Abstract Autophagy is closely related to the occurrence and development of human malignancies; however, the detailed mechanisms underlying autophagy in cervical cancer require further investigation. Previously, we found that the ectopic expression of NCAPH, a regulatory subunit of condensed protein complexes, significantly enhanced the proliferation of tumor cells; however, the underlying mechanisms were unclear. Here, we revealed that NCAPH is a novel autophagy-associated protein in cervical cancer that promotes cell proliferation by inhibiting autophagosome formation and reducing autophagy, with no effect on the cell cycle, apoptosis, or aging. Tripartite motif-containing protein 21 (TRIM21) is well known to be involved in inflammation, autoimmunity and cancer, mainly via its E3 ubiquitin ligase activity. Mass spectrometry and immunoprecipitation assays showed that TRIM21 interacted with NCAPH and decreased the protein stability of NCAPH via ubiquitination at the K11 lysine residue. Structural domain mutation analysis revealed that TRIM21 combined with NCAPH through its PRY/SPRY and CC domains and accelerated the degradation of NCAPH through the RING domain. Furthermore, TRIM21 promoted autophagosome formation and reduced cell proliferation by inhibiting NCAPH expression and the downstream AKT/mTOR pathway in cervical cancer cells. Immunohistochemical staining revealed that the protein expression of TRIM21 was negatively correlated with that of NCAPH and positively correlated with that of beclin-1 in cervical cancer tissues. Therefore, we provide evidence for the role of the TRIM21-NCAPH axis in cervical cancer autophagy and proliferation and the involvement of the AKT/mTOR signaling pathway in this process. These results deepen our understanding of the carcinogenesis of cervical cancer, broaden the understanding of the molecular mechanisms of TRIM21 and NCAPH, and provide guidance for individualized treatment of cervical cancer in the future.

To reduce the losses caused by insufficient preservation efforts during transportation, the preservation effort level has been the focus of research. In the fierce competition of online sales, it is particularly important to reduce the cost of damaged goods by improving the level of preservation efforts. Therefore, according to Stackelberg game theory, this article establishes five decision-making models and incorporates the damage rate and preservation effort level into the research. Finally, this article coordinates the online shipping supply chain (SC) through a joint contract. After comparing and analyzing the model results, research has found that: (1) in centralized model, the level of preservation effort reaches its optimal level and the system benefit is maximized; (2) under third-party logistics (TPL) leading decision-making, the different bearers of cargo damage costs will not affect the profits of both parties and the system; (3) among the four decentralized models, the level of preservation efforts and system profit are highest when the decision is led by online store and TPL bears the cost of damaged goods; and (4) under a given sharing ratio, when the logistics service quotation satisfies a certain range of condition, the online shopping SC can achieve Pareto improvement. This paper studies the differences and reasons for decision models in the supply and demand relationship between online stores and TPL, which provides fresh product e-commerce decision-makers with a theoretical basis.

Wooden cantilever covered bridges are known for their marvelous shapes and manufacturing technique. Many wooden cantilever covered bridges were constructed during Ming and Qing Dynasties in Anhua province, China, due to the “Tea-Horse Trade” policy. The excellent performance of wooden materials and exquisite building techniques have kept these wooden bridges well-preserved and worthy of investigation. This paper conducted a comprehensive review of wooden cantilever covered bridges in Anhua, especially for the eight bridges listed as historical and cultural heritage protected at the provincial or the national level. The discussions covered the historical background of the bridges, their locations and dimensions, and the details of their structures including the cantilever systems, corridors, and roofs. Moreover, the cultural background was introduced to better understand the meaning of the decorations carved on the bridges and the logic of location selections.

Presently, Non-alcoholic fatty liver disease (NAFLD) has become the most prevalent liver metabolic disease affecting the health of global citizens. The high activity and low toxicity of natural products make the use of dietary strategies to prevent NAFLD becoming a frontier hot spot. We investigated the broccoli, a natural food with lipid-lowering and anti-inflammatory properties, can ameliorate NAFLD in mice fed with Methionine and choline-deficient (MCD) diet for 6-weeks/8-weeks. The following mechanisms may be involved: I) Increased the mRNA expression of key lipolysis genes; II) Inhibited the secretion of TNF-α, IL-1β, IL-6 and increased the secretion of IL-10; III) Reduced the production of NO and enhanced the activity of Arginase. Regulating the M1/M2 polarization of macrophages may be related mechanisms that broccoli suppressed the inflammatory process of NAFLD. This experiment will provide a theoretical basis for dietary intervention with broccoli in delaying the occurrence and development of NAFLD.

Genetic modification (GM) offers an alternative strategy to conventional animal breeding. The present study was carried out to investigate the potential health effects of the consumption of growth hormone-transgenic triploid carp (GH-ttc) through a 90-day subchronic rodent feeding study. Wistar rats (n = 10/sex/group) were given formulated diets containing GH-ttc or non-GM carp at an incorporated rate of 2.5%, 5%, or 10% (w/w) for 90 days. An additional control group of rats (n = 10/sex/group) was fed a basic rodent diet. During the 90-day study, clinical observation, ophthalmic examination, body weight, and food intake were evaluated. At the end of the study, rats were killed, and the hematology, serum chemistry, urine test, necropsy, and histopathology were assessed. Compared with the non-GM carp and the basic control groups, no biologically significant differences were observed on clinical signs of toxicity, body weights, food intake, hematology, serum chemistry, urinalysis, organ weight, and histopathology on selected organs for the GH-ttc group. The results of this 90-day subchronic feeding study indicated that, at the dose level used in this study, consumption of GH-ttc showed no subchronic toxicity to Wistar rats.

Abstract Background Calorie restriction (CR), which has a potent anti-inflammaging effect, has been demonstrated to induce dramatic changes in the gut microbiota. Whether the modulated gut microbiota contributes to the attenuation of inflammation during CR is unknown, as are the members of the microbial community that may be key mediators of this process. Results Here, we report that a unique Lactobacillus-predominated microbial community was rapidly attained in mice within 2 weeks of CR, which decreased the levels of circulating microbial antigens and systemic inflammatory markers such as tumour necrosis factor alpha (TNF-α). Lactobacillus murinus CR147, an isolate in the most abundant operational taxonomic unit (OTU) enriched by CR, downregulated interleukin-8 production in TNF-α-stimulated Caco-2 cells and significantly increased the lifespan and the brood size of the nematode Caenorhabditis elegans. In gnotobiotic mice colonized with the gut microbiota from old mice, this strain decreased their intestinal permeability and serum endotoxin load, consequently attenuating the inflammation induced by the old microbiota. Conclusions Our study demonstrated that a strain of Lactobacillus murinus was promoted in CR mice and causatively contributed to the attenuation of ageing-associated inflammation.

Abstract Background Little is known about the link between the fungal community and phosphorus fractions when land use is converted from cropland to grassland in saline-alkali soil in northeastern China. Therefore, in this study, the diversity, composition, and function of fungi, as well as phosphorus fractions including Olsen-P and inorganic phosphorus (Pi), were investigated under land use conversions from maize cropland (MC) to alfalfa (Medicago sativa L.) (AG), Leymus chinensis (LG), and natural restored grasslands (RG). Results The results showed that the Pi fractions of Ca8-P, Fe-P, Ca2-P, and Ca10-P were closely related with Olsen-P. IntroductionThe sowing date plays a crucial role in influencing the growth and reproduction of plants, with its specific impact on biomass allocation and allometric growth remaining unclear. Understanding these effects is essential for optimizing agricultural practices and enhancing crop productivity. MethodsTo investigate the effects of sowing dates on biomass allocation and allometric growth, a field experiment was conducted with sequential sowings of Fagopyrum esculentum from April 12th to August 11th in 2018. Biomass measurements were taken across various plant organs, and corresponding allocation calculations were made. A detailed analysis of the allometric growth relationship involving organ biomass variations was performed. ResultsThe study revealed that the accumulation and allocation of organ biomass in buckwheat were significantly impacted by the sowing dates. Delayed planting led to reduced vegetative growth and increased biomass allocation towards reproduction. Allometric parameters such as exponent, constant, and individual size of buckwheat were notably affected by delayed planting. Interestingly, the allometric exponents governing the relationships between reproductive vs. vegetative biomass and belowground vs. aboveground biomass exhibited varying trends across different sowing dates. DiscussionNotably, late sowings resulted in significantly higher reproductive biomass compared to early and middle sowings. These findings highlight the nuanced relationship between plant size and reproductive biomass under different sowing dates, emphasizing the critical role of planting timing in shaping mature plant sizes and reproductive outcomes. The study underscores the importance of considering sowing dates in agricultural practices to optimize plant growth and productivity.

BackgroundTuberculosis is a worldwide epidemic disease, posing a serious threat to human health. To find effective drug action targets for Mycobacterium tuberculosis, differentially expressed genes in tuberculosis patients and healthy people were screened by mRNA sequencing in this study. A total of 556 differentially expressed genes in tuberculosis patients and healthy people were screened out by mRNA sequencing technology. 26 transcription factors and 66 corresponding target genes were screened out in the AnimalTFDB 3.0 database, and a transcription factor regulatory network was constructed.ResultsThree key transcription factors (TP53, KLF5 and GATA2) and one key gene (AKT1) were screened as new potential drug targets and diagnostic targets for tuberculosis by MCODE cluster analysis, and the key genes and key transcription factors were verified by RT-PCR. Finally, we constructed the and a key factor and KEGG signaling pathway regulatory network to clarify the possible molecular pathogenesis of tuberculosis.ConclusionThis study suggested M. tuberculosis may activate the AKT1 gene expression by regulating transcription factors TP53, KLF5, and GATA2, thus activating the B cell receptor signaling pathway to induce the infection and invasion of M. tuberculosis. AKT1, TP53, KLF5, and GATA2 can be used as new potential drug targets for tuberculosis.

Microscale robotics has emerged as a transformative field, offering unparalleled opportunities for innovation and advancement in various fields. Owing to the distinctive benefits of wireless operation and a heightened level of safety, magnetic actuation has emerged as a widely adopted technique in the field of microrobotics. However, factors such as Brownian motion, fluid dynamic flows, and various nonlinear forces introduce uncertainties in the motion of micro/nanoscale robots, making it challenging to achieve precise navigational control in complex environments. This paper presents an extensive review encompassing the trajectory from theoretical foundations of the generation and modeling of magnetic fields as well as magnetic field-actuation modeling to motion control methods of magnetic microrobots. We introduce traditional control methods and the learning-based control approaches for robotic systems at the micro/nanoscale, and then these methods are compared. Unlike the conventional navigation methods based on precise mathematical models, the learning-based control and navigation approaches can directly learn control signals for the actuation systems from data and without relying on precise models. This endows the micro/nanorobots with high adaptability to dynamic and complex environments whose models are difficult/impossible to obtain. We hope that this review can provide insights and guidance for researchers interested in automated magnetic microrobot control.

Abstract Abstract Images taken in low‐light conditions often suffer from reduced contrast, amplified noise, and colour distortions. Such degradation falls short of viewer expectations and negatively impacts high‐level tasks, such as object detection. In this paper, the above problems are addressed by leveraging the properties of the LAB colour space and a multi‐scale feature pyramid. Specifically, the model first generates a light attention map and a noise map, which are proficient at manipulating the luminance channel for brightness recovery while preserving the colour information and separating noise from the image content. Moreover, enriched contextual features is progressively extracted and then the low‐light images are refined by incorporating the light attention map and the noise map as priors into a coarse‐to‐fine architecture. The enhanced results are characterized by improved contrast, vibrant colour, and noise suppression.

IntroductionSymbiotic N fixation inhibition induced by N supply to legumes is potentially regulated by the relative N and P availability in soil. However, the specific responses of different legume species to changes in N:P availability remain unclear, and must be better understood to optimize symbiotic N fixation inputs under N enrichment. This study investigated mechanisms by which soil N and P supply influence the symbiotic N fixation of eight legume species, to quantify the inter-specific differences, and to demonstrate how these differences can be determined by the stoichiometric homeostasis in N:P ratios (HN:P).MethodsEight herbaceous legume species were grown separately in outdoor pots and treated with either no fertilizer (control), N fertilizer (14 g N m-2), P fertilizer (3.5 g P m-2) or both N and P fertilizer. Plant nutrients, stoichiometric characteristics, root biomass, non-structural carbohydrates (NSC), rhizosphere chemistry, P mobilization, root nodulation and symbiotic N fixation were measured.ResultsN addition enhanced rhizosphere P mobilization but drove a loss of root biomass and root NSC via exudation of P mobilization compound (organic acid), especially so in treatments without P addition. N addition also induced a 2-14% or 14-36% decline in symbiotic N fixation per plant biomass by legumes in treatments with or without P addition, as a result of decreasing root biomass and root NSC. The changes in symbiotic N fixation were positively correlated with stoichiometric homeostasis of N:P ratios in intact plants without root nodules, regardless of P additions.DiscussionThis study indicates that N addition can induce relative P limitations for growth, which can stimulate rhizosphere P mobilization at the expense of root biomass and carbohydrate concentrations, reducing symbiotic N fixation in legumes. Legume species that had less changes in plant N:P ratio, such as Lespedeza daurica and Medicago varia maintained symbiotic N fixation to a greater extent under N addition.

Abstract Background Cyclin-dependent kinase subunit 2 (CKS2) is a member of cyclin dependent kinase subfamily and the relationship between CKS2 and osteosarcoma (OS) remains to be further analyzed. Methods 80 OS and 41 non-tumor tissue samples were arranged to perform immunohistochemistry (IHC) to evaluate CKS2 expression between OS and non-tumor samples. The standard mean deviation (SMD) was calculated based on in-house IHC and tissue microarrays, and exterior high-throughput datasets for further verification of CKS2 expression trend in OS. The effect of CKS2 expression on clinicopathological parameters of OS patients, and single-cell in OS tissues was analyzed through public high-throughput datasets and functional enrichment analysis was conducted for co-expression genes of CKS2 in accordance with weighted correlation network analysis. Results A total of 217 OS samples and 87 non-tumor samples (including tissue and cell line) were obtained from in-house IHC, microarrays and exterior high-throughput datasets. The analysis of integrated expression status demonstrated up-regulation of CKS2 in OS (SMD = 1.57, 95%CI [0.27–2.86]) and the significant power of CKS2 expression in distinguishing OS samples from non-tumor samples (AUC = 0.97 95%CI [0.95–0.98]). Clinicopathological analysis of GSE21257 indicated that OS patients with higher CKS2 expression was more likely to suffer OS metastasis. Although Kaplan–Meier curves showed no remarkable difference of overall survival rate between OS patients with high and low-CKS2, CKS2 was found up-regulated in proliferating osteosarcoma cells. Co-expression genes of CKS2 were mainly assembled in function and pathways such as cell cycle, cell adhesion, and intercellular material transport. Conclusions In summary, up-regulation of CKS2 expression in OS tissue was found through multiple technical approaches. In addition, scRNA-seq and co-expression analysis showed that CKS2 may have an impact on important biological process linked with cell cycle, cell adhesion, and intercellular material transport. Present study on CKS2 in OS indicated a promising prospect for CKS2 as a biomarker for OS.

Mutual dislocation of seismogenic faults during strong earthquakes will result in a large relative displacement on both sides of the fault. It is of great significance to explore the influence of the collision effect between the main beam and the transverse shear key on the seismic response of the bridge under fault dislocation. In this paper, a series of cross-fault ground motions with different ground permanent displacements are artificially synthesized using a hybrid simulation method. Based on the contact element theory, the Kelvin–Voigt model is used to simulate the lateral collision effect. The effect of lateral collision on the seismic response of the continuous girder bridge is compared from the two aspects of fault dislocation position and fault dislocation degree. On this basis, the analysis of lateral collision parameters is carried out with the aim of reasonably regulating the seismic response of the structure. The results show that, compared with the near-fault bridge, the influence of lateral collision on the cross-fault bridge is stronger. The amplification of the bending moment of the central pier and the limitation of the bearing displacement are five times and two times, respectively, for the near-fault bridge. When the fault has a large dislocation, the weak point of the structural damage is the bending failure of the pier bottom and the residual torsion after the earthquake. The collision parameters of conventional bridges will aggravate the bending moment demand of the pier bottom of cross-fault bridges and limit their bearing displacement too much. Therefore, by appropriately reducing the collision stiffness and increasing the initial gap, the internal force and displacement response distribution of the cross-fault bridge structure can be more reasonable. The study in this paper has reference significance for seismic analysis of cross-fault bridges with transverse shear keys.

The variations of medium mineral elements in long-term grazing exclusion grassland have been few studied. Therefore, the contents, stocks, and proportions of calcium (Ca), magnesium (Mg), sulphur (S), and silicon (Si) in green plant-litter-root-soil continuum were evaluated by grazing exclusion for 18-, and 39-year (F18 and F39) and continue grazing (F0) in Mongolia grassland.
