiol. Presence of precancerous cervical-vaginal intraepithelial lesions, Ki67 and p16 expression levels, and CD8+ T cell proportions in the spleen were evaluated. Results: Two F0 generation mice exhibited correct the homologous recombination. Seven positive mice were identified in the F1 generation. After breeding and mating, 25 homozygous and 11 heterozygous HPV16-E6-E7-engineered mice were obtained from the F2 generation. After estradiol benzoate treatment, the cervical-vaginal epithelium appeared as precancerous lesions with positive Ki67 and p16 expression. The percentage of CD8+ T cells decreased. Conclusion: HPV16-E6-E7-Rosa26 induced low immune function in mice, and provides a good model for the basic research of the mechanisms of action of HPV infection-associated precancerous lesions or cervical cancer.

A core-shell-satellite nanostructure surface-enhanced Raman scattering (SERS) substrate based on magnetic nanoparticles, metal-organic frameworks (MOFs) and gold nanoparticles (Ag NPs) was established and applied in the simple, rapid and sensitive detection of the organophosphorus pesticide triazophos in agricultural products. MIL-100(Fe)-coated Fe3O4 was prepared by low-temperature cyclic self-assembly method, and Ag NPs were in-situ grown on the surface of MIL-100(Fe)-coated Fe3O4 by silver mirror cycling. The SERS sensing of triazophos residues in apples was carried out. The logarithmic value (Y) of the characteristic peak intensity of triazophos was linearly related to the logarithmic value (X) of its concentration in the range of 0.05–10 mg/L, which was described by the following equation: Y = 0.573 8X + 2.804 (R2 = 0.980). The limit of detection (LOD) was as low as 11.9 mg/L. The recoveries of the developed method ranged from 90.07% to 103.27% at spiked concentrations of 2, 5, and 10 mg/L. Therefore, the prepared SERS substrate has great potential for application in the detection of pesticide residues in foods.

In this article, gradient-distributed VCp-ZTAp/Fe45 composites were prepared by vacuum sintering, and three-body abrasive wear experiments were carried out to investigate its anti-wear performance. The composite casting of VCp-ZTAp/Fe45 was investigated by using the finite element method and experiments, and the bonding between the Fe45 substrate and 35SiMnCrMoNi steel was fully investigated. Results show the in situ formation of VCp in the VCp-ZTAp/Fe45 composite matrix during vacuum sintering. The anti-wear property of the VCp-ZTAp/Fe45 composite is ca. 7 times that of Hardox450 and NM450 and ca. 8 times of 30SiMn. There forms a solid metallurgical bonding between the Fe45 matrix of VCp-ZTAp/Fe45 and 35SiMnCrMoNi steel, with a ca. 80 μm thickness of the bonding area during the composite casting.

Objective To investigate the efficacy and safety of robot-assisted laparoscopic antegrade inguinal lymph node dissection for patients with penile cancer. Methods A retrospective study was performed on clinical data from 10 patients with penile cancer admitted to the First Affiliated Hospital of Army Medical University from March 2019 to January 2020. All of them were diagnosed with penis squamous cell carcinoma by pathological diagnosis. Robot-assisted laparoscopic antegrade inguinal lymphadenectomy was performed for all patients under general anesthesia. Results The surgery was successfully accomplished at totally 20 sides without intraoperative conversion to open surgery. The operation time was 87.90±11.58 (80~119) min, intraoperative blood loss was 12.50±8.90 (5~25) mL, and number of removed lymph nodes was 22.90±8.90 (10~35). The positive lymph nodes were 6 in 1 case at T2 stage, 3 in 1 case at T2 stage, 3 in 1 case at T1 stage and 2 in 1 case at T1 stage. The postoperative hospital stay was 2.80±1.23 (2~5) d, and the indwelling time of drainage tube was 17.10±3.63 (7~27) d. None of the patients experienced skin necrosis, bleeding, lymphatic fistula, infection and other complications. During the follow-up time of 16.40±6.79 (14~24) months, recurrence and metastasis were found in only 1 case, and not in the other 9 cases. Conclusion Robot-assisted laparoscopic antegrade bilateral inguinal lymphadenectomy is a safe and effective dissection for penile cancer, which can reduce complications and promote rapid recovery after surgery.

The physicochemical properties of the mixed gelatin gels with soy and whey proteins were investigated to develop the gel base with a soft texture and abundant essential amino acids for the elderly. Gelatin-only gel (control) was prepared at 6% ( w / v ), and mixed gelatin gels were formulated by replacing gelatin with soy protein isolate and whey protein concentrate at different mixing ratios [gelatin (G):soy protein isolate (S):whey protein concentrate (W)]. Results showed that replacing gelatin with the globular proteins in gelatin gels increased the pH value and processing yield ( p < 0.05). Moreover, the mixed gelatin gels, particularly the G2:S1:W3 treatment, showed significantly higher essential amino acids than the gelatin-only control. The partial replacement of gelatin with the globular proteins could decrease the hardness of gelatin gel ( p < 0.05), but there was no difference in hardness between the G2:G3:W1, G2:S2:W2, and G2:S1:W3 treatments ( p > 0.05). The results of protein pattern, x-ray diffraction, and microstructure had no clear evidence for specific protein-protein interaction in the mixed gelatin gels. Therefore, this study indicates that mixed gelatin gels with the globular proteins at specific mixing ratios could be a practical approach to providing a soft texture and high-level essential amino acids to the elderly.

This study aimed to isolate the proteolytic fraction from the silkworm thorn fruit ( Cudrania tricuspidata ) through ethanol precipitation at different ratios, and to determine its proteolytic activity and optimal activity conditions. Furthermore, the hydrolysis characteristics and antioxidant activity of soy protein isolate (SPI) and whey protein concentrate (WPC) hydrolyzates obtained through the enzymatic hydrolysis of freeze-dried silkworm thorn fruit powder (SF) were evaluated. For isolation and partial purification of proteolytic fraction, the water-solubilized fraction of the silkworm thorn fruit was purified through ethanol precipitation at four different ratios of 1:1, 1:2, 1:4, and 1:6 ( v/v ). The protein recovery rate, caseinolytic activity, protein pattern, and optimal activity (pH, temperature, and inhibitors) of fractional ethanol precipitate obtained from the silkworm thorn fruit (ESF) were evaluated. The proteolytic fraction obtained from silkworm thorn fruit exhibited a major protein band around 65-70 kDa and showed the highest proteolytic activity at a 1:4 ratio of ethanol precipitation ( p < 0.05). The optimal activity of the measured enzyme fraction was determined to be at pH 9.0 and 50 °C, and the proteolytic activity of ESF was almost inhibited by phenyl methyl sulphonyl fluoride (PMSF, 2 mM), a serine protease inhibitor. Compared to Alcalase and papain, extensively used as commercial enzymes, the silkworm thorn fruit powder was less effective in hydrolyzing SPI and WPC. Nevertheless, SPI and WPC hydrolyzates mediated with silkworm thorn fruit powder showed even better antioxidant activities than those mediated with Alcalase and papain. Thus, our results show the potential application of silkworm thorn fruit as a novel source of plant protease for producing human-grade protein hydrolyzates.

This study identified the aroma profile of salmon by-product for high utilization of by-products, including hydrolysates of head, frame, and skin were treated with reducing sugars and thermal processing. Electronic nose (E-nose) and gas chromatography-mass spectrometry (GC-MS) coupled with gas chromatography-olfactometry (GC-O) were used to analyzed the aroma profile. A total of 140 and 90 volatile compounds were detected through E-nose and GC-MS respectively, and the main volatile compounds were aldehydes. A total of 23 odor active compounds were recognized using GC-O, and 3-methyl-butanal, heptanal, benzaldehyde, octanal, furfural, and methoxy-phenyl-oxime were identified as the aroma of salmon. Using multivariate analysis, the pattern between the pretreated samples and aroma profiles was confirmed, and there were clear separations among the samples. Sous-vide is a cooking method used to improve the tenderness and juiciness of chicken breast. However, the comparative changes in meat quality attributes of sous-vide cooked chicken breast and thigh muscles are not fully understood. The objective of this study was to investigate the effects of sous-vide cooking conditions, based on collagen denaturation temperature of intramuscular connective tissue, on the physicochemical properties of chicken breasts and thighs. Chicken breast and thigh were cooked at four sous-vide cooking conditions (55 °C for 3/6 h and 65 °C for 3/6 h) and conventional cooking at 75 °C (core temperature of 71 °C) as control. No significant differences in pH and lightness were found between the sous-vide cooking conditions. Moisture content, cooking loss, protein solubility, shear force, myofibrillar fragmentation index, and lipid oxidation were affected by sous-vide cooking conditions ( p < 0.05). The decreased shear force and total collagen content of 65 °C sous-vide cooking treatment might be associated with collagen denaturation ( p < 0.05). Sous-vide cooking at 55 °C could decrease cooking loss, with higher moisture than sous-vide cooking at 65 °C ( p < 0.05). These tendencies on water-holding capacity and shear force at the two different temperatures were similarly observed for both chicken breast and thigh. Therefore, this study indicates that chicken breast and thigh are similarly affected by the sous-vide cooking conditions and suggests that a novel strategy to apply together two temperature ranges based on the thermal denaturation of intramuscular connective tissue would be required.

The objective of this study was to evaluate the general quality attributes and shelf stability of reduced-salt and sous-vide cooked chicken breast hams during 4 weeks of refrigerated storage (4°C). Four treatment groups of chicken breast ham were prepared using a 2 (salt level, 1.5% NaCl (regular) and 0.75% (reduced)) × 2 (cooking method, conventional and sous-vide) factorial arrangement. Based on each chicken breast weight, 20% NaCl solution was injected. Conventional cooking was done at 80°C until the core temperature reached 71°C, whereas sous-vide cooking was conducted at 60°C for 2 h. Sous-vide cooking could decrease cooking loss and shear force of reduced-salt chicken breast ham (P < 0.05). As a result, sensory scores for juiciness and tenderness of reduced-salt and sous-vide cooked chicken breast ham were similar to those of regular-salt and conventionally cooked chicken breast hams (P > 0.05). No adverse impacts on lipid oxidation and microbial safety were found in reduced-salt and sous-vide cooked chicken breast ham during 4 wk of refrigerated storage. Therefore, this study suggests that sous-vide cooking could be a practical thermal process for improving the water-holding capacity and texture of chicken breast ham without adverse impacts on shelf stability. Further studies on the combined application of sous-vide cooking with salt replacers would be warranted to improve the sensorial acceptance of saltiness of sous-vide cooked low-salt meat products., (Copyright © 2022 The Authors. The practical use of Korean native black goat skin as a source of gelatin extraction is limited. The objective of this study was to optimize the extraction temperature and time of gelatin from Korean native black goat skin, and to compare the quality characteristics of goat skin gelatin and other commercial gelatin products. Response surface methodology was applied to optimize the extraction temperature and time of gelatin obtained from native Korean black goat skin. The effects of temperature (50°C-70°C) and time (2-4 h) on extraction yield and gel strength were investigated using a face-centered central composite design with 13 experiments. Gelatin extraction from Korean native black goat skin was prepared through the serial processes of alkali pre-treatment, bleaching, neutralization, hot-water extraction, and freeze-drying. Using the optimization plot of Minitab software, the optimized conditions for extracting temperature and time of goat skin gelatin were 59.49°C and 3.03 h, and the optimized values of extraction yield and gel strength were 12.52% and 263.37 g, respectively. Based on a quality comparison of goat skin gelatin with commercial gelatin, the pH value of gelatin extracted from Korean native black goat skin was 5.57. The color of gelatin extracted from Korean native black goat skin was darker than that of commercial gelatin (p<0.05). Higher emulsifying properties and gel strength of goat skin gelatin were observed when compared to those of commercial gelatin (p<0.05). Image registration is a fundamental and crucial step in medical image analysis. However, due to the differences between mono-mode and multi-mode registration tasks and the complexity of the corresponding relationship between multimode image intensity, the existing unsupervised methods based on deep learning can hardly achieve the two registration tasks simultaneously. In this paper, we proposed a novel approach to register both mono- and multi-mode images $\color{blue}{\text{in a differentiable }}$. By approximately calculating the mutual information in a $\color{blue}{\text{differentiable}}$ form and combining it with CNN, the deformation field can be predicted quickly and accurately without any prior information about the image intensity relationship. The registration process is implemented in an unsupervised manner, avoiding the need for the ground truth of the deformation field. We utilize two public datasets to evaluate the performance of the algorithm for mono-mode and multi-mode image registration, which confirms the effectiveness and feasibility of our method. In addition, the experiments on patient data also demonstrate the practicability and robustness of the proposed method.

Next-generation flow (NGF) has detected minimal residual disease (MRD) in numerous myeloma patients who achieve a complete response (CR). However, when MRD is not detected via NGF in non-CR patients, its clinical meaning is uncertain. Here, we investigated the correlation between MRD findings on NGF and the response criteria, paying special attention to patients with discrepant results. We performed NGS analysis of IgH rearrangements on bone marrow samples from the non-CR patients with negative MRD on NGF. Atmospheric samples were collected using polyurethane foam (PUF) passive air sampling device for every 3 months from June 2012 to May 2013 in Shanghai rural regions in order to investigate the concentrations, profiles, spatial distributions, and seasonal variations of polybrominated diphenyl ethers (PBDEs). Twelve PBDE congeners (BDE-17, BDE-28, BDE-47, BDE-49, BDE-66, BDE-85, BDE-99, BDE-100, BDE-138, BDE-153, BDE-154, and BDE-183) were measured and analyzed by GC-MS. The results showed that detectable PBDEs were examined in all air samples, which indicated that these pollutants are widespread in the research areas. The ∑ 12 PBDE concentrations in Shanghai rural air ranged from 4.49 to 77.5 pg m -3 , with mean value up to 26.7 pg m -3 . The highest concentration was found at Jinshan sampling site in summer (from June to August in 2012). Furthermore, among the PBDE compounds investigated, the most frequently detected and the major congeners were BDE-17, BDE-28, BDE-47, and BDE-99. And the lower brominated diphenyl ethers (accounting for 75.0%) were the majority of the PBDE congeners. Finally, the result of principal component analysis (PCA) revealed that the lower and higher brominated diphenyl ethers in Shanghai rural regions were emitted from different pollutant sources.
