Background: Amygdalin is the essential component of the traditional Chinese medicinal drug peach kernel. The early studies of amygdalin are mainly focused on antitussive and tumor anti-tumor, but there are few studies on the treatment of neuroinflammation. Objectives: The purpose of this study was to explore the anti-inflammatory effect of amygdalin on lipopolysaccharide (LPS)-induced neuron–microglia and its protective mechanism for toll-like receptors (TLRs) signaling pathways. Materials and Methods: Griess, quantitative PCR, and flow cytometry were used to detect the influence of amygdalin on the release of inflammatory factors from BV2 mouse microglia (BV2 cells). The anti-inflammatory pathway of amygdalin was further explored by western blotting. At the same time, the effect of amygdalin on LPS-induced oxidative stress damage in BV2 cells was detected by WST-8 and fluorescence probe. Results: After LPS induction, the pro-inflammatory factors such as nitric oxide, interleukin-6, tumor necrosis factor-alpha, interleukin-1β, and recombinant NLR Family, Pyrin Domain Containing Protein3 were significantly increased. However, the levels were reversed significantly after treatment with amygdalin. In addition, amygdalin significantly inhibited the increase of TLR4, TLR2, and its downstream signal myeloid differentiation primary response gene 88 and P-NF-κB p65 in BV2 cells induced by LPS. Amygdalin can also reverse the abnormal expression of LPS-induced oxidative stress indexes such as cyclooxygenase-2, reactive oxygen species, and superoxide dismutase. Conclusion: Amygdalin may reduce the LPS-induced accumulation of pro-inflammatory substances via the down-regulating of the TLR2/TLR4-nuclear factor kappa-B signaling pathway. [ABSTRACT FROM AUTHOR]

BACKGROUND: Anaerobic ammonium oxidation (anammox) is a promising process for nitrogen removal, but it cannot always be used successfully in the presence of inorganic matter. The feasibility of using the anammox process for treating wastewater that contains fluoridewas examined in this study. The short-term inhibitory effectwas tested at various fluoride concentrations and exposure times, and diverse pre-exposure strategieswere performed in special serum bottles. Subsequently, the long-term effect of fluoride stress was continuously monitored and evaluated in an upflow anaerobic sludge blanket (UASB) reactor. RESULTS: The50%inhibitory concentration (IC50) of fluoride for anammoxbiomasswas calculated to be 767mgL-1 in batch tests with an initial total nitrogen (TN) level of 200mgL-1. A remarkable inhibitory effect of fluoride emergedwhen the substratewas supplied at >420 mg L-1 TN. The diameter and settling velocity of the granules significantly increased with the addition of 250 mg L-1 fluoride. CONCLUSION: The performance of the anammox system and the granule properties were abnormal at a fluoride concentration of 400 mg L-1. The anammox UASB reactor was able to treat wastewater containing up to 350 mg L-1 fluoride after 183 days. [ABSTRACT FROM AUTHOR]

Background: Adalimumab is used in an attempt to maintain remission for Ulcerative colitis. This study was to evaluate the efficacy and adverse events of adalimumab compared with placebo in inducing remission of Ulcerative colitis., Methods: MEDLINE, EMBASE, the Cochrane Controlled Trials Register, OVID, BIOSIS, CNKI, and Google were searched. All randomized trials comparing adalimumab with placebo in inducing remission of moderate-to-severe ulcerative colitis were included., Results: Two randomized controlled trials with a total of 754 participants met the inclusion criteria. The pooled risk ratio (RR) of clinical remission was 1.85 (95% confidence interval (CI) 1.26 to 2.72) following adalimumab treatment. RR of clinical response was 1.40 (95% CI 1.19 to 1.65) while that of mucosal healing was 1.23 (95% CI 1.03 to 1.47). RR of any adverse events was 1.00 (95% CI 0.93 to 1.09)., Conclusion: Compared with placebo, administration of adalimumab may increase the proportion of patients with moderate-to-severe ulcerative colitis attaining clinical remission, clinical response and mucosal healing. Adalimumab is also tolerated well in these patients.

This article presents some suggestions about how to strengthen the management of medical devices so as to remove the hidden perils and risks of medical safety.

Objective: To summarize the extracorporeal circulation(ECC) management experiences on 4 cases of orthotopic heart transplantation., Methods: All patients received moderate hypothermia and high flow rate perfusion. Many strategies were taken to maximize the protection of myocardium, lung, kidney and blood conservation. The donor hearts were arrested with aorto perfusion using modified St. Thomas solution at 4 degrees C. After being flushed with 5,000 mL cold saline, the donor hearts were perfused with 1,000 mL UW solution in 4 degrees C and preserved in ice saline. Ice mud was covered on the donor heart and cold saline was dripped into left atrium in the period of anastomosis., Results: The ECC time was 133.3+/-13.1 min, and the warm and cold ischemia time was respectively 1-3 min and 142.0+/-28.2 min. All the cases recovered heart beats automatically. Left ventricular ejection fractions were 60%-65% a month postoperatively. One patient died of acute rejection 150 days postoperatively. 3 cases survived., Conclusion: Proper ECC management, importment organs and donor heart protection are the key to success in heart transplant operations.
