Schizophyllum commune (S. commune) fermented supernatant with added Radix Puerariae (SC-RP) showed significant antioxidant activity in our previous work. However, the possible lifespan and healthspan extending the capacity of Caenorhabditis elegans (C. elegans) and the underlying mechanism were not illuminated. In this study, the effect of SC-RP on extending the lifespan and improving stress resistance of C. elegans were examined. Additionally, the underlying lifespan extending molecular mechanisms of SC-RP were explored. Treated with SC-RP at 10 μg/mL, the lifespan of C. elegans increased by 24.89% (P < 0.01). Also, SC-RP prolonged the healthspan of the nematode, including reducing lipofuscin levels, improving mobility and enhancing resistance to oxidative stress and heat shock. Moreover, superoxide dismutase and catalase activities were increased for SC-RP treated C. elegans. Meantime the intracellular levels of thiobarbituric acid reactive substances (TBARS) and reactive oxygen species (ROS) were attenuated. Express levels of eight genes including daf-2, daf-16, sod-3, skn-1, gst-4, clk-1, age-1 and mev-1 were analyzed by RT-PCR method for possible C. elegan anti-aging mechanisms of SC-RP. Expression levels of key genes daf-2, gst-4 and sod-3 were up-regulated, while that of daf-16, skn-1, and clk-1 were down-regulated. The results suggest that SC-RP could extend the lifespan and healthspan of C. elegans significantly, and the IIS pathway, SKN-1/Nrf2 pathway and mitochondrial metabolism pathway were primarily considered associated. Thus, SC-RP is a potential component to improve aging and aging-related symptoms as new functional materials.

Amphiphilic polysaccharides can be used as wall materials and applied to encapsulate hydrophobic active chemicals; moreover, there is significant demand for novel medical high-molecular-weight materials with various functions. In order to prepare amphiphilic schizophyllan (SPG), octenyl succinic anhydride (OSA) was chosen to synthesize OSA-modified schizophyllan (OSSPG) using an esterified reaction. The modification of OSSPG was demonstrated through FT-IR and thermal analysis. Moreover, it was found that OSSPG has a better capacity for loading curcumin, and the loading amount was 20 μg/mg, which was 2.6 times higher than that of SPG. In addition, a hydrogel made up of PVA, borax, and C-OSSPG (OSSPG loaded with curcumin) was prepared by means of the one-pot method, based on the biological effects of curcumin and the immune-activating properties of SPG. The mechanical properties and biological activity of the hydrogel were investigated. The experimental results show that the dynamic cross-linking of PVA and borax provided the C-OSSPG/BP hydrogel dressing with exceptional self-healing properties, and it was discovered that the C-OSSPG content increased the hydrogel’s swelling and moisturizing properties. In fibroblast cell tests, the cells treated with hydrogel had survival rates of 80% or above. Furthermore, a hydrogel containing C-OSSPG could effectively promote cell migration. Due to the excellent anti-inflammatory properties of curcumin, the hydrogel also significantly reduces the generation of inflammatory factors, such as TNF-α and IL-6, and thus has a potential application as a wound dressing medicinal material.

This study was aimed at increasing the capacity of fermented Tremella fuciformis extracellular polysaccharides (TEPS) for possible functional food applications. Thus, strain varieties, fermentation parameters and purification conditions, and the in vitro antioxidant activities of purified EPS fractions were investigated. An EPS high-yield strain Tf526 was selected, and the effects of seven independent fermentation factors (time, temperature, initial pH, inoculum size, shaking speed, carbon, and nitrogen source) on the EPS yield were evaluated. By single factor optimization test, yeast extract and glucose were chosen as nitrogen sources and carbon sources, respectively, and with initial pH of 6.0, inoculum size of 8%, shaking speed of 150 rpm, and culture at 25 °C for 72 h, the optimal yield of TEPS reached 0.76 ± 0.03 mg/mL. Additionally, A-722MP resin showed the most efficient decoloration ratio compared to six other tested resins. Furthermore, optimal decoloration parameters of A-722MP resin were obtained as follows: decoloration time of 2 h, resins dosage of 2 g, and temperature of 30 °C. Decoloration ratio, deproteinization ratio, and polysaccharide retention ratio were 62.14 ± 2.3%, 81.21 ± 2.13%, and 73.42 ± 1.96%, respectively. Furthermore, the crude TEPS was extracted and four polysaccharide fractions were isolated and purified as Tf1-a, Tf1-b, Tf2, and Tf3 by the DEAE-Sepharose FF column and the Sephasryl S100 column. In general, the antioxidant activities of the Lf1-a and Lf1-b were lower compared with Vc at the concentration of 0.1 to 3 mg/mL, but the FRAP assay, DPPH scavenging activity, and hydroxyl radical scavenging activity analysis still revealed that Tf1-a and Tf1-b possess significant antioxidant activities in vitro. At the concentration of 3 mg/mL, the reducing power of Lf1-a and Lf1-b reached 0.86 and 0.70, the maximum DPPH radical were 54.23 ± 1.68% and 61.62 ± 2.73%, and the maximum hydroxyl radicals scavenging rates were 58.76 ± 2.58% and 45.81 ± 1.79%, respectively. Moreover, there were significant correlations (r > 0.8) among the selected concentrations and antioxidant activities of TEPS major fractions Tf1-a and Tf1-b. Therefore, it is expected that Tf1-a and Tf1-b polysaccharide fractions from fermented TEPS may serve as active ingredients in functional foods.

This paper reviews recent advances in the circulation dynamics of the Kuroshio and its interaction with shelf currents in the East China Sea (ECS). The annually averaged Kuroshio volume transport varies between 19 and 24 Sv, based on different observations, but there is no consensus on which season its volume transport peaks. The Kuroshio is intensified over the central slope of the ECS from that off the northeast of Taiwan. The total Kuroshio intrusion into the ECS shelf is estimated to be 1.3–1.4 Sv, deduced from the observed volume transport of exchange flow in the Taiwan and Tsushima Straits, based on the assumption of volume conversation over the shelf. However, the uncertainty regarding this estimation remains due to the absence of sufficient observations and understanding of the Kuroshio dynamics. The Kuroshio intrusions over the shelf off the northeast of Taiwan and southwest of Kyushu are stimulated by planetary or topographic β -effect associated with the alongshore variations in the ECS slope topography and altered by variations in the Kuroshio intensity, shear stress, and baroclinicity. Multilayered exchanges between the Kuroshio and shelf currents were found between 100- and 200-m isobaths along the central ECS slope. The spatial variations in these exchanges are governed by cross-isobath transport by geostrophy, whereas bottom Ekman transport may play a predominant role in altering the integrated exchange flow along the slope. Although the intrusion is greatly modulated along the path of the Kuroshio in the ECS by variable slope topography, there are few observations on the spatial variations of these exchange flows. The characteristics and variations in the circulation and hydrographic properties of waters between 100- and 200-m isobaths significantly determine the general ECS circulation, about which consensus has still not been attained.

Advances on the circulation in the Changjiang Estuary and adjacent East China Sea (ECS) and Yellow Sea (YS) coastal waters in the recent decades (2000–2020) are synthesized in this review. The circulation over the complicated bathymetry in the region is locally driven by winds, tides, as well as riverine discharge, and is remotely influenced by shelf currents between the 50 and 100-m isobaths through the cross-shelf exchanges. The interchange of the momentum and the freshwater pathway inside the Changjiang Estuary are jointly determined by tides and seasonally varying discharge and winds over the shelf. The buoyant waters are trapped inside the bulge that forms and expands over the shelf to the west of the 30-m isobath in the vicinity of Hangzhou Bay and the Changjiang Estuary. These buoyant waters are exported offshore by the shelf current, tidal mixing, and variations of wind patterns, forming the Changjiang River plume, which shows notable seasonality due to the reversal of both winds and shelf currents in the ECS and YS. Extensive spatial irregularities in the form of freshwater patches are present along its pathway to the Tsushima Strait in summer and to the Taiwan Strait in winter, respectively. Tides and the bathymetry irregularity have recently been found to play critical roles in determining the cross-shelf exchanges of water mass and momentum along the pathway of the ECS coastal current, and along this pathway, a year-round upslope intrusion of shelf waters appears in both summer and winter. Tides also play an important role in altering the expansion of the Changjiang River plume, cross-shelf extrusion of waters, and variation in the Yellow Sea Coastal Current over the shallow Subei Shoal.
