Abstract Background The number of spikelets per spike is a key trait that affects the yield of bread wheat (Triticum aestivum L.). Identification of the QTL for spikelets per spike and its genetic effects that could be used in molecular assistant breeding in the future. Results In this study, four recombinant inbred line (RIL) populations were generated and used, having YuPi branching wheat (YP), with Supernumerary Spikelets (SS) phenotype, as a common parent. QTL (QSS.sicau-2 A and QSS.sicau-2D) related to SS trait were mapped on chromosomes 2 A and 2D through bulked segregant exome sequencing (BSE-Seq). Fourteen molecular markers were further developed within the localization interval, and QSS.sicau-2 A was narrowed to 3.0 cM covering 7.6 Mb physical region of the reference genome, explaining 13.7 − 15.9% the phenotypic variance. Similarly, the QSS.sicau-2D was narrowed to 1.8 cM covering 2.4 Mb physical region of the reference genome, and it explained 27.4 − 32.9% the phenotypic variance. These two QTL were validated in three different genetic backgrounds using the linked markers. QSS.sicau-2 A was identified as WFZP-A, and QSS.sicau-2D was identified a novel locus, different to the previously identified WFZP-D. Based on the gene expression patterns, gene annotation and sequence analysis, TraesCS2D03G0260700 was predicted to be a potential candidate gene for QSS.sicau-2D. Conclusion Two significant QTL for SS, namely QSS.sicau-2 A and QSS.sicau-2D were identified in multiple environments were identified and their effect in diverse genetic populations was assessed. QSS.sicau-2D is a novel QTL associated with the SS trait, with TraesCS2D03G0260700 predicted as its candidate gene.

Abstract R-loop-triggered collateral single-stranded DNA (ssDNA) nuclease activity within Class 1 Type I CRISPR-Cas systems holds immense potential for nucleic acid detection. However, the hyperactive ssDNase activity of Cas3 introduces unwanted noise and false-positive results. In this study, we identified a novel Type I-A Cas3 variant derived from Thermococcus siculi, which remains in an auto-inhibited state until it is triggered by Cascade complex and R-loop formation. This Type I-A CRISPR-Cas3 system not only exhibits an expanded protospacer adjacent motif (PAM) recognition capability but also demonstrates remarkable intolerance towards mismatched sequences. Furthermore, it exhibits dual activation modes—responding to both DNA and RNA targets. The culmination of our research efforts has led to the development of the Hyper-Active-Verification Establishment (HAVE, 惠父). This innovation enables swift and precise human papillomavirus (HPV) diagnosis in clinical samples, providing a robust molecular diagnostic tool based on the Type I-A CRISPR-Cas3 system. Our findings contribute to understanding type I-A CRISPR-Cas3 system regulation and facilitate the creation of advanced diagnostic solutions with broad clinical applicability.

Flood frequency in river source regions is significantly affected by rainfall and snowmelt as part of climatic changes. A traditional univariate flood frequency analysis cannot reflect the complexity of floods, and when used in isolation, it can only underestimate flood risk. For effective flood prevention and mitigation, it is essential to consider the combined effects of precipitation and snowmelt. Copula functions can effectively quantify the joint distribution relationship between floods and their associated variables without restrictions on their distribution characteristics. This study uses copula functions to consider a multivariate probability distribution model of flood peak flow (Q) with cumulative snowmelt (CSm) and cumulative precipitation (CPr) for the Hutubi River basin located in northern Xinjiang, China. The joint frequencies of rainfall and snowmelt floods are predicted using copula models based on the Coupled Model Intercomparison Project Phase 6 data. Abstract Carbon-based perovskite solar cells (c-PSCs) have attracted increasing attention due to their numerous advantages including ease of fabrication, the potential of assembling flexible devices, low manufacturing costs as well as large-scale production. However, c-PSCs suffer from the limited hole extraction and high charge carrier recombination due to the inadequate interface contact between the carbon electrode and perovskite film. Herein, we report the fabrication of planar c-PSCs with high efficiency and excellent stability by employing electrochemically produced large-area phosphorene flakes as a hole-transporting layer (HTL). Large-area phosphorene shows well-aligned band energy levels with the perovskite, and thus led to the efficient hole extraction and the reduced hysteresis behaviour. Consequently, while exhibiting excellent stability under various harsh testing conditions, the devices with phosphorene HTL delivered a power conversion efficiency of over 15% with an open-circuit voltage of 1.082 V, which is the highest reported value for c-PSCs without traditional hole transporting materials to date.

Abstract Accurate preoperative differentiation of the chronic rhinosinusitis (CRS) endotype between eosinophilic CRS (eCRS) and non-eosinophilic CRS (non-eCRS) is an important topic in predicting postoperative outcomes and administering personalized treatment. To this end, we have constructed a sinus CT dataset, which comprises CT scan data and pathological biopsy results from 192 patients of chronic rhinosinusitis with nasal polyps (CRSwNP), treated at the Second Affiliated Hospital of Shantou University Medical College between 2020 and 2022. To differentiate CRSwNP endotype on preoperative CT and improve efficiency at the same time, we developed a multi-view fusion model that contains a mini-architecture with each network of 10 layers by modifying the deep residual neural network. The proposed model is trained on a training set and evaluated on a test set. The multi-view deep learning fusion model achieved the area under the receiver-operating characteristics curve (AUC) of 0.991, accuracy of 0.965 and F1-Score of 0.970 in test set. We compared the performance of the mini-architecture with other lightweight networks on the same Sinus CT dataset. The experimental results demonstrate that the developed ResMini architecture contribute to competitive CRSwNP endotype identification modeling in terms of accuracy and parameter number.

Abstract Background N6-methyladenosine (m6A) modification plays an important role in lung cancer. However, methyltransferase-like 14 (METTL14), which serves as the main component of the m6A complex, has been less reported to be involved in the immune microenvironment of lung cancer. This study aimed to analyze the relationship between METTL14 and the immune checkpoint inhibitor programmed death receptor 1 (PD-1) in lung cancer. Methods CCK-8, colony formation, transwell, wound healing, and flow cytometry assays were performed to explore the role of METTL14 in lung cancer progression in vitro. Furthermore, syngeneic model mice were treated with sh-METTL14 andan anti-PD-1 antibody to observe the effect of METTL14 on immunotherapy. Flow cytometry and immunohistochemical (IHC) staining were used to detect CD8 expression. RIP and MeRIP were performed to assess the relationship between METTL14 and HSD17B6. LLC cells and activated mouse PBMCs were cocultured in vitro to mimic immune cell infiltration in the tumor microenvironment. ELISA was used to detect IFN-γ and TNF-α levels. Results The online database GEPIA showed that high METTL14 expression indicated a poor prognosis in patients with lung cancer. In vitro assays suggested that METTL14 knockdown suppressed lung cancer progression. In vivo assays revealed that METTL14 knockdown inhibited tumor growth and enhanced the response to PD-1 immunotherapy. Furthermore, METTL14 knockdown enhanced CD8+T-cell activation and infiltration. More importantly, METTL14 knockdown increased the stability of HSD17B6 mRNA by reducing its m6A methylation. In addition, HSD17B6 overexpression promoted the activation of CD8+ T cells. Conclusion The disruption of METTL14 contributed to CD8+T-cell activation and the immunotherapy response to PD-1 via m6A modification of HSD17B6, thereby suppressing lung cancer progression.

Interstitial free (IF) steels are characterized by their interstitial free ferritic matrix due to addition of micro-alloying elements that draw carbon and nitrogen out of matrix to form carbonitrides. The IF steels typically have excellent deep drawing ability, certain mechanical strength, and high plastic deformation performance. In this paper, the microstructure evolution and carbides precipitation behavior during the hot rolling, cold rolling and annealing processes have been studied using both experimental and thermodynamic and kinetic modeling approaches. The strengthening mechanism of Nb–Ti microalloyed IF steel has been investigated. The results of the thermodynamic simulation indicates that the addition of Nb into Ti bearing IF steel promotes recrystallization at 900 °C, leading to fine recrystallized grains. Due to the recrystallization of austenite during the final rolling process, Nb–Ti microalloyed hot rolled plates exhibit finer grain size, fewer substructures, and lower dislocation density compared to the hot rolled plates containing only Ti. Building upon the initial fine and uniform structure, Nb–Ti microalloyed steel maintains a refined structure after undergoing cold rolling and annealing. The study of the carbides precipitation behavior suggests that TiN precipitates at high temperature above 1100 °C, while a substantial amount of (Nb,Ti)C and TiC precipitate at about 900 °C in Nb–Ti and Ti bearing steels, respectively. There is no significantly difference between the amount of carbides in steels hot deformed at 900 °C and in the hot rolled plates, indicating that the carbides mainly precipitate during the hot rolling process. There are negligible carbides precipitated during the coiling process at 600 °C. In Nb–Ti containing steel, Nb and Ti in solid solution are significantly reduced due to the large amount precipitation of (Nb,Ti)C during the final rolling process. The reduction of solid solution Nb effectively promotes the recrystallization of austenite. Therefore, the grains of hot-rolled, cold-rolled, and annealed strips are refined. In addition, there are more small size carbides in Nb–Ti bearing steel than that in Ti bearing steel. Therefore, the higher strength of Nb–Ti steel is mainly from the strengthening effect of grain refinement and carbides precipitation.

BackgroundThe existing tools in China for assessing psychiatric nurses' knowledge， attitude and practice towards the use of protective restraint do not sufficiently consider the specialty in psychiatric practice， and the scale items are somewhat cumbersome to use， which together restrict their further promotion and application. Accordingly， there is an urgent need for developing a more scientific assessment tool.ObjectiveTo construct a scale for assessing psychiatric nurses' knowledge， attitude and practice towards the use of protective restraint and to verify its reliability and validity， so as to provide a generic tool for the assessment.MethodsBased on the theoretical framework of the knowledge-attitude-practice model， the preliminary scale was formed through systematic literature review， qualitative interview and Delphi method. From July to December 2021， a stratified sampling was utilized to select 729 psychiatric nurses from tertiary， secondary and primary （including unclassified medical institutions and grassroots hospitals） psychiatric hospitals in Guangdong Province. The formal scale was developed through item analysis， exploratory factor analysis and confirmatory factor analysis. The validity of the scale was evaluated by content validity and structure validity， and the reliability was verified by procedures including Cronbach's α coefficient， test-retest reliability， and split-half reliability.ResultsA total of 12 items of three dimensions （knowledge， attitude， practice） were included in the scale for assessing psychiatric nurses' knowledge， attitude and practice towards the use of protective restraint. The scale-level content validity index （S-CVI） was 0.941， and the item-level content validity index （I-CVI） ranged from 0.812 to 1.000. Exploratory factor analysis extracted three common factors， and the cumulative variance contribution rate was 62.948%. The confirmatory factor yielded adequate fit. Cronbach's α coefficient was found to be 0.887 for the scale， 0.819 for knowledge dimension， 0.842 for attitude dimension， and 0.831 for practice dimension. The split-half reliability and test-retest reliability for the scale were 0.712 and 0.922， respectively.ConclusionThe scale shows satisfactory reliability and validity， which can be used to assess psychiatric nurses' knowledge， attitude and practice towards the use of protective restraint. ［Funded by Guangdong Clinical Teaching Base Teaching Reform Research Project （number， 2021JD119）］

Abstract Maintaining sufficient cerebral oxygen metabolism is crucial for human survival, especially in challenging conditions such as high-altitudes. Human cognitive neural activity is sensitive to fluctuations in oxygen levels. However, there is a lack of publicly available datasets on human behavioural responses and cerebral dynamics assessments during the execution of conflicting tasks in natural hypoxic environments. We recruited 80 healthy new immigrant volunteers (males, aged 20 ± 2 years) and employed the Stroop cognitive conflict paradigm. After a two-week exposure to both high and low-altitudes, the behavioural performance, prefrontal oxygen levels, and electroencephalography (EEG) signals were recorded. Comparative analyses were conducted on the behavioural reaction times and accuracy during Stroop tasks, and statistical analyses of participants’ prefrontal oxygen levels and EEG signals were performed. We anticipate that our open-access dataset will contribute to the development of monitoring devices and algorithms, designed specifically for measuring cerebral oxygen and EEG dynamics in populations exposed to extreme environments, particularly among individuals suffering from oxygen deficiency.

Abstract Background Microbiota are closely associated with human health and disease. Metaproteomics can provide a direct means to identify microbial proteins in microbiota for compositional and functional characterization. However, in-depth and accurate metaproteomics is still limited due to the extreme complexity and high diversity of microbiota samples. It is generally recommended to use metagenomic data from the same samples to construct the protein sequence database for metaproteomic data analysis. Although different metagenomics-based database construction strategies have been developed, an optimization of gene taxonomic annotation has not been reported, which, however, is extremely important for accurate metaproteomic analysis. Results Herein, we proposed an accurate taxonomic annotation pipeline for genes from metagenomic data, namely contigs directed gene annotation (ConDiGA), and used the method to build a protein sequence database for metaproteomic analysis. We compared our pipeline (ConDiGA or MD3) with two other popular annotation pipelines (MD1 and MD2). In MD1, genes were directly annotated against the whole bacterial genome database; in MD2, contigs were annotated against the whole bacterial genome database and the taxonomic information of contigs was assigned to the genes; in MD3, the most confident species from the contigs annotation results were taken as reference to annotate genes. Annotation tools, including BLAST, Kaiju, and Kraken2, were compared. Based on a synthetic microbial community of 12 species, it was found that Kaiju with the MD3 pipeline outperformed the others in the construction of protein sequence database from metagenomic data. Similar performance was also observed with a fecal sample, as well as in silico mixed datasets of the simulated microbial community and the fecal sample. Conclusions Overall, we developed an optimized pipeline for gene taxonomic annotation to construct protein sequence databases. Our study can tackle the current taxonomic annotation reliability problem in metagenomics-derived protein sequence database and can promote the in-depth metaproteomic analysis of microbiome. The unique metagenomic and metaproteomic datasets of the 12 bacterial species are publicly available as a standard benchmarking sample for evaluating various analysis pipelines. Abstract Vanadium nitride (VN) is a transition-metal nitride with remarkable properties that have prompted extensive experimental and theoretical investigations in recent years. However, there is a current paucity of experimental research investigating the temperature-dependent electronic structure of single-crystalline VN. In this study, high-quality VN(111) films were successfully synthesized on $$\alpha$$ α -Al $$_2$$ 2 O $$_3$$ 3 (0001) substrates using magnetron sputtering. The crystal and electronic structures of the VN films were characterized by a combination of high-resolution X-ray diffraction, low-energy electron diffraction, resonant soft X-ray absorption spectroscopy, and ultraviolet photoelectron spectroscopy. The electrical transport measurements indicate that the superconducting critical temperature of the VN films is around 8.1 K. Intriguingly, the temperature-dependent photoelectron spectroscopy measurements demonstrate a weak temperature dependence in the electronic structure of the VN films, which is significant for understanding the ground state of VN compounds.

Abstract Background Acute myeloid leukemia (AML) with biallelic (CEBPA bi) as well as single mutations located in the bZIP region is associated with a favorable prognosis, but the underlying mechanisms are still unclear. Here, we propose that two isoforms of C/EBPα regulate DNA damage-inducible transcript 3 (DDIT3) transcription in AML cells corporately, leading to altered susceptibility to endoplasmic reticulum (ER) stress and related drugs. Methods Human AML cell lines and murine myeloid precursor cell line 32Dcl3 cells were infected with recombinant lentiviruses to knock down CEBPA expression or over-express the two isoforms of C/EBPα. Quantitative real-time PCR and western immunoblotting were employed to determine gene expression levels. Cell apoptosis rates were assessed by flow cytometry. CFU assays were utilized to evaluate the differentiation potential of 32Dcl3 cells. Luciferase reporter analysis, ChIP-seq and ChIP-qPCR were used to validate the transcriptional regulatory ability and affinity of each C/EBPα isoform to specific sites at DDIT3 promoter. Finally, an AML xenograft model was generated to evaluate the in vivo therapeutic effect of agents. Results We found a negative correlation between CEBPA expression and DDIT3 levels in AML cells. After knockdown of CEBPA, DDIT3 expression was upregulated, resulting in increased apoptotic rate of AML cells induced by ER stress. Cebpa knockdown in mouse 32Dcl3 cells also led to impaired cell viability due to upregulation of Ddit3, thereby preventing leukemogenesis since their differentiation was blocked. Then we discovered that the two isoforms of C/EBPα regulate DDIT3 transcription in the opposite way. C/EBPα-p30 upregulated DDIT3 transcription when C/EBPα-p42 downregulated it instead. Both isoforms directly bound to the promoter region of DDIT3. However, C/EBPα-p30 has a unique binding site with stronger affinity than C/EBPα-p42. These findings indicated that balance of two isoforms of C/EBPα maintains protein homeostasis and surveil leukemia, and at least partially explained why AML cells with disrupted C/EBPα-p42 and/or overexpressed C/EBPα-p30 exhibit better response to chemotherapy stress. Additionally, we found that a low C/EBPα p42/p30 ratio induces resistance in AML cells to the BCL2 inhibitor venetoclax since BCL2 is a major target of DDIT3. This resistance can be overcome by combining ER stress inducers, such as tunicamycin and sorafenib in vitro and in vivo. Conclusion Our results indicate that AML patients with a low C/EBPα p42/p30 ratio (e.g., CEBPA bi) may not benefit from monotherapy with BCL2 inhibitors. However, this issue can be resolved by combining ER stress inducers.

Despite advances in understanding the development and progression of cancer in recent years, there remains a lack of comprehensive characterization of the cancer glycoproteome. Glycoproteins play an important role in medicine and are involved in various human disease conditions including cancer. Glycan-moieties participate in fundamental cancer processes like cell signaling, invasion, angiogenesis, and metastasis. Aberrant N-glycosylation significantly impacts cancer processes and targeted therapies in clinic. Therefore, understanding N-glycosylation in a tumor is essential for comprehending disease progression and discovering anti-cancer targets and biomarkers for therapy monitoring and diagnosis. This review presents the fundamental process of protein N-glycosylation and summarizes glycosylation changes in tumor cells, including increased terminal sialylation, N-glycan branching, and core-fucosylation. Also, the role of N-glycosylation in tumor signaling pathways, migration, and metabolism are discussed. Glycoproteins and glycopeptides as potential biomarkers for early detection of cancer based on site specificity have been introduced. Collectively, understanding and exploring the cancer glycoproteome, along with its role in medicine, implication in cancer and other human diseases, highlights the significance of N-glycosylation in tumor processes, necessitating further research for potential anti-cancer targets and biomarkers.

Abstract Objective To investigate the clinical features, treatment strategies, and prognosis of neuroblastoma with bilateral blindness. Methods The clinical data of five patients with bilateral blindness neuroblastoma admitted to Beijing Children’s Hospital from April 2018 to September 2020 were retrospectively collected to summarize their clinical characteristics. Results All patients were female and the median age at presentation was 25 (23, 41) months. The median intervention time from the onset of symptoms of bilateral blindness to the start of treatment was 10 (10, 12) days. All five cases were staged as stage M and grouped as high risk. Four cases were MYCN gene amplification and one case was MYCN acquisition. Five children were treated according to a high-risk neuroblastoma treatment protocol. Four children did not recover their vision after treatment, and one case improved to have light perception. All patients were effectively followed up for a median of 20 (12, 31) months, with three deaths, one tumor-free survival, and one recurrent tumor-bearing survival. Conclusion Neuroblastoma with bilateral blindness is rare in the clinic, mostly in children of young age, and is often associated with MYCN amplification and multiple metastases. Early hormone shock therapy and optic nerve decompression are beneficial for preserving the child’s vision. A joint multi-disciplinary treatment may help in the formulation of treatment decisions. Achieving a balance between good visual preservation and survival within the short optic nerve neurotherapeutic window is extremely challenging.

Introduction: Chemicals, such as MNU (N-methyl-N-nitrosourea) and NaIO3 (sodium iodate), are widely used to induce retinal degeneration in rodents. Streptozotocin (STZ) is an analog of N-acetyl glucosamine in which an MNU moiety is linked to a hexose and has a special toxic effect on insulin-producing pancreatic β-cells. It is commonly used to induce hyperglycemia to model diabetes. While intracerebroventricular injection of STZ can produce Alzheimer's disease independent of hyperglycemia, most retinal studies using STZ focus on the effects of hyperglycemia on the retina, but whether STZ has any impact on retinal cells independent of hyperglycemia is unknown. We aimed to investigate the role of cytotoxicity of STZ in rat retina.Methods: Intravitreal or subcutaneous injection of STZ was performed on newborn rats. Electroretinogram (ERG) and H&E staining investigated retinal function and morphological changes. Retinal cell types, cell death, proliferation, inflammation, and angiogenesis were studied by immunostaining. RNA sequencing was performed to examine the transcriptome changes of retinal cells after intravitreal injection of STZ.Results: Intravitreal (5 μg or 10 μg) or subcutaneous (30 mg/kg) injection of STZ at the early stage of newborn rats couldn’t induce hyperglycemia but caused NSIR (Neonatal STZ-induced retinopathy), including reduced ERG amplitudes, retinal rosettes and apoptosis, cell cycle arrest, microglial activation, and delayed retinal angiogenesis. STZ did not affect the early-born retinal cell types but significantly reduced the late-born ones. Short-term and long-term hyperglycemia had no significant effects on the NSIR phenotypes. RNA sequencing revealed that STZ induces oxidative stress and activates the p53 pathway of retinal cells. Locally or systemically, STZ injection after P8 couldn’t induce SINR when all retinal progenitors exit the cell cycle.Conclusion: NSIR in rats is independent of hyperglycemia but due to STZ’s direct cytotoxic effects on retinal progenitor cells. NSIR is a typical reaction to STZ-induced retinal oxidative stress and DNA damage. This significant finding suggests that NSIR may be a valuable model for studying retinal progenitor DNA damage-related diseases, potentially leading to new insights and treatments.

The incompletely eliminated Treponema pallidum (T. pallidum) during primary syphilis chancre infection can result in the progression of secondary, tertiary, or latent syphilis in individuals, suggesting that T. pallidum has successfully evaded the immune response and spread to distant sites. The mechanism underlying the dissemination of T. pallidum is unclear. Here, a syphilitic rabbit model dorsal-injected with recombinant Tp0136 protein or Tp0136 antibody subcutaneously was used to demonstrate the role of Tp0136 protein in promoting the dissemination of T. pallidum to the testis and angiogenesis in vivo; vascular endothelial cell line HMEC-1 was employed to display that Tp0136 protein enhances the angiogenesis. Furthermore, the three-dimensional microfluidic angiogenesis system showed that the angiogenesis would heighten vascular permeability. Then transcriptome sequencing analysis, in conjunction with cell-level validation, elucidated the critical role of the PI3K-AKT signaling pathway in the promotion of angiogenesis by Tp0136 protein, resulting in heightened permeability. These findings elucidate the strategy employed by T. pallidum in evading immune clearance.

The bamboo is widely distributed around the world and the product process is developed well, the biomass bamboo fiber composite is potential to providing strong confinement for concrete, therefore in this paper, bamboo sheet twining tube-confined concrete columns with an inner original bamboo tube (BSTCB) is proposed. This column is a new structure that consists of an external bamboo fiber composite tube and an internal original bamboo tube filled with concrete between the outer and inner tubes. A total of 18 specimens were tested in axial compression. The effects of BCT thickness and original bamboo tube diameter on the mechanical properties of the specimens were analyzed. The experimental results suggested that BCT has a strong lateral confinement ability. The strength and deformability of concrete increased significantly when BCT thickness increased. When the number of BCT layers was in the range of 6–18 layers, the increase in peak stress ranged from 7.7 % to 12.8 % and the increase in ultimate strain ranged from 6.4 % to 24.0 %. The increase in the diameter of the original bamboo tubes had a positive effect to the deformation capacity, while limited effect on the concrete compressive strength. Within the change in diameter from 90 mm to 110 mm, the reduction in concrete strength is within 6.0 %, and the change in ultimate strain ranges from 8.5 % to 14.1 %. Finally, the peak stress and strain of the concrete were evaluated using the existing FRP-confined concrete model, and the full stress-strain curve of the concrete was obtained.

Ecological source areas play a pivotal role in nature preservation and soil conservation. These areas are critical in optimizing land-use patterns and fostering the establishment facilitating the establishment of protected ecological source areas. In this study, two types of state layers of ecological source area change were extracted based on land cover data and integrated with an intelligent algorithm to analyze the role of ecological factors contributing to land change. Recommendations are proposed to enhance the optimization of the ecological spatial pattern within the Yellow River Basin. The results indicate key findings: (1) The ecological source areas in the Daxia River Basin experienced Fluctuations between 1990 and 2020 due to the interconversion of grassland and unused land. The change was relatively smooth during 1990–2000, with the ecological source areas increasing slightly by 16.016 km2. From 2000 to 2010, the proportion of unutilized land transferred to grassland was 6.3%, which is significantly higher than the annual transfer average of 1.6%. This is mainly due to the ecological protection measures in the cities of Linxia and Hezuo. Between 2010 and 2020, economic construction resulted in the loss of grassland, and the percentage of grassland turned out was 7.3%, which was significantly higher than the annual average of 1.8%. (2) In the Daxia River Basin, rainfall, sunlight intensity, temperature, and elevation had a greater impact on land change during the period from 1990 to 2020, with contributions of −0.4, 0.09, 0.08, and −0.26, respectively.

Abstract Haplotype‐resolved 3D chromatin architecture related to allelic differences in avian skeletal muscle development has not been addressed so far, although chicken husbandry for meat consumption has been prevalent feature of cultures on every continent for more than thousands of years. Here, high‐resolution Hi‐C diploid maps (1.2‐kb maximum resolution) are generated for skeletal muscle tissues in chicken across three developmental stages (embryonic day 15 to day 30 post‐hatching). The sequence features governing spatial arrangement of chromosomes and characterize homolog pairing in the nucleus, are identified. Multi‐scale characterization of chromatin reorganization between stages from myogenesis in the fetus to myofiber hypertrophy after hatching show concordant changes in transcriptional regulation by relevant signaling pathways. Further interrogation of parent‐of‐origin‐specific chromatin conformation supported that genomic imprinting is absent in birds. This study also reveals promoter‐enhancer interaction (PEI) differences between broiler and layer haplotypes in skeletal muscle development‐related genes are related to genetic variation between breeds, however, only a minority of breed‐specific variations likely contribute to phenotypic divergence in skeletal muscle potentially via allelic PEI rewiring. Beyond defining the haplotype‐specific 3D chromatin architecture in chicken, this study provides a rich resource for investigating allelic regulatory divergence among chicken breeds.

Abstract Chorus subpackets are the wave packets with modulated amplitudes in chorus waves, commonly observed in the magnetospheres of Earth and other planets. Nonlinear wave‐particle interactions have been suggested to play an important role in subpacket formation, yet the corresponding electron dynamics remain not fully understood. In this study, we have investigated the electron trapping through cyclotron resonance with subpackets, using a self‐consistent general curvilinear plasma simulation code simulation model in dipole fields. The electron trapping period has been quantified separately through electron dynamic analysis and theoretical derivation. Both methods indicate that the electron trapping period is shorter than the subpacket period/duration. We have further established the relation between electron trapping period and subpacket period through statistical analysis using simulation and observational data. Our study demonstrates that the nonlinear electron trapping through cyclotron resonance is the dominant mechanism responsible for subpacket formation.

Among the Acinetobacter genus, Acinetobacter pittii stands out as an important opportunistic infection causative agent commonly found in hospital settings, which poses a serious threat to human health. Recently, the high prevalence of carbapenem-resistant A. pittii isolates has created significant therapeutic challenges for clinicians. Bacteriophages and their derived enzymes are promising therapeutic alternatives or adjuncts to antibiotics effective against multidrug-resistant bacterial infections. However, studies investigating the depolymerases specific to A. pittii strains are scarce. In this study, we identified and characterized a capsule depolymerase, Dpo27, encoded by the bacteriophage IME-Ap7, which targets A. pittii. A total of 23 clinical isolates of Acinetobacter spp. were identified as A. pittii (21.91%, 23/105), and seven A. pittii strains with various K locus (KL) types (KL14, KL32, KL38, KL111, KL163, KL207, and KL220) were used as host bacteria for phage screening. The lytic phage IME-Ap7 was isolated using A. pittii 7 (KL220) as an indicator bacterium and was observed for depolymerase activity. A putative tail fiber gene encoding a polysaccharide-degrading enzyme (Dpo27) was identified and expressed. The results of the modified single-spot assay showed that both A. pittii 7 and 1492 were sensitive to Dpo27, which was assigned the KL220 type. After incubation with Dpo27, A. pittii strain was susceptible to killing by human serum; moreover, the protein displayed no hemolytic activity against erythrocytes. Furthermore, the protein exhibited sustained activity across a wide pH range (5.0–10.0) and at temperatures between 20 and 50°C. In summary, the identified capsule depolymerase Dpo27 holds promise as an alternative treatment for combating KL220-type A. pittii infections.

Abstract Background As treatment strategies differ according to endotype, rhinologists must accurately determine the endotype in patients affected by chronic rhinosinusitis with nasal polyps (CRSwNP) for the appropriate management. In this study, we aim to construct a novel deep learning model using paranasal sinus computed tomography (CT) to predict the endotype in patients with CRSwNP. Methods We included patients diagnosed with CRSwNP between January 1, 2020, and April 31, 2023. The endotype of patients with CRSwNP in this study was classified as eosinophilic or non-eosinophilic. Sinus CT images (29,993 images) were retrospectively collected, including the axial, coronal, and sagittal planes, and randomly divided into training, validation, and testing sets. A residual network-18 was used to construct the deep learning model based on these images. Loss functions, accuracy functions, confusion matrices, and receiver operating characteristic curves were used to assess the predictive performance of the model. Gradient-weighted class activation mapping was performed to visualize and interpret the operating principles of the model. Results Among 251 included patients, 86 and 165 had eosinophilic or non-eosinophilic CRSwNP, respectively. The median (interquartile range) patient age was 49 years (37–58 years), and 153 (61.0%) were male. The deep learning model showed good discriminative performance in the training and validation sets, with areas under the curves of 0.993 and 0.966, respectively. To confirm the model generalizability, the receiver operating characteristic curve in the testing set showed good discriminative performance, with an area under the curve of 0.963. The Kappa scores of the confusion matrices in the training, validation, and testing sets were 0.985, 0.928, and 0.922, respectively. Finally, the constructed deep learning model was used to predict the endotype of all patients, resulting in an area under the curve of 0.962. Conclusions The deep learning model developed in this study may provide a novel noninvasive method for rhinologists to evaluate endotypes in patients with CRSwNP and help develop precise treatment strategies.

Earth’s magnetopause is a thin boundary separating the shocked solar wind plasma from the magnetospheric plasmas, and it is also the boundary of the solar wind energy transport to the magnetosphere. Soft X-ray imaging allows investigation of the large-scale magnetopause by providing a two-dimensional (2-D) global view from a satellite. By performing 3-D global hybrid-particle-in-cell (hybrid-PIC) simulations, we obtain soft X-ray images of Earth’s magnetopause under different solar wind conditions, such as different plasma densities and directions of the southward interplanetary magnetic field. In all cases, magnetic reconnection occurs at low latitude magnetopause. The soft X-ray images observed by a hypothetical satellite are shown, with all of the following identified: the boundary of the magnetopause, the cusps, and the magnetosheath. Local X-ray emissivity in the magnetosheath is characterized by large amplitude fluctuations (up to 160%); however, the maximum line-of-sight-integrated X-ray intensity matches the tangent directions of the magnetopause well, indicating that these fluctuations have limited impact on identifying the magnetopause boundary in the X-ray images. Moreover, the magnetopause boundary can be identified using multiple viewing geometries. We also find that solar wind conditions have little effect on the magnetopause identification. The Solar wind Magnetosphere Ionosphere Link Explorer (SMILE) mission will provide X-ray images of the magnetopause for the first time, and our global hybrid-PIC simulation results can help better understand the 2-D X-ray images of the magnetopause from a 3-D perspective, with particle kinetic effects considered.

Abstract Background Rectovaginal fistula (RVF) is an abnormal channel formed by epithelial tissue between the anterior wall of the rectum and the posterior wall of the vagina, which manifests as vaginal gassing and defecation. It is one of the common complications of female pelvic surgeries. With the increased number of proctectomies for rectal cancer, the number of postoperative rectovaginal fistulas also increases. Once RVF occurs, the failure rate is still high with various treatments available. RVF causes great suffering to women and is still a major problem in treatment. Therefore, it is significant for female rectal cancer patients to prevent RVF after rectal cancer surgery. In this study, we introduce a new method to prevent RVF during rectal cancer radical operation. Methods In this randomized controlled trial (RCT), all operations are performed according to the principle of total mesorectal excision (TME) radical resection in rectal cancer surgery. All eligible participants will be divided into two groups: the experimental group and the control group. Experimental group: the anterior rectal wall of about 1 cm distal to the anastomosis was dislocated. Before the anastomosis of the rectal end, a fat flap (usually left side) containing the ovarian vascular pedicle was dislocated, measured by 10–15 cm in length and 2 cm in width. The fat flap containing the ovarian vascular pedicle was packed and fixed anterior to the anastomotic stoma with fibrin glue. Control group: surgery will be carried out in accordance with the TME principle. Participants will be compared on several variables, including the incidence of RVF after operation (primary outcomes), the occurrence time of postoperative RVF, the occurrence time of RVF after stoma closure, and other postoperative complications, such as anastomotic leakage, chylous leakage, and intestinal obstruction (secondary outcomes). The follow-up data collection will be conducted according to the follow-up time point, and the baseline data will also be collected for follow-up analysis. By comparing the incidence of rectovaginal leakage between the experimental group and the control group, we aim to explore the feasibility of this method for the prevention of postoperative RVF. Discussion This RCT will explore the feasibility of packing with a laparoscopic dislocated fat flap containing an ovarian vascular pedicle anterior to the anastomotic stoma after rectal cancer surgery to prevent RVF. Trial registration Chinese Clinical Trial Registry (ChiCTR) registration ChiCTR2000031449. Registered on June 26, 2019. All items of the WHO Trial registration data set can be found within the protocol.

Abstract Background A public health emergency can cause large numbers of deaths in a short period, with devastating social, economic and health consequences. Nurses are the main healthcare providers during such emergencies, and their competencies affect the control and outcomes of the situation. Studies on nurses’ competencies in public health emergencies vary between countries and healthcare systems. Therefore, we conducted a scoping review to identify the common domains of nurses’ competencies in public health emergencies worldwide. Methods We searched the PubMed, CINHAL, Scopus, Web of Science, Science Direct, Embase, Cochrane Library, WanFang and ECRI databases from their inception to 2023. All published articles on nurses’ competencies in public health emergencies that were published in English and Chinese were included. We mainly analyzed and synthesized nurses’ competencies, assessment instruments and the training described in the included studies. Results A total of 27 competency domains were identified following an analysis and summary. The most frequently cited domains were communication skills, self-protection skills, basic knowledge of a public health emergency, laws and ethics and the capacity for organizational collaboration. The Disaster Preparedness Evaluation Tool and the Emergency Preparedness Information Questionnaire were the most commonly used tools for assessing competencies. Most training was conducted online and the content that was covered varied by country. Conclusions Given the significant roles and responsibilities of nurses in public health emergencies, knowing the domains of their competencies is essential to evaluating, developing, and conducting clinical training.

It emphasizes the importance of metal atom types, host material selection, and coordination environment in SACs design. The article explains how SACs enhance photocatalytic H2O2 production by improving light absorption, charge generation, migration, and lowering energy barriers for reactant adsorption and activation. The review acknowledges challenges and future research directions in SACs for H2O2 photosynthesis.

Abstract A 2‐D GCPIC simulation in a dipole field system has been conducted to explore the excitation of oblique whistler mode chorus waves driven by energetic electrons with temperature anisotropy. The rising tone chorus waves are initially generated near the magnetic equator, consisting of a series of subpackets, and become oblique during their propagation. It is found that electron holes in the wave phase space, which are formed due to the nonlinear cyclotron resonance, oscillate in size with time during subpacket formation. The associated inhomogeneity factor varies accordingly, giving rise to various frequency chirping in different phases of subpackets. Distinct nongyrotropic electron distributions are detected in both wave gyrophase and stationary gyrophase. Landau resonance is found to coexist with cyclotron resonance. This study provides multidimensional electron distributions involved in subpacket formation, enabling us to comprehensively understand the nonlinear physics in chorus wave evolution.

In this study, we delved into the safety profile of alpelisib, an FDA-approved treatment for hormone receptor-positive, HER2-negative, PIK3CA-mutated advanced or metastatic breast cancer, and PIK3CA-Related Overgrowth Spectrum (PROS). Despite its approval, real-world, long-term safety data is lacking. Our research scrutinizes the FDA database to assess alpelisib 's safety. We retrospectively analyzed data from April 2019 to June 2023 using four algorithms. Among 7,609,450 reports, 6692 implicated alpelisib as the primary suspected drug, uncovering adverse events (AEs) across 26 organ systems. Notably, we identified 21 previously unlisted AEs. Furthermore, differences in AEs emerged between patients with PIK3CA-mutated breast cancer and those with PROS. This study provides vital insights for healthcare professionals to navigate AEs in clinical practice and informs future research for enhancing alpelisib 's safety profile.

IntroductionThe shifting living and working conditions have profound impacts on the residents’ mental health. However, current research in this field has not remarkable investigated.MethodsThis study used the China Labor-force Dynamic Survey (CLDS) dataset from 2018 and relied on a regression model to examine the effects of the built environment, work environment, and subjective perception on the mental health of Chinese citizens. It also considers the circumstances of both migrants and local residents.ResultsThis study revealed significant correlations between mental health and greening space rate, road network density, commuting time, work feelings, community trust, economic satisfaction, and other factors. Additionally, the mental health of local residents was shown to be significantly affected by community security, while it shows no significance in migrants. Furthermore, a significant spatial autocorrelation was found in terms of mental health within the central and eastern regions of China.DiscussionThe findings of this study offer valuable insights that can be used to facilitate measures aimed at improving the mental health of residents and promoting the development of healthy cities.

ObjectivesTo investigate the etiology, clinical manifestations, imaging features, and treatment of patients with infratentorial superficial siderosis (iSS), enhance clinicians' comprehension of this rare disease, and conduct oral deferiprone intervention and subsequent monitoring.MethodsSix patients diagnosed with iSS based on magnetic resonance imaging (MRI) and susceptibility weighted imaging (SWI) were enrolled from 2021 to 2023 at the First Affiliated Hospital of Fujian Medical University. Their clinical datas were summarized, and the etiology and imaging characteristics were analyzed. Follow-up was conducted through telephone or outpatient visits.ResultsAmong the 6 patients, there were 3 males and 3 females. The onset age ranged from 35 to 71 years, with an average onset age of 53 years. The clinical symptoms mainly included acoustic disturbances (6/6), gait imbalance (6/6), dysolfactory (6/6), cognitive impairment (2/6), epilepsy (2/6), and pyramidal tract sign (2/6). Evidence of superficial siderosis was observed on MRI across the cortex, brainstem, cerebellum, and spinal cord in all patients. T2-space sequence MRI revealed two instances of dural tear. During the follow-up period ranging from 1 month to 3 years, three patients who received oral deferiprone treatment showed improvement, whereas the remaining three patients who declined deferiprone treatment demonstrated progression.ConclusionThe primary clinical manifestations of iSS include bilateral sensorineural hearing disturbances, progressive cerebellar ataxia, and spinal cord lesions. The key diagnostic criteria involve the presence of linear hypointensity on T2-WI in the surface region of the nervous system. Dural tear caused by various factors is considered to be the most common cause of iSS, and its treatment mainly involves surgical intervention for hemorrhagic primary diseases as well as pharmacotherapy with deferiprone.

Controlled functionalization of 2D black phosphorus (BP) nanosheets provides unique opportunities to tune their chemical, physical, and electronic properties. Herein, the preparation of single‐atom nickel‐doped BP (Ni–BP) sheets using a simple solution‐based strategy is reported. Using the Ni–BP sheets as a passivation layer on top of a perovskite film leads to standard perovskite solar cells (PSCs) with improved performance. The standard n–i–p PSCs with the Ni–BP interlayer achieve maximum power conversion efficiencies of over 22%, with negligible hysteresis and a modest improvement in stability when subjected to different testing conditions. The perovskite films prepared with Ni–BP sheets‐based passivation are found to have reduced defect densities as well as improved charge‐transfer properties and carrier lifetimes. Density‐functional theory calculations support the experimental results through showing that the atomic Ni‐doping increases the work function of the BP interlayer, enabling better hole extraction, as well as increasing the surface hydrophobicity of the BP layer, hence reducing water sorption into the perovskite film.

Abstract Objective To summarize the clinical characteristics of children with adrenocortical carcinoma (ACC) and preliminarily explore the indications for and efficacy of neoadjuvant chemotherapy in certain patients. Methods The data of 49 children with adrenocortical tumors (ACT) in the past 15 years were retrospectively analyzed, and after pathology assessment using Weiss system grading, 40 children diagnosed with ACC were included. Response Evaluation Criteria in Solid Tumors (RECIST) 1.1 and three-dimensional (3D) reconstruction of contrast-enhanced computed tomography data were used to evaluate the response to neoadjuvant chemotherapy. Results Forty patients (17 males, 23 females) with ACC were enrolled. Abnormal hormone levels were common in children with ACC (n = 31), and in terms of clinical presentation, sexual precocity was the most common (n = 14, 35.0%), followed by Cushing’s syndrome (n = 12, 30.0%). Seven of 40 children received neoadjuvant chemotherapy due to a maximum lesion diameter greater than 10 cm (n = 4), invasion of surrounding tissues (n = 2), intravenous tumor thrombus (n = 2), and/or distant metastasis (n = 2); 2 patients achieved partial response, and 5 had stable disease according to the RECIST 1.1 standard. Furthermore, 3D tumor volume reconstruction was performed in 5 children before and after neoadjuvant chemotherapy. Tumor volumes were significantly reduced in all 5 children, with a median volume reduction of 270 (interquartile range, IQR 83, 293) (range: 49–413) ml. After surgery with/without chemotherapy, the 5-year overall survival rate for all children was 90.0% (95% CI-confidence interval 80.0–100.0%), and the 5-year event-free survival rate was 81.5% (95% CI 68.0–97.7%). Conclusion In the diagnosis and treatment of pediatric ACC, a comprehensive endocrine evaluation is necessary to facilitate early diagnosis. Surgery and chemotherapy are important components of ACC treatment, and neoadjuvant chemotherapy should be considered for children with ACC who meet certain criteria, such as a large tumor, distant metastases, or poor general condition.

Abstract Background Macrophage-like transformation of vascular smooth muscle cells (VSMCs) is a risk factor of atherosclerosis (AS) progression. Transcription factor homeobox A1 (HOXA1) plays functional roles in differentiation and development. This study aims to explore the role of HOXA1 in VSMC transformation, thereby providing evidence for the potential mechanism of AS pathogenesis. Methods High fat diet (HFD)-fed apolipoprotein E knockout (ApoE−/−) mice were applied as an in vivo model to imitate AS, while 1-palmitoyl-2-(5-oxovaleroyl)-sn-glycero-3-phosphocholine (POV-PC)-treated VSMCs were applied as an in vitro model. Recombinant adeno-associated-virus-1 (AAV-1) vectors that express short-hairpin RNAs targeting HOXA1, herein referred as AAV1-shHOXA1, were generated for the loss-of-function experiments throughout the study. Results In the aortic root of AS mice, lipid deposition was severer and HOXA1 expression was higher than the wide-type mice fed with normal diet or HFD. Silencing of HOXA1 inhibited the AS-induced weight gain, inflammatory response, serum and liver lipid metabolism disorder and atherosclerotic plaque formation. Besides, lesions from AS mice with HOXA1 knockdown showed less trans-differentiation of VSMCs to macrophage-like cells, along with a suppression of krüppel-like factor 4 (KLF4) and nuclear factor (NF)-κB RelA (p65) expression. In vitro experiments consistently confirmed that HOXA1 knockdown suppressed lipid accumulation, VSMC-to-macrophage phenotypic switch and inflammation in POV-PC-treated VSMCs. Mechanism investigations further illustrated that HOXA1 transcriptionally activated RelA and KLF4 to participate in the pathological manifestations of VSMCs. Conclusions HOXA1 participates in AS progression by regulating VSMCs plasticity via regulation of NF-κB p65 and KLF4. HOXA1 has the potential to be a biomarker or therapeutic target for AS.

Background: Driving fatigue is a significant concern in contemporary society, contributing to a considerable number of traffic accidents annually. This study explores novel methods for fatigue detection, aiming to enhance driving safety. Methods: This study utilizes electroencephalography (EEG) and functional near-infrared spectroscopy (fNIRS) to monitor driver fatigue during simulated driving experiments lasting up to 7 h. Results: Analysis reveals a significant correlation between behavioral data and hemodynamic changes in the prefrontal lobe, particularly around the 4 h mark, indicating a critical period for driver performance decline. Despite a small participant cohort, the study’s outcomes align closely with established fatigue standards for drivers. Conclusions: By integrating fNIRS into non-voluntary attention brain function experiments, this research demonstrates promising efficacy in accurately detecting driving fatigue. These findings offer insights into fatigue dynamics and have implications for shaping effective safety measures and policies in various industrial settings.

This study investigated the impacts of biochar addition on N conversion, humification, and bacterial community during Flammulina velutipes waste composting. The mixture of chicken manure and Flammulina velutipes waste was 4:6 (dry weight basis). The biochar was added into the mixture and mixed thoroughly at ratios of 0, 2.5, 5, and 7.5% (w/w) and labeled as CK, T1, T2, and T3, respectively. The results showed that the biochar treatment significantly improved the compost maturity by increasing humic substances and the conversion of NH4+-N to NO3−-N. With the increase in biochar supplemental level, the abundance, diversity, and uniformity of the microbial community were improved. The dominant taxa were Firmicutes, Bacteroidota, Actinobacteriota, Proteobacteria, and Gemmatimonadota, especially the Firmicutes and Bacteroidota. Biochar addition facilitated the proliferation of thermophilic bacteria such as Bacillus, Actinobacteriota, Parapedobacter, and Sphingobacterium, leading to enhanced organic decomposition to increase humus. The findings of this study highlighted the positive effects of biochar addition on the composting mixture of chicken manure and Flammulina velutipes waste. These results can help to produce high-quality biochar composting products by balancing organic decomposition and humification based on the bacterial community.

Chronic rhinosinusitis (CRS) is a global disease characterized by poor treatment outcomes and high recurrence rates, significantly affecting patients’ quality of life. Due to its complex pathophysiology and diverse clinical presentations, CRS is categorized into various subtypes to facilitate more precise diagnosis, treatment, and prognosis prediction. Among these, CRS with nasal polyps (CRSwNP) is further divided into eosinophilic CRSwNP (eCRSwNP) and noneosinophilic CRSwNP (non-eCRSwNP). However, there is a lack of precise predictive diagnostic and treatment methods, making research into accurate diagnostic techniques for CRSwNP endotypes crucial for achieving precision medicine in CRSwNP. This paper proposes a method using multiangle sinus computed tomography (CT) images combined with artificial intelligence (AI) to predict CRSwNP endotypes, distinguishing between patients with eCRSwNP and non-eCRSwNP. The considered dataset comprises 22,265 CT images from 192 CRSwNP patients, including 13,203 images from non-eCRSwNP patients and 9,062 images from eCRSwNP patients. Test results from the network model demonstrate that multiangle images provide more useful information for the network, achieving an accuracy of 98.43%, precision of 98.1%, recall of 98.1%, specificity of 98.7%, and an AUC value of 0.984. Compared to the limited learning capacity of single-channel neural networks, our proposed multichannel feature adaptive fusion model captures multiscale spatial features, enhancing the model’s focus on crucial sinus information within the CT images to maximize detection accuracy. This deep learning-based diagnostic model for CRSwNP endotypes offers excellent classification performance, providing a noninvasive method for accurately predicting CRSwNP endotypes before treatment and paving the way for precision medicine in the new era of CRSwNP.

This study combines photovoltaic (PV) technology with rural revitalization, constructs a new long-term mechanism by using a BOT model, and applies fuzzy number processing to the model of the real option method to dig out the potential value of the project. Based on the field research of X county, the article comprehensively analyzes the PV power generation project in terms of economic and social benefits and its impact on poverty alleviation. The results show that the total benefit of X county can reach 3720.34 million yuan in 30 years, and the average annual benefit is about 124.011 million yuan. The full power generation capacity of the project is 4,987 MWh, and the total income from electricity sales reaches 471,985,000 yuan. The PV power project has contributed significantly to environmental protection, with reductions in carbon dust, carbon dioxide, sulfur dioxide, and nitrogen oxides of 12.12 billion tons, 44.61 billion tons, 13.77 billion tons, and 750 million tons, respectively. The study also found that villages with larger administrative territory and suitable topographic conditions are more ideal for establishing large-scale PV ground power plants to help alleviate poverty. Meanwhile, villages with good grid quality are more efficient in reducing poverty when implementing PV poverty alleviation programs. This study provides an innovative perspective for village revitalization strategies and demonstrates the great potential of PV technology in promoting village economic development and environmental protection.

Currently, more than 170 modifications have been identified on RNA. RNA modification mainly regulates RNA splicing, intracellular transport, degradation, translation, and stability. Gynecologic cancer (GC) mainly includes cervical cancer (CCA), ovarian cancer (OC), Endometrial cancer (EMC), among others, is the leading cause of cancer-related death. At present, there is still a lack of effective means to eradicate such diseases, so it is important to conduct more in-depth research on gynecological cancers. Numerous studies have shown that a series of epigenetic changes occur during the development of gynecologic cancer. This article reviews the latest findings on the functional significance of RNA modification in gynecologic cancer and discusses the therapeutic potential of RNA modification-related inhibitors in the treatment of gynecologic cancer.

ObjectiveSince 2022, Omicron has been circulating in China as a major variant of the novel coronavirus, but the effects of infection with Omicron variants on pregnant women and newborns are unknown. The purpose of this study was to determine the clinical characteristics of Omicron infection during pregnancy and its effect on pregnancy outcomes.MethodsThis study retrospectively analyzed the data of 93 confirmed cases of novel coronavirus infection and 109 non-infected patients admitted to the isolation ward of Guangdong Maternal and Child Health Hospital from December 1, 2022 to January 31, 2023, and statistically analyzed the clinical features of Omicron variant infection during pregnancy and its impact on pregnancy outcomes. Further effects of underlying diseases on Omicron infection in pregnant women were analyzed.ResultsThe incubation period of COVID-19 infection was 0.99±0.86 days, 94.38% of patients had fever or other respiratory symptoms, the lymphocyte count in the infected group was lower than that in the uninfected group, and the lymphocyte count was further reduced in the patients with pregnancy complications or complications. Compared with the uninfected group, APTT and PT were prolonged, platelet count and fibrinogen were decreased in the infected group, all of which had statistical significance. COVID-19 infection during pregnancy increased the rate of cesarean section compared to uninfected pregnant patients, and COVID-19 infection in gestational diabetes resulted in a 4.19-fold increase in cesarean section rate. There was no statistically significant difference in gestational age between the two groups. The incidence of intrauterine distress, turbidity of amniotic fluid and neonatal respiratory distress were higher in the infection group. No positive cases of neonatal COVID-19 infection have been found.ConclusionThe patients infected with omicron during pregnancy often have febrile respiratory symptoms with lymphocyopenia, but the incidence of severe disease is low. Both Omicron infection and gestational diabetes further increase the incidence of cesarean section, and this study found no evidence of vertical transmission of Omicron.

The current energy system of Portugal is heavily reliant on fossil fuels, which accounted for 65.4 % of primary energy consumption in 2021. The low share of renewable energy in the industrial, transportation, and heating & cooling sectors is the primary reason for this dependence. To achieve carbon neutrality and ensure a successful energy transition, the Portuguese government has introduced the "Carbon Neutrality Roadmap 2050", a strategic plan to decarbonise the energy system entirely. This study presents technical solutions to achieve carbon neutrality in the overall energy sector, based on the guidelines of the Portuguese government plans and assuming a high share of green hydrogen utilisation in the industrial and transportation sectors. Hourly energy balances were calculated using the EnergyPLAN software, which revealed the substantial investment required in new renewable electricity sources to achieve decarbonisation of the industrial and transportation sectors. The study's findings emphasise the urgent need to transition to a low-carbon energy system and inform policymakers and energy experts in developing effective strategies towards achieving carbon neutrality.

AbstractNatural hazards always have significant impacts on the regional ecological environment, causing huge damages to biodiversity, soil erosion, and impoverishment. This study aimed to analyze the variation of ecological environment quality in Nepal from 2014 to 2018, with a specific focus on the impact and recovery of the 2015 Ms8.1 Nepal earthquake. We utilized Google Earth Engine (GEE) and remote sensing based ecological index (RSEI) to evaluate the ecological environment quality. Moreover, Moran’s I and Geodetector model were employed to explore spatial clustering and driving forces of post-earthquake recovery. The results indicate that after the 2015 Nepal earthquake, there was a decrease of 0.0225 in the mean RSEI in Nepal. The monitoring of the severe-affected areas showed that the regional environmental quality gradually improved within three years after the earthquake. After a span of three years, the ecological environment managed to recover to its pre-earthquake level. In terms of spatial distribution, there is a notable correlation in Nepal, with the ecological environment exhibiting higher quality in the southern regions and lower quality in the northern regions. By examining the driving forces behind the changes in ecological environment quality during the post-earthquake recovery period, it has been determined that altitude, precipitation, and their interaction serve as influential factors. These findings provide valuable insights into the state of ecological environment recovery after the 2015 Nepal earthquake. Furthermore, this study establishes a robust theoretical foundation for local decision-makers to develop ecological protection measures and enhance local disaster risk management.

Research on digital economy (DE) and carbon environment governance (CEG) holds paramount importance in attaining sustainable development objectives. By focusing on 30 provinces within China as specific instances, this research constructed a comprehensive evaluation framework to assess the overall progress of DE and CEG from 2011 to 2020. Moreover, it employed a coupling coordination model alongside spatio-temporal evolution patterns to examine the state of coupling coordination development (CCD) and the temporal and spatial features within DE-CEG. First, the overall levels of China's DE and CEG were both showing an upward trend, with the average level of CCD improving year by year. The spatio-temporal arrangement exhibited a distinctive “bell-shaped” pattern, characterized by narrow at both ends, wide in the middle. The average level of CCD in three major regions was east, central and west followed a descending order, underscoring notable spatial and temporal disparities within the DE-CEG. Second, kernel density estimation and exploratory spatial data analysis substantiated the spatio-temporal differentiation for the CCD within DE-CEG. Notably, these analyses unveiled prevailing spatial patterns of “high-high” and “low-low” agglomeration, spatial agglomeration effect became significant over time. Finally, among the absolute differences, the central region exhibited the least disparity, while the western region displayed the greatest variation, with absolute differences undergoing a fluctuating trajectory. Among the relative differences, the most pronounced contrast was observed between the eastern and western regions, whereas the disparities between the central-east and central-west regions were comparable. Remarkably, relative differences accounted for over 40% of the total variation, emerging as the predominant contributor to the overall divergence in CCD within China's DE-CEG framework. Consequently, it is imperative for future developmental efforts to focus on diminishing the relative disparities among regions. In light of the research findings, a set of strategies and recommendations had been formulated to propel the CCD of China's DE and CEG.

Aberrant Septin9 methylation in cervical cancer has been rarely studied. We aimed to identify its diagnostic value in cervical cancer using cervical scrapings, and its predictive potential in plasma for pelvic nodal metastasis of cervical cancer. The statuses of methylated Septin9 in fresh cervical lesions and cervical scrapings were first evaluated by using quantitative methylation-specific PCR. Subsequently, the relationship between Septin9 methylation in 113 plasma samples and pelvic nodal metastasis of cervical cancer was evaluated. Methylated Septin9 was detected in all cancerous tissues, but not in cervicitis. The degrees of Septin9 methylation increased with growing severity of cervical lesions in cervical scrapings. The sensitivity of methylated Septin9 was lower than that of cytology, while it yielded a high specificity and area under the curve in detecting high-grade squamous intraepithelial lesion or cervical cancer; and when Septin9 methylation combined with HPV16/18 genotyping, the sensitivity would increase from 70.42% to 82.39%. Plasma-based Septin9 methylation had a high discriminatory power in predicting pelvic nodal metastasis of cervical cancer, with an optimal specificity of 81.48%. In conclusion, we demonstrated methylated Septin9 to be an innovative diagnostic biomarker for cervical cancer and its non-invasive predictive potential in plasma for pelvic nodal metastasis of cervical cancer.Impact statement What is already known on this subject? The occurrence of cervical cancer is related to Septin9 methylation. In fresh specimens and cervical scrapings, we found the degrees of methylated Septin9 increased with growing severity of cervical lesions. Compared with HPV16/18 genotyping and cytological detection, Septin9 methylation had a better specificity and AUC in detecting ≥ HSIL. Furthermore, plasma-based Septin9 methylation also had a high specificity for pelvic lymphatic metastasis prediction. What the results of this study add? Methylation analysis of Septin9 indicated a similar sensitivity, specificity and AUC in detecting ≥ HSIL, relative to HPV16/18 genotyping. Compared with cytological method, Septin9 methylation also yielded a higher specificity and AUC in detecting ≥ HSIL. And we also found plasma-based Septin9 methylation had a high discriminatory power in predicting pelvic nodal metastasis of cervical cancer, with an optimal specificity of 81.48%; additionally an increasing sensitivity from 50% to nearly 80% was found when combined with SCCAg. What the implications are of these findings for clinical practice and/or further research? This study aimed to evaluate the relationship between Septin9 methylation and cervical cancer, and to explore the value of methylated Septin9 in the detection of cervical (pre)cancerous lesions. Moreover, we would explore plasma-based ctDNA biomarkers for pelvic lymphatic metastasis prediction of cervical cancer, to improve non-invasive predictive accuracy of pelvic nodal metastasis and reduce the complications caused by pelvic lymphadenectomy.

Studies have investigated PAX1 and SEPT methylation were closely associated with cervical cancer. For this study, we verified the expressions of PAX1 and SEPT9 methylation in 236 hrHPV women cervical exfoliated cells by using quantitative methylation-specific PCR and we further explored their diagnostic value in cervical (pre)cancer detection. Our results identified that the methylation rates and levels of PAX1 and SEPT9 increased with cervical lesion severity. For a diagnosis of cervical (pre)cancer, the area under the curve (AUC) of PAX1 methylation was 0.77 (95% CI 0.71–0.83) and the AUC of SEPT9 methylation was 0.86 (95% CI 0.81∼0.90). Analyses of the PAX1 and SEPT9 methylation statuses alone or combined with commonly used tests can efficiently identify cervical (pre)cancer. In particular, SEPT9 methylation might serve as an effective and powerful biomarker for the diagnosis of cervical (pre)cancer and as an alternative triage test in HPV-based cervical (pre)cancer screening programs.Impact Statement What is already known on this subject? This subject showed that PAX1 and SEPT9 methylation were closely associated with cervical cancer. The methylation rates and levels of PAX1 and SEPT9 increased with cervical lesion severity and reached a peak in cervical cancer exfoliated cells. We further assessed the diagnostic performances of PAX1 and SEPT9 methylation in cervical cancer screening. In detecting cervical (pre)cancer, the sensitivity values of PAX1 and SEPT9 methylation were up to 61.18% and 82.35%, respectively, and the specificity values of PAX1 and SEPT9 methylation were up to 95.36% and 86.75%, respectively. Moreover, the ROC curve analysis showed AUC values of 0.77 for PAX1 methylation and 0.86 for SEPT9 methylation tests, which were significantly superior to other commonly used tests. These findings suggest that PAX1 and SEPT9 methylation detection may have great clinical potential in cervical cancer screening. What the results of this study add? The rates and levels of PAX1 and SEPT9 methylation increased with the severity of the cervical lesions. For a diagnosis of cervical (pre)cancer, the area under the curve (AUC) of PAX1 methylation was 0.77 (95% CI 0.71–0.83), and the sensitivity and specificity values were 61.18% and 95.36%, respectively. The methylation levels of PAX1 and SEPT9 had a certain predictive effect on the severity of cervical lesions in hrHPV-positive women. In addition, SEPT9 methylation analysis performs better than PAX1 methylation analysis and commonly used tests in cervical exfoliated cells for detecting cervical (pre)cancer in hrHPV-positive women. SEPT9 methylation analysis merits consideration as an effective and objective, alternative triage test in HPV-based cervical (pre)cancer screening programs.

To enhance the repairability of lateral force resisting systems after earthquakes, a novel precast rocking wall with multiple steel energy dissipaters (PRW-MED) was devised, and a hierarchical energy dissipation (HED) mechanism was proposed based on this structure. Three quasistatic tests were conducted on the rocking wall specimens to evaluate the seismic performance of the PRW-MED. Simulations of the experiment were carried out in OpenSees software, and the influences of several parameters on the performance of the PRW-MED were investigated. The results indicated that the structure possessed excellent self-centering and deformation capabilities; the multiple dissipaters contributed sufficient energy dissipation capacity to the structure, and the parameters of the steel panel dampers (SPDs) exhibited significant effects on the seismic performance of the structure. Meanwhile, the experiment proved that the application of multi dissipaters led to the proposed HED mechanism. The parametric analysis demonstrated that the design parameters including initial stress of strands, thickness of SPD, and width of SPD etc. contributed significantly to the seismic performance of PRW-MED.

AbstractThe increase in the number and severity of seismic disasters has put communities in danger, especially in rapidly developing and densely populated areas. Traditional seismic vulnerability and risk assessment methods, including field investigation, are accurate at the building scale; however, their low-efficiency and high-cost characteristics limit the application of these methods in urban-scale regions with high-speed development and risk exposure. To address this issue, multisource remote sensing interpretation, support vector machine (SVM) and geographic information science (GIScience) technologies are combined to test the performance and efficiency of a urban-scale macroscopic seismic vulnerability and risk assessment method in the Lixia District of Jinan City, Shandong Province, China, which is characterized by rapid development, a variety of building types, and moderate-to-low seismic risk. First, a traditional field survey was conducted in Lixia District, and a building attribute information database was constructed. Second, the vulnerability proxies of building attribute information and building seismic vulnerability were estimated based on the EMS-98 standard and the SVM. Finally, vulnerability proxies established based on the RISK-UE model were applied to the Lixia database, and the vulnerability and risk assessment under different seismic intensities were estimated with the experimental accuracy verified. The results showed that the SVM method can obtain stable and accurate results in urban scale vulnerability assessment. The mean building vulnerability index in Lixia District is 0.43, which indicates that the overall seismic performance is good. Most of the area falls within the seismic intensity range of VII-X degrees and would experience slight to moderate damage. The results of the study contribute to enhancing the precision and efficacy of large-scale seismic risk assessment, and they can be used by relevant departments to create tailored emergency plans and reduce seismic hazard losses. Additionally, these results can aid in achieving the climate action goal (SDG13) of the United Nations Sustainable Development Goals (SDGs).

Abstract Chorus subpackets/subelements are the wave packets occurring at intervals of ∼10–100 msec and are suggested to play a crucial role in the formation of substructures within pulsating aurora. In this study, we investigate the evolution of subpackets from the upstream to downstream regions. Using Van Allen Probe A measurements, we have found that the frequency of the upstream subpackets increases smoothly, but that of the downstream subpackets remains almost unchanged. Through a simulation in the real‐size magnetosphere, we have reproduced the subpackets with characteristics similar to those in observations, and revealed that the frequency chirping is influenced by both resonant current of electrons and wave amplitude due to nonlinear physics. Although the resonant currents in the upstream and downstream regions are comparable, the wave amplitude increases significantly during evolution, resulting in lower sweep rate in the downstream region. Our findings provide a fresh insight into the evolution of chorus subpackets.

The purpose of the infrared and visible image fusion is to generate a fused image with rich information. Although most fusion methods can achieve good performance, there are still shortcomings in extracting feature information from source images, which make it difficult to balance the thermal radiation region information and texture detail information in the fused image. To address the above issues, an expectation maximization (EM) learning framework based on adversarial generative networks (GAN) for infrared and visible image fusion is proposed. The EM algorithm (EMA) can obtain maximum likelihood estimation for problems with potential variables, which is helpful in solving the problem of lack of labels in infrared and visible image fusion. The axial‐corner attention mechanism is designed to capture long‐range semantic information and texture information of the visible image. The multifrequency attention mechanism digs the relationships between features at different scales to highlight target information of infrared images in the fused result. Meanwhile, two discriminators are used to balance two different features, and a new loss function is designed to maximize the likelihood estimate of the data with soft class label assignments, which is obtained from the expectation network. Extensive experiments demonstrate the superiority of EMA‐GAN over the state‐of‐the‐art.

Knee osteoarthritis (KOA) is a common chronic disease in orthopedics, which brings great pain to patients' life and spirit. Therefore, it is necessary to elucidate the pathogenesis of KOA. The pathophysiology of KOA has been linked to numerous factors, including oxidative stress, apoptosis, cellular senescence, mitochondrial dysfunction, and inflammatory factors. Cellular senescence has grown in importance as a topic of study for age-related illnesses recently. KOA has also been discovered to be closely related to human aging, a process in which chondrocyte senescence may be crucial. Numerous researches have looked at the pathogenesis of KOA from the perspectives of mechanical stress abnormalities, oxidative stress, inflammatory overexpression, and mitochondrial dysfunction. Many studies have discovered that the primary pathogenesis of KOA is inflammatory overexpression and chondrocyte death brought on by an imbalance in the joint microenvironment. And abnormal mechanical stress is the initiating cause of oxidative stress, inflammation, and mitochondrial disorders. However, few findings have been reported in the literature on the relationship between these factors, especially for mechanical stress abnormalities, and chondrocyte senescence. This time, in order to better understand the pathogenesis of KOA and identify potential connections between chondrocyte senescence and these microenvironments in KOA, as well as oxidative stress, inflammatory overexpression, and mitochondrial dysfunction microenvironmental dysfunctions, we will use chondrocyte senescence as a starting point. This will allow us to develop new therapeutic approaches for KOA.
