Background: Atopic dermatitis (AD) is a chronic inflammatory skin condition characterized by recurrent eczematous lesions and severe pruritus. The economic burden and time penalty caused by the relapse of AD reduce patients’ life quality. Summary: AD has complex pathogenesis, including genetic disorders, epidermal barrier dysfunction, abnormal immune responses, microbial dysbiosis of the skin, and environmental factors. Recently, the role of innate immune cells in AD has attracted considerable attention. This review highlighted recent findings on innate immune cells in the onset and progression of AD. Key Messages: Innate immune cells play essential roles in the pathogenesis of AD and enough attention should be given for treating AD from the perspective of innate immunity in clinics.

Objectives To understand the sleep quality and its influencing factors in patients with type 2 diabetes mellitus (T2DM) who suffered diabetic peripheral neuropathy (DPN), and provide evidence for clinicians to carry out comprehensive intervention measures to improve the sleep quality of patients. Methods Patients who were admitted to the Endocrinology Department of Affiliated Hospital of Zunyi Medical University were recruited from May to December 2022, and the investigation were conducted by face-to-face interview. The questionnaires included PSQI questionnaire and influencing factors, such as lifestyle and health status. Results Among the 193 patients, 40.4% of the patients never took physical examination, 56.5% of the patients had duration of illness greater than 5 years, 61.7% of the patients had had an operation, 10.4% of the patients had bad dietary status, and 55.4% of the patients had physical pain. In addition, the PSQI general score was 8.34 ± 3.98, the occurrence rate of poor sleep quality (PSQI ≥ 8) was 54.4%, and the results showed that sleep quality of the physical pain group was worse than the no pain group. Moreover, the results of multivariate analysis revealed that the factors affecting sleep quality were lower frequency of exercise, bad dietary status, lower frequency of physical examination, longer duration of illness, and smoking, and the OR and 95% CI were [1.40, 1.04∼1.89], [3.42, 1.86∼6.29], [1.49, 1.01∼2.20], [1.78, 1.09∼2.92], [2.38, 1.17∼4.88], respectively. Conclusion Patients with DPN have higher risk of poor sleep quality. Moreover, there were many risk factors associated with poor sleep quality, clinicians and health policymakers should timely detect and effectively intervene in these factors to improve the sleep quality, which is important to enhance the quality of life of T2DM patients complicated with DPN.

To improve the detection sensitivity of neomycin (NEO) in milk, we produced a sensitive monoclonal antibody (mAb) against NEO and developed a lateral flow immunoassay based on amorphous carbon nanoparticles (ACNPs-LFA). First, we conjugated NEO to carrier protein to prepare mAbs. We obtained six mAbs: mAb 1C6, mAb 1D3, mAb 2D3, mAb 4D5, mAb 5D1, and mAb 5H1. We characterised the mAbs by indirect competitive enzyme-linked immunosorbent assay and selected the most sensitive mAb based on the half-maximal inhibitory concentration (IC50). We selected mAb 4D5 (IC50 = 0.15 ng/mL) for the development of LFA. MAb 4D5 was labelled with ACNPs (ACNPs-mAb 4D5) by electrostatic absorption. Under optimised conditions, 5.4 μg mAb 4D5 coupling with 1 mL ACNPs, NEO-OVA at concentration of 80 μg/mL, 3 μL ACNPs-mAb 4D5 were used to develop LFA. The cut-off value was 8 ng/mL. Therefore, our developed ACNPs-LFA is suitable for on-site detection of NEO residues.

New potential therapies for inflammatory bowel disease are needed as not all patients respond to or maintain a response to conventional therapies. Here the authors report that mannose supplementation ameliorates experimental colitis in male mice, potentially via effects on intestinal epithelium lysosomal integrity.

Abstract Objectives This study aims to investigate the association between CD4+ T cell count and combined antiretroviral therapy (cART) with the prevalence of anal human papillomavirus (HPV) infection among HIV-positive male cohort in China. Methods A survey was conducted in men from a HIV cohort in Taizhou, China between 2016 and 2019. A face-to-face questionnaire interview was administered, and an anal-canal swab was collected for HPV genotyping. Results A total of 766 HIV-positive men were recruited. The HPV prevalence was lower among those with increased CD4+ T cell count than those with decreased or unchanged (46.5 vs. 56.6%, p = 0.033) from baseline. In multivariable models, having the current CD4+ T cell count of 350–499 cells/µL (aOR 0.28, 95% CI 0.13–0.64), and of ≥ 500 cells/µL (aOR 0.26, 95% CI 0.11–0.60) were associated with lower prevalence of any type HPV infection compared with those with

This paper proposes a recursive delta-operator-based subspace identification method with fixed data size. The majority of existing subspace identification methods are constrained to discrete-time systems because of the disparity in Hankel matrices. Additionally, due to the storage cost, LQ-factorization and singular value decomposition in identification methods are best suited for batch processing rather than online identification. The continuous-time systems are transformed into state space models based on the delta-operator to address these issues. These models approach the original systems when the sampling interval approaches zero. The size of the data matrices is fixed to reduce the computing load. By fading the impact of past data on future data, the amount of data storage can be decreased. The effectiveness of the proposed method is illustrated by the continuous stirred tank reactor system.

This paper presents a closed-loop continuous-time subspace identification method using prior information. Based on a rational inner function, a generalized orthonormal basis can be constructed, and the transformed noises have ergodicity features. The continuous-time stochastic system is converted into a discrete-time stochastic system by using generalized orthogonal basis functions. As is known to all, incorporating prior information into identification strategies can increase the precision of the identified model. To enhance the precision of the identification method, prior information is integrated through the use of constrained least squares, and principal component analysis is adopted to achieve the reliable estimate of the system. Moreover, the identification of open-loop models is the primary intent of the continuous-time system identification approaches. For closed-loop systems, the open-loop subspace identification methods may produce biased results. Principal component analysis, which reliably estimates closed-loop systems, provides a solution to this problem. The restricted least-squares method with an equality constraint is used to incorporate prior information into the impulse response following the principal component analysis. The input–output algebraic equation yielded an optimal multi-step-ahead predictor, and the equality constraints describe the prior information. The effectiveness of the proposed method is provided by numerical simulations.

The lifting approach and the Hambo system transform are used to establish the equivalent nature of continuous and transformed discrete-time stochastic systems. The constrained least squares method is adopted to investigate the incorporation of prior knowledge in order to further increase the subspace identification algorithm’s accuracy. The input–output algebraic equation derives an optimal multistep forward predictor, and prior knowledge is expressed as equality constraints. In order to solve an optimization problem with equality constraints characterizing the prior knowledge, the proposed method reduces the computational burden. The effectiveness of the proposed method is provided by numerical simulations.

Clusiaceae plants contain a wide range of biologically active metabolites that have gotten a lot of interest in recent decades. The chemical compositions of these plants have been demonstrated to have positive effects on a variety of ailments. The species has been studied for over 70 years, and many bioactive compounds with antioxidant, anti-proliferative, and anti-inflammatory properties have been identified, including xanthones, polycyclic polyprenylated acylphloroglucinols (PPAPs), benzophenones, and biphenyls. Prenylated side chains have been discovered in many of these bioactive substances. To date, there have been numerous studies on PPAPs and xanthones, while no comprehensive review article on biphenyls from Clusiaceae has been published. The unique chemical architectures and growing biological importance of biphenyl compounds have triggered a flurry of research and interest in their isolation, biological evaluation, and mechanistic studies. In particular, the FDA-approved drugs such as sonidegib, tazemetostat, daclatasvir, sacubitril and trifarotene are closely related to their biphenyl-containing moiety. In this review, we summarize the progress and development in the chemistry and biological activity of biphenyls in Clusiaceae, providing an in-depth discussion of their structural diversity and medicinal potential. We also present a preliminary discussion of the biological effects with or without prenyl groups on the biphenyls.

Dispatch of battery storage systems for stationary grid applications is a topic of increasing interest: due to the volatility of power system's energy supply relying on variable renewable energy sources, one foresees a rising demand and market potential for both short- and long-term fluctuation smoothing via energy storage. While the potential revenue attainable via arbitrage trading may yet surpass the steadily declining cost of lithium-ion battery storage systems, profitability will be constrained directly by the limited lifetime of the battery system and lowered by dissipation losses of both battery and power electronic components. In this study, we present a novel three-dimensional mixed-integer program formulation allowing to model power, state of charge (SOC), and temperature dependence of battery dynamics simultaneously in a three dimensional space leveraging binary counting and union-jack triangulation. The inclusion of a state-of-the-art electro-thermal degradation model with its dependence on most influential physical parameters to the arbitrage revenue optimization allows to extend the battery lifetime by 2.2 years (or 40%) over a base scenario. By doing a profitability estimation over the battery's lifetime and using 2018 historical intraday market trading prices, we have shown that profitability of the system increases by 11.14% via introducing SOC awareness and another significant 12.64% via introducing thermal sensitivity, resulting in a total 25.19% increase over the base case optimization formulation. Lastly, through the open source publication of the optimization routines described herein, an adaption and development of the code to individual needs is facilitated.

The consumption of shellfish contaminated with brevetoxins, a family of ladder-frame polyether toxins formed during blooms of the marine dinoflagellate Karenia brevis, can cause neurotoxic poisoning, leading to gastroenteritis and neurotoxic effects. To rapidly monitor brevetoxin levels in oysters, we generated a broad-spectrum antibody against brevetoxin 2 (PbTx-2), 1 (PbTx-1), and 3 (PbTx-3) and developed a rapid indirect competitive enzyme-linked immunosorbent assay (icELISA). PbTx-2 was reacted with carboxymethoxylamine hemihydrochloride (CMO) to generate a PbTx-2-CMO hapten and reacted with succinic anhydride (HS) to generate the PbTx-2-HS hapten. These haptens were conjugated to keyhole limpet hemocyanin (KLH) and bovine serum albumin (BSA) to prepare immunogen and coating antigen reagents, respectively, using the active ester method. After immunization and cell fusion, a broad-spectrum monoclonal antibody (mAb) termed mAb 1D3 was prepared. The 50% inhibitory concentration (IC50) values of the icELISA for PbTx-2, PbTx-1, and PbTx-3 were 60.71, 52.61, and 51.83 μg/kg, respectively. Based on the broad-spectrum mAb 1D3, an icELISA was developed to determine brevetoxin levels. Using this approach, the limit of detection (LOD) for brevetoxin was 124.22 μg/kg and recoveries ranged between 89.08% and 115.00%, with a coefficient of variation below 4.25% in oyster samples. These results suggest that our icELISA is a useful tool for the rapid monitoring of brevetoxins in oyster samples.

Mannan-binding lectin (MBL) is a vital element in the host innate immune system, which is primarily produced by the liver and secreted into the circulation. Low serum level of MBL is reported to be associated with an increased risk of arthritis. However, the underlying mechanism by which MBL contributes to the pathogenesis of arthritis is poorly understood. In this study, we investigated the precise role of MBL on the course of experimental murine adjuvant-induced arthritis (AIA). MBL-deficient (MBL−/−) AIA mice showed significantly increased inflammatory responses compared with wild-type C57BL/6 AIA mice, including exacerbated cartilage damage, enhanced histopathological features and high level of tartrate-resistant acid phosphatase (TRAP)-positive cells. MBL protein markedly inhibited the osteoclast formation from human blood monocytes induced by receptor activator of nuclear factor-κB ligand (RANKL) and macrophage colony-stimulating factor (M-CSF) in vitro. Mechanistic studies established that MBL inhibited osteoclast differentiation via down-regulation of p38 signaling pathway and subsequent nuclear translocation of c-fos as well as activation of nuclear factor of activated T-cells c1 (NFATc1) pathway. Importantly, we have provided the evidence that concentrations of MBL correlated negatively with the serum levels of amino-terminal propeptide of type I procollagen (PINP) and C-terminal telopeptide of type I collagen (β-CTX), serum markers of bone turnover, in patients with arthritis. Our study revealed an unexpected function of MBL in osteoclastogenesis, thus providing new insight into inflammatory arthritis and other bone-related diseases in patients with MBL deficiency.

Xanthones are secondary metabolites found in plants, fungi, lichens, and bacteria from a variety of families and genera, with the majority found in the Gentianaceae, Polygalaceae, and Clusiaceae. They have a diverse range of bioactivities, including anti-oxidant, anti-bacterial, anti-malarial, anti-tuberculosis, and cytotoxic properties. Xanthone glucosides are a significant branch of xanthones. After glycosylation, xanthones may have improved characteristics (such as solubility and pharmacological activity). Currently, no critical review of xanthone glucosides has been published. A literature survey including reports of naturally occurring xanthone glucosides is included in this review. The isolation, structure, bioactivity, and synthesis of these compounds were all explored in depth.

This paper investigates the problem of H∞ stabilization for nonuniform sampling fuzzy systems. To achieve better performance, a parallel distributed compensation scheme is proposed to design a nonlinear controller. By taking the deviation bounds of asynchronous normalized membership functions into account, the H∞ stability criterion is first obtained, which is dependent on the deviation bounds of asynchronous normalized membership functions. To reduce the computational complexity, a method for eliminating slack variables and a structured vertex separator for reducing the number of linear matrix inequalities are provided, and then an H∞ stabilization criterion with less complexity and less conservatism is obtained. Finally, an illustrative example is given to show the effectiveness of the proposed method and the significant improvement on the existing results.

To achieve maximum profit by dispatching a battery storage system in an arbitrage operation, multiple factors must be considered. While revenue from the application is determined by the time variability of the electricity cost, the profit will be lowered by costs resulting from energy efficiency losses, as well as by battery degradation. In this paper, an optimal dispatch strategy is proposed for storage systems trading on energy arbitrage markets. The dispatch is based on a computationally-efficient implementation of a mixed-integer linear programming method, with a cost function that includes variable-energy conversion losses and a cycle-induced battery capacity fade. The parametrisation of these non-linear functions is backed by in-house laboratory tests. A detailed analysis of the proposed methods is given through case studies of different cost-inclusion scenarios, as well as battery investment-cost scenarios. An evaluation with a sample intraday market data set, collected throughout 2017 in Germany, offers a potential monthly revenue of up to 8762 EUR/MWh cap installed capacity, without accounting for the costs attributed to energy losses and battery degradation. While this is slightly above the revenue attainable in a reference application—namely, primary frequency regulation for the same sample month (7716 EUR/MWh cap installed capacity)—the situation changes if costs are considered: The optimisation reveals that losses in battery ageing and efficiency reduce the attainable profit by up to 36% for the most profitable arbitrage use case considered herein. The findings underline the significance of considering both ageing and efficiency in battery system dispatch optimisation.
