4. Phosphorylated Adapter RNA Export Protein Is Methylated at Lys 381 by an Methyltransferase-like 21C (METTL21C).

Abstract

Methyltransferase-like 21C (METTL21C) is a member of the non-histone methyltransferase superfamily, which mainly mediates the methylation of lysine (Lys) residues. The main types of modification are Lys dimethylation and trimethylation. However, at present, most of the studies on METTL21C are focused on humans and mice, and there are few reports on poultry. Therefore, chicken embryo fibroblasts (DF-1) were selected as the object of study. To explore the function of chicken METTL21C (chMETTL21C) in the proliferation of DF-1 cells, flow cytometry and qPCR were used to detect the function of chicken METTL21C in the proliferation of DF-1 cells. The results showed that overexpression of METTL21C blocked the cell cycle in the G1max S phase, thus inhibiting cell proliferation. In addition, based on proteomic analysis, stable overexpression of METTL21C may inhibit the proliferation of DF-1 cells by mediating lysine trimethylation of proliferation-related proteins phosphorylated adapter RNA export protein (PHAX), nucleoside diphosphate kinases (NDPKs), eukaryotic transcription extension factor (eukaryotic translation elongation factor 1A,e EF1A), and inversin (Invs). Through immunoprecipitation (co-IP) and liquid chromatography-mass spectrometry (LC-MS/MS) analysis, METTL21C-mediated PHAX Lys-381 methylation was confirmed to be involved in the regulation of DF-1 cell proliferation. The results of this study provide a reference for analyzing the methylation function of METTL21C and the mechanism of regulating the growth and development of chicken cells. [ABSTRACT FROM AUTHOR]

Abstract

Three polymeric manganese(II) complexes [ Mn ( H 2 O ) 2 ( NTA ) ] ∞ 2 1 , [ Mn ( H 2 O ) 2 ( TNA ) ] ∞ 2 2 and [ Mn ( H 2 O ) 3 ( FNA ) ] ∞ 1 3 (HNTA = 2-nitroterephthalic acid; HTNA = 3-nitrophthalic acid; HFNA = 4-nitrophthalic acid), were synthesized and characterized. Compounds 1, 2 and 3 are assembled into 3D supramolecular network via O–H⋯O hydrogen bonds interactions. The Variable-temperature (2–300 K) magnetic susceptibility measurements of 1–3 show overall antiferromagnetic interactions between the high-spin Mn(II) (S = 5/2) ions. Results about i.r. spectra, thermal and elemental analyses are presented.

Abstract

Hydrothermal reactions of 4,4´-bipyridine, benzoic acid/p-methylbenzoic acid, Ni(NO3)2 · 6H2O and NH4OH yielded [Ni(bpy)1.5(H2O)2(BA)2]·H2O (1) and [Ni(bpy)(HMBA)(MBA)2]n (2) (bpy = 4,4´-bipyridine, HBA = benzoic acid, HMBA = p-methylbenzoic acid). The Ni(II) ions of compound 1 are six-coordinated and bridged by 4,4´-bipyridine ligands to form a binuclear structure while complex 2 features polymeric chains which are connected by 4,4´-bipyridine molecules. Variabletemperature magnetic susceptibility measurements from 300 to 30 K showed the presence of weak ferromagnetic interactions between the Ni(II) (S = 1) ions via the 4,4´-bipyridine bridges.

Abstract

Reactions of CuCl2, m-nitrobenzoic acid (HNBA) and NaOH with 2,2′-bipyridine (bpy) or 1,10-phenanthroline (phen) in aqueous ethanol afforded two Cu(II) complexes Cu(bpy)(NBA)2, 1, and [Cu(phen)(H2O)2(NBA)](NBA), 2. The monomolecular Cu(bpy)(NBA)2 moieties are both bridged by hydrogen bonding interactions and interlayer π *–π *stacking interactions to form a 3D (3,4,6)-connected supramolecular architecture with the Schafli symbol of (44·62)(44·66·85)(63)2. Complex 2 crystallizes in a noncentrosymmetric space group P21 where all molecules show the same orientation along the polar b axis. Preliminary investigations suggest that 2 exhibits ferroelectric hysteresis loops at room temperature with remanent polarization (P r) of ca. 0.09 μC cm−2 and coercive electric fields of 2.53 kV cm−1, respectively. It may be a potential ferroelectric with a relatively large spontaneous polarization (P s) of 0.22 μC cm−2. Furthermore, permittivity property measurements reveal a dielectric constant (e r) of 6.36. Variable-temperature (2–300 K) magnetic susceptibility measurements showed the presence of weak ferromagnetic interactions between the Cu(II) ions for both 1 and 2.
