Abstract Background USP54, a ubiquitin-specific protease in the deubiquitinase (DUB) family, facilitates the malignant progression of several types of cancer. However, the role of USP54 in prostate cancer (PCa), especially castration-resistant prostate cancer (CRPC), remains unknown. Methods We established the CRPC LNCaP-AI cell line from the hormone-sensitive prostate cancer (HSPC) LNCaP cell line. RNA-Seq was utilized to explore DUB expression levels in LNCaP and LNCaP-AI. USP54 was knocked down, and its effects on cell growth were evaluated in vitro and in vivo. Bioinformatics analyses were conducted to explore signaling pathways affected by USP54 in PCa. Quantitative polymerase chain reaction was used to confirm key signaling pathways involved. Results USP54 was the most strongly upregulated DUB in LNCaP-AI cells compared with LNCaP cells. USP54 levels were higher in PCa than in normal tissues. USP54 silencing suppressed the proliferation of PCa cell lines, both in vitro and in vivo. USP54 expression was positively correlated with the androgen receptor (AR) signaling level in PCa samples, and USP54 knockdown inhibited AR signaling in PCa cells. Conclusions USP54 was upregulated during HSPC progression to CRPC. USP54 depletion suppressed CRPC cell proliferation both in vitro and in vivo. USP54 may facilitate PCa progression by regulating AR signaling.

Marius Hills volcanic complex (MHVC) comprises one of the densest volcanic structures on the Moon and is of significant interest for comprehending lunar volcanism. In this article, brightness temperature (TB), normalized TB, and brightness temperature difference were first generated based on microwave radiometer (MRM) data from the ChangE-2 Lunar Orbiter and (FeO+TiO2) abundance was calculated from Clementine ultraviolet-visible data to investigate the microwave thermal emissions and evolutionary history of the MHVC. The results are as follows. First, surface composition derived from the optical data is strongly influenced by impact events, and MRM data can better reflect the thermophysical properties of surface deposits. Second, a notable concentration of high TB values at daytime suggests an enrichment of volcanic activities. Third, later volcanic activities are indicated by the TB performances at daytime and night in multiple regions. Finally, a hot anomaly was detected by MRM data, suggesting the higher substrate temperature and the existence of a potential hidden volcanic structure. The results suggest that MRM data provide new insights into the evolutionary history of the MHVC, as well as an important perspective for lunar thermal evolution.

Land use change monitoring is a common theme in achieving sustainable development, while research on ecological barrier transition zones is relatively scarce. This study quantitatively analyzes the characteristics and patterns of land use change in Western Jilin, located in the transitional zone between the northeast forest belt and the northern sand prevention belt, from 1990 to 2020. Land dynamic change index and transition matrix are used to quantify the rates and intensities, and conversions between different land use types over time, respectively. Geodetector is adopted to analyze the impact of 12 factors on 12 types of land use change, such as using the factor detector to quantify the influence of temperature on the conversion from cropland to unused land. The results indicate that from 1990 to 2020, there have been noticeable changes in the area of various land use types in western Jilin. However, the conversion types are relatively limited, mainly involving interchanges between cropland, grassland, unused land, and water bodies. The cropland has increased by 20% overall, but 16% of that increase occurred from 1990–2000. The woodland area has steadily increased at a growth rate of 5–8% from 2000–2020, aligning with sustainable development strategies. Water bodies and grasslands are undergoing continuous recovery, and a positive growth trend is predicted to emerge by 2030. The built-up land is steadily expanding. The influencing factors vary for different types of land-use change. In a short time, policy factors play a significant role in land use, such as the implementation of the “River-lake Connection Project”, which has helped to reduce water-body fragmentation and enabled the stable recovery of water resources. However, in the long term, multiple topographic, climatic, and anthropogenic factors exhibit interactive effects in the land use change process in the area. Governments can take corresponding measures and management policies based on the influence of these factors to allocate and plan land use rationally.

Abstract Background The anti-tumor activities of Natural compounds and their derivatives are of great interest to pharmaceutical industries. Fisetin is one of prospective natural compounds in this regard but unfortunately with poor hydrophilicity. Methods The effects of unmodified and modified fisetin in cultured ovarian cancer cells were compared by transmission electronmicroscopy to determine apoptotic bodies, MTT assay to quantitate cell numbers, and fluorescence activated cell sorting analyse of various markers to determine the apoptotic state. In addition, the efficacy of fisetin and fisetin-micelles in vivo was determined by using immunocompromised mice. Apoptosis was measured by established markers using both western blot analysis and immunochemistry. Angiogenesis in a xenograft mouse model carring SKOV3 cells was evaluated by color Doppler ultrasound and immunohistochemistry. Result Multiple lines of evidence indicated that fisetin and fisetin micelles induce apoptosis in ovarian cancer cells in a dose-dependent manner. Histological analysis, terminal deoxynucleotidyltransferase-mediated nick-end labeling assay, western blot, immunohistochemical detection and microvessel density detection demonstrated that fisetin and fisetin micelles induced increased tumor apoptosis, proliferation suppression and antiangiogenesis activities. Conclusion As far as we know, the present study is the first time to demonstrate the potency of both fisetin and fisetin micelles inducing apoptosis in ovarian cancer cells. Further studies will be needed to validate the therapeutic potential of fisetin and fisetin micelles in ovarian cancer treatment.

Objective To investigate the efficacy of intravitreal injection of Ozurdex® in the treatment of refractory diabetic macular edema (DME) with different optical coherence tomography (OCT) patterns. Methods A total of 38 patients (41 eyes) who underwent fundus examination and were diagnosed with refractory DME in Department of Ophthalmology in our hospital from June 2018 to June 2020 were enrolled in this study, and according to the OCT pattern, they were divided into diffuse retinal thickening (DRT) group, cystoid macular edema (CME) group, and serous retinal detachment (SRD) group. Intravitreal injection of Ozurdex® was performed for all diseased eyes as needed, and best corrected visual acuity (BCVA) and OCT measurements were performed before treatment and at 1, 3, and 6 months after treatment. The three groups were compared in terms of BCVA, central foveal thickness (CFT), the integrity of external limiting membrane, ellipsoid zone integrity, and the number of hyperreflective dots (HRDs) before and after treatment. Results All three groups had a significant improvement in BCVA at 1, 3, and 6 months after treatment (Fintra-group=15.334-42.187,P0.05), and at 1, 3, and 6 months after treatment, there was a significant change in the proportion of diseased eyes with different numbers of HRDs (H=23.645-39.117,P
