BackgroundPorcine deltacoronavirus (PDCoV), a novel swine enteropathogenic coronavirus, challenges the global swine industry. Currently, there are no approaches preventing swine from PDCoV infection.MethodsA new PDCoV strain named JS2211 was isolated. Next, the dimer receptor binding domain of PDCoV spike protein (RBD-dimer) was expressed using the prokaryotic expression system, and a novel nanoparticle containing RBD-dimer and ferritin (SC-Fe) was constructed using the SpyTag/SpyCatcher system. Finally, the immunoprotection of RBD-Fe nanoparticles was evaluated in mice.ResultsThe novel PDCoV strain was located in the clade of the late Chinese isolate strains and close to the United States strains. The RBD-Fe nanoparticles were successfully established. Immune responses of the homologous prime-boost regime showed that RBD-Fe nanoparticles efficiently elicited specific humoral and cellular immune responses in mice. Notably, high level PDCoV RBD-specific IgG and neutralizing antibody (NA) could be detected, and the histopathological results showed that PDCoV infection was dramatically reduced in mice immunized with RBD-Fe nanoparticles.ConclusionThis study effectively developed a candidate nanoparticle with receptor binding domain of PDCoV spike protein that offers protection against PDCoV infection in mice.

Spectral technology is a scientific method used to study and analyze substances. In recent years, the role of spectral technology in the non-destructive testing (NDT) of fruits has become increasingly important, and it is expected that its application in the NDT of fruits will be promoted in the coming years. However, there are still challenges in terms of dataset collection methods. This article aims to enhance the effectiveness of spectral technology in NDT of citrus and other fruits and to apply this technology in orchard environments. Firstly, the principles of spectral imaging systems and chemometric methods in spectral analysis are summarized. In addition, while collecting fruit samples, selecting an experimental environment is crucial for the study of maturity classification and pest detection. Subsequently, this article elaborates on the methods for selecting regions of interest (ROIs) for fruits in this field, considering both quantitative and qualitative perspectives. Finally, the impact of sample size and feature size selection on the experimental process is discussed, and the advantages and limitations of the current research are analyzed. Therefore, future research should focus on addressing the challenges of spectroscopy techniques in the non-destructive inspection of citrus and other fruits to improve the accuracy and stability of the inspection process. At the same time, achieving the collection of spectral data of citrus samples in orchard environments, efficiently selecting regions of interest, scientifically selecting sample and feature quantities, and optimizing the entire dataset collection process are critical future research directions. Such efforts will help to improve the application efficiency of spectral technology in the fruit industry and provide broad opportunities for further research.

The application of beneficial microbial consortium can effectively improve plant disease resistance and its growth. Various fungi were compounded with Bacillus velezensis LJ02 and applied to watermelon plants in this paper. The results showed that the microbial consortium T2 (compounded Bacillus velezensis LJ02 with Aspergillus aculeatus 9) can effectively control gummy stem blight and powdery mildew in watermelon, while the control effect reached 83.56% and 70.93%, respectively (p < 0.05). Compound treatment improved the diversity and richness of the rhizosphere microbial community structure, and the relative abundance of Caulobacterales and Xanthomonadaceae significantly increased after applying T2 to the soil. Meanwhile, the internode length was significantly decreased 28% (p < 0.05), and the maximum leaf length increased 10.33% (p < 0.05). In addition, the microbial consortium delays the maturity of watermelon vegetables. By studying the effects of microbial consortium on watermelon seedlings, our study provides a theoretical basis for the popularization and application of the compound inoculant.

Abstract Table grapes are susceptible to external pathogens during postharvest storage. The resulting continuous oxidative stress causes damage and aging, thereby reducing the defense against disease. In this study, the effect of biocontrol yeast T‐2 on the storage performance of grapes was evaluated. After T‐2 treatment, the grapefruits rot rate and lesion diameter caused by Botrytis cinerea (B. cinerea) were significantly decreased at 2–5 days after inoculation (DAI). Additionally, the browning rate and shedding rate of grapefruit during storage were significantly reduced at 2–5 DAI, and the weight loss rate was significantly reduced at 3–5 DAI. The decreased malondialdehyde (MDA) content in grapefruits at 1–5 DAI with T‐2 indicated a reduction in oxidative damage. Furthermore, the activities of antioxidant enzymes such as peroxidase (POD), catalase (CAT), phenylalanin ammonia‐lyase (PAL) were significantly increased during most storage time after being treated with T‐2. Moreover, the contents of total phenolics and flavonoids and the expression levels of key enzyme genes in metabolic pathways were increased after T‐2 treatment during most postharvest storage time, providing evidence that T‐2 changed the biological process of phenolic flavonoid metabolism. The increase in enzymatic and nonenzymatic antioxidants after treatment with T‐2 reflected the strengthening of the antioxidant system, hence postponing fruit senescence and promoting storage performance under the stress of B. cinerea.

Alfalfa mosaic virus (AMV) is the most pervasive epidemic virus affecting alfalfa production. However, detailed investigations on the molecular population genetics and evolutionary dynamics of AMV are scarce. This study aimed to report on a large-scale long-term survey of genetic variability in AMV populations from China and perform a comparative analysis of AMV population genetics in the three most thoroughly studied countries to date: China, Iran, and Spain. The study was based on the analysis of the coat protein gene (cp) using two analytical approaches: an analysis of molecular variance (AMOVA) and a Bayesian Markov Chain Monte Carlo approach that investigates the association between geographical origin and phylogeny. Both analytical approaches found significant genetic differentiation within localities, but not among localities nor among provinces. This observation might result from inappropriate agronomical practices involving extensive exchange of plant materials followed by rapid viral diversification within localities. In the Chinese population, both methods found that genetic diversification in AMV was strongly associated with different bioclimatic zones. Rates of molecular evolution were similar in the three countries. The estimated epidemic exponential population size and growth rate suggest that the epidemics grew faster and with higher incidence in Iran, followed by Spain and China. Estimates of the time to the most recent common ancestors suggest that AMV was first seen in Spain by the beginning of the twentieth century and later on in eastern and central Eurasia. After ruling out the existence of recombination breakpoints within the cp gene, a codon-based selection analysis per population was performed and identified many codons under significant negative selection and a few under significant positive selection; the latter varied among countries, suggesting regional differences in selective pressures.

Group A rotaviruses (RVAs) are major etiologic agents of gastroenteritis in infants and young children worldwide. To study the prevalence and genetic characteristics of RVAs, a hospital-based surveillance study was conducted in Wuhan, China from June 2019 through May 2022. The detection rates of RVAs were 19.40% (142/732) and 3.51% (8/228) in children and adults, respectively. G9P[8] was the predominant genotype, followed by G8P[8] and G3P[8]. G8P[8] emerged and was dominant in the 2021–2022 epidemic season. The genome constellation of six G8P[8] strains was assigned to G8-P[8]-I2-R2-C2-M2-A2-N2-T2-E2-H2. Phylogenetic analysis revealed that the VP7, VP4, VP2, VP3, NSP1, NSP2, NSP3, and NSP5 genes of these G8P[8] strains clustered closely with those of the G8P[8] strains in Asia and were distant from those of the P[8] and G2P[4] strains simultaneously detected in Wuhan. In contrast, the VP1, VP6, and NSP4 genes were closely related to the typical G2P[4] rotavirus, including those of G2P[4] strains simultaneously detected in Wuhan. The detection rate of RVAs decreased in the COVID-19 pandemic era. It was deduced that the G8P[8] rotaviruses that emerged in China may be reassortants, carrying the VP6, VP1, and NSP4 genes derived from the G2P[4] rotavirus in the backbone of the neighboring DS-1-like G8P[8] strains represented by CAU17L-103.

The loss of tomatoes caused by Botrytis cinerea (B. cinerea) is one of the crucial issues restricting the tomato yield. This study screened the elicitor protein phosphopentomutase from Bacillus velezensis LJ02 (BvEP) which improves the tomato resistance to B. cinerea. Phosphatemutase was reported to play a crucial role in the nucleoside synthesis of various microorganisms. However, there is no report on improving plant resistance by phosphopentomutase, and the related signaling pathway in the immune response has not been elucidated. High purity recombinant BvEP protein have no direct inhibitory effect on B. cinerea in vitro,and but induce the hypersensitivity response (HR) in Nicotiana tabacum. Tomato leaves overexpressing BvEP were found to be significantly more resistant to B. cinerea by Agrobacterium-mediated genetic transformation. Several defense genes, including WRKY28 and PTI5 of PAMP-triggered immunity (PTI), UDP and UDP1 of effector-triggered immunity (ETI), Hin1 and HSR203J of HR, PR1a of systemic acquired resistance (SAR) and the SAR related gene NPR1 were all up-regulated in transgenic tomato leaves overexpressing BvEP. In addition, it was found that transient overexpression of BvEP reduced the rotting rate and lesion diameter of tomato fruits caused by B. cinerea, and increased the expression of PTI, ETI, SAR-related genes, ROS content, SOD and POD activities in tomato fruits, while there was no significant effect on the weight loss and TSS, TA and Vc contents of tomato fruits. This study provides new insights into innovative breeding of tomato disease resistance and has great significance for loss reduction and income enhancement in the tomato industry.

Subtilisin, a serine protease, can trigger defense responses in a wide variety of plants, both locally and systemically, to protect against pathogens. However, key residues of subtilisin to improve resistance to plant diseases remain unknown. In this study, Nicotiana benthamiana (N. benthamiana) leaves expressing subtilisin from Bacillus velezensis LJ02 were shown to improve protection against Botrytis cinerea (B. cinerea). Furthermore, the underlying mechanism that LJ02 subtilisin improved the protective effect was explored, and the direct inhibitory effect of subtilisin on B. cinerea was excluded in vitro. Subsequently, reactive oxygen species (ROS) burst and upregulation of resistance-related genes in systemic leaves of N. benthamiana further verified that subtilisin could induce systemic protection against B. cinerea. G307A/T308A and S213A/L214A/G215A subtilisin significantly reduced the ability to resist B. cinerea infection in N. benthamiana. Furthermore, the ROS content and expression levels of resistance-related genes of both mutants were significantly decreased compared with that of wild-type subtilisin. This work identified key residues essential for the activation function of subtilisin plant immunity and was crucial in inducing plant defense responses against B. cinerea.

This paper is a case study on a thermal protection system in a combustion chamber of a ramjet. A multilayer passive thermal protection system (MPTPS) with carbon-phenolic material was established, and the temperature distribution and insulation performance of MPTPS were studied. The problem is that the carbon-phenolic material is pyrolyzed when subjected to high temperatures, leading to the changes in its thermal properties (e.g., density, specific heat capacity, and thermal conductivity.). Moreover, the carbon-phenolic material absorbs heat during the pyrolysis procedure of phenolic resin. Therefore, in this paper, several experimental studies were conducted on carbon-phenolic composites to investigate its pyrolysis characteristics, including tube furnace heating experiment and Py-GC-MS experiment. On the other hand, a one-dimensional multilayer transient heat conduction approach was developed, many calculations were numerically performed to study the effects of negative heat sources and the thickness of carbon-phenolic composites on the thermal insulation performance of MPTPS. As a result, the carbon-phenolic material was pyrolyzed from 200 °C, and the mass-loss rate was 25% when the heating temperature increased up to 900 °C; the negative heat source delayed the temperature rising point and decreased the outer wall temperature; the thickness of the carbon-phenolic composites affected the final outer wall mainly by conduction.

Staphylococcus aureus has been recognized as an important human pathogen and poses a serious health threat worldwide. With the advent of antibiotic resistance, such as the increased number of methicillin-resistant Staphylococcus aureus (MRSA), there is an urgent need to develop new therapeutical agents. In this study, Chinese traditional medicine Tanreqing (TRQ) has been used as an alternative treating agent against MRSA and we aim to unravel the mode of action of TRQ underlying MRSA inhibition. TRQ treatment affected numerous gene expression as revealed by RNA-seq analysis. Meanwhile, TRQ targeted cell division to inhibit cell growth as shown by illumination microscopy. Besides, we confirmed that TRQ downregulates the expression of virulence factors such as hemolysin and autolysin. Finally, we used a murine model to demonstrate that TRQ efficiently reduces bacterial virulence. Altogether, we have proved TRQ formula to be an effective agent against S. aureus infections.

As a complementary analysis tool to conventional high-field nuclear magnetic resonance (NMR), zero- to ultralow-field (ZULF) NMR detects nuclear magnetization signals in the sub-microtesla regime. Spin-exchange relaxation-free (SERF) atomic magnetometers provide a new generation of sensitive detectors for ZULF NMR. Owing to features such as low cost, high resolution, and portability, ZULF NMR has recently attracted considerable attention in chemistry, biology, medicine, and tests of fundamental physics. This review describes the basic principles, methodology, and recent experimental and theoretical development of ZULF NMR as well as its applications in spectroscopy, quantum control, imaging, NMR-based quantum devices, and tests of fundamental physics. The future prospects of ZULF NMR are also discussed.

Surface flattening plays an important role in the whole process of garment design. We proposed a novel method by using three-dimensional triangle mesh flattening in this study. First, the three-dimensional triangle mesh is flattened to a two-dimensional plane to approximate the original surface. The initial flattening results are then used as preliminary guesses for subsequent optimizations. Considering that the deformation energy in the real woven fabric is related to tensile or shear deformation, a simplified fabric deformation model based on energy is proposed to update the energy distribution to determine the best two-dimensional pattern. An innovative unified axis system process is proposed to obtain the deformation energy, and energy relaxation in local flattening is proposed to release the distortion of flattening. Finally, the experimental results show that complex surfaces such as garments could achieve better flattening results. Compared with other energy-based methods in garment design, our proposed methods are more flexible and practical.

Glycosaminoglycan from Apostichopus japonicus (AHG) and its depolymerized fragments (DAHGs) are anticoagulant fucosylated chondroitin sulfate. The aim of this study was to further evaluate the anticoagulant and antithrombic activity of AHG and DAHGs, as well as reveal the dynamic relationship between exposure and effect in vivo. The results demonstrated that AHG100 (Mw~100 kDa), DAHG50 (Mw~50 kDa), and DAHG10 (Mw~10 kDa) exhibited potent anticoagulant activity by inhibiting intrinsic factor Xase complex (FXase) as well as antithrombin-dependent factor IIa (FIIa) and factor Xa (FXa). These glycosaminoglycans markedly prevented thrombosis formation and thrombin-induced platelet aggregation in a dose- and molecular weight-dependent manner in vitro and in vivo. The further bleeding time measurement indicated that DAHG10 exhibited obviously lower hemorrhage risks than native AHG100. Following oral administration, DAHG10 could be absorbed into blood, further dose-dependently prolonging activated partial thromboplastin time (APTT) and thrombin time (TT) as well as inhibiting FXa and FIIa partially through FXase. Anticoagulant activity was positively associated with plasma concentration following oral administration of DAHG10. Our study proposed a new point of view to understand the correlation between effects and exposure of fucosylated chondroitin sulfate as an effective and safe oral antithrombotic agent.

Aim of study: Quercus variabilis is a sclerophyllous oak with strong resprouting capabilities and whose regeneration is facilitated by the development of stump shoots following disturbance. During secondary forest regeneration, fine roots are important organs relative to changes in stand characteristics. Here, we aimed to provide novel insights into the chemical composition variations in roots with seasonality and root order hierarchy in a Q. variabilis forest at different periods of regeneration. Area of study: The forest is located next to the Baxianshan National Reserve in the southern part of the Yanshan Mountains, Tianjin, China. Materials and methods: Six plots were established in stands with either eight or 40 years of regeneration for the repeated sampling of fine roots during the growing season of 2019. All roots were classified by branch order. The first three root orders were collected to analyse the concentrations of nonstructural carbohydrate, carbon, and nitrogen. Main results: Short-term regeneration stands showed a reduction in soil moisture and an increase in soil temperature because of the lower canopy cover, compared to long-term stands. Soluble sugar and starch were lower in roots of short-term stands than in those of long-term stands, and the decreasing ratio of both parameters was observed in short-term stands. Less carbon and greater nitrogen concentrations of fine roots were found in short-term stands than in long-term stands, which resulted in weaker C/N ratio values. Nonstructural carbohydrate was stored more in higher order roots than terminal roots and presented greater sensitivity to forest regeneration. Redundancy discriminate analysis demonstrated that the nonstructural carbohydrate concentrations in roots were affected positively by canopy cover and negatively by soil temperature. Research highlights: The seasonal dynamics and branch allocation of chemical reserves in fine roots varied in the different periods of forest regeneration because of the discrepancy between the canopy cover and soil traits. Less nonstructural carbohydrate and a lower C/N ratio at the onset of forest regeneration may elevate the risk of root death. This work aimed to optimize the washcoat slurry for hydrotalcite-based lean NOx trap (LNT) catalyst. The effects of the slurry properties including pH, solid content, binder and additive on the hydrotalcite-based slurry viscosity were investigated. The particle size distribution of the optimal hydrotalcite-based slurry was measured. A cordierite material was used to coat the optimal slurry, and the washcoat was characterized by X-ray diffraction, scanning electron microscopy and N2 adsorption. The optimal slurry containing Pt and Ba was coated on the cordierite for the preparation of hydrotalcite-based LNT catalyst, and the performances of this catalyst were evaluated by NOx storage test, temperature programmed desorption and NOx reduction. For comparison, the performance of the commercial LNT catalyst with Pt/BaO/Al2O3 was analyzed. After coating, the hydrotalcite-based washcoat was closely contacted with the support, being the main phase MgO and presenting a specific surface area of 86.3 m2/g. The hydrotalcite-based LNT catalyst had better NOx storage and desorption ability, selectivity to N2 and LNT efficiency than the Pt/BaO/Al2O3 catalyst.

We previously reported the development of a biosensor platform that is capable of measuring biometabolites and environmental sensitive species, such as peroxide and nitrate/nitrate, to concentrations in the order of ppb (parts per billion) or lower. In this investigation, we modified our platform with dendrimers to enhance its performance. Zero and second generation of dendrimers were coated on the surface of a carbon glassy electrode which was then modified with l-glutamate dehydrogenase (GDH) and α-keto glutarate. The resulting electrode was tested with ammonium solutions, concentrations ranged from 2 to 300 nM at pH 7.4; the results were satisfactory. Measurements at lower concentrations had better resolution than at higher concentrations and it is believed that the measurement limit can be lower than 2 nM. This biosensor platform was proven to be versatile and can be employed as a platform for ultrasensitive detecting devices in many biomedical and environmental applications.

In this study, the immunomodulatory activities of Apostichopus japonicus glycosaminoglycan (AHG) on the nature killer (NK) cells, cytotoxic T lymphocytes (CTLs) and cyclophosphamide (CY)-treated mice were investigated. After stimulation with multiple concentrations of AHG (0–100 μg/mL), NK cells and CTLs displayed outperformance against YAC-1 and B16 cells, respectively. Furthermore, the mitogen-induced splenic lymphocyte proliferation in CY-induced immunosuppressed mice was significantly promoted by AHG. In addition, the administration of AHG dramatically increased the splenocytes Ca2+ concentration and the delayed-type hypersensitivity (DTH) reaction in a dose-dependent manner. Besides, AHG could strongly increase the total antioxidant capacity (T-AOC), the activities of superoxidase dismutase (SOD), catalase (CAT) as well as glutathione peroxidase (GSH-PX), and could decrease the malondialdehyde (MDA) level in the heart, kidney and liver. These findings indicated that AHG played an important role in the immune enhancement and protection against CY-induced immunosuppression and oxidative damage. Our findings provide experimental evidence for further research and possible immunostimulatory applications of AHG in clinical practice.

The present study is focused on the intestinal absorption of sea cucumber saponins. We determined the pharmacokinetic characteristics and bioavailability of Echinoside A and Holotoxin A1; the findings indicated that the bioavailability of Holotoxin A1 was lower than Echinoside A. We inferred that the differences in chemical structure between compounds was a factor that explained their different characteristics of transport across the intestine. In order to confirm the absorption characteristics of Echinoside A and Holotoxin A1, we examined their transport across Caco-2 cell monolayer and effective permeability by single-pass intestinal perfusion. The results of Caco-2 cell model indicate that Echinoside A is transported by passive diffusion, and not influenced by the exocytosis of P-glycoprotein (P-gp, expressed in the apical side of Caco-2 monolayers as the classic inhibitor). The intestinal perfusion also demonstrated well the absorption of Echinoside A and poor absorption of Holotoxin A1, which matched up with the result of the Caco-2 cell model. The results demonstrated our conjecture and provides fundamental information on the relationship between the chemical structure of these sea cucumber saponins and their absorption characteristics, and we believe that our findings build a foundation for the further metabolism study of sea cucumber saponins and contribute to the further clinical research of saponins.

Quantum amplification enables the enhancement of weak signals and is of great importance for precision measurements, such as biomedical science and tests of fundamental symmetries. Here, we observe a previously unexplored magnetic amplification using dark noble-gas nuclear spins in the absence of pump light. Such dark spins exhibit remarkable coherence lasting up to 6 min and the resilience against the perturbations caused by overlapping alkali-metal gas. We demonstrate that the observed phenomenon, referred to as "dark spin amplification," significantly magnifies magnetic field signals by at least three orders of magnitude. As an immediate application, we showcase an ultrasensitive magnetometer capable of measuring subfemtotesla fields in a single 500-s measurement. Our approach is generic and can be applied to a wide range of noble-gas isotopes, and we discuss promising optimizations that could further improve the current signal amplification up to 104 with 21Ne, 105 with 129Xe, and 106 with 3He. This work unlocks opportunities in precision measurements, including searches for ultralight dark matter with sensitivity well beyond the supernova-observation constraints. [ABSTRACT FROM AUTHOR]
