LIN28B plays a critical role in the Warburg effect. However, its underlying mechanism remains elusive. Recently, it has been reported that LIN28B could collaborate with IGF2BP3, which can bind to m6A-modified c-MYC transcripts. Therefore, this study investigated if LIN28B recognises methylated c-MYC mRNA to promote the Warburg effect in gastric cancer.Effects of LIN28B on gastric cancer were confirmed in vitro and in vivo. On the basis of bioinformatics analysis, the association between LIN28B and c-MYC mRNA was shown using RNA immunoprecipitation (RIP) and luciferase reporter assays. The role of m6A was identified by RNA pull-down assays. We further performed RIP-seq to search for long non-coding RNAs (lncRNAs) participating in the LIN28B binding process. Chromatin immunoprecipitation was used to show the impact of c-MYC on transcription of LIN28B and lncRNAs.LIN28B was identified to stabilize c-MYC mRNA by recognizing m6A. Furthermore, the interaction between c-MYC mRNA and LIN28B is speculated to be supported by LOC101929709, which binds to both LIN28B and IGF2BP3. Functional experiments revealed that LOC101929709 promotes the proliferation, migration and glycolysis of gastric cancer. Mechanistically, LOC101929709 enriched in the cytoplasm helps LIN28B stabilize c-MYC mRNA. Moreover, c-MYC promoted the transcription of both LOC101929709 and LIN28B. Additionally, LOC101929709 also activated the PI3K/AKT pathway.The c-MYC/LOC101929709/LIN28B axis promotes aerobic glycolysis and tumour progression. Thus, LOC101929709 can be a novel potential target for gastric cancer treatment.

In the present study, the conditions for Azotobacter chroococcum fermentation using Agaricus bisporus wastewater as the culture medium were optimized. We analyzed the total number of living A. chroococcum in the fermentation broth, using multispectral imaging flow cytometry. Single-factor experiments were carried out, where a Plackett-Burman design was used to screen out three factors from the original six processing factors wastewater solubility, initial pH, inoculum size, liquid volume, culture temperature, and rotation speed that affected the total number of viable A. chroococcum. The Box-Behnken response surface method was used to optimize the interactions between the three main factors and to predict the optimal fermentation conditions. Factors significantly affecting the total number of viable A. chroococcum, including rotation speed, wastewater solubility, and culture temperature, were investigated. The optimum conditions for A. chroococcum fermentation in A. bisporus wastewater were a rotation speed of 200 rpm, a solubility of 0.25%, a culture temperature of 26°C, an initial pH of 6.8, a 5% inoculation volume, a culture time of 48 h, and a liquid volume of 120 mL in a 250 mL flask. Under these conditions, the concentration of total viable bacteria reached 4.29 ± 0.02 ✕ 10

The aim of this study is to explore the active components of Anoectochilus roxburghii capable of inhibiting melanin formation using chemical separation and extraction and functional analysis. Anoectochilus roxburghii were extracted with alcohol and separated into three groups: the total extraction group, alcohol extracted group and alcohol precipitated group. Zebrafish embryos at 0.75 h post-fertilization were exposed to various concentrations of the three groups of extracts, and analyzed at 72 h, using semi-quantitative RT-PCR and in situ hybridization. The results showed that the alcohol extracts inhibit melanogenesis most significantly in the zebrafish embryos. The mRNAs of melanin-related genes, such as silv, tyr, tyrp1a, were down-regulated by the alcohol extracts spatially and temporally. The alcohol extracts also inhibited the activity of tyrosinase, a key enzyme in melanogenesis, in a dosage dependent manner. In addition, the alcohol extracts also display a remarkable inhibitory effect on melanin synthesis through down-regulation of mRNAs of melanin-related genes and tyrosinase activity in zebrafish embryos, in which a large amount of melanin has already been synthesized. Such inhibitory effect could be reversed after the withdrawal of the alcohol extracts. Our results showed that the alcohol extracts of Anoectochilus roxburghii can significantly inhibit zebrafish melanogenesis, supporting the notion that Anoectochilus roxburghii could potentially be used in the development and production of natural whitening products.

A highly nonlinear, dispersion flattened hybrid nanofiber with a silicon/silica cross-slot-structure nanocore is proposed and analyzed, which is insensitive to polarization for implementing quasi-TE and quasi-TM fundamental modes transmission due to the cross slot effect. Simulation results show that fundamental mode with ultra-small mode effective areas and high nonlinearity at TE and TM polarizations, which are confined in the narrow cross slot by four silicon ribs, can be achieved via this cross-slot-structure core. Moreover, the cladding of four large-air-holes contributes to the tailoring of the group velocity dispersion (GVD) and further enhancment of the nonlinearity. Our results indicate that ultra-small Aeff of 0.098μm2 and flat anomalous GVD with less than 13.5 ps.km-1.nm-1 dispersion ripple at C-band are realizable.

Krüppel-like factors (KLFs), such as KLF4, KLF2, KLF5 and KLF15, have been extensively investigated in multi-cancers. However, KLF16, a member of KLFs, hasn’t been well identified in cancer, especially in gastric cancer (GC). Here, we investigated the roles of KLF16 in GC. In present study, we found that KLF16 expression levels were significantly up-regulated in GC tissues compared to adjacent normal tissues both in protein and mRNA levels by using immunohistochemistry assays (IHC) and real-time quantitative PCR (qPCR). And KLF16 expression levels were positively correlated to tumor size, invasion depth, lymphatic metastasis and TNM stage. Furthermore, KLF16 expression also could predict prognosis in patients with GC. Moreover, the knock-down of KLF16 could significantly suppress proliferation via increasing p21 expression and decreasing CDK4 expression in GC cell lines. In summary, these findings demonstrate that KLF16 plays a significant role in GC progression and could be a new therapeutic target for GC patients.

Taking the air intake pipe in the engine as the target of the research, the software STAR-CDE is adopted to build a three-dimensional simulation model for the air intake pipe in the engine T12V190 with the aim to solve the problems of air input deficiency, Combustion deterioration and performance reduction of one cylinder caused by the non-uniformity. Moreover, the non-uniformity of the flux of air intake of the air intake pipe is mainly studied and analyzed through a calculation on the CFD of the inner flow field of the three dimension of the air intake pipe in the engine T12V190. In addition, a retrofit design with multiple schemes is made based on the cavity structure of the original mold for the air intake pipe. Finally, through a comparison among the three selected designs, a more feasible retrofit designing scheme and a designing thought on the air intake pipe in the engine with directional significance are got.

Aimed at the problems of low control accuracy and weak robustness influenced by external disturbance, friction, load changes, modeling errors and other issues in ammunition auto-loading manipulator control system, a new robust adaptive controller based on fuzzy compensation is proposed. The control architecture employs fuzzy systems to compensate adaptively for plant uncertainties to distinguish different disturbance compensation terms and approximate each of them respectively. As a result, the control system has great dynamic features and robust stability and meet the requirement that the actual motion of ammunition auto-loading manipulator quickly tracks the scheduled trajectory.

A new fuzzy adaptive sliding mode controller based on fuzzy compensation for robot is proposed. The control architecture employs fuzzy systems to compensate adaptively for plant uncertainties to distinguish different disturbance compensation terms and approximate each of them respectively. By analyzing and comparing the simulation results, it is obviously shown that the control system can lighten the effect on the control system caused by different disturbance factors and eliminate the system chattering than that of traditional SMC.

To obtain the approximately analytical solution of double-walled carbon nanotubes (DWNTs) nonlinear vibration. In this study, homotopy perturbation method (HPM) was used to solve nonlinear vibration equation of DWNTs. Novel and accurate analytical solutions for the frequency and displacement are derived. Comparison of the result obtained by the HPM with exact solutions reveals that only the first or second order approximation of the HPM leads to higher accurate solution.

In order to meet the demands of practical, convenient and quick charge requirements, a mathematical model of a certain type of vehicle starting lead-acid battery is established. Using the method of circuit analysis, the model parameters are identified by the known test data. In addition, battery charge model is simulated in each charge stage using the intelligent three-stage charge method, simulation waveform and test waveform fit very well, absolute errors between them reach to 10-6. Simulation results show that the mathematical model and its analysis method is proper for the charge characteristics of vehicle starting lead-acid battery. This has a good guidance to design intelligent charger and extend the battery life.

In the DC power system of the unwatched transformer substation, the battery charger is one of the key devices. The stability of its performances directly affect the stability of DC power system. In the paper, using the computer technologies and the real-time Ethernet technology to communicate with the battery charger monitor to achieve remote monitoring technology of the performances of the battery charger. It can not only remote monitor the operations and fault states of the battery charger on line, but also remote sensing the ripple factor and the stabilized voltage precision when the battery charger is under the states of the floating charge and the constant voltage equalizing charge, at the same time remote sensing the stabilized current precisions when the battery charger is under the stage of constant current equalizing charge, and the characteristics of equalizing current when the battery charger is in operation. So the remote monitoring technology of the performances of the battery charger is very important in the transformer substation.

Gunpowder was released in an instant when the pill fly out of the shell during the firing, and then formed a complicated flow fields about the muzzle when the gas expanded sharply. Using the 2 d axisymmetric Navier-Stokes equation combined with single equation turbulent model to conduct the numerical simulation of the process of gunpowder gass evacuating out of the shell without muzzle regardless of the pill’s movement. The numerical simulation result was identical with the experimental. Then simulated the evacuating process of gunpowder gass of an artillery with muzzle brake. The result showed complicated wave structure of the flow fields with the muzzle brake and analysed the influence of muzzle brake to the gass flow field distribution.

In this paper, using current advanced computer and real-time Ethernet technologies to design a set of high performances’ DC power dynamic management system for digitalized transformer substation. In the system, the operation states and fault conditions of DC power system can be remote monitored and controlled, the state informations of the feeder switches are real time displayed, the state transformations of DC generatrices are remote controlled, the states of equalizing charge and floating charge of the battery charger are remote controlled and remote transformed, the parameters of the battery charger are remote set, etc. In addition, the operation modes of the whole DC power system in the transformer substation are shown by way of simulation map installed in the upper management machine. This dynamic management system greatly contribute to the construction of unwatched, intelligentized, digitalized transformer substation.
