There were no previous studies investigating the impact of ASXL1 mutations in Ph-negative MPNs in Turkey. In the current study, we investigated the prognostic significance of ASXL1 mutations in Turkish MPN patients. We also aimed to determine the prognostic significance of JAK2V617F allele burden and the relationship of JAK2V617F mutation with ASXL1 mutations in Ph-negative MPNs. Methods: About 184 patients from a single center diagnosed with Ph-negative MPNs were screened for ASXL1, JAK2V617F mutations, and JAK2V617F allele burden: 107 ET and 77 PMF. Results: A total of 29 ASXL1 mutations were detected in 24.7% of PMF and 8.4% of ET patients. ASXL1-mutated ET patients showed a trend toward an increase in the incidence of cerebrovascular events and higher total leukocyte counts. ASXL1-mutation in PMF was associated with older age and a higher prevalence of bleeding complications. In univariate analysis, overall survival (OS) was significantly reduced in ASXL1-mutated PMF patients. In multivariate analysis, Dynamic International Prognostic Scoring System-plus high-risk category and ASXL1 mutation status were independently associated with shorter survival in PMF. In PMF, mutational status and allele burden of JAK2V617F showed no difference in terms of OS and leukemia-free survival. Conclusion: We conclude that ASXL1 mutations are molecular predictors of short OS in PMF. In recent years, the occurrence of forest fires has become almost inevitable worldwide due to the severe impact of climate change. Consequently, it is imperative to examine the consequences of these fires, pinpoint the affected areas, and take required measures following post-fire incidents. With the advent of modern remote sensing satellites, which allow for the rapid monitoring of vast expanses, they offer a valuable data source for addressing such emergency scenarios. Therefore, the main objective of this study is to evaluate the extent of air pollutant gas emissions resulting from the forest fires that occurred in Marmaris and Manavgat, Türkiye, in the summer of 2021. To this aim, using the data obtained from the TROPOspheric Monitoring Instrument (TROPOMI) sensor onboard the Sentinel-5P satellite, the amount of major air pollutants nitrogen dioxide (NO2), carbon monoxide (CO) and aerosol were investigated after the forest fires considering three different non-fire time periods. The findings of the experiments indicated that in Manavgat, there was a substantial rise in NO2 and average CO levels by 260.43% and 107.07%, respectively, when compared to the 10-day period preceding the forest fire event. Similarly, in Marmaris, there was an increase of 203.63% in NO2 levels and a 102.47% rise in average CO levels during the same period. Positive absorbing aerosol index (AAI) values were also observed during the events, which means that the amount of UV-absorbing aerosols increased due to the fire. The differenced Normalized Burn Ratio (dNBR) maps derived from the Sentinel-2 MSI imagery were also used to investigate the severity of the forest fires, and to observe the relationship between the fire severity level and the air pollutants investigated. Objective: Amikacin (AK) is a wide-spectrum antibiotic routinely used to treat gram-negative and some gram-positive bacterial infections. However, its use is limited due to its potential to cause nephrotoxicity due to an increase in reactive oxygen radicals. The main goal of this study was to investigate the effect of carvacrol (CAR) on AK-induced nephrotoxicity in rats. Methods: Thirty-two Sprague Dawley rats were randomly separated into four groups: the control (0.9% NaCl solution and sunflower oil), AK (400 mg/kg), CAR+AK (80 mg/kg CAR+400 mg/kg AK), and AK+CAR (400 mg/kg AK+80 mg/kg CAR) groups. AK and CAR were administered intramuscularly and orally, respectively for 7 days. Blood and kidney tissue samples were collected at the end of the experiment. The level of catalase, superoxide dismutase, malondialdehyde, and reduced glutathione, which are parameters of oxidative stress, were detected while comparing renal function and histopathological changes. Results: Histopathological findings (necrotic changes, dilatation and inflammatory cell infiltration) were significantly greater in the AK group than in the control group. OBJECTIVE: In patients with acute myocardial infarction, coronary collateral circulation (CCC) is associated with reduced infarct size, preserved cardiac function, and decreased mortality. An interarm blood pressure difference (IABPD) is shown to be independently associated with cardiovascular and all-cause of mortality. We aimed to determine the effect of IABPD on coronary collateral flow in patients with ST-segment elevation myocardial infarction (STEMI) who had undergone primary percutaneous coronary intervention (p-PCI). PATIENTS AND METHODS: We prospectively investigated 1,348 consecutive patients who were hospitalized for STEMI and underwent p-PCI. The Rentrop classification was used to assess CCC. According to this classification, we defined Rentrop 0 and 1 as poor CCC, and Rentrop 2 and 3 as good CCC. A 10 mm Hg difference is considered the upper limit of IABPD. RESULTS: Patients were divided into two groups according to the collateral circulation, 325 patients (24%) had good collateral, while 1,023 patients (76%) had poor collateral. IABPD was significantly higher in the poor collateral group (57 patients, 5.6%) than in the good collateral group (9 patients, 2.8%) (p=0.04). Pre-infarction angina and IABPD were identified as independent predictors of poor collateral (OR: 0.516, 95% CI 0.370-0.631, p=0.007; OR: 3.681, 95% CI: 1.773-7.461, p=0.01, respectively) in multivariate analysis. CONCLUSIONS: The IABPD was shown as an independent predictor of poor collateral circulation in patients with STEMI who underwent p-PCI. [ABSTRACT FROM AUTHOR]

The main aim of this study was to investigate the effects of an inactivated SARS-CoV-2 virus vaccine (Vero cells) on renal function in Balb/C Albino mice.Methods: 21 healthy, 6– 8 week old BALB/c male mice were divided into three equal groups, and 0.10 mL of intramuscular saline equal to the vaccine dose volume was administered to the first group. To the second group, a single dose of 0.10 mL 120 U of Vero cell inactive SARS COV-2 vaccine was administered intramuscularly. Group 3 received two consecutive doses of 0.10 mL 120 U intramuscular Vero cell inactive SARS COV-2 vaccine, 14 days apart. After administration, the clinical status, fecal and urine status, nutritional status and kidney histopathology of the mice were evaluated.Results: It was determined that no acute toxic symptoms were observed in the mice administered the vaccine, they were in good condition, and there was no significant stimulatory reaction related to the vaccine in the tissues of the injected local area. There was no difference in feed consumption, water consumption, and body weight gains between the control group, the groups that received a single dose of vaccine, and the groups that received two doses of vaccine (p> 0.05). No difference was found between the groups when urine and feces amounts were compared (p> 0.05). No difference was found between the groups when urinary urea, creatinine, and serum BUN, creatinine levels were compared (p> 0.05). No difference was found in the histopathological evaluation of the kidneys between the groups (p> 0.05).Conclusion: In conclusion, single or repeated injections of the SARS-CoV-2 vaccine (Vero cells) into mice were found to have no adverse effects on the animals’ overall clinical health, performance abilities and kidneys.Keywords: COVID-19, vaccine, SARS CoV-2, adverse effect, kidney

Members of the genus Ortholinea are among the worldwide distributed myxozoan parasites that mainly infect marine fish. In this study, a new myxosporean species, Ortholinea hamsiensis n. sp., was isolated from the urinary bladder of European anchovy Engraulis engrasicolus collected from the Sinop coasts of the Black Sea. The prevalence and density values of infection were 1.4% and 1–5 individuals in the field of view (1 + ), respectively. Mature myxospores are subspherical with slight tapering down to the less pronounced tip in the frontal view and subspherical in the sutural view. Myxospores measured 9.1 ± 0.25 (8.8–9.9) μm in length, 9.2 ± 0.11 (8.9–9.4) μm in thickness, and 8.4 ± 0.33 (8.2-9.1) μm in width. Two polar capsules equal in size measured 3.1 ± 0.11 (3.0–3.3) μm in length and 2.7 ± 0.11 (2.6–2.9) μm in width. The polar tubule had 3–4 coils. Along with morphological peculiarities, the results of the 18S rDNA also revealed it to be a new species for science compared to the other species of the genus. In this study, another myxosporean species O. gobiusi was also detected in round goby Neogobius melanostomus with a prevalence of infection value of 4.8% and a density of 1–5 individuals in the field of view (1 + ). The present study also provided the first data of 18S rDNA of O. gobiusi from N. melanostomus and type species of the genus O. divergens from Gobius niger and the phylogenetic relationships of these species with other Ortholinea species have been revealed.
