Abstract Background Preoperative discrimination between non-muscle-invasive bladder cancer (NMIBC) and the muscle invasive bladder cancer (MIBC) is a determinant of management. The purpose of this research is to employ radiomics to evaluate the diagnostic value in determining muscle invasiveness of compressed sensing (CS) accelerated 3D T2-weighted-SPACE sequence with high resolution and short acquisition time. Methods This prospective study involved 108 participants who underwent preoperative 3D-CS-T2-weighted-SPACE, 3D-T2-weighted-SPACE and T2-weighted sequences. The cohort was divided into training and validation cohorts in a 7:3 ratio. In the training cohort, a Rad-score was constructed based on radiomic features selected by intraclass correlation coefficients, pearson correlation coefficient and least absolute shrinkage and selection operator . Multivariate logistic regression was used to develop a nomogram combined radiomics and clinical indices. In the validation cohort, the performances of the models were evaluated by ROC, calibration, and decision curves. Results In the validation cohort, the area under ROC curve of 3D-CS-T2-weighted-SPACE, 3D-T2-weighted-SPACE and T2-weighted models were 0.87(95% confidence interval (CI):0.73-1.00), 0.79(95%CI:0.63–0.96) and 0.77(95%CI:0.60–0.93), respectively. The differences in signal-to-noise ratio and contrast-to-noise ratio between 3D-CS-T2-weighted-SPACE and 3D-T2-weighted-SPACE sequences were not statistically significant(p > 0.05). While the clinical model composed of three clinical indices was 0.74(95%CI:0.55–0.94) and the radiomics-clinical nomogram model was 0.88(95%CI:0.75-1.00). The calibration curves confirmed high goodness of fit, and the decision curve also showed that the radiomics model and combined nomogram model yielded higher net benefits than the clinical model. Conclusion The radiomics model based on compressed sensing 3D T2WI sequence, which was acquired within a shorter acquisition time, showed superior diagnostic efficacy in muscle invasion of bladder cancer. Additionally, the nomogram model could enhance the diagnostic performance.

Abstract Purpose To investigate the performance of histogram features of non-Gaussian diffusion metrics for diagnosing muscle invasion and histological grade in bladder cancer (BCa). Methods Patients were prospectively allocated to MR scanner1 (training cohort) or MR2 (testing cohort) for conventional diffusion-weighted imaging (DWIconv) and multi-b-value DWI. Metrics of continuous time random walk (CTRW), diffusion kurtosis imaging (DKI), fractional-order calculus (FROC), intravoxel incoherent motion (IVIM), and stretched exponential model (SEM) were simultaneously calculated using multi-b-value DWI. Whole-tumor histogram features were extracted from DWIconv and non-Gaussian diffusion metrics for logistic regression analysis to develop diffusion models diagnosing muscle invasion and histological grade. The models’ performances were quantified by area under the receiver operating characteristic curve (AUC). Results MR1 included 267 pathologically-confirmed BCa patients (median age, 67 years [IQR, 46–82], 222 men) and MR2 included 83 (median age, 65 years [IQR, 31–82], 73 men). For discriminating muscle invasion, CTRW achieved the highest testing AUC of 0.915, higher than DWIconv’s 0.805 (p = 0.014), and similar to the combined diffusion model’s AUC of 0.885 (p = 0.076). For differentiating histological grade of non-muscle-invasion bladder cancer, IVIM outperformed a testing AUC of 0.897, higher than DWIconv’s 0.694 (p = 0.020), and similar to the combined diffusion model’s AUC of 0.917 (p = 0.650). In both tasks, DKI, FROC, and SEM failed to show diagnostic superiority over DWIconv (p > 0.05). Conclusion CTRW and IVIM are two potential non-Gaussian diffusion models to improve the MRI application in assessing muscle invasion and histological grade of BCa, respectively. Critical relevance statement Our study validates non-Gaussian diffusion imaging as a reliable, non-invasive technique for early assessment of muscle invasion and histological grade in BCa, enhancing accuracy in diagnosis and improving MRI application in BCa diagnostic procedures. Water use efficiency (WUE) is an important metric for assessing regional sustainability and the carbon-water balance of ecosystems. Drought is a significant stressor affecting the carbon-water cycle. Nevertheless, the response of WUE to different types of droughts remains poorly understood. This study analyzed the variability characteristics of WUE, meteorological (SPEI), hydrological (SRI), and agricultural (SSMI) droughts. Additionally, it investigated the response of WUE to these three types of droughts over the Mongolian Plateau (1982–2021). The results indicated a non-significant decreasing trend in WUE, and a remarkable drying trend in SPEI, SRI, and SSMI. WUE was positively correlated with three drought indices, with average R (WUE, SPEI), R (WUE, SRI), and R (WUE, SSMI) at 0.060, 0.051, and 0.028, respectively; this indicated that WUE decreased as drought intensified. Regional WUE proved most sensitive to SPEI and was observed in coniferous forests, broadleaf forests, meadow steppes, typical steppes, and alpine grasslands. Sand land vegetation and desert steppes showed the highest sensitivity to SRI, whereas shrubs and croplands exhibited the highest sensitivity to SSMI. More importantly, SRI caused more severe losses in WUE than SSMI and SPEI. These findings offer valuable insights for future carbon sink management and drought risk assessment.

The regional water cycle reflects the dual processes of nature and society. A healthy and benign water cycle is a fundamental guarantee for achieving regional sustainable development.The Yangtze River Economic Belt, as a densely populated urban cluster with abundant water resources in China, plays a crucial role in promoting the sustainable development of Chinese urban clusters. It is of significant importance to optimize the water cycle health of the Yangtze River Economic Belt and explore its primary obstacles.This study integrates the natural-social attributes of the water cycle and selects 25 evaluation indicators from six dimensions: Drivers (D), Pressures (P), States (S), Impacts (I), Responses (R), and Management (M), to construct the water cycle evaluation system for the Yangtze River Economic Belt.Combining Analytic Hierarchy Process (AHP), Decision-Making Trial and Evaluation Laboratory (DEMATEL), and the Criteria Importance Through Intercriteria Correlation (CRITIC) method, the comprehensive weights of subjective and objective factors are obtained based on the principle of minimum relative entropy.Based on this, an Extended Cloud Model is constructed to evaluate the water cycle health status of the region from 2012 to 2022.Furthermore, the Obstacle Degree Model is introduced to identify the primary obstacles affecting the water cycle health in the region. Finally, the ARIMA model is applied to forecast the water cycle health status in the region from 2023 to 2030.The results indicate that the comprehensive water cycle health level in the region improved gradually from initially sub-pathological to sub-healthy status from 2012 to 2022.Per capita GDP, industrial water consumption per ten thousand yuan of industrial value added, per capita water consumption, forest coverage rate, sediment transport module, proportion of environmental protection investment to GDP, and proportion of water resources management investment to GDP are the primary obstacles affecting the improvement of health in this region.The forecast indicates that the overall water cycle health status in the region will remain at a sub-healthy level from 2023 to 2030.The research results can provide a scientific reference for further promoting the high-quality development of the Yangtze River Economic Belt, better supporting and serving China's modernization, and providing scientific reference basis.

Abstract Bacterial meningitis is a serious complication after neurosurgery and has a high mortality rate. Early diagnosis and timely treatment are crucial or will have disastrous consequences. The classic triad of bacterial meningitis lacks sensitivity and specificity. Therefore, the diagnosis of post-neurosurgical bacterial meningitis relies on cerebrospinal fluid. But traditional cerebrospinal fluid parameters are not completely reliable in diagnosing post-neurosurgical bacterial meningitis. Previous studies have found that CSF lactate concentration is related to bacterial meningitis. But, after the craniocerebral operation, the cerebrospinal fluid of most patients is bloody. Whether red blood cells interfere with diagnosing PNBM based on lactate concentration is limited. In the current study, we further analysis on whether red blood cells interfere with diagnosing PNBM based on lactate concentration. This study aimed to investigate the value of cerebrospinal fluid lactate concentrations in post-neurosurgical bacterial meningitis. A prospective observational study was performed on 62 patients at Kunshan First People’s Hospital’s intensive care unit affiliated with Jiangsu University. We found that erythrocytes do not affect cerebrospinal fluid lactate, and elevated lactate concentrations can be used as a marker for postoperative bacterial meningitis.

Hemolytic activity assessment is a widely used method to evaluate the toxicity of marine organisms, including jellyfish. However, there are some challenges associated with testing hemolytic activity. In this study, four methods were employed to compare the hemolytic activity of jellyfish tentacle extract (TE). Firstly, a microplate reader was used to compare the mouse hemoglobin solution at three different wavelengths (415 nm, 541 nm, and 576 nm), and the most sensitive wavelength was selected for further experiments. Secondly, photomicrograph counting was used to determine the number of complete red blood cells in the field of view. Thirdly, a microplate reader was used to test hemolytic activity in a 96-well plate at 415 nm. Fourthly, a Bicinchoninic Acid (BCA) kit was used to test the concentration of hemoglobin in the solution. Finally, a UV-Vis Spectrophotometer was used to test hemolytic activity at 415 nm. Among the three wavelengths tested, the absorption value was most sensitive at 415 nm. The photomicrograph counting method was able to reflect changes in the shape of Red Blood Cells (RBCs). The microplate reader method may exhibit deviations when the solution concentration is high, while external factors could influence the BCA kit when the toxin concentration in the experimental group is low. The spectrophotometer method was found to be relatively accurate and sensitive to changes. When optimizing the method, it is important to consider the applicability of the Beer-Lambert law and the concentration of solutions.

Psoriasis is a common chronic inflammatory skin disease, which has seriously affected human health. Berberine is a plant alkaloid with significant anti-inflammatory effect. In this study, we aimed to determine whether topical application of berberine could ameliorate skin inflammation in psoriasis and explore the potential molecular mechanism. Imiquimod (IMQ)-induced psoriasis-like dermatitis in mice was firstly used to reveal the potential pathogenic mechanism. The transcriptome analysis showed that Janus kinase (JAK)-Signal transducer and activator of transcription (STAT) signaling pathway was significantly enriched in IMQ-induced dermatitis, which included the key genes such as Janus kinase 1 (JAK1), Interleukin-2 (IL2), Phosphatidylinositol 4,5-bisphosphate 3-kinase catalytic subunit alpha isoform (PIK3CA), Phosphatidylinositol 4,5-bisphosphate 3-kinase catalytic subunit beta isoform (PIK3CB) and Signal transducer and activator of transcription 1 (STAT1). Network pharmacology and molecular docking then predicted that topical berberine may treat psoriasis by JAK-STAT signaling pathway, especially act on JAK1, IL2, PIK3CA and PIK3CB. Experimental studies in vivo further demonstrated that topical application of berberine could ameliorate IMQ-induced psoriasis-like skin inflammation by suppressing JAK-STAT signaling pathway. In addition, experimental studies in vitro showed that berberine could directly act on and enter into human immortalized keratinocytes (HaCaT cells). Meanwhile, berberine may inhibit the hyperproliferation and proinflammatory functions of HaCaT cells induced by Interferon-gamma (IFN-γ) via suppressing JAK1/STAT1 signaling pathway. In conclusion, this study suggested that berberine may be a promising topical agent to ameliorate skin inflammation in psoriasis.

Allergic diseases in children are major public health concerns due to their widespread and rising prevalence. Food-specific immunoglobulin G4(FS-IgG4) has been detected in patients with allergic diseases, but its clinical significance is still debated. In the present study, 407 children with allergic diseases were recruited and categorized into three groups according to the different systems involved: the respiratory system group, the skin system group, and a multiple system group, with the collection of clinical symptoms and serum antibodies, including total immunoglobulin E (IgE), house dust mite (HDM) IgE, food-specific IgE (FS-IgE), and FS-IgG4. Part of these patients were followed up with the intervention of FS-IgG4-guided diet elimination with or without add-on probiotics supplement. The analysis at baseline revealed distinct serum levels of different antibodies. The positive rate of FS-IgG4 in all groups was more than 80%, and the proportion of total IgE and FS-IgG4 both positive in the multi-system group was the highest (p=0.039). Currently, there are several treatments approaches available for lung cancer; however, patients who develop drug resistance or have poor survival rates urgently require new therapeutic strategies for lung cancer. In autophagy, damaged proteins or organelles are enclosed within autophagic vesicles with a bilayer membrane structure and transported to the lysosomes for degradation and recirculation. Autophagy is a crucial pathway involved in the clearance of reactive oxygen species (ROS) and damaged mitochondria. Meanwhile, inhibiting autophagy is a promising strategy for cancer treatment. In this study, we found for the first time that Cinchonine (Cin) can act as an autophagy suppressor and exert anti-tumor effects. Cin significantly inhibited the proliferation, migration, and invasion of cancer cells in vitro and the tumor growth and metastasis in vivo, without obvious toxic effects. We found that Cin suppressed the autophagic process by blocking autophagosome degradation through the inhibition of the maturation of lysosomal hydrolases. Cin-mediated autophagy inhibition resulted in the elevated ROS level and the accumulation of damaged mitochondria, which in turn promoted apoptosis. N-acetylcysteine, a potential ROS scavenger, significantly suppressed Cin-induced apoptosis. Additionally, Cin upregulated programmed death-ligand 1 (PD-L1) expression in lung cancer cells by inhibiting autophagy. Compared with monotherapy and control group, the combined administration of anti-PD-L1 antibody and Cin significantly reduced tumor growth. These results suggest that Cin exerts anti-tumor effects by inhibiting autophagy, and that the combination of Cin and PD-L1 blockade has synergistic anti-tumor effects. The data demonstrates the significant clinical potential of Cin in lung cancer treatment.

Abstract Background Multi-omics technology provides a good tool to analyze the protein toxin composition and search for the potential pathogenic factors of Solenopsis invicta, under the great harm of the accelerated invasion in southern China. Methods Species collection, functional annotation, toxin screening, and 3D modeling construction of three interested toxins were performed based on the successfully constructed transcriptome and proteome of S. invicta. Results A total of 33,231 unigenes and 721 proteins were obtained from the constructed transcriptome and proteome, of which 9,842 (29.62%) and 4,844 (14.58%) unigenes, as well as 469 (65.05%) and 71 (99.45%) proteins were annotated against the databases of Gene Ontology and Kyoto Encyclopedia of Genes and Genomes, respectively. After comparing with the uniprot toxin database, a total of 316 unigenes and 47 proteins (calglandulin, venom allergen 3, and venom prothrombin activator hopsarin-D, etc.) were successfully screened. Conclusions The update of annotations at the transcriptome and proteome levels presents a progression in the comprehension of S. invicta in China. We also provide a protein toxin list that could be used for further exploration of toxicity as well as its antagonistic strategy by S. invicta.

Abstract Background To construct a predictive model of immunotherapy efficacy for patients with lung squamous cell carcinoma (LUSC) based on the degree of tumor-infiltrating immune cells (TIIC) in the tumor microenvironment (TME). Methods The data of 501 patients with LUSC in the TCGA database were used as a training set, and grouped using non-negative matrix factorization (NMF) based on the degree of TIIC assessed by single-sample gene set enrichment analysis (GSEA). Two data sets (GSE126044 and GSE135222) were used as validation sets. Genes screened for modeling by least absolute shrinkage and selection operator (LASSO) regression and used to construct a model based on immunophenotyping score (IPTS). RNA extraction and qPCR were performed to validate the prognostic value of IPTS in our independent LUSC cohort. The receiver operating characteristic (ROC) curve was constructed to determine the predictive value of the immune efficacy. Kaplan–Meier survival curve analysis was performed to evaluate the prognostic predictive ability. Correlation analysis and enrichment analysis were used to explore the potential mechanism of IPTS molecular typing involved in predicting the immunotherapy efficacy for patients with LUSC. Results The training set was divided into a low immune cell infiltration type (C1) and a high immune cell infiltration type (C2) by NMF typing, and the IPTS molecular typing based on the 17-gene model could replace the results of the NMF typing. The area under the ROC curve (AUC) was 0.82. In both validation sets, the IPTS of patients who responded to immunotherapy were significantly higher than those who did not respond to immunotherapy (P = 0.0032 and P = 0.0451), whereas the AUC was 0.95 (95% CI = 1.00–0.84) and 0.77 (95% CI = 0.58–0.96), respectively. In our independent cohort, we validated its ability to predict the response to cancer immunotherapy, for the AUC was 0.88 (95% CI = 1.00–0.66). GSEA suggested that the high IPTS group was mainly involved in immune-related signaling pathways. Conclusions IPTS molecular typing based on the degree of TIIC in the TME could well predict the efficacy of immunotherapy in patients with LUSC with a certain prognostic value.

Background: Triple-negative breast cancer (TNBC) is one of the most prominent neoplasm disorders and lacks efficacious treatments yet. Luteolin (3′,4′,5,7-tetrahydroxyflavone), a natural flavonoid commonly presented in plants, has been reported to delay the progression of TNBC. However, the precise mechanism is still elusive. We aimed to elucidate the inhibition and molecular regulation mechanism of luteolin on TNBC.Methods: The effects of luteolin on the biological functions of TNBC cells were first evaluated using the corresponding assays for cell counting kit-8 assay, flow cytometry, wound-healing assay, and transwell migration assay, respectively. The mechanism of luteolin on TNBC cells was then analyzed by RNA sequencing and verified by RT-qPCR, Western blot, transmission electron microscopy, etc. Finally, in vivo mouse tumor models were constructed to further confirm the effects of luteolin on TNBC.Results: Luteolin dramatically suppressed cell proliferation, invasion, and migration while favoring cell apoptosis in a dose- and time-dependent manner. In TNBC cells treated with luteolin, SGK1 and AKT3 were significantly downregulated while their downstream gene BNIP3 was upregulated. According to the results of 3D modeling, the direct binding of luteolin to SGK1 was superior to that of AKT3. The inhibition of SGK1 promoted FOXO3a translocation into the nucleus and led to the transcription of BNIP3 both in vitro and in vivo, eventually facilitating the interaction between BNIP3 and apoptosis and autophagy protein. Furthermore, the upregulation of SGK1, induced by luteolin, attenuated the apoptosis and autophagy of the TNBC.Conclusion: Luteolin inhibits TNBC by inducing apoptosis and autophagy through SGK1-FOXO3a-BNIP3 signaling.

IntroductionRecent studies have demonstrated the effectiveness of Gonadotropin-releasing hormone (GnRH) in inhibiting testicular growth and development in male animals to achieve castration while improving the meat quality of various livestock species, including cattle, sheep, goats, and pigs.MethodsIn this research, a GnRH-Th vaccine was synthesized using the Fmoc solid-phase synthesis technique, and the T helper (Th) antigen was modified with palmitic acid to improve its efficacy. The vaccine was then coated with a water-in-oil-in-water adjuvant to improve stability and safety. After passing safety and stability tests, the vaccine was administered to 13-week-old boars.ResultsThe results showed that it was stable, safe, and effective for up to 15 months. Moreover, the vaccine did not negatively affect the growth rate and body weight of the pigs. The palmitic acid-modified “GnRH-Th epitope peptide immunocastration vaccine (Water-in-Oil-in-Water (W/O/W)) effectively reduced the testosterone concentration and achieved castration. The concentration of androstenone and skatole hormones significantly decreased, leading to improved meat quality in the boars. The impact of extreme climate change on terrestrial ecosystems continues to intensify. This study was conducted to understand extreme climate–vegetation interactions under exacerbated frequency, severity, and duration of extreme climatic events. The Inner Mongolian Plateau (IMP) was selected due to its sensitive natural location, which is particularly vulnerable to climate change. Based on the Normalized Difference Vegetation Index (NDVI) and daily meteorological station data from 1982 to 2020, changes in the patterns of vegetation and extreme climate in the three ecological zones (forest, steppe, and desert steppe) of the IMP were identified. Furthermore, the effects of extreme climate on vegetation were quantified using correlation analysis and a geographical detector. The results showed that the annual NDVI of 95.1%, 50.6%, and 19.5% of the area increased significantly in the forest, steppe, and desert steppe, respectively. The Tx90p (warm days) and Tn90p (warm nights) increased significantly at the rate of 0.21 and 0.235 day·yr−1, respectively, while the Tx10p (cold days) and Tn10p (cold nights) showed a significantly decreasing trend at the rate of −0.105 and −0.117 day·yr−1. An extreme warming phenomenon was observed in all extreme temperature indices on the IMP. The results of both the correlation analysis and factor detector indicated that extreme temperature intensity and frequency greatly affected forest vegetation. In contrast, extreme precipitation intensity and frequency were relatively more important to the vegetation of the desert steppe. The lag in NDVI response to extreme temperature intensity was not less than three months in the IMP; however, extreme precipitation intensity exhibited a two-month time lag in the NDVI. This study can improve our understanding of extreme climate–vegetation interactions, provide theoretical support for disaster mitigation, and aid in understanding the ecological environment of the IMP.

Abstract Drought has a great impact on global terrestrial ecosystems. A large number of studies have shown that the impact of drought on vegetation growth has a lagged and cumulative effect, but it is unclear how much it contributes to different vegetation types. Therefore, based on the standardized precipitation evapotranspiration index (SPEI) base version 2.5 and the Global Inventory Monitoring and Modeling System (GIMMS3g) normalized difference vegetation index (NDVI) datasets, this study aimed to analyze the response process of different vegetation types to the cumulative and lagged effects of drought in the Mongolian Plateau during 1982–2015 using Pearson correlation and the Mann–Kendall mutation method and deeply explore the magnitude of the contribution of drought cumulative and lagged effects on vegetation using the multiple regression method. Our results show that, from 1982 to 2015 as a whole, NDVI showed an insignificant increasing trend, and SPEI had a significant mutation in 1998 and showed an insignificant increasing trend before and after 1998. Before 1998, the cumulative months were shorter (1–3 months) in the central steppe and agricultural vegetation zones, and the lagged months were longer (10–12 months) in the southeastern steppe and northeastern forest zones; after 1998, the cumulative months of NDVI increased (7–12 months) and the lag months decreased (3–8 months) in most vegetation zones. A comparison of the contribution of drought accumulation and lag to NDVI revealed that the main driver of NDVI has shifted from lagged to cumulative effect.

Kimura disease (KD) is a rare and benign chronic inflammatory disease of unknown cause. It is characterized by subcutaneous granuloma of soft tissues in the head and neck region, increased eosinophil count, and elevated serum IgE. Currently, no definitive treatments are recommended. A 57-year-old Chinese man was diagnosed with KD after 7 years of slow subcutaneous masses growth. The patient underwent treatment of oral glucocorticoids for 1 year, but the masses recurred as the dosage was tapered down. Subsequent anti-IgE therapy of omalizumab administered subcutaneously at 450 mg/day at a 4-week interval did not show improvement. The size of masses and serum IgE and circulating eosinophils did not decrease significantly after 19 cycles of continuous treatment. Ultimately, switched strategy of dupilumab was applied at an initial dose of 600 mg, followed by 300 mg every 2 weeks for 4 months. This treatment demonstrated dramatical effects with reduced masses in each area and fast dropdown of eosinophil counts, while the high level of serum IgE remained without changes. Recently, different biologics including anti-IgE, anti-IL-5, and anti-IL-4/IL-13 have been applied to treat KD with satisfied results and help to explore the pathogenesis of this rare disease. To our knowledge, this is the first report that demonstrates the effects of two different biologics in the same patient and reveals the impressive clinical efficacy of dupilumab to treat KD independent of IgE. Therefore, further investigation of the underlying mechanism and the development of diagnosis and treatment of KD is valuable.

Histone deacetylase 1 (HDAC1) is a unique member of the classes I HDACs and helps to regulate acute and chronic adaptation to environmental stimuli such as allergen, stress. Allergic diseases are complex diseases resulting from the effect of multiple genetic and interacting foreign substances. Epigenetics play an important role in both pathological and immunomodulatory conditions of allergic diseases. To be consistent with this role, recent evidence strongly suggests that histone deacetylase 1 (HDAC1) plays a critical role in allergic response. HDAC1 expression is stimulated by allergen and attributes to increase T helper 2 (Th2) cytokine levels, decrease Th1/Th17 cells and anti-inflammatory cytokine Interleukin-10 (IL-10), and TWIK-related potassium channel-1 (Trek-1) expression. This review focuses on the contribution of HDAC1 and the regulatory role in characterizing allergic endotypes with common molecular pathways and understanding allergic multimorbidity relationships, as well as addressing their potential as therapeutic targets for these conditions.

Background: Cervical cancer (CC), as a serious menace to the health of women, has long been one of the most lethal gynecologic neoplasms throughout the world. Long non-coding RNA (LncRNA) NR2F1-AS1 has been documented to exert crucial functions in many malignant tumors. Nonetheless, the function and molecular mechanism of NR2F1-AS1 in CC remain completely unknown. Objective: This study aimed to explore the function and molecular mechanism of NR2F1-AS1 in CC. Methods: The expression levels of NR2F1-AS1, miR-642a-3p, NR2F1 in CC tissues, and cell lines were examined by reverse transcription real-time quantitative polymerase chain reaction. Cell viability, proliferation, migration, and invasion were detected by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide, colony formation and Transwell assays. The protein levels of epithelial-mesenchymal transition markers and NR2F1 in CC cells were assessed by Western blot analysis. The correlations among NR2F1-AS1, miR-642a-3p, and NR2F1 were estimated through luciferase reporter and RNA immunoprecipitation assays. Results: NR2F1-AS1 expression was clearly downregulated in CC tissues and cell lines. Molecular mechanistic experiments showed that NR2F1-AS1 overexpression upregulated NR2F1 expression in CC cells by directly binding to miR-642a-3p, and inhibiting by this way cell viability, proliferation, migration, and invasion in CC. Rescue assays showed that NR2F1 knockdown or miR-642a-3p overexpression offset NR2F1-AS1 upregulation-induced inhibition on CC cell malignant phenotypes. Conclusion: These findings revealed that NR2F1-AS1 played a tumor suppressor role in CC by mediating the miR-642a-3p/NR2F1 axis.

The role of N7-methylguanosine(m7G)-related miRNAs in lung adenocarcinoma (LUAD) remains unclear. We used LUAD data from The Cancer Genome Atlas (TCGA) to establish a risk model based on the m7G-related miRNAs, and divided patients into high-risk or low-risk subgroups. A nomogram for predicting overall survival (OS) was then constructed based on the independent risk factors. In addition, we performed a functional enrichment analysis and defined the oxidative stress-related genes, immune landscape as well as a drug response profile in the high-risk and low-risk subgroups. This study incorporated 28 m7G-related miRNAs into the risk model. The data showed a significant difference in the OS between the high-risk and low-risk subgroups. The receiver operating characteristic curve (ROC) predicted that the area under the curve (AUC) of one-year, three-year and five-year OS was 0.781, 0.804 and 0.853, respectively. The C-index of the prognostic nomogram for predicting OS was 0.739. We then analyzed the oxidative stress-related genes and immune landscape in the high-risk and low-risk subgroups. The data demonstrated significant differences in the expression of albumin (ALB), estimated score, immune score, stromal score, immune cell infiltration and functions between the high-risk and low-risk subgroups. In addition, the drug response analysis showed that low-risk subgroups may be more sensitive to tyrosine kinase inhibitor (TKI) and histone deacetylase (HDAC) inhibitors. We successfully developed a novel risk model based on m7G-related miRNAs in this study. The model can predict clinical prognosis and guide therapeutic regimens in patients with LUAD. Our data also provided new insights into the molecular mechanisms of m7G in LUAD.

Aims: To evaluate the utility of fasudil in a rat model of contrast-associated acute kidney injury (CA-AKI) and explore its underlying mechanism through multiparametric renal magnetic resonance imaging (mpMRI).Methods: Experimental rats (n = 72) were grouped as follows: controls (n = 24), CA-AKI (n = 24), or CA-AKI + Fasudil (n = 24). All animals underwent two mpMRI studies (arterial spin labeling, T1 and T2 mapping) at baseline and post iopromide/fasudil injection (Days 1, 3, 7, and 13 respectively). Relative change in renal blood flow (ΔRBF), T1 (ΔT1) and T2 (ΔT2) values were assessed at specified time points. Serum levels of cystatin C (CysC) and interleukin-1β (IL-1β), and urinary neutrophil gelatinase-associated lipocalin (NGAL) concentrations were tested as laboratory biomarkers, in addition to examining renal histology and expression levels of various proteins (Rho-kinase [ROCK], α-smooth muscle actin [α-SMA]), hypoxia-inducible factor-1α (HIF-1α), and transforming growth factor-β1 (TGF-β1) that regulate renal fibrosis and hypoxia.Results: Compared with the control group, serum levels of CysC and IL-1β, and urinary NGAL concentrations were clearly increased from Day 1 to Day 13 in the CA-AKI group (all p < 0.05). There were significant reductions in ΔT2 values on Days 1 and 3, and ΔT1 reductions were significantly more pronounced at all time points (Days 1–13) in the CA-AKI + Fasudil group (vs. CA-AKI) (all p < 0.05). Fasudil treatment lowered expression levels of ROCK-1, and p-MYPT1/MYPT1 proteins induced by iopromide, decreasing TGF-β1 expression and suppressing both extracellular matrix accumulation and α-SMA expression relative to untreated status (all p < 0.05). Fasudil also enhanced PHD2 transcription and inhibition of HIF-1α expression after CA-AKI.Conclusions: In the context of CA-AKI, fasudil appears to reduce renal hypoxia, fibrosis, and dysfunction by activating (Rho/ROCK) or inhibiting (TGF-β1, HIF-1α) certain signaling pathways and reducing α-SMA expression. Multiparametric MRI may be a viable noninvasive tool for monitoring CA-AKI pathophysiology during fasudil therapy.

ObjectiveThis study was conducted in order to establish a long non-coding RNA (lncRNA)-based model for predicting overall survival (OS) in patients with lung adenocarcinoma (LUAD).MethodsOriginal RNA-seq data of LUAD samples were extracted from The Cancer Genome Atlas (TCGA) database. Univariate Cox survival analysis was performed to select lncRNAs associated with OS. The least absolute shrinkage and selection operator (LASSO) regression analysis and multivariate Cox analysis were performed for building an OS-associated lncRNA prognostic model. Moreover, receiver operating characteristic (ROC) curves were generated to assess predictive values of the hub lncRNAs. Consequently, qRT-PCR was conducted to validate its prognostic value. The potential roles of these lncRNAs in immunotherapy and anti-angiogenic therapy were also investigated.ResultsThe lncRNA-associated risk score of OS (LARSO) was established based on the LASSO coefficient of six individual lncRNAs, including CTD-2124B20.2, CTD-2168K21.1, DEPDC1-AS1, RP1-290I10.3, RP11-454K7.3, and RP11-95M5.1. Kaplan–Meier analysis revealed that LUAD patients with higher LARSO values had a shorter OS. Furthermore, a new risk score (NRS), including LARSO, stage, and N stage, could better predict the prognosis of LUAD patients compared with LARSO alone. Evaluation of the prognostic model in our cohort demonstrated that patients with higher scores had a worse prognosis. In addition, correlation analysis between these six lncRNAs and immune checkpoints or anti-angiogenic targets suggested that LUAD patients with high LARSO might not be sensitive to immunotherapy or anti-angiogenic therapy.ConclusionsThis robust six-lncRNA prognostic signature may be used as a novel and powerful prognostic biomarker for lung adenocarcinoma.

Abstract Background Wogonin, a natural flavonoid-like chemical compound, exhibits anti-inflammatory, antitumor, antiviral, neuroprotective, and anxiolytic effects by modulating a variety of cellular signaling pathways including PI3K-Akt, p53, nuclear factor κB (NF-κB), mitogen-activated protein kinase (MAPK) pathways. In this study, its antiviral effect against herpes simplex virus (HSV) type 1 and 2 (HSV-1 and HSV-2) replication was investigated. Results Wogonin suppressed HSV-2-induced cytopathic effect (CPE) and reduced viral mRNA transcription, viral protein synthesis, and infectious virion particle titers in a dose-dependent manner. A time-of-drug-addition assay demonstrated that wogonin acted as a postentry viral inhibitor. Wogonin also significantly reduced HSV-induced NF-κB and MAPK pathway activation, which has previously been demonstrated to be important for viral replication. Conclusions Our results suggest that the anti-herpes effect of wogonin may be mediated by modulation of cellular NF-κB and JNK/p38 MAPK pathways and imply that wogonin may be useful as an anti-HSV agent.

The just noticeable distortion (JND) model reveals the visibility limitation. Human eyes hold different attention and sensitivity to different regions owing to different contributions to the perceptual quality. In this paper, a superpixel-wise JND model based on region (RJND) is proposed. First, an image is segmented into superpixels by the simple linear iterative clustering (SLIC). Then, region color contrast is calculated for each region and foveation regions are selected for the image. Based on the human visual perception, a region weighting model is established by incorporating region color contrast and foveation regions modulation. Considering the contrast masking (CM) effect is not perfect, we introduce the texture coarseness combined with CM effect for a more accurate visual masking effect. Finally, a new region JND model is established by combining the region weighting model and the coarseness modulation. The experimental results demonstrate the proposed RJND model can decrease PSNR more efficiently compared with some existing JND models when achieving nearly the same subjective perceptual quality. It can remove much more visual redundancy.

Desertification is one of the most harmful ecological disasters on the Mongolian Plateau, placing the grassland ecological environment under great pressure. Remote-sensing monitoring of desertification and exploration of the drivers behind it are important for effectively combating this issue. In this study, four banners/counties on the border of China and Mongolia on the Mongolian Plateau were selected as the target areas. We explored desertification dynamics and their drivers by using remote sensing imagery and a product dataset for the East Ujimqin Banner and three counties in Mongolia during the period 2000–2015. First, remote sensing information on desertification in the fourth phase of the study area was extracted using the visual interpretation method. Second, the dynamic change characteristics of desertification were analyzed using the intensity analysis method. Finally, the drivers of desertification and their explanatory powers were identified using the geographical detector method. The results show that the desertification of the East Ujimqin Banner has undergone a process of reversion, development, and mild development, with the main transition occurring between slight (SL) and non-desertified land (N), very serious desertified land (VS), and water areas. The dynamics of desertification in this region are influenced by a combination of natural and anthropogenic factors. Desertification in the three counties of Mongolia has undergone processes of development, mild development and mild development with SL and vs. as the main types. Desertification in Mongolia is mainly concentrated in Matad County, which is greatly affected by natural conditions and has little impact from anthropogenic activities. In addition, the change intensity of desertification dynamics in the study area showed a decreasing trend, and the interaction between natural and anthropogenic drivers could enhance the explanatory power of desertification dynamics. The research results provide a scientific basis for desertification control, ecological protection, and ecological restoration on the Mongolian Plateau.

In recent decades, frequent compound dry and hot events have posed a great threat to humans and the ecological environment, especially in Inner Mongolia, which has arid and semi-arid characteristics. In this study, monthly temperature and precipitation data from 115 meteorological stations in Inner Mongolia from 1982 to 2020 were used to establish a standardized dry and hot index (SDHI). Theil–Sen median trend analysis, Mann–Kendall test, partial correlation analysis, and stepwise multiple regression models were used to characterize the changes in compound dry and hot events and the normalized difference vegetation index (NDVI) from 1982 to 2020, and the relationship between the SDHI and NDVI was quantitatively evaluated. The results showed that the overall SDHI values in Inner Mongolia showed a significant decrease at a rate of 0.03/year from 1982 to 2020, indicating an increase in the severity of compound dry and hot events. NDVI values showed a significant increasing trend and NDVI showed mutated 2001. Among the grassland vegetation types, SDHI and NDVI trends were more significant in forests, and meadow steppe, desert steppe, and desert were more susceptible to compound dry and hot events, and forests had the greatest severity of compound dry and hot events. The results of the partial correlation analysis showed that the average value of the partial correlation coefficient between the SDHI and NDVI was 0.68, and the area of positive correlation was 84.13%. Spatially, it showed strong response characteristics in the middle and gradual weakening towards the east and west sides. The correlation between NDVI and climatic conditions varied greatly in different vegetation areas. The forest area is most sensitive to the influence of temperature, and the desert steppe area is most affected by compound dry and hot events. The overall vegetation growth in Inner Mongolia was most affected by temperature conditions, followed by compound dry and hot conditions, and the influence of drought conditions was the least significant. The results of the relative importance analysis confirmed this. The research results provide a more detailed understanding of compound dry and hot events in arid and semi-arid regions and useful insights and support for ecological protection.

Background: To evaluate the application of blood oxygenation level-dependent (BOLD)imaging and intravoxel incoherent motion (IVIM) magnetic resonance imaging (MRI) on assessing early contrast-induced acute kidney injury (CIAKI). Materials: Sixty rabbits were randomly chosen to undergo iohexol (1.0, 2.5, and 5.0 [gI/kg], respectively; n = 15 for each group) or saline injection (n = 15). In each group, 6 rabbits underwent MRI at 24 h before injection and after injection of iohexol or saline (1 h and 1, 2, 3, and 4 days); meanwhile, out of the remaining 9 rabbits, 3 were chosen for MRI acquisition, and then they were killed at specific time points (1 h, 1 day, and 3 days, respectively). Results: The strong attenuation of pure molecular diffusion (D), apparent diffusion coefficient (ADC), and perfusion fraction (f) was observed at 1 day, while pseudodiffusion coefficient (D*) showed a significant decrease at 1 h after iohexol injection. A distinct elevation of apparent transverse relaxation rate (R2*) reached the maximum levels on day 1, which was consistent with the expression of hypoxia-inducible factor-1α and vascular endothelial growth factor. ADC, D, and R2* correlated well with histopathological parameters and biochemical parameters. Conclusion: BOLD combined with IVIM is effective to monitor renal pathophysiology associated with CIAKI.

Since a large proportion of the information that is received daily is in the form of images, a highly effective no-reference stereo image quality assessment (SIQA) method is desired. This paper proposes an improved method that covers wide quality-aware features, including the structure, color, luminance, phase, and human visual system (HVS). To be specific, since human eyes are highly sensitive to the structure of images, the gradient magnitude (GM) and gradient orientation (GO) are extracted from the left and right views of the stereo image. Considering the influence of color distortions, the images are decomposed into the RGB channels to be processed, and the local gradient of the color image is obtained by adding up the RGB gradient vectors. In addition, according to the study of the two main visual channels, especially the cyclopean and disparity maps, the binocular-related images of position and phase congruency are generated. Correspondingly, two special dictionaries for the gradient and phase are trained to parse the high-dimensional sample sets. The experimental results show that the proposed metric always achieves high consistency with human subjective assessments for both symmetric and asymmetric distortions.

Objectives In this preclinical investigation, the feasibility of using diffusion tensor imaging (DTI) to study contrast-induced acute kidney injury (CIAKI) is explored, comparing radiographic outcomes with histopathologic and immunohistochemical findings after repeated animal exposures to iodinated contrast agent. Materials and Methods Forty-five male wistar rats were allocated to three groups (n = 15 each), each receiving two separate injections 1 day apart: group 1 (iodixanol then saline); group 2 (iodixanol twice); and control group (saline twice). Five rats were then randomly selected from each group at three separate time points (1 h, 24 h, and 120 h) for magnetic resonance imaging (MRI). Upon MRI completion, the animals were sacrificed, examining renal tissue and serum creatinine level. DTI data served to calculate fractional anisotropy (FA) and apparent diffusion coefficient (ADC). Results FA values were significantly lower in group 2 than in the others. Compared with controls, FA assessments at 1 h, 24 h, and 120 h after injections commenced were significantly lower in group 2; and ADC was significantly more pronounced at 24 h. Serum creatinine levels at 24 h were markedly elevated in both groups 1 and 2. Pearson correlation analysis revealed significant negative correlations between FA (r = −0.730; p < 0.05) or ADC (r = −0.827; p < 0.05) and tubular injury and between FA (r = −0.563; p < 0.05) or ADC (r = −0.805; p < 0.05) and hypoxia-inducible factor-1α. Conclusions Analytic approaches to DTI with better reproducibility should aid in monitoring the early pathophysiologic derangements of CIAKI, thus facilitating timely reversal of the detrimental effects.

Abstract Background Epidemiological studies have suggested that noise exposure may increase the risk of type 2 diabetes mellitus (T2DM), and experimental studies have demonstrated that noise exposure can induce insulin resistance in rodents. The aim of the present study was to explore noise-induced processes underlying impaired insulin sensitivity in mice. Methods Male ICR mice were randomly divided into four groups: a control group without noise exposure and three noise groups exposed to white noise at a 95-dB sound pressure level for 4 h/day for 1, 10, or 20 days (N1D, N10D, and N20D, respectively). Systemic insulin sensitivity was evaluated at 1 day, 1 week, and 1 month post-noise exposure (1DPN, 1WPN, and 1MPN) via insulin tolerance tests (ITTs). Several insulin-related processes, including the phosphorylation of Akt, IRS1, and JNK in the animals’ skeletal muscles, were examined using standard immunoblots. Biomarkers of inflammation (circulating levels of TNF-α and IL-6) and oxidative stress (SOD and CAT activities and MDA levels in skeletal muscles) were measured via chemical analyses. Results The data obtained in this study showed the following: (1) The impairment of systemic insulin sensitivity was transient in the N1D group but prolonged in the N10D and N20D groups. (2) Noise exposure led to enhanced JNK phosphorylation and IRS1 serine phosphorylation as well as reduced Akt phosphorylation in skeletal muscles in response to exogenous insulin stimulation. (3) Plasma levels of TNF-α and IL-6, CAT activity, and MDA concentrations in skeletal muscles were elevated after 20 days of noise exposure. Conclusions Impaired insulin sensitivity in noise-exposed mice might be mediated by an enhancement of the JNK/IRS1 pathway. Inflammation and oxidative stress might contribute to insulin resistance after chronic noise exposure.

With the pervasiveness of mobile devices, satisfying spatial-temporal coverage requirements in interested regions while considering the quality of the sensing data and the budget constraint is a major research challenge in mobile crowdsensing (MCS). In this paper, we define a novel coverage metric, quality coverage, which considers both the fraction of subareas covered by sensor readings and the quality of sensing data in each covered subarea, for diversified location-based MCS tasks. In our system, as participants with high reliability level can contribute a lot to sensing tasks, we design an incentive model that provides higher bonus to more reliable participants. After knowing the reward for the per amount of sensing data, each worker can submit an interested task set that contains several unit tasks. The platform assigns MCS tasks to these mobile workers by solving the task allocation optimization problems, which select the optimal mobile worker set under the budget of data requesters to maximize diverse and spatial coverage level in the whole sensing area. As the task allocation problems are proven to be NP-hard, we propose a heuristic and a polynomialtime greedy approximation algorithm to solve optimization problems in two different application scenarios. Simulations using both real and synthetic data sets show that our algorithms outperform existing approaches.

Abstract Background Condyloma acuminatum, infected by low-risk human papillomaviruses (e.g., HPV6 and HPV11), is one of the most widespread sexually transmitted diseases. Apolipoprotein B mRNA-editing enzyme catalytic polypeptide-like 3 proteins (APOBEC3s, A3s) are cellular cytidine deaminases acting as antiviral factors through hypermutation of viral genome. However, it remains unknown whether A3s results in HPV11 gene mutations and interferon-ω (IFN-ω) exhibits antiviral activities through the A3s system. Here we investigated whether enhanced APOBEC3A (A3A) resulted in the E6 gene mutations and explore the effects of recombinant human interferon-ω (rhIFN-ω) on A3s/E6 expression in HaCaT keratinocytes containing the genome of HPV 11 (HPV11.HaCaT cells). Methods A3A-overexpressed HPV11.HaCaT (A3A-HPV11.HaCaT) cells were established by lentiviral infection and verified by immunofluorescence and western-blotting. Cell cycle, E6 gene mutations, APOBEC3s/E6 gene expression and subcellular localization were detected by FACS, 3D-PCR and sequencing, qRT-PCR and immunofluorescence respectively. Results The results suggested that A3A-HPV11.HaCaT cells were successfully established. Enhanced A3A induced S-phase arrest, G > A/C > T mutations and obvious reduction of E6 mRNA expression. A3A/A3B mRNA expression was up-regulated at 6 h and 12 h and obvious A3A staining existed throughout HPV11.HaCaT cells after rhIFN-ω treatment. RhIFN-ω could also inhibit mRNA expression of HPV11 E6 significantly. Conclusions Enhanced A3A repressed HPV11 E6 expression through gene hypermutation, and rhIFN-ω might be an effective agent against HPV11 infection by up-regulation of A3A.

Inner Mongolia in China is a typically arid and semi-arid region with vegetation prominently affected by global warming and human activities. Therefore, investigating the past and future vegetation change and its impact mechanism is important for assessing the stability of the ecosystem and the ecological policy formulation. Vegetation changes, sustainability characteristics, and the mechanism of natural and anthropogenic effects in Inner Mongolia during 2000–2019 were examined using moderate resolution imaging spectroradiometer normalized difference vegetation index (NDVI) data. Theil–Sen trend analysis, Mann–Kendall method, and the coefficient of variation method were used to analyze the spatiotemporal variability characteristics and sustained stability of the NDVI. Furthermore, a trend estimation method based on a Seasonal Trend Model (STM), and the Hurst index was used to analyze breakpoints and change trends, and predict the likely future direction of vegetation, respectively. Additionally, the mechanisms of the compound influence of natural and anthropogenic activities on the vegetation dynamics in Inner Mongolia were explored using a Geodetector Model. The results show that the NDVI of Inner Mongolia shows an upward trend with a rate of 0.0028/year (p < 0.05) from 2000 to 2019. Spatially, the NDVI values showed a decreasing trend from the northeast to the southwest, and the interannual variation fluctuated widely, with coefficients of variation greater than 0.15, for which the high-value areas were in the territory of the Alxa League. The areas with increased, decreased, and stable vegetation patterns were approximately equal in size, in which the improved areas were mainly distributed in the northeastern part of Inner Mongolia, the stable and unchanged areas were mostly in the desert, and the degraded areas were mainly in the central-eastern part of Inner Mongolia, it shows a trend of progressive degradation from east to west. Breakpoints in the vegetation dynamics occurred mainly in the northwestern part of Inner Mongolia and the northeastern part of Hulunbuir, most of which occurred during 2011–2014. The future NDVI trend in Inner Mongolia shows an increasing trend in most areas, with only approximately 10% of the areas showing a decreasing trend. Considering the drivers of the NDVI, we observed annual precipitation, soil type, mean annual temperature, and land use type to be the main driving factors in Inner Mongolia. Annual precipitation was the first dominant factor, and when these four dominant factors interacted to influence vegetation change, they all showed interactive enhancement relationships. The results of this study will assist in understanding the influence of natural elements and human activities on vegetation changes and their driving mechanisms, while providing a scientific basis for the rational and effective protection of the ecological environment in Inner Mongolia.

Resveratrol (Res) is a multi-functional polyphenol compound that has protective functions in acute kidney diseases. Here, we examined whether the resveratrol could ameliorate post-contrast acute kidney injury (PC–AKI) following diabetic nephropathy (DN), and explored any underlying mechanism(s) in vivo and in vitro. Twenty-four rabbits with DN were randomly divided into four groups: control (Cont), resveratrol (Res), iohexol (PC–AKI), and resveratrol plus iohexol (Res+PC–AKI) groups. Functional magnetic resonance imaging, renal histology, blood and urinary biomarkers, silent information regulator l (SIRT1), peroxisome proliferator-activated receptor gamma coactivator-1 alpha (PGC-1α), hypoxia-inducible transcription factor-1α (HIF-1α), and apoptosis-associated protein expression were assessed ex vivo. For in vitro experiments, renal tubular epithelial (HK-2) cells subjected to high glucose conditions were treated with resveratrol, Ex527, an SIRT1 inhibitor, or 2-methoxyestradiol (2-MeOE2), HIF-1α inhibitor, before treatment with iohexol. With regard to the rabbit model of acute renal injury in DN, compared to the PC–AKI group, the Res+PC–AKI group showed decreased levels of cystatin C and urinary neutrophil gelatinase-associated lipocalin, increased pure molecular diffusion (D) and the fraction of water flowing in capillaries (f), a decreased apparent relaxation rate (R2*), renal injury score and apoptosis rate, increased protein expression levels of SIRT1 and PGC-1α, and decreased levels of HIF-1α and apoptosis-associated protein. In addition, iohexol decreased HK-2 cell survival and increased the cell apoptosis rate; results were reversed after treating cells with resveratrol. Resveratrol reduced renal hypoxia, mitochondrial dysfunction and renal tubular cell apoptosis by activating SIRT1–PGC–1α–HIF-1α signaling pathways in PC–AKI with DN.

In recent years, global warming and intense human activity have been responsible for significantly altering vegetation dynamics on the Mongolian Plateau. Understanding the long-term vegetation dynamics in this region is important to assess the impact of these changes on the local ecosystem. Long-term (1982–2015), satellite-derived normalized difference vegetation index (NDVI) datasets were used to analyse the spatio-temporal patterns of vegetation activities using linear regression and the breaks for additive season and trend methods. The links between these patterns and changes in temperature, precipitation (PRE), soil moisture (SM), and anthropogenic activity were determined using partial correlation analysis, the residual trends method, and a stepwise multiple regression model. The most significant results indicated that air temperature and potential evapotranspiration increased significantly, while the SM and PRE had markedly decreased over the past 34 years. The NDVI dataset included 71.16% of pixels showing an increase in temperature and evaporation during the growing season, particularly in eastern Mongolia and the southern border of the Inner Mongolia Autonomous region, China. The proportion indicating the breakpoint of vegetation dynamics was 71.34% of pixels, and the trend breakpoints mainly occurred in 1993, 2003, and 2010. The cumulative effects of PRE and SM in the middle period, coupled with the short-term effects of temperature and potential evapotranspiration, have had positive effects on vegetation greening. Anthropogenic factors appear to have positively impacted vegetation dynamics, as shown in 81.21% of pixels. We consider rapid economic growth, PRE, and SM to be the main driving factors in Inner Mongolia. PRE was the main climatic factor, and combined human and livestock populations were the primary anthropogenic factors influencing vegetation dynamics in Mongolia. This study is important in promoting the continued use of green projects to address environmental change in the Mongolian Plateau.

Epidemiological studies have revealed that noise exposure was associated with an increased risk of type 2 diabetes mellitus (T2DM). However, the exact nature of that association remains to be elucidated. The present study is designed to examine the effects of chronic noise exposure on the development of T2DM in combination with a high-fat-diet (HFD) in mice. Here we show that chronic noise exposure at 85 dB SPL (4 h /day, below the safety limit for occupational noise exposure) exaggerated multiple metabolic abnormalities induced by HFD in C57BL/6J male mice, including worsened glucose intolerance, insulin resistance, fasting hyperglycemia and dyslipidemia. Furthermore, noise exposure exhibited a paradoxical impact on fat accumulation and circulating levels of free fatty acid, indicating a potential stimulating effect of noise on lipolysis. These results provide first in vivo supporting evidence for the causative role of noise exposure in diabetogenesis and pinpoint a noise-associated increase in blood free fatty acid levels as a possible mediator accelerating the effect of noise on the development of insulin resistance and T2DM.

Agricultural flood disaster risk assessment plays a vital role in agricultural flood disaster risk management. Extreme precipitation events are the main causes of flood disasters in the Midwest Jilin province (MJP). Therefore, it is important to analyse the characteristics of extreme precipitation events and assess the flood risk. In this study, the Multifractal Detrended Fluctuation Analysis (MF-DFA) method was used to determine the threshold of extreme precipitation events. The total duration of extreme precipitation and the total extreme precipitation were selected as flood indicators. The copula functions were then used to determine the joint distribution to calculate the bivariate joint return period, which is the flood hazard. Historical data and flood indicators were used to build an agricultural flood disaster vulnerability surface model. Finally, the risk curve for agricultural flood disasters was established to assess the flood risk in the MJP. The results show that the proposed approaches precisely describe the joint distribution of the flood indicators. The results of the vulnerability surface model are in accordance with the spatiotemporal distribution pattern of the agricultural flood disaster loss in this area. The agricultural flood risk of the MJP gradually decreases from east to west. The results provide a firm scientific basis for flood control and drainage plans in the area.

Drought has an important influence on the hydrological cycle, ecological system, industrial and agricultural production, and social life. Based on the different time scales of characteristics of drought variability, the standardized precipitation evapotranspiration index (SPEI), a multi-timescale index with consideration of evaporation, was used in this study to estimate the spatial and temporal variability characteristics of drought. Climatic data from 15 meteorological stations across Horqin Sandy Land during 1964–2013 were used to calculate the SPEI of 1, 3, 6, and 12 months. In order to examine the relationship between droughts and other variables, 10 extreme climate indices were calculated based on the daily precipitation and maximum/mean/minimum temperature data of 15 meteorological stations, and linkages between SPEI-12 and atmosphere indices were established using by the cross wavelet transform method. The results indicated that the climate of Horqin Sandy Land had a tendency towards drought conditions, which is particularly apparent from the year 2000 onwards. During the study period, drought events were frequent in the region. Mild drought occurred in a quarter of the month, with that of moderate, severe, and extreme drought accounting for 0.11, 0.05, and 0.02 of the total months. The spatial trend of multi-timescale drought revealed that there was an increase in the severity of drought throughout Horqin Sandy Land, among which the magnitude in southern parts was larger than that of northern parts. The results also showed that the short time scale drought negatively correlated with precipitation extremes and positively correlated with temperature extremes. Furthermore, the long time scale drought (SPEI-12) was associated with atmosphere indices. Significant resonance periods were found between El Nino southern oscillation (ENSO), the East Asian summer monsoon index (EASMI), and SPEI-12.
