Abstract

Colorectal cancer (CRC) has become a serious threat to human life and health. Most patients are diagnosed at the late stage of advanced CRC, resulting in losing their best opportunity for surgical treatment. Chemotherapy plays a crucial role in the control and treatment of advanced CRC. However, the cytotoxicity of chemotherapeutic drugs can easily cause the imbalance of gut flora, damage the barrier of the gastrointestinal mucosa, and mediate mucosal inflammation of the digestive tract, which is called “gastrointestinal mucositis.” This mucositis can affect the quality of life of the host and even threaten their lives. Several studies reported the association between chemotherapy-mediated gastrointestinal mucositis in CRC and gut dysbiosis. However, the underlying mechanisms of this association are still unclear. The alternative or complementary treatments to reshape gut microbiota and slow down the side effects of chemotherapy have shown the improvement of gastrointestinal mucositis following chemotherapy in the CRC condition. This review will summarize and discuss the evidence of the association between chemotherapy-mediated gastrointestinal mucositis in CRC and altered gut microbiota from in vivo and clinical studies. The possible mechanisms of gastrointestinal mucositis, including the destruction of the gastrointestinal mucosal barrier, the induction of gut dysbiosis, and histopathological changes in the gut of CRC with chemotherapy will be illustrated. In addition, the nonpharmacological interventions and phytochemical extracts by using the manipulation of the microbial population for therapeutic purposes for relieving side effects of chemotherapy as well as a cancer treatment would be summarized and discussed in this review.

28. Use new PLGL-RGD-NGF nerve conduits for promoting peripheral nerve regeneration

Abstract

Abstract Background Nerve conduits provide a promising strategy for peripheral nerve injury repair. However, the efficiency of nerve conduits to enhance nerve regeneration and functional recovery is often inferior to that of autografts. Nerve conduits require additional factors such as cell adhesion molecules and neurotrophic factors to provide a more conducive microenvironment for nerve regeneration. Methods In the present study, poly{(lactic acid)-co-[(glycolic acid)-alt-(L-lysine)]} (PLGL) was modified by grafting Gly-Arg-Gly-Asp-Gly (RGD peptide) and nerve growth factor (NGF) for fabricating new PLGL-RGD-NGF nerve conduits to promote nerve regeneration and functional recovery. PLGL-RGD-NGF nerve conduits were tested in the rat sciatic nerve transection model. Rat sciatic nerves were cut off to form a 10 mm defect and repaired with the nerve conduits. All of the 32 Wistar rats were randomly divided into 4 groups: group PLGL-RGD-NGF, group PLGL-RGD, group PLGL and group autograft. At 3 months after surgery, the regenerated rat sciatic nerve was evaluated by footprint analysis, electrophysiology, and histologic assessment. Experimental data were processed using the statistical software SPSS 10.0. Results The sciatic function index value of groups PLGL-RGD-NGF and autograft was significantly higher than those of groups PLGL-RGD and PLGL. The nerve conduction velocities of groups PLGL-RGD-NGF and autograft were significantly faster than those of groups PLGL-RGD and PLGL. The regenerated nerves of groups PLGL-RGD-NGF and autograft were more mature than those of groups PLGL-RGD and PLGL. There was no significant difference between groups PLGL-RGD-NGF and autograft. Conclusions PLGL-RGD-NGF nerve conduits are more effective in regenerating nerves than both PLGL-RGD nerve conduits and PLGL nerve conduits. The effect is as good as that of an autograft. This work established the platform for further development of the use of PLGL-RGD-NGF nerve conduits for clinical nerve repair.

44. Functionalized Flexible Soft Polymer Optical Fibers for Laser Photomedicine.

Abstract

Abstract: Optical waveguides allow propagating light through biological tissue in optogenetics and photomedicine applications. However, achieving efficient light delivery to deep tissues for long‐term implantation has been limited with solid‐state optical fibers. Here, a method is created to rapidly fabricate flexible, functionalized soft polymer optical fibers (SPOFs) coupled with silica fibers. A step‐index core/cladded poly(acrylamide‐co‐poly(ethylene glycol) diacrylate)/Ca alginate SPOF is fabricated through free‐radical polymerization in a mold. The SPOF is integrated with a solid‐state silica fiber coupler for efficient light delivery. The cladded SPOF shows ≈1.5‐fold increase in light propagation compared to the noncladded fiber. The optical loss of the SPOF is measured as 0.6 dB cm−1 at the bending angle of 70° and 0.28 dB cm−1 through a phantom tissue. The SPOF (inner Ø = 200 µm) integrated with a 21 gauge needle (inner Ø = 514 µm) is inserted within a porcine tissue. The intensity of light decreases ≈60%, as the SPOF is implanted as deep as 2 cm. Doped with fluorescent dye and gold nanoparticles, the SPOF fiber exhibits yellow‐red and red illumination. Living cells can also be incorporated within the SPOF with viability. The flexible SPOFs may have applications in photodynamic light therapy, optical biosensors, and photomedicine. [ABSTRACT FROM AUTHOR]
