Objective: Tan lambs (n = 36, 3 mo old, 19.1±0.53 kg) were used to assess effects of dietary guanidinoacetic acid (GAA) and rumen-protected methionine (RPM) on growth performance, carcass traits, meat quality, and serum parameters.Methods: Lambs were randomly assigned to three treatment groups, with 6 pens per group and 2 lambs per pen. Dietary treatments were: basal diet alone (I); basal diet supplemented with 0.08% GAA+0.06% RPM (II); and basal diet supplemented with 0.08% GAA+0.08% RPM (III). Diets were provided three times a day for 90 d. Intake per pen was recorded daily and individual lamb body weight (BW) was measured monthly. The B-cell lymphoma 2 (BCL-2) protein family plays a pivotal role in regulating the apoptosis process. BCL-2, as an antiapoptotic protein in this family, mediates apoptosis resistance and is an ideal target for cell death strategies in cancer therapy. Traditional treatment modalities target BCL-2 by occupying the hydrophobic pocket formed by BCL-2 homology (BH) domains 1-3, while in recent years, the BH4 domain of BCL-2 has also been considered an attractive novel target. Herein, we describe the discovery and identification of DC-B01, a novel BCL-2 inhibitor targeting the BH4 domain, through virtual screening combined with biophysical and biochemical methods. Our results from surface plasmon resonance and cellular thermal shift assay confirmed that the BH4 domain is responsible for the interaction between BCL-2 and DC-B01. As evidenced by further cell-based experiments, DC-B01 induced cell killing in a BCL-2-dependent manner and triggered apoptosis via the mitochondria-mediated pathway. DC-B01 disrupted the BCL-2/c-Myc interaction and consequently suppressed the transcriptional activity of c-Myc. Moreover, DC-B01 inhibited tumor growth in vivo in a BCL‑2‑dependent manner. Collectively, these results indicate that DC-B01 is a promising BCL-2 BH4 domain inhibitor with the potential for further development.

Background The lack of effective prognostic models for colorectal cancer (CRC) makes it crucial to establish a reliable prognostic model for colorectal cancer. The aim of this study was to develop a reliable prognostic model in CRC. Using CRC samples from the TCGA database, a multi-gene combination model was screened for prognosis in CRC patients and the prognostic power of the multi-gene combination model was validated. We screened a multi-gene combination model for risk stratification and prognosis of CRC patients, which is important for individualised clinical treatment and long-term management of colorectal cancer patients. Results To solve these questions, we used data from colorectal cancer patients in the TCGA database to uncover differential genes, screened variables using one-way COX analysis and LASSO regression analysis, and then screened for the polygenic combination most associated with colorectal cancer and assessed its significance on survival events using multi-factor COX analysis and ROC curves to assess polygenic combinations. Finally, using gene enrichment analysis, the polygenic combinations were found to be associated with tRNA biosynthesis, homologous recombination, nucleotide excision repair, ribosome biogenesis in eukaryotes and DNA replication. Conclusions The multi-gene combination (FCGBP, GSTM1, IGLV4-69, IGSF9 and PCOLCE2) model screened in this study is capable of prognosticating colorectal cancer, and the findings of this study could shed new light on the prognosis of colorectal cancer in the clinical setting.

The promising photocatalytic conversion of CO2 into valuable fuel promotes the development of photocatalyst through various methods. In this work, TiO2 nanoparticle was composited with covalent porphyrin polymers (COP-Ps) to fabricate composite photocatalysts. The resultant COP-Ps/TiO2 composites by in situ hydrothermal method exhibit much improved photocatalytic activity for the conversion of CO2 into CO relative to two components, and it is attributable to improved charge transfer between two moieties led by strong interaction. Especially, TiO2 is composited more evenly with the sulfonated hollow COP-P (sh-COP-P). The resultant composite sh-COP-P/TiO2 performs best with a CO production rate of 5.70 μmol·g−1·h−1, which is approximately 20.4 times as high as that of pure TiO2 and 2.3 times of sh-COP-P polymer. For comparison, the simple physical mixture of sh-COP-P and TiO2 (sm-sh-COP-P/TiO2) was fabricated, and it performs more badly due to poor mixing uniformity. A Z-scheme photocatalytic mechanism was proposed for sh-COP-P/TiO2 composite on the basis of energy band analysis and hydroxyl radical test. This study provides a new in situ strategy to fabricate organic polymer/metal oxide composites of high photocatalytic activity for CO2 reduction.

Highly selective separation and purification of acetylene (C2H2) from ethylene (C2H4) and carbon dioxide (CO2) are daunting challenges in light of their similar molecule sizes and physical properties. Herein, we report a two-dimensional (2D) stable metal–organic framework (MOF), NUM-11 ([Cu(Hmpba)2]·1.5DMF) (H2mpba = 4-(3,5-dimethyl-1H-pyrazol-4-yl)benzoic acid), with sql topology, stacked together through π–π interactions for efficient separation of C2H2 from C2H4 and CO2. The 2D-MOF material offers high hydrolytic stability and good purification capacity; especially, it could survive in water for 7 months, even longer. This stable MOF selectively captures C2H2 from mixtures containing C2H4 and CO2, as determined by adsorption isotherms. The ideal adsorbed solution theory selectivity calculations and transient breakthrough experiments were performed to verify the separation capacity. The low isosteric heat of NUM-11a (desolvated NUM-11) (18.24 kJ mol–1 for C2H2) validates the feasibility of adsorbent regeneration with low energy footprint consumption. Furthermore, Grand Canonical Monte Carlo simulations confirmed that the pore surface of the NUM-11 framework enabled preferential binding of C2H2 over C2H4 and CO2 via multiple C–H···O, C–H···π, and C–H···C interactions. This work provides some insights to prepare stable MOF materials toward the purification of C2H2, and the water-stable structure, low isosteric heat, and good cycling stability of NUM-11 make it very promising for practical industrial application.

The separation of ethylene (C2H4) from C2 hydrocarbons is considered as one of the most difficult and important processes in the petrochemical industry. Heat-driven cryogenic distillation is still widely used in the C2 hydrocarbons separation realms, which is an energy intensive process and takes up immense space. In response to a greener, more energy-efficient sustainable development, we successfully synthesized a multifunction microporous Mg-based MOF [Mg2(TCPE)(µ2-OH2)(DMA)2].solvents (NUM-9) with C2H6/C2H2 selectivity based on a physical adsorption mechanism, and with outstanding stability; especially, it is stable up to 500 °C under an air atmosphere. NUM-9a (activated NUM-9) shows good performances in the separation of C2H6/C2H2 from raw ethylene gases. In addition, its actual separation potential is also examined by LAST and dynamic column breakthrough experiments. GCMC calculation results indicate that the unique structure of NUM-9a is primarily conducive to the selective adsorption of C2H6 and C2H2. More importantly, compared with C2H4, NUM-9a prefers to selectively adsorb C2H6 and C2H2 simultaneously, which makes NUM-9a as a sorbent have the capacity to separate C2H4 from C2 hydrocarbon mixtures under mild conditions through a greener and energy-efficient separation strategy.

AIM: To determine whether Houttuynia cordata Thunb (HCT) can increase the survival of the retinal ganglion cells (RGCs) and inhibit microglia activation following retinal ischemia-reperfusion (RIR) injury. METHODS: Rat model of RIR was induced by transient elevation of the intraocular pressure (IOP). HCT was orally administered for 2d before the performance of retinal RIR model and once a day for the next 14d. After 14d of RIR injury, the rats were sacrificed for further analysis. Survival RGCs were stained with haematoxylin and eosin (H&E). Apoptosis of RGCs was detected by TUNEL staining. Retinal function was examined by flash-electroretinography (F-ERG). Retinal microglia were labeled using Iba-1, one specific marker for microglia. The mRNA expression levels of inducible nitric oxide synthase (iNOS), tumor necrosis factor alpha (TNF-α), and interleukin 1 beta (IL-1β) were assessed by quantitative real time reverse transcription polymerase chain reaction (qRT-PCR). RESULTS: Systemic HCT treatment significantly reduced RGCs death by H&E staining and exhibited an anti-apoptotic effect as assessed by TUNEL staining at day 14 after RIR injury. HCT greatly improved the retinal function as examined by F-ERG. The number of activated microglia significantly increased after RIR injury, which was significantly attenuated by HCT treatment. Besides, RIR injury induced a strong upregulation of pro-inflammatory genes TNFα, iNOS and IL-1β mRNAs at day 14 post injury, which was suppressed by HCT. CONCLUSION: Neuroprotective effects of HCT encourage the survival of RGCs through inhibiting microglia activation due to RIR injury. Together these results support the use of HCT as promising therapy for the ischemic events of the retina diseases.

The Paleoproterozoic Khondalite Belt, located in the northwestern segment of North China Craton (NCC), is characterized by widespread high-temperature/ultrahigh-temperature (UHT) granulite/gneiss and large-scale magmatic activity. The tectonic evolution is still controversial. Here, we report new geochronological, elemental, and Hf-O-Sr-Nd isotopic data for a Paleoproterozoic layered complex in the Jining terrane to constrain the tectonic evolution of the Khondalite Belt. In situ zircon U-Pb dating indicates that the Sanchakou gabbros were emplaced between ~1.94 Ga and ~1.82 Ga, which might be the heat source of UHT metamorphism. The elemental and Hf-O-Sr-Nd isotopic analysis shows that the formation of Sanchakou gabbros is consistent with the assimilation and fractional crystallization (AFC) process. The magma originates from the 10%~20% partial melting of the spinel + garnet lherzolite mantle. The Sanchakou gabbros are magmatic crystallization products mixed with crustal wallrocks in the magma chamber. We have established a tectonic evolution model involving asthenosphere upwelling after the amalgamation of the Ordos and Yinshan Blocks at ~1.95 Ga.

Bambusa contracta L. C. Chia & H. L. Fung is widely distributed in the foothills of Guangxi province, China, and used as a raw material for the production of various bamboo weaving products. In the present work, the complete chloroplast genome sequence of B. contracta was characterized by Illumina pair-end sequencing. The chloroplast genome of B. contracta was 139,470 bp in total length, containing a large single-copy (LSC) region of 83,187 bp, a small single-copy (SSC) region of 12,897 bp, and two inverted repeats (IR) regions of 21,693 bp. The genome consisted of a total of 127 genes, with 83 protein-coding genes, 36 tRNA genes, and eight rRNA genes. Based on 14 chloroplast genomes, the phylogenetic analysis revealed that B. contracta is closely related to B. emeiensis in Bambusodae.

As a new kind of photocatalytic system, covalent organic polymers (COPs) exhibit wide spectral absorption and high surface area, but poorer photocatalytic performance relative to traditional inorganic semiconductor materials due mainly to the lower separation efficiency of photogenerated carriers. Herein, a tetrabiphenylporphyrin-based COP (COP-P) was investigated in photocatalyzing the reduction of CO2, and nearly exclusive production of CO was observed with a rate of 1.55 μmol g−1 within one hour, which is a little higher than commercial TiO2 (P25) with a rate of 0.92 μmol g−1. Meanwhile, hollowrization and post-sulfonation were utilized to modulate the photocatalytic performance of COP-P, resulting in just moderately improved and slightly deteriorated performance, respectively. Surprisingly, the synergistic action of these two modulations gives 4.8 times enhanced production of CO (7.54 μmol g−1 within 1 hour). Its revealed that post-sulfonation would improve the separation and transfer ability of photogenerated carriers based on transient amperometric I–t curve analyses, but reduces significantly the surface area of COP-P polymers and thereby decreases CO2 uptake through gas adsorption analyses, which fortunately could be remedied by hollowrization, leading to much improved photocatalytic production of CO. This work reveals the complicated influences of post-modification and paves a facile way to optimize the photocatalytic efficiency of organic polymers through synergetic effects.

Two-dimensional (2D) covalent organic frameworks (COFs) feature open and ordered one-dimensional column nanochannels which offer immense possibilities for incorporation of various guests for specific functions. However, the relatively low chemical stability of most COFs originating from the dynamic covalent linkages hinders their practical application. In this work, a highly crystalline and heteroporous dibenzo[g,p]chrysene-based COF (DBC-2P) was synthesized and served as a host material for ionic conduction. DBC-2P exhibits excellent stability both in strong acid and base due to the large conjugated DBC-based knot that reinforces the interlayer interactions. Subsequent encapsulation of linear polyethylene glycol (PEG) and PEG-LiBF4 salt into the nanochannels of DBC-2P affords a hybrid material with a high ionic conductivity of 2.31×10-3 S cm-1 . This work demonstrates an efficient post-synthetic strategy for the development of new COF-polymer composites with intriguing properties.

The propane (C3H8)-trapping adsorption behavior is considered as a potential performance to directly produce high-purity propylene (C3H6). Herein, we report an ultramicroporous Mn-based metal-organic framework (NUM-7) with a reverse C3H8-selective behavior in the low-pressure area. The pore structure of this material possesses more electronegative aromatic benzene rings for the stronger binding affinity to C3H8, and the material shows outstanding reverse ideal adsorbed solution theory (IAST) selectivity values. Single-component sorption isotherms preliminarily show the reverse adsorption behavior in the low-pressure part, and the moderate heat of adsorption further confirms this performance and exhibits less energy consumption for regeneration. In addition, the purification effect for the C3H8/C3H6 mixture is evaluated by the IAST selectivity and transient breakthrough curves, and the GCMC calculation results reveal that the fascinating C3H8-trapping behavior mainly depends on the multiple C-H···π interactions. Moreover, because C3H6 is the desired target product, the interesting C3H8-selective adsorption behavior of NUM-7 may provide its potential for one-step purification of C3H6, and this work can provide the method of developing C3H8-selective materials for the purification of C3H6.

The promising photocatalytic conversion of CO

Neonatal respiratory distress syndrome (NRDS) is a common disease that occurs in premature infants. However, the mechanisms underlying the disease remain unclear. microRNAs (miRNAs) have been indicated to play a crucial role in the development of NRDS. In this study, we aimed to explore the regulatory mechanisms of miR-296-5p in NRDS. The expression levels of miR-296-5p in preterm infants with NRDS were determined using quantitative reverse-transcription polymerase chain reaction (RT-qPCR). A549 cells were transfected with lentiviral vectors encoding miR-296-5p, and the transfection efficiency was determined using RT-qPCR. Flow cytometry and CCK8 assay were performed to measure apoptosis and proliferation of A549 cells, respectively. The protein levels of pulmonary surfactant SP-A (SFTPA1), SP-B, Wnt7b, and β-catenin were measured using western blotting. We demonstrated an upregulation of miR-296-5p in NRDS. The miR-296-5p was successfully overexpressed in A549 cells via lentivirus transfection, and the upregulation of miR-296-5p inhibited cell proliferation and secretion of SP-A and SP-B and also induced downregulation of the Wnt7b/β-catenin in vitro. Therefore, miR-296-5p inhibits cell proliferation and secretion of pulmonary surfactants in A549 cells via downregulation of Wnt7b/β-catenin signaling.

Mucin1 (MUC1) upregulation in colon cancer has been linked to poor patient outcomes and advanced stage at diagnosis. This is partially due to MUC1-mediated inhibition of T-cell proliferation affecting efficient lysis by cytotoxic lymphocytes, which contributes to escape from immune surveillance. In the present study, human colorectal cancer tissues were collected, and MUC1-positive and MUC1-negative colon cancer mouse models were prepared; subsequently, the number and function of immune cells in tumor tissues were measured using flow cytometry. The present study revealed that MUC1, as a tumor-associated antigen, can recruit more tumor-infiltrating lymphocytes into the tumor microenvironment compared with MUC1-negative colon cancer, but that these cells could not serve antitumor roles. Conversely, the present study demonstrated that MUC1-positive colon cancer attracted more regulatory T cells (Treg cells), myeloid-derived suppressor cells (MDSCs) and tumor-associated macrophages (TAMs) to the tumor site than MUC1-negative colon cancer. Furthermore, the data suggested that programmed death protein 1 (PD1)-programmed death ligand 1 (PDL1) expression is greater in MUC1-positive colon cancer. Blocking the PD1-PDL1 signaling pathway reduced the percentage of Treg cells, MDSCs and TAMs in the tumor microenvironment, enhanced T-cell cytotoxicity and inhibited tumor growth, prolonging the survival time of MUC1-positive tumor-bearing mice. Therefore, the present study elucidated the role of MUC1 in tumor immune escape and provides a foundation for the application of PDL1 inhibitors to MUC1-positive colon cancer.

Aimed at the exploration of luminescent coordination polymer (LCP) systems for highly tunable luminophores, a new series of Cd(II) LCPs, namely, [Cd2(tpt)2(CBA)2(H2O)2]·(PAH) (1), [Cd2(tpt)2(CBA)2]·(PAHs) (2–3) (tpt = 2,4,6-tri(pyridin-4-yl)-1,3,5-triazine, H2CBA = 4-(carboxymethyl) benzoic acid, PAHs = polycyclic aromatic hydrocarbons, coronene for 1, perylene for 2, and triphenylene for 3), are successfully constructed based on the host–guest charge transfer (HGCT) mechanism. Through the PAHs directed coordination assembly of Cd(II) ions, CBA2– and tpt ligands, compounds 1–3 exhibit the as-expected host–guest structures, in which PAHs-tpt donor–acceptor (D–A) systems are successfully achieved. Structural investigations of 1–3 show that the flexibility of the carboxylate ligand and the guest dependent donor–acceptor interaction affect their structures. A new fluorescent coordination polymer (NKU-115) was constructed under solvothermal condition based on Tb3+ ion and H2L (5-(4H-1,2,4-triazol-4-yl) benzene-1,3-dicarboxylic acid) ligand, featuring high solvent and water stability. Luminescent investigations revealed that NKU-115 shows fluorescence quenching response toward Fe3+ in aqueous solution and the mechanism of quenching was analyzed.

Research on selective C8 sorption has tended to focus upon rigid porous materials such as zeolites and MOFs but has revealed generally weak selectivity that is inconsistent across the range of C8 molecules. Nevertheless, there are a few recent examples of non-porous molecular materials that exhibit relatively high selectivity for p-xylene (pX) from pX/oX, approaching that of the current benchmark pX sorbent, the zeolite H/ZSM-5. Herein, we report that a L-shaped Ag(I) complex, AgLClO4 (M), which crystallizes as a non-porous molecular solid material, offering exceptional performance for pX selectivity across the range of C8 isomers with liquid extraction selectivity values of 24.0, 10.4 and 6.2 vs. oX, eB and mX, respectively. Three new Co(II) coordination polymers with formulas of {[Co2(L1)(1,4-NDC)2]·3H2O}n (1), [Co3(L2)2(HCOO)2(1,4-NDC)2]n (2) and [Co2(L2)(μ3-OH)(1,4-NDC)1.5]n (3) (1,4-H2NDC = Naphthalene-1,4-dicarboxylic acid, L1 = di(1H-imidazol-1-yl)methane, L2 = 1,4-di(1H-imidazol-1-yl)benzene) were solvothermal synthesized from 1,4-H2NDC with the aid of three different length-controllable auxiliary ligands and fully characterized. Their structures are determined by single-crystal X-ray diffraction, IR spectra, elemental analysis, powder X-ray diffraction and thermogravimetric analysis. Complexes 1 and 3 display 3D framework structures, corresponding to a 6-connected (412·63) net, a 8-connected (424·5·63) net, respectively. However, it is noteworthy that the complex 1 displays a 2-fold interpenetrating framework structure, complex 3 possesses a self-interpenetrating framework structure. Complex 2 displays 2D 4-connected undulating plane net structure. Moreover, magnetic studies indicate antiferromagnetic interactions between the Co(II) ions in the four complexes.

A newly constructed hexanuclear cadmium metal–organic framework (MOF) exhibits a highly selective and sensitive fluorescence‐quenching response toward Fe 3+ ions, which is attributable to the coaction of absorption competition and an energy‐transfer mechanism.

Aiming to develop an effective fluorescence probe for detecting nitroanilines compounds, a new3D “pillar-layered” metal–organic framework [Cd(dpa)(pta)]·nDMF (1) was constructed through solvothermal reaction of Cd2+ with 9,10-di(pyridine-4-yl)anthracene (dpa) and terephthalic acid (pta). As expected, 1 displays selective fluorescence quenching response toward nitroanilines, including 2-nitroaniline, 4-nitroaniline and 2,4-dinitroaniline, attributable to the cooperation of absorption competition and energy transfer mechanism, which can effectively distinguish nitroanilines from other nitrobenzene and anilines compounds. Furthermore, the quenching efficiency exhibits different varying trends for these three nitroanilines along with the change of exciting wavelength, which can be used to effectively distinguish them from each other.

We developed a simple method based on molecularly imprinted photonic polymers (MIPPs) for sensing of sulfonamides in egg white samples. The fabrication of MIPPs mainly involved a photonic crystal template method integrated with a molecular imprinting technique. Firstly, an opal photonic crystal template was self-assembled from monodisperse polystyrene colloids. And then the template was embedded with molecularly imprinted polymers, which was synthesized with acrylic acid and acrylamide as monomers, N,N′-methylenebisacrylamide as a cross-linker and sulfamerazine (SM1) or sulfamethazine (SM2) as imprinting molecules. After removal of the photonic crystal template and the imprinted molecules, the resultant MIPP sensor accommodated an inverse opal photonic crystal structure embedded in hydrogels. By using the as-prepared MIPP sensor, sulfonamides were sensitively and specifically recognized via a molecular imprinting technique and the recognition was detected through the readable Bragg diffraction shift from the photonic crystal structure. A 70 nm maximum Bragg diffraction blue-shift of the MIPP was observed with the concentrations of the sulfamerazine or sulfamethazine varying from 3.8 μM to 22.8 μM and function relationships were found between blue-shifts and the concentrations of sulfonamides in pH 5 acetic acid–sodium acetate buffer solution. The developed method has been applied successfully to detect sulfonamides in egg white samples without using labelling techniques and expensive instruments.

According to problems in manufacturing and measuring process of cycloidal gear used in RV reducer, the profile coordinates of cycloidal gear captured by three coordinates measuring instrument Surveying and mapping do not coincide with the designed tooth profile coordinates in theory, it is difficult to precisely capture the addendum and dedendum of each cycloidal gear teeth, furthermore, the captured addendum and dedendum do not coincide with the actual tooth items. For these problems, this paper introduced a method can accurately calculating the manufacturing error of cycloidal gear.Based on the captured coordinate points of cycloidal gear tooth profile, firstly, find out 39 approximate addendums by calculating and comparing the radial radius of all points, divide the whole scanning points into 39 profile points groups by the 39 approximate addendums, with each group include a whole single tooth profile. Then, in each profile points group, interpolate the spline and precisely calculate the addendums and dedendums by fitting and recalculating radial radius.Set up the coordinate system of surveying and mapping profile by the 39 addendums. calibrate the coordinate system of surveying and mapping profile with the new coordinate system through coordinate transformation, calculate the radial runout error at top and root of tooth profile, pitch error, adjacent pitch error and accumulated pitch error as well as other manufacturing errors of the cycloidial gear at last.Through experiment, this process solve the problem in measuring process of cycloidal gear that the captured point by three coordinates measuring instrument could not be directly used for calculating the errors items.

Aiming at the problems of pitting, shafts broken and so on in the process of the use of boom and crowning mechanism gear box of a large mining excavator, research found that the key problems of broken shafts and the large bearings clearance are caused by excessive shafts and gear teeth deformation, which result in serious corrosion occurred by partial load and serious bearing damage.

In order to elucidate the effect of the structure on stability and gas adsorption performance, three porous Zn(II) metal–organic frameworks, NKU-106, NKU-107, and NKU-108, have been constructed. NKU-106 and NKU-107 assembled from acylamide functionalized tetracarboxylate ligand exhibit low stability and thereby poor gas adsorption ability. However, when one isophthalate moiety of the tetracarboxylate ligand is replaced with a pyrazole group, and the resulting NKU-108 shows improved thermal and chemical stability as well as good adsorption capacities with respect to CO2 and C2 hydrocarbons. This result demonstrates that coordination group optimization is an effective strategy to improve the stability and gas adsorption properties of metal–organic frameworks (MOFs) and thus provides very valuable information for future design and synthesis of porous MOFs for practical application.
