Lung cancer is the most talked about cancer in the world. It is also one of the cancers that currently has a high mortality rate. The aim of our research is to find more effective therapeutic targets and prognostic markers for human lung cancer. First, we download gene expression data from the GEO database. We performed weighted co-expression network analysis on the selected genes, we then constructed scale-free networks and topological overlap matrices, and performed correlation modular analysis with the cancer group. We screened the 200 genes with the highest correlation in the cyan module for functional enrichment analysis and protein interaction network construction, found that most of them focused on cell division, tumor necrosis factor-mediated signaling pathways, cellular redox homeostasis, reactive oxygen species biosynthesis, and other processes, and were related to the cell cycle, apoptosis, HIF-1 signaling pathway, p53 signaling pathway, NF-κB signaling pathway, and several cancer disease pathways are involved. Finally, we used the GEPIA website data to perform survival analysis on some of the genes with GS > 0.6 in the cyan module. CBX3, AHCY, MRPL12, TPGB, TUBG1, KIF11, LRRC59, MRPL17, TMEM106B, ZWINT, TRIP13, and HMMR was identified as an important prognostic factor for lung cancer patients. In summary, we identified 12 mRNAs associated with lung cancer prognosis. Our study contributes to a deeper understanding of the molecular mechanisms of lung cancer and provides new insights into drug use and prognosis.

Leptin can indirectly regulate fatty‐acid metabolism and synthesis in muscle in vivo and directly in incubated muscle ex vivo. In addition, non‐synonymous mutations in the bovine leptin gene (LEP) are associated with carcass intramuscular fat (IMF) content. However, the effects of LEP on lipid synthesis of adipocytes have not been clearly studied at the cellular level. Therefore, this study focused on bovine primary intramuscular preadipocytes to investigate the effects of LEP on the proliferation and differentiation of intramuscular preadipocytes, as well as its regulatory mechanism in lipid synthesis. The results showed that both the LEP and leptin receptor gene (LEPR) were highly expressed in IMF tissues, and their mRNA expression levels were positively correlated at different developmental stages of intramuscular preadipocytes. The overexpression of LEP inhibited the proliferation and differentiation of intramuscular preadipocytes, while interference with LEP had the opposite effect. Additionally, LEP significantly promoted the phosphorylation level of AMPKα by promoting the protein expression of CAMKK2. Meanwhile, rescue experiments showed that the increasing effect of AMPK inhibitors on the number of intramuscular preadipocytes was significantly weakened by the overexpression of LEP. Furthermore, the overexpression of LEP could weaken the promoting effect of AMPK inhibitor on triglyceride content and droplet accumulation, and prevent the upregulation of adipogenic protein expression (SREBF1, FABP4, FASN, and ACCα) caused by AMPK inhibitor. Taken together, LEP acted on the AMPK signaling pathway by regulating the protein expression of CAMKK2, thereby downregulating the expression of proliferation‐related and adipogenic‐related genes and proteins, ultimately reducing intramuscular adipogenesis. [Objective] The effects of dietary crude protein (CP) levels and Saccharomyces cerevisiae (SC) supplementation on immunization and nitrogen and phosphorus metabolism of Qinchuan beef cattle were studied. [Method] Eighteen healthy 14-month-old female cattle with similar body weight of (298 ± 9.78) kg were selected for feeding experiments with two protein levels (13.23%(A1) and (10.59%(A2)) and three SC levels (0(B1), 4x108 CFU/kg(B2) and 8x108 CFU/kg(B3)). Nitrogen and phosphorus conents of feces and urine were determined using total feces (urine) collection method and blood samples were collected before morning feeding at the end of the trial, which lasted for 55 days. [Result] ➀ During 11-55 d of the experimental period, dietary supplementation of 8x108 CFU/kg SC significantly increased eed intake of Qinchuan beef cattle (P<0.05). The feed intake of Qinchuan beef cattle in group A1 B3 was he highest, which was significantly higher than that in group A1B1. ➁ TG content of Qinchuan beef cattle n group B3 was significantly lower than that in group B1 (P<0.05), and serum total protein, albumin, globulin, IgG and IgM of Qinchuan beef cattle in group A2B3 were significantly higher than those in group A2B1 (P<0.05). ➂ Compared with group B1, the apparent digestibility of CP and NDF in group B3 was ncreased by 8.44% and 16.90% (P<0.05), respectively. The apparent digestibility of CP, OM and ADF n group A2B3 was the highest. ➃ Compared with A2B1 group, the manure and urinary nitrogen emissions of Qinchuan beef cattle in A2B3 group were decreased by 31.28% and 19.67%, while nitrogen digestibility and nitrogen deposition rate were increased by 7.66% and 62.57%, respectively. ➄ The fecal phosphorus of Qinchuan beef cattle in B2 and B3 groups was significantly lower than that in B1 group, and phosphorus igestibility and deposition rate of Qinchuan beef cattle in group A2B3 were 35.96% and 45.94% higher han those in group A2B1 (P<0.05). [Conclusion] Dietary supplementation of 8 x 108 CFU/kg SC imroved feed intake, immune level and nutrient digestibility, while reduced nitrogen and phosphorus emisions of Qinchuan beef cattle. It is suggested to supplement 8x108 CFU/kg SC to diets of Qinchuan beef attle with 10.59% CP level. [ABSTRACT FROM AUTHOR]
