This study investigated effects of encapsulated blends of essential oils (EO) and organic acids (OA) on performance, intestinal morphology, cecal microflora, and jejunal mucosal enzyme activity and secretory IgA (s-IgA) level of broilers. Three hundred one-day-old male broilers were divided into 5 treatments with 5 replicates each. Control group received basal diet. Three additional groups received basal diets supplemented with 150, 200, or 250 mg/kg encapsulated blends of EO and OA. Antibiotic group received basal diet supplemented with 30 mg/kg bacitracin zinc. On days 21 and 70 of trial, 2 broilers from each replicate were weighed, and then blood, duodenum, jejunum, ileum, and cecum samples were collected. Results showed that broilers fed blends of EO and OA had greater average daily gain (ADG) (linear, P = 0.043) and gain-to-feed ratio (G : F) (linear, P = 0.052) at day 21 compared to broilers fed the control diet. Blends of EO and OA reduced cecal Escherichia coli and Salmonella level of 21- and 70-day-old broilers (linear, P < 0.01). Jejunum villus height-to-crypt depth ratio (VCR) of 21-day-old broilers was elevated by blends of EO and OA (linear, P = 0.041) compared to control. Blends of EO and OA promoted chymotrypsin activity at day 21 (quadratic, P = 0.014), jejunal mucosal s-IgA content at day 70 (linear, P = 0.012), α-amylase activity at day 70 (quadratic, P = 0.043), and chymotrypsin activity at day 70 (quadratic, P = 0.037). Compared to control, antibiotic group increased ADG at day 21 (P = 0.039), decreased cecal Salmonella level at day 21 (P = 0.018), enhanced jejunum VCR at day 21 (P = 0.049), and elevated jejunal mucosal s-IgA content at day 21 (P = 0.016). It can be stated that EO and OA blends enhanced performance, increased jejunal s-IgA level and enzyme activity, improved intestinal morphology, and balanced intestinal microflora of broilers.

H9N2 avian influenza virus (AIV) infection is a major problem in poultry industry worldwide. In this study, molecular characterizations and phylogenetic relationships of hemagglutinin (HA) gene sequences of H9N2 AIV of 5 Chinese isolates in 2014 recently available in GenBank, 3 widely used vaccine strains, and 52 novel isolates in China from 2013 to 2015 were analyzed. The homology analysis showed that the nucleotide sequences of HA gene of these recent Chinese H9N2 AIV isolates shared homologies from 94.1 to 99.9%. Phylogenetic analysis showed that all isolates belonged to AIV lineage h9.4.2.5. Fifty-six out of the 57 recent Chinese H9N2 AIV isolates had the motifs PSRSSR↓GLF at the cleavage sites within the HA protein, while one isolate PWH01 harbored LSRSSR↓GLF. Remarkably, all of the recent Chinese H9N2 AIV strains had the Q216L substitution in the receptor binding site, which indicated that they had potential to infect humans. Most of recent Chinese H9N2 AIV isolates lost the potential N-linked glycosylation site at residues 200–202 compared with vaccine strains. This present study demonstrated that AIV lineage h9.4.2.5 was more predominant in China than other lineages as it harbored all the H9N2 AIV isolated between 2013 and 2015. Also we showed the importance of continuous surveillance of emerging H9N2 AIV in China and update of vaccine formulation accordingly in order to prevent and control H9N2 AIV.

Influenza A viruses (IAV) still pose a threat to animals and humans. Currently, M2 protein ion channel inhibitors and neuraminidase inhibitors are the two main drugs for treating IAV infections by interrupting virus assembly or release respectively, but the emergence of viral resistance was a concern for their long term uses. In this study, the inhibition effect of alpha-lipoic acid (α-LA) on IAV propagation has been evaluated in vitro. The results showed that α-LA inhibited IAV replication in MDCK cells at 2mM, and also reduced nucleus translocation of nuclear factor κB (NF-κB) p65 at the concentration above 1mM. Additionally, it was found that caspase-3 activity was remarkably inhibited and type I interferons (IFNs) were up-regulated following α-LA treatment. This study indicated that α-LA might be a potential anti-influenza virus agent worthy of further investigations.

As part of our ongoing influenza surveillance program in South China, 19 field strains of H9N2 subtype avian influenza viruses (AIVs) were isolated from dead or diseased chicken flocks in Guangdong province, South China, between 2012 and 2013. Hemagglutinin (HA) genes of these strains were sequenced and analyzed and phylogenic analysis showed that 12 of the 19 isolates belonged to the lineage h9.4.2.5, while the other seven belonged to h9.4.2.6. Specifically, we found that all of the viruses isolated in 2013 belonged to lineage h9.4.2.5. The lineage h9.4.2.5 viruses contained a PSRSSR ↓ GLF motif at HA cleavage site, while the lineage h9.4.2.6 viruses contained a PARSSR ↓ GLF at the same position. Most of the isolates in lineage h9.4.2.5 lost one potential glycosylation site at residues 200-202, and had an additional one at residues 295-297 in HA1. Notably, 19 isolates had an amino acid exchange (Q226L) in the receptor binding site, which indicated that the viruses had potential affinity of binding to human like receptor. The present study shows the importance of continuing surveillance of new 119N2 strains to better prepare for the next epidemic or pandemic outbreak of H9N2 AIV infections in chicken flocks. In order to develop a desirable inexpensive, effective and safe vaccine against the very virulent infectious bursal disease virus (vvIBDV), we tried to take advantage of the emerging T4 bacteriophage surface protein display system. The major immunogen protein VP2 from the vvIBDV strain HK46 was fused to the nonessential T4 phage surface capsid protein, a small outer capsid (SOC) protein, resulting in the 49 kDa SOC-VP2 fusion protein, which was verified by sodium dodecylsulfate polyacrylamide gel electrophoresis and Western blot. Immunoelectromicroscopy showed that the recombinant VP2 protein was successfully displayed on the surface of the T4 phage. The recombinant VP2 protein is antigenic and showed reactivities to various monoclonal antibodies (mAbs) against IBDV, whereas the wild-type phage T4 could not react to any mAb. In addition, the recombinant VP2 protein is immunogenic and elicited specific antibodies in immunized specific pathogen free (SPF) chickens. More significantly, immunization of SPF chickens with the recombinant T4-VP2 phage protected them from infection by the vvIBDV strain HK46. When challenged with the vvIBDV strain HK46 at a dose of 100 of 50% lethal dose (LD50) per chicken 4 weeks after the booster was given, the group vaccinated with the T4-VP2 recombinant phage showed no clinical signs of disease or death, whereas the unvaccinated group and the group vaccinated with the wild-type T4 phage exhibited 100% clinical signs of disease and bursal damages, and 30%-40% mortality. This study investigated the effects of xanthophylls (containing 40% lutein and 60% zeaxanthin; Juyuan Biochemical Co., Ltd., GuangZhou, China) on gene expression associated with carotenoid cleavage enzymes (β-carotene 15, 15'-monooxygenase, BCMO1; and β-carotene 9', 10'-dioxygenase, BCDO2) and retinoid metabolism (lecithin:retinol acyl transferase (LRAT) and STRA6) of breeding hens and chicks. In experiment 1, 432 hens were divided into 3 groups and fed diets supplemented with zero (as the control group), 20, or 40 mg/kg xanthophyll. The liver, duodenum, jejunum, and ileum were sampled at d 35 of the trial. Results showed that 40 mg/kg xanthophyll supplementation increased BCDO2 mRNA in the liver, duodenum, and jejunum; LRAT mRNA in the jejunum; and STRA6 mRNA in the liver, while it decreased LRAT mRNA in the liver. Experiment 2 was a 2 x 2 factorial design. Male chicks hatched from a zero or 40 mg/kg xanthophyll diet of hens were fed a diet containing either zero or 40 mg/kg xanthophylls. The liver, duodenum, jejunum, and ileum were sampled at zero, 7, 14, and 21 d after hatching. Results showed that in ovo xanthophyll modulated carotenoid and retinoid metabolism mainly within one wk after hatching. The maternal effects gradually vanished and dietary effects began to work one to 2 wk after hatching. Dietary xanthophyll regulated carotenoid and retinoid metabolism mainly from 2 wk onward. The xanthophyll regulation of carotenoid and retinoid metabolism also revealed strong tissue specificity. In conclusion, xanthophyll supplementation could modulate carotenoid and retinoid metabolism in different tissues of hens and chicks. [ABSTRACT FROM AUTHOR]

Cell-mediated cytotoxic responses are critical for control of Marek's disease virus (MDV) infection and tumour development. However, the mechanisms of virus clearance mediated by cytotoxic responses in the bursa of Fabricius of chickens during MDV infection are not fully understood. In this study, the host cytotoxic responses during MDV infection in the bursa were investigated by examining the expression of genes in the cell lysis pathways. Partial up-regulation existed in the expression of the important cytolytic molecule granzyme A (GzmA), Fas, NK lysin and DNA repair enzyme Ape1, whereas little or no expression appeared in other cytolytic molecules, including perforin (PFN) and Fas ligand (FasL), and molecules involved in DNA repair and apoptosis in the bursa during MDV infection. These results suggest that less sustained cytotoxic activities are generated in the bursa of MDV-infected chickens.
