Diabetic retinopathy is one of the main causes of blindness in human eyes, and lesion segmentation is an important basic work for the diagnosis of diabetic retinopathy. Due to the small lesion areas scattered in fundus images, it is laborious to segment the lesion of diabetic retinopathy effectively with the existing U-Net model. In this paper, we proposed a new lesion segmentation model named FFU-Net (Feature Fusion U-Net) that enhances U-Net from the following points. Firstly, the pooling layer in the network is replaced with a convolutional layer to reduce spatial loss of the fundus image. Then, we integrate multiscale feature fusion (MSFF) block into the encoders which helps the network to learn multiscale features efficiently and enrich the information carried with skip connection and lower-resolution decoder by fusing contextual channel attention (CCA) models. Finally, in order to solve the problems of data imbalance and misclassification, we present a Balanced Focal Loss function. In the experiments on benchmark dataset IDRID, we make an ablation study to verify the effectiveness of each component and compare FFU-Net against several state-of-the-art models. In comparison with baseline U-Net, FFU-Net improves the segmentation performance by 11.97%, 10.68%, and 5.79% on metrics SEN, IOU, and DICE, respectively. We describe a method for the differentiation of carboxylate anions on disposable paper supports (common printer paper, filter paper, chromatography paper), based on differential patterns of interactions between carboxylates and a fluorescent sensing system. The sensor was built from commercially available components, namely a polycationic fifth generation amine-terminated poly(amidoamine) dendrimer (PAMAM G5) and a small organic fluorophore (calcein) through non-covalent interactions. The assay’s physical dimensions were chosen to conform to the microwell plate standard so detection could be carried out on widely available plate reader instrumentation. The sensing complex was first deposited in spots on a paper support to prepare the sensor strip; a carboxylate solution was then loaded on each spot. Nuanced changes in fluorescence were associated with carboxylate binding to the PAMAM dendrimer, characteristic of the structure and affinity of each carboxylate. Such signal changes, interpreted through Linear Discriminant Analysis (LDA), contained enough information to recognize and successfully discriminate most anions in the panel. Among the substrates we tested, chromatography paper was the most promising. The relationship between the structure of the carboxylates and the patterns giving rise to their differentiation was also discussed. Background: Subjects with elevated Epstein-Barr virus (EBV) immunoglobulin A (IgA) titers have a higher risk of developing nasopharyngeal carcinoma (NPC), indicating that reactivation of EBV in the local mucosa might be important for NPC carcinogenesis. Cigarette smoking appears to be one of the environmental risk factors for NPC. However, it remains unclear whether smoking-induced nasopharyngeal carcinogenesis acts through reactivating EBV in the nasopharyngeal mucosa. Therefore, this study aims to investigate the association between cigarette smoking and nasopharyngeal EBV reactivation in a NPC high-risk population.Methods: A NPC high-risk cohort study, established from a population-based NPC screening program of 22,816 subjects, consisted of 1045 subjects with elevated serum IgA antibodies against EBV viral capsid antigen (VCA/IgA). Among high-risk subjects, information on detailed cigarette smoking history was collected among 313 male subjects. The associations between cigarette smoking and EBV antibody levels, EBV DNA load of the nasopharynx were analyzed.Results: No significant association was observed between either nasopharyngeal EBV DNA load or serum VCA/IgA titers and smoking status, age at smoking initiation, daily smoking intensity, smoking duration, cigarette type, or pack-years of smoking. Cigarette smoking characteristics in all subgroups did not correlate with nasopharyngeal EBV DNA positivity or EBV VCA/IgA seropositivity.Conclusions: In a population at high risk of NPC, our study suggests that cigarette smoking is neither associated with nasopharyngeal EBV DNA load nor serum VCA/IgA antibody level. Morphology plays an essential role in chemistry through the segregation of atoms and/or molecules into different phases, delineated by interfaces. This is a general process in materials synthesis and exploited in many fields including colloid chemistry, heterogeneous catalysis, and functional molecular systems. To rationally design complex materials, we must understand and control morphology evolution. Toward this goal, we utilize cryogenic transmission electron microscopy (cryoTEM), which can track the structural evolution of materials in solution with nanometer spatial resolution and a temporal resolution of <1 s. In this Account, we review examples of our own research where direct observations by cryoTEM have been essential to understanding morphology evolution in macromolecular self-assembly, inorganic nucleation and growth, and the cooperative evolution of hybrid materials. These three different research areas are at the heart of our approach to materials chemistry where we take inspiration from the myriad examples of complex materials in Nature. Biological materials are formed using a limited number of chemical components and under ambient conditions, and their formation pathways were refined during biological evolution by enormous trial and error approaches to self-organization and biomineralization. By combining the information on what is possible in nature and by focusing on a limited number of chemical components, we aim to provide an essential insight into the role of structure evolution in materials synthesis. Bone, for example, is a hierarchical and hybrid material which is lightweight, yet strong and hard. It is formed by the hierarchical self-assembly of collagen into a macromolecular template with nano- and microscale structure. This template then directs the nucleation and growth of oriented, nanoscale calcium phosphate crystals to form the composite material. Fundamental insight into controlling these structuring processes will eventually allow us to design such complex materials with predetermined and potentially unique properties. CaCO3-based biominerals usually possess sophisticated hierarchical structures and promising mechanical properties. Recent researches imply that vaterite may play an important role in the formation of CaCO3-based biominerals. However, as a less common polymorph of CaCO3, the growth mechanism of vaterite remains unclear. Here we report the growth of a pumpkin-shaped vaterite hierarchical structure with a six fold symmetrical axis and lamellar microstructure. We demonstrate that the growth is controlled by supersaturation and the intrinsic crystallographic anisotropy of vaterite. For the scenario of high supersaturation, the nucleation rate is higher than the lateral extension rate, favoring the "double-leaf" spherulitic growth. Meanwhile, nucleation occurs preferentially in 〈1120〉 as determined by the crystalline structure of vaterite, modulating the grown products with a hexagonal symmetry. The results are beneficial for an in-depth understanding of the biomineralization of CaCO3. Objective. Ceruloplasmin was elevated in patients with coronary heart disease, but the relationship between ceruloplasmin and heart failure was still unknown. We aimed to evaluate ceruloplasmin in heart failure patients and assess association between ceruloplasmin and the extent of heart failure. Methods and Results. 202 heart failure patients were divided into ischemic (78 with coronary stenosis) and nonischemic groups (124 without coronary stenosis). 94 subjects without heart failure were included as controls. The extent of heart failure was defined according to NYHA classification. Ceruloplasmin levels in ischemic (P < 0.001) and nonischemic groups (P < 0.001) were higher than those in control group. Ceruloplasmin had a positive linear correlation with C-reactive protein (P < 0.01) and a negative linear correlation with LVEF (P < 0.05). In nonischemic group, CP levels were significantly different among different NYHA subgroups (P < 0.05). The correlation between ceruloplasmin and extent of heart failure was calculated by binary logistic regression. Ceruloplasmin showed an independent association with the extent of heart failure in nonischemic cardiomyopathy patients (P < 0.05). Conclusions. Ceruloplasmin was significantly elevated in patients with ischemic or nonischemic cardiomyopathy and had linear correlation with C-reactive protein and LVEF. TRIM32 is often aberrantly expressed in many types of cancers. Kaposi’s sarcoma-associated herpesvirus (KSHV) is linked with several human malignancies, including Kaposi’s sarcoma and primary effusion lymphomas (PELs). Increasing evidence has demonstrated the crucial role of KSHV lytic replication in viral tumorigenesis. However, the role of TRIM32 in herpesvirus lytic replication remains unclear. Here, we reveal that the expression of TRIM32 is upregulated by KSHV in latency, and reactivation of KSHV lytic replication leads to the inhibition of TRIM32 in PEL cells. Strikingly, RTA, the master regulator of lytic replication, interacts with TRIM32 and dramatically promotes TRIM32 for degradation via the proteasome systems. Inhibition of TRIM32 induces cell apoptosis and in turn inhibits the proliferation and colony formation of KSHV-infected PEL cells and facilitates the reactivation of KSHV lytic replication and virion production. Thus, our data imply that the degradation of TRIM32 is vital for the lytic activation of KSHV and is a potential therapeutic target for KSHV-associated cancers. IMPORTANCE TRIM32 is associated with many cancers and viral infections; however, the role of TRIM32 in viral oncogenesis remains largely unknown. In this study, we found that the expression of TRIM32 is elevated by Kaposi’s sarcoma-associated herpesvirus (KSHV) in latency, and RTA (the master regulator of lytic replication) induces TRIM32 for proteasome degradation upon viral lytic reactivation. In order to evaluate the URTPSQ (Urban Rail Transit Passenger Service Quality) comprehensively, find the shortage of URTPSQ, find out the difference between the actual service situation and the passenger's expectation and demand, and provide passengers with better travel services, a passenger-oriented KANO-Entropy-TOPSIS (Technique for Order of Preference by Similarity to Ideal Solution) method is proposed and applied in this paper. Firstly, a KANO model is applied to select the service quality indicators from the 24 URTPSQ evaluation sub-indicators, according to the selection results, the KANO service quality indicators of URTPSQ are constructed. Then the sensitivity of the KANO service quality indicators based on the KANO model are calculated and ranked, the PS (Passenger Satisfaction) of each KANO service quality indicator by using the Entropy-TOPSIS method is calculated and ranked. Based on the difference between the sensitivity degree rank and the satisfaction degree rank of each KANO service quality indicator, determine the service quality KANO indicators of the URTPSQ that need to be improved significantly. A case study is conducted by taking the Chengdu subway system in China as a background. The results show that the Chengdu subway operation enterprises should pay attention to the must-be demand first, then the one-dimensional demand, finally the attractive demand. The three indicators, including transfer on the same floor in the station, service quality of staffs of urban rail transit enterprises, and cleanness in the station and passenger coach, need to be improved urgently. For the managers and operators of urban rail transit system, the passengers' must-be demand should be satisfied first if the KANO model is applied to evaluate the service. The indicators with highest sensitivity degree and lowest TOPSIS value should be improved based on the KANO-Entropy-TOPSIS model. [ABSTRACT FROM AUTHOR]

Context: Amauroderma rugosum (Blume & T. Nees) Torrend (Ganodermataceae) is an edible mushroom with medicinal properties. However, the effects of A. rugosum on gastric ulcer remain unclear. Objective: To investigate the gastroprotective efficacy of water extract of A. rugosum (WEA) on gastric ulcer. Materials and methods: Sprague-Dawley rats were randomly grouped as control, model, lansoprazole and 200, 100 and 50 mg/kg of WEA. After pre-treatment for seven days, ethanol- and indomethacininduced gastric ulcer models were established. The gastric ulcer and histopathology were investigated. Enzyme-linked immunosorbent assay (ELISA), quantitative polymerase chain reaction (Q-PCR) and Western blot assays were conducted to explore the potential anti-inflammatory effect and mechanism of WEA. Additionally, the pyloric ligation model was used to explore the influence of WEA on gastric acid and mucus. Results: Pre-treatment with WEA (200, 100 and 50mg/kg) effectively reduced ulcerous area in both ethanol-induced (71%, 88% and 71%) and indomethacin-induced (77%, 65% and 86%) gastric ulcer model. The gastric levels of tumour necrosis factor-alpha (TNF-a) (34% and 50mg/kg), interleukin-6 (IL-6) (32% and 100mg/kg) and interleukin-1b (IL-1b) (36%, 45% and 41%) were reduced significantly (p<0.05) by WEA. Serum nitric oxide was decreased significantly (p<0.05) at 200 and 50mg/kg and PGE2 concentration was increased remarkably (p<0.05) at 100 mg/kg. Gene expression of inflammasome Nlrp3, and the nuclear translocation of nuclear factor-jB (NF-jB) P65 were significantly decreased by WEA pre-treatment. However, the pH of gastric acid and secretion of mucus did not show any significant change. Conclusions: The gastroprotective effect of WEA on gastric damage is attributed to anti-inflammation through the inhibition on NF-jB P65 nuclear migration and Nlrp3 gene expression. Background Phylogenetic networks are employed to visualize evolutionary relationships among a group of nucleotide sequences, genes or species when reticulate events like hybridization, recombination, reassortant and horizontal gene transfer are believed to be involved. In comparison to traditional distance-based methods, quartet-based methods consider more information in the reconstruction process and thus have the potential to be more accurate. Results We introduce QuartetSuite, which includes a set of new quartet-based methods, namely QuartetS, QuartetA, and QuartetM, to reconstruct phylogenetic networks from nucleotide sequences. We tested their performances and compared them with other popular methods on two simulated nucleotide sequence data sets: one generated from a tree topology and the other from a complicated evolutionary history containing three reticulate events. We further validated these methods to two real data sets: a bacterial data set consisting of seven concatenated genes of 36 bacterial species and an influenza data set related to recently emerging H7N9 low pathogenic avian influenza viruses in China. Conclusion QuartetS, QuartetA, and QuartetM have the potential to accurately reconstruct evolutionary scenarios from simple branching trees to complicated networks containing many reticulate events. Two subtypes of influenza A virus (IAV), avian-origin canine influenza virus (CIV) H3N2 (CIV-H3N2) and equine-origin CIV H3N8 (CIV-H3N8), are enzootic in the canine population. Dogs have been demonstrated to seroconvert in response to diverse IAVs, and naturally occurring reassortants of CIV-H3N2 and the 2009 H1N1 pandemic virus (pdmH1N1) have been isolated. We conducted a thorough phenotypic evaluation of CIV-H3N2 in order to assess its threat to human health. Using ferret-generated antiserum, we determined that CIV-H3N2 is antigenically distinct from contemporary human H3N2 IAVs, suggesting that there may be minimal herd immunity in humans. We assessed the public health risk of CIV-H3N2 × pandemic H1N1 (pdmH1N1) reassortants by characterizing their in vitro genetic compatibility and in vivo pathogenicity and transmissibility. Using a luciferase minigenome assay, we quantified the polymerase activity of all possible 16 ribonucleoprotein (RNP) complexes (PB2, PB1, PA, NP) between CIV-H3N2 and pdmH1N1, identifying some combinations that were more active than either parental virus complex. Using reverse genetics and fixing the CIV-H3N2 hemagglutinin (HA), we found that 51 of the 127 possible reassortant viruses were viable and able to be rescued. Nineteen of these reassortant viruses had high-growth phenotypes in vitro, and 13 of these replicated in mouse lungs. A single reassortant with the NP and HA gene segments from CIV-H3N2 was selected for characterization in ferrets. The reassortant was efficiently transmitted by contact but not by the airborne route and was pathogenic in ferrets. Our results suggest that CIV-H3N2 reassortants may pose a moderate risk to public health and that the canine host should be monitored for emerging IAVs. [ABSTRACT FROM AUTHOR]

Abstract. Grape seed proanthocyanidin (GSPA) consists of catechin, epicatechin and epicatechin gallate, which are strong antioxidants that are beneficial to health and may attenuate or prevent Alzheimer's disease (AD). In the present study, the effects of GSPA on pheochromocytoma (PC12) cell viability were determined using cell counting kit-8 and lactate dehydrogenase (LDH) assays, whereas apoptosis and mitochondrial membrane potential (ψm) were measured via low cytometry analysis. The effect of GSPA administration on the behavior and memory of amyloid precursor protein (APP)/presenilin-1 (PS-1) double transgenic mice was assessed using a Morris water maze. APP Aβ peptides and tau hyperphosphorylation were examined by western blotting; whereas the expression levels of PS-1 were evaluated by reverse transcription-quantitative polymerase chain reaction and compared with pathological sections stained with hematoxylin-eosin and Congo red. Data from the in vitro experiments demonstrated that GSPA significantly alleviated Aβ25-35 cytotoxicity and LDH leakage ratio, inhibited apoptosis and increased ψm. The findings from the in vivo experiments showed a significant enhancement in cognition and spatial memory ability, an improvement in the pathology of APP and tau protein and a decrease in PS-1 mRNA expression levels. Therefore, the results of the present study indicated that GSPA may be a novel therapeutic strategy for the treatment of AD or may, at the very least, improve the quality of life of patients with AD.
