Klotho is an anti-aging, single-pass transmembrane protein found mainly in the kidney. Although aging is likely to be associated with DNA damage, the involvement of Klotho in protecting cells from DNA damage is still unclear. In this study, we examined DNA damage in human kidney cells and mouse kidney tissue after ionizing radiation (IR). The depletion and overexpression of Klotho in human kidney cells reduced and increased the cell survival rates after IR, respectively. The formation of γ-H2AX foci, representing DNA damage, was significantly elevated immediately after IR in cells with Klotho depletion and decreased in cells overexpressing Klotho. These results were confirmed in mouse renal tissues after IR. Quantification of DNA damage by a comet assay revealed that the Klotho knockdown significantly increased the amount of DNA damage immediately after IR, suggesting that Klotho protects chromosomal DNA from the induction of damage, rather than facilitating DNA repair. Consistent with this notion, Klotho was detected in both the nucleus and cytoplasm. In the nucleus, Klotho may serve to protect chromosomal DNA from damage, leading to its anti-aging effects.

A new earwig species of the genus Liparura Burr, 1907 of the family Forficulidae, namely L. chongqingensis sp. nov., is described from Chongqing, southwestern China. The new species is characterized by the male forceps basally approached to each other, and each with a small tooth protruding dorsad. A key to the species of Liparura is provided.

Animal genitalia have changed substantially and rapidly during evolution, and functionally interacting anatomical structures complementarily match between the sexes. Several hypotheses have been proposed to explain how such structure-matching evolved. A test of these hypotheses would require a detailed analysis of male and female genitalia among closely related species and a comparison of the functional aspects of the interacting structures between the sexes. Therefore, here we document genital coupling and copulatory wounds in the four species of the Drosophila auraria complex. The position of the protrusion of the median gonocoxite of males relative to the female terminalia differed among the species, which may reflect differences in protrusion morphology. Species-specific female structures were discovered on the membrane between the genitalia and analia and on the vaginal wall. The former makes contact with the protrusion, and the latter makes contact with appendages of the aedeagus. Copulatory wounds, which are produced during copulation, were seen at three locations on females: depressions near the genital orifice, the membrane between the genitalia and analia, and the vaginal wall. Some of the copulatory wounds were located at sites that could potentially make contact with the species-specific structures that we identified. We speculate that the female structures that differ between species of the D. auraria complex evolved in concert with the genitalia of male conspecifics.

Animals can show bias in their use of laterally paired organs that do not have any conspicuous anatomical differentiation between the right and left organs. Like right handedness in humans, males of the giant earwig Labidura riparia (Labiduridae: Labidurinae) preferentially (~90%) use the right one of their laterally paired penises for copulation. To elucidate the evolutionary origin of this lateralization, patterns of penis use were examined for the related species of the genus Nala (Labiduridae: Nalinae). In multiple populations and broods of both Nala lividipes and Nala nepalensis, males that were ready to use the right or left penis were equally frequent, providing a striking contrast to Labidura. Surgical ablation of one of the two penises revealed that both penises are functionally competent in N. lividipes. Nevertheless, each male almost consistently used only one of the paired penises, either the right or the left one. Changes in penis use were estimated to occur only once per 64–143 days per male. The present study is the first report of individual-level lateralization for animal genitalia that do not show any conspicuous anatomical differentiation between the right and left organs. Possible advantages of lateralization are discussed in relationship to co-evolution of the genitalia between the sexes.

A 2012–13 survey on Penang Island, Malaysia, revealed the existence of both Drosophila ananassae and Drosophila parapallidosa, the latter of which carries chromosomes Y and 4 from D. ananassae and thus is of hybrid origin. We collected the flies again from the same location in 2018. The hybrid population remained present, which suggests that the D. parapallidosa of hybrid origin does not represent a mere transient population but is stable. Why do these two species coexist irrespective of gene flow? We realized that body size is generally larger in D. ananassae than in D. parapallidosa, which constitutes a new character with which to discriminate these species; previously the number of sex comb teeth was the only diagnostic trait. Character displacement was not detected, however, for those traits. We crossed these two species, which resulted in offspring that had an altered genomic constitution. The body size of D. ananassae was dominant, and the presence of chromosomes Y and 4 did not have a significant effect on body size. By contrast, the presence of chromosome 4 from D. ananassae significantly affected the number of sex comb teeth. Even flies having a genomic constitution similar to that of the Penang D. parapallidosa exhibited a number of sex comb teeth that was intermediate between the two species. We propose that the D. parapallidosa sex comb character underwent selection during evolution of the Penang Island population. Reproductive interference between the species, presumably caused by signal jamming, was detected.

The model organism Drosophila melanogaster has become a focal system for investigations of rapidly evolving genital morphology as well as the development and functions of insect reproductive structures. To follow up on a previous paper outlining unifying terminology for the structures of the male terminalia in this species, we offer here a detailed description of the female terminalia of D. melanogaster. Informative diagrams and micrographs are presented to provide a comprehensive overview of the external and internal reproductive structures of females. We propose a collection of terms and definitions to standardize the terminology associated with the female terminalia in D. melanogaster and we provide a correspondence table with the terms previously used. Unifying terminology for both males and females in this species will help to facilitate communication between various disciplines, as well as aid in synthesizing research across publications within a discipline that has historically focused principally on male features. Our efforts to refine and standardize the terminology should expand the utility of this important model system for addressing questions related to the development and evolution of animal genitalia, and morphology in general.

Some diopsid flies have sexually dimorphic eye stalks that are assumed to require considerable nutrition for growth but are advantageous in competition and courtship. According to the handicap theory, the eye span in some dimorphic species serves as a reliable signal of individual quality to an opponent. However, it is not well understood how well eye span represents energy source storage. In this study, we focused on two species: Sphyracephala detrahens, which has weak dimorphism, and Cyrtodiopsis dalmanni, which has moderate dimorphism. We found that the eye stalks of the former species contained more fat bodies than those of the latter species. When the flies were starved, the fat body cells in the eye stalks underwent autophagy. A strong positive correlation was consistently found between eye span and starvation tolerance for S. detrahens, while a weak correlation was found for C. dalmanni. Furthermore, starvation decreased the contest winning rate between S. detrahens pairs with similar eye spans. These findings suggest that the presentation of resource holding potential may be larger than the actual storage ability and that the fidelity of nutritional storage signaling varies; the signal presented by S. detrahens is more reliable than that presented by C. dalmanni.

The evolution of laterality, that is the biased use of laterally paired, morphologically symmetrical organs, has attracted the interest of researchers from a variety of disciplines. It is, however, difficult to quantify the fitness benefits of laterality because many organs, such as human hands, possess multimodal functions. Males of the earwig Labidura riparia (Insecta: Dermaptera: Labiduridae) have morphologically similar laterally paired penises, only one of which is used for inseminating the female during a single copulation bout, and thus provide a rare opportunity to address how selection pressure may shape the evolution of population-level laterality. Our population studies revealed that in 10 populations, located at 2.23-43.3° north, the right penis is predominantly used for copulating (88.6%). A damaged penis was found in 23% of rare left-handers, suggesting that the left penis can function as a spare when the right one is damaged. By pairing L. riparia females with surgically manipulated males, we found that males forced to use the right penis outperformed left-handed males in copulation (the probability of establishing genital coupling during the 1-hr observation period: odds ratio [OR] of 3.50) and insemination (probability of transferring a detectable amount of sperm: OR of 2.94). This right-handed advantage may be due to the coiled morphology of the sperm storage organ with a right-facing opening. Thus, female genital morphology may play a significant role in the evolution of handedness and may have acted as a driving force to reduce penis number in related taxa.

The puzzling sex ratio behavior of Melittobia wasps has long posed one of the greatest questions in the field of sex allocation. Laboratory experiments have found that, in contrast to the predictions of theory and the behavior of numerous other organisms, Melittobia females do not produce fewer female-biased offspring sex ratios when more females lay eggs on a patch. We solve this puzzle by showing that, in nature, females of Melittobia australica have a sophisticated sex ratio behavior, in which their strategy also depends on whether they have dispersed from the patch where they emerged. When females have not dispersed, they lay eggs with close relatives, which keeps local mate competition high even with multiple females, and therefore, they are selected to produce consistently female-biased sex ratios. Laboratory experiments mimic these conditions. In contrast, when females disperse, they interact with nonrelatives, and thus adjust their sex ratio depending on the number of females laying eggs. Consequently, females appear to use dispersal status as an indirect cue of relatedness and whether they should adjust their sex ratio in response to the number of females laying eggs on the patch.

Many male animals donate nutritive materials during courtship or mating to their female mates. Donation of large-sized gifts, though costly to prepare, can result in increased sperm transfer during mating and delayed remating of the females, resulting in a higher paternity Nuptial gifting sometimes causes severe female-female competition for obtaining gifts (i.e., sex-role reversal in mate competition) and female polyandry, changing the intensity of sperm competition and the resultant paternity gains. We built a theoretical model to analyze such coevolutionary feedbacks between nuptial gift size (male trait) and propensity for multiple mating (female trait). Our genetically explicit, individual-based computer simulations demonstrate that a positive correlation between donated gift size and the resultant paternity gain is a requisite for the co-occurrence of large-sized gifts and females’ competitive multiple mating for the gifts. When donation of gifts imposes monandry, exaggeration in nuptial gift size also occurs under the assumption that the last male monopolizes paternity. We also analyzed the causes and consequences of the evolution of a female persistence trait in trading of nuptial gifts, that is, double receptacles for nuptial gifts known to occur in an insect group with a “female penis” (Neotrogla spp.).

Mechanical incompatibility of male and female genitalia is common in animals with internal fertilization. However, our knowledge regarding the precise mechanisms is limited. One key question regards the susceptibility of the match between male and female genitalia to morphological modification. To address this issue, we generated six different second-chromosome introgression lines possessing partially Drosophila mauritiana-like genital morphology in multiple structures in D. simulans background. Three of the six introgression males showed elevated mobility at some stages during copulation with D. simulans females; this was assumed to be an indication of genital mismatch. Notably, one of the introgression males with D. mauritiana-like enlarged anal plates showed occasional leakage of adhesive ejaculate on the body surface when mated with pure D. simulans females, suggesting apparent structural incompatibility in genital coupling. These observations suggested that both sexual and natural selection shape the anal plate morphology, highlighting the role of this structure as an important component of mechanical isolation. Partial replacement (introgression) by a sibling species genome can induce perturbations in genital coupling mechanics, suggesting that genital compatibility can be susceptible to subtle genomic changes at the early stages of divergence in these species.

The mechanisms driving the coevolution of male and female genital morphologies are still debated. Female genitalia in Drosophila species bear membranous "pouches" or hardened "shields," which the male genital armature contact during copulation. Although shield-like structures likely serve to mitigate the effects of harmful mating, some authors have suggested that soft pouches, which do not prevent male genitalia from inflicting wounds, represent a congruent sensory organ. To elucidate the evolutionary forces responsible for the development of such organs, I examined the effects of artificial damage to various genital parts of female Drosophila erecta on reproductive success. Despite a high survival rate among females, damage to the ovipositor plate resulted in frequent failure of insemination and in the embedment of eggs into the substrate. Damage to the vaginal shield resulted in increased mortality and frequent failure of egg embedment, with an egg blocking the vagina under the damaged shield in some females. Wounding of the pouch had less of an effect on both mating and oviposition success, suggesting that the structure "lures" the male trauma-causing organs to areas where the resultant wounds do not interfere with insemination or oviposition. These data show that the dual functions of female genitalia (mating and oviposition) mediate genital coevolution.

Limited attention has been given to ecological factors influencing the coevolution of male and female genitalia. The innovative ovipositor of Drosophila suzukii , an invading fruit pest, represents an appealing case to document this phenomenon. The serrated saw-like ovipositor is used to pierce the hard skin of ripening fruits that are not used by other fruit flies that prefer soft decaying fruits. Here, we highlight another function of the ovipositor related to its involvement in genital coupling during copulation. We compared the morphology and coupling of male and female genitalia in this species to its sibling species, Drosophila subpulchrella , and to an outgroup species, Drosophila biarmipes . These comparisons and a surgical manipulation indicated that the shape of male genitalia in D. suzukii has had to be adjusted to ensure tight coupling, despite having to abandon the use of a hook-like structure, paramere, because of the more linearly elongated ovipositor. This phenomenon demonstrates that ecological niche exploitation can directly affect the mechanics of genital coupling and potentially cause incompatibility among divergent forms. This model case provides new insights towards elucidating the importance of the dual functions of ovipositors in other insect species that potentially induce genital coevolution and ecological speciation.

The cave-dwelling psocid tribe Sensitibillini ( Afrotrogla , Neotrogla and Sensitibilla ) is of special morphological and evolutionary interest because of its possession of reversed copulatory organs: i.e. females of Afrotrogla and Neotrogla have a penis-like organ. The female penis structure is highly variable among taxa, as is the case of the male penis in animals with normal copulatory organs. Here, we present the first molecular phylogeny of Sensitibillini and analyse the evolutionary pattern of their genitalia. Afrotrogla and Neotrogla did not form a monophyletic clade, and their female penis structures are significantly different, suggesting two independent origins of the female penis within Sensitibillini. In Neotrogla , the species that has a simple female penis is embedded among species that have an elaborate penis, and detailed structures of the female penis elaborations are in exact agreement among species, suggesting a secondary simplification of the female penis. A correlated evolutionary pattern between male and female genitalia was also detected. This coevolution of genitalia may suggest that sexual conflict or cryptic ‘male’ choice drove the diversity of the female penis, as is the case of male penile diversity in animals with conventional genitalia.

Chloroplast DNA is organized into DNA–protein conglomerates called chloroplast nucleoids, which are replicated, transcribed, and inherited. We applied live-imaging technology with a microfluidic device to examine the nature of chloroplast nucleoids in Chlamydomonas reinhardtii. We observed the dynamic and reversible dispersion of globular chloroplast nucleoids into a network structure in dividing chloroplasts. In the monokaryotic chloroplast (moc) mutant, in which chloroplast nucleoids are unequally distributed following chloroplast division due to a defect in MOC1, the early stages of chloroplast nucleoid formation occurred mainly in the proximal area. Ovoviviparity or viviparity has evolved independently in animals and involves adaptations in females to accommodate developing embryos for a prolonged duration in their bodies, a condition which has likely to have influenced the evolution of the male genitalia. We aimed to ascertain whether the elongated male genitalia of the ovoviviparous free-living earwig species Marava arachidis (Dermaptera: Spongiphoridae) delivers sperm directly to the female ovaries where fertilization occurs. Males mated coercively with females by grabbing the female antenna with their mouth parts. Although females resisted the mating attempts, pairs mated 3.3 times on average over 15 h. The elongated intromittent organ, known as a virga, was inserted into the long-tubed spermatheca during insemination. Surgical ectomy of the spermatheca confirmed that sperm migrated from here to the ovaries with a variable delay. A pair of sclerites in the male genitalia frequently inflicted wounds near the spermathecal opening, while the single, thin virga sometimes broke off during mating. However, unlike earwigs bearing a ‘spare’ virga, damage was restricted to the tip of the virga, without which the males could still inseminate the females. We discuss the evolution of the genitalia in this insect in the light of sexual selection and sexual conflict over mating and fertilization.

We report a functional switching valve within the female genitalia of the Brazilian cave insect Neotrogla. The valve complex is composed of two plate-like sclerites, a closure element, and in-and-outflow canals. Females have a penis-like intromittent organ to coercively anchor males and obtain voluminous semen. The semen is packed in a capsule, whose formation is initiated by seminal injection. It is not only used for fertilization but also consumed by the female as nutrition. The valve complex has two slots for insemination so that Neotrogla can continue mating while the first slot is occupied. In conjunction with the female penis, this switching valve is a morphological novelty enabling females to compete for seminal gifts in their nutrient-poor cave habitats through long copulation times and multiple seminal injections. The evolution of this switching valve may have been a prerequisite for the reversal of the intromittent organ in Neotrogla., eLife digest In dry caves of southeastern Brazil, live a group of insects named Neotrogla that are perhaps best known because the egg-producing females have penises while the sperm-producing males have vaginas. The sex roles of these Brazilian cave insects are also reversed: females compete over the males, who in turn are selective of their female partners. This sex role reversal likely evolved within Neotrogla because the males’ semen represents a rich and reliable source of energy within a nutrient-poor cave environment. When females are not using semen to fertilize their eggs, they consume it. Yet, while other animals show sex role reversal, Neotrogla species alone have reversed sexual organs. Neotrogla penises are spiky and may have evolved so that females can anchor themselves to males and then mate for prolonged periods. This would allow the females to stock up on the nutritious semen. Compared to their closest relatives, Neotrogla species can hold twice as much semen within their sperm storage organs. Scientists have speculated that a valve-like structure within this organ enables this extra storage by allowing the female to redirect semen into two separate chambers. But the organ’s small size has made it difficult to determine its inner workings, and scientists have yet to discover a switching valve that serves such a purpose within the animal kingdom. Yoshizawa et al. examined three Neotrogla species using advanced imaging technology and detected the first example of a biological switching valve. Neotrogla females can control this valve, switching the flow of semen between two slots. In this way, females can store two batches of semen at once. Seemingly exploiting this adaptation, the females’ spiky penises help them restrain males until they have received multiple semen injections. Yoshizawa et al. therefore suggest the emergence of this valve within the sperm storage organ may have promoted the evolution of the female penis. Along with giving insight into the lives of cave insects, these findings are of interest to engineers, who face challenges when constructing valves on a microscopic scale. The unique switching valve of female Neotrogla may one day inspire new man-made machinery that could advance a range of industries.

The sex ratio behavior of parasitoid wasps in the genus Melittobia is scandalous. In contrast to the prediction of Hamilton's local mate competition theory, and the behavior of numerous other species, their extremely female-biased sex ratios (1-5% males) change little in response to the number of females that lay eggs on a patch. We examined the mating structure and fitness consequences of adjusting the sex ratio in M. australica and found that (1) the rate of inbreeding did not differ from that expected with random mating within each patch; (2) the fitness of females that produced less female-biased sex ratios (10 or 20% males) was greater than that of females who produced the sex ratio normally observed in M. australica. These results suggest that neither assortative mating nor asymmetrical competition between males can explain the extreme sex ratios. More generally, the finding that the sex ratios produced by females led to a decrease in their fitness suggests that the existing theory fails to capture a key aspect of the natural history of Melittobia, and emphasizes the importance of examining the fitness consequences of different sex ratio strategies, not only whether observed sex ratios correlate with theoretical predictions.

SummarySex-specific elaborations are common in animals and have attracted the attention of many biologists, including Darwin [1]. It is accepted that sexual selection promotes the evolution of sex-specific elaborations. Due to the faster replenishment rate of gametes, males generally have higher potential reproductive and optimal mating rates than females. Therefore, sexual selection acts strongly on males [2], leading to the rapid evolution and diversification of male genitalia [3]. Male genitalia are sometimes used as devices for coercive holding of females as a result of sexual conflict over mating [4, 5]. In contrast, female genitalia are usually simple. Here we report the reversal of intromittent organs in the insect genus Neotrogla (Psocodea: Prionoglarididae) from Brazilian caves. Females have a highly elaborate, penis-like structure, the gynosome, while males lack an intromittent organ. The gynosome has species-specific elaborations, such as numerous spines that fit species-specific pouches in the simple male genital chamber. During prolonged copulation (∼40–70 hr), a large and potentially nutritious ejaculate is transferred from the male via the gynosome. The correlated genital evolution in Neotrogla is probably driven by reversed sexual selection with females competing for seminal gifts. Nothing similar is known among sex-role reversed animals.

The evolution of a female penis is an extremely rare event and is only known to have occurred in a tribe of small cave insects, Sensitibillini (Psocodea: Trogiomorpha: Prionoglarididae). The female penis, which is protrudable and inserted into the male vagina-like cavity during copulation to receive semen, is thought to have evolved independently twice in this tribe, in the Brazilian Neotrogla and the African Afrotrogla. These findings strongly suggest that there are some factors unique to Sensitibillini that have facilitated female penis evolution. Here, several hypothetical factors are presented that may have enabled the evolution of the female penis in Sensitibillini. The female-female competition for nutritious semen, the oligotrophic environment, and the twin insemination slots with switching valve are considered to be the driving factors for female penis evolution. Additionally, the following factors are considered responsible for relaxing the constraint against female penis evolution: preexistence of the female-above mating position, the elongated duct connecting the female pre-penis with the sperm storage organ, and the small male genital cavity accepting the female genital tubercle bearing the opening of this duct. Understanding the factors enabling female penis evolution may also shed light on the evolution of the male penis.

Dermaptera (earwigs) is a relatively small polyneopteran order with approximately 2200 described species. They are characterized by a pair of forceps, which are hardened, unsegmented cerci at the caudal end of the abdomen. In most species, males have more exaggerated forceps than females, indicating an effect of sexual selection on them. Earwigs also exhibit astonishing diversity in the number, laterality and size of both male and female genital components. This characteristic has promoted the study of postcopulatory sexual selection in several representative species. Here, previous studies of earwigs that examined pre- and postcopulatory sexual selection are reviewed in detail. Related topics included here are sexually antagonistic coevolution, evolution of laterally asymmetrical morphologies, and developmental aspects of intra-sexually dimorphic traits. A new terminology system for male genitalia is also proposed.

Female genitalia often show complex morphologies that cannot be explained by sperm reception and storage functions. However, our understanding of the forces underlying genital exaggeration in females is limited. Female earwigs, Euborellia plebeja , are promiscuous and their highly elongated sperm storage organs allow only partial removal and displacement of stored sperm by shorter male genitalia, resulting in only a 20% gain in paternity per copulation with a sperm-saturated female. This study examined the significance and optimality of restricted sperm displacement for females. A staged mating experiment with a paternity success analysis revealed that large males dominated male–male competition for burrows housing females, resulting in repeated copulations with the same female. Despite the low paternity gain per copulation, such repeated copulations resulted in a significant increase in paternity for larger males with higher resource-holding potential. A numerical simulation based on the relationship between male body size and copulation frequency showed that restricted sperm displacement (about 20% per copulation) is optimal for promiscuous females to accumulate sperm effectively from larger males. Because male body size is heritable in this species, females were estimated to benefit from a 1.4% increase in their sons' mating success. This genetic benefit disappeared when only a single copulation per male–female encounter was assumed. Since no measurable costs of mating for females have been detected in this species, the combination of promiscuity and restricted sperm displacement is best explained by the cryptic female choice hypothesis.
