Abstract

The cell cycle transcriptional program imposes order on events of the cell-cycle and is a target for signals that regulate cell-cycle progression, including checkpoints required to maintain genome integrity. Neither the mechanism nor functional significance of checkpoint regulation of the cell-cycle transcription program are established. We show that Nrm1, an MBF-specific transcriptional repressor acting at the transition from G1 to S phase of the cell cycle, is at the nexus between the cell cycle transcriptional program and the DNA replication checkpoint in fission yeast. Phosphorylation of Nrml by the Cds1 (Chk2) checkpoint protein kinase, which is activated in response to DNA replication stress, promotes its dissociation from the MBF transcription factor. Abstract

Previous investigations into the mechanisms that control RNA Polymerase (Pol) I transcription have primarily focused on the process of transcription initiation, thus little is known regarding postinitiation steps in the transcription cycle. Spt4p and Spt5p are conserved throughout eukaryotes, and they affect elongation by Pol II. We have found that these two proteins copurify with Pol I and associate with the rDNA in vivo. Disruption of the gene for Spt4p resulted in a modest decrease in growth and rRNA synthesis rates at the permissive temperature, 30[degrees]C. Furthermore, biochemical and EM analyses showed clear defects in rRNA processing. These data suggest that Spt4p, Spt5p, and, potentially, other regulators of Pol I transcription elongation play important roles in coupling rRNA transcription to its processing and ribosome assembly. yeast Saccharomyces cerevisiae

Abstract

Pancreatic β cell physiology changes substantially throughout life, yet the mechanisms that drive these changes are poorly understood. Here, we performed comprehensive in vivo quantitative proteomic profiling of pancreatic islets from juvenile and 1-year-old mice. The analysis revealed striking differences in abundance of enzymes controlling glucose metabolism. We show that these changes in protein abundance are associated with higher activities of glucose metabolic enzymes involved in coupling factor generation as well as increased activity of the coupling factor-dependent amplifying pathway of insulin secretion. Nutrient tracing and targeted metabolomics demonstrated accelerated accumulation of glucose-derived metabolites and coupling factors in islets from 1-year-old mice, indicating that age-related changes in glucose metabolism contribute to improved glucose-stimulated insulin secretion with age. Together, our study provides an in-depth characterization of age-related changes in the islet proteome and establishes metabolic rewiring as an important mechanism for age-associated changes in β cell function.

28. Fatigue crack initiation in AA2024: A coupled micromechanical testing and crystal plasticity study

Abstract

A new combined experimental and modelling approach has been developed in order to understand the physical mechanisms that lead to crack nucleation in a polycrystalline aluminium alloy AA2024 undergoing cyclic loading. Four-point bending low-cycle fatigue tests were performed inside the chamber of a scanning electron microscope on specimens with a through-thickness central hole, introduced to localize stresses and strains in a small region on the top surface of the sample. Fatigue crack initiation and small crack growth mechanisms were analyzed through high-resolution scanning electron microscope images, local orientation measurements using electron-back-scattered-diffraction, and local strain measurements using digital image correlation. A crystal plasticity finite element model was developed to simulate the cyclic deformation behaviour of AA2024. Two-dimensional Voronoi-based microstructures were generated, and the material parameters for the constitutive equations (including both isotropic and kinematic hardening) were identified using monotonic and fully reversed cyclic tests. A commonly used fatigue crack initiation criterion found in the literature, the maximum accumulated plastic slip, was evaluated in the crystal plasticity finite element model but could not predict the formation of cracks away from the edge of the hole in the deformed specimens. A new criterion combining 2 parameters: The maximum accumulated slip over each individual (critical) slip system and the maximum accumulated slip over all slip systems were formulated to reproduce the experimental locations of crack nucleation in the microstructure.

Abstract

The oxygen sites in the tellurite glasses show no spectroscopic distinction, even when comparing bridging and non-bridging sites. It is achieved by high field (super 17)O solid-state nuclear magnetic resonance (NMR) and by ab initio calculations of both the NMR and the oxygen 1s photoelectron spectra.
