Wolbachia is a genus of maternally transmitted bacteria having an endosymbiotic relationship with arthropod and nematode species. These bacteria manipulate host development, sex-determination, and reproduction. In addition, they are known to potentially suppress vector-borne diseases by interfering with pathogen transmission. Although the occurrence of Wolbachia infection in insects has been known, the underlying mechanisms that mediate their interactions remain unclear. To examine the influence of Wolbachia adaptation on the host, we infected wAlbA and wAlbB strain from Aedes albopictus into the C6/36 cell line derived from Ae. albopictus. The transient Wolbachia infection was characterized by induction of cell migration without cell proliferation. The production of nitrite and reactive oxygen species (ROS) was induced by transient Wolbachia infection. Cells with transient Wolbachia infection exhibited elevated expression of Toll-like receptor 6 (TLR6) and myeloid differentiation primary response 88 (Myd88). Conversely, the expression of TLR2, TLR4, TLR7, Cactus, Ankyrin, and Argonaute2 (AGO2) was inhibited upon Wolbachia infection. These results suggest that Wolbachia has an influence on the cell migration ability as well as host innate immune response in vitro. Considering these results, transient Wolbachia strain transfer in C6/36 cells might be an important approach for studying Wolbachia-host interactions and might help gain a deeper understanding of the early adaptation of Wolbachia in the original host insect.

The number of coronavirus disease (COVID-19)-positive cases has increased in Jeju Island, Republic of Korea. Identification and monitoring of new mutations in severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) are extremely important to fighting the global pandemic. We report a breakout of the B.1.620 lineage, harboring the E484 mutation in the virus spike protein in a general hospital on Jeju Island. A cluster of cases was detected between August 4 and September 10, 2021, involving 20 patients positive for COVID-19 of 286 individuals exposed to the virus, comprising hospital patients, staff, and caregivers. We analyzed the epidemiological characteristics and spike proteins mutation sites using Sanger sequencing and phylogenetic analysis on these 20 patients. By analyzing genomic variance, it was confirmed that 12 of the confirmed patients harbored the SARS-CoV-2 B.1.620 lineage. The breakthrough rate of infection was 2% in fully vaccinated individuals among these patients. Next clade analysis revealed that these SARS-CoV-2 genomes belong to clade 20A. This is the first reported case of SARS-CoV-2 sub-lineage B.1.620, although the B.1.617.2 lineage has prevailed in August and September in Jeju, which has a geographical advantage of being an island. We reaffirm that monitoring the spread of SARS-CoV-2 variants with characteristic features is indispensable for controlling COVID-19 outbreaks.

Flaviviruses, for example, dengue virus (DENV) and Zika virus (ZIKV), and Alphaviruses, for example, chikungunya virus (CHIKV), attracted little attention until 2000 when increasingly higher numbers of infections were reported in urban settings throughout much of the tropical and subtropical areas of the world. While Aedes aegypti is the primary vector in most areas, Aedes albopictus, which is present in relatively high numbers in forested areas of the Republic of Korea (ROK), as well as limited urban environments (e.g., where used tires are improperly stored), is considered a secondary vector. Aedes albopictus gained considerable attention in 2016 when > 100 imported cases of DENV were reported annually since 2010 in the ROK as travelers returned home from endemic countries,1 in addition to 16 imported cases of ZIKV in 2016. ZIKV, a member of the Family Flaviviridae, was first isolated in Uganda, Africa, from monkeys in 1947 and from Aedes africanus in 1948.2 While there were occasional cases and outbreaks, its recent adaptation to efficiently infect Ae. aegypti, Ae. albopictus, and several other Aedes spp. in the Pacific Islands has led to its widespread dispersal and outbreaks in many of the tropical and semitropical regions of the world, including nine countries in Asia, 47 in Central and South America, one in North America, 10 in Oceania, and two in Africa, with the potential to spread to temperate regions during “mosquito seasons”.3–6 ZIKV is a monkey-mosquito-monkey forest cycle zoonotic disease that has been adapted to an urban Ae. aegypti-man-Ae. aegypti (and related Aedes species) cycle with the potential for emergence in nonendemic areas where competent mosquito vectors (e.g., Ae. albopictus) are present.7 Worldwide, Ae. aegypti, and Ae. albopictus to a lesser degree because of its broader range of hosts, have been identified as primary and secondary vectors of ZIKV.8 Vector competence studies of Ae. albopictus and Ae. aegypti exposed to the same mice infected by ZIKV showed that the viremia levels in mice produced disseminated infections. However, Ae. albopictus infection rates were more dose dependent, requiring higher blood meal titers than titers required for Ae. aegypti infections.9 Unfortunately, as with other arboviruses, for example, West Nile virus and CHIKV, there are no government approved human vaccines or other specific treatments for ZIKV.10 Therefore, it is essential to implement effective vector control for target species near residences where imported ZIKV-infected patients in the ROK are identified to decrease the potential human-vector-human transmission risks during the late spring to fall mosquito season. An example of this type of approach occurred in Chiba City, Japan, in 2014, when emergency vector surveillance and control strategies of Ae. albopictus were implemented due to local transmission of DENV. The result was that Ae. albopictus populations were significantly reduced after adult control strategies were implemented, which also reduced the risks for future transmission of DENV.11 A total of 16 imported ZIKV cases, including one asymptomatic case, were identified from January to October 2016 in the ROK. Nine of the cases were reported during the primary mosquito season from June to October.12 All ZIKV cases were attributed to transmission by mosquitoes during their travels to foreign ZIKV endemic countries, with 77% of the cases attributed to travel to Southeast Asia (Philippines, Vietnam, and Thailand).13 While the Korean Centers for Disease Control & Prevention (KCDC) has established vector control guidelines for vivax malaria and Japanese encephalitis that are endemic in the ROK, there were no established guidelines for vector control of nonendemic vector-borne diseases, for example, ZIKV, DENV, and CHIKV, imported from tropical and subtropical countries where they are endemic. To reduce the potential for autochthonous transmission of imported mosquito-borne viruses during the mosquito season in the ROK, the KCDC established guidelines in 2016 for the control of Ae. albopictus, the primary vector in Korea.4 However, the efficacies of these guidelines have not been fully evaluated.

Spleen traps malaria-infected red blood cells, thereby leading to splenomegaly. Splenomegaly induces impairment in splenic function, i.e., rupture. Therefore, splenomegaly inhibition is required to protect the spleen. In our previous study, genistein was found to have an influence on malaria-induced splenomegaly. However, the effect of genistein in malaria-induced splenomegaly, especially on the function of spleen, has not been fully investigated. In this study, hematoxylin and eosin (H&E) staining images show that genistein partially prevents malaria-induced architectural disruption of spleen. In addition, genistein decreases transgenic Plasmodium parasites accumulation in the spleen. Genistein treatment can protect splenic function from impairment caused by malaria infection. To examine the functions of malaria-infected spleen, we employed single-photon emission computed tomography/computed tomography (SPECT/CT) technology. Red blood cells are specifically radiolabeled with Technetium-99m pertechnetate (99mTcO4-) and trapped inside the spleen. The standardized uptake values (SUVs) in the spleen of infected mice are higher than those of naive and genistein-treated mice. However, genistein reduces the malaria-induced trapping capacity of spleen for heat-damaged radiolabeled RBCs, while exhibiting a protective effect against malaria. Considering these results, we suggested that genistein could be effectively used in combination therapy for malaria-induced splenic impairment.

Red blood cell (RBC) deformability has been considered a potential biomarker for monitoring pathological disorders. High throughput and detection of subpopulations in RBCs are essential in the measurement of RBC deformability. In this paper, we propose a new method to measure RBC deformability by evaluating temporal variations in the average velocity of blood flow and image intensity of successively clogged RBCs in the microfluidic channel array for specific time durations. In addition, to effectively detect differences in subpopulations of RBCs, an air compliance effect is employed by adding an air cavity into a disposable syringe. The syringe was equally filled with a blood sample (V(blood) = 0.3 mL, hematocrit = 50%) and air (V(air) = 0.3 mL). Owing to the air compliance effect, blood flow in the microfluidic device behaved transiently depending on the fluidic resistance in the microfluidic device. Based on the transient behaviors of blood flows, the deformability of RBCs is quantified by evaluating three representative parameters, namely, minimum value of the average velocity of blood flow, clogging index, and delivered blood volume. The proposed method was applied to measure the deformability of blood samples consisting of homogeneous RBCs fixed with four different concentrations of glutaraldehyde solution (0%-0.23%). The proposed method was also employed to evaluate the deformability of blood samples partially mixed with normal RBCs and hardened RBCs. Thereafter, the deformability of RBCs infected by human malaria parasite Plasmodium falciparum was measured. As a result, the three parameters significantly varied, depending on the degree of deformability. In addition, the deformability measurement of blood samples was successfully completed in a short time (∼10 min). Therefore, the proposed method has significant potential in deformability measurement of blood samples containing hematological diseases with high throughput and precise detection of subpopulations in RBCs.

The parasite Plasmodium falciparum causes severe malaria and is the most dangerous to humans. However, it exhibits resistance to their drugs. Farnesyltransferase has been identified in pathogenic protozoa of the genera Plasmodium and the target of farnesyltransferase includes Ras family. Therefore, the inhibition of farnesyltransferase has been suggested as a new strategy for the treatment of malaria. However, the exact functional mechanism of this agent is still unknown. In addition, the effect of farnesyltransferase inhibitor (FTIs) on mitochondrial level of malaria parasites is not fully understood. In this study, therefore, the effect of a FTI R115777 on the function of mitochondria of P. falciparum was investigated experimentally. As a result, FTI R115777 was found to suppress the infection rate of malaria parasites under in vitro condition. It also reduces the copy number of mtDNA-encoded cytochrome c oxidase III. In addition, the mitochondrial membrane potential (ΔΨm) and the green fluorescence intensity of MitoTracker were decreased by FTI R115777. Chloroquine and atovaquone were measured by the mtDNA copy number as mitochondrial non-specific or specific inhibitor, respectively. Chloroquine did not affect the copy number of mtDNA-encoded cytochrome c oxidase III, while atovaquone induced to change the mtDNA copy number. These results suggest that FTI R115777 has strong influence on the mitochondrial function of P. falciparum. It may have therapeutic potential for malaria by targeting the mitochondria of parasites.

Disease-mediated mosquitoes have been receiving much attention, as the World Health Organization recently declared the Zika virus a global public health emergency. Mosquitoes transmit pathogens that cause various tropical diseases including malaria, dengue fever and yellow fever as well as Zika virus. The vector efficiency of mosquitoes depends on their blood-feeding characteristics and the mechanics of their blood-sucking pump system, but only a few studies have attempted to investigate these key issues. In this study, we demonstrate the rapid and gluttonous liquid-feeding characteristics of Ae. albopictus which transmits Zika virus can be explained by similar proportion of two blood-sucking pumps and accelerated liquid intake driven by fast expanding of pumps. Our results provide insight into the vector efficiency of Ae. albopictus in terms of feeding velocity, pumping frequency, liquid-intake rate, and wall shear stress.

Mosquitoes act as a vector for the transmission of disease. The World Health Organization has recommended strict control of mosquito larvae because of their “few, fixed, and findable” features. The respiratory system of mosquito larvae and pupae in the water has a weak point. As aquatic organisms, mosquito larvae and pupae inhale atmosphere oxygen. However, the mosquito pupae have a non-feeding stage, unlike the larvae. Therefore, detailed study on the tracheal system of mosquito pupae is helpful for understanding their survival strategy. In this study, the three-dimensional (3D) structures of the tracheal systems of Anopheles sinensis and Aedes togoi pupae were comparatively investigated using synchrotron X-ray microscopic computed tomography. The respiratory frequencies of the dorsal trunks were also investigated. Interestingly, the pupae of the two mosquito species possess special tracheal systems of which the morphological and functional features are distinctively different. The respiratory frequency of Ae. togoi is higher than that of An. sinensis. These differences in the breathing phenomena and 3D structures of the respiratory systems of these two mosquito species provide an insight into the tracheal systems of mosquito pupae.

Mosquito-borne diseases, such as malaria, dengue fever, and Zika virus, are serious global health issues. Vector control may be an important strategy in reducing the mortality caused by these diseases. The respiratory system of mosquito larvae in the water has to inhale atmospheric oxygen as aquatic organisms. In this study, the three-dimensional (3D) structures of the dorsal longitudinal trunks (DLTs) of the tracheal systems of Anopheles sinensis and Aedes togoi were compared using synchrotron X-ray microscopic computed tomography. DLT respiratory frequencies were also investigated. Interestingly, the larvae of the two mosquito species exhibit tracheal systems that are both morphologically and functionally distinct. A. sinensis hangs horizontally under the water surface, and has a smaller DLT volume than A. togoi. In contrast, A. togoi hangs upside down using a siphon by fixing its tip to the water surface. The frequency of peristaltic movement in A. togoi is higher than that of A. sinensis. These differences in the structures and breathing behaviors of the respiratory systems of mosquito larvae provide new insights into the tracheal systems of mosquito larvae, which should help develop novel effective control strategies targeting mosquito larvae.

Inflammation is a biological response to various external stimuli that induces vascular permeability via signaling pathways. Lipopolysaccharide (LPS) activates the acute inflammatory response and induces inflammatory mediators. Genistein has an anti-inflammation activity, however, whether genistein inhibits vascular permeability using the same signaling pathways at different development stages in chick embryos is unclear. Inflammatory processes in the chorioallantoic membrane (CAM) after LPS and genistein treatments at different immune stages were investigated. Genistein suppresses LPS-induced vascular permeability in chick embryos at the immature stage. RTPCR and western blotting were performed for LPSinduced toll-like receptor 4 (TLR4) signaling pathways in the CAM. The LPS-induced TLR4 pathways are attenuated by genistein. Different inflammation mechanisms in TLR4 pathways are shown in embryos in the immature stage. Genistein probably inhibits the LPS-induced inflammatory response through different signaling pathways and has a strong therapeutic potential associated with inflammationinduced vascular permeability in chick embryos with immature immune capacities.

We examined the correlation between laminin 332 and malignancy in bladder cancer patients, and, using a strain of invasive bladder cancer cells, determined whether laminin 332 causes bladder cancer motility and invasion. To investigate the correlation between laminin 332 g2 distribution and patient outcome, we performed a semiquantitative immunohistochemical analysis of 35 paraffin-embedded samples using the antibody D4B5, which is specific for the laminin 5 γ2 chain. To evaluate the role of laminin 332 in NBT-II cell motility and invasion, we used a scratch assay and the Boyden chamber chemoinvasion system. Tumor stage and grade were significantly correlated with a loss of laminin 332 γ2 chain from the basement membrane (p = 0.001) and its retention in the cytoplasm (p = 0.001) (Kruskal–Wallis test). Kaplan–Meier survival curves revealed an association between the risk of progression and cytoplasmic retention of the laminin 332 γ2 chain. In addition, an in vitro scratch assay showed an increase in the migration of cells treated with laminin 332 from their cluster. The Boyden chamber assay showed that laminin 332 potentiated NBT-II cell invasion. Immunohistochemistry results showed that bladder cancer patients with a higher malignancy expressed more laminin 332. The in vitro scratch and invasion assay showed that laminin 332 stimulated the motility and invasion of bladder cancer cells. The invasion assay explains the correlation between laminin 332 expression and bladder cancer malignancy.

Female mosquitoes transmit various diseases as vectors during liquid-feeding. Identifying the determinants of vector efficiency is a major scientific challenge in establishing strategies against these diseases. Infection rate and transmission efficiency are interconnected with the mosquito-induced liquid-feeding flow as main indexes of vector efficiency. However, the relationship between liquid-feeding characteristics and pathogen remains poorly understood. The liquid-feeding behavior of Aedes togoi and Anopheles sinensis was comparatively investigated in conjunction with vector efficiency via micro-particle image velocimetry. The flow rates and ratio of the ejection volume of Aedes togoi were markedly higher than those of Anophels sinensis. These differences would influence pathogen re-ingestion. Wall shear stresses of these mosquito species were also clearly discriminatory affecting the infective rates of vector-borne diseases. The variations in volume of two pump chambers and diameter of proboscis of these mosquito species were compared to determine the differences in the liquid-feeding process. Liquid-feeding characteristics influence vector efficiency; hence, this study can elucidate the vector efficiency of mosquitoes and the vector-pathogen interactions and contribute to the development of strategies against vector-borne diseases.

Summary Background Ischemic preconditioning (IPC) is a powerful mechanism for limiting myocardial infarction during or after ischemia–reperfusion (IR) injury. However, effective target genes and proteins for IPC are unknown. We characterized global changes in gene expression in the heart during IR, and identified effective target genes for IPC. Methods Hearts were isolated from Sprague-Dawley rats under control, IR, and IPC conditions. We generated expressed-sequence-tags (ESTs) for each group and investigated their functions and the major biological processes in which they are involved using the eukaryotic clusters of orthologous groups (KOG) database and bioinformatics analysis tools. Results IR modified the expression of 126 genes. Of these, 62 were upregulated, 64 were downregulated, and 77 were found to be effective target genes for IPC. In KOG analysis, most of the genes whose expression was modified were involved in energy production and conversion and the cytoskeleton. A gene-to-gene interaction map revealed that IR modified the expression of genes in four major functional modules: electron transport chain/oxidative phosphorylation; tricarboxylic acid cycle/glucose metabolism/amino acid metabolism; cellular structure and contraction; and gene transcription, translation, and protein folding. At the individual gene level, the genes encoding mitochondrial cytochrome c oxidase subunits 2 and 3 were downregulated, and those encoding the major cytoskeleton components tropomyosin, myosin light chain, myomesin 2, and myosin regulatory light chain 2, as well as the gene encoding the iron-storage protein ferritin, were upregulated, and thus were identified as potential target genes. Real time PCR evaluated expression patterns of three mitochondrial IPC effective genes. Two-dimensional electrophoresis proteomic analyses revealed altered expression of 14 target proteins. The expression patterns of six proteins matched the corresponding EST expression patterns. Conclusion The global profiling of cardiac ischemia-related genes provides the possible mechanisms of IR and IPC and ways of treating IR injury.

Phosphatase of regenerating liver (PRL)-3, a member of a subgroup of protein tyrosine phosphatases that can stimulate the degradation of the extracellular matrix, is over-expressed in metastatic colorectal cancer (CRC) relative to primary tumors. To determine whether PRL-3-induced enhancement of migration and invasion is dependent on the expression of matrix metalloproteinases (MMPs), PRL-3 was expressed in DLD-1 human CRC cells. The motility, migration and invasion characteristics of the cells were examined, and metastasis to the lung was confirmed in a nude mouse using PRL-3-overexpressing DLD-1 cells [DLD-1 (PRL-3)]. Migration and invasion of the cells were inhibited by phosphatase and farnesyltransferase inhibitors. Expression of MMPs was enhanced 3- to 10-fold in comparison to control cells, and migration and invasion were partially inhibited by small interfering RNA (siRNA) knockdown of MMP-2, -13 or -14. Importantly, siRNA knockdown of MMP-7 completely inhibited the migration and invasion of DLD-1 (PRL-3) cells, whereas overexpression of MMP-7 increased migration. The expression of MMP-7 was also downregulated by phosphatase and farnesyltransferase inhibitors. It was found that PRL-3 induced MMP-7 through oncogenic pathways including PI3K/AKT and ERK and that there is a relationship between the expression of PRL-3 and MMP-7 in human tumor cell lines. The expression of MMP-13 and -14 was very sensitive to the inhibition of farnesyltransferase; however, the migration and invasion of DLD-1 (PRL-3) cells did not strongly depend on the expression of MMP-13 or -14. These results suggest that the migration and invasion of PRL-3-expressing CRC cells depends primarily on the expression of MMP-7.

Mosquitoes act as vectors for severe tropical diseases. Mosquito-borne diseases are affected by various factors such as environmental conditions, host body susceptibility and mosquito feeding behavior. Among these factors, feeding behavior is affected by the feeding pump system located inside the mosquito head and also depends on the species of mosquito. Therefore, the 3D morphological structures of the feeding pumps of Aedes togoi and Anopheles sinensis were comparatively investigated using synchrotron X-ray microscopic computed tomography. In addition, the feeding behaviors of their pumping organs were also investigated using a 2D X-ray micro-imaging technique. An. sinensis, a malarial vector mosquito, had a larger feeding pump volume than Ae. togoi in the static or resting position. Interestingly, the two species of mosquitoes exhibited different feeding behaviors. Ae. togoi had a higher feeding frequency and expansion ratio than An. sinensis. Ae. togoi also exhibited F-actin localization more clearly. These distinctive variations in feeding volumes and behaviors provide essential insight into the blood-feeding mechanisms of female mosquitoes as vectors for tropical diseases.

Plectin, a linker protein that organizes the cytoskeleton, is critical for cell migration and wound healing. It is specifically expressed in epithelial cells, muscles, and other tissues. Numerous studies have shown that curcumin (diferuloylmethane) has anti-cancer potential. Curcumin can inhibit cancer cell migration and invasion through various signaling pathways. In this study, the down-regulation of plectin expression by siRNA was observed to induce the migration and invasion of human breast and lung cancer cell lines. Interestingly, the down-regulation of plectin expression by siRNA reversed the curcumin-induced anti-migration and anti-invasion effects in both cancer cell lines while α-tocopherol and genistein inhibited knockdown of plectin induced cancer cell migration. Curcumin did not affect the expression of plectin; however, α-tocopherol and genistein inhibited migration in plectin down-regulated cancer cells. These findings support the novel role of plectin and its effect on the anti-migration and anti-invasion potential of curcumin.

Our previous study showed that a methanol extract of Inula helenium had the potential to induce detoxifying enzymes such as quinone reductase (QR) and glutathione S-transferase (GST) activity. In this study the methanol extract was further fractionated using silica gel chromatography and vacuum liquid chromatography, to yield pure compounds alantolactone and isoalantolactone as QR inducers. Alantolactone caused a dose-dependent induction of antioxidant enzymes including QR, GST, gamma-glutamylcysteine synthase, glutathione reductase, and heme oxygenase 1 in hepa1c1c7 mouse hepatoma cells. The compound increased the luciferase activity of HepG2-C8 cells, transfectants carrying antioxidant response element (ARE)-luciferase gene, in a dose-dependent manner, suggesting ARE-mediated transcriptional activation of antioxidant enzymes. Alantolactone also stimulated the nuclear accumulation of Nrf2 that was inhibited by phosphatidylinositol 3-kinase (PI3K) inhibitors. In conclusion, alantolactone appears to induce detoxifying enzymes via activation of PI3K and JNK signaling pathways, leading to translocation of Nrf2, and subsequent interaction between Nrf2 and ARE in the encoding genes.

Malaria parasites induce morphological, biochemical, and mechanical changes in red blood cells (RBCs). Mechanical variations are closely related to the deformability of individual RBCs. The deformation of various RBCs, including healthy and malaria-infected RBCs (iRBCs), can be directly observed through quantitative phase imaging (QPI). The effects of chloroquine treatment on the mechanical property variation of iRBCs were investigated using time-resolved holographic QPI of single live cells on a millisecond time scale. The deformabilities of healthy RBCs, iRBCs, and drug-treated iRBCs were compared, and the effect of chloroquine on iRBC restoration was experimentally examined. The present results are beneficial to elucidate the dynamic characteristics of iRBCs and the effect of the antimalarial drug on iRBCs.

Employing quantitative phase imaging technique, the shear moduli of healthy RBC, RBC infected by Plasmodium falciparum (iRBC), drug-treated iRBC are compared. This study on antimalarial drug effects would be helpful for understanding the effects of drug on iRBCs.

Spleen plays an important role in removing old and damaged red blood cells and malaria-infected erythrocytes. When malaria parasites invade the spleen and induce splenomegaly, splenic function tends to be impaired. Thus, the inhibition of splenomegaly is strongly required to protect the spleen. In this study, malaria-induced splenomegaly is inhibited by injecting genistein into a Plasmodium berghei-infected ICR mouse. To explain this phenomenon, the effect of genistein in spleen and liver of malaria-infected mice was evaluated by histological examination. Malaria parasites disrupted splenic architecture. After treating genistein, the disrupted architecture in which red and white pulp regions were clearly separated in recovered to uninfected ones. Changes in biophysical properties of blood were studied by measuring the viscosity of blood collected from malaria-infected and uninfected mice using a microfluidic viscometer. Genistein also had a negligible influence on variation in blood viscosity. The enzymatic activity and expression pattern of proteins were then investigated to explain the genistein effect on malaria-induced splenomegaly. Genistein is a potential drug for splenomegaly in P. berghei-infected mouse.

Problem Inflammation is an essential process in the pathogenesis of endometriosis. Method of study Serum and peritoneal fluid (PF) samples were collected from women with endometriosis (n = 31) and women without endometriosis (n = 48). Chemerin, tumor necrosis factor-alpha (TNF-α), and interleukin-6 (IL-6) levels in serum and PF samples were determined with a specific enzyme-linked immunosorbent assay. Eutopic endometrial tissue from controls and ovarian endometriotic cysts were obtained during surgery. Expression of chemerin and chemerin receptors in ectopic and eutopic endometrial tissues was measured on real-time reverse transcriptase–polymerase chain reaction. Protein expression was examined with Western blot and densitometric analysis. Results Chemerin concentrations were higher in PF from women with endometriosis than that in that from controls. PF chemerin concentrations were significantly correlated with both TNF-α and IL-6 levels in PF. The mRNA and protein of chemerin and its receptor were significantly increased in the ovarian endometrioma tissue compared with eutopic endometrium of controls. Conclusion These findings suggest that chemerin plays a role in endometriosis-related pelvic inflammation.

PROBLEM Inflammation is a key process in the establishment and progression of endometriosis. Resistin, an adipocytokine, has biological properties linked to immunologic functions, but its role in endometriosis is unclear. METHOD OF STUDY Resistin gene expression was examined in eutopic and ectopic endometrial tissues from women with (n=25) or without (n=25) endometriosis. Resistin mRNA and protein levels were determined in endometrial tissue using quantitative real-time reverse transcription PCR and Western blotting, following adipokine profiling arrays. RESULTS Resistin protein was detected in human endometrial tissues using an adipokine array test. Resistin mRNA and protein levels were significantly higher in ectopic endometrial tissue of patients with endometriosis than in normal eutopic endometrial tissue. CONCLUSION Our results indicate that resistin is differentially expressed in endometrial tissues from women with endometriosis and imply a role for resistin in endometriosis-associated pelvic inflammation.

Background: In this study, we investigated the effect of the extracellular matrix on endothelial dysfunction by careful observation of human umbilical vein endothelial cells (HUVECs) cultured on denatured collagen film. Results: HUVECs on denatured collagen film showed relatively high surface roughness compared with normal HUVECs. The expression levels of MMP-1, MMP-2 and CD146 increased in the ECs on denatured collagen film. In addition, we examined the accumulation of fluorescent beads on HUVEC layers subjected to circulatory flow. The number of accumulated fluorescent beads increased on the disorganized HUVEC layers. Conclusions: The proposed in vitro study using bio inspired collagen films could potentially be used in the size- and ligand-based design of drugs to treat endothelial dysfunction caused by circulatory vascular diseases.

Mosquitoes secrete saliva that contains biological substances, including anticoagulants that counteract a host’s hemostatic response and prevent blood clotting during blood feeding. This study aimed to detect heparin, an anticoagulant in Aedes togoi using an immunohistochemical detection method, in the salivary canal, salivary gland, and midgut of male and female mosquitoes. Comparisons showed that female mosquitoes contained higher concentrations of heparin than male mosquitoes. On average, the level of heparin was higher in blood-fed female mosquitoes than in non-blood-fed female mosquitoes. Heparin concentrations were higher in the midgut than in the salivary gland. This indicates presence of heparin in tissues of A. togoi.

Phase holographic microscopy was applied for the phase imaging of red blood cells (RBCs) and human umbilical vein endothelial cells (HUVECs). HUVECs and its abnormal deformation influenced by collagenase were observed.

Background Malaria is one of the world’s public health problems in terms of medical emergency with a high risk of mortality. The protozoan malaria parasites are transmitted by infected female mosquitoes [1]. The most pathogenic human malaria parasite, Plasmodium falciparum, has gradually expanded in last three decades [2]. Choloroquine is the cheapest and the most widely used drug in almost endemic countries. However, Plasmodium falciparum exhibits resistance to their drug. Resistance to the combination of sulfadoxine-pyrimethamine was also already emerged [3]. Farnesyltransferase have been identified in eukaryotic organisms, including pathgenic protozoa of the genera Plasmodium [3,4]. Therefore, the inhibition of farnesyltransferase has been suggested as a new strategy for the malaria treatment. However, the exact mechanism of action of this class of agents is still unknown [5]. In addition, the effect of farnesyltransferase inhibitor on malaria mitochondria level is not fully understood. In this study, the effect of farnesyltransferase inhibitor on the function of mitochondria of Plasmodium falciparum were investigated experimentally. The oxygen distribution and the morphological shape of farnesyltransferase inhibitor-treated mitochondria were examined under in vitro condition. From this study, we found farnesyltransferase inhibitor is very important to understand the mitochondrial function of Plasmodium falciparum as an antimalarial drug.

MicroRNAs (miRs) are small noncoding RNAs that have been reported to be promising diagnostic tools. We used quantitative real-time reverse transcription PCR (RT-qPCR) to analyze differentially expressed miRNAs in prostate tumor samples to determine its prognostic value. From 2007 to 2009, tumor tissues were obtained from 73 radical prostatectomy specimens. Differentially expressed miR-96, -145 and -221 were validated by TaqMan RT-qPCR using all 73 tissues. The prognostic value was assessed in terms of biochemical recurrence using Kaplan-Meier and Cox regression analyses. For our patient cohort, the mean age was 64.7 years (50-76 years) and the mean prostate-specific antigen (PSA) was 7.5 ng ml(-1). During the follow-up period (mean, 19.4 months), 14 of 73 (19.2%) patients developed biochemical recurrence. Expression of miR-96, -145 and -221 correlated strongly with each other, but there were no correlations between miRNA expression and clinicopathologic parameters. Few protein biomarkers for oxidative stress have been reported. In this study, we attempted to identify the proteins selectively overexpressed in human colon tumor cells by treating with hydrogen peroxide as oxidative stress. A proteomic analysis followed by western blotting showed that phosphoglycerate kinase 1 (PGK1) was induced by hydrogen peroxide in a dose-dependent manner, while its expression was suppressed by a co-treatment with delphinidin, a known antioxidant. Furthermore, several antioxidants, including alpha-tocopherol, butylated hydroxytoluene (BHT), and Trolox, also inhibited the PGK1 induction caused by hydrogen peroxide. The data suggest that PGK1 might be a potential protein biomarker of intracellular oxidative status.

The pathogenic mechanism of granulomatous amebic encephalitis (GAE) and amebic keratitis (AK) by Acanthamoeba has yet to be clarified. Protease has been recognized to play an important role in the pathogenesis of GAE and AK. In the present study, we have compared specific activity and cytopathic effects (CPE) of purified 33 kDa serine proteinases from Acanthamoeba strains with different degree of virulence (A. healyi OC-3A, A. lugdunensis KA/E2, and A. castellanii Neff). Trophozoites of the 3 strains revealed different degrees of CPE on human corneal epithelial (HCE) cells. The effect was remarkably reduced by adding phenylmethylsulfonylfluoride (PMSF), a serine proteinase inhibitor. This result indicated that PMSF-susceptible proteinase is the main component causing cytopathy to HCE cells by Acanthamoeba. The purified 33 kDa serine proteinase showed strong activity toward HCE cells and extracellular matrix proteins. The purified proteinase from OC-3A, the most virulent strain, demonstrated the highest enzyme activity compared to KA/E2, an ocular isolate, and Neff, a soil isolate. Polyclonal antibodies against the purified 33 kDa serine proteinase inhibit almost completely the proteolytic activity of culture supernatant of Acanthamoeba. In line with these results, the 33 kDa serine proteinase is suggested to play an important role in pathogenesis and to be the main component of virulence factor of Acanthamoeba.

In order to evaluate the possible roles of secretory proteases in the pathogenesis of amoebic keratitis, we purified and characterized a serine protease secreted by Acanthamoeba lugdunensis KA/E2, isolated from a Korean keratitis patient. The ammonium sulfate-precipitated culture supernatant of the isolate was purified by sequential chromatography on CM-Sepharose, Sephacryl S-200, and mono Q-anion exchange column. The purified 33 kDa protease had a pH optimum of 8.5 and a temperature optimum of 55 degrees C. Phenylmethylsulfonylfluoride and 4-(2- Aminoethyl)-benzenesulfonyl-fluoride, both serine protease specific inhibitors, inhibited almost completely the activity of the 33 kDa protease whereas other classes of inhibitors did not affect its activity. The 33 kDa enzyme degraded various extracellular matrix proteins and serum proteins. Our results strongly suggest that the 33 kDa serine protease secreted from this keratopathogenic Acanthamoeba play important roles in the pathogenesis of amoebic keratitis, such as in corneal tissue invasion, immune evasion and nutrient uptake.

We demonstrate sheath-free vertical concentration and positioning of cells induced by fluid viscoelasticity in a rectangular channel. The high uniformity in depth position and the reduction in cell overlap eliminate out-of-focus blurring and increase detection sensitivity. Additionally, orientational ordering of red blood cells improves the monitoring and counting performance of an optical imaging system. As a proof of concept of vertical focusing and cell ordering, we demonstrate counting and monitoring of red blood cells flowing in a microchannel using phase-contrast holographic microscopy for quantitative phase imaging.

Objectives Utilisation of high-frequency drills is known to increase noise induced hearing loss due to increasing the damages of inner ear cells. This study aimed to investigate whether preconditioning by using dexmedetomidine (DEX) decreased the occurrence of ischemia in inner cells of the ear. Methods We utilised a transgenic zebrafish line Brn3C, and the embryos were collected from breeding adult zebrafish. Five-day-old larvae were cultured at the density of 50 embryos, and the larvae were classified into 4 groups: control, cisplatin group, DEX group, and DEX+yohimbine; adrenoreceptor blocker group. The DEX group was categorised into 3 subgroups by dosage; 0.1, 1, and 10 µM. Preconditioning was performed for 150 minutes and then exposed to cisplatin for 6 hours. The experiment was performed in 7 replicates for each group and the number of hair cells in 3 parts of the neuromasts of each fish was determined. Results Hair cell apoptosis by cisplatin was attenuated more significantly in the DEX preconditioning group than in the control group. However, the preconditioning effects were not blocked by yohimbine. Conclusion The results of this study suggest that hearing loss caused by vibration-induced noise could be reduced by using DEX and may occur through other mechanisms rather than adreno-receptors.

Objective: To evaluate the effect of early exercise therapy on the natural killer cell cytotoxic activity (NKCA) of patients who had undergone curative resection of stomach cancer. Design: Prospective study. Patients: Thirty-five stomach cancer patients who had undergone curative surgery were randomly divided into an exercise group ( n = 17) and a control group ( n = 18). Intervention: From postoperative day 2, moderated exercise using arm and bicycle ergometers performed twice a day, 5 times a week, for 14 days. The intensity of exercise was 60% of maximal heart rate. Venous blood samples were obtained on postoperative days 1, 7, and 14. Main Outcome Measure: Mean sequential change of NKCA. Results: The mean sequential change of NKCA decreased until postoperative day 7 and then increased. Mean NKCA of day 7 decreased in both groups, compared with that at postoperative day 1. Genetic diversity of 18 Acanthamoeba isolates from ocean sediments was evaluated by comparing mitochondrial (mt) DNA RFLP, 18S rDNA sequences and by examining their cytopathic effects on human corneal epithelial cells versus reference strains. All isolates belonged to morphologic group II. Total of 16 restriction phenotypes of mtDNA from 18 isolates demonstrated the genetic diversity of Acanthamoeba in ocean sediments. Phylogenetic analysis using 18s rDNA sequences revealed that the 18 isolates were distinct from morphological groups I and III. Fifteen isolates showed close relatedness with 17 clinical isolates and A. castellanii Castellani and formed a lineage equivalent to T4 genotype of Byers' group. Two reference strains from ocean sediment, A. hatchetti BH-2 and A. griffini S-7 clustered unequivocally with these 15 isolates. Diversity among isolates was also evident from their cytopathic effects on human corneal cells. This is the first time describing Acanthamoeba diversity in ocean sediments in Korea.
