In underground engineering, shear failure is a common failure type in coal-rock mass under medium and low strain-rate disturbance loads. Analyzing the shear failure mechanical properties of coal-rock mass under dynamic normal load is significant. In order to reveal the influence of disturbance load on the shear mechanical properties of coal rock, a dynamic and static load coupling electro-hydraulic servo testing machine was used to conduct the shear tests of coal-like rock materials under dynamic and constant normal load. The amplitude of dynamic load is 10 kN and the frequency is 5 Hz. The damage process of the specimens was detected by the acoustic emission (AE) detection system. The results imply that the shear failure process of coal-like rock materials under constant normal load can be divided into four stages. The normal disturbance decreased the shear strength of the specimens and increased the shear modulus of the specimens. With the increase in normal load, the influence of disturbance on the shear strength of the specimen decreased. By analyzing the AE parameters, it was found that the dynamic load made the internal damage of the specimen more severe during the shear failure process. The damage variable was calculated by AE cumulative energy, and the damage evolution was divided into three stages. The shear failure mechanism of the specimen was judged by RA (rise time/amplitude) and AF (average frequency). It was found that from the elastic deformation stage to the unstable development fracture stage, the proportion of shear fracture increased. When the dynamic normal load was 10 kN and 30 kN, the fracture was mainly shear fracture; When the dynamic normal load was 50 kN, the fracture was mainly tensile or mixed fracture. The dynamic normal load affects the shear strength and failure mechanism. Therefore, the influence of disturbance load on coal-rock mass strength cannot be ignored in underground engineering. In this paper, the privacy-preserving-based bipartite consensus control problem is investigated for a class of discrete-time nonlinear multi-agent systems (MASs). The classical bipartite consensus control implementation relies on the explicit information exchange between agents, which will lead to the possible leakage of agents' private information, especially the initial states. Therefore, to prevent data disclosure to neighbors inside the MASs and the outside eavesdroppers, the data is encrypted by means of Paillier encryption scheme during information transmission. This means each agent first sends the relative data in the form of ciphertext to its neighbors for further encryption process, and then restores the control protocol based on the returned encrypted information from the neighbors. Since homomorphic encryption only works for integers, the data must be quantized before encryption, and it will inevitably result in the quantization error. On the other hand, for the purpose of saving communication resources, both decentralized and distributed event-triggered schemes are also proposed. Resorting to the Lyapunov stability theory and algebraic graph theory, sufficient conditions are derived ensuring that the MASs subject to a confidential communication agreement can achieve the bounded bipartite consensus. In this paper, we are concerned with the sampled-based consensus problem of multi-agent systems with bounded distributed time-delays and quantisation effects. A dynamic quantisation scheme is introduced to save the constrained communication resources, where some adjustable parameters are introduced to exclude the phenomena of quantiser saturation. In addition, to reduce the actuation burden of controller and quantiser cost, a sampled-data control strategy is employed to investigate the consensus problem. Several sufficient criteria are derived to achieve consensus of multi-agent systems based on the matrix theory and Lyapunov control techniques. This article investigates the optimal control problem via reinforcement learning for a class of nonlinear discrete‐time systems. The nonlinear system under consideration is assumed to be partially unknown. A new learning‐based algorithm, T‐step heuristic dynamic programming with eligibility traces (T‐sHDP(λ)), is proposed to tackle the optimal control problem for such partially unknown system. First, the concerned optimal control problem is turned into its equivalence problem, that is, solving a Bellman equation. Then, the T‐sHDP(λ) is utilized to get an approximate solution of Bellman equation, and a rigorous convergence analysis is also conducted as well. Instead of the commonly used single step update approach, the T‐sHDP(λ) stores finite step past returns by introducing a parameter, and then utilizes these knowledge to update the value function (VF) of multiple moments synchronously, so as to achieve higher convergence speed. For implementation of T‐sHDP(λ), a neural network‐based actor‐critic architecture is applied to approximate VF and optimal control scheme. Backfill mining is an effective technique that can prevent and control the impact of dynamic disasters which can occur in deep underground coal mines. This technique allows to reduce the surface subsidence and achieve a greener mining. The backfill shear mechanical properties under mining disturbances, are of great significance for ensuring the surrounding rock stability in deep mining. Therefore, studying the shear mechanical properties of the cemented gangue backfill body (CGBB) under coupled dynamic and static loads actions in deep coal mining is of great importance. In this paper, shear mechanical tests and the acoustic emission (AE) technology were used to study the shear mechanical response characteristics of CGBB under low-frequency dynamic normal load conditions. The results show that a dynamic normal load with an amplitude of 5 kN improves the shear CGBB strength, while a dynamic load with an amplitude greater than 10 kN will cause a shear strength deterioration, and the degree of deterioration is positively correlated with the load amplitude. Under low-frequency dynamic normal load conditions, the shear CGBB stress-strain curve exhibits classical characteristics. AE indicate that the main CGBB damage changes from a nonlinear deformation to an elastic stage under low-frequency dynamic normal load conditions. During the shear failure process, the cumulative AE counts are positively correlated with the load amplitude. The crack type identification based on RA-AF shows that the low-frequency dynamic normal load affects the CGBB crack type. Under constant normal load (CNL) conditions, tensile or mixed cracks account for 63.95% and are the main fracture mode. Conversely, under low-frequency dynamic normal load conditions, shear cracks account for more than 60% and are the main failure mode. In this paper, the intermittent dynamic event-based state estimation problem is investigated for a class of delayed complex dynamic networks (CDNs). The estimate is implemented based on the measurements from a fraction of network nodes. In the framework of aperiodic intermittent measurement outputs, a dynamic event-triggered mechanism is introduced to save communication resources and reduce actuation burden. The aim of this work is to design a dynamic event-based state estimator by adopting intermittent dynamic event-triggered (IDET) strategy, such that the dynamics of estimation error system is exponentially stable. By resorting to Halanay inequality and switched system method, the sufficient conditions are derived for ensuring the existence of the desired state estimator, and are characterized by the ratio of the average working time to the average rest time. In the meanwhile, the estimator gains for partial nodes are explicitly obtained by solving some matrix inequalities. Furthermore, it is also proven that Zeno behavior can be excluded under the proposed IDET strategy. Finally, a numerical simulation is provided to verify the effectiveness of the derived results. Xenotransplantation from pigs to human beings is viewed as a potential solution for the acute organ shortage. However, consequent xenorejection induced by Gal alpha 1,3 Gal (a Gal, Gal antigen) prevents xenotransplantation from clinical application. Thus, the most attracting attempt to prevent xenorejection is the elimination of Gal. Our study suggested that compared with the human alpha 1,2 fucosyltransferase (FT) gene and the porcine antisense alpha 1,3 galactosyltransferase gene, sequence-specific siRNA targeting Gal was capable of suppressing Gal expression markedly, and therefore, significantly inhibiting xenoreactivity and the complement activation with human serum in PIEC cells. We also demonstrated the concordant inhibitory effect of siRNA and the human FT gene on Gal and corresponding functions, which implied a practical significance of combined transgenic strategy. This paper mainly investigates the exponential stability of switched neural networks (SNNs) with partial state reset and time-varying delays, in which partial state reset means that only a fraction of the states can be reset at each switching instant. Moreover, both stable and unstable subsystems are also taken into account and therefore, switched systems under consideration can take several switched systems as special cases. The comparison principle, the Halanay-like inequality, and the time-dependent switched Lyapunov function approach are used to obtain sufficient conditions to ensure that the considered SNNs with delays and partial state reset are exponentially stable. Summary: Synthetic biology offers new tools and capabilities of engineering cells with desired functions for example as new biosensing platforms leveraging engineered microbes. In the last two decades, bacterial cells have been programmed to sense and respond to various input cues for versatile purposes including environmental monitoring, disease diagnosis and adaptive biomanufacturing. Despite demonstrated proof‐of‐concept success in the laboratory, the real‐world applications of microbial sensors have been restricted due to certain technical and societal limitations. Yet, most limitations can be addressed by new technological developments in synthetic biology such as circuit design, biocontainment and machine learning. Here, we summarize the latest advances in synthetic biology and discuss how they could accelerate the development, enhance the performance and address the present limitations of microbial sensors to facilitate their use in the field. Stem cells play critical roles in cell therapies and tissue engineering for nerve repair. However, achieving effective delivery of high cell density remains a challenge. Here, a novel cell delivery platform termed the hyper expansion scaffold (HES) is developed to enable high cell loading. HES facilitated self‐promoted and efficient cell absorption via a dual driving force model. In vitro tests revealed that the HES rapidly expanded 80‐fold in size upon absorbing 2.6 million human amniotic epithelial stem cells (hAESCs) within 2 min, representing over a 400% increase in loading capacity versus controls. This enhanced uptake benefited from macroscopic swelling forces as well as microscale capillary action. In spinal cord injury (SCI) rats, HES–hAESCs promoted functional recovery and axonal projection by reducing neuroinflammation and improving the neurotrophic microenvironment surrounding the lesions. In summary, the dual driving forces model provides a new rationale for engineering hydrogel scaffolds to facilitate self‐promoted cell absorption. In order to study the weakening mechanism and mechanical behaviors of hard lamprophyre of Carboniferous Permian coal-bearing strata in China's mining area, lamprophyre samples were subjected to static rock dissolution experiments with pH values of 0, 2, and 4. The acid corrosion mechanism of lamprophyre was revealed from the weight changes of samples, characteristics of solution ion concentration, and macro-mechanical properties. The experimental results show that reaction occurred between lamprophyre and acid solution. With the increasing concentration of H+, the reaction was more intense, the degree of acid etching was higher, and the weight loss was greater. The internal damage induced by acid etching results in the slow extension of the compaction stage of stress–strain curve of uniaxial compression, and the obvious deterioration of mechanical properties of the lamprophyre. The uniaxial compressive strength of the lamprophyre in the dry state is 132 MPa, which decreased to 39 MPa under the acid etching condition, showing significant mudding characteristics. Dolomite (CaMg(CO3)2 with 19.63%) and orthoclase (KAlSi3O8 with 31.4%) in lamprophyre are the major minerals constituents involved in acidification reaction. Photomicrograph recorded from SEM studies reveals that the dissolution effect was directly related to the concentration of H+ in the solution. The dissolution effect was from the surface to the inside. The small dissolution pores became larger and continuously expanded, then finally formed a skeleton structure dominated by quartz. The content of K+, Ca2+, and Mg2+ in the solution after acid etching reaction indicates that the acidified product of orthoclase is colloidal H2SiO3, which adhered to the surface of samples during acid etching and hinders the further acidification of minerals. The dissolution of dolomite and orthoclase under acidic conditions directly leads to the damage of their structure and further promotes the water–rock interaction, which is the fundamental reason for the weakening of the mechanical properties of lamprophyre. Spinal cord injury (SCI) results in devastating consequences for the motor and sensory function of patients due to neuronal loss and disrupted neural circuits, confronting poor prognosis and lack of effective therapies. A new therapeutic strategy is urgently required. Here, human amniotic epithelial cells (hAEC), featured with immunocompatibility, non-tumorgenicity and no ethical issues, were induced into neural-like cells by a compound cocktail, as evidenced with morphological change and the expression of neural cell markers. Interestingly, the hAEC-neural-like cells maintain the characteristic of low immunogenicity as hAEC. Aiming at SCI treatment in vivo, we constructed a 3D-printed GelMA hydrogel biomimetic spinal cord scaffold with micro-channels, in which hAEC-neural-like cells were well-induced and grown. In a rat full transection SCI model, hAEC-neural-like cell scaffolds that were implanted in the lesion demonstrated significant therapeutic effects; the neural circuit and hindlimb locomotion were partly recovered compared to little affection in the SCI rats receiving an empty scaffold or a sham implantation operation. The loss of photoreceptors is a major event of retinal degeneration that accounts for most cases of untreatable blindness globally. To date, there are no efficient therapeutic approaches to treat this condition. In the present study, we aimed to investigate whether human amniotic epithelial stem cells (hAESCs) could serve as a novel seed cell source of photoreceptors for therapy. Here, a two–step treatment with combined Wnt, Nodal, and BMP inhibitors, followed by another cocktail of retinoic acid, taurine, and noggin induced photoreceptor–like cell differentiation of hAESCs. The differentiated cells demonstrated the morphology and signature marker expression of native photoreceptor cells and, intriguingly, bore very low levels of major histocompatibility complex (MHC) class II molecules and a high level of non–classical MHC class I molecule HLA–G. Importantly, subretinal transplantation of the hAESCs–derived PR–like cells leads to partial restoration of visual function and retinal structure in Royal College of Surgeon (RCS) rats, the classic preclinical model of retinal degeneration. Together, our results reveal hAESCs as a potential source of functional photoreceptor cells; the hAESCs–derived photoreceptor–like cells could be a promising cell–replacement candidate for therapy of retinal degeneration diseases. The osteogenic potential of bone marrow mesenchymal stem cells (BMSCs) is critical for bone formation and regeneration. A high non-/delayed-union rate of fracture healing still occurs in specific populations, implying an urgent need to discover novel targets for promoting osteogenesis and bone regeneration. Long non-coding (lnc)RNAs are emerging regulators of multiple physiological processes, including osteogenesis. Based on differential expression analysis of RNA sequencing data, we found that lncRNA AC132217.4, a 3'UTR-overlapping lncRNA of insulin growth factor 2 (IGF2), was highly induced during osteogenic differentiation of BMSCs. Afterward, both gain-of-function and loss-of-function experiments proved that AC132217.4 promotes osteoblast development from BMSCs. As for its molecular mechanism, we found that AC132217.4 binds with IGF2 mRNA to regulate its expression and downstream AKT activation to control osteoblast maturation and function. Furthermore, we identified two splicing factors, splicing component 35 KDa (SC35) and heterogeneous nuclear ribonucleoprotein A1 (HNRNPA1), which regulate the biogenesis of AC132217.4 at the post-transcriptional level. We also identified a transcription factor, ALX1, which regulates AC132217.7 expression at the transcriptional level to promote osteogenesis. Importantly, in-vivo over-expression of AC132217.4 essentially promotes the bone healing process in a murine tibial drill-hole model. Parkinson's disease (PD) remains a substantial clinical challenge due to the progressive loss of midbrain dopaminergic (DA) neurons in nigrostriatal pathway. In this study, human amniotic epithelial stem cells (hAESCs)-derived dopaminergic neuron-like cells (hAESCs-DNLCs) were generated, with the aim of providing new therapeutic approach to PD. hAESCs, which were isolated from discarded placentas, were induced to differentiate into hAESCs-DNLCs by following a "two stages" induction protocol. The differentiation efficiency was assessed by quantitative real-time PCR (qRT-PCR), immunocytochemistry (ICC), and ELISA. Immunogenicity, cell viability and tumorigenicity of hAESCs-DNLC were analyzed before in vivo experiments. Subsequently, hAESCs-DNLCs were transplanted into PD rats, behavioral tests were monitored after graft, and the regeneration of DA neurons was detected by immunohistochemistry (IHC). Furthermore, to trace hAESCs-DNLCs in vivo , cells were pre-labeled with PKH67 green fluorescence. hAESCs were positive for pluripotent markers and highly expressed neural stem cells (NSCs) markers. Based on this, we established an induction method reliably generates hAESCs-DNLCs, which was evidenced by epithelium-to-neuron morphological changes, elevated expressions of neuronal and DA neuronal markers, and increased secretion of dopamine. Moreover, hAESCs-DNLCs maintained high cell viability, no tumorigenicity and low immunogenicity, suggesting hAESCs-DNLCs an attractive implant for PD therapy. Transplantation of hAESCs-DNLCs into PD rats significantly ameliorated motor disorders, as well as enhanced the reinnervation of TH+ DA neurons in nigrostriatal pathway. Abstract Background aims The chronic inflammation of autoimmune diseases develops repetitive localized destruction or systemic disorders, represented by Hashimoto's thyroiditis (HT) and Systemic lupus erythematosus (SLE) respectively. Currently, there are no efficient ways to treat these autoimmune diseases. Therefore, it is critically important to explore new therapeutic strategies. The aim of this study was to investigate the therapeutic efficacy of human amniotic epithelial cells (hAECs) in murine models of HT and SLE. Methods Experimental autoimmune thyroiditis (EAT) was induced in female CBA/J mice by immunization with porcine thyroglobulin (pTg). hAECs were intravenously administered at different time points during the disease course. MRL-Fas lpr mice, a strain with spontaneously occurring SLE, were intravenously administered hAECs when their sera were positive for both anti-nuclear antibodies (ANAs) and anti-double-stranded DNA (anti-dsDNA) antibodies. Two weeks after the last cell transplantation, blood and tissue samples were collected for histological examination and immune system analysis. Results hAECs prevented lymphocytes infiltration into the thyroid and improved the damage of thyroid follicular in EAT mice. Correspondingly, hAECs administration reduced anti-thyroglobulin antibodies (TGAb), anti-thyroid peroxidase antibodies (TPOAb) and thyroid stimulating hormone (TSH) levels. SLE mice injected with hAECs appeared negative for ANAs and anti-dsDNA antibodies and showed reduced immunoglobulin profiles. Mechanically, hAECs modulated the immune cells balance in EAT and SLE mice, by downregulating the ratios of Th17/Treg cells in both EAT and SLE mice and upregulating the proportion of B10 cells in EAT mice. This was confirmed by in vitro assay, in which hAECs inhibited the activation of EAT mice-derived splenocytes. Moreover, hAECs improved the cytokine environment in both EAT and SLE mice, by suppressing the levels of IL-17A and IFN-γ and enhancing TGF-β. Conclusion These results demonstrated the immunoregulatory effect of hAECs for inflammation inhibition and injury recovery in HT and SLE murine models. Background and Purpose: Cardiovascular disease associated with antiretroviral therapy (ART) has become a major clinical challenge for HIV-positive patients. However, the role of ART in blood vessel growth is largely unknown. Here, we examined an integral component of ART, nucleoside/nucleotide reverse transcriptase inhibitors (NRTIs) and investigated their effects on key microvascular functions, including angiogenesis and lymphangiogenesis.Experimental Approach: The angiogenesis/lymphangiogenesis capability of endothelial cells (ECs) was evaluated using migration, proliferation and tube formation assays in vitro, and mouse ear and Matrigel plug assays in vivo. Expressions of signalling molecules and mitochondrial antioxidant catalases were determined using Western blotting. Receptor tyrosine kinase (RTK) internalization and endocytosis were examined using flow cytometry and confocal immunofluorescence microscopy respectively. Mitochondrial DNA copy number and ROS were determined using quantitative real-time PCR and MitoSOX staining respectively.Key Results: Pharmaceutical doses of NRTIs [azidothymidine (AZT), tenofovir disoproxil fumarate (TDF) and lamivudine (3TC)] inhibited angiogenesis and lymphangiogenesis both in vivo and in vitro by affecting the proliferation and migration of ECs. Correspondingly, NRTIs selectively attenuated the activation and transduction of endothelial RTK signals, VEGFR2 and FGFR1 pathways, in vascular ECs and the VEGFR3 pathway in lymphatic ECs. Both TDF and 3TC restrained RTKs' endocytosis into early endosomes but not internalization, while AZT blocked the protein maturation of RTKs. This study aimed to investigate whether conventional predisposing factors modify the associations of homocysteine with blood pressure levels and hypertension. A total of 2615 adults were recruited from Liaoning province. An elevated homocysteine level was significantly associated with increased hypertension risk and blood pressure (all P<.05). Interaction analyses showed that homocysteine acted synergistically with age, overweight/obesity, dyslipidemia, and family history of hypertension to affect hypertension risk, and the relative excess risk due to interaction was 1.21 (95% confidence interval, 0.07-2.35), 0.72 (95% confidence interval, 0.07-1.36), 0.45 (95% confidence interval, 0.06-0.85), and 1.87 (95% confidence interval, 0.77-2.97), respectively. Increases in blood pressure were higher in patients who were overweight/obese or had a family history of hypertension than in their counterparts (all Pinteraction <.05). Objectives: The aim of this study was to determine the role of suppressor of cytokine signaling 1 (SOCS1) in graft arteriosclerosis (GA). Background: GA, the major cause of late cardiac allograft failure, is initiated by immune-mediated endothelial activation resulting in vascular inflammation and consequent neointima formation. SOCS1, a negative regulator of cytokine signaling, is highly expressed in endothelial cells (ECs) and may prevent endothelial inflammatory responses and phenotypic activation. Methods: Clinical specimens of coronary arteries with GA, with atherosclerosis, or without disease were collected for histological analysis. SOCS1 knockout or vascular endothelial SOCS1 (VESOCS1) transgenic mice were used in an aorta transplant model of GA. Mouse aortic ECs were isolated for in vitro assays. Results: Dramatic but specific reduction of endothelial SOCS1 was observed in human GA and atherosclerosis specimens, which suggested the importance of SOCS1 in maintaining normal endothelial function. SOCS1 deletion in mice resulted in basal EC dysfunction. After transplantation, SOCS1-deficient aortic grafts augmented leukocyte recruitment and neointima formation, whereas endothelial overexpression of SOCS1 diminished arterial rejection. Induction of endothelial adhesion molecules in early stages of GA was suppressed by the VESOCS1 transgene, and this effect was confirmed in cultured aortic ECs. Moreover, VESOCS1 maintained better vascular function during GA progression. Mechanistically, endothelial SOCS1, by modulating both basal and cytokine-induced expression of the adhesion molecules platelet/endothelial cell adhesion molecule-1, intercellular adhesion molecule-1, and vascular cell adhesion molecule-1, restrained leukocyte adhesion and transendothelial migration during inflammatory cell infiltration. Perinatal stem cells have been regarded as an attractive and available cell source for medical research and clinical trials in recent years. Multiple stem cell types have been identified in the human placenta. Recent advances in knowledge on placental stem cells have revealed that human amniotic epithelial stem cells (hAESCs) have obvious advantages and can be used as a novel potential cell source for cellular therapy and clinical application. hAESCs are known to possess stem-cell-like plasticity, immune-privilege, and paracrine properties. In addition, non-tumorigenicity and a lack of ethical concerns are two major advantages compared with embryonic stem cells (ESCs) and induced pluripotent stem cells (iPSCs). All of the characteristics mentioned above and other additional advantages, including easy accessibility and a non-invasive application procedure, make hAESCs a potential ideal cell type for use in both research and regenerative medicine in the near future. The application of RF cavity beam position monitors (CBPM) is essential for precise beam position measurements in X-ray Free-Electron Lasers (XFEL) and linear colliders. This paper addresses the solution to the inter-bunch signal interference when the bunch separation is as short as nanoseconds. The signals can overlap from bunch to bunch, which can induce appreciable error in evaluating beam position. We propose a conceptual method to eliminate the dominant signal overlap under a quadrature phase offset condition. Using this method, the elimination automatically occurs in the process of signal digitalization. The dominant interference that is contributed by the last bunch can be reduced over an order of magnitude in a conventional CBPM system. In addition, the required bandwidth can be effectively reduced by a factor of 2, and the design challenge for a wideband CBPM is relaxed. The method was demonstrated in an analytic approach by simulating the signal evolution in the RF front end of electronics built in MATLAB code. In the practical concern of applying this method, the factors that perturb the elimination have been examined as well. Based on the numerical result, the CBPM prototype has been designed for the pre-research plan of future high repetition rate hard XFEL at Chinese Academy of Engineering Physics. The combined simulations in CST and MATLAB on the CBPM prototype indicate agreement with the result obtained in the analytic approach. [ABSTRACT FROM AUTHOR]
