9. MoS2 Quantum Dots Alter Macrophage Plasticity and Induce Mitophagy to Attenuate Endothelial Barrier Dysfunctions.

Abstract

Molybdenum disulfide (MoS2), as a member of transition metal dichalcogenides (TMDs), presents enormous potential in biomedical applications due to its unique chemical–physical properties, photo-electronic properties, and biocompatibility. MoS2 nanostructures have recently been shown to be multifunctional nanotheranostic agents for neuroprotection and maintenance of endothelial homeostasis. However, whether ultrasmall MoS2 quantum dots (QD) participate in preventing the impairment of the endothelial barrier elicited by pro-inflammatory stimuli and the associated underlying mechanisms remain elusive yet. Herein, we set out to explore the protective effects of MoS2 QD on inflammatory endothelial barrier dysfunction and macrophage plasticity. We revealed an inhibitory effect of MoS2 QD on endothelial barrier impairment and monocyte–endothelial cell interactions, including monocyte adhesion and transmigration, and this might be associated with a reduced activation of focal adhesion kinase (FAK). Importantly, using an in vitro macrophage polarization system, we revealed that MoS2 QD altered macrophage plasticity via inhibiting M1-macrophage polarization, and the reduction in nuclear factor-κB (NF-κB) activation and promotion of mitophagy by MoS2 QD was involved in preventing M1 macrophage polarization. Taken together, these findings suggested potential therapeutic applications of MoS2-based bioactive nanoagents to attenuate inflammatory endothelial barrier dysfunctions that are provoked by macrophage-mediated endothelial activation. 10. Unsaturated phospholipid modified FeOCl nanosheets for enhancing tumor ferroptosis.

Abstract

Iron-dependent accumulation of reactive oxygen species (ROS) and lipid peroxidation play key roles in ferroptosis, which has been an attractive strategy to kill tumor cells. However, the rapid annihilation of hydroxyl radicals (˙OH) produced from the Fenton reaction has become a major obstacle in inducing lipid peroxidation in cells. In this study, we develop a nano-delivery system of unsaturated phospholipid (Lip) and polyacrylic acid (PAA) functionalized FeOCl nanosheets (FeOCl@PAA-Lip). In this system, the ˙OH radicals produced from the Fenton reaction between FeOCl nanosheets and endogenous H2O2 of tumor cells attack Lip on the nanosheets in situ to initiate the lipid peroxidation chain reaction, which not only realizes free radical conversion but also leads to the amplification of ROS and lipid peroxides, thus enhancing tumor ferroptosis. The in vitro and in vivo results confirmed that FeOCl@PAA-Lip nanosheets exhibited specific tumor cell-killing effects, good biocompatibility, long circulation time, low side effects, high tumor targeting and an excellent tumor inhibition rate (73%). The Lip functionalization strategy offers a paradigm of enhancing ferroptosis treatment by conversion of ˙OH/phospholipid radicals/lipid peroxyl radicals and strengthening lipid peroxidation. 22. Endogenous Fe2+-activated ROS nanoamplifier for esterase-responsive and photoacoustic imaging-monitored therapeutic improvement.

Abstract

Chemodynamic therapy (CDT) is well acknowledged as potent reactive oxygen species (ROS)-mediated anticancer strategy. Especially, the study about labile iron pool (LIP) as endogenous ferrous catalyzer has paved the way for future CDT development. However, limited H2O2 expression, mild acidity, reduced glutathione (GSH) ablation of ROS, etc., all require employing alternate peroxo-complex to achieve enhanced CDT effect. As a non-Fenton-type substrate, artesunate (ART) has been utilized as a source of free radicals through decomposition of endoperoxide bridges catalyzed by ferrous ions, nonetheless, the non-tumor-specific delivery, inferior pharmacokinetics, and hydrophobic nature minimize the efficacy of ART in physiological systems. Herein, we devise a PPA nanoamplifier by conjugating ART with PEG-functionalized Pd@Pt nanoplates (PP NPs) to form ester linkage, ensuring specific intratumoral esterase-responsive ART release. Significantly, the PPA nanoamplifier combines the in situ decomposition of ART's endoperoxide bridges by Fe2+ to superoxide anions (O2·−) and peroxidase (POD)-like enzymatic catalysis of endogenous H2O2 by PP to hydroxyl radicals (·OH), thus achieving amplified ROS-mediated tumor therapy. Besides, PPA displays GSH destruction potential, thereby protecting ROS from the cleavage by GSH oxidation. In addition, the strong absorption of PPA in near-infrared (NIR) region also endows PPA with photoacoustic property to realize imaging-guided CDT. In short, by taking advantages of the high enrichment and esterase- responsive drug release at tumor sites, PPA amplified ROS signals via dual pathways, killing tumor cells in vitro and inhibiting tumor growth in vivo, thereby realizing high-efficiency non-Fenton CDT. Abstract

A bio-inspired approach was used to integrate an anti-tumor drug into nanocrystalline assemblies for sustained drug release., Delicate mesoscopic architectures, bearing complex forms with multiple hierarchy levels, lead to significant functions in biogenic minerals. Herein, a bio-inspired approach was developed to fabricate comet-shaped assemblies of an anti-tumor drug – 10-hydroxycamptothecin (HCPT). The anti-solvent co-precipitation of HCPT and the excipient – PEG-b-PLGA – within the emulsifier leads to the immediate nucleation of comet bundles, followed by a secondary nucleation to generate the comet head, which is an assembly of nanofibers aligned almost in parallel. The continuous manufacturing furnishes drug–excipient hybrid particles with high drug-loading and a sustained drug release profile. This simple and efficient bio-inspired approach led to a promising sustained local drug delivery system, and could be extended to the fabrication of other functional organic materials bearing mesoscopic structural units.

35. Molybdenum Disulfide Nanoparticles Resist Oxidative Stress-Mediated Impairment of Autophagic Flux and Mitigate Endothelial Cell Senescence and Angiogenic Dysfunctions

Abstract

The impairment of autophagy involves oxidative stress-induced cellular senescence, leading to endothelial dysfunctions and the onset of cardiovascular diseases. As molybdenum disulfide nanoparticles (MoS2NPs), representative transition metal dichacogenide materials, have great potential as a multifunctional therapeutic agent against various disorders, the present study aimed to investigate whether MoS2NPs prevents hydrogen peroxide (H2O2)-induced endothelial senescence by modulating autophagic process. Our results showed that pretreatment with MoS2NPs inhibited H2O2-induced endothelial senescence and improved endothelial functions. Exposure of H2O2increased p62 level and blocked the fusion of autophagosomes with lysosomes, indicating of impaired autophagic flux in senescent endothelial cells. However, MoS2NPs pretreatment efficiently suppressed cellular senescence through triggering autophagy and resisting impaired autophagic flux. Furthermore, the genetic inhibition of autophagy by siRNA against Beclin 1 or ATG-5 directly abrogated the protective action of MoS2NPs on endothelial cells against H2O2-induced senescence.Thus, these results suggested that MoS2NPs rescue endothelial cells from H2O2-induced senescence by improving autophagic flux, and provide valuable information for the rational design of MoS2-based nanomaterials for therapeutic use in senescence-related diseases.
