Abstract Objective Currently, there are still some shortcomings in EBM education in China.The study aimed to investigate the effectiveness of the novel evidence-based medicine (EBM) learning model of “autonomy-collaboration.” Methods A total of 91 undergraduate students majoring in clinical medicine at Zhongshan Clinical College of Dalian University from the 2019 batch were selected as the participants in this study. They were instructed to follow the EBM learning model of “autonomy-collaboration.” Upon completion of the course, questionnaires, records of participants’ sentiments and insights, and evidence-based clinical practice reports were used as indicators to evaluate the effectiveness of the training. Results This learning modality effectively enhanced independent learning ability of the students, stimulated their interest in learning, and strengthened the communication between students and teachers, thereby improving the quality of teaching. Conclusion The novel EBM learning model of “autonomy-collaboration,” exhibited robust effectiveness in instruction and facilitated the seamless integration of theoretical knowledge with clinical practice. Consequently, its widespread adoption is strongly recommended.

Abstract Intron retention (IR) is the most common alternative splicing event in Arabidopsis. An increasing number of studies have demonstrated the major role of IR in gene expression regulation. The impacts of IR on plant growth and development and response to environments remain underexplored. Here, we found that IR functions directly in gene expression regulation on a genome-wide scale through the detainment of intron-retained transcripts (IRTs) in the nucleus. Nuclear-retained IRTs can be kept away from translation through this mechanism. COP1-dependent light modulation of the IRTs of light signaling genes, such as PIF4, RVE1, and ABA3, contribute to seedling morphological development in response to changing light conditions. Furthermore, light-induced IR changes are under the control of the spliceosome, and in part through COP1-dependent ubiquitination and degradation of DCS1, a plant-specific spliceosomal component. Our data suggest that light regulates the activity of the spliceosome and the consequent IRT nucleus detainment to modulate photomorphogenesis through COP1.

Objective: To analyze the current practice of labeling common food with added strains in China, explore the existing problems, and provide a reference for the management of labeling food that contains edible strains. Methods: Label information for prepackaged foods labeled "live bacteria" "lactic acid bacteria" and "probiotics" (excluding health foods and traditional fermented foods) was collected via online and in-person markets and then organized. Results: A total of 526 samples that met the requirements were examined. 65.24% of products used the term "probiotics" in their labels or promotional language. 91.06% of the samples were labeled with the strain name on the packaging, 49.81% of the samples were labeled with the strain name. In addition, 72.43% of the samples were labeled with a bacterial count, with 95% of the product packaging indicating a bacterial count greater than 106 CFU/g (CFU/mL). Further, 29.28% of the samples used functional claims and related descriptions of similar functional claims in labels or promotional language, and 46.9% of the samples indicated an unsuitable intended audience of the product on the label. Conclusion: There are many types of edible mushroom products added to common food in China, and the positions of the labels and methods for naming mushrooms and the quantities that appear in products greatly differ. Some products incorrectly use the term "probiotics", while other products state claims for improper use. A management system in China for general labeling and for the specific labeling of food that contains "lactic acid bacteria" "probiotics" needs to be standardized.

Carbon nanotubes (CNTs), as potent reinforcements in composites, have demonstrated excellent strengthening effects when combined with copper in numerous recent studies. Challenges remain in the application of these composites and in fully leveraging the reinforcing capabilities of CNTs to achieve comprehensive performance enhancement. The performance of CNTs/Cu composites can be flexibly regulated owing to the unique structure and properties of CNTs. To achieve the fabrication of high-performance and diverse CNTs/Cu composites, a profound understanding of the reinforcement mechanisms of CNTs in the composites is essential, along with the consideration of key influencing factors on performance. This article provides a comprehensive overview of the reinforcement mechanisms of CNTs on the mechanical, electrical, and thermal properties of CNTs/Cu composites. Factors influencing the effectiveness of CNT reinforcement in composites are discussed, including the attributes and dispersion of CNTs, the architectures of composites, and the interface between CNTs and Cu. Furthermore, this study explores the role of CNTs in addressing the trade-off between high strength and high conductivity as well as between high strength and high ductility in the copper matrix.

ABSTRACT: Objectives: Ceftolozane-tazobactam (C/T) is a combination of a cephalosporin and a β-lactamase inhibitor with activity against Gram-negative bacilli (GNB). The study aims were to evaluate the activity of C/T in vitro vs. comparators against clinical GNB isolated from Chinese paediatric patients. Methods: From 2017–2021, 660 GNB isolates were collected from 20 hospitals across China. The minimum inhibitory concentrations were tested using a Trek Diagnostic System (Thermo Fisher Scientific). Susceptibility was determined by CLSI broth microdilution and the results were interpreted according to CLSI M100 (2021) breakpoints. Results: GNB isolates were obtained from paediatric patients < 18 years old, mainly from the bloodstream (n = 146), intraperitoneal cavity (n = 138), lower respiratory (n = 278) and urinary tract (n = 96). Overall, C/T was active against 76.6% of 436 Enterobacterales, with a descending susceptibility rate of 100.0% to S. marcescens, 92.2% to E. coli, 83.3% to K. oxytoca, 66.7% to K. aerogenes, 66.7% to P. mirabilis, 58.6% to K. pneumoniae and 57.1% to E. cloacae. The susceptibility of P. aeruginosa to C/T was 89.4%, which was the highest among the β-lactam antibiotics and was second only to amikacin (92.9%). Based on experiments and first-principles calculations, the microstructures and mechanical properties of as-cast and solution treated Mg-10Gd-4Y-xZn-0.6Zr (x = 0, 1, 2, wt.%) alloys are investigated. The transformation process of long-period stacking ordered (LPSO) structure during solidification and heat treatment and its effect on the mechanical properties of experimental alloys are discussed. Results reveal that the stacking faults and 18R LPSO phases appear in the as-cast Mg-10Gd-4Y-1Zn-0.6Zr and Mg-10Gd-4Y-2Zn-0.6Zr alloys, respectively. After solution treatment, the stacking faults and 18R LPSO phase transform into 14H LPSO phase. The Enthalpies of formation and reaction energy of 14H and 18R LPSO are calculated based on first-principles. Results show that the alloying ability of 18R is stronger than that of 14H. The reaction energies show that the 14H LPSO phase is more stable than the 18R LPSO. The elastic properties of the 14H and 18R LPSO phases are also evaluated by first-principles calculations, and the results are in good agreement with the experimental results. The precipitation of LPSO phase improves the tensile strength, yield strength and elongation of the alloy. After solution treatment, the Mg-10Gd-4Y-2Zn-0.6Zr alloy has the best mechanical properties, and its ultimate tensile strength and yield strength are 278.7 MPa and 196.4 MPa, respectively. The elongation of Mg-10Gd-4Y-2Zn-0.6Zr reaches 15.1, which is higher than that of Mg-10Gd-4Y0.6Zr alloy. The improving mechanism of elastic modulus by the LPSO phases and the influence on the alloy mechanical properties are also analyzed.

We proposed a unique wire-production process by rolling up and drawing chemical vapor deposition copper/graphene (Cu/Gr) foils, and a good strength-conductivity trade-off was achieved originating from the dispersed Gr in Cu matrix and the high-quality heterointerfaces. The 1.14 mm cold-drawn composite wire exhibited a tensile strength of 455 ± 5 MPa and a conductivity of 98.18 ± 0.16 % of the International Annealed Copper Standard (IACS), and the tensile strength was 250 ± 2 MPa with a satisfied electrical conductivity of 101.68 ± 0.52 % IACS upon annealing. The microstructure involution during the preparing process was revealed, and the reinforcement mechanisms in the yield strength and conductivity due to the introduced Gr were clarified. The results indicated that the Gr plays a role in pinning dislocations and preventing grain boundary movement during deformation and the subsequent annealing, thereby enhancing the strength of the Cu matrix. Meanwhile, the Cu-Gr coupling interfaces exhibited an electronic doping effect, enhancing the conductive properties. Our work presented a feasible method for preparing Cu/Gr composite wire with comprehensive electrical conductivity and strength optimization.

Abstract Background The study aims were to evaluate the species distribution and antimicrobial resistance profile of Gram-negative pathogens isolated from specimens of intra-abdominal infections (IAI), urinary tract infections (UTI), respiratory tract infections (RTI), and blood stream infections (BSI) in emergency departments (EDs) in China. Methods From 2016 to 2019, 656 isolates were collected from 18 hospitals across China. Minimum inhibitory concentrations were determined by CLSI broth microdilution and interpreted according to CLSI M100 (2021) guidelines. In addition, organ-specific weighted incidence antibiograms (OSWIAs) were constructed. Results Escherichia coli (E. coli) and Klebsiella pneumoniae (K. pneumoniae) were the most common pathogens isolated from BSI, IAI and UTI, accounting for 80% of the Gram-negative clinical isolates, while Pseudomonas aeruginosa (P. aeruginosa) was mainly isolated from RTI. ABSTRACT Acinetobacter baumannii is one of the most important pathogens worldwide. The intrinsic and acquired resistance of A. baumannii, coupled with the slow pace of novel antimicrobial drug development, poses an unprecedented and enormous challenge to clinical anti-infective therapy of A. baumannii. Recent studies in the field of pathogenicity, antibiotic resistance, and biofilms of A. baumannii have focused on the model strains, including ATCC 17978, ATCC 19606, and AB5075. However, these model strains represent only a limited portion of the heterogeneity in A. baumannii. Furthermore, variants of these model strains have emerged that show significant diversity not only at the genotypic level but also reflected in differences at the phenotypic levels of capsule, virulence, pathogenicity, and antibiotic resistance. Research on A. baumannii, a key pathogen, would benefit from a standardized approach, which characterizes heterogeneous strains in order to facilitate rapid diagnosis, discovery of new therapeutic targets, and efficacy assessment. Our study provides and describes a standardized, genomically and phenotypically heterogeneous panel of 45 different A. baumannii strains for the research community. In addition, we performed comparative analyses of several phenotypes of this panel. We found that the sequence type 2 (ST2) group showed significantly higher rates of resistance, lower fitness cost for adaptation, and yet less biofilm formation. The Macrocolony type E (MTE, flat center and wavy edge phenotype reported in the literature) group showed a less clear correlation of resistance rates and growth rate, but was observed to produce more biofilms. Our study sheds light on the complex interplay of resistance fitness and biofilm formation within distinct strains, offering insights crucial for combating A. baumannii infection.IMPORTANCEAcinetobacter baumannii is globally notorious, and in an effort to combat the spread of such pathogens, several emerging candidate therapies have already surfaced. However, the strains used to test these therapies vary across studies (the sources and numbers of test strains are varied and often very large, with little heterogeneity). The variation complicates the studies. Furthermore, the limited standardized resources of A. baumannii strains have greatly restricted the research on the physiology, pathogenicity, and antibiotic resistance. Therefore, it is crucial for the research community to acquire a standardized and heterogeneous panel of A. baumannii. Our study meticulously selected 45 diverse A. baumannii strains from a total of 2,197 clinical isolates collected from 64 different hospitals across 27 provinces in China, providing a scientific reference for the research community. This assistance will significantly facilitate scientific exchange in academic research.

ABSTRACT To monitor the resistance rate and gain a deeper understanding of the resistance mechanisms, we conducted over a 2-year surveillance focusing on the Klebsiella pneumoniae associated with the clinical usage of ceftazidime-avibactam (CZA) in a teaching hospital. A total of 4,641 K. pneumoniae isolates were screened to identify the CZA resistance through antimicrobial susceptibility testing. Comprehensive analyses, including homology analysis, conjugation experiments, clone assays, and whole genome sequencing, were furtherly performed on the CZA-resistant strains. In total, four CZA-resistant K. pneumoniae (CZA-R-Kp) strains were separated from four patients, in which three of them received CZA treatment during the hospitalization, accounting for a 4% (3/75) resistance development rate of K. pneumoniae under CZA stress. All CZA-R-Kp isolates were found to possess variants of blaKPC-2. The identified mutations included blaKPC-33, blaKPC-86, and a novel variant designated as blaKPC-129, all of which were located in the Ω loop of the KPC enzyme. These mutations were found to impact the amino acid sequence and spatial structure of the enzyme’s active center, consequently affecting KPC carbapenemase activity. This study underscores the importance of active surveillance to monitor the emergence of resistance to CZA, highlighting the need for ongoing research to develop effective strategies for combating antimicrobial resistance. Understanding the mechanisms behind resistance is crucial in maintaining the efficacy of CZA, a vital tool in the battle against multidrug-resistant infections.IMPORTANCEAs an effective drug for the treatment of carbapenem-resistant Klebsiella pneumoniae, ceftazidime-avibactam (CZA) began to develop resistance in recent years and showed an increasing trend. In order to effectively monitor the resistance rate of CZA and understand its resistance mechanism, we monitored K. pneumoniae for more than 2 years to find CZA-resistant strains. Through comprehensive analysis of the selected CZA-resistant strains, it was found that all the CZA-resistant strains had mutation, which could affect the activity of KPC carbapenemase. This study highlights the importance of proactive surveillance to monitor the emergence of CZA resistance, which highlights the need for ongoing research to develop effective strategies to combat antimicrobial resistance. Understanding the mechanisms behind resistance is critical to maintaining the effectiveness of CZA, an important tool in the fight against multidrug-resistant infections.

BackgroundMultiple sclerosis (MS) and Cerebral Small Vessel Disease (CSVD) exhibit some similarities in Magnetic resonance imaging (MRI), potentially leading to misdiagnosis and delaying effective treatment windows. It is unclear whether CSVD can be detected with Paramagnetic Rim Lesions (PRL), which is special in MS.ObjectiveWe aimed to investigate whether PRL can serve as a neuroimaging marker for discriminating between MS and CSVD.MethodsIn this retrospective study, 49 MS and 104 CSVD patients underwent 3.0 T Magnetic resonance imaging (MRI). Visual assessment of 37 MS patients and 89 CSVD patients with or without lacunes, cerebral microbleeds (CMBs), enlarged perivascular spaces (EPVS), white matter hyperintensity (WMH), central vein sign (CVS), and PRL. IntroductionThe dissemination of carbapenem-resistant Enterobacteriales (CRE) in nosocomial settings is primarily associated with the horizontal transfer of plasmids. However, limited research has focused on the in-host transferability of carbapenem resistance. In this study, ten isolates were collected from gut specimens of five individuals, each hosting two different species, including Escherichia coli, Klebsiella pneumoniae, Klebsiella aerogenes, Enterobacter cloacae, or Citrobacter koseri.MethodsSpecies identification and antimicrobial susceptibility were determined by MALDI-TOF MS and broth microdilution method. Carbapenemase genes were detected and localized using PCR, S1-PFGE and southern blot. The transferability of carbapenemase genes between species was investigated through filter mating experiments, and the genetic contexts of the plasmids were analyzed using whole genome sequencing.Results and discussionOur results revealed that each of the ten isolates harbored a carbapenemase gene, including blaNDM-5, blaNDM-1, or blaKPC-2, on a plasmid. Five different plasmids were successfully transferred to recipient cells of E. coli, K. pneumoniae or A. baumannii by transconjugation. The genetic contexts of the carbapenemase gene were remarkably similar between the two CRE isolates from each individual. This study highlights the potential for interspecies plasmid transmission in human gut, emphasizing the colonization of CRE as a significant risk factor for the dissemination of carbapenemase genes within the host. These findings underscore the need for appropriate intestinal CRE screening and colonization prevention.

Abstract Since ancient times, humans have learned to use fire and other heating methods to fight against dangerous pathogens, like cooking raw food, sterilizing surgical tools, and disinfecting other pathogen transmission media. However, it remains difficult for current heating methods to achieve extremely fast and highly efficient sterilization simultaneously. Herein, an ultrafast and uniform heating‐based strategy with outstanding bactericidal performance is proposed. Ultra‐precise laser manufacturing is used to fabricate the Joule heater which can be rapidly heated to 90 °C in 5 s with less than 1 °C fluctuation in a large area by real‐time temperature feedback control. An over 98% bactericidal efficiency on S. aureus for 30 s and on E. coli for merely 5 s is shown. The heating strategy shows a 360 times faster acceleration compared to the commonly used steam sterilization from the suggested guidelines by the Centers for Disease Control and Prevention (CDC), indicating that high temperatures with short duration can effectively disinfect microorganisms. As a proof of concept, this heating strategy can be widely applied to sterilizing cash and various objects to help protect the public from bacteria in daily life.

Semantic segmentation of remote sensing images (SSRSIs), which aims to assign a category to each pixel in remote sensing images, plays a vital role in a broad range of applications, such as environmental monitoring, urban planning, and land resource utilization. Recently, with the successful application of deep learning in remote sensing, a substantial amount of work has been aimed at developing SSRSI methods using deep learning models. In this survey, we provide a comprehensive review of SSRSI. First, we review the current mainstream semantic segmentation models based on deep learning. Next, we analyze the main challenges faced by SSRSI and comprehensively summarize the current research status of deep-learning-based SSRSI, especially some new directions in SSRSI are outlined, including semisupervised and weakly-supervised SSRSI, unsupervised domain adaption in SSRSI, multimodal data-fusion-based SSRSI, and pretrained models for SSRSI. Then, we examine the most widely used datasets and metrics and review the quantitative results and experimental performance of some representative methods of SSRSI. Finally, we discuss promising future research directions in this area.

The application of a hybrid-fruit-peel (HFP) coagulant used as an external carbon source (ECS) in both simulated water and real sewage having a low carbon source treated with sequencing batch reactor (SBR) was studied, compared with that of sodium acetate (NaAc). The impact of HFP on sludge properties (such as extracellular polymer substance (EPS), dehydrogenase activity (DHA), charged property, size, microscopic images and bacteria phase) was characterized. The results showed that as an ECS, HFP basically gave similar nitrogen removal to NaAc and also gave a similar developing trend of both dissolved oxygen (DO) and pH. HFP promoted more proliferation of microorganisms and posed higher levels of protein (PN) and polysaccharide (PS) than NaAc, but gave slightly lower DHA than NaAc. After HFP was added as an ECS, the types and quantities of microorganisms increased significantly, the effluent qualities were improved and the sludge size and extensibility became larger, which was conducive to direct contact and remove pollutants. HFP played a similar role to NaAc as ECS and can be used as a quality and slow-releasing ECS for low carbon source wastewaters. HIGHLIGHTS Application of HFP coagulant as an external carbon source (ECS) for low C/N wastewater was studied and gave a similar effect to NaAc.; HFP can be used as a quality and slow-releasing ECS for low C/N wastewater.; HFP produced sludge has a larger size and better extensibility.; HFP promoted more proliferation of microorganisms and gave better effluent qualities.;

Abstract Nasopharyngeal carcinoma (NPC) is a malignant tumor that is highly prevalent in Southeast Asia, especially in South China. The pathogenesis of NPC is complex, and genetic alterations of tumor suppressors and proto-oncogenes play important roles in NPC carcinogenesis. p53 is unexpectedly highly expressed in NPC and possesses an uncommon mutation of R280T, which is different from a high frequency of hotspot mutations or low expression in other tumors. However, the mechanism of p53 loss of function and its correlation with R280T in NPC are still unclear. In this study, p53 amyloid aggregates were found to be widespread in NPC and can be mainly induced by the R280T mutation. Aggregated p53-R280T impeded its entry into the nucleus and was unable to initiate the transcription of downstream target genes, resulting in decreased NPC cell cycle arrest and apoptosis. In addition, NPC cells with p53-R280T amyloid aggregates also contributed aggressively to tumor growth in vivo. Transcriptome analysis suggested that p53 amyloid aggregation dysregulated major signaling pathways associated with the cell cycle, proliferation, apoptosis, and unfolded protein response (UPR). Further studies revealed that Hsp90, as a key molecular chaperone in p53 folding, was upregulated in NPC cells with p53-R280T aggregation, and the upregulated Hsp90 facilitated p53 aggregation in turn, forming positive feedback. Therefore, Hsp90 inhibitors could dissociate p53-R280T aggregation and restore the suppressor function of p53 in vitro and in vivo. In conclusion, our study demonstrated that p53-R280T may misfold to form aggregates with the help of Hsp90, resulting in the inability of sequestered p53 to initiate the transcription of downstream target genes. These results revealed a new mechanism for the loss of p53 function in NPC and provided novel mechanistic insight into NPC pathogenesis.

ObjectiveIn this work, we propose a novel method for constructing whole-brain spatio-temporal multilayer functional connectivity networks (FCNs) and four innovative rich-club metrics.MethodsSpatio-temporal multilayer FCNs achieve a high-order representation of the spatio-temporal dynamic characteristics of brain networks by combining the sliding time window method with graph theory and hypergraph theory. The four proposed rich-club scales are based on the dynamic changes in rich-club node identity, providing a parameterized description of the topological dynamic characteristics of brain networks from both temporal and spatial perspectives. The proposed method was validated in three independent differential analysis experiments: male–female gender difference analysis, analysis of abnormality in patients with autism spectrum disorders (ASD), and individual difference analysis.ResultsThe proposed method yielded results consistent with previous relevant studies and revealed some innovative findings. For instance, the dynamic topological characteristics of specific white matter regions effectively reflected individual differences. The increased abnormality in internal functional connectivity within the basal ganglia may be a contributing factor to the occurrence of repetitive or restrictive behaviors in ASD patients.ConclusionThe proposed methodology provides an efficacious approach for constructing whole-brain spatio-temporal multilayer FCNs and conducting analysis of their dynamic topological structures. The dynamic topological characteristics of spatio-temporal multilayer FCNs may offer new insights into physiological variations and pathological abnormalities in neuroscience.

Purpose: The overall diagnostic value of fine-needle aspiration (FNA) is not as excellent as that of core needle biopsy (CNB). Limited research has investigated small cervical lymph nodes inaccessible to ultrasound-guided CNB due to technical challenges associated with their small size. Therefore, this study aimed to evaluate the accuracy of ultrasound-guided FNA in determining the etiology of small cervical lymph nodes. Methods: A retrospective analysis was conducted on patients who underwent FNA between May 2018 and May 2021 at our hospital. Cytological, histopathological, and clinical follow-up data were analyzed. The diagnostic yield of FNA was assessed based on sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), and accuracy calculations. Results: This study included 505 patients, each with a small cervical lymph node under evaluation (total number of lymph nodes: 505). The average maximal diameter of the lymph nodes was 14.6 ± 6.2 mm. According to the Sydney system, the cytology results were as follows: Category I in 26 lymph nodes (5.1 %); Category II in 269 (53.3 %); Category III in 35 (6.9 %); Category IV in 17 (3.4 %); and Category V in 158 (31.3 %). We identified 212 malignant cases (203 metastases and 9 lymphomas) and 293 benign lymph nodes. FNA achieved high sensitivity (88.8 %), specificity (99.6 %), PPV (99.4 %), NPV (91.8 %), and overall accuracy (94.8 %) in determining the etiology of small cervical lymph nodes. Conclusion: FNA cytology is suitable for small lesions inaccessible by CNB and provides a diagnostic basis for implementing clinically appropriate treatment measures.

Summary: Jasmonate (JA) is a well-known phytohormone essential for plant response to biotic stress. Recently, a crucial role of JA signaling in salt resistance has been highlighted; however, the specific regulatory mechanism remains largely unknown. In this study, we found that the NUCLEAR FACTOR-Y (NF-Y) subunits NF-YA1, NF-YB2, and NF-YC9 form a trimeric complex that positively regulates the expression of salinity-responsive genes, whereas JASMONATE-ZIM DOMAIN protein 8 (JAZ8) directly interacts with three subunits and acts as the key repressor to suppress both the assembly of the NF-YA1-YB2-YC9 trimeric complex and the transcriptional activation activity of the complex. When plants encounter high salinity, JA levels are elevated and perceived by the CORONATINE INSENSITIVE (COI) 1 receptor, leading to the degradation of JAZ8 via the 26S proteasome pathway, thereby releasing the activity of the NF-YA1-YB2-YC9 complex, initiating the activation of salinity-responsive genes, such as MYB75, and thus enhancing the salinity tolerance of plants.

Fermentation is a vital process occurred under the premise of rolling affecting black tea quality. Theoretically, re-rolling during fermentation will remodel the biochemical conditions of tea leaves, and thus influence black tea quality. Herein, we studied the effect of re-rolling on black tea taste and liquor color. Sensory evaluation showed that re-rolling significantly weakened the astringency taste and improved the redness and luminance of liquor. With re-rolling, the color attributes of a* and L* and the contents of theaflavins and thearubigins were significantly improved. Metabolomics analysis showed that the contents of 110 non-volatile compounds were significantly different among black teas with different rolling treatments. In summary, re-rolling accelerated the oxidation of polyphenols into pigments, the hydrolysis of proteins into amino acids, and the metabolism of alkaloids, organic acids, glycosidically-bound volatiles, and lipids during the fermentation period. Our study provided a novel and simple way to improve black tea quality.

Abstract The atomic-thick anticorrosion coating for copper (Cu) electrodes is essential for the miniaturisation in the semiconductor industry. Graphene has long been expected to be the ultimate anticorrosion material, however, its real anticorrosion performance is still under great controversy. Specifically, strong electronic couplings can limit the interfacial diffusion of corrosive molecules, whereas they can also promote the surficial galvanic corrosion. Here, we report the enhanced anticorrosion for Cu simply via a bilayer graphene coating, which provides protection for more than 5 years at room temperature and 1000 h at 200 °C. Such excellent anticorrosion is attributed to a nontrivial Janus-doping effect in bilayer graphene, where the heavily doped bottom layer forms a strong interaction with Cu to limit the interfacial diffusion, while the nearly charge neutral top layer behaves inertly to alleviate the galvanic corrosion. Our study will likely expand the application scenarios of Cu under various extreme operating conditions.

Abstract Controllable growth of two-dimensional (2D) single crystals on insulating substrates is the ultimate pursuit for realizing high-end applications in electronics and optoelectronics. However, for the most typical 2D insulator, hexagonal boron nitride (hBN), the production of a single-crystal monolayer on insulating substrates remains challenging. Here, we propose a methodology to realize the facile production of inch-sized single-crystal hBN monolayers on various insulating substrates by an atomic-scale stamp-like technique. The single-crystal Cu foils grown with hBN films can stick tightly (within 0.35 nm) to the insulating substrate at sub-melting temperature of Cu and extrude the hBN grown on the metallic surface onto the insulating substrate. Single-crystal hBN films can then be obtained by removing the Cu foil similar to the stamp process, regardless of the type or crystallinity of the insulating substrates. Our work will likely promote the manufacturing process of fully single-crystal 2D material-based devices and their applications.

In the work, the cryogenic and room temperature drawing behaviors of Cu–Te alloy with a wide range of accumulative drawing true strain up to 1.1 are systematically investigated. For the first time, the storage of geometrically necessary dislocations (GND) and statically stored dislocations (SSD), occurrence of deformation twinning, as well as their respective roles in strain hardening in the drawn Cu–Te alloy at cryogenic and room temperature have been studied. The results show that the liquid nitrogen temperature drawn (LNT-D) Cu–Te alloy presents higher yield strength (YS), ultimate tensile strength than the room temperature drawn (RT-D) Cu–Te alloy. Microstructural investigation reveals that cryogenic wire drawing of Cu–Te alloy promotes the formation of simultaneous twinning and shear bands, accompanied by the fragmentation of Cu2Te phase. The LNT-D Cu–Te alloy exhibits planar slip characteristics, while RT-D Cu–Te alloy presents wave slip characteristics. Compared with RT-D Cu–Te alloy, the higher YS of LNT-D Cu–Te alloy can be mainly attributed to dislocation strengthening and twinning strengthening, which account for 76.25–80.45% and 4.79–4.91% of the total YS respectively. During room temperature drawing, both GND and SSD play an important role in the hardening of Cu–Te alloy. During cryogenic temperature drawing, work hardening is predominantly through rapid multiplication of SSDs.

Objectives: Cervical discomfort and other symptoms may be attributable to the middle cervical sympathetic ganglion. The aim of this study was to explore the sonographic features of this ganglion in anatomical specimens and cadavers and evaluate the feasibility of its visualization using high-resolution ultrasonography. Methods: We examined three cervical sympathetic-ganglion specimens and two fresh cadavers using high-resolution ultrasound to explore the sonographic features of this ganglion. Basic imaging characteristics examined included the shape, echo intensity, and location of the ganglion. Core-needle biopsy was performed to examine the suspected middle cervical sympathetic ganglion in the two fresh cadavers and verify the accuracy of the sonographic identification via pathological examination. Results: The middle cervical sympathetic ganglion appeared on high-resolution ultrasonography as an oval-shaped hypoechoic structure, with at least one continuous hypoechoic line connected to each ending in the anatomical specimens and fresh cadavers, and it was distinctly different from the adjacent lymph nodes. Discussion: Based on an adequate understanding of both its location and sonographic features, the direct visualization of the middle cervical sympathetic ganglion using high-resolution ultrasonography is feasible.

Abstract Flexible perovskite solar cells (f‐PSCs) as a promising power source have grabbed surging attention from academia and industry specialists by integrating with different wearable and portable electronics. With the development of low‐temperature solution preparation technology and the application of different engineering strategies, the power conversion efficiency of f‐PSCs has approached 24%. Due to the inherent properties and application scenarios of f‐PSCs, the study of strain in these devices is recognized as one of the key factors in obtaining ideal devices and promoting commercialization. The strains mainly from the change of bond and lattice volume can promote phase transformation, induce decomposition of perovskite film, decrease mechanical stability, etc. However, the effect of strain on the performance of f‐PSCs has not been systematically summarized yet. Herein, the sources of strain, evaluation methods, impacts on f‐PSCs, and the engineering strategies to modulate strain are summarized. Furthermore, the problems and future challenges in this regard are raised, and solutions and outlooks are offered. This review is dedicated to summarizing and enhancing the research into the strain of f‐PSCs to provide some new insights that can further improve the optoelectronic performance and stability of flexible devices.

Abstract Background Nitrogen (N), phosphorus (P) and potassium (K) are critical macronutrients in crops, such that deficiency in any of N, P or K has substantial effects on crop growth. However, the specific commonalities of plant responses to different macronutrient deficiencies remain largely unknown. Methods Here, we assessed the phenotypic and physiological performances along with whole transcriptome and metabolomic profiles of rapeseed seedlings exposed to N, P and K deficiency stresses. Results Quantities of reactive oxygen species were significantly increased by all macronutrient deficiencies. N and K deficiencies resulted in more severe root development responses than P deficiency, as well as greater chlorophyll content reduction in leaves (associated with disrupted chloroplast structure). Transcriptome and metabolome analyses validated the macronutrient-specific responses, with more pronounced effects of N and P deficiencies on mRNAs, microRNAs (miRNAs), circular RNAs (circRNAs) and metabolites relative to K deficiency. Tissue-specific responses also occurred, with greater effects of macronutrient deficiencies on roots compared with shoots. We further uncovered a set of common responders with simultaneous roles in all three macronutrient deficiencies, including 112 mRNAs and 10 miRNAs involved in hormonal signaling, ion transport and oxidative stress in the root, and 33 mRNAs and 6 miRNAs with roles in abiotic stress response and photosynthesis in the shoot. 27 and seven common miRNA-mRNA pairs with role in miRNA-mediated regulation of oxidoreduction processes and ion transmembrane transport were identified in all three macronutrient deficiencies. No circRNA was responsive to three macronutrient deficiency stresses, but two common circRNAs were identified for two macronutrient deficiencies. Combined analysis of circRNAs, miRNAs and mRNAs suggested that two circRNAs act as decoys for miR156 and participate in oxidoreduction processes and transmembrane transport in both N- and P-deprived roots. Simultaneously, dramatic alterations of metabolites also occurred. Associations of RNAs with metabolites were observed, and suggested potential positive regulatory roles for tricarboxylic acids, azoles, carbohydrates, sterols and auxins, and negative regulatory roles for aromatic and aspartate amino acids, glucosamine-containing compounds, cinnamic acid, and nicotianamine in plant adaptation to macronutrient deficiency. Conclusions Our findings revealed strategies to rescue rapeseed from macronutrient deficiency stress, including reducing the expression of non-essential genes and activating or enhancing the expression of anti-stress genes, aided by plant hormones, ion transporters and stress responders. The common responders to different macronutrient deficiencies identified could be targeted to enhance nutrient use efficiency in rapeseed.

A semi-solid Al alloy with superior comprehensive performance was obtained via alloying design concept and rheo-diecasting (RDC). The semi-solid alloy slurry was obtained via the vibrating contraction sloping plate. As the contents of Cu and Mg elements increased, the secondary dendrite arm spacing (SDAS) decreased and solubility of Cu and Mg elements increased, which improved the mechanical property and decreased the thermal conductivity of as-cast alloys. The effect of microstructure evolution induced by pouring temperature on mechanical properties at room and elevated temperatures, thermal conductivity, and corrosion behavior of the RDC alloy was investigated. The results demonstrated that the low porosity and refined primary α1-Al grains, secondary α2-Al grains, β-Al5FeSi phases, eutectic Si phases played a significant role in enhancing mechanical properties of the alloy at room temperature. Meanwhile, some submicron β-Al5FeSi phases effectively hindered grain boundary sliding, resulting in the increased mechanical properties of the alloy at elevated temperatures. The improvement in thermal conductivity was related to the decreased electron scattering due to the reduced porosity and small-sized secondary phases. The decrease in the potential difference between the β-Al5FeSi phase and matrix helped improve the corrosion resistance of the alloy.

This research focuses on 72 approved varieties of colored wheat from different provinces in China. Utilizing coefficients of variation, structural equation models, and correlation analyses, six agronomic traits of colored wheat were comprehensively evaluated, followed by further research on different dwarfing genes in colored wheat. Using the entropy method revealed that among the 72 colored wheat varieties, 10 were suitable for cultivation. Variety 70 was the top-performing variety, with a comprehensive index of 87.15%. In the final established structural equation model, each agronomic trait exhibited a positive direct effect on yield. Notably, plant height, spike length, and flag leaf width had significant impacts on yield, with path coefficients of 0.55, 0.40, and 0.27. Transcriptome analysis and real-time fluorescence quantitative polymerase chain reaction (RT-qPCR) validation were used to identify three dwarfing genes controlling plant height: Rht1, Rht-D1, and Rht8. Subsequent RT-qPCR validation clustering heatmap results indicated that Rht-D1 gene expression increased with the growth of per-acre yield. Rht8 belongs to the semi-dwarf gene category and has a significant positive effect on grain yield. However, the impact of Rht1, as a dwarfing gene, on agronomic traits varies. These research findings provide crucial references for the breeding of new varieties.

Clostridium perfringens is the main pathogen of chicken necrotic enteritis (NE) causing huge economic losses in the poultry industry. Although dietary secondary bile acid deoxycholic acid (DCA) reduced chicken NE, the accumulation of conjugated tauro-DCA (TDCA) raised concerns regarding DCA efficacy. In this study, we aimed to deconjugate TDCA by bile salt hydrolase (BSH) to increase DCA efficacy against the NE pathogen C. perfringens. Assays were conducted to evaluate the inhibition of C. perfringens growth, hydrogen sulfide (H2S) production, and virulence gene expression by TDCA and DCA. BSH activity and sequence alignment were conducted to select the bsh gene for cloning. The bsh gene from Bifidobacterium longum was PCR-amplified and cloned into plasmids pET-28a (pET-BSH) and pDR111 (pDR-BSH) for expressing the BSH protein in E. coli BL21 and Bacillus subtilis 168 (B-sub-BSH), respectively. His-tag-purified BSH from BL21 cells was evaluated by SDS-PAGE, Coomassie blue staining, and a Western blot (WB) assays. Secretory BSH from B. subtilis was analyzed by a Dot-Blot. B-sub-BSH was evaluated for the inhibition of C. perfringens growth. C. perfringens growth reached 7.8 log10 CFU/mL after 24 h culture. C. perfringens growth was at 8 vs. 7.4, 7.8 vs. 2.6 and 6 vs. 0 log10 CFU/mL in 0.2, 0.5, and 1 mM TDCA vs. DCA, respectively. Compared to TDCA, DCA reduced C. perfringens H2S production and the virulence gene expression of asrA1, netB, colA, and virT. BSH activity was observed in Lactobacillus johnsonii and B. longum under anaerobe but not L. johnsonii under 10% CO2 air. After the sequence alignment of bsh from ten bacteria, bsh from B. longum was selected, cloned into pET-BSH, and sequenced at 951 bp. After pET-BSH was transformed in BL21, BSH expression was assessed around 35 kDa using Coomassie staining and verified for His-tag using WB. After the subcloned bsh and amylase signal peptide sequence was inserted into pDR-BSH, B. subtilis was transformed and named B-sub-BSH. The transformation was evaluated using PCR with B. subtilis around 3 kb and B-sub-BSH around 5 kb. Secretory BSH expressed from B-sub-BSH was determined for His-tag using Dot-Blot. Importantly, C. perfringens growth was reduced greater than 59% log10 CFU/mL in the B-sub-BSH media precultured with 1 vs. 0 mM TDCA. In conclusion, TDCA was less potent than DCA against C. perfringens virulence, and recombinant secretory BSH from B-sub-BSH reduced C. perfringens growth, suggesting a new potential intervention against the pathogen-induced chicken NE.

Predators are dependent on the capture of prey to meet their energetic and nutritive requirements, which brings the risk of predation to prey. The predation risk is divided into consumptive and non-consumptive effects. Non-consumptive effects may manifest through altered growth and ontogenetic trajectories in prey species, a dynamic modulated by olfactory or other sensory cues from predators. Bactrocera dorsalis Hendel represents a major invasive threat to global horticulture. While earlier research was primarily centered on the consumptive interactions between B. dorsalis and its natural enemies, the potential consequences of non-consumptive interactions on the development of B. dorsalis have been overlooked. In this study, we investigated the impact of predation risk effects, induced by both visual exposure to the predatory mantis Hierodula patellifera Serville and its associated odor, on the life history traits of B. dorsalis. Female B. dorsalis demonstrated a reduced developmental time in the presence of a caged predator (H. patellifera) or predator odors, but showed significantly increased fecundity. Conversely, males displayed no significant change in developmental time. Additionally, neither the female nor male body weight at death was significantly influenced by the predation risk from the caged predator or predator odors. This study investigated the effects of predation risk on the development and reproduction of B. dorsalis, emphasizing the potential importance of odor risk in biological and pest control.

A Cu/AlCoCrFeNi2.1 composite was fabricated via solid-liquid method. In order to acquire the good interface bonding, the pulsed magnetic field (PMF) treatment was applied during the preparation of the Cu/AlCoCrFeNi2.1 composite. The effects of different processes and parameters, including pulse frequency on the interface microstructure, element distribution, diffusion thickness, and mechanical properties of the composite were investigated. The results demonstrated that PMF treatment effectively decreased the diffusion layer thickness of the composite, and improved mechanical properties of the Cu side. The diffusion thickness of the composite reduced from 31.22 μm to 6.20 μm after applying PMF. The action mechanism of PMF during preparation was revealed. The ultimate tensile strength, yield strength, and elongation of the Cu/AlCoCrFeNi2.1 composite via PMF were 192.6 MPa, 121.7 MPa, and 43.8%, respectively, which were higher than that of the composite without PMF. The maximum shear strengths of the composites with and without PFM were 120.2 MP and 137.4 MPa, respectively. When the pulse frequency was 2 Hz, the macro-segregation was eliminated to form the high-quality interface. This work paves a way for the performance improvement and complex parts preparation of layer composite by using PMF.

Effective weed control in the field is essential for maintaining favorable growing conditions and rapeseed yields. Sulfonylurea herbicides are one kind of most widely used herbicides worldwide, which control weeds by inhibiting acetolactate synthase (ALS). Molecular markers have been designed from polymorphic sites within the sequences of ALS genes, aiding marker-assisted selection in breeding herbicide-resistant rapeseed cultivars. However, most of them are not breeder friendly and have relatively limited application due to higher costs and lower throughput in the breeding projects. The aims of this study were to develop high throughput kompetitive allele-specific PCR (KASP) assays for herbicide resistance. We first cloned and sequenced BnALS1 and BnALS3 genes from susceptible cultivars and resistant 5N (als1als1/als3als3 double mutant). Sequence alignments of BnALS1 and BnALS3 genes for cultivars and 5N showed single nucleotide polymorphisms (SNPs) at positions 1676 and 1667 respectively. These two SNPs for BnALS1 and BnALS3 resulted in amino acid substitutions and were used to develop a KASP assay. These functional markers were validated in three distinct BC1F2 populations. The KASP assay developed in this study will be valuable for the high-throughput selection of elite materials with high herbicide resistance in rapeseed breeding programs.

Computational efficiency is significantly enhanced using artificial neural network-based computing. A two-terminal memristive device is a powerful electronic device that can mimic the behavior of a biological synapse in addition to storing information and performing logic operations. This work focuses on the fabrication of a memristive device that utilizes a resistive switching layer composed of polyvinyl alcohol infused with ZnO nanoparticles. By incorporating ZnO nanoparticles into the polymer film, the fabricated memristive devices exhibit functionalities that closely resemble those of biological synapses, including short-term and long-term plasticity, paired-pulse facilitation, and spike time-dependent plasticity. These findings establish the ZnO nanoparticle-polymer nanocomposite as a highly promising material for future neuromorphic systems.

IntroductionStreptococcus pneumoniae is an important human pathogen that may cause severe invasive pneumococcal diseases (IPDs) in young children and the elderly. A comprehensive comparative whole-genome analysis of invasive and non-invasive serotype strains offers great insights that are applicable to vaccine development and disease control.MethodsIn this study, 58 invasive (strains isolated from sterile sites) and 71 non-invasive (serotypes that have not been identified as invasive in our study) pneumococcal isolates were identified among the 756 pneumococcal isolates obtained from seven hospitals in Zhejiang, China (2010–2022). Serotyping, antimicrobial resistance tests, and genomic analyses were conducted to characterize these strains.Results and discussionThe three most invasive serotypes were 23F, 14, and 6B. The invasive pneumococcal isolates' respective resistance rates against penicillin, ceftriaxone, tetracycline, and erythromycin were 34.5%, 15.5%, 98.3%, and 94.7%. Whole-genome sequencing indicated that the predominant invasive clonal complexes were CC271, CC876, and CC81. The high rate of penicillin non-susceptible Streptococcus pneumoniae (PNSP) is related to the clonal distribution of resistance-conferring penicillin-binding proteins (PBP). Interestingly, we found a negative correlation between invasiveness and resistance in the invasive pneumococcal serotype strains, which might be due to the proclivity of certain serotypes to retain their β-lactam resistance. Moreover, the mutually exclusive nature of zmpC and rrgC+srtBCD suggests their intricate and potentially redundant roles in promoting the development of IPD. These findings reveal significant implications for pneumococcal vaccine development in China, potentially informing treatment strategies and measures to mitigate disease transmission.

Objective To investigate the risk factors associated with lower extremity deep vein thrombosis (LEDVT) and to establish a predictive model for patients who undergo gynecologic laparoscopic surgery. Methods A review of clinical data was conducted on patients who underwent gynecologic laparoscopic surgery between November 1, 2020, and January 31, 2022. Patients who developed LEDVT after surgery were included as the observation group, while the control group comprised patients who did not experience complications. Multivariate forward stepwise logistic regression models were used to identify independent risk factors associated with LEDVT. A nomogram model was then developed based on these risk factors. Results A total of 659 patients underwent gynecologic laparoscopic surgery during the study period, and 52 (7.89%) of these patients developed postoperative LEDVT. Ectopic mammary gland tissue in the vulva is an exceptionally rare disease. We present a case of a 62-year-old woman with a left vulvar mass of 30 years duration that progressively increased in size. The patient reported having pressure and discomfort, especially during movement. Surgical excision was performed, and a histopathological examination revealed a well-differentiated ectopic breast. We also review other cases of vulvar ectopic breast to further comprehend the characteristics of this rare disease. Clinicians and pathologists should always consider it as a differential diagnosis when presented with a vulvar mass.

Abstract The Maf proteins (Mafs) belong to basic leucine zipper transcription factors and are members of the activator protein-1 (AP-1) superfamily. There are two subgroups of Mafs: large Mafs and small Mafs, which are involved in a wide range of biological processes, such as the cell cycle, proliferation, oxidative stress, and inflammation. Therefore, dysregulation of Mafs can affect cell fate and is closely associated with diverse diseases. Accumulating evidence has established both large and small Mafs as mediators of tumor development. In this review, we first briefly describe the structure and physiological functions of Mafs. Then we summarize the upstream regulatory mechanisms that control the expression and activity of Mafs. Furthermore, we discuss recent studies on the critical role of Mafs in cancer progression, including cancer proliferation, apoptosis, metastasis, tumor/stroma interaction and angiogenesis. We also review the clinical implications of Mafs, namely their potential possibilities and limitations as biomarkers and therapeutic targets in cancer.

ABSTRACTApplication of titanium alloys is restricted due to their low hardness and poor wear resistance. Duplex surface treatment consisting of depositing of Al coating by arc ion plating (AIP) plus micro-arc oxidation (MAO) was carried out on Ti6Al4V alloy to improve its tribological performance. Ceramic coating mainly composed of α-Al2O3, γ-Al2O3 and mullite was fabricated on the aluminum-coated titanium alloy after the MAO treatment, and its thickness, compactness and hardness increased with the MAO time before the titanium alloy was oxidized. After the substrate titanium alloy was locally oxidized due to oxygen anions inward migration, a thin transition layer containing titanium oxides and aluminum titanate formed and incorporation of Ti in the alumina coating occurred. Occurrence of the strong eruptible discharges at these positions was facilitated probably due to the large difference in permittivity of the alumina and the titanium oxides, which led to the formation of penetrating cracks in the MAO coating. The hardness and compactness of the ceramic coating decreased. Tensile test results showed bonding strength of MAO ceramic coating/aluminum layer/titanium alloy substrate system was more than 40 MPa, and the maximum value could reach around 68 MPa. The bonding strength of the ceramic coating/titanium alloy substrate system decreased to around 34 MPa due to the weak bonding between the alumina ceramic coating and the titanium alloy substrate. Tribological test results show wear resistance of titanium alloy was greatly improved due to the fabrication of hard alumina ceramic coating on titanium alloy.

Large amounts of natural gas hydrates have been discovered in the Qiongdongnan Basin (QDNB), South China Sea. The chemical and stable carbon isotopic composition shows that the hydrate-bound gas was a mixture of thermogenic and microbial gases. It is estimated that microbial gas accounts for 40.96% to 60.58%, showing a trend of decrease with the increase in burial depth. A significant amount of gas hydrates is thought to be stored in the mass transport deposits (MTDs), exhibiting vertical superposition characteristics. The stable carbon isotopic values of methane (δ13C1) in the MTD1, located near the seabed, are less than −55‰, while those of the methane below the bottom boundary of MTD3 are all higher than −55‰. The pure structure I (sI) and structure II (sII) gas hydrates were discovered at the depths of 8 mbsf and 145.65 mbsf, respectively, with mixed sI and sII gas hydrates occurring in the depth range 58–144 mbsf. In addition, a series of indigenous organic matters and allochthonous hydrocarbons were extracted from the hydrate-bearing sediments, which were characterized by the origin of immature terrigenous organic matter and low-moderate mature marine algal/bacterial materials, respectively. More allochthonous (migrated) hydrocarbons were also discovered in the sediments below the bottom boundary of MTD3. The gas hydrated is “wet gas” characterized by a low C1/(C2 + C3) ratio, from 2.55 to 43.33, which was mainly derived from a deeply buried source kitchen at a mature stage. There is change in the heterogeneity between the compositions of gas and biomarkers at the site GMGS5-W08 along the depth and there is generally a higher proportion of thermogenic hydrocarbons at the bottom boundary of each MTDs, which indicates a varying contribution of deeply buried thermogenic hydrocarbons. Our results indicate that the MTDs played a blocking role in regulating the vertical transportation of hydrate-related gases and affect the distribution of gas hydrate accumulation in the QDNB.

Multiple sclerosis (MS) is a condition that affects the veins and small blood vessels. Previous research suggests that individuals with MS have an increased risk of vascular events and higher mortality rates. However, the relationship between MS and cerebral small vessel disease (CSVD) remains uncertain. This study aims to investigate the association between MS and lacunes. A prospective observational study was conducted, including a total of 112 participants, of which 46 had MS and 66 had CSVD. All participants underwent an MRI scan and a battery of neurological functional assessments. The presence of definite lacunes and black holes was determined through the analysis of T2-weighted, T1-weighted, and FLAIR images. The occurrence of lacunes in MS patients was found to be 19.6%. Notably, the duration of MS was identified as the sole risk factor for the development of lacune lesions in MS patients [odds ratio (OR) = 1.3, 95% confidence interval (CI) = 1.1–1.6, p = 0.008]. Comparatively, MS patients with lacunes exhibited a higher frequency of attacks and larger volumes of T2 lesions compared to MS patients without lacunes. Further analysis using receiver operating characteristic (ROC) curves showed that lacune lesions had limited ability to discriminate between MS and CSVD when disease duration exceeded 6 years. The presence of small arterial lesions in the brain of individuals with MS, along with the duration of the disease, contributes to the development of lacunes in MS patients.

Abstract As an emerging non‐invasive and easy‐to‐use toolkit, salivary analysis has garnered significant attention recently. By leveraging the latest discoveries in salivary biomarkers, salivary analysis has the potential to revolutionize the field of healthcare by providing valuable information for monitoring an individual's health status, diagnosing disease progression, and enabling personalized treatment. However, the transitional development of breakthrough salivary analysis remains an intriguing challenge. Only a few innovations have progressed to new transformative healthcare diagnosis and monitoring tools. In this review, we attempt to illustrate a bird's eye view of the emerging field of salivary analysis by exploring an assorted selection of seminar progress that can potentially shape the developing trajectory of the field of salivary analysis. The fundamental nature of saliva and the versatile robust uses of salivary analysis as a novel non‐invasive method in various healthcare diagnostic and monitoring fields will be thoroughly discussed. To inspire next‐stage development in this thriving and fruitful field, we also crystalize key findings, pinpoint challenges that remain to be addressed and present an outlook on the future transitional development of salivary analysis, invigorating future research of groundbreaking non‐invasive healthcare diagnosis and monitoring strategies, and beyond.

Drosophila melanogaster Down syndrome cell adhesion molecule 1 (Dscam1) encodes 19,008 diverse ectodomain isoforms via the alternative splicing of exon 4, 6, and 9 clusters. However, whether individual isoforms or exon clusters have specific significance is unclear. Here, using phenotype-diversity correlation analysis, we reveal the redundant and specific roles of Dscam1 diversity in neuronal wiring. A series of deletion mutations were performed from the endogenous locus harboring exon 4, 6, or 9 clusters, reducing to 396 to 18,612 potential ectodomain isoforms. Of the 3 types of neurons assessed, dendrite self/non-self discrimination required a minimum number of isoforms (approximately 2,000), independent of exon clusters or isoforms. In contrast, normal axon patterning in the mushroom body and mechanosensory neurons requires many more isoforms that tend to associate with specific exon clusters or isoforms. We conclude that the role of the Dscam1 diversity in dendrite self/non-self discrimination is nonspecifically mediated by its isoform diversity. In contrast, a separate role requires variable domain- or isoform-related functions and is essential for other neurodevelopmental contexts, such as axonal growth and branching. Our findings shed new light on a general principle for the role of Dscam1 diversity in neuronal wiring.

With the continuous development of the economy and society, the production of solid waste is increasing. Construction and demolition waste (CDW), used tires and silica fume are the main components of solid waste. Recycling these materials will reduce resource pressure, decrease carbon emissions and protect the environment, while bringing certain economic benefits. This study focused on the influence of the synergistic effect of recycled steel fibers (RSF) and silica fume on the mechanical properties and impact resistance of recycled aggregate concrete (RAC). The mathematical statistical model of Weibull distribution was adopted to depict the impact results. In addition, the microstructure of the interfacial transition zone (ITZ) between the cement paste and the aggregate/fiber was researched by scanning electron microscopy (SEM). Results revealed that the synergistic effect of RSF and silica fume significantly improved the impact resistance of RAC. Especially the specimen containing 10% silica fume and 0.75% RSF performed the best properties, the first crack impact and failure impact energy increased by 421.9% and 461.3%, the net-gain in them by the synergistic action of silica fume and RSF with values of 102.7% and 115.3%. The distribution law of the impact life for RSF reinforced RAC can better describe by the two-parameter Weibull distribution function. Impact resistance life was predicted corresponding to reliability levels. SEM investigations revealed that the ITZ between cement and aggregate/RSF significantly improved by mixing silica fume.

Background: Kidney renal papillary cell carcinoma (KIRP) is a rare malignancy with a very poor prognosis. Anoikis is a specific form of apoptosis involved in carcinogenesis, but the role of anoikis in KIRP has not been explored.Methods: Anoikis-related genes (ARGs) were obtained from the GeneCards database and Harmonizome database and were used to identify different subtypes of KIRP and construct a prognostic model of KIRP. In addition, we also explored the immune microenvironment and enrichment pathways among different subtypes by consensus clustering into different subtypes. Drug sensitivity analysis was used to screen for potential drugs. Finally, we verified the mRNA and protein expression of the independent prognostic gene PLK1 in patient tissues and various cells and further verified the changes in relevant prognostic functions after constructing a PLK1 stable knockdown model using ShRNA.Results: We identified 99 differentially expressed anoikis-related genes (DEGs) associated with KIRP survival, and selected 3 genes from them to construct a prognostic model, which can well predict the prognosis of KIRP patients. Consensus clustering divided KIRP into two subtypes, and there was a significant difference in survival rates between the two subtypes. Immune profiling revealed differing immune statuses between the two subtypes, and functional analysis reveals the differential activity of different functions in different subtypes. Drug sensitivity analysis screened out 15 highly sensitive drugs in the high-risk group and 11 highly sensitive drugs in the low-risk group. Univariate and multivariate Cox regression analysis confirmed that PLK1 was an independent prognostic factor in KIRP, and its mRNA and protein expression levels were consistent with gene differential expression levels, both of which were highly expressed in KIRP. Functional verification of PLK1 in KIRP revealed significant results. Specifically, silencing PLK1 inhibited cell proliferation, clonogenicity, and migration, which indicated that PLK1 plays an important role in the proliferation and migration of KIRP.Conclusion: The prognosis model constructed by ARGs in this study can accurately predict the prognosis of KIRP patients. ARGs, especially PLK1, play an important role in the development of KIRP. This research can help doctors provide individualized treatment plans for KIRP patients and provide researchers with new research ideas.

ABSTRACT The treatment of extensively drug-resistant (XDR) A. baumannii has emerged as a major problem. Tigecycline (TGC) and sulbactam (SUL) are both effective antibiotics against XDR A. baumannii. Here, we investigated the in-host evolution and mechanism of collateral sensitivity (CS) phenomenon in development of tigecycline resistance accompanied by a concomitant increase of sulbactam susceptibility. A total of four XDR A. baumannii strains were sequentially isolated from the same patient suffering from bacteremia. Core-genome multilocus sequence typing separated all the strains into two clusters. Comparative analysis of isolate pair 1 revealed that multiplication of blaOXA-23 within Tn2006 on the chromosome contributed to the change in the antimicrobial susceptibility phenotype of isolate pair 1. Additionally, we observed the emergence of CS to sulbactam in isolate pair 2, as demonstrated by an 8-fold increase in the TGC MIC with a simultaneous 4-fold decrease in the SUL MIC. Compared to the parental strain Ab-3557, YZM-0406 showed partial deletion in the two-component system sensor adeS. Reconstruction of the adeS mutant in Ab-3557 in situ suggested that TGC resistance and CS to SUL were mainly caused by the mutation of adeS. Overall, our study reported a novel CS combination of TGC and SUL in A. baumannii and further revealed a mechanism of CS attributed to the mutation of adeS. This study provides a valuable foundation for developing effective regimens and sequential combinations of tigecycline and sulbactam against XDR A. baumannii. IMPORTANCE Collateral sensitivity (CS) has become an increasingly common evolutionary trade-off during adaptive bacterial evolution. Here, we report a novel combination of tigecycline (TGC) resistance and CS to sulbactam (SUL) in A. baumannii. TGC and SUL are both effective antibiotics against XDR A. baumannii, and it is essential to reveal the mechanism of CS between TGC and SUL. In our study, the partial deletion of adeS, a two-component system sensor, was confirmed to be the key factor contributing to this CS phenomenon. This study provides a valuable foundation for developing effective regimens and sequential combinations of tigecycline and sulbactam against XDR A. baumannii.

Cement-based materials in underground structures, bridge foundation, and offshore platforms often face sulfate environments. It has been widely reported that the compressive strength (CS) of these materials post sulfate attack is crucial for structural stability and durability. However, existing experimental and analytical methods require extensive resources and lengthy period. For addressing this challenge, therefore, a novel stacking model is proposed to accurately predict the CS of cement base under sulfate attack in this study. The stacking model integrates Support Vector Regression (SVR), Random Forest (RF), and Extreme Gradient Boosting (XGB) as base learners, with Linear Regression (LR) as the meta-learner. In this novel model training, the 330 data sets used are obtained from the relevant literatures. The water-cement ratio, cement content, water content, sand content, sulfate concentration, wetting temperature, wetting time, drying temperature, drying time, exposure time are selected as inputs while the output is CS of cement-based materials subjected to sulphate deterioration. To improve the forecast capability, particle swarm optimization (PSO) is adopted for optimizing hyperparameters of stacking model, which can be implemented by reducing the discrepancy between model prediction and measured CS. By comparing with the results of experiments and the solely machine learning (ML) models, it is found that the stacking model optimized by PSO presents the best prediction accuracy of R²=0.992, RMSE=2.248 MPa, and MAE=1.782 MPa. Moreover, the examination of the influence of individual input variables on the CS of cement-based material subjected to sulfate attack is conducted through the application of Shapley Additive Explanations (SHAP) and Partial Dependence Plot (PDP) analyses. The PSO optimized stacking model proposed in this study can effectively predict the CS of cement-based materials under sulfate attack. Our study provides a new reference for future research in this field.
