Primary sclerosing cholangitis (PSC) is a chronic liver disease characterized by inflammatory responses and fibrotic scar formation leading to cholestasis. Ductular reaction and liver fibrosis are typical liver changes seen in human PSC and cholestasis patients. The current study aimed to clarify the role of liver-specific microRNA-34a in the cholestasis-associated ductular reaction and liver fibrosis. We demonstrated that miR-34a expression was significantly increased in human PSC livers along with the enhanced ductular reaction, cellular senescence, and liver fibrosis. A liver-specific miR-34a knockout mouse was established by crossing floxed miR-34a mice with albumin-promoter-driven Cre mice. Bile duct ligation (BDL) induced liver injury characterized by necrosis, fibrosis, and immune cell infiltration. In contrast, liver-specific miR-34a knockout in BDL mice resulted in decreased biliary ductular pathology associated with the reduced cholangiocyte senescence and fibrotic responses. The miR-34a-mediated ductular reactions may be functioning through Sirt-1-mediated senescence and fibrosis. The hepatocyte-derived conditioned medium promoted LPS-induced fibrotic responses and senescence in cholangiocytes, and miR-34a inhibitor suppressed these effects, further supporting the involvement of paracrine regulation. In conclusion, we demonstrated that liver-specific miR-34a plays an important role in ductular reaction and fibrotic responses in a BDL mouse model of cholestatic liver disease. Heat shock protein 47 (Hsp47) is a collagen-specific molecular chaperone that is indispensable for molecular maturation of collagen. In this study, hsp47 and hsp47-like cDNAs were cloned and characterized in grass carp (Ctenopharyngodon idella). The cDNAs were 1212 and 1218 base pairs long, respectively, and included an open reading frame encoding 403 and 405 amino acids. The molecular phylogeny based on the deduced amino acid sequences indicated that the correct sequences of the hsp47 and hsp47-like cDNA were obtained and the deduced proteins clustered distinctly into teleost clades. Primary structure analysis and characterization of Hsp47 and Hsp47-like shared the basic structure and biofunctions of Hsp47 in vertebrates. The spatial pattern of gene expression revealed that hsp47 and hsp47-like were relatively ubiquitous in different tissues and highly expressed in heart and skin. The expression levels of hsp47 and hsp47-like and type I collagen mRNAs varied similarly in different tissues. Type I collagen content increased significantly with the increase of water velocity in the muscle of grass carp in response to aerobic exercise. Among the gene expression changes of hsp47, hsp47-like, col1a1 and col1a2 in muscle that occurred in response to aerobic exercise, the change of type I collagen was most strongly correlated with hsp47 expression. Additionally, col1a1 showed the highest correlation with hsp47-like and col1a2 showed the highest correlation with hsp47. These findings suggest that grass carp Hsp47 and Hsp47-like are closely related to type I collagen synthesis. Cathepsin S (CTSS) is a widely expressed cysteinyl protease that has garnered attention because of its enzymatic and non-enzymatic functions under inflammatory and metabolic pathological conditions. Here, we examined whether CTSS participates in stress-related skeletal muscle mass loss and dysfunction, focusing on protein metabolic imbalance. Eight-week-old male wildtype (CTSS+/+) and CTSS-knockout (CTSS-/-) mice were randomly assigned to non-stress and variable-stress groups for 2 weeks, and then processed for morphological and biochemical studies. Compared with non-stressed mice, stressed CTSS+/+ mice showed significant losses of muscle mass, muscle function, and muscle fiber area. In this setting, the stress-induced harmful changes in the levels of oxidative stress-related (gp91phox and p22phox,), inflammation-related (SDF-1, CXCR4, IL-1β, TNF-α, MCP-1, ICAM-1, and VCAM-1), mitochondrial biogenesis-related (PPAR-γ and PGC-1α) genes and/or proteins and protein metabolism-related (p-PI3K, p-Akt, p-FoxO3α, MuRF-1, and MAFbx1) proteins; and these alterations were rectified by CTSS deletion. Metabolomic analysis revealed that stressed CTSS-/- mice exhibited a significant improvement in the levels of glutamine metabolism pathway products. Thus, these findings indicated that CTSS can control chronic stress-related skeletal muscle atrophy and dysfunction by modulating protein metabolic imbalance, and thus CTSS was suggested to be a promising new therapeutic target for chronic stress-related muscular diseases. We used whole cell recordings from slice preparations of mouse cortex to identify various inputs to neurons of layer 1. Two sensory cortical areas were targeted: a primary somatosensory area, namely, the barrel cortex of S1, and a higher order visual area, namely, V2M. Results were similar from both areas. By activating local inputs using photostimulation with caged glutamate, we also identified glutamatergic (and possibly GABAergic) inputs from all lower layers plus GABAergic inputs from nearby layer 1 neurons. However, the patterns of such inputs to layer 1 neurons showed great variation among cells. In separate experiments, we found that electrical stimulation of axons running parallel to the cortical surface in layer 1 also evoked a variety of convergent input types to layer 1 neurons, including glutamatergic "drivers" and "modulators" plus classic modulatory inputs, including serotonergic, nicotinic, α-and β-adrenergic, from subcortical sites. Given that these layer 1 cells significantly affect the responses of other cortical neurons, especially via affecting the apical dendrites of pyramidal cells so important to cortical functioning, their role in cortical processing is significant. Plant immunity is initiated by extracellular detection of pathogen-associated molecular patterns (PAMPs) through surface-localized pattern recognition receptors (PRRs). PRR activation induces many responses including the activation of mitogen-activated protein kinases (MAPKs) that ultimately limit bacterial growth. Previous work identified Arabidopsis MAP kinase phosphatase 1 (MKP1) as a negative regulator of signaling pathways required for some, but not all, of PAMP-initiated responses. Specifically, loss of MAPK MPK6 in an mkp1 background suppressed a subset of the mkp1-dependent biological phenotypes, indicating the requirement for MPK6 in MKP1-dependent signaling. To further genetically separate the outputs of PAMP-responsive signaling pathways, we performed a transcriptome analysis in Arabidopsis wild type, mkp1 and mkp1 mpk6 seedlings treated with the bacterially derived PAMP elf26 for 0, 30, and 90 min. Using differential genetic and temporal clustering analyses between and within genotypes, we identified and separated 6963 elf26-responsive transcripts based on both genetic requirements of MKP1 (with or without a requirement for MPK6) and temporal transcriptional accumulation patterns, and some of these novel response markers were validated by qRT-PCR over a more extended time course. A prior study demonstrated that icariin (ICA) could repress angiotensin II-induced apoptosis in H9c2 cells. The activation of mitochondrial apoptotic pathways may play a crucial role in this phenomenon. In this study, we explored the potential protective roles of ICA in apoptosis in cardiomyocytes, cardiac remodelling, and the underlying mechanisms with regard to the mitochondrial apoptotic pathway in rats with spontaneous hypertension. The oral administration of ICA (20 and 40 mg/kg/d) inhibited cardiomyocyte apoptosis and ameliorated left heart ventricle remodelling and abnormal mitochondria. ICA also decreased the blood pressure of model rats. ICA treatment increased the expression of Bcl-2 and decreased the expression of p53, Bax, Bok and cleaved caspase 3 in model rats, which suggests the potential mechanism underlying this effect. In summary, ICA inhibits the apoptosis of cardiomyocytes and ameliorates cardiac remodelling. The potential mechanism may relate to the inhibition of the mitochondrial apoptotic pathway. Love wave sensors have been widely used for sensing applications. In this work, we introduce the theoretical analysis of the monolayer and double-layer waveguide Love wave sensors. The velocity, particle displacement and energy distribution of Love waves were analyzed. Using the variations of the energy repartition, the sensitivity coefficients of Love wave sensors were calculated. To achieve a higher sensitivity coefficient, a thin gold layer was added as the second waveguide on top of the silicon dioxide (SiO2) waveguide–based, 36 degree–rotated, Y-cut, X-propagating lithium tantalate (36° YX LiTaO3) Love wave sensor. The Love wave velocity was significantly reduced by the added gold layer, and the flow of wave energy into the waveguide layer from the substrate was enhanced. By using the double-layer structure, almost a 72-fold enhancement in the sensitivity coefficient was achieved compared to the monolayer structure. Additionally, the thickness of the SiO2 layer was also reduced with the application of the gold layer, resulting in easier device fabrication. Ability to characterize the heat transfer in flowing gases is important for a wide range of applications involving micro- and nanoscale devices. Gas flows away from the continuum limit can be captured using the Boltzmann equation, whose analytical solution poses a formidable challenge. An efficient and accurate numerical simulation of the Boltzmann equation is thus highly desirable. In this article, the linearized Boltzmann Bhatnagar-Gross-Krook equation is used to develop a hierarchy of thermal lattice Boltzmann (LB) models based on half-space Gaussian-Hermite (GH) quadrature ranging from low to high algebraic precision, using double distribution functions. Simplified versions of the LB models in the continuum limit are also derived, and are shown to be consistent with existing thermal LB models for noncontinuum heat transfer reported in the literature. Accuracy of the proposed LB hierarchy is assessed by simulating thermal Couette flows for a wide range of Knudsen numbers. Effects of the underlying quadrature schemes (half-space GH vs full-space GH) and continuum-limit simplifications on computational accuracy are also elaborated. The numerical findings in this article provide direct evidence of improved computational capability of the proposed LB models for modeling noncontinuum flows and heat transfer at small length scales. The thalamic reticular nucleus (TRN) is a thin layer of GABAergic cells lying rostral and lateral to the dorsal thalamus, and its projection to thalamic relay cells (i.e., the reticulothalamic pathway) strongly inhibits these cells. In an attempt to extend earlier studies of reticulothalamic connections to sensory thalamic nuclei, we used laser-scanning photostimulation to study the reticulothalamic projections to the main motor thalamic relays, the ventral anterior and lateral (VA and VL) nuclei, as well as to the nearby central lateral (CL) thalamic nucleus. VA/VL and the earlier studied somatosensory thalamic nuclei are considered "core" nuclei with topographic thalamocortical projections, whereas CL is thought to be a "matrix" nucleus with diffuse thalamocortical projections. We found that the TRN input footprints to VA/VL and CL are spatially localized and topographic and generally conform to the patterns established earlier for the TRN projections to sensory thalamic relays. These remarkable similarities suggest similar organization of reticulothalamic pathways and TRN regulation of thalamocortical communication for motor and sensory systems and perhaps also for core and matrix thalamus. Furthermore, we found that VA/VL and CL shared overlapping TRN input regions, suggesting that CL may also be involved in the relay of motor information. [ABSTRACT FROM AUTHOR]

Genes encoding the virulence-promoting type III secretion system (T3SS) in phytopathogenic bacteria are induced at the start of infection, indicating that recognition of signals from the host plant initiates this response. However, the precise nature of these signals and whether their concentrations can be altered to affect the biological outcome of host-pathogen interactions remain speculative. Here we use a metabolomic comparison of resistant and susceptible genotypes to identify plant-derivedmetabolites that induce T3SS genes in Pseudomonas syringae pv tomato DC3000 and report that mapk phosphatase 1 (mkp1), an Arabidopsis mutant that is more resistant to bacterial infection, produces decreased levels of these bioactive compounds. Consistent with these observations, T3SS effector expression and delivery by DC3000 was impaired when infecting the mkp1 mutant. The addition of bioactive metabolites fully restored T3SS effector delivery and suppressed the enhanced resistance in the mkp1 mutant. Pretreatment of plants with pathogen-associated molecular patterns (PAMPs) to induce PAMPtriggered immunity (PTI) also restricts T3SS effector delivery and enhances resistance by unknown mechanisms, and the addition of the bioactive metabolites similarly suppressed both aspects of PTI. Together, these results demonstrate that DC3000 perceives multiple signals derived from plants to initiate its T3SS and that the level of these host-derived signals impacts bacterial pathogenesis. Probiotic bacteria are known to exert a wide range of beneficial effects on their animal hosts. Therefore, the present study explored the effect of the supernatants obtained from Lactobacillus delbrueckii fermentation (LBF) on colon cancer. The results indicated that the proliferation of LBF solution-treated colon cancer SW620 cells was arrested and accumulated in the G1 phase in a concentration-dependent manner. The LBF solution efficiently induced apoptosis through the intrinsic caspase 3-depedent pathway, with a corresponding decreased expression of Bcl-2. The activity of matrix metalloproteinase 9, which is associated with the invasion of colon cancer cells, was also decreased in the LBF-treated cells. In conclusion, the results demonstrate the antitumor effect of LBF in vitro and may contribute to the development of novel therapies for the treatment of colon cancer. [ABSTRACT FROM AUTHOR]

Oscillatory gas flows are commonly generated by micro- and nanoelectromechanical systems. Due to their small size and high operating frequencies, these devices often produce noncontinuum gas flows. Theoretical analysis of such flows requires solution of the unsteady Boltzmann equation, which can present a formidable challenge. In this article, we explore the applicability of the lattice Boltzmann (LB) method to such linear oscillatory noncontinuum flows; this method is derived from the linearized Boltzmann Bhatnagar-Gross-Krook (BGK) equation. We formulate four linearized LB models in the frequency domain, based on Gaussian-Hermite quadratures of different algebraic precision (AP). The performance of each model is assessed by comparison to high-accuracy numerical solutions to the linearized Boltzmann-BGK equation for oscillatory Couette flow. The numerical results demonstrate that high even-order LB models provide superior performance over the greatest noncontinuum range. Our results also highlight intrinsic deficiencies in the current LB framework, which is incapable of capturing noncontinuum behavior at high oscillation frequencies, regardless of quadrature AP and the Knudsen number. Background: Reversible assembly and disassembly of nanostructures has important function in controllable construction of nanodevices. There are several methods to achieve reversible assembly/disassembly, such as pH, temperature, DNA hybridization and so on. Among these methods, pH driven reversible assembly presents superiority due to its ease-of-use and no waste produced. Herein we report a novel design that use two single-stranded (ss) DNAs wrapped single walled carbon nanotubes (SWCNTs) for the pH controlled assembly of SWCNTs without generation of waste. Results: Both of the two DNAs with a same wrapping sequence of d(GT)20 and different free terminals showed a very high tendency to wrap around carbon nanotubes. The assembly was driven by the hybridization between the two free terminals of wrapped DNAs on the neighboring SWCNTs: i-motif (four-stranded C-quadruplex) and its complemental stranded G-quadruplex which would form tight tetraplexes and break the hybridization under slightly acidic conditions. Thus the assembly and disassembly are reversibly controlled by pH. And this assembly/ disassembly process can be easily distinguished by naked eyes. Gel electrophoresis and Atomic Force Microscope are used to demonstrate the assembly and disassembly of SWCNTs at different pH. Conclusions: A novel pH induced reversible assembly and disassembly of SWCNTs was realized which may have potential applications in the area of controlled assembly of nanostructures. MicroRNAs are a class of small noncoding RNAs that function as key regulators of gene expression at the post-transcriptional level. In this study, we demonstrate that miR-214 is frequently down-regulated in cervical cancer, and its expression reduces the proliferation, migration, and invasiveness of cervical cancer cells, whereas inhibiting its expression results in enhanced proliferation, migration, and invasion. miR-214 binds to the 3'-UTR of UDP-N-acetyl-α-D-galactosamine: polypeptide N-acetylgalactosaminyltransferase 7 (GALNT7), thereby repressing GALNT7 expression. Furthermore, we are the first to show, using quantitative real-time PCR, that GALNT7 is frequently up-regulated in cervical cancer. The knockdown of GALNT7 markedly inhibits cervical cancer cell proliferation, migration, and invasion, whereas ectopic expression of GALNT7 significantly enhances these properties, indicating that GALNT7 might function as an oncogene in cervical cancer. The restoration of GALNT7 expression can counteract the effect of miR-214 on cell proliferation, migration, and invasiveness of cervical cancer cells. Together, these results indicate that miR-214 is a new regulator of GALNT7, and both miR- 214 and GALNT7 play important roles in the pathogenesis of cervical cancer. [ABSTRACT FROM AUTHOR]

Modeling gas flows generated by micro- and nano-devices often requires the use of kinetic theory. To facilitate implementation, various approximate formulations have been proposed based on the Bhatnagar-Gross-Krook (BGK) kinetic model, including most recently, the lattice Boltzmann (LB) method. While there exists a comprehensive numerical data set for the hard sphere linearized Boltzmann equation for steady Couette flow, no such set of data is available for the Boltzmann-BGK equation. The purpose of this article is to present a high accuracy data set for the linearized Boltzmann-BGK equation over the full range of Knudsen numbers and normalized oscillation frequencies - this encompasses both steady and unsteady Couette flows. This data set is expected to be of particular value in the benchmarking and validation of computational methods such as the LB method and other approaches based on the Boltzmann-BGK equation. Most axons connecting the thalamus and cortex in both directions pass through the thalamic reticular nucleus (TRN), a thin layer of GABAergic cells adjacent to the thalamus, and innervate neurons there. The TRN, therefore, is in a strategic location to regulate thalamocortical communication. We recorded neurons of the somatosensory region of the TRN in a thalamocortical slice preparation and studied the spatial organization of their thalamic input using laser scanning photostimulation. We show that the thalamoreticular pathway is organized topographically for most neurons. The somatosensory region of the TRN can be organized into three tiers. From the inner (thalamoreticular) border to the outer, in a manner roughly reciprocal to the reticulothalamic pathway, each of these tiers receives its input from one of the somatosensory relays of the thalamus-the posterior medial, ventroposterior medial, and ventroposterior lateral nuclei. What is surprising is that approximately a quarter of the recorded neurons received input from multiple thalamic regions usually located in different nuclei. These neurons distribute evenly throughout the thickness of the TRN. Our results, therefore, suggest that there exist a subpopulation of TRN neurons that receive convergent inputs from multiple thalamic sources and engage in more complex patterns of inhibition of relay cells. We propose these neurons enable the TRN to act as an externally driven "searchlight" that integrates cortical and subcortical inputs and then inhibits or disinhibits specific thalamic relay cells, so that appropriate information can get through the thalamus to the cortex. BoNTs (botulinum neurotoxins) are both deadly neurotoxins and natural toxins that are widely used in protein therapies to treat numerous neurological disorders of dystonia and spinal spasticity. Understanding the mechanism of action and substrate specificity of BoNTs is a prerequisite to develop antitoxin and novel BoNT-derived protein therapy. To date, there is a lack of detailed information with regard to how BoNTs recognize and hydrolyse the substrate VAMP-2 (vesicle-associated membrane protein 2), even though it is known to be cleaved by four of the seven BoNT serotypes, B, D, F, G and TeNT (tetanus neurotoxin). In the present study we dissected the molecular mechanisms of VAMP-2 recognition by BoNT serotype F for the first time. The initial substrate recognition was mediated through sequential binding of VAMP-2 to the B1, B2 and B3 pockets in LC/F (light chain of BoNT serotype F), which directed VAMP-2 to the active site of LC/F and stabilized the active site substrate recognition, where the P2, P1′ and P2′ sites of VAMP-2 were specifically recognized by the S2, S1′ and S2′ pockets of LC/F to promote substrate hydrolysis. The understanding of the molecular mechanisms of LC/F substrate recognition provides insights into the development of antitoxins and engineering novel BoNTs to optimize current therapy and extend therapeutic interventions. Ethnopharmacological relevance: This study examined the protective effects of total saponins from Ornithogalum saundersiae (Liliaceae) on d-galactosamine (d-GalN) and lipopolysaccharide (LPS) – induced fulminant hepatic failure. Materials and methods: Total saponins of Ornithogalum saundersiae (Liliaceae) (OC) were prepared with ethyl alcohol extract from bulbs of the plant. Mice were given an intraperitoneal injection of d-GalN (700mg/kg)/LPS (10μg/kg). OC (100mg/kg, 200mg/kg and 300mg/kg) was administered orally for 3 days continuously, and at the last day at 1h before the d-GalN/LPS injection. Mice were sacrificed at 8h after the d-GalN/LPS injection. The liver injury was assessed biochemically, investigating aspartate aminotransferase (AST), alanine aminotransferase (ALT), malondialdehyde (MDA), glutathione (GSH) activities, and the expressions of caspase-3 and hypoxia inducible factor-1α (HIF-1α) as well. Tumor necrosis factor (TNF-α) content was measured after d-GalN/LPS induced 1h by ELISA assay. The survival rates after application of OC in 24h also were observed. Results: d-GalN/LPS increased the serum aminotransferase levels and lipid peroxidation, while decreased the reduced glutathione level. The pretreatment with OC attenuated these changes in a dose-dependent manner. Elevation of TNF-α level and activation of caspase-3, HIF-1α were observed in the d-GalN/LPS group, which was attenuated by OC. The survival rate of the OC groups was significantly higher than that of the d-GalN/LPS group. Conclusions: Protection afforded by OC against d-GalN/LPS-induced fulminant hepatic failure is the result of reduced oxidative stress, inhibited expression of caspase-3, HIF-1α, and anti-apoptotic activity. In this work, we report a multiplexed electrochemical DNA sensor for highly specific single-nucleotide polymorphism (SNP) detection by using oligonucleotide-incorporated nonfouling surfaces (ONS). A typical “sandwich” scheme was employed to perform the SNP assay. The presence of the target DNA templated the ligation between the capture probe DNA anchored on gold electrodes and the tandem reporter probe tagged with a biotin moiety, which could be transduced to peroxidase-based catalyzed amperometric signals. This method could effectively differentiate SNP sites with only one-base mismatch. Importantly, the differentiation ratio was significantly higher than that with surfaces without the nonfouling property, which clearly demonstrated the superiority of the ONS strategy. Background: MicroRNAs (miRNAs) are a new class of naturally occurring, small, non-coding RNAs that regulate protein-coding mRNAs by causing mRNA degradation or repressing translation. The roles of miRNAs in lineage determination and proliferation, as well as the localization of several miRNA genes at sites of translocation breakpoints or deletions, have led to speculation that miRNAs could be important factors in the development or maintenance of the neoplastic state. Results: We showed that miR-9 was downregulated in human gastric adenocarcinoma. Overexpression of miR-9 suppressed the growth of human gastric adenocarcinoma cell line MGC803 cell as well as xenograft tumors derived from them in SCID mice. Bioinformatics analysis indicated a putative miR-9 binding site in the 3'- untranslated region (3'UTR) of the tumor-related gene κ1 mRNA. In an EGFP reporter system, overexpression of miR-9 downregulated EGFP intensity, and mutation of the miR-9 binding site abolished the effect of miR-9 on EGFP intensity. Furthermore, both the κ1 mRNA and protein levels were affected by miR-9. Finally, knockdown of κ1 inhibited MGC803 cell growth in a time-dependent manner, while ectopic expression of κ1 could rescue MGC803 cell from growth inhibition caused by miR-9. Conclusion: These findings indicate that miR-9 targets κ1 and regulates gastric cancer cell growth, suggesting that miR-9 shows tumor suppressive activity in human gastric cancer pathogenesis. Heterogeneous palladium catalysts have been supported on the ordered mesoporous carbons (Pd/OMC) with bimodal pores which are prepared by the surfactant-templating approach. Characterization using XRD, TEM, XPS, H2 chemisorption, and N2 sorption techniques reveals that the Pd/OMC catalysts have the ordered 2-D hexagonal mesostructure (space group of p6 mm), extremely high surface areas (~1800 m2/g), large pore volumes (~1.64 cm3/g), bimodal pores (6.3 nm of primary mesopores and 1.7 nm of secondary mesopores inside the pore walls), hydrophobic carbon surface, and small metal particles well-dispersed inside the secondary small mesopores. This catalyst exhibits a high yield of 43% for biphenyl from the Ullmann coupling reaction of chlorobenzene in water at 100 °C without assistance of any phase transfer catalyst and can be reused up to 10 times, providing potential opportunities for industrial applications such as coupling and hydrogenation reactions. A linear fluorinated bis-phenol-A novolac resin (LFAR) for optical waveguide was synthesized based on 4,4′-(hexafluoro-isopropylidene)diphenol, epoxy chloropropane and formaldehyde. Negative fluorinated photoresist (FP) was made by composing the LFAR, diphenyl iodonium salt and solvent. The film, which was made by spin-coating FP, had good UV light lithograph sensitivity, large hardness and high glass transition temperature ( T g>170°C, after crosslinking). Low-loss optical waveguides with very smooth top surface were fabricated from the resulting FP by direct UV exposure and chemical development. We demonstrate, for the first time, the application of ordered mesoporous carbons with large pore sizes prepared from the surfactant-templating approach in efficient disposal of wastewater containing bulky dye molecules. The adsorption amount for the bulky dye (methylthionine chloride, fuchsin basic, rhodamine B, brilliant yellow, methyl orange, or Sudan G) is almost twice that of the activated carbon in which mesopores contribute almost 100% to the total surface area and volume. The ordered mesoporous carbon adsorbent has a high adsorption rate (>99.9%) for low-concentration dyes, good performance in decoloration regardless of the dye nature, including basic, acidic, or azo dyes, and high stability after dye elution. To establish the relationship between the pore texture and adsorption properties, three kinds of ordered mesoporous carbons with different pore sizes, surface areas, and pore volumes have been synthesized by using phenolic resins as carbon sources and triblock copolymer as a structure-directing agent. The XRD, TEM, and N2sorption measurements reveal that all mesoporous carbonaceous materials have the highly ordered 2D hexagonal mesostructure, high surface areas (398−2580 m2/g), large pore volumes (0.51−2.16 cm3/g), and uniform pore sizes ranging from 4.5 to 6.4 nm. The adsorption capacities are compared and the pore occupation is estimated to understand the adsorption behaviors in the ordered mesopores with different diameters and models. The spatial effect of dye molecules is the determinative factor for the adsorption in ordered mesoporous carbons with various pore textural properties. The mesoporous carbon with an extremely high surface area (2580 m2/g), a large pore volume (2.16 cm3/g), and bimodal pores (6.4 and 1.7 nm) prepared from the silica−carbon composite shows the highest adsorption capacities for bulky basic dyes among the three ordered mesoporous carbons. In this work, we report an enzyme-based F-DNA sensor for the sequence-specific detection of nucleic acids. This DNA sensor employs a "stem-loop" DNA probe dually labeled with biotin and digoxigenin (DIG). The probe is immobilized at an avidin-modified electrode surface via the biotin-avidin bridge, and the DIG serves as an affinity tag for the enzyme binding. In the initial state of the sensor, the probe adopts the stem-loop structure, which shields DIG from being approached by a bulky horseradish peroxidase- linked-anti-DIG antibody (anti-DIG-HRP) due to the steric effect. After hybridization, the probe undergoes a significant conformational change, forcing DIG away from the electrode. As a result, the DIG label becomes accessible by the anti-DIG-HRP, and the target hybridization event can be sensitively transduced via the enzyrnatically amplified electrochemical current signal. By using this new strategy, we demonstrate that the prototype E-DNA sensor has been able to detect as low as femtomolar DNA targets with excellent differentiation ability for even single mismatches. Ordered mesostructured LaF 3nanoarrays have been, for the first time, synthesized via a one-step nanocasting process using La(CF 3COO) 3as a precursor and mesoporous silica as a hard template. The characterization of SAXS and XRD patterns and TEM and SEM images shows that the LaF 3nanowire arrays have long-range regularity of hexagonal mesostructure ( p6 mm) and single-crystalline feature. N 2-sorption isotherms reveal that ordered mesoporous LaF 3products have high BET specific surface area (∼75 m 2/g), large pore volume (0.15 cm 3/g), and narrow pore-size distribution (the mean value of 4.3 nm). The upconversion fluorescence has been realized in the Yb 3+/Er 3+(red/green) and Yb 3+/Tm 3+(blue) codoped LaF 3nanoarrays by upconversion (UC) excitation in the near-infrared region. The UC emission population for 4F 9/2, 2H 11/2, 4S 3/2, and 2H 9/2levels in the Yb 3+/Er 3+codoped LaF 3nanowire array matrixes depends on Er 3+concentration, the excitation density, and the specific surface areas. Supramolecular aggregateʼs self-assembling approach has derived diverse mesostructured inorganic solids. Recent progresses involve ordered mesoporous carbonaceous frameworks. This review paper summarizes the synthesis of ordered mesoporous polymers and carbons based on the supramolecular aggregates as templates. Block copolymers are mainly introduced here. Both the aggregates of block copolymers themselves and the assembling with thermosetting resins have the abilities to organize ordered mesostructures. The understanding of the synthesis is on the formation of supramolecular arrangement of molecules, templating, cross-linking, removal of templates, and carbonization. In addition, the morphological control and the functionalization of ordered mesoporous carbonaceous materials are briefly summarized. Highly ordered mesoporous carbons have been synthesized from a direct triblock-copolymer-templating process of organic–organic self-assembly. The organic precursors are phenol, formaldehyde, and a functional monomer of p-fluorophenol, and the organic structure directing agent is triblock copolymer Pluronic F127. Characterization using X-ray diffraction (XRD), transmission electron microscopy (TEM), Raman, Fourier transform infrared (FTIR), and nitrogen adsorption techniques reveals that the fluorinated carbons possess highly ordered mesostructures, high surface areas (693–998 m 2/g), large pore sizes (3.0–4.4 nm), large pore volumes (0.43–0.70 cm 3/g), and C–F covalent bonds after high-temperature carbonization at 900 °C. Various mesostructures such as 2D hexagonal and 3D body-centered cubic structures can be synthesized by simply tuning the ratios of p-fluorophenol/phenol or phenol/triblock copolymer. The fluorinated mesoporous carbon modified glassy carbon (GC) electrode exhibits higher electron transfer rate than both pure mesoporous carbon FDU-15 modified and bare GC electrodes. The thalamic reticular nucleus is a layer of GABAergic neurons that occupy a strategic position between the thalamus and cortex. Here we used laser scanning photostimulation to compare in young mice (9–12 days old) the organization of the reticular inputs to first- and higher-order somatosensory relays, namely, the ventral posterior lateral nucleus and posterior nucleus, respectively. The reticulothalamic input footprints to the ventral posterior lateral nucleus neurons consisted of small, single, topographically organized elliptical regions in a tier away from the reticulothalamic border. In contrast, those to the posterior nucleus were complicated and varied considerably among neurons: although almost all contained a single elliptical region near the reticulothalamic border, in most cases, they consisted of additional discontinuous regions or relatively diffuse regions throughout the thickness of the thalamic reticular nucleus. Our results suggest two sources of reticular inputs to the posterior nucleus neurons: one that is relatively topographic from regions near the reticulothalamic border and one that is relatively diffuse and convergent from most or all of the thickness of the thalamic reticular nucleus. We propose that the more topographic reticular input is the basis of local inhibition seen in posterior nucleus neurons and that the more diffuse and convergent input may represent circuitry through which the ventral posterior lateral and posterior nuclei interact. A high-temperature reductive sulfuration method is demonstrated to synthesize highly ordered mesoporous metal sulfide crystallites by using mesoporous silica as hard templates. H2S gas is utilized as a sulfuration agent to in situ convert phosphotungstic acid H3PW12O40·6H2O to hexagonal WS2 crystallites in the silica nanochannels at 600 °C. Upon etching silica, mesoporous, layered WS2 nanocrystal arrays are produced with a yield as high as 96 wt %. XRD, nitrogen sorption, SEM, and TEM results reveal that the WS2 products replicated from the mesoporous silica SBA-15 hard template possess highly ordered hexagonal mesostructure (space group, p6mm) and rodlike morphology, analogous to the mother template. The S-W-S trilayers of the WS2 nanocrystals are partially oriented, parallel to the mesochannels of the SBA-15 template. This orientation is related with the reduction of the high-energy layer edges in layered metal dichalcogenides and the confinement in anisotropic nanochannels. The mesostructure can be 3-D cubic bicontinuous if KIT-6 (Ia3d) is used as a hard template. Mesoporous WS2 replicas have large surface areas (105-120 m²/g), pore volumes (~0.20 cm³/g), and narrow pore size distributions (~4.8 nm). By one-step nanocasting with the H3PMo12O40·6H2O (PMA) precursor into the mesochannels of SBA-15 or KIT-6 hard template, highly ordered mesoporous MoS2 layered crystallites with the 2-D hexagonal (p6mm) and 3-D bicontinuous cubic (Ia3d) structures can also be prepared via this high-temperature reductive sulfuration route. When the loading amount of PMA precursor is low, multiwalled MoS2 nanotubes with 5-7 nm in diameter can be obtained. The synthesis of mesoporous silicate from the surfactant templating approach is demonstrated with the aim of controllable synthesis on the micro-, meso-, and macroscale. An atmosphere-assisted heating process was utilized to transform mesostructured SiC−C nanocomposites in situ into ordered mesoporous SiOC and SiCN ceramics with relatively small structural shrinkage. The mesostructured SiC−C nanocomposites were fabricated using commercially available polycarbosilane (PCS) as a ceramic precursor and mesoporous carbon CMK-3 as a hard template that itself was prepared by a nanocasting procedure from mesoporous silica SBA-15. Reactive gases including air and ammonia were employed to simultaneously incorporate O or N into SiC ceramics and oxidize or reduce the carbon template and excess carbon deposits. The procedure was carried out at 500 °C for 10 h and at 1000 °C for 10 h for air- and ammonia-assisted in situ transformations, respectively. SAXS, XRD, N2sorption, and TEM analyses revealed that the mesoporous SiOC and SiCN ceramics exhibit open, continuous frameworks similar to that of the primary template ordered mesoporous SBA-15. The ordered mesoporous SiOC and SiCN ceramics have high surface areas (200−400 m2g-1), large pore volumes (0.4−0.8 cm3g-1), and narrow pore size distributions (4.9−10.3 nm). The structural shrinkage from mesostructured SiC−C composites to mesoporous SiC-based ceramics decreased with increasing initial pyrolysis temperature for SiC−C nanocomposites owing to the improvement of structural rigidity. This shrinkage was found to be as low as 2.6% from mesostructured SiC−C pyrolysis at 1400 °C to mesoporous SiCN-1400 via ammonia-assisted in situ transformation. [ABSTRACT FROM AUTHOR]

The continual needs for improved performances in applications derived by diversified compositions and mesostructures have pushed forward the development of mesoporous solids. The nonionic-surfactant-templating approach has been a critical route in this advancement. A large number of nonionic surfactants widely used in industries and featured with low cost, low toxicity, bio-degradation and ordered microdomains can be utilized as effective templates to the design and synthesis of abundant mesoporous solids. This feature article provides recent reports on the use of nonionic surfactant self-assembly as examples to fabricate high-quality ordered mesoporous solids which illustrates advances in synthesis and understanding of formation mechanisms. It includes the selection of surfactants, a summary of the effects of synthetic parameters, the current understanding of the synthetic pathways and related mechanisms with some emphasis on evaporation induced self-assembly (EISA), as well as the design and synthesis on the microscale (atomic and molecular compositions) and mesoscale (mesostructures). The thalamic reticular nucleus is strategically located in the axonal pathways between thalamus and cortex, and reticular cells exert strong, topographic inhibition on thalamic relay cells. Although evidence exists that reticular neurons are interconnected through conventional and electrical synapses, the spatial extent and relative strength of these synapses are unclear. To address these issues, we used uncaging of glutamate by laser-scanning photostimulation to provide precisely localized and consistent activation of reticular cell bodies and dendrites in an in vitro slice preparation from the rat as a means to study reticulo-reticular connections. Among the 47 recorded reticular neurons, 29 (62%) received GABAergic axodendritic input from an area immediately surrounding each of the recorded cell bodies, and 8 (17%) responded with depolarizing spikelets, suggesting inputs through electrical synapses. We also found that TTX completely blocked all evoked IPSCs, implying that any dendrodendritic synapses between reticular cells either are relatively weak, have no nearby glutamatergic receptors, or are dependent on back-propagation of action potentials. Finally, we showed that the GABAergic connections between reticular cells are weaker than those from reticular cells to relay cells. Our results suggest that the GABAergic axodendritic synapse is the dominant form of reticulo-reticular connectivity, and because they are much weaker than the reticulo-relay cell synapses, their functional purpose may be to regulate the spatial extent of the reticular inhibition on relay cells. We used laser scanning photostimulation through a focused UV laser of caged glutamate in an in vitro slice preparation through the rat's somatosensory thalamus to study topography and connectivity between the thalamic reticular nucleus and ventral posterior lateral nucleus. This enabled us to focally stimulate the soma or dendrites of reticular neurons. We were thus able to confirm and extend previous observations based mainly on neuroanatomical pathway tracing techniques: the projections from the thalamic reticular nucleus to the ventral posterior lateral nucleus have precise topography. The reticular zone, which we refer to as a "footprint," within which photostimulation evoked inhibitory postsynaptic currents (IPSCs) in relay cells, was relatively small and oval, with the long axis being parallel to the border between the thalamic reticular nucleus and ventral posterior lateral nucleus. These evoked IPSCs were large, and by using appropriate GABA antagonists, we were able to show both GABA(A) and GABA(B) components. This suggests that photostimulation strongly activated reticular neurons. Finally, we were able to activate a disynaptic relay cell-to-reticular-to- relay cell pathway by evoking IPSCs in relay cells from photostimulation of the region surrounding a recorded relay cell. This, too, suggests strong responses of relay cells, responses strong enough to evoke spiking in their postsynaptic reticular targets. The regions of photostimulation for these disynaptic responses were much larger than the above-mentioned reticular footprints, and this suggests that reticulothalamic axon arbors are less widespread than thalamoreticular arbors, that there is more convergence in thalamoreticular connections than in reticulothalamic connections, or both. Thrombocytopenia caused by long- term radiotherapy and chemotherapy exists in cancer treatment. Previous research demonstrates that 5- Hydroxtrayptamine (5- HT) and its receptors induce the formation of megakaryocytes (MKs) and platelets. However, the relationships between 5- HT1A receptor (5- HTR1A) and MKs is unclear so far. We screened and investigated the mechanism of vilazodone as a 5- HTR1A partial agonist in promoting MK differentiation and evaluated its therapeutic effect in thrombocytopenia. We employed a drug screening model based on machine learning (ML) to screen the megakaryocytopoiesis activity of Vilazodone (VLZ). The effects of VLZ on megakaryocytopoiesis were verified in HEL and Meg- 01 cells. Tg (itga2b: eGFP) zebrafish was performed to analyze the alterations in thrombopoiesis. Moreover, we established a thrombocytopenia mice model to investigate how VLZ administration accelerates platelet recovery and function. We carried out network pharmacology, Western blot, and immunofluorescence to demonstrate the potential targets and pathway of VLZ. VLZ has been predicted to have a potential biological action. Meanwhile, VLZ administration promotes MK differentiation and thrombopoiesis in cells and zebrafish models. Progressive experiments showed that VLZ has a potential therapeutic effect on radiation- induced thrombocytopenia in vivo. The network pharmacology and associated mechanism study indicated that SRC and MAPK signaling are both involved in the processes of megakaryopoiesis facilitated by VLZ. Furthermore, the expression of 5- HTR1A during megakaryocyte differentiation is closely related to the activation of SRC and MAPK. Our findings demonstrated that the expression of 5- HTR1A on MK, VLZ could bind to the 5- HTR1A receptor and further regulate the SRC/MAPK signaling pathway to facilitate megakaryocyte differentiation and platelet production, which provides new insights into the alternative therapeutic options for thrombocytopenia. This study presents a rather valuable finding that vilazodone can restore the normal platelet level through regulating 5- HT1A receptor. The evidence supporting the claims of the authors is solid, although inclusion of more cell lines and more detailed analysis of the results would have strengthened the study. Premise of the study: Microsatellite markers were developed for the common Mediterranean shrub Smilax aspera to study the population genetics of this species. Methods and Results: Using the compound microsatellite marker technique, a total of 14 pairs of primers were developed for S. aspera , of which 11 were polymorphic, and three were monomorphic. Levels of polymorphism in the 11 markers were checked in 43 individuals collected from two populations in Greece and Italy. The number of alleles per locus ranged from 10 to 26, with an average of 15.55 alleles per locus. All of these primers also could be amplified from a second species, S. hispida . Conclusions: These microsatellite markers can be used for future studies of genetic diversity in S. aspera, as well as in other related taxa, and will help us to improve our understanding of the microevolutionary processes of this species in the Mediterranean region. The catalytic asymmetric [3 + 2] cycloaddition of 2-indolylmethanols to p-hydroxystyrenes was established in the presence of a chiral phosphoramide, and this reaction provided chiral cyclopenta[b]indole scaffolds in generally high yields and with good enantioselectivities (up to 98% yield, 99:1 er). The control experiments demonstrated that the dual hydrogen-bonding activation mode of the chiral catalyst toward the two substrates played an important role in the reaction. In addition, the large-scale reaction indicated that this catalytic asymmetric [3 + 2] cycloaddition could be scaled up for the synthesis of chiral cyclopenta[b]indole derivatives. C/EBP-homologous protein (CHOP) and histone H3 lysine 4 (H3K4) methylation have been verified to be correlated with apoptosis, whereas their biological function in arsenic-induced hepatocyte apoptosis through the mitochondrial pathway is still unclear. This study aimed to explore the specific regulatory mechanism of CHOP and H3K4me1/2 in arsenic-induced mitochondrial apoptosis in hepatocytes. Apoptosis and proliferation results showed arsenic promoted apoptosis and inhibited cell growth in BRL-3A cells. Meanwhile, arsenic treatment significantly upregulated the 78-kDa glucose-regulated protein (GRP78), CHOP, su(var)-3-9,enhancer-of-zeste,trithorax (SET) domain containing 7/9 (SET7/9), H3K4me1/2, BIM and BAX expression, while markedly downregulated lysine-specific histone demethylase 1 (LSD1) and BCL2 expression. After down-regulating CHOP, LSD1, and (su(var)-3-9,enhancer-of-zeste,trithorax) domain-containing protein 7/9 (SET7/9) in BRL-3A cells by siRNA, silencing CHOP and SET7/9 notably attenuated the pro-apoptotic and anti-proliferative effects of arsenic treatment on BRL-3A cells, which was reversed after inhibiting LSD1. In addition, our results suggested that knockdown of CHOP altered the expression of mitochondrial-associated proteins BCL2 and BIM, whereas knockdown of LSD1 and SET7/8 regulated the level of H3K4me1/2 modification and BAX protein. Coupled with chromatin immunoprecipitation results, we found that the level of CHOP in the promoter regions of BCL2 and BIM was significantly increased in BRL-3A cells exposed to 30 µmol/L NaAsO2 for 24 h, whereas the levels of H3K4me1/2 in the promoter regions of BAX were unchanged. Abstract The odour-induced population response in the in vivo turtle (Terepene sp.) olfactory bulb consists of three oscillatory components (rostral, middle and caudal) that ride on top of a DC signal. In an initial step to determine the functional role of these four signals, we compared the signals elicited by different odorants. Most experiments compared isoamyl acetate and cineole, odorants which have very different maps of input to olfactory bulb glomeruli in the turtle and a different perceptual quality for humans. We found substantial differences in the response to the two odours in the rise-time of the DC signal and in the latency of the middle oscillation. The rate of rise for cineole was twice as fast as that for isoamyl acetate. Similarly, the latency for the middle oscillation was about twice as long for isoamyl acetate as it was for cineole. On the other hand, a number of characteristics of the signals were not substantially different for the two odorants. These included the latency of the rostral and caudal oscillation, the frequency and envelope of all three oscillations and their locations and spatial extents. A smaller number of experiments were carried out with hexanone and hexanal; the oscillations elicited by these odorants did not appear to be different from those elicited by isoamyl acetate and cineole. Qualitative differences between the oscillations in the turtle and those in two invertebrate phyla suggest that different odour processing strategies may be used. [ABSTRACT FROM AUTHOR]

An emulsification method was developed for fabricating core-shell microspheres with a thick shell layer. Kartogenin (KGN) and platelet-derived growth factor BB (PDGF-BB) were respectively loaded into the core portion and the shell layer of the microspheres with high loading efficiency. The optimally built microspheres were combined with chitosan (CH) and silk fibroin (SF) to construct a new type of composite hydrogel with enhanced strength and elasticity, using genipin or/and tyrosinase as crosslinkers for the intended use in cartilage tissue engineering. The composite hydrogels were found to be thermo-responsive at physiological temperature and pH with well-defined injectability. Rheological measurements revealed that they had an elastic modulus higher than 6 kPa with a high ratio of elastic modulus to viscous modulus, indicative of their mechanically strong features. Compressive measurements demonstrated that they possessed well-defined elasticity. In addition, some gels had the ability to administer the temporal separation release of PDGF-BB and KGN in an approximately linear manner for several weeks. The released PDGF-BB was found to be bioactive based on its effects on Balb/c 3T3 cells. The composite gels supported the growth of seeded chondrocytes while preserving their phenotype. Background Obesity increases the risk for insulin resistance and metabolic syndrome in both adults and children. SAA is a member of apolipoprotein and plays an important role in maintaining glucose and lipid homeostasis. The purpose of this study was to assess SAA1 allelic variants with obesity in young school-age children. Methods A total of 520 consecutive children ages 5-15 years were recruited. Children were divided based on BMI z score into Obese (OB; BMI z score ⩾1.65; n = 253) and non-obese (NOB; n = 267). Four SNPs of the human SAA1 gene (rs12218, rs4638289, rs7131332 and rs11603089) were genotyped by use of polymerase chain reaction - restriction fragment length polymorphism (PCR-RFLP) method. Results Compared to NOB, circulating SAA levels were increased in OB, as were LDL-C, TG and TC concentration. Obese children showed increased frequency of rs12218 and rs4638289 polymorphism compared to control children. There were no differences between OB and NOB for the other 2 polymorphisms. Only the rs4638289 polymorphism showed significant contributions to higher SAA plasma levels. CDA-2 (cell differentiation agent 2), a urinary preparation, has potent anti- proliferative and pro-apoptotic properties in cancer cells. However, the mechanisms of tumor inhibitory action of CDA-2 are far from clear, and especially there was no report on lung cancer. Here we demonstrate that CDA-2 and its main component phenylacetylglutamine (PG) reduce the metastatic lung tumor growth, and increases survival time after inoculation with Lewis lung carcinoma (LLC) cells in a dosedependent manner in C57BL6 mice. Proliferative program analysis in cancer cells revealed a fundamental impact of CDA-2 and PG on proliferation and apoptosis, including Bcl-2, Bcl-XL, cIAP1, Survivin, PCNA, Ki-67 proteins and TUNEL assays. CDA- 2 and PG significantly reduced NF-κB DNA-binding activity in lung cancer cells and in alveolar macrophages of tumor bearing mice and especially decreased the release of inflammatory factors including TNFa, IL-6, and KC. Furthermore, CDA-2 and PG decrease the expressions of TLR2, TLR6, and CD14, but not TLR1, TLR3, TLR4, and TLR9 in bone-marrow-derived macrophages (BMDM) of mice stimulated by LLC-conditioned medium (LLC-CM). Over-expressing TLR2 in BMDM prevented CDA-2 and PG from inhibiting NF-κB activation, as well as induction of TNFa and IL-6. TLR2:TLR6 complexes mediate the effect of NF-κB inactivation by CDA-2. In conclusion, CDA-2 potently inhibits lung tumor development by reduction of the inflammation in lung through suppression of NF-κB activation in myeloid cells, associating with modulation of TLR2 signaling. Conjugates of DNA and gold nanop articles (AuNPs) typically exploit the strong Au-S chemistry to self-assemble thiolated oligonucleotides at AuNPs. How- ever, it remains challenging to precisely control the orientation and conformation of surface-tethered oligonu- cleotides and finely tune the hybridization ability. We herein report a novel strategy for spatially controlled functionalization of AuNPs with designed diblock oligonucleotides that are free of modifications. We have demonstrated that poly adenine (polyA) can serve as an effective anchoring block for preferential binding with the AuNP surface, and the appended recognition block adopts an upright conformation that favors DNA hybridization. The lateral spacing and surface density of DNA on AuNPs can also be systematically modulated by adjusting the length of the polyA block. Significantly, this diblock oligonucleotide strategy results in DNA-AuNPs nano- conjugates with high and tunable hybridization ability, which form the basis of a rapid plasmonic DNA sensor. The sensitivity of aptamer-based electrochemical sensors is often limited by restricted target accessibility and surface-induced perturbation of the aptamer structure, which arise from imperfect packing of probes on the heterogeneous and locally crowded surface. In this study, we have developed an ultrasensitive and highly selective electrochemical aptamer-based cocaine sensor (EACS), based on a DNA nanotechnology-based sensing platform. We have found that the electrode surface decorated with an aptamer probe-pendant tetrahedral DNA nanostructure greatly facilitates cocaine-induced fusion of the split anticocaine aptamer. This novel design leads to a sensitive cocaine sensor with a remarkably low detection limit of 33 nM. It is also important that the tetrahedra-decorated surface is protein-resistant, which not only suits the enzyme-based signal amplification scheme employed in this work, but ensures high selectivity of this sensor when deployed in sera or other adulterated samples. Simulation of noncontinuum gas flows presents tremendous challenges, especially for nanoscale devices that usually exhibit low speeds and isothermal conditions. Such simulations are often achieved through use of the Boltzmann Bhatnagar-Gross-Krook equation, which forms the foundation for the lattice Boltzmann (LB) method. Accuracy of the LB method in noncontinuum flows is widely assumed to depend on the order of quadrature used. Here, we study noncontinuum Couette flow and discover that interaction of the lattice with the solid boundaries is the dominant mechanism controlling accuracy—quadrature order plays a comparatively minor role. This suggests the applicability of low-order quadrature in LB simulation of wall bounded isothermal noncontinuum flows, and leads to a framework and rationale for accurate implementation of LB models in noncontinuum flows. The adsorption and desorption behavior of tetracycline on an Alfisol and an Ultisol was investigated at varying soil/solution ratios (1:10, 1:50, and 1:100) and different cation types (CaCl2, KCl, and NaCl) and H2O using a batch equilibration experiment. Kinetic studies revealed that the adsorption of tetracycline on the two soils was rate limited, and the adsorption kinetics could be well described by the Elovich equation and exponent equation. Changes in the soil/solution ratio affected the relative equilibration time in the Ultisol, but there was no effect of the initial tetracycline concentration. The adsorption rate in the soils decreased as the soil/solution ratio increased from 1:50 to 1:10. The adsorption and desorption data on tetracycline in the two soils could be described by the Freundlich isotherm equation, with r values >0.952 under all experimental treatments. The Ultisol always had stronger adsorption (higher distribution coefficient [Kd] value) than the Alfisol under all experimental conditions. The Kd values decreased with increasing soil/solution ratios in the two tested soils at 1 or 1.25 mg L-1 of tetracycline; however, this trend did not occur at 25 mg L-1 of tetracycline. The adsorption isotherm had an S-type curve that changed into an L-type curve with decreasing soil/solution ratios from 1:10 to 1:100. The presence of cations in the soil solution, especially CaCl2, can significantly reduce the adsorption of tetracycline in soils. Soy isoflavone preparations, such as purified genistein and a soy extract (Novasoy), were reported previously to exert beneficial effects on bones. Our purpose in this study was to compare the effects of genistein and Novasoy on 3-dimensional trabecular bone parameters and the expression of bone-specific genes in ovariectomized (OVX) mice. The sham-operated mice were fed the control diet and OVX mice were fed diets containing genistein or Novasoy or the control diet, with or without 17β-estradiol treatment, for 5 wk. Trabecular bone parameters of tibias were measured by microcomputed tomography and gene expression was assayed by real-time PCR. Consumption of diets containing genistein or Novasoy partially prevented the ovariectomy-induced increase in body weight but did not alter the uterus weight of the OVX mice. Novasoy, but not purified genistein, significantly preserved trabecular bone mass, bone volume, and trabecular bone separation in the proximal tibial metaphysis. Purified genistein decreased mRNA expression of receptor activator of nuclear factor-κB ligand (RANKL), carbonic anhydrase II, and cathepsin K and enhanced the ratio of osteoprotegrin:RANKL mRNA expression in the tibial head of the OVX mice. In contrast, the diet containing Novasoy suppressed the OVX-induced increase in serum alkaline phosphatase but did not alter bone-specific gene expression of tibia. Our study demonstrated that a soy extract containing a similar level of genistein in the form of Novasoy is more effective than purified genistein in improving tibial trabecular bone quality in OVX mice, but the mechanism of action might be distinct from that of genistein.
