Building a more resilient response system to climate change for sustainable development and reducing uncertainty in China's food markets, requires access to historical research gaps and mapping future research progress for decision making. However, the lack of quantitative and objective analyses to ensure the stability and development of agroecosystems increases the complexity of agro-climatic mechanisms, which leads to uncertainty and undesirable consequences. In this paper, we review the characteristics of climate change in China (1951–2020), reveal the mechanisms of agroecosystem structure in response to climate, and identify challenges and opportunities for future efforts in the context of research progress. The aim is to improve the scientific validity and relevance of future research by clarifying agro-climatic response mechanisms. The results show that surface temperature, precipitation, and frequency of extreme weather events have increased to varying degrees in major agricultural regions of China in 1951–2020. And they have strong geographic variation, which has resulted in droughts in the north and floods in the south. Moreover, climate change has complicated the mechanisms of soil moisture, Net Primary Productivity (NPP), soil carbon pool, and crop pest structure in agroecosystems. This lends to a reduction in soil water holding capacity, NPP, soil carbon content, and the number of natural enemies of diseases and insects, which in turn affects crop yields. However, human interventions can mitigate the deterioration of these factors. We have also realized that the methodology and theory of historical research poses a great challenge to future agroecosystem. Historical and projected climate trends identified current gaps in interdisciplinary integration and multidisciplinary research required to manage diverse spatio-temporal climate change impacts on agroecosystems. Future efforts should highlight integrated management and decision making, multidisciplinary big data coupling, and numerical simulations to ensure sustainable agricultural development, ecological security, and food security in China. Porcine reproductive and respiratory syndrome (PRRS) is an important viral disease, causing significant economic losses to the swine industry worldwide. Atypical cases caused by highly pathogenic PRRS virus (HP-PRRSV) emerged in 2006 in China. The vaccine strain HuN4-F112 has been developed from the wild-type HP-PRRSV HuN4 through repeated passages on MARC-145 cells. However, the mechanisms of attenuation have yet to be defined. Previous studies have shown that the vaccine strain HuN4-F112 could not effectively replicate in porcine alveolar macrophages (PAMs). In the present study, a series of chimeric and mutant PRRSVs were constructed to investigate regions associated with the virus attenuation. Firstly, the corresponding genome regions (ORF1a, ORF1b and ORFs 2-7) were exchanged between two infectious clones of HuN4 and HuN4-F112, and then the influence of small regions in ORF1a and ORF2-7 was evaluated, then influence of specific amino acids on NSP2 was tested. NSP2 was determined to be the key gene that regulated infection efficiency on PAMs, and amino acids at 893 and 979 of NSP2 were the key amino acids. The results of in vivo study indicated that NSP2 was not only important for infection efficiency in vitro, but also influenced the virulence, which was indicated by the results of survival rate, temperature, viremia, lung score and tissue score. We investigate the dual-band unidirectional reflectionless phenomenon and near perfect absorption using high-order plasmon resonators at exceptional points based on near-field coupling in plasmonic waveguide system. The system consists of two high-order plasmon resonators end-coupled to a metal-insulator-metal plasmonic waveguide. The reflection for forward (backward) direction is close to 0.85 (0.74), while for backward (forward) direction is approximate to 0 (0) at a frequency of 199.8 THz (206.1 THz), and the quality factor of absorption spectrum is about 257.6 (217.2) at 206.1 THz (199.8 THz) for forward (backward) direction. In recent years, the conversion of saline-alkali land to rice fields has become the most dominant land use change feature in western Jilin, leading to significant surface greening. Saline–alkali land and paddy fields have distinct surface biophysical properties; however, there is a lack of systematic assessment of the moderating effect of planting rice on saline–alkali land on regional climate by changing surface properties. In this paper, multiscale data on the surface temperature of saline–alkali land and paddy fields were obtained using 1 km MODIS product, 30 m Landsat 8 satellite imagery and centimeter-scale UAV imagery in Da'an City, western Jilin as the study area, and the various characteristics of the surface temperature of saline-alkali land and paddy fields in different months of the year and at different times of the day were analyzed. Furthermore, the effect of rice cultivation in saline–alkali land on the local surface temperature was assessed using a space-for-time approach. The results based on satellite observations including both MODIS and Landsat showed that the surface temperature of saline–alkali land was significantly higher than that of paddy fields during the crop growing season, especially in July and August. The high temporal resolution MODIS LST data also indicated the paddy fields cool the daytime surface temperature, while warming the nighttime surface temperature, which was in contrast for saline–alkali land during the growing season. High-resolution UAV observations in July confirmed that the cooling effect of paddy fields was most significant at the middle of day. From the biophysical perspective, the reclamation of saline–alkali land into paddy fields leads to an increase in leaf area index, followed by a significant increase in evapotranspiration. Meanwhile, rice cultivation in saline–alkali land reduces surface albedo and increases surface net radiation. The trade-off relationship between the two determines the seasonal difference in the surface temperature response of saline–alkali land for rice cultivation. At the same time, the daily cycle of crop evapotranspiration and the thermal insulation effect of paddy fields at night are the main reasons for the intraday difference in surface temperature between saline–alkali land and paddy field. Based on the multiscale assessment of the impact of rice cultivation in saline-alkali land on surface temperature, this study provides a scientific basis for predicting future regional climate change and comprehensively understanding the ecological and environmental benefits of saline–alkali land development. Although desertification has greatly increased across the Mongolian Plateau during the last decades of the 20th century, recent satellite records documented increasing vegetation growth since the 21st century in some areas of the Mongolian Plateau. Compared to the study of desertification, the opposite characteristics of land use and vegetation cover changes and their different effects on regional land–atmosphere interaction factors still lack enough attention across this vulnerable region. Using long-term time-series multi-source satellite records and regional climate model, this study investigated the climate feedback to the observed land surface changes from the 1990s to the 2010s in the Mongolia Plateau. Model simulation suggests that vegetation greening induced a local cooling effect, while the warming effect is mainly located in the vegetation degradation area. For the typical vegetation greening area in the southeast of Inner Mongolia, latent heat flux increased over 2 W/m2 along with the decrease of sensible heat flux over 2 W/m2, resulting in a total evapotranspiration increase by 0.1~0.2 mm/d and soil moisture decreased by 0.01~0.03 mm/d. For the typical vegetation degradation area in the east of Mongolia and mid-east of Inner Mongolia, the latent heat flux decreased over 2 W/m2 along with the increase of sensible heat flux over 2 W/m2 obviously, while changes in moisture cycling were spatially more associated with variations of precipitation. It means that precipitation still plays an important role in soil moisture for most areas, and some areas would be at potential risk of drought with the asynchronous increase of evapotranspiration and precipitation. To mitigate the impacts of land degradation and achieve sustainable development, several large-scale conservation programs including the Key Shelterbelt Construction Program, Natural Forest Conservation Program, and the Grain to Green Program have been formulated into action in China since the late 1970s and 1998, respectively [1]. However, a study based on an ESM indicated that in North and Southeast China, the increased precipitation from vegetation greening, although statistically insignificant, may cancel out enhanced evapotranspiration, resulting in weak impact on soil moisture [5]. • Crop greening slightly decreased regional soil moisture but showed spatial variations. • Landscape pattern can affect how rainfall and soil moisture responds to crop greening. • Background precipitation increase masked the moisture deficit caused by crop greening. The discrepancy between water resource availability and agricultural production has become a dominant constraint on the sustainable development of agriculture. Recent satellite records have documented increasing vegetation growth since the 21st century, and this so-called "vegetation greening" has been unprecedented in the agricultural region of Northeast China. However, the influence of increased crop growth on regional water resources through the alteration of hydrological cycling remains unclear. This study investigated the impact of crop greening on the regional water balance in Northeast China using time-series satellite and observational records, machine learning, and numerical scenario simulations from WRF. Our results indicate that the enhanced water cycling during 2001–2020 in this region was characterized by a significant increasing trend in evapotranspiration, precipitation, and soil moisture. We found a complex sensitivity to the leaf area index (LAI) across Northeast China, with positive sensitivity in the southwestern regions and negative sensitivity in the typical black soil zones. The downwind precipitation induced by crop greening suggests that at the regional scale, the landscape pattern may play a more significant role in determining the water cycling response to crop greening. Our data-driven and model-driven results indicate that the background precipitation increase was the dominant contributor to the soil moisture increase, which masked the water deficit caused by crop greening. We also found that the continuous precipitation increasing trend projected by CMIP6 could further relieve conflicts between agriculture and water resources. In summary, our study provides insights into the impact of crop greening on the regional water balance in Northeast China and highlights the importance of considering both landscape patterns and background precipitation changes in understanding the water cycling response to crop greening. Over the last six decades, paddy fields on the Sanjiang Plain have experienced rapid expansion and aggregation. In our study, land use and land cover changes related to paddy fields were studied based on information acquired from topographic maps and remote-sensing images. Paddy field expansion and aggregation were investigated through landscape indices and trajectory codes. Furthermore, the possible causes of paddy field expansion and aggregation were explored. Results indicated that such fields have increased by approximately 42,704 ha·y-1 over the past six decades. Approximately 98% of paddy fields in 2015 were converted from other land use types. In general, the gravity center moved 254.51 km toward the northeast, at a rate of approximately 4.17 km·y-1. The cohesion index increased from 96.8208 in 1954 to 99.5656 in 2015, and the aggregation index grew from 91.3533 in 1954 to 93.4448 in 2015, indicating the apparent aggregation of paddy fields on the Sanjiang Plain. Trajectory analyses showed that the transformations from marsh as well as from grassland to dry farmland and then into paddy fields were predominant. Climate warming provided a favorable environment for rice planting. The snow cover extent in mid-high latitude areas of the Northern Hemisphere has significantly declined corresponding to the global warming, especially since the 1970s. Snow-climate feedbacks play a critical role in regulating the global radiation balance and influencing surface heat flux exchange. However, the degree to which snow cover changes affect the radiation budget and energy balance on a regional scale and the difference between snow-climate and land use/cover change (LUCC)-climate feedbacks have been rarely studied. In this paper, we selected Heilongjiang Basin, where the snow cover has changed obviously, as our study area and used the WRF model to simulate the influences of snow cover changes on the surface radiation budget and heat balance. In the scenario simulation, the localized surface parameter data improved the accuracy by 10 % compared with the control group. The spatial and temporal analysis of the surface variables showed that the net surface radiation, sensible heat flux, Bowen ratio, temperature and percentage of snow cover were negatively correlated and that the ground heat flux and latent heat flux were positively correlated with the percentage of snow cover. The spatial analysis also showed that a significant relationship existed between the surface variables and land cover types, which was not obviously as that for snow cover changes. Finally, six typical study areas were selected to quantitatively analyse the influence of land cover types beneath the snow cover on heat absorption and transfer, which showed that when the land was snow covered, the conversion of forest to farmland can dramatically influence the net radiation and other surface variables, whereas the snow-free land showed significantly reduced influence. Furthermore, compared with typical land cover changes, e.g., the conversion of forest into farmland, the influence of snow cover changes on net radiation and sensible heat flux were 60 % higher than that of land cover changes, indicating the importance of snow cover changes in the surface-atmospheric feedback system. In the context of global climate change, the extent of snow cover in Siberia has significantly decreased since the 1970s, especially in spring. The changes of snow cover at middle and high latitudes have significant impacts on the meteorological and hydrological processes because the snow cover can affect the surface energy, water balance, and the development of the atmospheric boundary layer. In this paper, the temporal and spatial changes in snow cover were firstly estimated based on a long time series of remote sensing snow cover data, both showing a decreased trend. Based on this, we estimated the radiative forcing caused by the snow cover changes from the 1970s to the 2010s and compared it with the radiative forcing caused by the vegetation cover changes over the same time period in Siberia, indicating that the snow cover changes in Siberia can accelerate climate warming and the vegetation cover changes here have the opposite effect. Furthermore, the snow cover changes may play a more important role than the vegetation cover changes in regulating the surface radiation balance in Siberia on the regional scale. Understanding the interactions between temperature regimes and vegetation cover is one of the key scientific topics in global environmental change and land surface process research. Recently, the prominent global greening trend has inspired considerable interest in examining climate feedbacks to vegetation changes. However, the impacts of recent widespread vegetation cover changes on soil temperature have been less documented and have received insufficient attention. Here, a high-resolution regional climate model was used to examine the potential impact of vegetation cover changes over the past 37 years, obtained as satellite observational data, on regional air and soil temperatures across the Heilong-Amur River Basin. Our sensitivity experiments indicated that the 1.85% increase in the regional fractional vegetation cover (FVC) from 1982 to 2018 cooled the air temperature by 0.045 °C and the soil temperature by 0.19 °C and that this cooling effect would continue in the near future, from 2016 to 2053. We found that soil temperatures were more sensitive to vegetation cover changes than air temperatures, particularly in mid-high latitude forests; this finding helps to explain the nonsignificant decrease in soil temperatures in forests over the past 40 years detected in the ERA5-Land reanalysis. Northeast China is an important commodity grain base in China and plays an important role in maintaining national and local food security. The intensifying climate variability and strengthening of extreme weather events threaten crop growth and yields in this region. The dynamic crop model coupled into the land surface models showed advances in simulating crop phenology and crop growth response to climate change, however, which was mainly validated for the United States. By comparing with the satellite observed crop growth cycles, we firstly calibrated the parameters for the Weather Research and Forecasting (WRF) coupled Noah-MP-Crop model. Then, we evaluated the performance of the calibrated WRF-Noah-MP-Crop model with the default crop models and the dynamic vegetation models coupled with WRF by comparing our simulations with corresponding satellite measurements and meteorological observations. Our results indicated the augmentation of the Noah-MP-Crop model in WRF significantly improved the crop phenology simulation at the beginning and ending time of the growing season compared with the dynamic vegetation model using unmanaged crops. The satellite-calibrated crop parameters substantially improved the simulation of crop growth, plant physiology, and biomass accumulation for both corn and soybean. Coupling the localized dynamic crop model into the WRF led to considerable decreases in the simulated mean-absolute-errors (MAEs) and biases of the leaf area index, evapotranspiration, and gross primary production compared with the MODIS observed values. Compared with the statistical yield from each province, the modified crop model underestimated the corn yield from 11.1% to 48.6%, whereas overestimated the soybean yield from 16.5% to 162.6%. The coupled WRF-Noah-MP-Crop model with flexible resolutions holds unmatched advantages in estimating and projecting crop growth and yield under future climate change scenarios, which would be critical in ensuring sustainable agricultural development and maintaining future food security. Land cover change, as one of the most important driving forces to climate change, has become the research focus of the global environmental change research and global land project. More researchers studied on the global influence of Land-Use and Land-Cover Change and proved that land use change occurred at different temperature zones may produce different climate effects. For example, deforestation in tropical areas would lead to higher temperatures as the decreasing of evapotranspiration caused by the reduction of roughness and the decreasing of drag coefficient and leaf area index while, in boreal areas, similar deforestation would cause lower temperature as the increasing of albedo particularly during winter with the snow cover. However, the impact of deforestation in the temperate regions on the climate still existed uncertainty and the impacts of deforestation at different humidity conditions on climate has not explored yet. From this perspective, this article used Weather Research and Forecasting model to simulate the impact of deforestation on the temperature of Northeastern China. In this study, we designed two scenarios in July and December, respectively: One was simulated without human intervention, and the second one was simulated with the current forest covers. The results showed that the temperature in both summer and winter showed a decreasing trend when the conversion of forest to farmland occurred in northeastern China. In order to further explore the humidity impacts on the temperature, we performed sample analysis on humid, sub-humid, and semi-arid regions. According to the results, the maximum variation of temperature was found in humid areas, especially in December when the temperature decreased around 4-5 °C, while the change in semi-arid and sub-humid areas is relatively small. Ecotones have received great attention due to its critical function in energy flux, species harbor, global carbon sequestration, and land-atmosphere interaction. This study investigated land use pattern and spatial heterogeneity of the ecotones among agricultural land, forest land, and grassland of the southeastern Da Hinggan Mountains in the northeastern China. The change of these delineated ecotones under different slopes and aridity conditions was analyzed by two landscape indices, edge density (ED) and core area percentage of landscape (CPL), to explore the inter-linkage between spatial structure of ecotones and socioeconomic development and land management. Specifically, the ecotones such as agriculture-forest (AF) ecotone, forest-grassland (FG) ecotone, and agriculture-forest-grassland (AFG) ecotone moved from the arid southeast to the humid northwest. The flat area with small slope is more edge-fragmented than the steep area since the ED decreases as the slope increases. The AF ecotone mostly found in the humid region is moving to more humid areas while the agriculture-grassland (AG) ecotone mostly found in the dry region is moving towards the drier region. Porcine epidemic diarrhea virus (PEDV), a member of the genera alphacoronavirus, causes acute watery diarrhea and dehydration in suckling piglets and results in enormous economic losses in the swine industry worldwide. Identification and characterization of different cell lines are not only invaluable for PEDV entry and replication studies but also important for the development of various types of biological pharmaceuticals against PEDV. In this study, we present an approach to identify suitable permissive cell lines for PEDV research. Human cell lines were screened for a high correlation coefficient with the established PEDV infection model Huh7 based on RNA-seq data from the Cancer Cell Line Encyclopedia (CCLE). Experimentally testing permissiveness towards PEDV infection, three highly permissive human cell lines, HepG2, Hep3B217, and SNU387 were identified. The replication kinetics of PEDV in HepG2, Hep3B217, and SNU387 cells were similar to that in Vero and Huh7 cells. Additionally, the transcriptomes analysis showed robust induction of transcripts associated with the innate immune in response to PEDV infection in all three cell lines, including hundreds of inflammatory cytokine and interferon genes. Moreover, the expression of inflammatory cytokines and interferons were confirmed by qPCR assay. Our findings indicate that HepG2, Hep3B217, and SNU387 are suitable cell lines for PEDV replication and innate immune response studies. [ABSTRACT FROM AUTHOR]

Studying land use change and its associated climate effects is important to understand the role of human activities in the regulation of climate systems. By coupling remote sensing measurements with a high-resolution regional climate model, this study evaluated the land surface changes and corresponding climate impact caused by planting rice on saline-alkali land in western Jilin (China). Our results showed that paddy field expansion became the dominant land use change in western Jilin from 2015 to 2019, 25% of which was converted from saline-alkali land; this percentage is expected to increase in the near future. We found that saline-alkali land reclamation to paddy fields significantly increased the leaf area index (LAI), particularly in July and August, whereas it decreased albedo, mainly in May and June. Our simulation results showed that planting rice on saline-alkali land can help decrease the air temperature and increase the relative humidity. The temperature and humidity effects showed different magnitudes during the growing season and were most significant in July and August, followed by September and June. The nonradiative process, rather than the radiative process, played a dominant role in regulating the regional climate in this case, and the biophysical competition between evapotranspiration (ET) and albedo determined the temperature and relative humidity response differences during the growing season. • Widespread greening cools surface and increases rainfall in summer over China. • Climatic responses to the greening showed significant regional variability. • Vegetation greening would decrease the risk of extreme climate events. Changes in land-use management and natural climate variability has led to significant vegetation greening in China over the past decades, as evidenced by satellite observations. In this study, we used a high-resolution land-atmosphere coupled regional climate model to assess the impacts of recent greening on China's regional summer climate. Our results inferred a country-averged surface cooling of 0.11 °C and a precipitation increase of 0.02 mm/d during summer in response to China's widespread greening. We also identified significant regional variability in the climatic responses to vegetation change. The regions of northern China showed significant cooling, including the Northeast China Plain, Loess Plateau, and the eastern areas of the Northern arid and semi-arid region; however, no significant temperature changes were simulated in southern China. Summer precipitation decreased in southern China and increased in the Huang-Huai-Hai Plain and the northern Loess Plateau. Variations in moisture flux convergence had a stronger influence on the precipitation patterns relative to evapotranspiration. Furthermore, our results suggested that vegetation greening would influence the occurrence of extreme climate events by decreasing the number of heatwave days (by 1–3 days, particularly in northern China), reducing the number of continuous drought events in northern China, and minimizing the risks of floods in southern China. Land use and land cover change (LUCC) has been increasingly recognized as having important effects on climate systems. Paddy fields, one kind of artificial wetland, have seen a significant increase in the Sanjiang Plain, China since 2000 and have become the most typical LUCC at the regional scale. Against this background, in this paper, we discuss the effects of this artificial wetland increase on surface temperature, in addition to its driving mechanisms. Firstly, the spatiotemporal variations of land surface temperature (LST) and its two driving variables (albedo and latent heat flux (LE)) in the Sanjiang Plain are analyzed and assessed based on remote sensing observation information from 2001 to 2015. Our results from both spatial distribution difference and time series analysis show that paddy field expansion led to day-time cooling and night-time warming over the study area. However, the LST changes show different characteristics and magnitudes in the spring (May to June) compared to the other months of the growing season (July to September). The daytime cooling trend is found to be −0.3842 K/year and the warming trend at night 0.1988 K/year during the period 2001 to 2015, resulting in an overall cooling effect in May and June. In July–September, the LST changes have the same sign but a smaller magnitude, with a −0.0686 K/year temperature trend seen for the day-time and a 0.0569 K/year increase for the night-time. As a consequence, a pronounced decrease in the diurnal temperature range is detected in the growing season, especially in spring. Furthermore, albedo and LE are demonstrated to be very sensitive to land use changes, especially in the earlier periods of the growing season. Early and accurate prediction and simulation of grain crop yield can help maximize the revision and development of regional food policy, which is crucial for ensuring national food security. The development of unmanned aerial vehicle (UAV) technology is gradually gaining an advantage over satellite remote sensing at the field scale. In this study, we predicted maize yield using canopy vegetation indices (VIs) and crop phenology metrics obtained through UAV with ordinary least squares (OLS), stepwise multiple linear regression (SMLR) and gradient‐boosted regression tree (GBRT). The results reveal that the VIs extracted from UAV imagery had a high correlation with yield (R = 0.92), facilitating crop yield estimation. Additionally, coupling crop phenology significantly improved the prediction accuracy of SMLR, with the highest R2 and lowest RMSE of 0.894, 1.238 × 103 kg ha−1, respectively. But, the enhancement of GBRT by this method was slender. Its simulation outperformed OLS and SMLR with dramatic R2, RMSE, and MAE of 0.892, 1.189 × 103 kg ha−1, and 9.150 × 102 kg ha−1, respectively. Moreover, the blister stage was deemed the optimal stage for maize yield prediction with an accuracy rate exceeding 81%. Porcine reproductive and respiratory syndrome (PRRS) is one of the major drivers of economic loss in the swine industry worldwide. In commercial pig production, vaccination is the first option in an attempt to control infectious diseases. Pigs are therefore often immunized with different vaccines, and almost all of them are delivered via the intramuscular (IM) route. However, the IM injection may result in physical damage, stress reactions, and is labor demanding. An alternative route is urgently needed to reduce the disadvantages of conventional vaccination. In this study, a needle-free intradermal (ID) delivery system was evaluated for delivering a live PRRS vaccine as compared with the traditional needle-syringe method. Fifty-two 4-week-old piglets were divided into six groups: piglets in groups A-C were immunized using ID delivery system with 104, 105 and 106 TCID 50 of PRRS candidate vaccine strain rHN-NP49, respectively; piglets in group D were immunized IM with 105 TCID 50 of rHN-NP49; and group E and F were used as challenge and control groups, respectively. At 28 days post vaccination, piglets in group A to E were challenged with a lethal dose of highly-pathogenic PRRSV. Similar results were found in viremia and antibody response among the ID and IM groups during the immunization stage. After challenge, similar results were found in average body weight gain, viral shedding, serum viral load, and clinical score among the immunization groups, with a higher protection ratio in the ID group compared with IM group with the same immunization dose. These results demonstrated that the ID delivery system could provide similar or even better protection compared with IM route, and could be an effective route for PRRS vaccination. The irrigated paddy fields have expanded greatly at semi-arid western Jilin province of China in recent over ten years, the sources of which are rain-fed cornfields, swamp meadow and saline alkali land mainly. Based on regional land use data, remote sensing data and meteorological data, this paper evaluates the land surface temperature changes response to land surface biophysical processes changes resulting from land use change (LUC), and dissociates the effect of radiative change (albedo) and non-radiative change (evapotranspiration and turbulent process) quantitatively using the energy redistribution factor. The results show that, the total land surface temperature changes based on energy redistribution factor are consistent with that based on remote sensing data on the whole, which have significant and different seasonal variations for agriculture adjustment of rain-fed cornfields to irrigated paddy fields and nature land reclamation. Generally, the largest Land surface temperature changes (ΔTs) are most pronounced in May and June for agriculture adjustment of rain-fed cornfields to irrigated paddy fields, which is −1.85 K averagely. Notable decline of albedo from saline alkali land to irrigated paddy fields in April to June greatly counteracts the cooling effect of non-radiative processes changes, while the largest ΔTs is found of −2. 54 K in dry summer months of July and August. For swamp meadows to irrigated paddy fields, non-radiative process is strengthened from June to September, the cooling effect of which is −1.69 K averagely. This study provides a case reference of local temperature change and obvious changes of land surface non-radiative terms at semi-arid area for adjustment of agricultural activities and land use changes. Numerous studies have documented the effects of irrigation on local, regional, and global climate. However, most studies focused on the cooling effect of irrigated dryland in semiarid or arid regions. In our study, we focused on irrigated paddy fields in humid regions at mid to high latitudes and estimated the effects of paddy field expansion from rain-fed farmland on local temperatures based on remote sensing and observational data. Our results revealed much significant near-surface cooling in spring (May and June) rather than summer (July and August) and autumn (September), which was −2.03 K, −0.73 K and −1.08 K respectively. Non-radiative mechanisms dominated the local temperature response to paddy field expansion from rain-fed farmland in the Sanjiang Plain. The contributions from the changes to the combined effects of the non-radiative process were 123.6%, 95.5%, and 66.9% for spring (May and June), summer (July and August), and autumn (September), respectively. Due to the seasonal changes of the biogeophysical properties for rain-fed farmland and paddy fields during the growing season, the local surface temperature responses, as well as their contributions, showed great seasonal variability. Our results showed that the cooling effect was particularly obvious during the dry spring instead of the warm, wet summer, and indicated that more attention should be paid to the seasonal differences of these effects, especially in a region with a relatively humid climate and distinct seasonal variations. Porcine reproductive and respiratory syndrome virus (PRRSV) is globally prevalent and seriously harms the economic efficiency of pig farming. Because of its immunosuppression and high incidence of mutant recombination, PRRSV poses a great challenge for disease prevention and control. Nonstructural protein 2 (NSP2) is the most variable functional protein in the PRRSV genome and can generate NSP2N and NSP2TF variants due to programmed ribosomal frameshifts. These variants are broad and complex in function and play key roles in numerous aspects of viral protein maturation, viral particle assembly, regulation of immunity, autophagy, apoptosis, cell cycle and cell morphology. In this paper, we review the structural composition, programmed ribosomal frameshift and biological properties of NSP2 to facilitate basic research on PRRSV and to provide theoretical support for disease prevention and control and therapeutic drug development. Diarrhea outbreaks in piglets on pig farms are commonly attributed to porcine epidemic diarrhea virus (PEDV) infection. This research analyzed the S gene prevalence variation and recombination patterns in PEDV GII strains. Throughout the previous two years, 172 clinical samples of piglet diarrhea have been collected, from which 24 PEDV isolates have been isolated. Analysis of the evolutionary relationships among all 24 S genes revealed that 21 were most closely related to strains within the GII-a subgroup. The 2 isolates grouped into one clade with the GII-b subgroup. According to the mutation analysis of the amino acids (aa) that encode the S protein, 43 of the common aa in strains of the GII subtype were found to have undergone a change in polarity or charge, and 36 of these aa had a mutation frequency of more than 90%. Three different aa mutation sites were identified as exclusive to GII-a subtype strains. The genomes of three PEDV isolates were sequenced, and the resulting range in genome length was 28,035−28,041 nt. The results of recombination analysis showed that the SD1 isolate is a novel strain recombinant from the foreign S-INDEL strain and a domestic GII subtype strain. Based on the findings, the PEDV GII-a strain has been the most circulating strain in several parts of China during the previous two years. Our study reveals for the first time the unique change of aa mutations in the S protein of the GII-a subtype strain and the new characteristics of the recombination of foreign strains and domestic GII subtype strains, indicating that it is crucial to monitor the epidemic dynamics of PEDV promptly to prevent and control the occurrence of PED effectively. The Fifth IPCC Assessment Report indicates that climate change will affect crop growth and threaten the stability of food systems. Accumulated temperature, which is closely related to vegetation phenology and cropping systems, is an important indicator of heat in a region. Studying the history and future accumulated temperature changes can provide scientific reference for the change of crop phenology and cropping system, which is important for the improvement of grain production in China. Based on the MK trend test, MK abrupt change test and interpretable machine learning model, this study analyzes the spatial and temporal variation of accumulated temperature in China from 1979 to 2018, predicts its future variation based on CMIP6, and investigates the dominant influencing factors among different agricultural regions. The study found that (1) the accumulated temperature belt shows a northward shift and retreat trend toward higher altitudes, and the area of the high accumulated temperature belt increases year by year, leading to the narrowing of the area of the low accumulated temperature belt year by year, and the trend remains unchanged under the future scenario; meanwhile, the northward shift trend of the accumulated temperature belt is greatly mitigated and curbed under the SSP126 scenario. (2) The changes of accumulated temperature belt are mainly influenced by the increase of accumulated temperature duration days, and secondarily by the increase of temperature. The contribution brought by the first day of accumulated temperature from 1979 to 2018 is greater than that brought by the last day of accumulated temperature, while in the future scenario, on the contrary, changes in vegetation phenology delay should be given more attention. The Northeast China is one of typical regions experiencing intensive human activities within short time worldwide. Particularly, as the significant changes of agriculture land and forest, typical characteristics of pattern and process of agroforestry ecotone change formed in recent decades. The intensive land use change of agroforestry ecotone has made significant change for regional land cover, which had significant impact on the regional climate system elements and the interactions among them. This paper took agroforestry ecotone of Nenjiang River Basin in China as study region and simulated temperature change based on land cover change from 1950s to 1978 and from 1978 to 2010. The analysis of temperature difference sensitivity to land cover change based on Weather Research and Forecasting (WRF) model showed that the land cover change from 1950s to 1978 induced warming effect over all the study area, including the change of grassland to agriculture land, grassland to deciduous broad-leaved forest, and deciduous broad-leaved forest to shrub land. The land cover change from 1978 to 2010 induced cooling effect over all the study area, including the change of deciduous broad-leaved forest to agriculture land, grassland to agriculture land, shrub land to agriculture land, and deciduous broad-leaved forest to grassland. Highly pathogenic porcine reproductive and respiratory syndrome virus PRRSV (HP-PRRSV) was one of the most devastating diseases of the pig industry, among various strategies, vaccination was one of the most useful tools for PRRS control. Attenuated live vaccine was used worldwide, however, the genetic basis of HP-PRRSV virulence change during attenuation remain to be determined. Here, to identify virulence associated regions of HP-PRRSV during attenuation in vitro, six full-length infectious cDNA clones with interchanges of 5′UTR + ORF1a, ORF1b, and ORF2-7 + 3′UTR regions between HP-PRRSV strain HuN4-F5 and its attenuated vaccine strain HuN4-F112 were generated, and chimeric viruses were rescued. Piglets were inoculated with chimeric viruses and their parental viruses, and rectal temperature were recorded daily, and serum were collected for future experiments. Our results showed that ORF1a played an important role on virus replication, cytokine response and lung damage, the exchange of ORF1b and ORF2-7 in different backbone led to different exhibition on virus replication in vivo/vitro and cytokine response. Among 9 PRRSV attenuated series, consistent amino acid changes during PRRSV attenuation were found in NSP4, NSP9, GP2, E, GP3 and GP4. Our study provides a fundamental data for the investigation of PRRSV attenuation, the different results of the virulence change among different studies indicated that different mechanisms might be used during PRRSV virulence enhancement in vivo and attenuation in vitro. Feline coronavirus (FCoV) is an unsegmented, single-stranded RNA virus belonging to the Alphacoronavirus genus. It can cause fatal feline infectious peritonitis (FIP) in cats of any ages. Currently, there are no effective prevention and control measures to against FCoV. In this study, we developed a recombinant adenovirus vaccine, AD5-N, based on the nucleocapsid(N) protein of FCoV. The immunogenicity of AD5-N was evaluated through intramuscular immunization in 6-week-old Balb/c mice and 9–12 months old cats. Compared to the control group, AD5-N specifically induced a significant increase in IgG and SIgA levels in the vaccinated mice. Furthermore, AD5-N not only effectively promoted strong cellular immune responses in cats but also induced high levels of specific SIgA, effectively helping cats resist FCoV infection. Tripartite motif protein 21 (TRIM21) is an E3 ubiquitin ligase and cytosolic antibody receptor of the TRIM family. Previous reports have indicated that TRIM21 plays an important role during viral infection. This study aimed at examining the role of TRIM21 in the replication of porcine epidemic diarrhea virus (PEDV) and showed that TRIM21 inhibits PEDV proliferation by targeting and degrading the nucleocapsid (N) protein through the proteasomal pathway. Furthermore, the endogenous expression of TRIM21 was found to be downregulated by PEDV infection in Vero and LLC-PK1 cells. Overexpression of TRIM21 inhibited PEDV replication, whereas knockdown of TRIM21 increased viral titers and N protein levels. TRIM21 was found to interact and colocalize with the N protein, and the TRIM21-mediated antiviral effect was dependent on its ubiquitin ligase activity, which engages in polyubiquitination and degradation of the N protein in a proteasome-dependent manner. MiR-7-5p has been demonstrated to inhibit tumorigenesis by limiting tumor cell proliferation, migration and invasion. However, its role in countering hydroquinone (HQ)-induced malignant phenotype of TK6 cells has remained unclear. The present study aimed to investigate whether miR-7-5p overexpression could restrain the malignant phenotype in TK6 cells exposed to HQ. The results displayed that HQ suppressed the expression of miR-7-5p and promoted cell cycle progression. Further investigations confirmed that miR-7-5p could decelerate the cell cycle progression by targeting Rb after acute HQ exposure. Through the regulation of the Rb/E2F1 signaling pathway, the overexpression of miR-7-5p mitigated HQ-induced malignant phenotype in TK6 cells by impeding cell cycle progression. In conclusion, miR-7-5p overexpression appears to be involved in HQ-induced malignant transformation by suppressing Rb/E2F1 signaling pathway, resulting in a deceleration of the cell cycle progression. We have previously demonstrated that rPRRSV-E2 is a promising live, virus-vectored vaccine that provides 100% protection against highly pathogenic PRRSV (HP-PRRSV) and CSFV. Here, we evaluated the duration of immunity (DOI) of the vaccine strain, rPRRSV-E2. Vaccine or cell culture medium was administered to piglets at 4 weeks of age. All immunized piglets developed high levels of antibodies, which could maintain for up to 23 weeks, against PRRSV and CSFV. All immunized pigs were well protected from the challenge of HP-PRRSV or CSFV at 20 weeks and 24 weeks post vaccination. The vaccine protection rate was still 100% at 24 weeks after immunization. Here, we screened a porcine alveolar macrophage (PAM) cDNA library with a yeast two-hybrid system to reveal that galectin-1 (Gal-1), an endogenous innate immune protein encoded by LGALS1 , interacts with nonstructural protein 11 (Nsp11) of PRRSV. Western blotting and viral titer assays indicated that Gal-1 overexpression suppressed replication in multiple PRRSV strains (P < 0.001), whereas Gal-1 knockdown or knockout increased viral titer and nucleocapsid protein expression. The Gal-1-specific anti-PRRSV effect was associated with the endoribonuclease domain of Nsp11 through inactivation of interferon-antagonist function and stimulation of interferon-stimulated gene expression. Additionally, Gal-1 interacted with PRRSV E protein but not with PRRSV glycoproteins, and recombinant Gal-1 treatment inhibited PRRSV in PAMs and MARC-145 cells. Furthermore, Gal-1 inhibited replication in multiple viruses, including equine arteritis virus, porcine epidemic diarrhea virus, pseudorabies virus, Japanese encephalitis virus, and classical swine fever virus, suggesting its potential broad application for antiviral strategies. Abstract In recent years, the outbreaks of porcine epidemic diarrhea (PED) caused by the highly virulent porcine epidemic diarrhea virus (PEDV) variants occurred frequently in China, resulting in severe economic impacts to the pork industry. In this study, we selected and analyzed the genetic evolution of 15 PEDV representative strains that were identified in fecal samples of diarrheic piglets in 10 provinces and cities during 2011–2017. The phylogenetic analysis indicated that all the 15 PEDV isolates clustered into G2 genotype associated with the current circulating strains. Compared with the genome of the prototype strain CV777, these strains had 103–120 amino acid mutations in their S proteins, most of which were in the N terminal domain of S1 (S1-NTD). We also found 37 common mutations in all these 15 strains, although these strains shared 96.9–99.7% nucleotide homology and 96.3–99.8% amino acid homology in the S protein compared with the other original pandemic strains. Computational analysis showed that these mutations may lead to remarkable changes in the conformational structure and asparagine (N)-linked glycosylation sites of S1-NTD, which may be associated with the altered pathogenicity of these variant PEDV strains. We evaluated the pathogenicity of the PEDV strain FJzz1 in piglets through oral and intramuscular infection routes. Compared with oral infection, intramuscular infection could also cause typical clinical signs but with a slightly delayed onset, confirming that the variant PEDV isolate FJzz1 was highly pathogenic to suckling piglets. In conclusion, we analyzed the genetic variation and pathogenicity of the emerging PEDV isolates of China, indicating that G2 variant PEDV strains as the main prevalent strains that may mutate continually. This study shows the necessity of monitoring the molecular epidemiology and the etiological characteristics of the epidemic PEDV isolates, which may help better control the PED outbreaks. Understanding future climate change is crucial for securing and managing water supplies necessary for agricultural activities in China, particularly in rainfed agriculture regions. This paper evaluates the projected climate change for nine major agricultural zones in China, using ensembles of regional climate models (RCMs) simulated under the Coordinated Regional Downscaling Experiment-East Asia second phase (CORDEX-EA-II) framework. The simulations were conducted for two climate scenarios (RCP2.6 and RCP8.5) at a 25 km grid spacing, with three RCMs (RegCM4, REMO, and WRF) driven by multiple CMIP5 global climate models (GCMs). The study compares the RCM simulations to the driving CMIP5 and the recent CMIP6 ensembles, focusing on surface temperature, precipitation, and surface relative humidity for the present day (1981–2010), mid-future (2036–2065), and far future (2070–2099) periods. The model evaluations indicate that the GCM and RCM simulations are consistent with observations, with RCMs showing better performance than the driving GCMs. The results show that both GCMs and RCMs project increased temperature and precipitation in most agricultural zones of China in the future, with the CMIP6 ensembles projecting the largest temperature increase and the CORDEX-EA-II ensemble showing the finest spatial details. Relative humidity is projected to decrease over major rice-producing areas of South China in the CORDEX-EA-II, CMIP5, and CMIP6 ensembles. Porcine reproductive and respiratory syndrome (PRRS) and classical swine fever (CSF) are economically significant diseases that affect the swine industry worldwide. However, the current vaccination strategy, which uses two single live attenuated vaccines, can result in interference for each other. In addition, the universally used CSFV vaccine C-strain does not allow for differentiation of infected and vaccinated animals. In this study, rPRRSV-E2, PRRS virus (PRRSV) expressing CSF virus (CSFV) E2, was constructed by reverse genetics. The E2 gene of CSFV was inserted between ORF1b and ORF2 in the genome of the PRRS vaccine virus, HuN4-F112. A copy of transcriptional regulatory sequence 6 was inserted at the 3′ terminal of the exogenous gene to produce CSFV E2 as a unique subgenomic mRNA transcript. The rPRRSV-E2 was stable for at least 25 serial cell passages. Single-shot intramuscular immunization of rPRRSV-E2 into pigs induced PRRSV-specific and CSFV-specific antibodies and fully protected pigs from lethal challenge with highly-pathogenic PRRSV and CSFV. These results demonstrate that a novel strategy for recombinant PRRSV production is effective, and suggest that rPRRSV-E2 is a promising live, virus-vectored vaccine against PRRS and a marker vaccine against CSF. The marshes have undergone dramatic loss in the Sanjiang Plain since the 1950s. This paper analyzed the spatiotemporal changes of the marshes using the transition probabilities index, loss rate, and landscape indices. We also used a trajectory analysis method to trace every location of marshes over multiple points in time and to quantitatively estimate the impact of human activities on the marsh changes since the 1950s. This study indicates that the marsh area declined sharply by 79.4% (approximately 2.99 million ha) from 1954 to 2015. A large area of marsh was reclaimed as cultivated land from 1954 to 2015. The changes in the related landscape indices showed the large-scale loss of the marsh area and marsh fragmentation in the study period. Human activities were the dominant factor (87.86%) that influenced the marsh changes in the Sanjiang Plain during the last 60 years compared with natural factors. These findings can provide valuable information for better understanding wetlands changes and implementing sustainable management strategies for wetlands (such as wetland restoration). Wetland databases can provide the basic data that necessary for the protection and management of wetlands. A large number of wetland databases have been established in the world as well as in China. In this paper, we review China's wetland databases based on remote sensing (RS) technology after introducing the background theory to the application of RS technology in wetland surveys. A key conclusion is that China's wetland databases are far from sufficient in fulfilling protection and management needs. Our recommendations focus on the use of the hyper-spectral imagery, microwave data, multi-temporal images, and automatic classifications in order to improve the accuracy and efficiency of wetland inventory. Further, attention should also be paid to detect major biophysical features of wetlands and build wetland databases in years after the 1980s in China. Considering that great gap exists between RS experts and wetland experts, further cooperation between wetland scientists and RS scientists are needed to promote the application of RS in the foundation of wetland databases. KLF16, a member of KLFs (Krüppel-like factors), contributes to the progression of a variety of cancer types. There is, however, still uncertain regarding the role of KLF16 in viral replication and the signaling mechanism of type I IFN. It was discovered that KLF16 inhibited the replication of porcine epidemic diarrhea virus (PEDV) through the type I IFN signaling pathway. Besides, it can also be found that the expression of KLF16 was down-regulated after PEDV infection of LLC-PK1 cells. Furthermore, overexpression of KLF16 inhibited the replication of PEDV in Vero cells as well as LLC-PK1 cells, whereas the replication of PEDV was promoted by the knockdown of KLF16. KLF16 up-regulated the expression of interferons (IFNs) via the TRAF6-pTBK1-pIRF3 pathway with the aim of promoting the host antiviral innate immune response. Autophagy-related 4B (ATG4B) is found to exert a vital function in viral replication, although the mechanism through which ATG4B activates type-I IFN signaling to hinder viral replication remains to be explained, so far. The current work revealed that ATG4B was downregulated in porcine epidemic diarrhea virus (PEDV)-infected LLC-PK1 cells. In addition, ATG4B overexpression inhibited PEDV replication in both Vero cells and LLC-PK1 cells. On the contrary, ATG4B knockdown facilitated PEDV replication. Moreover, ATG4B was observed to hinder PEDV replication by activating type-I IFN signaling. Further detailed analysis revealed that the ATG4B protein targeted and upregulated the TRAF3 protein to induce IFN expression via the TRAF3-pTBK1-pIRF3 pathway. MicroRNAs (miRNAs) play important roles in viral infections, especially by modulating the expression of cellular factors essential to viral replication or the host innate immune response to infection. To identify host miRNAs important to controlling porcine reproductive and respiratory syndrome virus (PRRSV) infection, we screened 15 miRNAs that were previously implicated in innate immunity or antiviral functions. Over-expression of the miR-26 family strongly inhibited PRRSV replication in vitro, as shown by virus titer assays, Western blotting, and qRT-PCR assays. MiR-26a inhibited the replication of both type 1 and type 2 PRRSV strains. Mutating the seed region of miR-26 restored viral titers. Luciferase reporters showed that miR-26a does not target the PRRSV genome directly but instead affects the expression of type I interferon and the IFN-stimulated genes MX1 and ISG15 during PRRSV infection. These results demonstrate the important role of miR-26a in modulating PRRSV infection and also support the possibility of using host miR-26a to achieve RNAi-mediated antiviral therapeutic strategies. Land use and land cover change (LUCC) modeling has continuously been a major research theme in the field of land system science, which interprets the causes and consequences of land use dynamics. In particular, models that can obtain long-term land use data with high precision are of great value in research on global environmental change and climate impact, as land use data are important model input parameters for evaluating the effect of human activity on nature. However, the accuracy of existing reconstruction and prediction models is inadequate. In this context, this study proposes an integrated convolutional neural network (CNN) LUCC reconstruction and prediction model (CLRPM), which meets the demand for fine-scale LUCC reconstruction and prediction. This model applies the deep learning method, which far exceeds the performance of traditional machine learning methods, and uses CNN to extract spatial features and provide greater proximity information. Taking Baicheng city in Northeast China as an example, we verify that CLRPM achieved high-precision annual LUCC reconstruction and prediction, with an overall accuracy rate 9.38% higher than that of the existing models. Additionally, the error rate was reduced by 49.5%. Moreover, this model can perform multilevel LUCC classification category reconstructions and predictions. This study casts light on LUCC models within the high-precision and fine-grained LUCC categories, which will aid LUCC analyses and help decision-makers better understand complex land-use systems and develop better land management strategies. The temperate forests in Northeast China are an important ecological barrier. However, the way in which temperate forests regulate the regional temperature and water cycling remains unclear. In this study, we quantitatively evaluated the role that temperate forests play in the regulation of the regional temperature and precipitation by combining remote sensing observations with a state-of-the-art regional climate model. Our results indicated that the forest ecosystem could slightly warm the annual air temperature by 0.04 ± 0.02 °C and bring more rainfall (17.49 ± 3.88 mm) over Northeast China. The temperature and precipitation modification function of forests varies across the seasons. If the trees were not there, our model suggests that the temperature across Northeast China would become much colder in the winter and spring, and much hotter in the summer than the observed climate. Interestingly, the temperature regulation from the forest ecosystem was detected in both forested regions and the adjacent agricultural areas, suggesting that the temperate forests in Northeast China cushion the air temperature by increasing the temperature in the winter and spring, and decreasing the temperature in the summer over the whole region. Our study also highlights the capacity of temperate forests to regulate regional water cycling in Northeast China. With high evapotranspiration, the forests could transfer sufficient moisture to the atmosphere. Combined with the associated moisture convergence, the temperate forests in Northeast China brought more rainfall in both forest and agricultural ecosystems. The increased rainfall was mainly concentrated in the spring and summer; these seasons accounted for 93.82% of the total increase in rainfall. • rPRRSV-E2 is a marker vaccine for CSFV. • rPRRSV-E2 is promising as a virus-vectored vaccine against both HP-PRRSV and CSFV infection in swine herds by single vaccination. • Maternal derived antibodies (MDAs) do not interfere with the immune efficacy of rPRRSV-E2. Currently live attenuated porcine reproductive and respiratory syndrome (PRRS) and classical swine fever (CSF) vaccines are widely used in Chinese swine herds. However, the mutual effects of vaccination procedures and severe stress caused by successive vaccinations harm piglets and make it difficult to stimulate robust and effective immune responses. In our previous study, a recombinant PRRS virus (PRRSV) vectored vaccine candidate rPRRSV-E2, which expresses CSF virus (CSFV) E2 protein, has been demonstrated being able to protect piglets against lethal challenge of highly-pathogenic (HP)-PRRSV and CSFV. In this study, we determine whether preexisting maternally derived antibodies (MDA) interfere with the immune efficacy of rPRRSV-E2. 8 experimental groups of piglets, with or without PRRSV MDAs or CSFV MDAs were immunized with a single dose of 105 TCID 50 rPRRSV-E2 or DMEM and challenged with HP-PRRSV or CSFV. Clinical characteristics, PRRSV- or CSFV-specific antibodies, viremia and pathological changes were monitored, examined and analyzed. The results showed that rPRRSV-E2-vaccinated piglets, either with or without MDAs directed against PRRSV or CSFV were completely protected from the lethal challenge of HP-PRRSV or CSFV. These results demonstrate that the MDAs do not interfere with the immune efficacy of rPRRSV-E2, which indicates that rPRRSV-E2 could have great significance in the effective prevention and control of HP-PRRSV and CSFV. Porcine reproductive and respiratory syndrome virus (PRRSV) poses a significant threat to the swine industry worldwide, and the development of effective and sustainable measures to control PRRSV transmission remains a pressing problem. The function of PRRSV nonstructural protein 12 (Nsp12), which might play essential roles in viral replication and production, remains unknown. In this study, we identified a new host-restricted factor, porcine RING finger protein 114 (RNF114), as an inhibitor of PRRSV replication through its degradation of viral Nsp12. Western blot, quantitative real-time polymerase chain reaction, and viral titer assays indicated that RNF114 overexpression suppressed PRRSV replication, whereas RNF114 knockdown increased viral titer and nucleocapsid protein levels. Additionally, we observed that PPRSV infection led to increased RNF114 levels during the middle and late phases of infection in both porcine alveolar macrophages and MARC-145 cells. Moreover, screening of PRRSV Nsps showed that RNF114 interacted with viral Nsp12, and that RNF114-specific anti-PRRSV effects were associated with its ubiquitin ligase activity, which involves K27-linked polyubiquitination and degradation of Nsp12 through a proteasome-dependent pathway. Porcine reproductive and respiratory syndrome virus (PRRSV) is one of the most important pathogens in the swine industry worldwide. Our previous study had indicated that proprotein convertase subtilisin/kexin type 9 (PCSK9) was a responsive gene in porcine alveolar macrophages (PAMs) upon PRRSV infection. However, whether PCSK9 impacts the PRRSV replication and how the PRRSV modulates host PCSK9 remains elusive. Here, we demonstrated that PCSK9 protein suppressed the replication of both type-1 and type-2 PRRSV species. More specifically, the C-terminal domain of PCSK9 was responsible for the antiviral activity. Besides, we showed that PCSK9 inhibited PRRSV replication by targeting the virus receptor CD163 for degradation through the lysosome. In turn, PRRSV could down-regulate the expression of PCSK9 in both PAMs and MARC-145 cells. By screening the nonstructural proteins (nsps) of PRRSV, we showed that nsp11 could antagonize PCSK9's antiviral activity. Furthermore, mutagenic analyses of PRRSV nsp11 revealed that the endoribonuclease activity of nsp11 was critical for antagonizing the antiviral effect of PCSK9.
