Abstract

Amino acids are very important for oolong tea brisk-smooth mouthfeel which is mainly associated with bruising and withering treatment (BWT). In this study, metabolome and transcriptome analyses were performed to comprehensively investigate the changes in abundance of amino acids and the expression pattern of relevant genes during BWT of oolong tea manufacturing. Levels of most amino acids increased during BWT in the leaves harvested from 4 cultivars, while expression of the relevant function genes responsible for synthesis and transformation of amino acids up-regulated accordingly. Upstream hub genes including receptor-like protein kinase IKU2, serine/threonine-protein kinase PBL11, MYB transcription factor MYB2, ethylene-responsive transcription factor ERF114, WRKY transcription factor WRKY71, aspartate aminotransferase AATC, UDP-glycosyltransferase U91D1, and 4-hydroxy-4-methyl-2-oxoglutarate aldolase 2 RRAA2, were predicted to be involved in regulation of the function genes expression and the amino acids metabolism through weighted gene coexpression network analysis. A modulation mechanism for accumulation of amino acids during BWT was also proposed. These findings give a deep insight into the metabolic reprogramming mechanism of amino acids during BWT of oolong tea.

Abstract

This article has introduced four different numeralization methods for the Complete Characteristic Curves of Pump’s, and these methods are applied for the calculation of water hammer that caused by pump-stopping. Then by using the performance curves of pumps with different specific speeds we had compared those four methods, and determined the optimal method for the reference of related engineering and technical personnel.

According to the situation of Chinese urban water supply, this article has analyszed the possibility and computing method for water hammer that caused by pump stop in large diameter and high-lift water pipelines. In addition the impact of Full-featured Pump Curve on the water hammer calculation are also discussed. Finally, using the water hammer calculation of the primary pumping stations within a water-supply project which located in northeast China as an example, we had made corresponding conclusion.

Abstract

In this paper，by transforming the fully characteristic curve of pump with eighty specific speed to be numeralization he obtained fehleranalyse error of discrete data were analyzed based on the pump similarity theory and the difference between certain assumed condition and real situation in numeralization process. Then the influence of the fehleranalyse error of numeralization method on the pressure boost caused by Pump-off water hammer was computed by a Pump-off water hammer calculation within an real engineering project. The reliability of numeric calculation method of fully characteristic curve of Pump is proved.

Abstract

Abstract: This study was aimed at investigating the effects of lncRNA AK139328 on myocardial ischaemia/reperfusion injury (MIRI) in diabetic mice. Ischaemia/reperfusion (I/R) model was constructed in normal mice (NM) and diabetic mice (DM). Microarray analysis was utilized to identify lncRNA AK139328 overexpressed in DM after myocardial ischaemia/reperfusion (MI/R). RT‐qPCR assay was utilized to investigate the expressions of lncRNA AK139328 and miR‐204‐3p in cardiomyocyte and tissues. Left ventricular end diastolic diameter (LVEDD), left ventricular end systolic diameter (LVESD), left ventricular ejection fraction (LVEF) and fractioning shortening (FS) were obtained by transthoracic echocardiography. Haematoxylin‐eosin (HE) staining and Masson staining were utilized to detect the damage of myocardial tissues degradation of myocardial fibres and integrity of myocardial collagen fibres. Evans Blue/TTC staining was used to determine the myocardial infarct size. TUNEL staining was utilized to investigate cardiomyocyte apoptosis. The targeted relationship between lncRNA AK139328 and miR‐204‐3p was confirmed by dual‐luciferase reporter gene assay. MTT assay was used for analysis of cardiomyocyte proliferation. Western blot was utilized to investigate the expression of alpha smooth muscle actin (α‐SMA), Atg7, Atg5, LC3‐II/LC3‐I and p62 marking autophagy. Knockdown of lncRNA AK139328 relieved myocardial ischaemia/reperfusion injury in DM and inhibited cardiomyocyte autophagy as well as apoptosis of DM. LncRNA AK139328 modulated miR‐204‐3p directly. MiR‐204‐3p and knockdown of lncRNA AK139328 relieved hypoxia/reoxygenation injury via inhibiting cardiomyocyte autophagy. Silencing lncRNA AK139328 significantly increased miR‐204‐3p expression and inhibited cardiomyocyte autophagy, thereby attenuating MIRI in DM. [ABSTRACT FROM AUTHOR]

29. Abstract 17159: Two Birds One Stone: Salvianolic Acid B Inhibits Coagulation And Platelet Aggregation

Abstract

Introduction:Danshen (Salvia miltiorrhizaroot extracts) have been reported for centuries to control cardiovascular diseases in Chinese medicine. Danshen depside salts are currently approved in China to treat coronary heart disease and angina and are under evaluation in phase II clinical trials outside of China. Salvianolic acids have been identified as the active compounds of danshen and salvianolic acid B (SAB), the most abundant salvianolic acid, has been previously shown to exhibit anti-platelet and anti-thrombotic properties in animal models. However, the mechanism of action has not been adequately explored.Results:We demonstrate that SAB attenuates ADP-, collagen-, and thrombin receptor activating peptide (TRAP)-induced human platelet aggregation. Interestingly, SAB inhibits thrombin-induced platelet aggregation far more potently than other agonists. Furthermore, using our intravital microscopy thrombosis models, we demonstrate that SAB decreases thrombus growth in vivo. Using a series of in vitrocoagulation assays, we found that SAB significantly reduced clot weight in human whole blood, and delayed coagulation in human cell-free plasma using thromboelastography. In addition, SAB reduces fibrin network density in cell-free plasma. Through structural analysis, we found that SAB contains structural similarities to trisubstituted benzimidazole thrombin inhibitors, such as dabigatran. In silicomolecular modeling predicts that SAB binds within the thrombin active site - interacting with similar residues as dabigatran. Using isothermal titration calorimetry and kinetic thrombin inhibition assays, we corroborate these findings and report SAB as a direct competitive thrombin inhibitor.Conclusions:These data establish a novel mechanism of SAB in the inhibition of both platelet aggregation and blood coagulation. These unique characteristics position salvianolic acids as safe and potent herb-derived anti-thrombotic agents.
