Abstract. Background:. Chronic kidney disease (CKD) is associated with common pathophysiological processes, such as inflammation and fibrosis, in both the heart and the kidney. However, the underlying molecular mechanisms that drive these processes are not yet fully understood. Therefore, this study focused on the molecular mechanism of heart and kidney injury in CKD. Methods:. We generated an microRNA (miR)-26a knockout (KO) mouse model to investigate the role of miR-26a in angiotensin (Ang)-II-induced cardiac and renal injury. We performed Ang-II modeling in wild type (WT) mice and miR-26a KO mice, with six mice in each group. In addition, Ang-II-treated AC16 cells and HK2 cells were used as in vitro models of cardiac and renal injury in the context of CKD. Histological staining, immunohistochemistry, quantitative real-time polymerase chain reaction (PCR), and Western blotting were applied to study the regulation of miR-26a on Ang-II-induced cardiac and renal injury. Immunofluorescence reporter assays were used to detect downstream genes of miR-26a, and immunoprecipitation was employed to identify the interacting protein of LIM and senescent cell antigen-like domain 1 (LIMS1). We also used an adeno-associated virus (AAV) to supplement LIMS1 and explored the specific regulatory mechanism of miR-26a on Ang-II-induced cardiac and renal injury. Dunnett’s multiple comparison and t-test were used to analyze the data. Results:. Compared with the control mice, miR-26a expression was significantly downregulated in both the kidney and the heart after Ang-II infusion. Our study identified LIMS1 as a novel target gene of miR-26a in both heart and kidney tissues. Downregulation of miR-26a activated the LIMS1/integrin-linked kinase (ILK) signaling pathway in the heart and kidney, which represents a common molecular mechanism underlying inflammation and fibrosis in heart and kidney tissues during CKD. Furthermore, knockout of miR-26a worsened inflammation and fibrosis in the heart and kidney by inhibiting the LIMS1/ILK signaling pathway; on the contrary, supplementation with exogenous miR-26a reversed all these changes. Conclusions:. Our findings suggest that miR-26a could be a promising therapeutic target for the treatment of cardiorenal injury in CKD. This is attributed to its ability to regulate the LIMS1/ILK signaling pathway, which represents a common molecular mechanism in both heart and kidney tissues.

Abstract Background Workplace may not only increase the risk of heat-related illnesses and injuries but also compromise work efficiency, particularly in a warming climate. This study aimed to utilize machine learning (ML) and deep learning (DL) algorithms to quantify the impact of temperature discomfort on productivity loss among petrochemical workers and to identify key influencing factors. Methods A cross-sectional face-to-face questionnaire survey was conducted among petrochemical workers between May and September 2023 in Fujian Province, China. Initial feature selection was performed using Lasso regression. The dataset was divided into training (70%), validation (20%), and testing (10%) sets. Six predictive models were evaluated: support vector machine (SVM), random forest (RF), extreme gradient boosting (XGBoost), Gaussian Naive Bayes (GNB), multilayer perceptron (MLP), and logistic regression (LR). The most effective model was further analyzed with SHapley Additive exPlanations (SHAP). Results Among the 2393 workers surveyed, 58.4% (1,747) reported productivity loss when working in high temperatures. Lasso regression identified twenty-seven predictive factors such as educational level and smoking. All six models displayed strong prediction accuracy (SVM = 0.775, RF = 0.760, XGBoost = 0.727, GNB = 0.863, MLP = 0.738, LR = 0.680). GNB model showed the best performance, with a cutoff of 0.869, accuracy of 0.863, precision of 0.897, sensitivity of 0.918, specificity of 0.715, and an F1-score of 0.642, indicating its efficacy as a predictive tool. SHAP analysis showed that occupational health training (SHAP value: -3.56), protective measures (-2.61), and less physically demanding jobs (-1.75) were negatively associated with heat-attributed productivity loss, whereas lack of air conditioning (1.92), noise (2.64), vibration (1.15), and dust (0.95) increased the risk of heat-induced productivity loss. Conclusions Temperature discomfort significantly undermined labor productivity in the petrochemical sector, and this impact may worsen in a warming climate if adaptation and prevention measures are insufficient. To effectively reduce heat-related productivity loss, there is a need to strengthen occupational health training and implement strict controls for occupational hazards, minimizing the potential combined effects of heat with other exposures.

The blood-brain barrier (BBB) constitutes a unique anatomical and physiological interface between the central nervous system (CNS) and the peripheral circulation. It is of critical importance for maintaining the stability of the CNS. This article provides an overview of the changes in the structure of the BBB during the progression of diseases such as aging, neurological disorders, and cancer, with a focus on the effects of gut microbiota and their metabolites on the BBB. Environmental factor-induced changes in the composition of the gut flora disrupt BBB integrity through multiple pathways, whereas gut microbial metabolites have positive, negative, or bidirectional modulatory effects on the BBB. For example, lipopolysaccharide and hydrophobic bile acids have negative effects on the BBB by disrupting cellular structure, decreasing tight junction protein expression, and exacerbating inflammatory responses. The short chain fatty acids, 3-indolepropionic acid, and hydrophilic bile acids have positive effects on the BBB by regulating the intestinal flora, increasing tight junction protein expression, reducing inflammation and exerting antioxidant effects. The trimethylamine oxide has a two-way regulatory effects on the BBB. In addition, different probiotic strains are described to directly or indirectly improve BBB integrity by altering the composition of the intestinal flora, regulating short chain fatty acid levels, enhancing tight junction protein expression, as well as anti-inflammatory and antioxidant pathways. This article provides new strategies and theoretical materials for the treatment and improvement of neurological diseases.

Abstract Polycomb repressive complex 2 (PRC2) catalyzes the writing of the tri-methylated histone H3 at Lys27 (H3K27me3) epigenetic marker and suppresses the expression of genes, including tumor suppressors. The function of the complex can be partially antagonized by the SWI/SNF chromatin-remodeling complex. Previous studies have suggested that PRC2 is important for the proliferation of tumors with SWI/SNF loss-of-function mutations. In the present study, we have developed an EED-directed allosteric inhibitor of PRC2 termed BR0063, which exhibits anti-proliferative properties in a subset of solid tumor cell lines harboring mutations of the SWI/SNF subunits, SMARCA4 or ARID1A. Tumor cells sensitive to BR0063 exhibited several distinct phenotypes, including cell senescence, which was mediated by the up-regulation of CDKN2A/p16. Further experiments revealed that the expression of p16 was suppressed in the BR0063-resistant cells via DNA hypermethylation in the CpG island (CGI) promoter region, rather than via PRC2 occupancy. The expression of TET1, which is required for DNA demethylation, was found to be inversely correlated with p16 CGI methylation, and this may serve as a biomarker for the prediction of resistance to PRC2 inhibitors in SWI/SNF LOF tumors.

Abstract The objective of this research is to address the rut problems in asphalt pavements and to resist the permanent plastic deformation with the increasing heavy traffic loads. In this paper, a new type materials of high modulus asphalt was developed by incorporating styrene–butadiene–styrene (SBS) and zinc oxide nanoparticles (nano ZnO) with an (EME)-type high modulus modifier, guided by the synergistic effects and the preparation methods of High-speed shear. The basic road performance, mechanical response and thermal stability of the new high modulus asphalt materials were analysed through basic physical indicator tests, dynamic shear rheometer tests, thermogravimetric analysis (TGA), the time–temperature superposition principle, the Refutas model, and the Christensen-Andersen-Marasteanu model. The optimal results demonstrate that the optimal blend ratio of the developed asphalt is 12% EME/8% SBS/1.5% ZnO. Under this composition, the road performance indicator values of softening point, penetration and ductility of the modified asphalt met the standard requirements. The dynamic shear rheometer tests demonstrates that the inclusion of SBS and nano ZnO considerably enhanced the shear resistance and recovery deformation capacity of EME, effectively improving the high-temperature deformation resistance of asphalt. Furthermore,the Refutas and the Christensen-Andersen-Marasteanu model fitting results showed that adding SBS and nano ZnO considerably improved the temperature sensitivity of the EME types high modulus modified asphalt and exhibiting low frequency sensitivity. Compared to PR Module-type high modulus modifier(PRM),TGA reveals that the maximum thermal weight loss of EME-SBS-ZnO decreased by 3.5441%, indicating better thermal stability and the major character of SBS,EME and asphalt is physical reaction. Moreover, EME-nano ZnO-SBS high modulus asphalt at 64 °C shows Jnr-diff = 9.5% and passes the "E" extreme traffic grade. Additionally, its cost is 4.67% lower than that of the PRM high modulus modified asphalt, presenting considerable economic benefits.

BackgroundOlder adults with hypertension are at an increased risk of depression. Social support and coping style significantly influence this risk, with social cognitive theory suggesting that social support can affect an individual’s coping style, and coping style can affect the effective use of social support. However, the mediating role of coping style in the relationship between social support and depression in older hypertensive patients remains unclear. This study aimed to explore the inter-relationships between social support, coping style and depression in older hypertensive patients within a community setting, and to investigate the mediating effects of coping style.MethodsA cross-sectional questionnaire survey was conducted with 4211 older hypertensive patients from Fuzhou, Fujian Province, China. Participants were assessed for depression, social support, coping styles, and general demographic information. Pearson correlation analysis was employed to test the correlation between variables. Mediation effect test was conducted using SPSS macro program PROCESS with Bootstrap based Model 4, after controlling for confounding factors.ResultsA total of 4211 valid questionnaires were collected, yielding an effective response rate of 98.6%. The average depression score among participants was 7.99 ± 4.92 points, with 29.5% of respondents experiencing varying degrees of depression. Both social support and coping style were associated with depression in older hypertensive patients. Specifically, objective support, subjective support, support utilization, and positive coping style were negatively correlated with depression, with correlation coefficients of -0.159, -0.160, -0.145, and -0.163, respectively. Conversely, negative coping style was positively correlated with depression, with a correlation coefficient of 0.170. Mediating effect analysis showed that coping style played a mediating role between social support and depression. Social support negatively moderated depression through positive coping style (moderating effect =-0.020, Bootstrap 95%CI [-0.027, -0.138], mediating effect value was 15.87%), while it positively influenced depression through negative coping style (moderating effect =0.012, Bootstrap 95%CI [0.008,0.017], intermediate effect value =9.52%).ConclusionSocial support exerts a direct negative effect on depression, while coping styles mediate the relationship between social support and depression. Interventions to reduce depression in this population should focus on enhancing social support (across objective, subjective, and utilization aspects) and promoting positive coping strategies, thus strengthening psychological resilience and improving overall health and quality of life for older adults with hypertension.

ObjectiveThis meta-analysis aims to evaluate the effects of blood flow restriction (BFR) training on cardiometabolic health and body composition in adults with overweight and obesity.MethodFollowing PRISMA guidelines, a systematic search of PubMed (MEDLINE), EMBASE, Web of Science, Cochrane, and Scopus databases was conducted on 15 March 2024. Pooled effects for each outcome were summarized using Hedge’s g (g) through meta-analysis-based random effects models, and subgroup analyses were used to explore moderators.ResultsA total of 11 studies with 242 participants (Age:32.6 ± 3.6, BMI:27.2 ± 3.5) were included. Regarding cardiometabolic health, BFR training significantly reduced systolic blood pressure (g = 0.62 [0.08, 1.16], p = 0.02), while no significant differences were observed in maximal oxygen uptake (g = 0.48 [-0.21, 1.17], p = 0.17) or diastolic blood pressure (g = 0.31 [-0.22, 0.84], p = 0.25). Regarding body composition, BFR training significantly reduced body fat percentage (g = 0.30 [0.01, 0.58]; p = 0.04), while no significant differences (p > 0.05) were observed in body weight (g = 0.14 [-0.14, 0.42]), body mass index (g = 0.08 [-0.21, 0.38]), waist circumference (g = 0.13 [-0.28, 0.53]), or waist-to-hip ratio (g = 0.48 [-0.19, 1.15]). Subgroup analysis revealed no significant difference in improving systolic blood pressure (g = 0.57 [-0.10, 1.24] vs. g = 0.70 [-0.18, 1.59]) and body fat percentage (g = 0.20 [-0.20, 0.61] vs. g = 0.45 [-0.05, 0.95]) between BFR resistance training and BFR aerobic training. In all selected studies, the overall risk of bias was categorized as “some concern”. The certainty of evidence for the BFR outcomes was low.ConclusionBFR training shows promise in improving cardiometabolic health and body composition, indicating that it may serve as a beneficial, individualized exercise prescription for improving cardiovascular disease risk and fat loss in adults with excess body weight and obesity.Systematic Review Registration:https://archive.org/details/osf-registrations-uv6jx-v1.

Abstract Background The contribution of cholinergic degeneration to gait disturbance in Parkinson’s disease (PD) is increasingly recognized, yet its relationship with dopaminergic-resistant gait parameters has been poorly investigated. We investigated the association between comprehensive gait parameters and cholinergic nucleus degeneration in PD. Methods This cross-sectional study enrolled 84 PD patients and 69 controls. All subjects underwent brain structural magnetic resonance imaging to assess the gray matter density (GMD) and volume (GMV) of the cholinergic nuclei (Ch123/Ch4). Gait parameters under single-task (ST) and dual-task (DT) walking tests were acquired using sensor wearables in PD group. We compared cholinergic nucleus morphology and gait performance between groups and examined their association. Results PD patients exhibited significantly decreased GMD and GMV of the left Ch4 compared to controls after reaching HY stage > 2. Significant correlations were observed between multiple gait parameters and bilateral Ch123/Ch4. After multiple testing correction, the Ch123/Ch4 degeneration was significantly associated with shorter stride length, lower gait velocity, longer stance phase, smaller ankle toe-off and heel-strike angles under both ST and DT condition. For PD patients with HY stage 1–2, there were no significant degeneration of Ch123/4, and only right side Ch123/Ch4 were corrected with the gait parameters. However, as the disease progressed to HY stage > 2, bilateral Ch123/Ch4 nuclei showed correlations with gait performance, with more extensive significant correlations were observed in the right side. Conclusions Our study demonstrated the progressive association between cholinergic nuclei degeneration and gait impairment across different stages of PD, and highlighting the potential lateralization of the cholinergic nuclei’s impact on gait impairment. These findings offer insights for the design and implementation of future clinical trials investigating cholinergic treatments as a promising approach to address gait impairments in PD.

Objectives: Efforts to shorten rifampicin-resistant tuberculosis (RR-TB) treatment have led to concerns about hepatotoxicity in shorter regimens. We evaluated hepatotoxicity in two novel regimens against the standard shorter regimen recommended by the World Health Organization (WHO). Methods: Participants from the TB-TRUST and TB-TRUST plus trials were assigned to the WHO shorter regimen, a levofloxacin (Lfx)-based regimen, or a bedaquiline (Bdq)-based regimen. Liver function was tested bi-weekly in the first month, then monthly until treatment ended. Eligibility required receiving at least one drug dose and undergoing at least two liver function tests. Results: Of 429 patients, hepatotoxicity was most prevalent in the WHO shorter group (26.7% of 169), compared to 4.7% in the Lfx group (172 patients), and 5.7% in the Bdq group (88 patients). The median peak alanine aminotransferase levels were 1.67 × upper limit of normal (ULN) for WHO, 0.82 × ULN for Lfx, and 0.88 × ULN for Bdq groups. The incidence of drug-induced liver injury was significantly higher in the WHO group (18.3%) than in the Lfx (3.5%) and Bdq (4.6%) groups. The time to significant alanine aminotransferase elevation was about 2.8 months, with no differences between groups. Conclusions: Two novel regimens demonstrated lower hepatotoxicity compared to the WHO's shorter regimen. Entire course management monitoring is recommended in RR-TB treatment.

Plant lead (Pb) tolerance and accumulation are key characteristics affecting phytoremediation efficiency. Bermudagrass is an excellent candidate for the remediation of Pb-polluted soil, and it needs to be mowed regularly. Here, we explored the effect of different mowing frequencies on the remediation of Pb-contaminated soil using bermudagrass. Mowing was found to decrease the biomass and photosynthetic efficiency of bermudagrass under Pb stress, thereby inhibiting its growth. Although mowing exacerbated membrane peroxidation, successive mowing treatments alleviated peroxidation damage by regulating enzymatic and nonenzymatic systems. A comprehensive evaluation of Pb tolerance revealed that all the mowing treatments reduced the Pb tolerance of bermudagrass, and a once-per-month mowing frequency had a less negative effect on Pb tolerance than did more frequent mowing. In terms of Pb enrichment, mowing significantly increased the Pb concentration, total Pb accumulation, translocation factor (TF), and bioenrichment factor (BCF) of bermudagrass. The total Pb accumulation was greatest under the once-a-month treatment, while the TF and BCF values were greatest under the three-times-a-month mowing treatment. Additionally, the decrease in soil pH and DOC were significantly correlated with the soil available Pb content and plant Pb accumulation parameters. The results showed that changes in the rhizosphere are crucial factors regulating Pb uptake in bermudagrass during mowing. Overall, once-a-month mowing minimally affects Pb tolerance and maximizes Pb accumulation, making it the optimal mowing frequency for soil Pb remediation by bermudagrass. This study provides a novel approach for the remediation of Pb-contaminated soil with bermudagrass based on mowing.

Background: Filamin A (FLNA) is a member of the filamin family and has been found to be critical for the progression of several cancers. However, its biological function in papillary thyroid cancer (PTC) remains largely unexplored. Methods: Data from The Cancer Genome Atlas (TCGA) databases were utilized to analyze the FLNA expression level and its influence on the clinical implications of patients with PTC. Gene Expression Omnibus (GEO) and qRT-PCR was used to verify the expression levels of FLNA in PTC. Kaplan–Meier survival analysis was conducted to evaluate the prognostic value of FLNA in PTC. Transwell assays and wound healing were performed to examine the biological function of FLNA knockdown in PTC cells. Gene set enrichment analysis (GSEA) and Western blotting were conducted to investigate the potential mechanisms underlying the role of FLNA in PTC progression. In addition, the relationship between FLNA expression and the tumor immune microenvironment (TME) in PTC was explored. Results: FLNA was significantly upregulated in PTC tissues. High expression levels of FLNA was correlated with advanced TNM stage, T stage, and N stage, as well as poor disease-free interval (DFI) and progression-free interval (PFI) time in PTC patients. Moreover, we found that FLNA knockdown inhibited the migration and invasion of PTC cells. Mechanistically, FLNA knockdown inhibited epithelial–mesenchymal transition (EMT) in PTC and affected the activation of the FAK/AKT signaling pathway. In addition, FLNA expression was associated with TME in PTC. Conclusion: FLNA may be regarded as a new therapeutic target for PTC patients.

Abstract Community-acquired respiratory infection is the commonest cause of sepsis presenting to emergency departments. Yet current experimental animal models simulate peritoneal sepsis with intraperitoneal (I.P.) injection of lipopolysaccharide (LPS) as the predominant route. We aimed to compare the progression of organ injury between I.P. LPS and intranasal (I.N.) LPS in order to establish a better endotoxemia murine model of respiratory sepsis. Eight weeks old male BALB/c mice received LPS-Escherichia coli doses at 0.15, 1, 10, 20, 40 and 100 mg per kg body weight (e.g. LPS-10 is a dose of 10 mg/kg body weight). Disease severity was monitored by a modified Mouse Clinical Assessment Score for Sepsis (M-CASS; range 0–21). A M-CASS score ≥ 10 or a weight reduction of ≥ 20%, was used as a criterion for euthanasia. The primary outcome was the survival rate (either no death or no need for euthanasia). The progression of disease was specified as M-CASS, body weight, blood glucose, histopathological changes to lung, liver, spleen, kidney, brain and heart tissues. Survival rate in I.P. LPS-20 mice was 0% (2/3 died; 1/3 euthanized with M-CASS > 10) at 24 h. Survival rate in all doses of I.N. LPS was 100% (20/20; 3–4 per group) at 96 h. 24 h mean M-CASS post-I.P. Abstract The abundant genetic variation harbored by wild rice (Oryza rufipogon) has provided a reservoir of useful genes for rice breeding. However, the genome of wild rice has not yet been comprehensively assessed. Here, we report the haplotype-resolved gapless genome assembly and annotation of wild rice Y476. In addition, we develop two sets of chromosome segment substitution lines (CSSLs) using Y476 as the donor parent and cultivated rice as the recurrent parents. By analyzing the gapless reference genome and CSSL population, we identify 254 QTLs associated with agronomic traits, biotic and abiotic stresses. We clone a receptor-like kinase gene associated with rice blast resistance and confirm its wild rice allele improves rice blast resistance. Collectively, our study provides a haplotype-resolved gapless reference genome and demonstrates a highly efficient platform for gene identification from wild rice.

Since market uncertainty, or volatility, serves as a crucial gauge for assessing the traits of market fluctuations, the link between stock market volume and price continues to be a focal point of interest in finance. This study examines the dynamic, nonlinear correlations between Chinese stock volatility, trading volume, and return using a hybrid approach that combines the Markov-switching regime with the vector autoregressive model (MS-VAR). The empirical findings are as follows: (1) The Chinese stock market can be divided into three regional systems: steady downward, steady upward, and high volatility. The three states have similar frequencies of occurrence, and their corresponding stable probabilities are not high, indicating that the Chinese stock market is unstable. (2) Asymmetric dynamic relationships exist between market volatility, investment return, and trading volume. For different regimes, while the effect of trading volume on volatility and return appears to be insignificant, the impacts of volatility and return on trading volume are considerably strong. (3) A regime-dependent, contemporaneous correlation between volatility and return is observed, which also reflects the behavior of the Chinese stock market “chasing up and down”. However, a positive contemporaneous correlation always exists between volatility and trading volumes in different regimes, indicating that uncertainty in the Chinese stock market is closely related to information inflow.

Abstract Introduction Observational studies and clinical trials have supported the association between gut microbiota and psoriatic arthritis. However, the causal link between gut microbiota and psoriatic arthritis is still unclear. Methods A two-sample bi-directional Mendelian randomization analysis was performed using the summary statistics of gut microbiota from the largest available genome-wide association study meta-analysis (n = 13,266) conducted by the MiBioGen consortium. The summary statistics of psoriatic arthritis were extracted directly from the FinnGen consortium, which consists of 3186 psoriatic arthritis patients and 24,086 controls. Sensitivity analyses were conducted to assess the validity of our findings. Enrichment analyses were used to investigate the biofunction and pathways. Results Inverse variance weighted (IVW) estimates suggested that family Rikenellaceae (P = 0.032) and genus Ruminococcaceae UCG011 (P = 0.014) had a detrimental effect on psoriatic arthritis. We also noticed the negative association between the class Methanobacteria (P = 0.032), order Methanobacteriales (P = 0.032), family Methanobacteriaceae (P = 0.032), genus Eubacterium fissicatena group (P = 0.010), genus Methanobrevibacter (P = 0.031), and genus Butyricicoccus (P = 0.041) with psoriatic arthritis. Sensitivity analyses showed that genus Butyricicoccus had pleiotropy and heterogeneity. According to the results of reverse MR analysis, the causal effect of psoriatic arthritis was found on six taxa, respectivelyc family Clostridiaceae1, family Defluviitaleaceae, genus Butyrivibrio, genus Defluviitaleaceae UCG011, genus Clostridium sensu stricto1, and genus Ruminococcaceae UCG011. Conclusion This two-sample bidirectional Mendelian randomization analysis suggested that the gut microbiota had a causal effect on psoriatic arthritis and implied the potential role of probiotics in the management and prevention of psoriatic arthritis.

The traditional Chinese herbal prescription Buyang Huanwu decoction (BHD), effectively treats atherosclerosis. However, the mechanism of BHD in atherosclerosis remains unclear. We aimed to determine whether BHD could alleviate atherosclerosis by altering the microbiome-associated metabolic changes in atherosclerotic mice. An atherosclerotic model was established in apolipoprotein E-deficient mice fed high-fat diet, and BHD was administered through gavage for 12 weeks at 8.4 g/kg/d and 16.8 g/kg/d. The atherosclerotic plaque size, composition, serum lipid profile, and inflammatory cytokines, were assessed. Mechanistically, metabolomic and microbiota profiles were analyzed by liquid chromatography-mass spectrometry and 16S rRNA gene sequencing, respectively. Furthermore, intestinal microbiota and atherosclerosis-related metabolic parameters were correlated using Spearman analysis. Atherosclerotic mice treated with BHD exhibited reduced plaque area, aortic lumen occlusion, and lipid accumulation in the aortic root. Nine perturbed serum metabolites were significantly restored along with the relative abundance of microbiota at the family and genus levels but not at the phylum level. Gut microbiome improvement was strongly negatively correlated with improved metabolite levels. BHD treatment effectively slows the progression of atherosclerosis by regulating altered intestinal microbiota and perturbed metabolites.

Degradation prediction for aerospace electronic systems plays a crucial role in maintenance work. This paper proposes a concise and efficient framework for multivariate time series forecasting that is capable of handling diverse sequence representations through a Channel-Independent (CI) strategy. This framework integrates a decomposition-aware layer to effectively separate and fuse global trends and local variations and a temporal attention module to capture temporal dependencies dynamically. This design enables the model to process multiple distinct sequences independently while maintaining the flexibility to learn shared patterns across channels. Additionally, the framework incorporates probabilistic distribution forecasting using likelihood functions, addressing the dynamic variations and uncertainty in time series data. The experimental results on multiple real-world datasets validate the framework’s effectiveness, demonstrating its robustness and adaptability in handling diverse sequences across various application scenarios.

Sakuranetin, a flavonoid phytoalexin in rice, plays a crucial role in defense against pathogen infection. While MYB-type transcription factors are well-known to regulate plant growth, development, secondary metabolism, and adaptation to environmental stresses, the function of rice MYB-related transcription factors in sakuranetin biosynthesis and sakuranetin-mediated defense remains unclear. In this study, we identified and characterized OsMYB1R, a novel single repeat MYB transcription factor that acts as a transcriptional activator in sakuranetin biosynthesis. Protein-DNA binding and activation assays revealed that OsMYB1R directly regulates the gene promoter of OsNOMT, a key enzyme in sakuranetin synthesis. Molecular analyses and infection studies using OsMYB1R-overexpressing (OsMYB1R-OE) and OsMYB1R-knockout (Osmyb1r, generated using CRISPR/Cas9) plants demonstrated that OsMYB1R increases sakuranetin production and decreases Magnaporthe oryzae infection by transcriptionally regulating OsNOMT expression. This finding indicates a positive regulation of sakuranetin biosynthesis and antifungal resistance by the OsMYB1R-OsNOMT crosstalk. Interestingly, the alteration of OsMYB1R expression did not affect yield-related agronomic traits. Our results reveal a novel and positive role of 1R-MYB in secondary metabolite biosynthesis and pathogen defense, suggesting that OsMYB1R is a potential gene for effectively enhancing rice resistance without compromising yield.

BackgroundImatinib is the most widely used tyrosine kinase inhibitor (TKI) in patients with newly diagnosed chronic-phase chronic myeloid leukemia(CML-CP). However, failure to achieve optimal response after imatinib administration, and subsequent switch to second-generation TKI therapy results in poor efficacy and induces drug resistance. In the present study, we developed and validated a nomogram to predict the efficacy of imatinib in the treatment of patients newly diagnosed with CML-CP in order to help clinicians truly select patients who need 2nd generation TKI during initial therapy and to supplement the risk score system.MethodsWe retrospectively analyzed 156 patients newly diagnosed with CML-CP who met the inclusion criteria and were treated with imatinib at the Second Affiliated Hospital of Xi’an Jiao Tong University from January 2012 to June 2022. The patients were divided into a poor-response cohort (N = 60)and an optimal-response cohort (N = 43) based on whether they achieved major molecular remission (MMR) after 12 months of imatinib treatment. Using univariate and multivariate logistic regression analyses, we developed a chronic myeloid leukemia imatinib-poor treatment (CML-IMP) prognostic model using a nomogram considering characteristics like age, sex, HBG, splenic size, and ALP. The CML-IMP model was internally validated and compared with Sokal, Euro, EUTOS, and ELTS scores.ResultsThe area under the curve of the receiver operator characteristic curve (AUC)of 0.851 (95% CI 0.778–0.925) indicated satisfactory discriminatory ability of the nomogram. The calibration plot shows good consistency between the predicted and actual observations. The net reclassification index (NRI), continuous NRI value, and the integrated discrimination improvement (IDI) showed that the nomogram exhibited superior predictive performance compared to the Sokal, EUTOS, Euro, and ELTS scores (P < 0.05). Magnetic metals have attracted extensive attention in microwave absorption for comprehensive merits of high magnetic loss and suitability for mass productions. Nevertheless, the applications have been limited by shortcomings of high conductivity and inadequate impedance matching. In this study, we have successfully synthesized a biphasic magnetic Sm2O3@Co composite through a facile one-step precipitation method. The resultant features a well-designed structure with spherical cores and flaky shells, exhibiting highly-tunable absorption performance over 2–18 GHz. Particularly, under 20 wt% filling ratio with paraffin, the Sm2O3@Co composite achieved minimum reflection loss (RLmin) of −30.1 dB and effective absorbing bandwidth (EAB) of 6.22 GHz at only 1.7 mm. The dielectric and magnetic losses could be easily modulated by simply changing the induced Co ratios. Through a systematic study, the microwave absorption mechanisms of the Sm2O3@Co magnetic composite have been comprehensively elucidated. This research offers a fresh perspective on developing highly efficient electromagnetic wave absorbents based on traditional magnetic metals.

As renewable energy continues to grow, more inverter-based distributed generators are being integrated into power grids, in which virtual synchronous generator (VSG) is widely adopted to regulate system voltage and stabilize frequency. However, grid voltage imbalances can cause serious oscillations in active power and system frequency, thus severely limiting the effectiveness of VSGs. Consequently, this study proposes an adaptive control strategy to enhance the dynamic performance of VSG under an imbalanced grid voltage. Compared to conventional VSG control methods, the proposed approach enables dynamic adjustment of the inertia and damping coefficients based on the output angular frequency oscillations, so as to shorten the settling time and reduce active power overshoot. The error-driven adaptive mechanism forms a closed-loop control, which effectively keeps the system stable regardless of parameter variations. Additionally, it combines with balanced output current (BOC), constant active power (CAP), and constant reactive power (CRP) control modes to further improve effectiveness. Finally, verifications have been conducted in the MATLAB/SIMULINK and hardware-in-the-loop (HIL) environment to demonstrate the superiority and feasibility of the proposed method.

Hot water extractable organic carbon (HWOC), the labile carbon component, is often used to indicate soil organic carbon (SOC) dynamics. Nevertheless, few studies have been carried out in arid climate areas which affects our full understanding of HWOC. Here, we investigated the change in HWOC in the topsoil of different ecosystems in the southern part of the Loess Plateau in the semiarid region of China and compared it with that in other regions. The HWOC concentrations of the study area (0-10 cm) were 0.27 ± 0.12 g C kg−1 and 0.19 ± 0.04 g C kg−1 in the natural and agricultural systems respectively, and the HWOC proportions were 1.38 ± 0.38% and 2.18 ± 0.22%. The HWOC concentration and proportion in the study area were much lower than the reported data in other areas, which may be affected by drought conditions. Irrigation could weaken the difference in HWOC between agricultural systems in different regions. Since HWOC is easily lost due to the impact of the arid climate, the soil carbon balance and carbon sequestration in arid and semiarid areas are relatively unstable, indicating that soil management should be improved in combination with water management.

The integration of robotics in the garment industry remains relatively limited, primarily due to the challenges in the highly deformable nature of garments. The objective of this study is thus to explore a vision-based garment recognition and environment reconstruction model to facilitate the application of robots in garment processing. Object SLAM (Simultaneous Localization and Mapping) was employed as the core methodology for real-time mapping and tracking. To enable garment detection and reconstruction, two datasets were created: a 2D garment image dataset for instance segmentation model training and a synthetic 3D mesh garment dataset to enhance the DeepSDF (Signed Distance Function) model for generative garment reconstruction. In addition to garment detection, the SLAM system was extended to identify and reconstruct environmental planes, using the CAPE (Cylinder and Plane Extraction) model. The implementation was tested using an Intel Realsense® camera, demonstrating the feasibility of simultaneous garment and plane detection and reconstruction. This study shows improved performance in garment recognition with the 2D instance segmentation models and an enhanced understanding of garment shapes and structures with the DeepSDF model. The integration of CAPE plane detection with SLAM allows for more robust environment reconstruction that is capable of handling multiple objects. The implementation and evaluation of the system highlight its potential for enhancing automation and efficiency in the garment processing industry.

To further understand the influence of n-heptane on the ignition process of ammonia, an isotope labeling method was applied in the current investigation to decouple the influence of the chemical effect, the thermal effect, and the effect of O radical from the oxidation of n-heptane on the ignition delay times (IDTs) of ammonia. An analysis of the time evolution of fuel, analysis of the time evolution of temperature, rate of consumption and production (ROP) analysis, and sensitivity analysis were conducted to gain a further understanding of the mechanism of the influence of the chemical effect, the thermal effect, and the effect of O radical on the ignition of ammonia. The results showed that the negative temperature coefficient (NTC) behavior of n-heptane is mitigated by the blending of ammonia, and this mitigated effect of ammonia is mainly due to the chemical effect. The IDTs of ammonia under low and medium temperatures are significantly shortened by the chemical effect at a n-heptane mass fraction of 10%. The promoting effect of the chemical effect decreases when the n-heptane mass fraction increases. The time evolution of n-heptane for NC7H16/ND3-G can be classified into three stages at 800 K, and the rapid consumption stage is mitigated by an increase in temperature. The rapid consumption stage is suppressed by the chemical effect of ammonia, while O radical has a promoting effect on the rapid consumption stage. The chemical effect will enhance the sensitivities of reactions associated with ammonia. As the n-heptane mass fraction increases, the sensitivities of reactions associated with n-heptane are enhanced. Correspondingly, the effect of reactions associated with ammonia is weakened. When the n-heptane mass fraction is 30%, only reactions related to n-heptane have a great influence on the ignition of ammonia/n-heptane fuel blends under the thermal effect + the effect of O radical or only the thermal effect.

This study focuses on 19 provinces in the Yangtze River Basin of China. It gathers relevant data indicators from 2010 to 2021 and constructs an evaluation index system centered on agricultural science and technology innovation. The study evaluates the relationship between agricultural “science and technology innovation-economy-ecology” systems and identifies key obstacle factors using the obstacle degree model. The study draws the following conclusions: Firstly, the comprehensive development level index of the agricultural science and technology innovation system shows an overall linear upward trend (values range from 0.121 to 0.382). Secondly, the comprehensive development level index of the agricultural economic system exhibits an upward trend but with a relatively small overall magnitude (values range from 0.248 to 0.322). Thirdly, the comprehensive development level index of the agricultural ecological system demonstrates significant overall fluctuations, with notable regional disparities (values range from 0.384 to 0.414). Fourthly, the overall agricultural SEE (Science and technological innovation, Economy, Ecology) complex system exhibits a characteristic of “high coupling, low coordination”, identifying the main obstacle factors influencing agricultural SEECS based on a formulated approach. Subsequently, the following policy recommendations are proposed: Firstly, enhance the agricultural technological innovation system and promote green and efficient agricultural technology research and development. Secondly, to accelerate the transformation and upgrading of modern agriculture, achieving green and high-quality development of the agricultural economy. Thirdly, to strengthen agricultural ecological environment protection, laying a solid foundation for the healthy and sustainable development of agriculture.

Estimating the 6D pose and size of objects is crucial in the task of visual grasping for robotic arms. Most current algorithms still require the 3D CAD model of the target object to match with the detected points, and they are unable to predict the object’s size, which significantly limits the generalizability of these methods. In this paper, we introduce category priors and extract high-dimensional abstract features from both the observed point cloud and the prior to predict the deformation matrix of the reconstructed point cloud and the dense correspondence between the reconstructed and observed point clouds. Furthermore, we propose a staged geometric correction and dense correspondence refinement mechanism to enhance the accuracy of regression. In addition, a novel lightweight attention module is introduced to further integrate the extracted features and identify potential correlations between the observed point cloud and the category prior. Ultimately, the object’s translation, rotation, and size are obtained by mapping the reconstructed point cloud to a normalized canonical coordinate system. Through extensive experiments, we demonstrate that our algorithm outperforms existing methods in terms of performance and accuracy on commonly used benchmarks for this type of problem. Additionally, we implement the algorithm in robotic arm-grasping simulations, further validating its effectiveness.

Interface dislocation networks have a great influence on the mechanical properties of the new Ni-based single-crystal alloy (NSC) containing Re, but it is difficult to find out the structural evolution behaviors at the micro-level. Thus, molecular dynamics (MD) simulation is used to analyze the atomic potential energy change and dislocation evolution mechanism, and non-linear characteristic parameters are used to analyze the microstructure evolution of NSC. First, a new model of Ni-Al-Re that is closer to the real properties of the material is established using the MD method according to the optimal volume ratio of matrix phase to precipitate phase. Then, the MD models of NSC with different contents of Re are calculated and analyzed under compressive and tensile loads. The results show that with an increase in Re atoms, the atomic potential energy at the interface dislocation networks is reduced; thus, the stability of the system is enhanced, and the hindrance of the interface dislocation networks to the dislocation movement of the matrix phase is strengthened. At the same time, the number of HCP structures and OISs formed by the destruction of the intact FCC structures also decreases. In the non-linear ultrasonic experiment, with the increase in Re atoms, the non-linear enhancement of the microstructure of the NSC leads to an increase in the corresponding non-linear characteristic parameters. Accordingly, the microstructural evolution behaviors of the phase interface of the new NSC can be effectively explored using the combination of MD simulation and non-linear ultrasonic experimentation. The results of this study lay a foundation for the subsequent research of the microscopic defects of NSCs by using ultrasonic phased-array technology.

Walking robots have been widely applied in complex terrains due to their good terrain adaptability and trafficability. However, in some environments (such as disaster relief, field navigation, etc.), although a single strategy can adapt to various environments, it is unable to strike a balance between speed and stability. Existing control schemes like model predictive control (MPC) and traditional incremental control can manage certain environments. However, they often cannot balance speed and stability well. These methods usually rely on a single strategy and lack adaptability for dynamic adjustment to different terrains. To address this limitation, this paper proposes an innovative double-layer reinforcement learning algorithm. This algorithm combines Deep Double Q-Network (DDQN) and Proximal Policy Optimization (PPO), leveraging their complementary strengths to achieve both fast adaptation and high stability in complex terrains. This algorithm utilizes terrain information and the robot’s state as observations, determines the walking speed command of the quadruped robot Unitree Go1 through DDQN, and dynamically adjusts the current walking speed in complex terrains based on the robot action control system of PPO. The speed command serves as a crucial link between the robot’s perception and movement, guiding how fast the robot should walk depending on the environment and its internal state. By using DDQN, the algorithm ensures that the robot can set an appropriate speed based on what it observes, such as changes in terrain or obstacles. PPO then executes this speed, allowing the robot to navigate in real time over difficult or uneven surfaces, ensuring smooth and stable movement. Then, the proposed model is verified in detail in Isaac Gym. Wecompare the distances walked by the robot using six different control methods within 10 s. The experimental results indicate that the method proposed in this paper demonstrates excellent speed adjustment ability in complex terrains. On the designed test route, the quadruped robot Unitree Go1 can not only maintain a high walking speed but also maintain a high degree of stability when switching between different terrains. Ouralgorithm helps the robot walk 25.5 m in 10 s, outperforming other methods.

Abstract Background Dietary intake has been shown to have a causal relationship with various lung diseases, such as lung cancer and asthma. However, the causal relationship between dietary intake and idiopathic pulmonary fibrosis (IPF) remains unclear. We conducted a two-sample Mendelian Randomization (MR) study to investigate the causal relationship between dietary intake and IPF. Methods The exposure datasets included meat, fruit, vegetable, and beverage intake from the UK Biobank. IPF data came from the EBI database of 451,025 individuals. All data in this study were obtained from the IEU Open GWAS Project. The inverse variance weighted (IVW), MR-Egger, and weighted median methods were used as the primary methods. Sensitivity analyses were performed to ensure the validity of the results. Results Oily fish intake [odds ratio (OR):0.995; 95% confidence interval (CI): 0.993–0.998; p = 6.458E-05] and Dried fruit intake (OR:0.995;95%CI:0.991–0.998; p = 0.001) were discovered as protective factors. There was also a suggestive correlation between Beef intake (OR:1.006;95%Cl:1.001–1.012; p = 0.023) and IPF. Sensitivity analysis did not reveal any contradictory results. No causal relationship was found between IPF and the rest of the dietary exposures. Conclusions Our study found that Oily fish and Dried fruit intake were associated with the risk of IPF, while Beef intake was suggestively associated with the risk of IPF. Other studies are still needed to confirm the results in the future.

BackgroundRecent studies have demonstrated a strong association between acute kidney injury (AKI) and chronic kidney disease (CKD), while the unresolved inflammation is believed to be a driving force for this chronic transition process. As a transmembrane pattern recognition receptor, Mincle (macrophage-inducible C-type lectin, Clec4e) was identified to participate in the early immune response after AKI. However, the impact of Mincle on the chronic transition of AKI remains largely unclear.MethodsWe performed single-cell RNA sequencing (scRNA-seq) with the unilateral ischemia-reperfusion (UIR) murine model of AKI at days 1, 3, 14 and 28 after injury. Potential effects and mechanism of Mincle on renal inflammation and fibrosis were further validated in vivo utilizing Mincle knockout mice.ResultsThe dynamic expression of Mincle in macrophages and neutrophils throughout the transition from AKI to CKD was observed. For both cell types, Mincle expression was significantly up-regulated on day 1 following AKI, with a second rise observed on day 14. Notably, we identified distinct subclusters of Minclehigh neutrophils and Minclehigh macrophages that exhibited time-dependent influx with dual peaks characterized with remarkable pro-inflammatory and pro-fibrotic functions. Moreover, we identified that Minclehigh neutrophils represented an “aged” mature neutrophil subset derived from the “fresh” mature neutrophil cluster in kidney. Additionally, we observed a synergistic mechanism whereby Mincle-expressing macrophages and neutrophils sustained renal inflammation by tumor necrosis factor (TNF) production. Mincle-deficient mice exhibited reduced renal injury and fibrosis following AKI.ConclusionThe present findings have unveiled combined persistence of Minclehigh neutrophils and macrophages during AKI-to-CKD transition, contributing to unresolved inflammation followed by fibrosis via TNF-α as a central pro-inflammatory cytokine. Targeting Mincle may offer a novel therapeutic strategy for preventing the transition from AKI to CKD.

Abstract A great challenge for electrochemical CO2 reduction is to improve energy efficiency, which requires reducing overpotential while increasing product Faraday efficiency. Here, we designedly synthesize a hybrid electrocatalyst consisting of Fe nanoparticles, pyrrole-type Fe-N4 sites and less-oxygenated carbon supports, which exhibits a remarkable CO Faraday efficiency above 99% at an ultralow overpotential of 21 mV, reaching the highest cathode energy efficiency of 97.1% to date. The catalyst also can afford a CO selectivity nearly 100% with a high cathode energy efficiency (>90%) at least 100 h. The combined results of control experiments, in situ characterizations and theoretical calculations demonstrate that introducing Fe nanoparticles can reduce the overpotential by accelerating the proton transfer from CO2 to *COOH and lowering the free energy for *COOH formation, constructing pyrrole-type Fe-N4 sites and limiting oxygen species on carbon supports can increase CO Faraday efficiency through inhibiting the H2 evolution, thus achieving energy-efficient electrochemical CO2 reduction to CO.

Abstract Objective This study aimed to investigate the clinical effects of recombinant human interleukin-11 (rhIL-11) gargle on preventing and treating oral mucositis (OM) after chemotherapy for acute leukemia. Methods This single-site, prospective, observer-blinded, nonrandomized controlled trial was conducted on 74 patients with acute leukemia, who were divided into the experimental and control groups. The patients in the experimental group were treated with IL-11 gargle, and those in the control group were treated with sodium bicarbonate gargle. We examined the time and severity of oral mucositis, severity and duration of associated pain, healing time of mucositis, effects of OM on eating, and levels of T-cell subset indicators before and after treatment to evaluate the effects of IL-11 treatment. Results The proportion of patients with severe OM was significantly lower in the experimental group than in the control group. Mucositis occurred later in the experimental group compared with the control group. The degree and duration of pain, ulcer healing time, and effects on eating were lower in the experimental group compared with the control group. Following treatment, the levels of all T-cell subset indicators improved in each of the two groups. Gut microbial "dark matter" refers to the microorganism that exists in low or rare abundance in the gastrointestinal tract and affects human health, without a clear disclosure of its influence and the related mechanism. Most current studies on the relationship among diet, gut microbiota, and host health have focused on the high-abundance gut microbiota, while the role of gut microbial "dark matter" has often been neglected. Therefore, this paper reviews the interactions among dietary nutrition, gut microbial "dark matter", and host health, as well as the new advances in microbiomics technologies, discusses the importance of exploring the acting mechanisms of microbial "dark matter" on human health, and provides new ideas for future dietary interventions to influence gut microbiota for preventing or even improving human diseases.

Abstract Background Pathogen infections seriously affect host health, and the use of antibiotics increases the risk of the emergence of drug-resistant bacteria and also increases environmental and health safety risks. Probiotics have received much attention for their excellent ability to prevent pathogen infections. Particularly, explaining mechanism of action of probiotics against pathogen infections is important for more efficient and rational use of probiotics and the maintenance of host health. Results Here, we describe the impacts of probiotic on host resistance to pathogen infections. Our findings revealed that (I) the protective effect of oral supplementation with B. velezensis against Aeromonas hydrophila infection was dependent on gut microbiota, specially the anaerobic indigenous gut microbe Cetobacterium; (II) Cetobacterium was a sensor of health, especially for fish infected with pathogenic bacteria; (III) the genome resolved the ability of Cetobacterium somerae CS2105-BJ to synthesize vitamin B12 de novo, while in vivo and in vitro metabolism assays also showed the ability of Cetobacterium somerae CS2105-BJ to produce vitamin B12; (IV) the addition of vitamin B12 significantly altered the gut redox status and the gut microbiome structure and function, and then improved the stability of the gut microbial ecological network, and enhanced the gut barrier tight junctions to prevent the pathogen infection. Conclusion Collectively, this study found that the effect of probiotics in enhancing host resistance to pathogen infections depended on function of B12 produced by an anaerobic indigenous gut microbe, Cetobacterium. Furthermore, as a gut microbial regulator, B12 exhibited the ability to strengthen the interactions within gut microbiota and gut barrier tight junctions, thereby improving host resistance against pathogen infection. As technology rapidly evolves, the application of bipedal robots in various environments has widely expanded. These robots, compared to their wheeled counterparts, exhibit a greater degree of freedom and a higher complexity in control, making the challenge of maintaining balance and stability under changing wind speeds particularly intricate. Overcoming this challenge is critical as it enables bipedal robots to sustain more stable gaits during outdoor tasks, thereby increasing safety and enhancing operational efficiency in outdoor settings. To transcend the constraints of existing methodologies, this research introduces an adaptive bio-inspired exploration framework for bipedal robots facing wind disturbances, which is based on the Deep Deterministic Policy Gradient (DDPG) approach. This framework allows the robots to perceive their bodily states through wind force inputs and adaptively modify their exploration coefficients. Additionally, to address the convergence challenges posed by sparse rewards, this study incorporates Hindsight Experience Replay (HER) and a reward-reshaping strategy to provide safer and more effective training guidance for the agents. Simulation outcomes reveal that robots utilizing this advanced method can more swiftly explore behaviors that contribute to stability in complex conditions, and demonstrate improvements in training speed and walking distance over traditional DDPG algorithms.

Background: This study aimed to examine the epidemiological characteristics of imported infections and assess the effectiveness of border health screening in detecting imported diseases. Methods: We obtained infection data for 2016 to 2019 from the Fuzhou Changle International Airport Infection Reporting System. The demographic, temporal, and spatial characteristics of travel-related infections were analyzed using r×c contingency tables, the Cochran–Armitage trend test, and seasonal-trend decomposition using LOESS (STL). Detection rates were used as a proxy for the effectiveness of border health-screening measures. Results: Overall, 559 travel-related infections were identified during the study period, with 94.3% being imported infections. Airport health screening demonstrated an overall effectiveness of 23.7% in identifying travel-associated infections. Imported infections were predominantly identified in males, with 55.8% of cases occurring in individuals aged 20–49. The peak periods of infection importation were from January to February and from May to August. The infectious diseases identified were imported from 25 different countries and regions. All dengue fever cases were imported from Southeast Asia. Most notifiable infections (76.0%) were identified through fever screening at the airport. Conclusion: The increasing number of imported infections poses a growing challenge for public health systems. Multifaceted efforts including surveillance, vaccination, international collaboration, and public awareness are required to mitigate the importation and spread of infectious diseases from overseas sources.

Magnetic metal absorbing materials have exhibited excellent absorptance performance. However, their applications are still limited in terms of light weight, low thickness and wide absorption bandwidth. To address this challenge, we design a broadband and low-profile multilayer absorber using cobalt–iron (CoFe) alloys doped with rare earth elements (REEs) lanthanum (La) and Neodymium (Nd). An improved estimation of distribution algorithm (IEDA) is employed in conjunction with a mathematical model of multilayer absorbing materials (MAMs) to optimize both the relative bandwidth with reflection loss (RL) below −10 dB and the thickness. Firstly, the absorption performance of CoFe alloys doped with La/Nd with different contents is analysed. Subsequently, IEDA is introduced based on a mathematical model to achieve an optimal MAM design that obtains a balance between absorption bandwidth and thickness. To validate the feasibility of our proposed method, a triple-layer MAM is designed and optimized to exhibit wide absorption bandwidth covering C, X, and Ku bands (6.16–12.82 GHz) and a total thickness of 2.39 mm. Then, the electromagnetic (EM) absorption mechanisms of the triple-layer MAMs are systematically investigated. Finally, the triple-layer sample is further fabricated and measured. The experimental result is in good agreement with the simulated result. This paper presents a rapid and efficient optimization method for designing MAMs, offering promising prospects in microwave applications, such as radar-stealth technology, EM shielding, and reduced EM pollution for electronic devices.

The ever-rising resistance in Botrytis cinerea has appeared as the awkward agricultural challenge that could be effectively resolved by developing novel fungicides featuring disparate action mechanisms. Aiming to explore novel fungicidal leads inhibiting B. cinerea, quinazolin-4(3H)-one bionic-alkaloids bearing an 1,3,4-oxadiazole fragment were conceived, synthesized, and systematically optimized under the guidance of anti-B. cinerea activities. The aforementioned optimization on molecular structures generated the anti-B. cinerea candidate I25 owning the promising in vitro EC50 value (0.76 μg/mL) that was fantastically superior to those of boscalid, penthiopyral, pyrimethanil and imazalil (0.86, 1.03, 15.91 and 2.15 µg/mL). Whereafter, the in vivo anti-B. cinerea preventative efficacy of an active molecule I25 was noticeably evaluated as 69.3 % at 200 µg/mL, which was megascopically better than that of boscalid (60.6 %). Furthermore, the preliminary investigation on action mechanisms indicated that the fungicidal molecule I25 could induce the conspicuous wrinkle on hyphal surfaces and increase the membrane permeability of B. cinerea cells. The above results have emerged as an imperative reference to developing the novel fungicides that could effectively control gray mold caused by B. cinerea.

Abstract The Kuroshio intrusion (KI) is a northwestward‐flowing branch of the Kuroshio Current, which enters the South China Sea (SCS) and regulates its temperature, salinity, and water mass exchanges. However, limited direct observations hinder our understanding of KI's mechanisms and responses to climate change. Here, we present a 60‐year bi‐monthly resolved coral oxygen isotope (δ18Oc) record from Dongsha Atoll, the northern SCS. The dry‐season (December–March) δ18Oc record reveals interannual to decadal variabilities of the KI. The impact of the East Asian winter monsoon (EAWM) on Dongsha δ18Oc was more pronounced during the 1970s and 1980s and after the early 2000s, while the influence of the El Niño‐Southern Oscillation (ENSO) on Dongsha δ18Oc was higher between the 1980s and 1990s. The Pacific Decadal Oscillation (PDO) may have a relatively minor effect on KI strength or may indirectly modulate KI strength through its influence on ENSO activities. Our Dongsha δ18Oc record highlight the importance of the EAWM, ENSO, and PDO in predicting future KI changes.

Abstract Background Standard treatment for drug-susceptible tuberculosis (DS-TB) includes a multidrug regimen requiring at least 6 months of treatment, and this lengthy treatment easily leads to poor adherence. There is an urgent need to simplify and shorten treatment regimens to reduce interruption and adverse event rates, improve compliance, and reduce costs. Methods ORIENT is a multicenter, randomized controlled, open-label, phase II/III, non-inferiority trial involving DS-TB patients to evaluate the safety and efficacy of short-term regimens compared with the standardized six-month treatment regimen. In stage 1, corresponding to a phase II trial, a total of 400 patients are randomly divided into four arms, stratified by site and the presence of lung cavitation. Investigational arms include 3 short-term regimens with rifapentine 10 mg/kg, 15 mg/kg, and 20 mg/kg, while the control arm uses the standardized six-month treatment regimen. A combination of rifapentine, isoniazid, pyrazinamide, and moxifloxacin is administered for 17 or 26 weeks in rifapentine arms, while a 26-week regimen containing rifampicin, isoniazid, pyrazinamide, and ethambutol is applied in the control arm. After the safety and preliminary effectiveness analysis of patients in stage 1, the control arm and the investigational arm meeting the conditions will enter into stage 2, which is equivalent to a phase III trial and will be expanded to recruit DS-TB patients. If all investigational arms do not meet the safety conditions, stage 2 will be canceled. In stage 1, the primary safety endpoint is permanent regimen discontinuation at 8 weeks after the first dose. The primary efficacy endpoint is the proportion of favorable outcomes at 78 weeks after the first dose for both two stages. Discussion This trial will contribute to the optimal dose of rifapentine in the Chinese population and suggest the feasibility of the short-course treatment regimen containing high-dose rifapentine and moxifloxacin for DS-TB. Trial registration The trial has been registered on ClinicalTrials.gov on 28 May 2022 with the identifier NCT05401071.

Abstract Chronic low-grade inflammation has been identified as a major contributor in the development of atherosclerosis. Nuclear Factor-κappa B (NF-κB) is a critical transcription factors family of the inflammatory pathway. As a major catalytic subunit of the IKK complex, IκB kinase β (IKKβ) drives canonical activation of NF-κB and is implicated in the link between inflammation and atherosclerosis, making it a promising therapeutic target. Various natural product derivatives, extracts, and synthetic, show anti-atherogenic potential by inhibiting IKKβ-mediated inflammation. This review focuses on the latest knowledge and current research landscape surrounding anti-atherosclerotic drugs that inhibit IKKβ. There will be more opportunities to fully understand the complex functions of IKKβ in atherogenesis and develop new effective therapies in the future.

Abstract Ultrafine particles, defined as particles with a diameter (dp) smaller than 100 nm, serve as an important component of cloud condensation nuclei, in addition to impacting human health. The dominant sources of ultrafine particles include traffic emissions and nucleation. Singapore is a tropical city that hosts petrochemical industries. To identify the sources of ultrafine particles, a year-long observation of the number size distribution was conducted in Singapore in 2018 and 2019. The concentrations of CO, CO2, CH4, and SO2 were also monitored. The particle number concentration during the southwest monsoon season was high, while that during the northeast monsoon period was relatively low. The CO concentration increased during the morning traffic rush hours, which was associated with relatively minor enhancements in ultrafine particle number concentration. The events for a high number concentration of the Aitken mode particles (dp < 50 nm) were identified during high SO2 concentration periods. The SO2 concentration was high during the afternoon because the sea breeze transported the emissions from the coastal industrial area to the observation site. The enhancements in CH4 from its background level (ΔCH4) and SO2 had a quasi-inverse relationship, as the major emission sources of these two chemical species were different. The particle number concentration (dp > 50 nm) correlated with the enhancements in CO concentration (ΔCO) for CH4-dominant air masses, suggesting that incomplete combustion processes, such as traffic emission, are important for the size range. Conversely, the number concentration of the Aitken mode particles (dp < 50 nm) increased for SO2-dominant air masses, suggesting the importance of industrial plume.

To gain an insight into the lattice and band structure of blue perovskite NC emitters, doping organic cations (FA+) reduces nonradiative recombination at room temperature. The FA+ significantly decreases exciton quenching and phonon energy scattering via manipulating hot carriers and electron filling in band structure.

The combined application of steel–FRP composite bars (SFCBs) and seawater sea-sand concrete (SSSC) in marine engineering not only solves the problem of resource scarcity and reduces the construction cost but also avoids the problems of chloride corrosion of steel reinforcement in seawater sea-sand concrete and the lack of ductility of FRP bars. At the same time, the addition of glass fiber (GF) and expansion agent (EA) in appropriate amounts improves the crack resistance and seepage resistance of concrete. However, the durability of SFCB with GF- and EA-reinforced SSSC in freezing–thawing environment remains unclear, which limits its potential application in cryogenic marine engineering. This study investigates the bonding properties between SFCB and GF-EA-SSSC interfaces using eccentric pullout experiments under different thicknesses of concrete protective cover and a number of freezing–thawing cycles. The results showed that the compressive strength and dynamic elastic modulus of SSSC decrease, while the mass loss increases with an increasing number of freezing–thawing cycles. Additionally, the bond strength and stiffness between SFCB and SSSC decrease, leading to an increase in relative slip. However, the rate of bond strength and stiffness loss decreases with an increase in the thickness of the concrete protective cover. Furthermore, formulas for bond strength, relative slip, and bond stiffness are established to quantify the effects of the thickness of the concrete protective cover and the number of freezing–thawing cycles. The experimental values obtained verify the accuracy of these formulas, with a relative error of less than 5%. Moreover, a bond stress–slip constitutive model is developed for SFCB and GF-EA-SSSC, and the fitting results closely resemble the experimental values, demonstrating a high level of model fit.

Intestinal barrier hyperpermeability, which is characterised by impaired tight junction proteins, is associated with a variety of gastrointestinal and systemic diseases. Therefore, maintaining intestinal barrier integrity is considered one of the effective strategies to reduce the risk of such disorders. This study aims to investigate the potential benefits of two probiotic strains (Lactiplantibacillus plantarum ST-III and Lacticaseibacillus rhamnosus KF7) on intestinal barrier function by using a physiologically relevant in vitro model of the intestinal epithelium. Our results demonstrate that both strains increased transepithelial electrical resistance, a measure of intestinal barrier integrity. Immunolocalisation studies indicated that this improvement in barrier function was not due to changes in the co-localisation of the tight junction (TJ) proteins ZO-1 and occludin. However, we observed several modifications in TJ-related genes in response to the probiotics, including the upregulation of transmembrane and cytosolic TJ proteins, as well as TJ signalling proteins. Gene expression modulation was strain- and time-dependent, with a greater number of differentially expressed genes and higher fold-change being observed in the L. plantarum ST-III group and at the latter timepoint. Further studies to investigate how the observed gene expression changes can lead to enhanced barrier function will aid in the development of probiotic foods to help improve intestinal barrier function.

Meat and meat products are of high nutritional values and are the main source of high-quality proteins for human beings. However, meat and meat products are susceptible to contaminate with various microorganisms during processing and storage, leading to meat spoilage and even foodborne diseases. High pressure carbon dioxide (HPCD) is a novel non-thermal processing technology. It can achieve significant antimicrobial effect under relatively mild pressure and temperature conditions. This paper summarized the inactivation mechanism of HPCD, and reviewed the application of HPCD in meat pasteurization and preservation in the past years. The effects of HPCD separately and its combination with other methods on antimicrobial activity, physiochemical characteristics, texture properties, and microbial stability of meat and meat products were also described in detail. In view of the existing problems, this paper also looked into the development prospect of HPCD technology.

ABSTRACT Prediction of susceptibility to pyrazinamide (PZA) directly from sputum has been challenging. The MeltPro MTB/PZA assay, based on melting curve analysis, can simultaneously detect Mycobacterium tuberculosis and the resistance to PZA from sputum. We aimed to evaluate the MeltPro MTB/PZA assay to predict PZA resistance among rifampin-resistant tuberculosis (RR-TB) patients. We prospectively enrolled RR-TB patients in the registered trials, and their baseline sputum samples were obtained to perform the assay and culture. DNA sequencing of culture isolates was analyzed and used as the reference standard. Sanger sequencing was performed for samples with discrepant results between next-generation sequencing (NGS) and the investigational assay. The main analysis was conducted in the population of patients with interpretable results by both NGS and the assay. A total of 239 patients with RR-TB were screened, and 220 underwent the MeltPro MTB/PZA assay. The assay provided no information for 25 of 220 patients (11.4%). Among the remaining 195 patients, 13 had negative culture or insufficient raw NGS sequencing data, and 15 had indeterminate assay results. A total of 167 patients were included in the main analysis. Against DNA sequencing, the sensitivity, specificity, and negative predictive value of the assay for detecting resistance to PZA were 91.4% (95% confidence interval [CI], 87.1% to 95.6%), 89.9% (95% CI, 85.3% to 94.5%), and 95.2% (95% CI, 91.9% to 98.4%), respectively. In conclusion, the MeltPro MTB/PZA assay is a fast semiautomatic molecular platform to rapidly predict resistance to PZA from sputum and holds promise as a screening tool with satisfactory sensitivity. IMPORTANCE This study evaluated the accuracy of the MeltPro MTB/PZA assay at detecting the presence of PZA resistance through registered clinical trials. Compared to DNA sequencing, the assay had high sensitivity and negative predictive value, suggesting its potential utility as a screening tool in clinical practice. The assay could serve as an ideal primary screening tool in low PZA-resistant M. tuberculosis prevalence settings and could be used as an additional test to identify PZA resistance rapidly and initially in the RR-TB population.

Abstract Manipulating Gilbert damping is of critical importance in spintronic devices. Here, this work reports the damping anisotropy of Heusler‐alloy Co3−xFexAl single crystalline thin films using inductive ferromagnetic resonance. A colossal Gilbert damping anisotropy ratio of up to 1400%, nearly three times higher than the highest value reported among known metallic ferromagnets, is observed in the optimized sample Co1.4Fe1.6Al. The Gilbert damping anisotropy with strong fourfold symmetry is attributed to the variation of the spin–orbit coupling, which is further corroborated by the current‐orientation‐dependent anisotropic magnetoresistance ratio in Co1.4Fe1.6Al thin film. These results provide a practical approach to adjust Gilbert damping continuously and show great promise for future spintronic applications.
