Abstract Background Endometrial cancer is one of the major gynecological cancers, with increasing incidence and mortality in the past decades. Emerging preclinical and clinical data have indicated its close association with obesity and dyslipidemia. Metabolism reprogramming has been considered as the hallmark of cancer, to satisfy the extensive need of nutrients and energy for survival and growth. Particularly, lipid metabolism reprogramming has aroused the researchers’ interest in the field of cancer, including tumorigenesis, invasiveness, metastasis, therapeutic resistance and immunity modulation, etc. But the roles of lipid metabolism reprogramming in endometrial cancer have not been fully understood. This review has summarized how lipid metabolism reprogramming induces oncogenesis and progression of endometrial cancer, including the biological functions of aberrant lipid metabolism pathway and altered transcription regulation of lipid metabolism pathway. Besides, we proposed novel therapeutic strategies of targeting lipid metabolism pathway and concentrated on its potential of sensitizing immunotherapy and hormonal therapy, to further optimize the existing treatment modalities of patients with advanced/metastatic endometrial cancer. Moreover, we expect that targeting lipid metabolism plus hormone therapy may block the endometrial malignant transformation and enrich the preventative approaches of endometrial cancer. Conclusion Lipid metabolism reprogramming plays an important role in tumor initiation and cancer progression of endometrial cancer. Targeting the core enzymes and transcriptional factors of lipid metabolism pathway alone or in combination with immunotherapy/hormone treatment is expected to decrease the tumor burden and provide promising treatment opportunity for patients with advanced/metastatic endometrial cancer.

Introduction: Proton beam therapy (PBT) may reduce the number of adverse events in treatment of patients with pediatric cancer. However, it is difficult to evaluate whether the actual therapeutic effect is truly equivalent to that of photon radiotherapy. To compare photon radiotherapy and PBT, a meta-analysis and systematic review were performed. Methods: The meta-analysis used papers from 1990 to 2023 in which postoperative local photon radiotherapy or PBT was performed for pediatric intracranial ependymomas. Fifteen articles (5 PBT, 9 photon radiotherapy, one both) were selected based on administration of radiotherapy as local irradiation. Results: Among the 15 chosen articles, the 1- to 5-year overall survival (OS) rates (photon radiotherapy vs. PBT) were 95.4 % (95 % confidence interval (CI) 92.8–97.1 %) vs. 97.2 % (95.7–98.2 %); 88.3 % (85.0–90.9 %) vs. 93.5 % (91.4–95.1 %); 81.2 % (76.9–84.8 %) vs. 91.1 % (88.4–93.2 %); 76.9 % (71.2–81.6 %) vs. 86.1 % (81.9–89.4 %); and 73.8 % (68.3–78.5 %) vs. 84.7 % (79.9–88.5 %), respectively. The 1- to 5-year local control (LC) rates (photon radiotherapy vs. PBT) were 90.9 % (95 % CI 83.9–94.9 %) vs. 91.0 % (88.7–92.9 %); 81.5 % (68.9–89.4 %) vs. 85.7 % (82.0–88.6 %); 77.3 % (62.8–86.8 %) vs. 82.6 % (79.1–85.5 %); 74.6 % (57.7–85.6 %) vs. 78.3 % (71.6–83.5 %); and 72.6 % (51.4–85.8 %) vs. 79.0 % (73.4–83.5 %), respectively. The meta-regression analysis identified relationships of modality (photon radiotherapy vs. PBT), age at irradiation, pathology (Grade 2 vs. Grade 3), and tumor removal (complete resection vs. none) with significantly better 3-year OS after PBT and better 1- to 5-year LC at a younger age. Conclusion: In postoperative local irradiation of ependymomas in children, proton beam therapy had outcomes comparable to those of photon radiotherapy.

Loquat leaves are widely used in traditional Chinese medicine and food processing as raw materials. To provide a theoretical basis for suitable harvesting periods based on the utilization of distinct components, the change patterns of major functional components in the annual growth of loquat leaves were investigated. In this study, the summer loquat leaves were sampled regularly during their annual growth, and the contents of seven main functional components and 13 monomer components were determined. The results showed that there was a regular change pattern for major functional components during loquat annual growth from Jul 2020 to Jun 2021; however, the change trend was different among different components. Among them, the contents of total flavonoids, total phenols, total triterpenoid acids, vitamin C, asiatic acid, quillaic acid, corosolic acid, and chlorogenic acid presented an overall upward trend. The content of maslinic acid showed a downward trend, whereas the content of neochlorogenic acid did not change much throughout the whole growth period. The content changes in the loquat leaves were affected by the light durations and temperature changes in different seasons, with mutual transit during the flowering and fruit growth processes. Sampling in May to June was found to be a most optimized option when applied in Chinese medicine processing; however, the degree of fibrosis is another indicator when it is used as food raw material. This study revealed the change pattern of major functional components of loquat leaves during the annual growth period and provides a theoretical basis for the reasonable harvesting period of loquat leaves as raw materials for processing different types of products.

BackgroundRecent epidemiological studies have indicated a correlation between platelet indices and pulmonary arterial hypertension (PAH), yet the causality between them remains unclear. To explore the causal relationship between four platelet indices and PAH, with the aim of providing a theoretical basis for clinical prevention and treatment.MethodsSingle-nucleotide polymorphisms (SNPs) associated with platelet-related traits were selected as exposure factors from published genome-wide association studies (GWAS), including: platelet count (PLT), plateletcrit (PCT), mean platelet volume (MPV), and platelet distribution width (PDW). Summary-level data for PAH were obtained from the FinnGen study (248 cases and 289,117 controls). Two-sample and multivariable Mendelian randomization (MR) analyses were conducted to assess the causal relationship between exposure factors and the risk of outcomes. The inverse variance weighted (IVW) method was utilized as the primary MR analysis approach, supplemented by weighted median, mode-based estimation, MR-Egger regression, and the MR Pleiotropy Residual Sum and Outlier (MR-PRESSO) test to detect and adjust for pleiotropy, ensuring the reliability of the results through sensitivity analysis.Results(1) The IVW results from the two-sample MR analysis showed a positive causal association between PLT and the risk of developing PAH [(OR = 1.649, 95%CI: 1.206–2.256, P = 0.0017)], with the sensitivity analysis confirming the robustness of the causal relationship. The MR-Egger intercept analysis did not detect potential pleiotropy (P = 0.879). (2) The MVMR results showed no statistically significant causal relationship between these four markers and the risk of developing PAH. After adjusting for collinearity, a direct positive causal association was observed between PLT and the risk of developing PAH (OR = 1.525, 95%CI: 1.063–2.189, P = 0.022).ConclusionThe positive correlation between PLT and the risk of PAH suggests that correcting elevated platelet levels may reduce the risk of developing PAH.

Background: Numerous studies have shown a strong correlation between disulfidptosis and various cancers. However, the expression and function of RPN1, a crucial gene in disulfidptosis, remain unclear in the context of cancer. Methods: Gene expression and clinical information on lung adenocarcinoma were obtained from The Cancer Genome Atlas (TCGA) and Genotype-Tissue Expression (GTEx) databases. RPN1 expression was analyzed using the Timer2.0 and the Human Protein Atlas (HPA) databases. Prognostic significance was assessed using Cox regression analysis and Kaplan–Meier curves. Genetic mutations and methylation levels were examined using the cBioPortal and UALCAN platforms, respectively. The relationship between RPN1 and tumor mutation burden (TMB) and microsatellite instability (MSI) across different cancer types was analyzed using the Spearman correlation coefficient. The relationship between RPN1 and immune cell infiltration was analyzed using the Timer2.0 database, whereas variations in drug sensitivity were explored using the CellMiner database. Receiver operating characteristic curves validated RPN1’s diagnostic potential in glioma, and its correlation with immune checkpoint inhibitors (ICIs) was assessed using Spearman’s correlation coefficient. Single-sample gene set enrichment analysis elucidated a link between RPN1 and immune cells and pathways. In addition, a nomogram based on RPN1 was developed to predict patient prognosis. The functional impact of RPN1 on glioma cells was confirmed using scratch and Transwell assays. Result: RPN1 was aberrantly expressed in various cancers and affected patient prognosis. The main mutation type of RPN1 in the cancer was amplified. RPN1 exhibited a positive correlation with myeloid-derived suppressor cells, neutrophils, and macrophages, and a negative correlation with CD8+ T cells and hematopoietic stem cells. RPN1 expression was associated with TMB and MSI in various cancers. The expression of RPN1 affected drug sensitivity in cancer cells. RPN1 was positively correlated with multiple ICIs in gliomas. RPN1 also affected immune cell infiltration into the tumor microenvironment. RPN1 was an independent prognostic factor for gliomas, and the nomogram demonstrated excellent predictive performance. Interference with RPN1 expression reduces the migratory and invasive ability of glioma cells. Conclusion: RPN1 exerts multifaceted effects on different stages of cancer, including immune infiltration, prognosis, and treatment outcomes. RPN1 expression affects the prognosis and immune microenvironment infiltration in patients with glioma, making RPN1 a potential target for the treatment of glioma.

Autumn rain in western China is a unique and complex precipitation phenomenon that takes place during the autumn season in the western region of China， which is closely related to various aspects of economy， society and human life.This natural phenomenon has a profound impact on agriculture， transportation and other aspects of social production and life， sometimes even causes the phenomenon of autumn flood， which deserves full attention.Based on the basic understanding of the phenomenon in the 20th century， this paper compares and reviews the main research achievements of autumn rain in western China over the past 20 years from multiple perspectives.First of all， the spatial distribution range and temporal extent and precipitation intensity of autumn rain in West China are thoroughly explored， quantitative criteria and indexes such as ARI （Autumn Rain Index）， DARI （Daily Autumn Rain Index） and MARI （Modified Autumn Rain Index）are also proposed.In addition to this， the significant interannual differences， the "positive - negative - positive" interdecadal variations and the 4~8 years cycle of autumn rain in western China fully demonstrate its complexity and inhomogeneity.Secondly， the different but important influences of atmospheric circulation systems at low， middle， and high latitudes， tropical sea temperature forcing including Pacific Ocean， Indian Ocean， and Atlantic Ocean as well as the Qinghai-Xizang （Tibet） Plateau effect and other aspects on the autumn rain in western China are pointed out.These different factors all play significant roles in the evolution of the phenomenon， but the mechanisms are different.This leads to the recognition of the significance of the multi-system and multi-factor synergistic effect on the anomalous evolution of autumn rain in western China， which enriches the understanding of the autumn rain and its impact.Finally， according to the current cognition of autumn rain in western China， the main problems that exist in the combination of different monsoon systems like Indian monsoon， East Asian monsoon and Plateau monsoon， cold and warm air currents， evolution causes， weather processes， night rain characteristics， regional responses to the global warming and other aspects are pointed out.These problems highlight the challenges that researchers face in studying this complex phenomenon.However， despite the challenges， there are several promising areas for future research.The main development trend in the future about the research of autumn rain in western China is the comprehensive and elaborate analysis of the multi-scale synergy of autumn rain in western China and its physical mechanism under the complex and important topography of Qinghai-Xizang （Tibet） Plateau.

Pelvic organ prolapse (POP) is a benign disease characterized by the descent of pelvic organs due to weakened pelvic floor muscles and fascial tissues. Primarily affecting elderly women, POP can lead to various urinary and gastrointestinal tract symptoms, significantly impacting their quality of life. The pathogenesis of POP predominantly involves nerve–muscle damage and disorders in the extracellular matrix metabolism within the pelvic floor. Recent studies have indicated that genetic factors may play a crucial role in this condition. Focusing on linkage analyses, single-nucleotide polymorphisms, genome-wide association studies, and whole exome sequencing studies, this review consolidates current research on the genetic predisposition to POP. Advances in epigenetics are also summarized and highlighted, aiming to provide theoretical recommendations for risk assessments, diagnoses, and the personalized treatment for patients with POP.

Abstract Purpose To discuss the optimal treatment modality for inoperable locally advanced Non-Small Cell Lung Cancer patients with poor physical status, impaired cardio-pulmonary function, and negative driver genes, and provide clinical evidence. Materials and methods Retrospective analysis of 62 cases of locally advanced non-small cell lung cancer patients with negative driver genes treated at Tsukuba University Hospital(Japan) and Qingdao University Affiliated Hospital(China).The former received proton therapy with concurrent chemotherapy, referred to as the proton group, with 25 cases included; while the latter underwent X-ray therapy with concurrent chemoradiotherapy followed by 1 year of sequential immunomodulatory maintenance therapy, referred to as the X-ray group, with 37 cases included.The treatment response and adverse reactions were assessed using RECIST v1.1 criteria and CTCAE v3.0, and radiotherapy planning and evaluation of organs at risk were performed using the CB-CHOP method.All data were subjected to statistical analysis using GraphPad Prism v9.0, with a T-test using P 0.05).(3)Treatment-related toxicities: The incidence of grade 3 or higher adverse events in the proton group and X-ray group was 28.6% and 4.2%, respectively, with a statistically significant difference (P 0.05), and the DCR was 92.2% and 86.4%, respectively (P > 0.05), indicating no significant difference in short-term efficacy between the two treatment modalities.(5)Survival status: The PFS in the proton group and X-ray group was 31.6 ± 3.5 months (95% CI: 24.7 ~ 38.5) and 24.9 ± 1.55 months (95% CI: 21.9 ~ 27.9), respectively (P > 0.05), while the OS was 51.6 ± 4.62 months (95% CI: 42.5 ~ 60.7) and 33.1 ± 1.99 months (95% CI: 29.2 ~ 37.1), respectively (P 0.05).The OS rates for the first to third years in both groups were as follows: 100%, 88.2%, 76.4% for the proton group vs. 100%, 91.4%, 46.3% for the X-ray group. Abstract Recurrence is the main cause of death in patients with endometrial cancer (EC). This study aimed to construct and validate a nomogram to predict the recurrence-free survival of patients with EC. This was a multicenter retrospective study. A total of 812 patients from Wuhan Tongji Hospital were divided into training and validation cohorts, and 347 and 580 patients from People’s Hospital of Peking University and Qilu Hospital of Shandong, respectively, were used for validation. Univariate and multivariate Cox regression analyses were used to construct a nomogram for predicting recurrence-free survival of EC. Calibration curves, receiver operating characteristic (ROC) curves, and consistency indexes (C-indexes) were used to estimate the performance of the model. IntroductionChondrosarcoma is a rare malignant bone tumor. Particle beam therapy (PT) can concentrate doses to targets while reducing adverse events. A meta-analysis based on a literature review was performed to examine the efficacy of PT and photon radiotherapy for skull base chondrosarcoma.MethodsThe meta-analysis was conducted using 21 articles published from 1990 to 2022.ResultsAfter PT, the 3- and 5-year overall survival (OS) rates were 94.1% (95% confidence interval [CI]: 91.0-96.2%) and 93.9% (95% CI: 90.6-96.1%), respectively, and the 3- and 5-year local control rates were 95.4% (95% CI: 92.0-97.4%) and 90.1% (95% CI: 76.8-96.0%), respectively. Meta-regression analysis revealed a significant association of PT with a superior 5-year OS rate compared to three-dimensional conformal radiotherapy (p < 0.001). In the studies used in the meta-analysis, the major adverse event of grade 2 or higher was temporal lobe necrosis (incidence 1-18%, median 7%).ConclusionPT for skull base chondrosarcoma had a good outcome and may be a valuable option among radiotherapy modalities. However, high-dose postoperative irradiation of skull base chondrosarcoma can cause adverse events such as temporal lobe necrosis.

Over 70% of leiomyoma (LM) harbor MED12 mutations, primarily in exon 2 at c.130-131 (GG). Myometrial cells are the cell origin of leiomyoma, but the MED12 mutation status in non-neoplastic myometrial cells is unknown. In this study, we investigated the mutation burden of MED12 in myometrium. As traditional Sanger or even NGS sequencing may not be able to detect MED12 mutations that are lower than 0.1% in the testing sample, we used duplex deep sequencing analysis (DDS) to overcome this limitation. Tumor-free myometria (confirmed by pathology evaluation) were dissected, and genomic DNA from MED12 exon 2 (test) and TP53 exon 5 (control) were captured by customer-designed probe sets, followed by DDS. Notably, DDS demonstrated that myometrial cells harbored a high frequency of mutations in MED12 exon 2 and predominantly in code c.130-131. In contrast, the baseline mutations in other coding sequences of MED12 exon 2 as well as in the TP53 mutation hotspot, c.477-488 were comparably low in myometrial cells. This is the first report demonstrating a non-random accumulation of MED12 mutations at c.130-131 sites in non-neoplastic myometrial cells which provide molecular evidence of early somatic mutation events in myometrial cells. This early mutation may contribute to the cell origin for uterine LM development in women of reproductive age.

Polymer fibers are attracting increasing attention as a type of fundamental material for a wide range of products. However, to incorporate novel functionality, a crucial challenge is to simultaneously manipulate their structuring across multiple length scales. In this research, a facile and universal approach is proposed by directly drawing a pre-gel feedstock embedding a cellulose cholesteric liquid crystal (CLC). An in situ photo-polymerization process is applied, which not only allows for the continuous drawing of the filaments without breakup but also makes the final CLC fibers a colored appearance. More importantly, the multiscale properties of the fibers, such as their diameter, morphology, and the internal liquid crystalline ordering of the molecules (and thus structural color), can be manipulated by several controlling parameters. Combining this cross-scale tunability with a smart functional hydrogel system results in the formation of fibers with structural coloration, self-healing, electrical conduction, and thermal-sensing abilities. We believe that this platform can be extended to other hydrogel systems and will help unlock a wide variety of real-life applications.

The purpose of this study was to analyze renal late effects after proton beam therapy (PBT) for pediatric malignant tumors. A retrospective study was performed in 11 patients under 8 years of age who received PBT between 2013 and 2018. The kidney was exposed in irradiation of the primary lesion in all cases. Kidney volume and contour were measured on CT or MRI. Dose volume was calculated with a treatment-planning system. The median follow-up was 24 months (range, 11–57 months). In irradiated kidneys and control contralateral kidneys, the median volume changes were −5.63 (−20.54 to 7.20) and 5.23 (−2.01 to 16.73) mL/year; and the median % volume changes at 1 year were −8.55% (−47.52 to 15.51%) and 9.53% (−2.13 to 38.78%), respectively. The median relative volume change for irradiated kidneys at 1 year was −16.42% (−52.21 to −4.53%) relative to control kidneys. Kidneys irradiated with doses of 10, 20, 30, 40, and 50 GyE had volume reductions of 0.16%, 0.90%, 1.24%, 2.34%, and 8.2% per irradiated volume, respectively. The larger the irradiated volume, the greater the kidney volume was lost. Volume reduction was much greater in patients aged 4–7 years than in those aged 2–3 years. The results suggest that kidneys exposed to PBT in treatment of pediatric malignant tumor show continuous atrophy in follow-up. The degree of atrophy is increased with a higher radiation dose, greater irradiated volume, and older age. However, with growth and maturation, the contralateral kidney becomes progressively larger and is less affected by radiation.

Abstract Background Epigenetic modifications established in mammalian gametes are largely reprogrammed during early development, however, are partly inherited by the embryo to support its development. In this study, we examine CpG island (CGI) sequences to predict whether a mouse blastocyst CGI inherits oocyte-derived DNA methylation from the maternal genome. Recurrent neural networks (RNNs), including that based on gated recurrent units (GRUs), have recently been employed for variable-length inputs in classification and regression analyses. One advantage of this strategy is the ability of RNNs to automatically learn latent features embedded in inputs by learning their model parameters. However, the available CGI dataset applied for the prediction of oocyte-derived DNA methylation inheritance are not large enough to train the neural networks. Results We propose a GRU-based model called CMIC (CGI Methylation Inheritance Classifier) to augment CGI sequence by converting it into variable-length k-mers, where the length k is randomly selected from the range $$k_{\min }$$ k min to $$k_{\max }$$ k max , N times, which were then used as neural network input. N was set to 1000 in the default setting. In addition, we proposed a new embedding vector generator for k-mers called splitDNA2vec. The randomness of this procedure was higher than the previous work, dna2vec. Conclusions We found that CMIC can predict the inheritance of oocyte-derived DNA methylation at CGIs in the maternal genome of blastocysts with a high F-measure (0.93). We also show that the F-measure can be improved by increasing the parameter N, that is, the number of sequences of variable-length k-mers derived from a single CGI sequence. This implies the effectiveness of augmenting input data by converting a DNA sequence to N sequences of variable-length k-mers. This approach can be applied to different DNA sequence classification and regression analyses, particularly those involving a small amount of data.

Highlights High rate of MED12 mutations and leiomyoma burden are clearly associated with myometrial oxidative stress. MED12 mutations are driven by misrepair of 8-OHdG at c.130-c.131 detected by deep sequencing analysis both in vitro and in vivo.

Background: This study aimed to investigate the effect of increased HER-2 expression on tumor-infiltrating lymphocytes (TILs) and determine its impact on the prognosis of colorectal cancer (CRC) patients; Methods: HER-2, CD4, CD8, CD19, LY6G, CD56, CD68, CD11b, and EpCam expression in CRC tissues and adjacent paracancerous tissues were assessed using multiplex fluorescence immunohistochemical staining. The correlation between HER-2 expression and the number of TILs in CRC tissues was analyzed. Kaplan–Meier and Cox proportional hazards models were used to analyze survival outcomes; Results: The expression of HER-2 in tumor tissues was higher than that in paracancerous tissues (1.31 ± 0.45 vs. 0.86 ± 0.20, p < 0.05). Additionally, there was an increase in the numbers of CD4+, CD8+, CD19+, and CD68+ cells in CRC tissues (14.11 ± 1.10 vs. 3.40 ± 0.18, p < 0.005; 0.16 ± 0.12 vs. 0.04 ± 0.04, p < 0.005; 0.71 ± 0.46 vs. 0.25 ± 0.13, p < 0.0005; 0.27 ± 0.24 vs. 0.03 ± 0.11, p < 0.05). An increase in HER-2 expression was positively correlated with an increase in CD4, CD8, and CD19 (p < 0.0001). In HER-2-positive CRC tissues, CD68 expression was increased (0.80 ± 0.55 vs. 0.25 ± 0.22, p < 0.05). In HER-2-upregulated CRC tissues, CD4, CD8, CD19, CD68, CD11b, Ly6G, and CD56 expressions were elevated (0.70 ± 0.37 vs. 0.32 ± 0.17, p = 0.03; 0.22 ± 0.13 vs. 0.09 ± 0.06, p = 0.03; 0.31 ± 0.19 vs. 0.12 ± 0.08, p = 0.02; 1.05 ± 0.62 vs. 0.43 ± 0.21, p < 0.01; 1.34 ± 0.81 vs. 0.53 ± 0.23, p < 0.01; 0.50 ± 0.31 vs. 0.19 ± 0.10, p < 0.01; 1.26 ± 0.74 vs. 0.52 ± 0.24, p < 0.01). Furthermore, increased HER-2 expression was an independent risk factor for recurrence-free survival (RFS) in patients (p < 0.01, HR = 3.421); Conclusions: The increased expression of HER-2 and its relationship with immune cells will provide new insights for immunotherapy in CRC patients.

Abstract Macrophages are among the most abundant immune cells in colorectal cancer (CRC). Re-educating tumor-associated macrophages (TAMs) to switch from protumoral to anti-tumoral activity is an attractive treatment strategy that warrants further investigation. However, little is known about the key pathway that is activated in TAMs. In this study, infitrating CD206+ TAMs in CRC were sorted and subjected to RNA-seq analysis. Differentially expressed genes were found to be enriched in unfolded protein response/endoplasmic reticulum stress response processes, and XBP1 splicing/activation was specifically observed in TAMs. XBP1 activation in TAMs promoted the growth and metastasis of CRC. Ablation of XBP1 inhibited the expression of the pro-tumor cytokine signature of TAMs, including IL-6, VEGFA, and IL-4. Simultaneously, XBP1 depletion could directly inhibit the expression of SIRPα and THBS1, thereby blocking “don’t eat me” recognition signals and enhancing phagocytosis. Therapeutic XBP1 gene editing using AAV2-sgXBP1 enhanced the anti-tumor activity. Together, XBP1 activation in TAMs drives CRC progression by elevating pro-tumor cytokine expression and secretion, as well as inhibiting macrophage phagocytosis. Targeting XBP1 signaling in TAMs may be a potential strategy for CRC therapy.

Abstract To study the carbon components in indoor and outdoor PM2.5, the samples of PM2.5 were collected from Nankai University in December 2015. The contents of eight carbon components were analyzed to use the thermo-optical reflection method. The results indicated that organic carbon (OC) mass concentration was 17.01, 19.48 and 18.92 µg/m3 in outdoor, dormitory and laboratory; elemental carbon (EC) mass concentration was 7.97, 3.56 and 3.53 µg/m3 in outdoor, dormitory and laboratory; and the total carbon aerosol was the proportion of more than 23% of PM2.5 samples. Lower wind speed and higher relative humidity were helpful to the accumulation of PM2.5. The ratio of OC/EC was > 2, and the SOC/OC ratio was > 30%, indicating that SOC was a crucial component indoors and outdoors. About 72% and 85% of the outdoor OC entering dormitory and laboratory environment, and about 59% and 71% of the outdoor EC entering dormitory and laboratory environment. Factor analysis of the eight carbon fractions indicated that the sources of OC and EC in outdoor, dormitory and laboratory is different.

Background: Hepatic hemangiomas are benign tumors with a favorable prognosis, but giant hepatic hemangiomas can cause abdominal symptoms and are indicated for treatment. Most cases are treated with surgery, but radiotherapy has also been used. However, to date, there have been no reports of proton beam therapy for a hepatic hemangioma. Case presentation: A 46-year-old woman had a tumor of 80 × 80 mm in the left medial lobe of the liver, which was diagnosed as a giant hemangioma based on the contrast pattern. Therapy was required for a giant hepatic hemangioma with symptoms, but the patient refused blood transfusion due to religious reasons, which made surgical resection difficult. Therefore, she was referred to our hospital for proton beam therapy. At her first visit, liver function was Child-Pugh A (5 points) and there was no elevation of tumor markers. Proton beam therapy of 28.6 Gy (RBE) given in 13 fractions was performed without interruption. The only observed acute radiation toxicity was Grade 1 dermatitis. One year after proton beam therapy, the hemangioma had significantly decreased, and a complete response has been maintained for 15 years based on ultrasound and MRI. Conclusion: This case is the first reported use of proton beam therapy for a hepatic hemangioma. The outcome suggests that this treatment may be effective for a giant liver hemangioma.

In this paper, the effects of quartz fibers on the resistivity, complex permittivity, complex permeability, and S-parameters of high-alumina cement pastes are investigated. Meanwhile, the change of microstructure after adding quartz fibers were analyzed by mercury intrusion porosimetry (MIP) and scanning electron microscope (SEM). The results show that with the increase the content of quartz fiber, the real part of complex permittivity decreases, while the imaginary part of complex permittivity, dielectric loss tangent, and resistivity increased. At the same time, there are more small pores, and the interface situation is more complicated after adding quartz fiber. Therefore, the reflectivity decreases and the absorptivity increases, but the scope of increase in the absorptivity is smaller than the decrease in the reflectivity, resulting in an increase in the transmittivity. When the content of quartz fiber is 0.15, 0.30, 0.45, and 0.60%, the average transmittance of high-alumina cement paste is increased by 3.69, 6.18, 15.51, and 21.03% respectively. It is indicated that the quartz fibers can improve the wave transmission properties of the high-alumina cement paste.

Cervical cancer remains a leading cause of cancer death in women, seriously threatening their physical and mental health. It is an easily preventable cancer with early screening and diagnosis. Although technical advancements have significantly improved the early diagnosis of cervical cancer, accurate diagnosis remains difficult owing to various factors. In recent years, artificial intelligence (AI)-based medical diagnostic applications have been on the rise and have excellent applicability in the screening and diagnosis of cervical cancer. Their benefits include reduced time consumption, reduced need for professional and technical personnel, and no bias owing to subjective factors. We, thus, aimed to discuss how AI can be used in cervical cancer screening and diagnosis, particularly to improve the accuracy of early diagnosis. The application and challenges of using AI in the diagnosis and treatment of cervical cancer are also discussed.

This study aims to propose a new treatment model for glioblastoma (GBM). The combination of chemotherapy, molecular targeted therapy and radiotherapy has been achieved in a highly simultaneous manner through the application of a safe, non-toxic, locally sustained drug-releasing composite Nanofiber mesh (NFM). The NFM consisted of biodegradable poly(ε-caprolactone) with temozolomide (TMZ) and 17-allylamino-17-demethoxygeldanamycin (17AAG), which was used in radiation treatment. TMZ and 17AAG combination showed a synergistic cytotoxicity effect in the T98G cell model. TMZ and 17AAG induced a radiation-sensitization effect, respectively. The NFM containing 17AAG or TMZ, known as 17AAG-NFM and TMZ-NFM, enabled cumulative drug release of 34.1% and 39.7% within 35 days. Moreover, 17AAG+TMZ-NFM containing both drugs revealed a synergistic effect in relation to the NFM of a single agent. When combined with radiation, 17AAG+TMZ-NFM induced in an extremely powerful cytotoxic effect. These results confirmed the application of NFM can simultaneously allow multiple treatments to T98G cells. Each modality achieved a significant synergistic effect with the other, leading to a cascading amplification of the therapeutic effect. Due to the superior advantage of sustained drug release over a long period of time, NFM has the promise of clinically addressing the challenge of high recurrence of GBM post-operatively.

The PIWI-interacting RNA (piRNA) pathway mainly consists of evolutionarily conserved protein factors. Intriguingly, many mutations of piRNA pathway factors lead to meiotic arrest during spermatogenesis. The majority of piRNA factor-knockout animals show arrested meiosis in spermatogenesis, and only a few show post-meiosis male germ cell arrest. It is still unclear whether the majority of piRNA factors expressed in spermatids are involved in long interspersed nuclear element-1 repression after meiosis, but future conditional knockout research is expected to resolve this. In addition, recent hamster knockout studies showed that a piRNA factor is necessary for oocytes—in complete contrast to the findings in mice. This species discrepancy allows researchers to reexamine the function of piRNA in female germ cells. This mini-review focuses on the current knowledge of protein factors derived from mammalian knockout studies and summarizes their roles in the biogenesis and function of piRNAs.

Mullerian adenosarcoma (MAS) is a biphasic tumor with malignant stroma. It is most commonly of endometrial origin but occasionally originates in the cervix, ovary, or other pelvic/peritoneal sites. The typical MAS is low grade with an indolent clinical course; however, tumors with sarcomatous overgrowth (SO) or a high-grade sarcoma tend to be aggressive. Tumor etiology is largely unknown. To better understand the global genome alterations and gene mutations in MAS, whole-genome sequencing (WGS) and target validation analysis were performed. MAS showed remarkable chromosome (chr) copy number variation (CNV), specifically, gains in chr 1q, 5p, 12p, 12q, and 17q and losses in chr 3p, 3q, 9p, and 11q. Gain of chr 12q13-15 was present in 50% of cases. The selected gene products in gain regions were upregulated as measured by immunohistochemistry. HMGA2 overexpression was significantly correlated with SO. While the structural variation (SV) rate was relatively low overall, a disproportionally high rate of break-ends at chr 7 was noted involving 6 in-frame rearrangement fusion genes. Among 40 frequently mutated genes detected by WGS and validated in 29 MAS by next generation sequencing (NGS), KMT2C, and BCOR were frequently seen in MAS both with and without SO, while MAGEC1 and KDM6B were strongly associated with SO. Overall, a higher rate of frequently mutated genes was found in MAS with SO (33%) than MAS without (11%). This study uncovers the complex and specific genetic alterations in this malignancy. The findings provide a tool for future investigation of these molecular changes in tumorigenesis and target therapies.

Aqueous zinc-ion batteries (AZIBs) have received increasing attention in large-scale energy storage systems because of their appealing features with respect to safety, cost, and scalability. Although vanadium oxides with different compositions demonstrate promising potential as cathodes for AZIBs, the narrow interlayer spacing, inferior electronic conductivity, and high dissolution in electrolyte seriously restrict their practical applications. Here we design an ingenious bimetallic-ion (Mg2+ and Al3+) co-intercalation strategy to boost the performance of AZIBs using V6O13·1.31H2O (VOH). The bimetallic-ion intercalation expands the interlayer spacing, increases electronic conductivity, and more importantly stabilizes the vanadium-oxygen bond in VOH, thus promoting ion/electron transport kinetics and restraining vanadium oxide dissolution. As expected, MgAl-VOH cathodes deliver ultrahigh specific capacities of 524.9 and 275.6 mA h g-1 at current densities of 0.1 and 5 A g-1, respectively, comparable to the highest value reported for vanadium oxides. The underlying zinc-ion storage mechanism is unambiguously clarified with the aid of density functional theory calculations and in situ structural characterization. Objective: We previously elucidated that long non-coding RNA Promoter of CDKN1A Antisense DNA damage Activated RNA (PANDAR) as a p53-dependent oncogene to promote cisplatin resistance in ovarian cancer (OC). Intriguingly, high level of p53-independent PANDAR was found in cisplatin-resistant patients with p53 mutation. Here, our study probed the new roles and the underlying mechanisms of PANDAR in p53-mutant OC cisplatin-resistance. Methods: A2780 and A2780-DDP cells were served as OC cisplatin-sensitive and cisplatinresistant cells. HO-8910PM cells were subjected to construct chemotherapy-induced extracellular vesicles (Chemo-EVs). Transmission electron microscopy (TEM) and nanoparticle tracking analysis were employed to evaluate Chemo-EVs. Cell viability was assessed using cell counting kit-8 and colony formation assays. Cell apoptosis was assessed using Annexin V and propidium iodide staining. The relationships between PANDAR, serine and arginine-rich premRNA splicing factor 9 (SRSF9) were verified by RNA immunoprecipitation and fluorescence in situ hybridization. Tumor xenograft experiment was employed to evaluate the effects of PANDAR-Chemo-EVs on OC cisplatin-resistance in vivo. Immunofluorescent staining and immunohistochemistry were performed in tumor tissue. Results: PANDAR level increased in OC patients with p53-mutation. PANDAR efflux enacted via exosomes under cisplatin conditions. Additionally, exosomes from OC cell lines carried PANDAR, which significantly increased cell survival and chemoresistance in vitro and tumor progression and metastasis in vivo. During cisplatin-induced stress, SRSF9 was recruited to nuclear bodies by increased PANDAR and muted apoptosis in response to cisplatin. Besides, SRSF9 significantly increased the ratio of SIRT4/SIRT6 mRNA in OC. Conclusion: Cisplatin-induced exosomes transfer PANDAR and lead to a rapid adaptation of OC cell survival through accumulating SRSF9 following cisplatin stress exposure. [ABSTRACT FROM AUTHOR]

Abstract Background Castleman disease (CD) is a rare lymphoproliferative disorder. This study is to evaluate the correlation between 18F-flurodeoxyglucose (18F-FDG) positron emission tomography-computed tomography (PET/CT) and clinical features in CD patients, and exploring its value in distinguishing disease severity and assisting in risk stratification. Methods We retrospectively enrolled 93 patients with newly diagnosed CD. Traditional semi-quantitative 18F-FDG PET/CT parameters including the maximum standardized uptake value (SUVmax), total metabolic lesion volume (MLV), total lesion glycolysis (TLG) were measured, and the lymph node to liver ratio of SUVmax (LLR), lymph node to mediastinal blood pool of SUVmax (LMR), spleen to liver ratio of SUVmax (SLR) and No. of involved lymph node stations (LNS) were calculated. The correlation between these metabolic parameters and clinical features were studied using a univariate analysis. The influencing factors of CD severity were determined by univariate and multivariate analysis. The optimal cut-off values for metabolic parameters were obtained by receiver operating characteristic (ROC) curve. Results A total of 20 unicentric CD (UCD) and 73 multicentric CD (MCD) cases were included, with the highest SUVmax of Lymph nodes ranged 1.40 ~ 28.18 (median, 4.86).
