Abstract Human coronaviruses exhibit a spectrum of symptoms, ranging from mild seasonal colds to severe respiratory manifestations. Despite progress in understanding the host's innate defense mechanisms against coronaviruses, how these viruses manipulate the immune response to promote inflammation remains elusive. In this study, we unveil the role of the coronavirus nonstructural protein 14 (NSP14) in leveraging the host's linear ubiquitin chain assembly complex (LUBAC) to instigate NF-κB activation, thereby triggering proinflammatory responses. Our findings uncover that HOIL-1-interacting protein (HOIP) directly engages with NSP14, conferring linear polyubiquitin chains onto NSP14. Consequently, ubiquitinated NSP14 recruits NEMO and initiates the activation of the IKK complex. This NSP14-induced NF-κB activation stimulates the expression of proinflammatory factors but not type I interferon, leading to a skewed host innate immune response tilting to inflammation. Collectively, our study sheds light on a virus-initiated linear ubiquitination pathway that induces NF-κB signaling and provokes proinflammatory responses.

Summary: Glioblastoma represents one of the most aggressive cancers, characterized by severely limited therapeutic options. Despite extensive investigations into this brain malignancy, cellular and molecular components governing its immunosuppressive microenvironment remain incompletely understood. Here, we identify a distinct neutrophil subpopulation, termed disease-specific suppressive granulocytes (DSSGs), present in human glioblastoma and lower-grade gliomas. DSSGs exhibit the concurrent expression of multiple immunosuppressive and immunomodulatory signals, and their abundance strongly correlates with glioma grades and poor clinical outcomes. Genetic disruption of neutrophil recruitment in immunocompetent mouse models of gliomas, achieved through Cxcl1 knockout in glioma cells or host-specific Cxcr2 deletion or diphtheria toxin A-mediated neutrophil depletion, can significantly enhance antitumor immunity and prolong survival. Further, we reveal that the skull bone marrow and meninges can be the primary sources of neutrophils and DSSGs in human and mouse glioma tumors. These findings demonstrate a critical mechanism underlying the establishment of the immunosuppressive microenvironment in gliomas.

Analysis of transcription factors and chromatin modifications at the genome-wide level provides insights into gene regulatory processes, such as transcription, cell differentiation and cellular response. Chromatin immunoprecipitation is the most popular and powerful approach for mapping chromatin, and other enzyme-tethering techniques have recently become available for living cells. Among these, Cleavage Under Targets and Tagmentation (CUT&Tag) is a relatively novel chromatin profiling method that has rapidly gained popularity in the field of epigenetics since 2019. It has also been widely adapted to map chromatin modifications and TFs in different species, illustrating the association of these chromatin epitopes with various physiological and pathological processes. Scalable single-cell CUT&Tag can be combined with distinct platforms to distinguish cellular identity, epigenetic features and even spatial chromatin profiling. In addition, CUT&Tag has been developed as a strategy for joint profiling of the epigenome, transcriptome or proteome on the same sample. In this review, we will mainly consolidate the applications of CUT&Tag and its derivatives on different platforms, give a detailed explanation of the pros and cons of this technique as well as the potential development trends and applications in the future.

Abstract The inadequate stability of organic–inorganic hybrid perovskites remains a significant barrier to their widespread commercial application in optoelectronic devices. Aging phenomena profoundly affect the optoelectronic performance of perovskite-based devices. In addition to enhancing perovskite stability, the real-time detection of aging status, aimed at monitoring the aging progression, holds paramount importance for both fundamental research and the commercialization of organic–inorganic hybrid perovskites. In this study, the aging status of perovskite was real-time investigated by using terahertz time-domain spectroscopy. Our analysis consistently revealed a gradual decline in the intensity of the absorption peak at 0.968 THz with increasing perovskite aging. Furthermore, a systematic discussion was conducted on the variations in intensity and position of the terahertz absorption peaks as the perovskite aged. These findings facilitate the real-time assessment of perovskite aging, providing a promising method to expedite the commercialization of perovskite-based optoelectronic devices. Graphical Abstract

The development of communication technology will promote the application of Internet of Things, and Beyond 5G will become a new technology promoter. At the same time, Beyond 5G will become one of the important supports for the development of edge computing technology. This paper proposes a communication task allocation algorithm based on deep reinforcement learning for vehicle-to-pedestrian communication scenarios in edge computing. Through trial and error learning of agent, the optimal spectrum and power can be determined for transmission without global information, so as to balance the communication between vehicle-to-pedestrian and vehicle-to-infrastructure. The results show that the agent can effectively improve vehicle-to-infrastructure communication rate as well as meeting the delay constraints on the vehicle-to-pedestrian link.

In order to investigate the impact mechanism of branch slope on fire smoke in underground bifurcation spaces, a series of small-scale fire experiments were conducted. This study measured relevant data under natural ventilation conditions for four different branch slopes and six fire power levels, focusing on the highest ceiling temperature in the tunnel and the attenuation pattern of ceiling temperatures along the longitudinal direction in both the main and branch tunnels. It also analyzed the temperature field, velocity field, and pressure field at the bifurcation. The results indicate that a decrease in branch slope leads to an increase in the highest temperature in the main tunnel, primarily due to the enhanced chimney effect that intensifies the entrainment of smoke and accelerates the flow of fire smoke. A modified model for the highest temperature, considering the influence of branch slope, was proposed. Branch slope has a certain impact on the longitudinal temperature attenuation of the tunnel ceiling, with a relatively weaker effect on the main tunnel. Based on empirical formulas, a formula for longitudinal temperature attenuation involving slope, including both the main and branch tunnels, was developed. Fire smoke accelerates at the bifurcation point, a phenomenon that intensifies with an increase in the slope of the branch tunnel. The research in this paper provides a reference for fire safety issues in bifurcated tunnel structures.

Understanding the spatial morphological characteristics and driving factors of rural settlements in ethnic minority areas is crucial for the conservation and tourism development of ethnic villages. Accordingly, this study employs Shuanglang Town, China as a case study, adopting an integrated approach that combines spatial syntax analysis, the optimal parameter geodetector model, and GIS spatial analysis techniques. This comprehensive methodology systematically investigates the spatial morphological features, differentiation characteristics, and influencing factors of ethnic villages. The findings reveal the logical lineage and formation mechanisms underlying the overall layout, street network, and public spaces of the villages. Specifically, the results demonstrate (1) a discernible gradation in spatial configurations, transitioning from compact “back mountain villages” in the northeast to more dispersed “seaside villages” in the southwest, with notable disparities in accessibility among different villages; (2) topography, water distribution, and water quality as the dominant factors shaping village spatial patterns; (3) the interactive and heterogeneous effects of multiple natural and anthropogenic factors, including topography, water resources, agricultural practices, and ethnic cultural traditions, significantly influencing the spatial morphology of villages; and (4) common principles governing the site selection of different ethnic village typologies, reflecting the villagers’ understanding and intelligent utilization of the natural environment. This study contributes to comprehending the spatial characteristics of rural settlements in ethnic minority areas and provides a theoretical and practical foundation for advancing analogous rural revitalization initiatives. The findings offer insights into the spatial logic and formation processes of ethnic villages, informing conservation efforts and sustainable tourism development strategies.

Abstract Adequacy, especially in the power system with high penetration of renewable energy sources, is a crucial consideration in power system planning and operational dispatching. A multi‐time scale adequacy evaluation method based on empirical mode decomposition (EMD) is proposed, taking into account the multi‐time scale characteristics of renewable energy sources and the response characteristics of flexible regulation resources. In this method, a net load curve is decomposed into several component curves by the EMD algorithm. Then, the adequacy demand on each time scale is obtained by waveform recognition. Furthermore, the available adequacy resources on different time scales are quantified according to the regulation models of flexible resources. By analysing the adequacy demand and available adequacy resources of the power system on the same time scale, the adequacy evaluation indices on each time scale can be calculated and weighted to form the comprehensive indices. Finally, the adequacy evaluation indices on each time scale with different capacities of renewable energy sources and energy storage systems are compared and analysed using a practical power system as a case. Simulation results show the validity of the multi‐time scale adequacy evaluation method.

IntroductionAt present, the use of beneficial microorganisms to control cucumber Fusarium wilt is a widely used method, and the rhizosphere microecological reset is one of the mechanisms involved. However, how biocontrol strains reshape cucumber rhizosphere microecology remains to be further studied.MethodsThe composition changes of cucumber root exudates induced by biocontrol strain 1JN2, the microbial ecology of cucumber rhizosphere and the colonization ability of biocontrol strain 1JN2 in cucumber rhizosphere were analyzed through UHPLC-MS/MS analysis, Illumina high-throughput sequencing and SEM, respectively.ResultsFirst, cucumber plants treated with biocontrol Bacillus 1JN2 reduced the disease severity of Fusarium wilt by 60%. Significant changes in cucumber root exudates were found after 1JN2 inoculation and the contents of four amino acids including glutamine, tryptophan, glycine and glutamic acid were significantly increased. Second, It was found that the bacterial diversity in the rhizosphere of cucumber was significantly increased in both the strain treatment group and the amino acid mixture treatment group, The number of Bacillus was the largest in all dominant populations, exceeded 20% in all treatment groups. The bacteria of Hydrogenispora and Vicinamibacteria were significantly increased after treatment.DiscussionOverall, the results demonstrated that amino acid substances in cucumber root exudates induced by biocontrol strain 1JN2 can shift the cucumber root microenvironment and prevent the occurrence of Fusarium wilt disease.

The rapid expansion of urban land has altered land use/land cover (LULC) types, affecting land surface temperatures (LSTs) and intensifying the urban heat island (UHI) effect, a prominent consequence of urbanization. This study, which focuses on Harbin, a representative city in a cold region, employs the patch-generating land use simulation (PLUS) model to predict LULC changes and a Bidirectional Long Short-Term Memory (Bi-LSTM) model to predict LST. The PLUS model exhibits a high prediction accuracy, evidenced by its FoM coefficient of 0.15. And the Bi-LSTM model also achieved high accuracy, with an R2 value of 0.995 and 0.950 and a root mean square error (RMSE) of 0.199 and 0.390 for predictions in winter and summer, respectively, surpassing existing methods. This study analyzed the trends in LULC, LST, and the urban thermal field variance index (UTFVI) to assess the relationships among LST, LULC, and UTFVI. The results show that urban land increased by 27.81%, and woodland and grassland decreased by 61.07% from 2005 to 2030. Areas with high temperatures increased by 40.86% in winter and 60.90% in summer. The proportion of the medium UTFVI zone (0.005–0.010) in urban land increased by 50.71%, and the proportion of areas with medium UTFVI values and above (>0.005) decreased at a rate of 84.70%. This finding suggests that the area affected by the UHI has decreased, while the UHI intensity in some regions has increased. This study provides a technical reference for future urban development and thermal environment management in cold regions.

Cell-cultured meat holds significant environmental value as an alternative protein source. Throughout the 21st century, cell-cultured meat has progressively penetrated commercial markets. However, a systematic review encompassing the entire field needs improvement. Employing Citespace, Vosviewer, and R-Bibliometrix software, a bibliometric analysis was used to present the research progress and general development trends of 484 articles on cell-cultured meat from 2000 to 2022 based on countries, authors, institutions, and keywords. This analysis provides ideas for the future development of cell-cultured meat in different countries or regions worldwide. Research on cell-cultured meat from 2000 to 2022 has undergone two phases: fluctuating growth (2000–2013) and rapid growth (2013–2022). Noteworthy contributions to cell-cultured meat studies emerge from author groups in the United States of America, the United Kingdom, and China, with influential institutions like the University of Bath significantly impacting pertinent research. Furthermore, over the past two decades, research has leaned towards exploring topics such as “biomaterials”, “cultured”, “land use”, “public opinion”, “animal welfare”, and “food safety”. Furthermore, this study reveals differences in nomenclature between regions and institutions. “Cultured meat” is more popular in some countries than in other forms. Institutions in Asia use “cultured meat” more frequently; however, institutions in the Americas adopt “cultivated meat” and rarely adopt “in vitro meat”, and institutions in the European region have no particularly prominent tendency towards a specific nomenclature. Future research should emphasize aligning the labeling of cell-cultured meat with effective management strategies and referencing regulatory policies across various countries. For the first time, we use three different bibliometric methods to analyze temporal and spatial variation in research on cellular meat. The results of this study have a multiplier effect. We provide a theoretical basis and a practical reference for the identification of alternatives in the dual context of “food crisis and food security” and “climate crisis”. At the same time, we also provide a reference for the sustainable development of the food system.

Abstract Insects pose significant challenges in cotton‐producing regions. Here, they describe a high‐throughput CRISPR/Cas9‐mediated large‐scale mutagenesis library targeting endogenous insect‐resistance‐related genes in cotton. This library targeted 502 previously identified genes using 968 sgRNAs, generated ≈2000 T0 plants and achieved 97.29% genome editing with efficient heredity, reaching upto 84.78%. Several potential resistance‐related mutants (10% of 200 lines) their identified that may contribute to cotton‐insect molecular interaction. Among these, they selected 139 and 144 lines showing decreased resistance to pest infestation and targeting major latex‐like protein 423 (GhMLP423) for in‐depth study. Overexpression of GhMLP423 enhanced insect resistance by activating the plant systemic acquired resistance (SAR) of salicylic acid (SA) and pathogenesis‐related (PR) genes. This activation is induced by an elevation of cytosolic calcium [Ca2+]cyt flux eliciting reactive oxygen species (ROS), which their demoted in GhMLP423 knockout (CR) plants. Protein‐protein interaction assays revealed that GhMLP423 interacted with a human epidermal growth factor receptor substrate15 (EPS15) protein at the cell membrane. Together, they regulated the systemically propagating waves of Ca2+ and ROS, which in turn induced SAR. Collectively, this large‐scale mutagenesis library provides an efficient strategy for functional genomics research of polyploid plant species and serves as a solid platform for genetic engineering of insect resistance.

IntroductionColorectal cancer is one of the most common gastrointestinal cancers and the second leading cause of cancer-related death. Although colonoscopy screening has greatly improved the early diagnosis of colorectal cancer, its recurrence and metastasis are still significant problems. Tumour cells usually have the hallmark of metabolic reprogramming, while fatty acids play important roles in energy storage, cell membrane synthesis, and signal transduction. Many pathways of fatty acid metabolism (FAM) are involved in the occurrence and development of colon cancer, and the complex molecular interaction network contains a variety of genes encoding key enzymes and related products.MethodsClinical information and RNA sequencing data were collected from TCGA and GEO databases. The prognosis model of colon cancer was constructed by LASSO-Cox regression analysis among the selected fatty acid metabolism genes with differential expression. Nomogram for the prognosis model was also constructed in order to analyze its value in evaluating the survival and clinical stage of the colon cancer patients. The differential expression of the selected genes was verified by qPCR and immunohistochemistry. GSEA and GSVA were used to analyze the enrichment pathways for high- and low-risk groups. CIBERSORT was used to analyze the immune microenvironment of colon cancer and to compare the infiltration of immune cells in the high- and low-risk groups. The “circlize” package was used to explore the correlation between the risk score signature and immunotherapy for colon cancer.ResultsWe analysed the differential expression of 704 FAM-related genes between colon tumour and normal tissue and screened 10 genes with prognostic value. Subsequently, we constructed a prognostic model for colon cancer based on eight optimal FAM genes through LASSO Cox regression analysis in the TCGA-COAD dataset, and its practicality was validated in the GSE39582 dataset. Moreover, the risk score calculated based on the prognostic model was validated as an independent prognostic factor for colon cancer patients. We further constructed a nomogram composed of the risk score signature, age and American Joint Committee on Cancer (AJCC) stage for clinical application. The colon cancer cohort was divided into high- and low-risk groups according to the optimal cut-off value, and different enrichment pathways and immune microenvironments were depicted in the groups.DiscussionSince the risk score signature was significantly correlated with the expression of immune checkpoint molecules, the prognostic model might be able to predict the immunotherapy response of colon cancer patients. In summary, our findings expand the prognostic value of FAM-related genes in colon cancer and provide evidence for their application in guiding immunotherapy.

Terahertz (THz) technology has seen significant advancements in the past decades, encompassing both fundamental scientific research, such as THz quantum optics, and highly applied areas like sixth-generation communications, medical imaging, and biosensing. However, the progress of on-chip THz integrated waveguides still lags behind that of THz sources and detectors. This is attributed to issues such as ohmic losses in microstrip lines, coplanar and hollow waveguides, bulky footprints, and reflection and scattering losses occurring at sharp bends or defects in conventional dielectric waveguides. Inspired by the quantum Hall effects and topological insulators in condensed matter systems, recent discoveries of topological phases of light have led to the development of topological waveguides. These waveguides exhibit remarkable phenomena, such as robust unidirectional propagation and reflectionless behavior against impurities or defects. As a result, they hold tremendous promise for THz on-chip applications. While THz photonic topological insulators (PTIs), including wave division, multiport couplers, and resonant cavities, have been demonstrated to cover a wavelength range of 800–2500 nm, research on tunable THz PTIs remains limited. In this perspective, we briefly reviewed a few examples of tunable PTIs, primarily concentrated in the infrared range. Furthermore, we proposed how these designs could benefit the development of THz on-chip PTIs. We explore the potential methods for achieving tunable THz PTIs through optical, electrical, and thermal means. Additionally, we present a design of THz PTIs for potential on-chip sensing applications. To support our speculation, several simulations were performed, providing valuable insights for future THz on-chip PTI designs.

Hepatocellular carcinoma (HCC) is the fourth leading cause of cancer-associated death worldwide. Angiogenesis, the process of formation of new blood vessels, is required for cancer cells to obtain nutrients and oxygen. HCC is a typical hypervascular solid tumor with an aberrant vascular network and angiogenesis that contribute to its growth, progression, invasion, and metastasis. Current anti-angiogenic therapies target mainly tyrosine kinases, vascular endothelial growth factor receptor (VEGFR), and platelet-derived growth factor receptor (PDGFR), and are considered effective strategies for HCC, particularly advanced HCC. However, because the survival benefits conferred by these anti-angiogenic therapies are modest, new anti-angiogenic targets must be identified. Several recent studies have determined the underlying molecular mechanisms, including pro-angiogenic factors secreted by HCC cells, the tumor microenvironment, and cancer stem cells. In this review, we summarize the roles of pro-angiogenic factors; the involvement of endothelial cells, hepatic stellate cells, tumor-associated macrophages, and tumor-associated neutrophils present in the tumor microenvironment; and the regulatory influence of cancer stem cells on angiogenesis in HCC. Furthermore, we discuss some of the clinically approved anti-angiogenic therapies and potential novel therapeutic targets for angiogenesis in HCC. A better understanding of the mechanisms underlying angiogenesis may lead to the development of more optimized anti-angiogenic treatment modalities for HCC.

Abstract It is crucial to understand why people comply with measures to contain viruses and their effects during pandemics. We provide evidence from 35 countries (N total = 12,553) from 6 continents during the COVID-19 pandemic (between 2021 and 2022) obtained via cross-sectional surveys that the social perception of key protagonists on two basic dimensions—warmth and competence—plays a crucial role in shaping pandemic-related behaviors. Firstly, when asked in an open question format, heads of state, physicians, and protest movements were universally identified as key protagonists across countries. Secondly, multiple-group confirmatory factor analyses revealed that warmth and competence perceptions of these and other protagonists differed significantly within and between countries. Thirdly, internal meta-analyses showed that warmth and competence perceptions of heads of state, physicians, and protest movements were associated with support and opposition intentions, containment and prevention behaviors, as well as vaccination uptake. Our results have important implications for designing effective interventions to motivate desirable health outcomes and coping with future health crises and other global challenges.

Soy whey wastewater(SWW) is produced in the production processes of tofu and soybean protein isolates. It has a large discharged volume and contains rich organic compounds such as whey soy proteins, oligosaccharides and soy isoflavones. At present, SWW is discharged into sewage plants and then treated via biochemical methods by most factories. This results in an increase in the production costs of tofu and soybean protein isolate and a large loss of the organics compounds. Therefore, the recycling of SWW has become a burning problem. On this ground, the studies on the recycling of SWW in recent years were reviewed from two perspectives: recovery of active compounds from SWW and biotransformation of SWW. The advantages and disadvantages of the two perspectives were also described. The results showed that most of the methods for recycling SWW were limited in a lab-scale. Only the recovery of whey soy proteins as animal feeds and biotransformation of SWW into biogas were operated in the industrial scale. On this basis, three future research directions were proposed for promoting the recycling of SWW: reducing the separation costs and increasing the use values of active compounds in SWW, increasing the transformation efficiency and rate of SWW and added values of corresponding products and evaluating the economic benefits of recycling methods to determine the feasibility of their industrialization.

Epitranscriptomics has emerged as another level of epigenetic regulation similar to DNA and histone modifications. N6-methyladenosine (m6A) is one of the most prevalent and abundant posttranscriptional modifications, widely distributed in many biological species. The level of N6-methyladenosine RNA methylation is dynamically and reversibly regulated by distinct effectors including methyltransferases, demethylases, histone modification and metabolites. In addition, N6-methyladenosine RNA methylation is involved in multiple RNA metabolism pathways, such as splicing, localization, translation efficiency, stability and degradation, ultimately affecting various pathological processes, especially the oncogenic and tumor-suppressing activities. Recent studies also reveal that N6-methyladenosine modification exerts the function in immune cells and tumor immunity. In this review, we mainly focus on the regulatory mechanisms of N6-methyladenosine RNA methylation, the techniques for detecting N6-methyladenosine methylation, the role of N6-methyladenosine modification in cancer and other diseases, and the potential clinical applications.

The study examined how children's self-regulation skills measured by the strengths and weaknesses of ADHD symptoms and normal behavior rating are associated with story comprehension and how verbal engagement and e-book discussion prompts moderate this relation. Children aged 3-7 (N = 111, 50% female, Chinese as first language) read an interactive Chinese-English bilingual story e-book with or without discussion prompts twice with their parents (2020-2021). Results demonstrated that the lower children's self-regulation skills, the more they struggled with story comprehension. Critically, our data suggest that embedding e-book discussion prompts and more verbalization in English can mitigate this negative association for children with inattention/hyperactivity. These findings have critical implications for future e-book design, interventions, and home reading practice for children with inattention/hyperactivity and those at risk for attention deficit/hyperactivity disorder.

Cotton (Gossypium hirsutum) is an important fiber crop worldwide. Insect attack causes cotton yield and quality losses. However, little is known about the mechanism of cotton response to insect attack. We simulated insect feeding by applying insect oral secretions (OS) to wounds, and combined transcriptome and metabolome analysis to investigate how OS from two major pest species (Helicoverpa armigera and Spodoptera litura) affect cotton defense responses. We found that respectively 12,668 and 13,379 genes were differentially expressed in comparison with wounding alone. On addition of OS, the jasmonic acid signaling pathway was rapidly and strongly induced, whereas genes involved in salicylic acid biosynthesis were downregulated. On constructing a coexpression gene network, we identified a hub gene encoding a leucine-rich repeat receptor kinase that may play an important role in early signal recognition and transduction. OS from the two insect species altered the abundance of flavonoid-related compounds in different patterns. Gossypol remained in lower concentration after OS application than after wounding alone, suggesting a suppressive effect of OS on cotton defense response. This study illustrated transcriptional and metabolic changes of cotton in responding to OS from two chewing insect species, identified potential key response genes, and revealed evidence for OS inhibition of wounding-induced cotton defense response.

Abstract Two-dimensional (2D) lead-free (K, Na)NbO3 (KNN) micro/nano structures with controllable K/Na ratio were successfully fabricated via a two-step molten salt synthesis (MSS). In this work, the reaction factors, including the proportion of molten salts, the types of carbonates, the sintering temperature, and the sintering time, were discussed in detail and the optimized condition was identified. The microstructure of KNN was confirmed by confocal Raman spectroscopy, while piezoresponse force microscopy (PFM) was applied to measure three-dimensional (3D) morphology and piezoelectric properties of KNN particles. The as-synthesized KNN platelets apparently possess anisotropic morphology and uniform structure, the size of which reaches 5–20 µm in length/width and 0.5–1 µm in thickness. It should be noted that the K/Na ratios of the KNN crystals are basically consistent while the proportion of salts changes within a certain range. The enrichment of Na element in the products is also observed, which owes to the smaller ionic radius of Na+ comparing to that of K+. This result provides a reference for the further preparation of textured ceramics and flexible piezoelectric generators.

We demonstrate an ultrabroadband electrically controllable terahertz modulator based on a Schottky diode structure formed with periodic metal microslits on an n-doped GaAs substrate. The mechanism of our design is different from that of the traditional Schottky diode-based THz electrical modulator, which uses free charge carriers in a substrate to control the resonant behavior of metamaterials. In our device, the modulation is based on free-carrier absorption on the THz wave and therefore broadband. The charge carrier concentration between the metal microslits is actively modified by applying a reverse bias voltage to generate a direct modulation of THz waves. The modulation performance is enhanced by the THz non-resonant electric field enhancement effect from the metal microslits. The experimental results indicate that the modulation depth is positively correlated with the electric field enhancement ratio at the depletion region in the gap and the number of microslits in the THz light spot-covered area. An averaged modulation depth of ∼40% in the measurable frequency range from 0.4 to 1.4 THz was achieved by the device with a metal microslits gap width of 2 µm and a period of 20 µm. A maximum modulation depth of ∼75% was achieved by stacking two devices back-to-back with a 3-dB down bandwidth modulation speed of ∼100 kHz. Further improvements of the device can be achieved by optimizing the parameters such as the free-carrier density in the doping layer, the active area size, and the specifications of the metal microslits.

Abstract Anthropogenic factors play an important role in shaping the distribution of wildlife species and their habitats, and understanding the influence of human activities on endangered species can be key to improving conservation efforts as well as the implementation of national strategies for sustainable development. Here, we used species distribution modeling to assess human impacts on the endangered red panda (Ailurus fulgens) in high‐altitude regions of Nepal. We found that the distance to paths (tracks used by people and animals), livestock density, human population density, and annual mean temperature were the most important factors determining the habitat suitability for red pandas in Nepal. This is the first study that attempts to use comprehensive environmental and anthropogenic variables to predict habitat suitability for the red pandas at a national level. The suitable habitat identified by this study is important and could serve as a baseline for the development of conservation strategies for the red panda in Nepal.

Ribulose 5-phosphate (Ru5P) and guanosine 5′-triphosphate (GTP) are two key precursors of riboflavin, whereby Ru5P is also a precursor of GTP. Ribulose 5-phosphate 3-epimerase (Rpe) catalyzes the conversion of ribulose 5-phosphate into xylulose 5-phosphate. Inactivation of Rpe can reduce the consumption of Ru5P, enhancing the carbon flux toward riboflavin biosynthesis. Here we investigated the effect of mutation of rpe and other related genes on riboflavin production, physiological and metabolic phenotypes in Bacillus subtilis LY (BSLY). Introducing single nucleotide deletion (generated BSR) or nonsense mutation (generated BSRN) on the genomic copy of rpe, resulting in more than fivefold increase of riboflavin production over the parental strain. BSR process 62% Rpe activity, while BSRN lost the entire Rpe activity and had a growth defect compared with the parent strain. BSR and BSRN exhibited increases of the inosine and guanine titers, in addition, BSRN exhibited an increase of inosine 5′-monophosphate titer in fermentation. The transcription levels of most oxidative pentose phosphate pathway and purine synthesis genes were unchanged in BSR, except for the levels of zwf and ndk, which were higher than in BSLY. The production of riboflavin was increased to 479.90 ± 33.21 mg/L when ribA was overexpressed in BSR. The overexpression of zwf, gntZ, prs, and purF also enhanced the riboflavin production. Finally, overexpression of the rib operon by the pMX45 plasmid and mutant gnd by pHP03 plasmid in BSR led to a 3.05-fold increase of the riboflavin production (977.29 ± 63.44 mg/L), showing the potential for further engineering of this strain.

Abstract Background Current knowledge and data on miRNA-lncRNA interactions is still limited and little effort has been made to predict target lncRNAs of miRNAs. Accumulating evidences suggest that the interaction patterns between lncRNAs and miRNAs are closely related to relative expression level, forming a titration mechanism. It could provide an effective approach for characteristic feature extraction. In addition, using the coding non-coding co-expression network and sequence data could also help to measure the similarities among miRNAs and lncRNAs. By mathematically analyzing these types of similarities, we come up with two findings that (i) lncRNAs/miRNAs tend to collaboratively interact with miRNAs/lncRNAs of similar expression profiles, and vice versa, and (ii) those miRNAs interacting with a cluster of common target genes tend to jointly target at the common lncRNAs. Methods In this work, we developed a novel group preference Bayesian collaborative filtering model called GBCF for picking up a top-k probability ranking list for an individual miRNA or lncRNA based on the known miRNA-lncRNA interaction network. Results To evaluate the effectiveness of GBCF, leave-one-out and k-fold cross validations as well as a series of comparison experiments were carried out. GBCF achieved the values of area under ROC curve of 0.9193, 0.8354+/− 0.0079, 0.8615+/− 0.0078, and 0.8928+/− 0.0082 based on leave-one-out, 2-fold, 5-fold, and 10-fold cross validations respectively, demonstrating its reliability and robustness. Conclusions GBCF could be used to select potential lncRNA targets of specific miRNAs and offer great insights for further researches on ceRNA regulation network.

A better understanding of the mode of action of postharvest biocontrol agents on fruit surfaces is critical for the advancement of successful implementation of postharvest biocontrol products. This is due to the increasing importance of biological control of postharvest diseases over chemical and other control methods. However, most of the mechanisms involved in biological control remain unknown and need to be explored. Yarrowia lipolytica significantly inhibited blue mold decay of apples caused by Penicillium expansum. The findings also demonstrated that Y. lipolytica stimulated the activities of polyphenoloxidase, peroxidase, chitinase, l-phenylalanine ammonia lyase involved in enhancing defense responses in apple fruit tissue. Proteomic and transcriptomic analysis revealed a total of 35 proteins identified as up- and down-regulated in response to the Y. lipolytica inducement. These proteins were related to defense, biotic stimulus, and stress responses, such as pathogenesis-related proteins and dehydrin. The analysis of the transcriptome results proved that the induced resistance was mediated by a crosstalk between salicylic acid (SA) and ethylene/jasmonate (ET/JA) pathways. Y. lipolytica treatment activated the expression of isochorismate synthase gene in the SA pathway, which up-regulates the expression of PR4 in apple. The expression of 1-aminocyclopropane-1-carboxylate oxidase gene and ET-responsive transcription factors 2 and 4, which are involved in the ET pathway, were also activated. In addition, cytochrome oxidase I, which plays an important role in JA signaling for resistance acquisition, was also activated. However, not all of the genes had a positive effect on the SA and ET/JA signal pathways. As transcriptional repressors in JA signaling, TIFY3B and TIFY11B were triggered by the yeast, but the gene expression levels were relatively low. Taken together, Y. lipolytica induced the SA and ET/JA signal mediating the defense pathways by stimulating defense response genes, such as peroxidase, thaumatin-like protein, and chitinase 4-like, which are involved in defense response in apple.[Graphic: see text] Copyright © 2017 The Author(s) This is an open access article distributed under the CC BY-NC-ND 4.0 International license.

Different from conventional single-task optimization, the recently proposed multitasking optimization (MTO) simultaneously deals with multiple optimization tasks with different types of decision variables. MTO explores the underlying similarity and complementarity among the component tasks to improve the optimization process. The well-known multifactorial evolutionary algorithm (MFEA) has been successfully introduced to solve MTO problems based on transfer learning. However, it uses a simple and random inter-task transfer learning strategy, thereby resulting in slow convergence. To deal with this issue, this paper presents a two-level transfer learning (TLTL) algorithm, in which the upper-level implements inter-task transfer learning via chromosome crossover and elite individual learning, and the lower-level introduces intra-task transfer learning based on information transfer of decision variables for an across-dimension optimization. The proposed algorithm fully uses the correlation and similarity among the component tasks to improve the efficiency and effectiveness of MTO. Experimental studies demonstrate the proposed algorithm has outstanding ability of global search and fast convergence rate.

Evidences increasingly indicate the involvement of long non-coding RNAs (lncRNAs) in various biological processes. As the mutations and abnormalities of lncRNAs are closely related to the progression of complex diseases, the identification of lncRNA-disease associations has become an important step toward the understanding and treatment of diseases. Since only a limited number of lncRNA-disease associations have been validated, an increasing number of computational approaches have been developed for predicting potential lncRNA-disease associations. However, how to predict potential associations precisely through computational approaches remains challenging. In this study, we propose a novel two-side sparse self-representation (TSSR) algorithm for lncRNA-disease association prediction. By learning the self-representations of lncRNAs and diseases from known lncRNA-disease associations adaptively, and leveraging the information provided by known lncRNA-disease associations and the intra-associations among lncRNAs and diseases derived from other existing databases, our model could effectively utilize the estimated representations of lncRNAs and diseases to predict potential lncRNA-disease associations. The experiment results on three real data sets demonstrate that our TSSR outperforms other competing methods significantly. Moreover, to further evaluate the effectiveness of TSSR in predicting potential lncRNAs-disease associations, case studies of Melanoma, Glioblastoma, and Glioma are carried out in this paper. The results demonstrate that TSSR can effectively identify some candidate lncRNAs associated with these three diseases.

The mud pump piston is a key part for providing mud circulation, but its sealing performance often fails under complex working conditions, which shorten its service life. Inspired by the ring segment structure of earthworms, the bionic striped structure on surfaces of the mud pump piston (BW-160) was designed and machined, and the sealing performances of the bionic striped piston and the standard piston were tested on a sealing performance testing bench. It was found the bionic striped structure efficiently enhanced the sealing performance of the mud pump piston, while the stripe depth and the angle between the stripes and lateral of the piston both significantly affected the sealing performance. The structure with a stripe depth of 2 mm and angle of 90° showed the best sealing performance, which was 90.79% higher than the standard piston. The sealing mechanism showed the striped structure increased the breadth and area of contact sealing between the piston and the cylinder liner. Meanwhile, the striped structure significantly intercepted the early leaked liquid and led to the refluxing rotation of the leaked liquid at the striped structure, reducing the leakage rate.

The efficacy of Sporidiobolus pararoseus Y16 in controlling postharvest blue mold caused by Penicillium expansum on apples and the defense response involved were evaluated. The results suggested that the decay incidence of blue mold of apples treated by S. pararoseus Y16 was significantly reduced compared with the control. In vitro testing indicated that germination of spores and germ tube length of P. expansum were markedly inhibited by S. pararoseus Y16. Meanwhile, polyphenol oxidase (PPO), peroxidase (POD), phenylalanine ammonia lyase (PAL), and catalase (CAT) activities and several pathogenesis-related (PR) gene expression levels (including PR3, PR4, PR5, and PR9) were determined. In apples, the activities of PPO, POD, CAT, and PAL were significantly induced by S. pararoseus Y16 treatment compared with the control fruits. The relative expression levels of PR3 and PR4 were significantly induced at 4 and 6 d, while PR5 was significantly induced at 4 and 6 d and PR9 was significantly induced at 4 d. Therefore, the reduction in apple fruit decay by S. pararoseus Y16 treatment could be related to the increased activities of related enzymes and proteins involved in the defense against pathogens, which suggest that S. pararoseus Y16 is a potential antagonistic yeast.

The Crested Ibis Nipponia nippon was once thought to be extinct in the wild until seven birds were discovered in a remote mountain village in China in 1981. Studies suggested that winter-flooded rice fields play an essential role in nest site selection by the Crested Ibis and hence in their survival. Considerable efforts were therefore made to conserve the winter-flooded rice fields, but these have caused conflicts between the agricultural and conservation communities. The population and geographical range of the wild Crested Ibis has expanded greatly since 1981, but there is no spatial information on the winter-flooded rice fields, nor on the current association of nest sites and winter-flooded rice fields. We mapped winter-flooded rice fields across the entire current range of Crested Ibis using innovative remote sensing and geographical information systems (GIS) techniques. The spatial relationships between the nest site clusters and winter-flooded rice fields were quantified using Ward's hierarchical clustering method and Ripley's K-function. We show that both have significantly clumped distribution patterns and that they are positively associated. However, the dependence of Crested Ibis on the winter-flooded rice fields varied significantly among the nest site clusters and has decreased over the years, indicating the absence of winter-flooded rice fields is not constraining their recovery and population expansion. We therefore recommend that efforts should be made to protect the existing winter-flooded rice fields and to restore the functionality of natural and semi-natural wetlands, to encourage both in-situ conservation and the re-introduction of the Crested Ibis. In addition, we recommend that caution should be exercised when interpreting the habitat requirements of species with a narrow distribution, particularly when that interpretation is based only on their current habitat.

T-rays is sensitive to covalently cross-linked proteins and can be used to probe unique dynamic properties of water surrounding a protein. In this paper, we demonstrate the unique absorption properties of the dynamic hydration shells determined by hemagglutinin (HA) protein in terahertz frequency. We study the changes arising from different concentrations in detail and show that nonlinear absorption coefficient is induced by the dynamic hydration water. The binary and ternary component model were used to interpret the nonlinearity absorption behaviors and predict the thickness of the hydration shells around the HA protein in aqueous phase.

The GP2 peptide is derived from the Human Epidermal growth factor Receptor 2 (HER2/nue), a marker protein for breast cancer present in saliva. In this paper we study the temperature dependent behavior of hydrated GP2 at terahertz frequencies and find that the peptide undergoes a dynamic transition between 200 and 220 K. By fitting suitable molecular models to the frequency response we determine the molecular processes involved above and below the transition temperature (T(D)). In particular, we show that below T(D) the dynamic transition is dominated by a simple harmonic vibration with a slow and temperature dependent relaxation time constant and that above T(D), the dynamic behavior is governed by two oscillators, one of which has a fast and temperature independent relaxation time constant and the other of which is a heavily damped oscillator with a slow and temperature dependent time constant. Furthermore a red shifting of the characteristic frequency of the damped oscillator was observed, confirming the presence of a non-harmonic vibration potential. Our measurements and modeling of GP2 highlight the unique capabilities of THz spectroscopy for protein characterization.
