Context Yi-Shen-Hua-Shi (YSHS) is a traditional Chinese medicine that treats chronic kidney disease (CKD). However, its efficacy in reducing proteinuria and underlying mechanisms is unknown.Objective This single-center randomized controlled trial explored whether YSHS could improve proteinuria and modulate the gut microbiota.Materials and methods 120 CKD patients were enrolled and randomized to receive the renin-angiotensin-aldosterone system (RAAS) inhibitor plus YSHS (n = 56) or RAAS inhibitor (n = 47) alone for 4 months, and 103 patients completed the study. We collected baseline and follow-up fecal samples and clinical outcomes from participants. Total bacterial DNA was extracted, and the fecal microbiome was analyzed using bioinformatics.Results Patients in the intervention group had a significantly higher decrease in 24-h proteinuria. After 4 months of the YSHS intervention, the relative abundance of bacteria that have beneficial effects on the body, such as Faecalibacterium, Lachnospiraceae, Lachnoclostridium, and Sutterella increased significantly, while pathogenic bacteria such as the Eggerthella and Clostridium innocuum group decreased. However, we could not find these changes in the control group. Redundancy analysis showed that the decline in 24-h proteinuria during follow-up was significantly correlated with various taxa of gut bacteria, such as Lachnospiraceae and the Lachnoclostridium genus in the YSHS group. KEGG analysis also showed the potential role of YSHS in regulating glycan, lipid, and vitamin metabolism.Discussion and conclusion The YSHS granule reduced proteinuria associated with mitigating intestinal microbiota dysbiosis in CKD patients. The definite mechanisms of YSHS to improve proteinuria need to be further explored.Trial registration ChiCTR2300076136, retrospectively registered.

To achieve efficient degradation of crustacean raw materials and eco-preparation of N-acetyl-oligosaccharides, YS-54 strain screened from offshore soil in Qingdao was used for chitinase preparation in the present study. Chitinase was prepared through 60% ammonium sulfate precipitation and Sephadex G-100 gel filtration chromatography. Enzymatic properties such as optimal temperature, optimal pH, and substrate specificity were characterized. Mass spectrometry was used to determine the polymerization degree of enzymatic products. The 16S rRNA identification result showed that YS-54 strain belonged to the Aeromonas species. Chitinase from Aeromonas sp. YS-54 showed a specific activity of 23.44 U/mg. The molecular weights of chitinase were 63 and 75 kDa as shown by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Chitinase was stable under the condition of 40 ℃ and pH5.0. Objective This study aimed to compare the efficacy and safety of intravascular ultrasound (IVUS)-guided coronary intravascular lithotripsy and rotational atherectomy in treating severe coronary artery calcification. Methods A retrospective analysis of 60 patients who underwent percutaneous coronary intervention at the General Hospital of Ningxia Medical University from October 2022 to August 2023 was conducted. The patients were divided into two groups: 30 received IVUS-guided coronary intravascular lithotripsy and 30 underwent IVUS-guided rotational atherectomy. During the operation of the Salt Cavern Flow Battery (SCFB) system, the rock surrounding a salt cavern is subjected to erosion by the electrolyte. To study the creep characteristics of electrolyte-bearing salt rock under long-term triaxial cyclic loading in SCFB, a triaxial creep experiment with a cycle period of 1 day was conducted. The results indicated that, when not subjected to failure, the axial stress-strain curve of electrolyte-bearing sample undergoes only two phases of “sparse-dense”, entering dense phase approximately 4 cycles earlier than that of sample without electrolyte. Under the same stress conditions, the strain generated in electrolyte-bearing salt rock surpasses that of sample without electrolyte, demonstrating an initial rapid increase followed by a gradual stabilization trend. The stress-strain curve of electrolyte-bearing sample in a single cycle can be divided into six stages. The number of cycles has almost no effect on the axial strain in stages I, IV, V and VI, and the axial strain in stages IV and VI is basically 0. Additionally, the elastic deformation generated in stage I is basically recovered in stage V. The strain in stage II gradually decreases and disappears in the 4th cycle, which is 13 cycles earlier than that of the sample without electrolyte. The creep rate of electrolyte-bearing sample shows a trend of “gradual decrease—basically stabilization” as the number of cycles increases, and the creep experiment contains only the decay creep stage and steady creep stage. Irreversible deformation of electrolyte-bearing sample exhibits a gradual decrease followed by stabilization with increasing number of cycles. The research findings hold significant implications for the stability analysis of SCFB systems.

In order to investigate the distribution and accumulation characteristics of metabolites and mineral elements in different parts of Peganum harmala L. (P. harmala), and the synergistic or antagonistic effects between them. In this study, nuclear magnetic resonance (NMR), high performance liquid chromatography (HPLC) and inductively coupled plasma optical emission spectrometer (ICP-OES) were used to determine the contents of metabolites (proline, phosphorylcholine, choline, lysine, 4-hydroxyisoleucine, asparagine, acetic acid, sucrose, harmaline and vasicine) and mineral elements (Ca, Mg, K, P, Na, Cr, Cu, Fe, Zn, Mn, Ni, C, N) in five parts of P. harmala, including root, seed, testa, stem and leaf, and to analyze the relationship among the contents of metabolites and mineral elements. The results showed that the contents of acetic acid, proline, lysine, sucrose and Fe in the root were higher than those in other parts, and the contents of harmaline, phosphorylcholine, P, C, N and Zn in the seeds were the highest. The leaves were rich in vasicine, Na, K, Ca, Mg and Mn. The principal component analysis (PCA) showed that the cumulative variance contribution of the first two principal components was 69.00 %, and the loading values of K, Cu and sucrose were higher, which was consistent with the results of biplot and cluster analysis(HCA). Correlation analysis (CA) results showed that there was a strong overall correlation between the different components of seeds and leaves, and the correlation was greater than that of other parts. The results of this study are helpful to understand the correlation of functional traits among different parts of plants, and determine the internal mechanism of controlling functional traits and the proportional relationship between traits, so as to provide a reference for the resource utilization of plants.

Cardiac arrest and successful resuscitation cause whole-body ischemia and reperfusion, leading to brain injury and extracerebral multiple organ dysfunction. Brain injury is the leading cause of death and long-term disability in resuscitated survivors, and was conceptualized and treated as an isolated injury, which has neglected the brain-visceral organ crosstalk. Extracerebral organ dysfunction is common and is significantly associated with mortality and poor neurological prognosis after resuscitation. However, detailed description of the characteristics of post-resuscitation multiple organ dysfunction is lacking, and the bidirectional interactions between brain and visceral organs need to be elucidated to explore new treatment for neuroprotection. This review aims to describe current concepts of post-cardiac arrest brain injury and specific characteristics of post-resuscitation dysfunction in cardiovascular, respiratory, renal, hepatic, adrenal, gastrointestinal, and neurohumoral systems. Additionally, we discuss the crosstalk between brain and extracerebral organs, especially focusing on how visceral organ dysfunction and other factors affect brain injury progression. We think that clarifying these interactions is of profound significance on how we treat patients for neural/systemic protection to improve outcome.

Offshore wind power construction has seen significant development due to the high density of offshore wind energy and the minimal terrain restrictions for offshore wind farms. However, integrating this energy into the grid remains a challenge. The scientific community is increasingly focusing on hydrogen as a means to enhance the integration of these fluctuating renewable energy sources. This paper reviews the research on renewable energy power generation, water electrolysis for hydrogen production, and large-scale hydrogen storage. By integrating the latest advancements, we propose a system that couples offshore wind power generation, seawater electrolysis (SWE) for hydrogen production, and salt cavern hydrogen storage. This coupling system aims to address practical issues such as the grid integration of offshore wind power and large-scale hydrogen storage. Regarding the application potential of this coupling system, this paper details the advantages of developing renewable energy and hydrogen energy in Jiangsu using this system. While there are still some challenges in the application of this system, it undeniably offers a new pathway for coastal cities to advance renewable energy development and sets a new direction for hydrogen energy progress.

To address the inherent intermittency and instability of renewable energy, the construction of large-scale energy storage facilities is imperative. Salt caverns are internationally recognized as excellent sites for large-scale energy storage. They have been widely used to store substances such as natural gas, oil, air, and hydrogen. With the global transition in energy structures and the increasing demand for renewable energy load balancing, there is broad market potential for the development of salt cavern energy storage technologies. There are three types of energy storage in salt caverns that can be coupled with renewable energy sources, namely, salt cavern compressed air energy storage (SCCAES), salt cavern hydrogen storage (SCHS), and salt cavern flow battery (SCFB). The innovation of this paper is to comprehensively review the current status and future development trends of these three energy storage methods. Firstly, the development status of these three energy storage methods, both domestically and internationally, is reviewed. Secondly, according to the characteristics of these three types of energy storage methods, some key technical challenges are proposed to be focused on. The key technical challenge for SCCAES is the need to further reduce the cost of the ground equipment; the key technical challenge for SCHS is to prevent the risk of hydrogen leakage; and the key technical challenge for SCFB is the need to further increase the concentration of the active substance in the huge salt cavern. Finally, some potential solutions are proposed based on these key technical challenges. This work is of great significance in accelerating the development of salt cavern energy storage technologies in coupled renewable energy.

This article addresses the common issues of insufficient computing power and path congestion for automated guided vehicles (AGVs) in real-world production environments, as well as the shortcomings of traditional path-planning algorithms that mainly consider the shortest path while ignoring vehicle turning time and stability. We propose a secondary path-planning method based on an improved directed weighted graph theory integrated with an ROS. Firstly, the production environment is modeled in detail to identify the initial position of the AGV. Secondly, the operational area is systematically divided, key nodes are selected and optimized, and a directed weighted graph is constructed with optimized weights. It is integrated with the ROS for path planning, using the Floyd algorithm to find the optimal path. The effectiveness and superiority of this method have been demonstrated through simulation verification and actual AGV operation testing. The path planning strategy and fusion algorithm proposed in this article that comprehensively considers distance and angle steering are simple and practical, effectively reducing production costs for enterprises. This method is suitable for logistics sorting and small transport AGVs with a shorter overall path-planning time, higher stability, and limited computing power, and it has reference significance and practical value.

Salt cavern flow batteries (SCFBs) are an energy storage technology that utilize salt caverns to store electrolytes of flow batteries with a saturated NaCl solution as the supporting electrolyte. However, the geological characteristics of salt caverns differ significantly from above-ground storage tanks, leading to complex issues in storing electrolytes within salt caverns. Therefore, investigating and summarizing these issues is crucial for the advancement of SCFB technology. This paper’s innovation lies in its comprehensive review of the current state and development trends in SCFBs both domestically and internationally. First, the current development status of SCFB energy storage technology both domestically and internationally is summarized. Then, eight main issues are proposed from the perspectives of salt cavern geological characteristics (tightness, conductivity, ions, and temperature) and electrolyte properties (selection, permeability, corrosion, and concentration). Finally, a novel SCFB system is proposed to address the most critical issue, which is the low concentration and uneven distribution of active materials in the current SCFB system. The review in this paper not only comprehensively summarizes the development status of SCFBs both domestically and internationally, but also points out the direction for the future research focussing on SCFBs.

This paper proposes a nonlinear Model Predictive Control (MPC) method based on Physics-Informed Neural Networks (PINNs), aimed at enhancing the trajectory tracking performance of Automated Guided Vehicles (AGVs) in complex dynamic environments. Traditional physical models often face the challenges of computational inefficiency and insufficient control precision when dealing with complex dynamic systems. However, by integrating physical laws directly into the training process of neural networks, PINNs can effectively learn and capture the kinematic characteristics of vehicles, replacing traditional nonlinear ordinary differential equation models and thus significantly enhancing computational efficiency and control performance. During the model-training phase, this study further incorporates the Theory of Functional Connections (TFC) and adaptive loss balancing strategies to efficiently solve ODE problems without relying on numerical integration and optimize the control strategy. This combined approach not only reduces computational complexity, but also improves the robustness and precision of the control strategy in varying environments. Numerical simulations demonstrate that this method offers significant advantages in AGV trajectory-tracking tasks, manifested in higher computational efficiency and precise control performance. The proposal of the PINN-MPC method provides new theoretical support and innovative methods for real-time complex system control, with important research and application potential, and is expected to play a key role in future intelligent control systems.

The sweetpotato whitefly, Bemisia tabaci (Gennadius) Middle East–Asia Minor 1 (MEAM1), causes significant losses to vegetable crops directly by sap-feeding, inducing plant physiological disorders, and elevating the build-up of sooty mold, and indirectly by transmitting plant viruses. In this study, we evaluated the susceptibility of 20 yellow squash and zucchini (Cucurbita pepo) cultivars to MEAM1, across three growing seasons in the southeastern United States. Weekly sampling of the numbers of MEAM1 adults, nymphs, and eggs were conducted from the fourth week after seed sowing and across 6 weeks during the summer and fall of 2021 and five weeks during the fall of 2022. In general, adult whitefly populations were high during the first week of sampling but decreased as the seasons progressed. The zucchini cultivar ‘Black Beauty’ harbored the most adults, while ‘Green Eclipse Zucchini’ was the least attractive zucchini cultivar to the adults in fall 2022. For yellow squash, ‘Early Summer’ (summer 2021) and ‘Amberpic 8455’ (summer 2021 and fall 2022) were the cultivars with the highest adult populations, while ‘Lioness’ (summer 2021) and ‘Gourmet Gold Hybrid’ (fall 2022) harbored the lowest adult counts. The whitefly egg counts across both vegetables trailed those of adults and peaked in the second week of sampling. The counts of nymphs increased as the seasons progressed, but there was a decline after the second week during fall 2021. For the yellow squash cultivars, ‘Gourmet Gold Hybrid’, (summer 2021 and fall 2022), ‘Lioness’, and ‘Fortune’ (summer 2021) recorded the highest yields. For zucchini, ‘Golden Glory’ (summer 2021) was the top performer. These results provide valuable information for whitefly management in yellow squash and zucchini based on host plant susceptibility and yield.

Mycotoxins are toxic secondary metabolites produced by certain fungi, which can contaminate various food commodities, including fruits and their derived products. Patulin and Alternaria toxins are among the most commonly encountered mycotoxins in fruit and their derived products. In this review, the sources, toxicity, and regulations related to these mycotoxins, as well as their detection and mitigation strategies are widely discussed. Patulin is a mycotoxin produced mainly by the fungal genera Penicillium, Aspergillus, and Byssochlamys. Alternaria toxins, produced by fungi in the Alternaria genus, are another common group of mycotoxins found in fruits and fruit products. The most prevalent Alternaria toxins are alternariol (AOH) and alternariol monomethyl ether (AME). These mycotoxins are of concern due to their potential negative effects on human health. Ingesting fruits contaminated with these mycotoxins can cause acute and chronic health problems. Detection of patulin and Alternaria toxins in fruit and their derived products can be challenging due to their low concentrations and the complexity of the food matrices. Common analytical methods, good agricultural practices, and contamination monitoring of these mycotoxins are important for safe consumption of fruits and derived products. And Future research will continue to explore new methods for detecting and managing these mycotoxins, with the ultimate goal of ensuring the safety and quality of fruits and derived product supply.

As a highly economic small fruit crop, blueberry is enjoyed by most people in terms of color, taste, and rich nutrition. To better understand its coloring mechanism on the process of ripening, an integrative analysis of the metabolome and transcriptome profiles was performed in three blueberry varieties at three developmental stages. In this study, 41 flavonoid metabolites closely related to the coloring in blueberry samples were analyzed. It turned out that the most differential metabolites in the ripening processes were delphinidin-3-O-arabinoside (dpara), peonidin-3-O-glucoside (pnglu), and delphinidin-3-O-galactoside (dpgal), while the most differential metabolites among different varieties were flavonols. Furthermore, to obtain more accurate and comprehensive transcripts of blueberry during the developmental stages, PacBio and Illumina sequencing technology were combined to obtain the transcriptome of the blueberry variety Misty, for the very first time. Finally, by applying the gene coexpression network analysis, the darkviolet and bisque4 modules related to flavonoid synthesis were determined, and the key genes related to two flavonoid 3′, 5′-hydroxylase (F3′5′H) genes in the darkviolet module and one bHLH transcription factor in the bisque4 module were predicted. It is believed that our findings could provide valuable information for the future study on the molecular mechanism of flavonoid metabolites and flavonoid synthesis pathways in blueberries.

Abstract Background Cardiac Arrest (CA) is one of the leading causes of mortality worldwide. The present study aimed to establish a simple and stable rat model of CA induced by transesophageal cardiac pacing for the investigation of cerebral resuscitation. Materials and Methods A total of 26 healthy adult male Sprague-Dawley rats were randomly allocated into two groups: Sham-operated (n = 6) and experimental (n = 20) groups. High-frequency cardiac pacing (50 Hz, 2 ms and 30 V) was maintained for 3 min to induce CA. Providing CA was not achieved, an additional 2 min of pacing was performed 30 min later. After 4 min following the onset of CA, Cardiopulmonary Resuscitation (CPR) was initiated. Results CA was successfully induced in all 20 rats by this setting of high-frequency cardiac pacing. Among them, CA was induced in six rats after 2 min of pacing; the remaining 14 rats underwent CA after 3 min of pacing. When electrical stimulation was terminated the rate of Pulseless Electrical Activity (PEA) was 85% (17/20), the rate of Ventricular Fibrillation (VF) was 15% (3/20) and no asystole occurred. Of the 17 PEA rats, 16 were successfully resuscitated and the average duration of CPR was 106.75 ± 30.81 s. To increase natural gas storage capacity and further utilize salt mine resources, salt cavern gas storage in the Yunying salt mine, Hubei Province, China, was simultaneously constructed in two different mining layers (K3 and K4). The purpose of this study was to investigate the long-term feasibility of operating salt caverns for gas storage in two mining layers. Based on the geological conditions and sonar test results, the geometric parameters for the salt caverns in the two mining layers were designed, and a 3D geomechanical model was built to predict the cavern stability. The corresponding evaluation index included the displacement, volume shrinkage rate, equivalent strain, and dilatancy factor. The results show that simultaneously operating salt cavern gas storage in two mining layers is feasible, and the operational pressures for the salt caverns in mining layers K3 and K4 should be no less than 4–9 and 7–12 MPa, respectively, to satisfy the stability requirements. The surrounding rock of the salt caverns presents a larger displacement and volume reduction compared with cases in which the salt caverns are operated in a single mining layer. Increasing the injection–withdrawal frequency increases the deformation of the surrounding rock.

Peaches are the most significant temperate fruit crop worldwide. However, peach fruits are susceptible to fungal and mycotoxin contamination. Consequently, monitoring the residual levels of multiple mycotoxins in peaches and related products is essential. In this study, a novel method based on QuEChERS extraction, followed by ultra-high-performance liquid chromatography–tandem mass spectrometry (UPLC-MS/MS) detection, was developed for analyzing 14 mycotoxins in peaches and peach products from China. Matrix-matched calibrations were employed to accurately quantify the mycotoxins and compensate for matrix effects. Recoveries for the target analytes ranged from 84.6% to 117.6%, with intra-day and inter-day precision below 20%. The limits of quantification were 2 or 5 μg/L for the 14 mycotoxins. This method was utilized to detect the presence of target mycotoxins in 109 fresh peaches, 100 diseased peaches, and 89 peach products from China. Six mycotoxins were identified in the rotten parts of the diseased peaches, with concentrations ranging from 5.2 to 1664.3 µg/kg. In the remaining parts of the diseased peach samples, only two toxins, alternariol (AOH) and alternariol monomethyl ether (AME), were quantified at levels of 15.3 µg/kg and 15.5 µg/kg, respectively. No mycotoxins were detected in fresh peaches. For peach products, all contamination levels were below the quantitative limits and significantly lower than the maximum legal limits established for the products.

Peripheral nerve regeneration requires stepwise and well-organized establishment of microenvironment. Since local delivery of VEGF-A in peripheral nerve repair is expected to promote angiogenesis in the microenvironment and Schwann cells (SCs) play critical role in nerve repair, combination of VEGF and Schwann cells may lead to efficient peripheral nerve regeneration. VEGF-A overexpressing Schwann cells were established and loaded into the inner wall of hydroxyethyl cellulose/soy protein isolate/polyaniline sponge (HSPS) conduits. When HSPS is mechanically distorted, it still has high durability of strain strength, thus, can accommodate unexpected strain of nerve tissues in motion. A 10 mm nerve defect rat model was used to test the repair performance of the HSPS-SC (VEGF) conduits, meanwhile the HSPS, HSPS-SC, HSPS-VEGF conduits and autografts were worked as controls. The immunofluorescent co-staining of GFP/VEGF-A, Ki67 and MBP showed that the VEGF-A overexpressing Schwann cells could promote the proliferation, migration and differentiation of Schwann cells as the VEGF-A was secreted from the VEGF-A overexpressing Schwann cells. The nerve repair performance of the multifunctional and flexible conduits was examined though rat behavioristics, electrophysiology, nerve innervation to gastrocnemius muscle (GM), toluidine blue (TB) staining, transmission electron microscopy (TEM) and NF200/S100 double staining in the regenerated nerve. The results displayed that the effects on the repair of peripheral nerves in HSPS-SC (VEGF) group was the best among the conduits groups and closed to autografts. HSPS-SC (VEGF) group exhibited notably increased CD31+ endothelial cells and activation of VEGFR2/ERK signaling pathway in the regenerated nerve tissues, which probably contributed to the improved nerve regeneration. Altogether, the comprehensive strategy including VEGF overexpressing Schwann cells-mediated and HSPS conduit-guided peripheral nerve repair provides a new avenue for nerve tissue engineering.

Hydrogels with adhesive properties have the potential for rapid haemostasis and wound healing in uncontrolled non-pressurized surface bleeding. Herein, a typical hydrogen bond-crosslinked hydrogel with the above functions was constructed by directly mixing solutions of humic acid (HA) and polyvinylpyrrolidone (PVP), in which the HA worked as a crosslinking agent to form hydrogen bonds with the PVP. By altering the concentration of HA, a cluster of stable and uniform hydrogels were prepared within 10 s. The dynamic and reversible nature of the hydrogen bonds gave the HA/PVP complex (HPC) hydrogels injectability and good flexibility, as well as a self-healing ability. Moreover, the numerous functional groups in the hydrogels enhanced the cohesion strength and interaction on the interface between the hydrogel and the substrate, endowing them with good adhesion properties. The unique chemical composition and cross-linking mechanism gave the HPC hydrogel good biocompatibility. Taking advantage of all these features, the HPC hydrogels obtained in this work were broadly applied as haemostatic agents and showed a good therapeutic effect. This work might lead to an improvement in the development of multifunctional non-covalent hydrogels for application to biomaterials.

Snap bean is an important crop in the United States. Insecticides are commonly used against pests on snap bean, but many pests have developed resistance to the insecticides and beneficials are threatened by the insecticides. Therefore, host plant resistance is a sustainable alternative. Population dynamics of insect pests and beneficials were assessed on 24 snap bean cultivars every week for six weeks. The lowest number of sweetpotato whitefly (Bemisia tabaci) eggs was observed on cultivar ‘Jade’, and the fewest nymphs were found on cultivars ‘Gold Mine’, ‘Golden Rod’, ‘Long Tendergreen’, and ‘Royal Burgundy’. The numbers of potato leafhopper (Empoasca fabae) and tarnished plant bug (Lygus lineolaris) adults were the lowest on cultivars ‘Greencrop’ and ‘PV-857′. The highest numbers of adults were found in Week 1 (25 days following plant emergence) for B. tabaci and Mexican bean beetle (Epilachna varivestis); Week 3 for cucumber beetle, kudzu bug (Megacopta cribraria), and E. fabae; Weeks 3 and 4 for thrips; Week 4 for L. lineolaris; and Weeks 5 and 6 for bees. Temperature and relative humidity correlated with B. tabaci, E. varivestis, bee, and predator ladybird beetle populations. These results provide valuable information on the integrated pest management of snap beans.

Underground salt caverns are widely used in large-scale energy storage, such as natural gas, compressed air, oil, and hydrogen. In order to quickly build large-scale natural gas reserves, an unusual building method was established. The method involves using the existing salt caverns left over from solution mining of salt to build energy storages. In 2007, it was first applied to Jintan Natural Gas Storage of China. Based on this successful project, several existing salt caverns were screened to build energy storages in China. Engineering experience indicates that the key to successful reusing is how to select the most suitable of the numerous available caverns and confirm it. This paper summarizes and reviews relevant theories and testing methods, including: (1) the primary selection principle for using existing salt caverns to build energy storage, (2) the testing method and evaluation theory of tightness of the existing salt cavern, and (3) the typical project case of using the existing salt caverns to build energy storage in China. From the practical application results, the selection principle proposed in this paper can quickly screen available existing salt caverns with energy storage potential, and the brine injection method can effectively evaluate their tightness. It provides a technical roadmap for the subsequent implementation of existing salt cavern utilization projects on a large scale.

Forsythiae Fructus (Lianqiao in Chinese) is widely used in traditional Chinese medicine. The lipid components in Forsythiae Fructus are the basis of plant growth and active metabolism. Samples were collected at two growth stages for a comprehensive study. Transcriptome and lipidomics were performed by using the RNA-seq and UPLC-Q-TOF-MS techniques separately. For the first time, it was reported that there were 5802 lipid components in Lianqiao comprised of 31.7% glycerolipids, 16.57% phospholipids, 13.18% sphingolipids, and 10.54% fatty acids. Lipid components such as terpenes and flavonoids have pharmacological activity, but their content was low. Among these lipids which were isolated from Forsythiae Fructus, 139 showed significant differences from the May and July harvest periods. The lipids of natural products are mainly concentrated in pregnenolones and polyvinyl lipids. RNA-Seq analysis revealed 92,294 unigenes, and 1533 of these were differentially expressed. There were 551 differential genes enriched in 119 KEGG pathways. The de novo synthesis pathways of terpenoids and flavonoids were explored. Combined with the results of lipidomics and transcriptomics, it is hypothesized that in the synthesis of abscisic acid, a terpenoid, may be under the dynamic regulation of genes EC: 1.1.1.288, EC: 1.14.14.137 and EC: 1.13.11.51 in balanced state. In the synthesis of gibberellin, GA20-oxidase (GA20ox, EC: 1.14.11.12), and GA3-oxidase (GA3ox, EC: 1.14.11.15) catalyze the production of active GAs, and EC: 1.14.11.13 is the metabolic enzymes of active GAs. In the synthesis of flavonoids, MF (multifunctional), PAL (phenylalanine ammonia-lyase), CHS (chalcone synthase), ANS (anthocyanidin synthase), FLS (flavonol synthase) are all key enzymes. The results of the present study provide valuable reference information for further research on the metabolic pathways of the secondary metabolites of Forsythia suspensa.

Ground-penetrating radar (GPR) is widely used in the detection and positioning of underground facilities. Through the inversion analysis of echo signals of GPR, information such as pipe material, burial depth and location of pipelines can be obtained. Unfortunately, underground pipelines are cylindrical, the ladder approximation method used in traditional forward models produces certain errors. In this study, an accurate and efficient numerical model of GPR forward model in underground pipelines is established using symplectic Euler algorithm, graphics processing unit (GPU) acceleration technology and surface conformal technology. With a Ricker wavelet pulse as the GPR source, the convolution perfectly matched layer (CPML) is incorporated in the symplectic Euler algorithm and shown to be effective to truncate the symplectic Euler computational domain. Through the simulation study of different underground pipeline models, GPR image characteristics of the metal pipeline, plastic pipeline and concrete pipeline filled with air and water are obtained. According to the numerical simulation results, parallel conformal symplectic Euler algorithm effectively reduces the false diffracted waves caused by ladder approximation and improves the computational efficiency of the model in metallic and non-metallic media.

Abstract Background Hepatitis C virus (HCV) infection is commonly associated with a disturbance of glucose metabolism. However, there have been conflicting reports on whether the clearance of the HCV may be followed by changes of serum blood glucose and insulin resistance. The aim of the present study was to evaluate the impact of HCV and antiviral treatment on serum glucose levels and other glucose metabolism parameters. Methods A retrospective analysis of 306 HCV-infected patients was performed. Fasting serum blood glucose (FBG) levels in these patients were compared with that of 325 healthy individuals. Serum parameters of glucose metabolism were measured in 183 patients with chronic hepatitis C at baseline, at the end of interferon α-2b plus ribavirin treatment, and at 24 weeks post-treatment. Chemical investigation of the psychrophilic fungus Pseudogymnoascus sp. HDN17-933 derived from Antarctica led to the discovery of six new tetrapeptides psegymamides A–F (1–6), whose planar structures were elucidated by extensive NMR and MS spectrometric analyses. Structurally, psegymamides D–F (4–6) possess unique backbones bearing a tetrahydropyridoindoles unit, which make them the first examples discovered in naturally occurring peptides. The absolute configurations of structures were unambiguously determined using solid-phase total synthesis assisted by Marfey’s method, and all compounds were evaluated for their inhibition of human (h) nicotinic acetylcholine receptor subtypes. Compound 2 showed significant inhibitory activity. A preliminary structure–activity relationship investigation revealed that the tryptophan residue and the C-terminal with methoxy group were important to the inhibitory activity. Further, the high binding affinity of compound 2 to hα4β2 was explained by molecular docking studies.

Ground Penetrating Radar (GPR) is a shallow geophysical method for detecting and locating subsurface targets. The GPR image echo characteristics of complex underground structures can be obtained by carrying out GPR forward modeling research. The traditional finite-difference time-domain (FDTD) method has low efficiency and accuracy. The alternating direction implicit FDTD (ADI-FDTD) algorithm surmounts the stability limitations of the traditional FDTD method, making it possible to select a larger time step for higher computational efficiency. For circular underground structures, a pseudowave produced by the ladder approximation method can be corrected using the surface conformal technique. This paper proposes a high-efficiency and high-accuracy GPR forward modeling method that combines the ADI-FDTD algorithm and surface conformal technology. The performance of the conformal ADI-FDTD algorithm is verified by a simple two-layer model. Based on the proposed algorithm, the GPR image features of three complex underground structure models are obtained. Finally, a field experiment is used to support the accuracy and usefulness of the conformal ADI-FDTD algorithm. The numerical simulation results and experimental results show that the conformal ADI-FDTD algorithm reduces the pseudodiffraction wave caused by the ladder approximation method and can significantly improve the computing efficiency for complex underground structure models.

A functional Chinese fermented glutinous rice has been developed with the supplementation of Fu brick tea (CRW-FBT). In this study, we aimed to evaluate its effect on the growth of potential probiotic strains in the Lactobacillus, Streptococcus, and Weissella genus, compared with traditional Chinese fermented glutinous rice (CRW). The growth profiles of lactic acid bacteria were analyzed based on fermentations in vitro, and the optical densities were recorded at 600 nm during the whole fermentation. Growth curve, maximum OD600 nm, and growth rate were measured and compared among samples with different ratios of CRW-FBT and CRW addition. Through the multiple analysis of growth parameters, we found that all the tested strains obtained better growth results when CRW-FBT was supplemented to the media, compared with the CRW and basic media. The bacterial growth was promoted by exhibiting the shortened lag time, prolonged logarithmic phase and stationary phase, and increased growth rate and cell density, as well as the better performance after 24 h and 48 h fermentation. Besides, short-chain fatty acids and organic acids in CRW-FBT were founded. Our work demonstrated the positive effect of Fu brick tea supplemented in the CRW and illustrated its beneficial role in the food fermentation industry for the purpose of microorganism enrichment and the improvement of microbial metabolism.

Peripheral nerve injury is a common clinical neurological disease. In our previous study, highly oriented poly (L-lactic acid) (PLLA)/soy protein isolate (SPI) nanofiber nerve conduits were constructed and exhibited a certain repair capacity for peripheral nerve injury. In order to further improve their nerve repairing efficiency, the bone mesenchymal stem cells (BMSCs) overexpressing brain derived neurotrophic factor (BDNF) and glial cell line-derived neurotrophic factor (GDNF) were introduced into the conduits as seed cells and then were used to repair the 10-mm sciatic nerve defects in rats. The nerve repair efficiency of the functional nerve conduits was evaluated by gait experiment, electrophysiological test, and a series of assays such as hemotoxylin-eosin (HE) staining, immunofluorescence staining, toluidine blue (TB) staining, transmission electron microscopy (TEM) observation of regenerated nerve and Masson’s trichrome staining of gastrocnemius muscle. The results showed that the conduits containing BMSCs overexpressing BDNF and GDNF double-factors group had better nerve repairing efficiency than blank BMSCs and single BDNF or GDNF factor groups, and superior to autografts group in some aspects. These data demonstrated that BDNF and GDNF produced by BMSCs could synergistically promote peripheral nerve repair. This study shed a new light on the conduits and stem cells-based peripheral nerve repair.

The venoms of toxic animals are chemical pools composed of various proteins, peptides, and small organic molecules used for predation and defense, in which the peptidic toxins have been intensively pursued mining modulators targeting disease-related ion channels and receptors as valuable drug pioneers. In the present study, we uncovered the molecular diversity of peptide toxins in the venom of the spider Heteropoda pingtungensis (H. pingtungensis) by using a combinatory strategy of venom gland cDNA library and transcriptome sequencing (RNA-seq). An amount of 991 high-quality expressed sequence tags (ESTs) were identified from 1138 generated sequences, which fall into three categories, such as the toxin-like ESTs (531, 53.58%), the cellular component ESTs (255, 25.73%), and the no-match ESTs (205, 20.69%), as determined by gene function annotations. Of them, 190 non-redundant toxin-like peptides were identified and can be artificially grouped into 13 families based on their sequence homology and cysteine frameworks (families A–M). The predicted mature toxins contain 2–10 cysteines, which are predicted to form intramolecular disulfide bonds to stabilize their three-dimensional structures. Bioinformatics analysis showed that toxins from H. pingtungensis venom have high sequences variability and the biological targets for most toxins are unpredictable due to lack of homology to toxins with known functions in the database. Furthermore, RP-HPLC and MALDI-TOF analyses have identified a total of 110 different peptides physically existing in the H. pingtungensis venom, and many RP-HPLC fractions showed potent inhibitory activity on the heterologously expressed NaV1.7 channel. Most importantly, two novel NaV1.7 peptide antagonists, µ-Sparatoxin-Hp1 and µ-Sparatoxin-Hp2, were characterized. In conclusion, the present study has added many new members to the spider toxin superfamily and built the foundation for identifying novel modulators targeting ion channels in the H. pingtungensis venom.

Bemisia tabaci Gennadius (Hemiptera: Aleyrodidae) is among the most economically important insect pests of various vegetable crops in the Southern United States. This insect is considered a complex of at least 40 morphologically indistinguishable cryptic species. Bemisia tabaci Middle East-Asia Minor 1 (MEAM1) was initially introduced in the United States around 1985 and has since rapidly spread across the Southern United States to Texas, Arizona, and California, where extreme field outbreaks have occurred on vegetable and other crops. This pest creates extensive plant damage through direct feeding on vegetables, secreting honeydew, causing plant physiological disorders, and vectoring plant viruses. The direct and indirect plant damage in vegetable crops has resulted in enormous economic losses in the Southern United States, especially in Florida, Georgia, and Texas. Effective management of B. tabaci on vegetables relies mainly on the utilization of chemical insecticides, particularly neonicotinoids. However, B. tabaci has developed considerable resistance to most insecticides. Therefore, alternative integrated pest management (IPM) strategies are required, such as cultural control by manipulation of production practices, resistant vegetable varieties, and biological control using a suite of natural enemies for the management of the pest.

Inflammatory bowel disease (IBD) has been gradually considered a public health challenge worldwide. Sulfated polysaccharides, extracted from seaweed, have been shown to have an anti-inflammatory effect on the disease. In this study, LMW-ulvan, a unique sulfate Ulva polysaccharide with low molecular weight, was prepared using the enzymatic method. The structural characterization of LMW-ulvan and its protective effect on colitis induced by dextran sulfate sodium (DSS) were studied. The results showed that LMW-ulvan with molecular weight of 2.56 kDa consists of 57.23% rhamnose (Rha), 28.76% xylose (Xyl), 7.42% glucuronic acid (GlcA), and 1.77% glucose (Glc). Its backbone contains (1→3,4)-linked Rha, (1→4)-linked Xyl, and (1→4)-linked GlcA with small amounts of (1→4)-linked Rha residues; sulfate substitution was at C-3 of Rha. LMW-ulvan was found to reduce DSS-induced disease activity index, colon shortening, and colonic tissue damage, which were associated with decreased oxidative stresses and inflammation, thus improving the expression of tight junction proteins. These results indicate that LMW-ulvan is able to improve colitis and may be a promising application for IBD.

The pantograph catenary system plays an important role in the power performance of electric mining vehicles. A pantograph catenary system combining both a pantograph and a catenary is one of the most promising solutions. As a case study, this paper discusses the dynamic performance and the stable current collection of a pantograph catenary system for a 14 ton underground overhead wire electrical actuated load, haul, dump machine (LHD). First, based on the optimized finite element simulation process, finite element models of the pantograph system and the catenary system are established. Second, the motion equation of the catenary is improved, and the finite element model of the pantograph catenary system is established. Finally, a dynamic simulation experiment is performed to determine the dynamic performance of the pantograph catenary system. The results show that when the radius of the contact wire is set to 0.00564 m and the tension of contact wire is set to 30 KN, the current collection indexes of the pantograph catenary system meet the requirements of stable current collection and are superior to the simulation results of related references. Therefore, the validity of the finite element model is verified; thus, the pantograph catenary system can stably charge and supply energy for the trolley wire overhead electrically actuated LHD and ensure sufficient power.

Xerophytes play an active role in preventing soil denudation and desertification in arid and semi-arid areas. Peganum harmala L. (Zygophyllaceae family), a seasonally growing, poisonous and drought-tolerant plant, is widely distributed in the Xinjiang Uygur Autonomous Region and used as a traditional herbal medicine as well as, in winter, a fodder source. Previous research has focused on the pharmacological activity of isolated compounds and stress responses to growth environments. However, the metabolic profile of P. harmala and variations in its metabolites, including medicinally active and stress resistance components, have not been illustrated during different growth stages. Here, we collected plant samples in May, August, October and December. We determined the metabolic composition of methanol extracts using NMR spectroscopy, and comparisons of four growth stages were accomplished by applying statistical analysis. The results showed that vasicine, choline and sucrose were significantly elevated in samples harvested in May. Significantly higher amounts of betaine, lysine, 4-hydroxyisoleucine and proline were found in samples collected in August than in samples collected in other months, and the concentrations of phosphorylcholine, glucose, acetic acid and vasicinone were highest in December. The relationships between differential biomarkers and plant physiological states affected by diverse growth environmental factors were discussed. Our result deepened the understanding of metabolic mechanisms in plant development and confirmed the advantage of using NMR-based metabolomic treatments in quality evaluation when P. harmala is used for different purposes.

Objectives. We aim to identify the key biomarker of acute rejection (AR) after kidney transplantation via bioinformatics methods. Methods. The gene expression data GSE75693 of 30 samples with stable kidney transplantation recipients and 15 AR samples were downloaded and analyzed by the limma package to identify differentially expressed genes (DEGs). Then, Gene Ontology (GO) functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis were done to explore the biological functions and potential important pathways of DEGs. Finally, protein-protein interactions (PPIs) and literature mining were applied to construct the cocitation network and to select the hub protein. Results. A total of 437 upregulated genes and 353 downregulated genes were selected according to P1.0. DEGs of AR are mainly located on membranes and impact the activation of receptors in immune responses. In the PPI network, Src kinase, lymphocyte kinase (LCK), CD3G, B2M, interferon-γ, CD3D, tumor necrosis factor, VAV1, and CD3E in the T cell receptor signaling pathway were selected as important factors, and LCK was identified as the hub protein. Conclusion. LCK, via acting on T-cell receptor, might be a potential therapeutic target for AR after kidney transplantation.

A nuclear magnetic resonance- (NMR-) based metabolomics method was used to identify differential metabolites of methanol extracts obtained from six parts of Peganum harmala L. (P. harmala), namely, the root, stem, leaf, flower, testa, and seed. Two multivariate statistical analysis methods, principal component analysis (PCA) and partial least squares-discriminant analysis (PLS-DA), were combined to clearly distinguish among the P. harmala samples from the six different parts. Eleven differential components were screened by the PLS-DA loading plot, and the relative contents were calculated by univariate analysis of variance. Chemometric results showed significant differences in the metabolites of the different parts of P. harmala. The seeds contained large amounts of harmaline, harmine, and vasicine compared to other organs. The acetic acid, proline, lysine, and sucrose contents of the roots were significantly higher than those of the other parts. In the testa, the vasicine, asparagine, choline, and 4-hydroxyisoleucine contents were clearly dominant. The obtained data revealed the distribution characteristics of the metabolomes of the different P. harmala parts and provided fundamental knowledge for the rational development of its medicinal parts.

Background and Aims. There is scarcity of data in literature regarding the treatment response to sofosbuvir- (SOF-) based therapies in Chinese patients with chronic Hepatitis C Virus (HCV) infection. The aim of this study was to evaluate the efficacy and safety of SOF-based regimens for chronic hepatitis C (CHC) patients without cirrhosis in a real-world setting in mainland China. Methods. A total of 226 patients receiving SOF plus daclatasvir (DCV), ledipasvir (LDV), or velpatasvir (VEL) were enrolled from December 2014 to June 2017. The primary observation point was the percentage of patients with a sustained virologic response (SVR) at posttreatment week 12 (SVR12), and all adverse events were monitored during treatment and follow-up period. Results. The overall SVR12 rate was 96% (216/226), and individual SVR12 ranged from 93% to 100% in different treatment groups. No significant differences of efficacy were detected between genotypes 1b and 6a (98% for GT 1b versus 100% for GT 6a, P=0.322). Comparing the high success rates in GT 1b and 6a patients, SVR12 was relatively low in GT 3a and 3b patients. Soft corals are common marine organisms that inhabit tropical and subtropical oceans. They are shown to be rich source of secondary metabolites with biological activities. In this work, soft corals from two geographical locations were investigated using 1H-NMR spectroscopy coupled with multivariate statistical analysis at the metabolic level. A partial least-squares discriminant analysis showed clear separation among extracts of soft corals grown in Sanya Bay and Weizhou Island. The specific markers that contributed to discrimination between soft corals in two origins belonged to terpenes, sterols and N-containing compounds. The satisfied precision of classification obtained indicates this approach using combined 1H-NMR and chemometrics is effective to discriminate soft corals collected in different geographical locations. The results revealed that metabolites of soft corals evidently depended on living environmental condition, which would provide valuable information for further relevant coastal marine environment evaluation.

A linear theoretical model is established for the dynamics of a hanging vertical cantilevered pipe which is subjected concurrently to internal and reverse external axial flows. Such pipe systems may have instability by flutter (amplified oscillations) or static divergence (buckling). The pipe system under consideration is a slender flexible cantilevered pipe hanging concentrically within an inflexible external pipe of larger diameter. From the clamped end to the free end, fluid is injected through the annular passage between the external pipe and the cantilevered pipe. When exiting the annular passage, the fluid discharges in the counter direction along the cantilevered pipe. The inflexible external pipe has a variable length and it can cover a portion of the length of the cantilevered pipe. This pipe system has been applied in the solution mining and in the salt cavern underground energy storage industry. The planar motion equation of the system is solved by means of a Galerkin method, and Euler–Bernoulli beam eigenfunctions are used as comparison functions. Calculations are conducted to quantify the effects of different confinement conditions (i.e., the radial confinement degree of the annular passage and the confined-flow length) on the cantilevered pipe stability, for a long leaching-tubing-like system. For a long system, an increase in the radial confinement degree of the annular passage and the confined-flow length gives rise to a series of flutter and divergence. Additionally, the effect of the cantilevered pipe length is studied. Increasing the cantilevered pipe length results in an increase of the critical flow velocity while a decrease of the associated critical frequency. For a long enough system, the critical frequency almost disappears.

A number of large underground oil storage spaces will be constructed in deep salt mines in China in the coming years. According to the general geological survey, the first salt cavern oil storage base of China is planned to be built in Jintan salt mine. In this research, the geological feasibility of the salt mine for oil storage is identified in detail as follows. (1) The characteristics of regional structure, strata sediment, and impermeable layer distribution of Jintan salt mine were evaluated and analyzed. (2) The tightness of cap rock was evaluated in reviews of macroscopic geology and microscopic measuring. (3) According to the geological characteristics of Jintan salt mine, the specific targeted formation for building underground oil storage was chosen, and the sealing of nonsalt interlayers was evaluated. (4) Based on the sonar measuring results of the salt caverns, the characteristics of solution mining salt caverns were analyzed. In addition, the preferred way of underground oil storage construction was determined. (5) Finally, the results of closed well observation in solution mining salt caverns were assessed. The research results indicated that Jintan salt mine has the basic geological conditions for building large-scale underground oil storage.
