Abstract Albeit extensive studies of translation universals at lexical and grammatical levels, there has been scant research at the syntactic-semantic level. To bridge this gap, this study employs semantic role labeling and textual entailment analysis to compare Chinese translations with English source texts and non-translated Chinese original texts. The research has found substantial evidence for translation universals like explicitation, simplification, and levelling out at the syntactic-semantic level, which is illustrated by significant differences between syntactic-semantic features of Chinese translations and those of English source texts and Chinese original texts. This suggests a distinct syntactic-semantic uniqueness of Chinese translations, wherein the overall features exhibit an “eclectic” characteristic, showcasing contrasting outcomes such as explicitation identified as S-universal and implicitation deemed T-universal. This could be attributed to the gravitational pull from the two language systems. In the inspection of specific semantic roles, features of agents and discourse markers are found to be evidence for both S-explicitation and T-explicitation, potentially reflecting the role of socio-cultural factors in shaping the uniqueness of syntactic-semantic features of Chinese translations. These findings further underscore the complexity inherent in translation, highlighting its function as a dynamic balance system.

Abstract Objective Brachytherapy has been indicated as an alternative option for treating cystic craniopharyngiomas (CPs). The potential benefits of brachytherapy for CPs have not yet been clarified. The purpose of this work was to conduct a meta-analysis to analyze the long-term efficacy and adverse reactions profile of brachytherapy for CPs. Materials and methods The relevant databases were searched to collect the clinical trials on brachytherapy in patients with CPs. Included studies were limited to publications in full manuscript form with at least 5-year median follow-up, and adequate reporting of treatment outcomes and adverse reactions data. Stata 12.0 was used for data analysis. Results According to the inclusion and exclusion criteria, a total of 6 clinical trials involving 266 patients with CPs were included in this meta-analysis. The minimum average follow-up was 5 years. The results of the meta-analysis showed that 1-year, 2–3 years and 5 years progression free survival rates (PFS) are 75% (95%CI: 66-84%), 62% (95%CI: 52-72%) and 57% (95%CI: 22-92%), respectively. At the last follow-up, less than 16% of patients with visual outcomes worser than baseline in all included studies. While, for endocrine outcomes, less than 32% of patients worser than baseline level. Conclusion In general, based on the above results, brachytherapy should be considered as a good choice for the treatment of CP.

Abstract Extensive studies have described the linguistic features of human translations and verified the existence of the simplification translation universal. However, little has been known about the linguistic features of machine translations, although machine translation, as a unique modality of translation, has become an integral part of translation practice. This study is intended to test whether the simplification translation universal observed in human translations also holds true for machine translations. If so, are simplification features in machine translations different significantly from those in human translations? And does genre significantly affect simplification features? To this end, we built a balanced comparable corpus containing three text varieties, i.e., machine translations, human translations and target-language originals across three genres namely contemporary novels, government documents and academic abstracts. Based on the corpus, we conducted a systematic comparison of lexical diversity, as a proxy for simplification, of different text varieties. The results show that simplification is corroborated overall in both machine and human translations when compared with target-language originals, and machine translations are more simplified than human translations. Additionally, genre is found to exert a significant influence on the lexical diversity of different text varieties. This study is expected to expand the scope of corpus-based translation studies on the one hand and to offer insights into the improvement of machine translation systems on the other hand.

Abstract Background Definitive concurrent chemoradiotherapy (dCCRT) is the gold standard for the treatment of locally advanced esophageal squamous cell carcinoma (ESCC). However, the potential benefits of consolidation chemotherapy after dCCRT in patients with esophageal cancer remain debatable. Prospective randomized controlled trials comparing the outcomes of dCCRT with or without consolidation chemotherapy in patients with ESCC are lacking. In this study, we aim to generate evidence regarding consolidation chemotherapy efficacy in patients with locally advanced, inoperable ESCC. Methods This is a multicenter, prospective, open-label, phase-III randomized controlled trial comparing non-inferiority of dCCRT alone to consolidation chemotherapy following dCCRT. In total, 600 patients will be enrolled and randomly assigned in a 1:1 ratio to receive either consolidation chemotherapy after dCCRT (Arm A) or dCCRT alone (Arm B). Overall survival will be the primary endpoint, whereas progression-free survival, locoregional progression-free survival, distant metastasis-free survival, and treatment-related toxicity will be the secondary endpoints. Discussion This study aid in further understanding the effects of consolidation chemotherapy after dCCRT in patients with locally advanced, inoperable ESCC. Trial registration ChiCTR1800017646.

Abstract The present study revisits the unique item hypothesis (UIH) from the perspective of translation directionality in the Chinese–English(C–E) language pair. Phrasal verb (PV) is used as the linguistic feature to investigate whether UIH holds true in C–E translations and whether translation directionality plays a role in the representation of unique items, based on a self-built parallel corpus of Lu Xun’s short stories and their English translations done by two L1 and two L2 translators, and a reference corpus of BNC short stories as the non-translated reference. It is found PVs are significantly over-represented in C–E translated texts when compared with English non-translated texts, and this overrepresentation is mainly attributed to the remarkable use of PVs by L1 translators; and there is a significant difference in the use of PVs by translators of different directionality, while no significant difference is found within the same direction. Additionally, L2 translators tend to use a limited range of PVs and prefer transparent PVs to semi-transparent and opaque ones. The results falsify the UIH in general and suggest that UIH is a conditional translation tendency constrained by translation directionality, or UIH is directionality-dependent. Gravitational pull model is used to analyze and explain the divergence between different translation directions.

Tigecycline serves as a last-resort antimicrobial agent against severe infections caused by multidrug-resistant bacteria. Tet(X) and its numerous variants encoding flavin-dependent monooxygenase can confer resistance to tigecycline, with tet(X4) being the most prevalent variant. This study aims to investigate the prevalence and characterize tigecycline resistance gene tet(X) in E. coli isolates from various origins in Yangzhou, China, to provide insights into tet(X) dissemination in this region. In 2022, we tested the presence of tet(X) in 618 E. coli isolates collected from diverse sources, including patients, pig-related samples, chicken-related samples, and vegetables in Yangzhou, China. The antimicrobial susceptibility of tet(X)-positive E. coli isolates was conducted using the agar dilution method or the broth microdilution method. Whole genome sequencing was performed on tet(X)-positive strains using Illumina and Oxford Nanopore platforms. Four isolates from pig or pork samples carried tet(X4) and exhibited resistance to multiple antimicrobial agents, including tigecycline. They were classified as ST542, ST10, ST761, and ST48, respectively. The tet(X4) gene was located on IncFIA8-IncHI1/ST17 (n=2), IncFIA18-IncFIB(K)-IncX1 (n=1), and IncX1 (n=1) plasmids, respectively. These tet(X4)-carrying plasmids exhibited high similarity to other tet(X4)-bearing plasmids with the same incompatible types found in diverse sources in China. They shared related genetic environments of tet(X4) associated with ISCR2, as observed in the first identified tet(X4)-bearing plasmid p47EC. In conclusion, although a low prevalence (0.65%) of tet(X) in E. coli strains was observed in this study, the horizontal transfer of tet(X4) among E. coli isolates mediated by pandemic plasmids and the mobile element ISCR2 raises great concerns. Thus, heightened surveillance and immediate action are imperative to curb this clinically significant resistance gene and preserve the efficacy of tigecycline.

Abstract Photo‐rechargeable zinc‐ion batteries (PRZIBs) have attracted much attention in the field of energy storage due to their high safety and dexterity compared with currently integrated lithium‐ion batteries and solar cells. However, challenges remain toward their practical applications, originating from the unsatisfactory structural design of photocathodes, which results in low photoelectric conversion efficiency (PCE). Herein, a flexible MoS2/SnO2‐based photocathode is developed via constructing a sunflower‐shaped light‐trapping nanostructure with 3D hierarchical and self‐supporting properties, enabled by the hierarchical embellishment of MoS2 nanosheets and SnO2 quantum dots on carbon cloth (MoS2/SnO2 QDs@CC). This structural design provides a favorable pathway for the effective separation of photogenerated electron‐hole pairs and the efficient storage of Zn2+ on photocathodes. Consequently, the PRZIB assembled with MoS2/SnO2 QDs@CC delivers a desirable capacity of 366 mAh g−1 under a light intensity of 100 mW cm−2, and achieves an ultra‐high PCE of 2.7% at a current density of 0.125 mA cm−2. In practice, an integrated battery system consisting of four series‐connected quasi‐solid‐state PRZIBs is successfully applied as a wearable wristband of smartwatches, which opens a new door for the application of PRZIBs in next‐generation flexible energy storage devices.

Abstract Background mcr-1-positive Escherichia coli has emerged as a significant threat to human health, veterinary health, and food safety in recent years. After the prohibition of colistin as a feed additive in animal husbandry in China, a noticeable reduction in both colistin resistance and the prevalence of mcr-1 was observed in E. coli from animals and humans. Objectives To assess the prevalence of the colistin resistance gene mcr-1 and characterize its genetic context in E. coli strains derived from fecal and meat samples from food-producing animals in China. Methods A total of 1,353 fecal samples and 836 food samples were collected between 2019 and 2020 in China. E. coli isolates were identified using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry and their susceptibility to colistin were determined using the broth microdilution method. The colistin-resistant E. coli isolates were screened for the presence of mcr by PCR analysis and sequencing. The minimal inhibitory concentrations (MICs) of 15 antimicrobial agents against the mcr-1-positive strains were further tested using the agar dilution method, conjugation assays were performed, and whole genome sequencing was performed using Illumina HiSeq. Results In total, 1,403 E. coli strains were isolated. Thirteen isolates from chicken meat (n = 7), chickens (n = 3), and pigs (n = 3) were resistant to colistin with MIC values of 4 to 16 mg/L, and carried mcr-1. All mcr-1-positive strains, except for isolate AH20PE105, contained multiple resistance genes and exhibited multidrug-resistant phenotypes. They belonged to 10 sequence types (STs), including a novel ST (ST14521). mcr-1 was located on IncI2 (n = 9), IncX4 (n = 2), and IncHI2 (n = 2) plasmids, which were highly similar to other mcr-1-carrying plasmids sharing the same incompatibility type. Seven mcr-1-carrying plasmids could be successfully conjugally transferred to E. coli C600. Conclusions While the low prevalence of mcr-1 (0.93%) identified in this study may not immediately seem alarming, the very emergence of this gene merits attention given its implications for colistin resistance and public health. Hence, ongoing surveillance of mcr-1 in E. coli remains crucial.

The loss of feature information during scale propagation in the deep learning method usually causes a big misclassification rate for many complex urban scenes. For this reason, this article presents a new deep learning method, called “Feature combination and promotion network (FCPNet).” This method consists of an end-to-end feature learning layer for obtaining multiscale depth features of point clouds, an external feature combination module for obtaining more fine-grained point cloud features, and a mutiheaded separable self-attention module for learning connections between features to obtain more globally informative features. When compared with PointNet++, the proposed FCPNet improved OA, MIOU, F1-score, and Kappa in the NPM3D dataset by 1.75%, 17.02%, 2.13%, and 0.0263, respectively. When compared with KpConv, the proposed FCPNet improved OA, mIOU, F1-score, and Kappa in the NPM3D dataset by 0.36%, 12.11%, 0.77%, and 0.0085, respectively. Especially, the proposed FCPNet is able to classify the objects with fewer point cloud data, such as pedestrians and cars, whose OA can reach 88.04% and 96.42%, respectively. These experimental results demonstrated the proposed FCPNet has rescued much lost information that happened in the traditional PointNet++. In addition, the adaptability to point cloud density variations for the proposed method is verified as well. The results demonstrated that when the total density of point cloud data decreases from 731.3 to 52.2 ${\text{pcs/}}{{\text{m}}^{2}}$, the OA of classification with the proposed FCPNet method only decreases by 3.07%. This means that the proposed FCPNet method is capable of being adaptive to the point cloud density changes.

The Charpy impact toughness is a crucial mechanical parameter in the application of materials within the realm of engineering. This study represents the pioneering investigation into the influence of different building directions on the Charpy impact toughness of NiTi shape memory alloys (SMAs) produced via laser powder bed fusion (LPBF). Texture analysis reveals that C0 and C90 exhibit a pronounced // building direction (BA), while C45 demonstrates a strong texture along and . The impact test results demonstrate that C0 exhibits superior impact toughness with an absorbed energy of 12.87 J. The fractured surfaces are examined using the scanning electron microscope (SEM) and transmission electron microscope (TEM). Among them, the electron backscatter diffraction (EBSD) results indicate that the samples with higher geometrically necessary dislocation (GND) and kernel average misorientation (KAM) values can effectively reflect superior impact toughness. In contrast, fewer high-angle grain boundaries (HAGBs) are preferable. Due to the adiabatic effect, the phase transformation of the samples from austenite to martensite doesn't appear under the impact loading. Additionally, this paper discusses the impact of texture orientation, defects, and the relationship between loading direction and deposited layers on the impact toughness of the samples. These findings on impact properties would provide valuable guidance for engineering applications of NiTi SMAs.

Abstract As a “third code”, translational language attracts considerable attention in linguistics research due to its distinctive features. Adopting the quantitative linguistic approach, the current study examines its features by investigating the mean dependency distance (MDD), as well as the probability distribution of the individual dependency distances (DDs) and distribution of a high-frequency dependency type in translational language. The MDD and the distributions were tested in a self-built corpus which contains parallel and comparable language materials in both Chinese-English and English-Chinese translations. The results show that: (1) compared with source texts and native texts, translated texts in both translation directions yield an MDD in between; (2) both the distribution of DDs and that of the dependency type nsubj follow the Zipf-Alekseev distribution in translated texts, as in source texts and native texts; (3) the in-between feature is further confirmed by parameters a and b in Chinese-English translation materials when fitting the distribution of DDs to Zipf-Alekseev distribution; (4) translational texts in both directions show higher a and lower b than their source and native texts when fitting the DD Distribution of dependency type nsubj to Zipf-Alekseev distribution. These findings suggest that, on the one hand, dependency distance minimization (DDM) occurs in translational language, which is consistent with native language and reflects a general tendency of natural languages to reduce cognitive load; on the other hand, translational language presents distinctive feature in nsubj type, but in most cases, it is subject to the gravitational pull of both source and target language systems, exhibiting a “compromise” feature in between. The current study highlights the contribution of syntactic quantitative methods to deeper understanding of the complexity of translational language and its cognitive underpinnings.

Abstract Cancer-associated fibroblasts (CAFs) play a role in ovarian cancer (OV) evolution, immunosuppression and promotion of drug resistance. Exploring the value of CAFs-related biomarker in OV is of great importance. In the present work, we developed a CAFs-related index (CAFRI) based on an integrated analysis of single-cell and bulk RNA-sequencing and highlighted the value of CAFRI in predicting clinical outcomes in individuals with OV, tumour immune microenvironment (TIME) and response to immune checkpoint inhibitors (ICIs). The GSE151214 cohort was used for cell subpopulation localization and analysis, the TCGA-OV patients as a training set. Moreover, the ICGC-OV, GSE26193, GSE26712 and GSE19829 cohorts were used for the validation of CAFRI. The TIMER 2.0, CIBERSORT and ssGSEA algorithms were used for analysis of TIME characteristics based on the CAFRI. The GSVA, GSEA, GO, KEGG and tumour mutation burden (TMB) analyses were used for mechanistic exploration. Additionally, the IMvigor210 cohort was conducted to validate the predictive value of CAFRI on the efficacy of ICIs. Finally, CAFRI-based antitumour drug sensitivity was analysed. The findings demonstrate that the CAFRI can served as an excellent predictor of prognosis for individuals with OV, as well as identifying patients with different TIME characteristics, differentiating between immune ‘hot’ and ‘cold’ tumour populations, and providing new insights into the selection of ICIs and personalised treatment regimens. CAFRI provides new perspectives for the development of novel prognostic and immunotherapy efficacy predictive biomarkers for OV.

The discipline of plant immunity has developed rapidly in the 21st century and is marked by substantial progress toward understanding the molecular mechanisms of plant immune recognition and the pathogenicity of microorganisms. Here, we highlight the recent advances in plant immunity studies made by Yule Liu and his team at Tsinghua University in Beijing, China.

Background: Diesel exhaust particles (DEPs), a predominant component of ambient particulate matter (PM), are classified as ultrafine particles with the capacity to penetrate the cerebral blood-brain barrier (BBB). This penetration is implicated in the pathogenesis of central nervous system (CNS) disorders. The integrity of the BBB is inextricably linked to cerebrovascular homeostasis and the development of neurodegenerative disease, highlighting the importance of studying the effects and mechanisms of DEPs on BBB function damage. Methods and results: Utilizing mouse cerebral microvascular endothelial cells (bEnd.3 cells) as an in vitro model of the BBB, we explored the detrimental effects of DEPs exposure on BBB permeability and integrity, with particular focus on inflammation, cell apoptosis, and miRNA expression profiles. Our findings revealed that exposure to DEPs at varying concentrations for 48 h resulted in the inhibition of bEND.3 cell proliferation, induction of cell apoptosis, and an upregulation in the secretion of inflammatory cytokines/chemokines and adhesion molecules. The BBB integrity was further compromised, as evidenced by a decrease in trans-epithelial electrical resistance(TEER), a reduction in cytoskeletal F-actin, , and diminished tight junction (TJ) protein expression. Microarray analysis revealed that 23 miRNAs were upregulated and 11 were downregulated in response to a 50 μg/mL DEPs treatment, with miR-466d-3p being notably differentially expressed. Wnt3 was identified as a target of miR-466d-3p, with the Wnt signaling pathway being significantly enriched. We validated that miR-466d-3p expression was downregulated, and the protein expression levels of Wnt/β-catenin and Wnt/PCP signaling components were elevated. The modulation of the Wnt signaling pathway by miR-466d-3p was demonstrated by the transfection of miR-466d-3p mimic, which resulted in a downregulation of Wnt3 and β-catenin protein expression, and the mRNA level of Daam1, as well as an enhancement of TJ proteins ZO-1 and Claudin-5 expression. Conclusions: Our study further confirmed that DEPs can induce the disruption of BBB integrity through inflammatory processes. We identified alterations in the expression profile of microRNAs (miRNAs) in endothelial cells, with miR-466d-3p emerging as a key regulator of tight junction (TJ) proteins, essential for maintaining BBB integrity. Additionally, our findings primarily demonstrated that the Wnt/ β-catenin and Wnt/PCP signaling pathway can be activated by DEPs and are regulated by miR-466d-3p. Under the combined effects of Wnt/PCP and inflammation, there is an ultimate increase in BBB hyperpermeability. To explore how chemical, physical and biological process impact the adsorption capacity of biochar for cations and anions, five kinds of spent mushroom substrate biochars including PB (carbonized Auricularia auricula substrate at 600 °C for 6 h), NB (Aged PB at 25 °C and 40 % moisture for 12 mon), AB (PB treated with 20 % mixed acid solution at 70 °C for 6 h), FB (PB subjected to 16 freeze–thaw cycles between − 20 °C and 25 °C) and MB (PB incubated with microbes from contaminated soil for 15 cycles) were prepared under simulated conditions, the internal mechanisms were revealed. Results showed that all aging processes caused a variation in crystal quantity and structure on different biochar. AB, NB and MB had higher hydrophilicity and polarity, and more positive charges than PB. Moreover, AB and MB had a higher cation exchange capacity (CEC) (70.7 and 75.3 cmol/kg) and more oxygen-containing functional groups than PB. MB has the maximum adsorption capacity for Cd(II) (24.2 mg/g), followed by PB (23.5 mg/g), FB (22.7 mg/g), NB (22.2 mg/g), and AB (22.0 mg/g). Langmuir model better described the Cd(II) adsorption onto NB, FB, and PB, while Freundlich better described AB and MB. As for Cr(VI), the maximum adsorption capacity followed the order: MB (24.1 mg/g) > AB (23.7 mg/g) > FB (23.5 mg/g) > PB (23.0 mg/g) > NB (22.7 mg/g). Langmuir model was better for Cr(VI) adsorption onto FB and PB, while Freundlich was better for AB, NB and MB. The pseudo-first-order kinetic model fits for Cd adsorption by FB，while the pseudo-second-order kinetic model fit for the adsorption of Cd and Cr by other biochars. The adsorption of Cr by all biochars was significantly related to the content of lactone group. Acidification and organic acids from microorganisms increased the pHPZC values of AB and MB, which was not conducive to the adsorption of Cd. The surface fragmentation of FB caused by the freezing and thawing process promoted its adsorption of Cd.

Bone defects and non-union are prevalent in clinical orthopedy, and the outcomes of current treatments are often suboptimal. Bone tissue engineering offers a promising approach to treating these conditions effectively. Bone morphogenetic protein 9 (BMP9) can commit mesenchymal stem cells to osteogenic lineage, and a knowledge of the underlying mechanisms may help advance the field of bone tissue engineering. Leucine-rich repeats containing G protein-coupled receptor 4 (LGR4), a member of G protein-coupled receptors, is essential for modulating bone development. This study is aimed at investigating the impact of LGR4 on BMP9-induced osteogenesis in mesenchymal stem cells as well as the underlying mechanisms. Bone marrow stromal cells from BMP9-knockout mice exhibited diminished LGR4 expression, and exogenous LGR4 clearly restored the impaired osteogenic potency of the bone marrow stromal cells. Furthermore, LGR4 expression was increased by BMP9 in C3H10T1/2 cells. LGR4 augmented the benefits of BMP9-induced osteogenic markers and bone formation, whereas LGR4 inhibition restricted these effects. Meanwhile, the BMP9-induced lipogenic markers were increased by LGR4 inhibition. The protein levels of Raptor and p-Stat3 were elevated by BMP9. Raptor knockdown or p-Stat3 suppression attenuated the osteoblastic markers and LGR4 expression brought on by BMP9. LGR4 significantly reversed the blocking effect of Raptor knockdown or p-Stat3 suppression on the BMP9-induced osteoblastic markers. Raptor interacts with p-Stat3, and p-Stat3 activates the LGR4 promoter activity. Background: Cushing disease (CD) is a rare clinical neuroendocrine disease. CD is characterized by abnormal hypercortisolism induced by a pituitary adenoma with the secretion of adrenocorticotropic hormone. Individuals with CD usually exhibit atrophy of gray matter volume. However, little is known about the alterations in topographical organization of individuals with CD. This study aimed to investigate the structural covariance networks of individuals with CD based on the gray matter volume using graph theory analysis. Methods: High-resolution T1-weighted images of 61 individuals with CD and 53 healthy controls were obtained. Gray matter volume was estimated and the structural covariance network was analyzed using graph theory. Network properties such as hubs of all participants were calculated based on degree centrality. Results: No significant differences were observed between individuals with CD and healthy controls in terms of age, gender, and education level. The small-world features were conserved in individuals with CD but were higher than those in healthy controls. The individuals with CD showed higher global efficiency and modularity, suggesting higher integration and segregation as compared to healthy controls. The hub nodes of the individuals with CD were Short insular gyri (G_insular_short_L), Anterior part of the cingulate gyrus and sulcus (G_and_S_cingul-Ant_R), and Superior frontal gyrus (G_front_sup_R). Conclusions: Significant differences in the structural covariance network of patients with CD were found based on graph theory. These findings might help understanding the pathogenesis of individuals with CD and provide insight into the pathogenesis of this CD.

Abstract Fluorescent materials have attracted widespread attention for information encryption owing to their stimuli-responsive color-shifting. However, the 2D encoding of fluorescent images poses a risk of information leakage. Herein, inspired by the mimic octopus capable of camouflage by changing colors and shapes, we develop a thermadapt shape-memory fluorescent film (TSFF) for integrating 2D/3D encoding in one system. The TSFF is based on anthracene group with reversible photo-cross-linking and poly (ethylene-co-vinyl acetate) network with thermadapt shape-memory properties. The reversible photo-cross-linking of anthracene is accompanied by repeatable fluorescence-shifting and enables rewritable 2D encoding. Meanwhile, the thermadapt shape-memory properties not only enables the reconfiguration of the permanent shape for creating and erasing 3D patterns, i.e., rewritable 3D information, but also facilitates recoverable shape programming for 3D encoding. This rewritable 2D/3D encoding strategy can enhance information security because only designated inspectors can decode the information by providing sequential heating for shape recovery and UV exposure. Overall, TSFF capable of rewritable 2D/3D encoding will inspire the design of smart materials for high-security information carriers.

Abstract Introduction Oral histopathology is a bridge course connecting oral basic medicine and clinical dentistry. However, the application of outcomes-based education via flipped classroom (FC) in oral histopathology has not been well explored. This study has assessed the efficacy of outcomes-based education via FC in undergraduate oral histopathology module learning in Nanjing Medical University of China. Materials and methods A total of 214 third-year students were enrolled and assigned to the FC group of the batch 2022-23 (n = 110) and the traditional classroom (TC) group of the batch 2021-22 (n = 104) to participate the oral histopathology sessions respectively in the study. The FC group were required to preview the online course materials pre-class, followed by in-class quizz, in-class interactive group discussion, and slides microscopic observation. The outcomes-based formative and summative assessments for FC were designed. The TC group attended traditional laboratory class for the same glass slides microscopic observation. In addition, a questionnaire was performed to investigate the satisfaction of learning. Along with this, the performances of FC group in written theory tests and oral histopathology slide tests were compared with TC group. Abstract Topological photonic crystals, which offer topologically protected and back-scattering-immune transport channels, have recently gained significant attention for both scientific and practical reasons. Although most current studies focus on dielectric materials with weak dispersions, this study focuses on topological phases in dispersive materials and presents a numerical study of Chern insulators in gaseous-phase plasma cylinder cells. We develop a numerical framework to address the complex material dispersion arising from the plasma medium and external magnetic fields and identify Chern insulator phases that are experimentally achievable. Using this numerical tool, we also explain the flat bands commonly observed in periodic plasmonic structures, via local resonances, and how edge states change as the edge termination is periodically modified. This work opens up opportunities for exploring band topology in new materials with non-trivial dispersions and has potential radio frequency (RF) applications, ranging from plasma-based lighting to plasma propulsion engines.

To study the preservation effect of chitosan coating with different molecular weights on postharvest passion fruit, the "Qinmi No.9" was coated with chitosan of molecular weights of 30, 50, 100, 150 and 200 kDa (1.5%, w/v) to determine the quality of passion fruit during storage. The results showed that chitosan coating with different molecular weights was able to delay the shrinkage and yellowing, reduce the weight loss rate and inhibit the decay of passion fruit. Moreover, chitosan with a larger molecular weight was more conducive to delaying the ripening and senescence of passion fruit, as well as reducing shrinkage, and decay. At the end of storage, the weight loss of fruits coated with 200 kDa chitosan was nearly 10% less than that coated with 30 kDa chitosan, and the fruits coated with 150 and 200 kDa chitosan did not decay. The lower molecular weight (30 and 50 kDa) and higher molecular weight (150 kDa) chitosan were more effective in inhibiting weight loss, total soluble solids and soluble sugar metabolism, and maintaining titratable acid, flavonoid and total phenol contents of fruit during storage. The chitosan with 150 kDa had the best effect in maintaining the vitamin C content, which was 1.12 times higher than the control group at the end of storage. In conclusion, chitosan with different molecular weights was effective to delay senescence, slow down water loss and shrink of passion fruit and maintain the quality, chitosan with 150 kDa was more suitable to maintain the quality of postharvest passion fruit.

Objective·To investigate the correlation between intramuscular adipose tissue (IMAT) content and glucose metabolism in male patients with obesity.Methods·Eighty male patients with obesity were recruited from the Endocrinology Department of Renji Hospital, Shanghai Jiao Tong University School of Medicine from December 2019 to December 2020. According to the results of oral glucose tolerance test (OGTT), they were divided into normal glucose tolerance (NGT) group and impaired glucose regulation (IGR) group. Abstract Developing stable and efficient electrocatalysts is vital for boosting oxygen evolution reaction (OER) rates in sustainable hydrogen production. High-entropy oxides (HEOs) consist of five or more metal cations, providing opportunities to tune their catalytic properties toward high OER efficiency. This work combines theoretical and experimental studies to scrutinize the OER activity and stability for spinel-type HEOs. Density functional theory confirms that randomly mixed metal sites show thermodynamic stability, with intermediate adsorption energies displaying wider distributions due to mixing-induced equatorial strain in active metal-oxygen bonds. The rapid sol-flame method is employed to synthesize HEO, comprising five 3d-transition metal cations, which exhibits superior OER activity and durability under alkaline conditions, outperforming lower-entropy oxides, even with partial surface oxidations. The study highlights that the enhanced activity of HEO is primarily attributed to the mixing of multiple elements, leading to strain effects near the active site, as well as surface composition and coverage.

Abstract Background Although sex bias has been reported in the development and progression of renal cell carcinoma (RCC), the underlying mechanisms remain enigmatic. Here, we investigated the sex differences in the tumor microenvironment (TME) of RCC and explored a promising combination drug regimen to enhance the efficacy of immunotherapy. Methods Single-cell RNA sequencing (scRNA-seq) data from four published datasets were analyzed to investigate the sex differences in RCC patients, and tumor tissues were collected to validate the sex differences using multiplex immunofluorescence (MxIF) and flow cytometry (FCM). The function of the androgen–androgen receptor axis in sex differences was explored in vivo and in vitro experiments. Results Our analysis of scRNA-seq data from 220,156 cells, as well as MxIF and FCM assays, revealed that CD8+ T-cells infiltrated highly in the TME of male RCC, but were mostly in an exhausted and dysfunctional state. In vitro and in vivo experiments indicated that the dysfunction and exhaustion of CD8+ T-cells in male TME were induced by androgen. Clinically, higher serum androgen was significantly associated with a worse prognosis in male RCC patients receiving immunotherapy. Androgen receptor inhibitors could activate tumor-infiltrating CD8+ T-cells and enhance the efficacy of immunotherapy of RCC in vivo. Conclusions Our study delineated the difference in TME between male and female patients with RCC, and demonstrated that the androgen–androgen receptor axis plays an important role in immunosuppression in male RCC. Our findings suggest that androgen receptor inhibitors in combination with immunotherapy may be a promising treatment option for male RCC patients.

Due to its rich nutritional composition, biogas slurry can serve as a special liquid fertilizer. However, the application of slurry in agricultural fields currently faces challenges such as reliance on skilled famers’ experience, low precision, and difficulty in accurately controlling the irrigation dosage. To address these issues, an agricultural biogas slurry mixing agricultural machinery and its system has been designed and developed with the aim of enhancing the precision and safety of slurry application. The structure of the device has been designed, filter components have been selected, and improvements have been made to the structure of traditional connectors. Taking into account factors such as soil and humidity, an algorithm based on biogas slurry conductivity for slurry mixing decisions and the feedback control mechanism has been designed. After assembling the prototype, experiments were conducted, and the results showed that after processing by the system, compared to the calculated theoretical optimum, the concentration error of each component in the mixed fertilizer was controlled within 10%, and the conductivity fluctuation range was within 5%. This indicates that the overall ratio accuracy and stability of the biogas slurry mixing system are high. The biogas slurry mixing agricultural machinery and its system provide a novel intelligent equipment solution for the precise application of slurry, effectively enhancing the accuracy and safety of slurry application, reducing the use of chemical fertilizers during agricultural irrigation, and minimizing pollution to the environment and soil.

Ginseng, an important medicinal plant, is characterized by its main active component, ginsenosides. Among more than 40 ginsenosides, Rg1 is one of the ginsenosides used for measuring the quality of ginseng. Therefore, the identification and characterization of genes for Rg1 biosynthesis are important to elucidate the molecular basis of Rg1 biosynthesis. In this study, we utilized 39,327 SNPs and the corresponding Rg1 content from 344 core ginseng cultivars from Jilin Province. We conducted a genome-wide association study (GWAS) combining weighted gene co-expression network analysis (WGCNA), SNP-Rg1 content association analysis, and gene co-expression network analysis; three candidate Rg1 genes (PgRg1-1, PgRg1-2, and PgRg1-3) and one crucial candidate gene (PgRg1-3) were identified. Functional validation of PgRg1-3 was performed using methyl jasmonate (MeJA) regulation and RNAi, confirming that this gene regulates Rg1 biosynthesis. The spatial–temporal expression patterns of the PgRg1-3 gene and known key enzyme genes involved in ginsenoside biosynthesis differ. Furthermore, variations in their networks have a significant impact on Rg1 biosynthesis. This study established an accurate and efficient method for identifying candidate genes, cloned a novel gene controlling Rg1 biosynthesis, and identified 73 SNPs significantly associated with Rg1 content. This provides genetic resources and effective tools for further exploring the molecular mechanisms of Rg1 biosynthesis and molecular breeding.

BackgroundBrain atrophy is a type of neurological and psychiatric disorder characterized by a decrease in brain tissue volume and weight for various reasons and can have a serious impact on the quality of life of patients. Although there are many studies on brain atrophy, there is a lack of relevant bibliometric studies. Therefore, this study aims to provide a visual analysis of global trends in brain atrophy research over the past 16 years.MethodsCiteSpace and VOSviewer were used to visually analyze publication output, scientific collaborations, cocitations, publishing journals, and keywords to determine the current status and future trends of brain atrophy research. Materials published from 2008 to 2023 were collected from the Web of Science Core Collection (WoSCC) database. This study placed no restrictions on the types of literature and focused on English language publications.ResultsA total of 3,371 publications were included in the analysis. From 2008 to 2023, the number of publications increased annually. In terms of national and academic institutions, universities in the United States and University College London rank first in publication out. Barkhof Frederik and Zivadinov Robert are the most prolific researchers in this field. The publication with the highest cocitation strength is “Deep gray matter volume loss drives disability worsening in multiple sclerosis.” Keyword clustering analysis showed that “Alzheimer’s disease” and “multiple sclerosis” are current popular topics. The analysis of emergent words indicates that “cerebral small vessel disease,” “neurodegeneration,” and “cortex/gray matter volume” may become hot research topics in the coming years.ConclusionThis study analyses papers on brain atrophy from the past 16 years, providing a new perspective for research in this field. In the past 16 years, research on brain atrophy has received increasing attention. The quality of articles in this field is generally high. Extensive national cooperation already exists. The statistical results indicate that a stable core author group in the field of brain atrophy has almost formed.

The building automation system (BAS) of a subway is the core component of monitoring and managing urban rail transit systems. For the current problems such as low control efficiency, insufficient accuracy, and poor stability of metro BAS, this article proposes a cooperative control framework based on an improved fuzzy proportional-integral-derivative (PID) algorithm. Firstly, the concept of an integrated supervisory control system (ISCS) for subways is introduced by summarizing the previously implemented engineering construction and combining it with advanced automation technology. The system’s overall design under the ISCS framework is also improved by integrating it with the fire alarm system (FAS) with the BAS as the core unit of the reliance. Then, an improved seeker optimization algorithm (ISOA) is employed to optimize the parameters of the fuzzy PID control algorithm to achieve a coordinated control of the system based on considering the time lag problem. Finally, the accuracy, efficiency, and stability of the coordinated control response of the BAS under the ISCS framework are tested experimentally. The results suggest that the proposed cooperative control solution of BAS employing the improved fuzzy PID algorithm has good control accuracy and response efficiency and can also ensure the BAS’s higher stability in the coordinated control process, which thus greatly improves the automation level of the subway and provides a safer and more reliable high-performance for the ISCS of the subway in the urban rail transportation industry.

Abstract Statistics show that more than 80 applications are installed on each android smartphone. Vulnerability research on Android applications is of critical importance. Recently, academic researchers mainly focus on single bug patterns, while few of them investigate the relations between multiple bugs. Industrial researchers proposed a series of logic exploit chains leveraging multiple logic bugs. However, there is no general model to evaluate the chaining abilities between bugs. This paper presents a formal model to elucidate the relations between multiple bugs in Android applications. To prove the effectiveness of the model, we design and implement a prototype system named AppChainer. AppChainer automatically identifies attack surfaces of Android applications and investigates whether the payloads entering these attack surfaces are “chainable”. Experimental results on 2138 popular Android applications show that AppChainer is effective in identifying and chaining attacker-controllable payloads. It identifies 14467 chainable payloads and constructs 5458 chains both inside a single application and among various applications. The time cost and resource consumption of AppChainer are also acceptable. For each application, the average analysis time is 317 s, and the average memory consumed is 2368 MB. Compared with the most relevant work Jandroid, the experiment results on our custom DroidChainBench show that AppChainer outperforms Jandroid at the precision rate and performs equally with Jandroid at the recall rate.

Abstract Background Impaired sensitivity to thyroid hormones is a newly proposed clinical entity associated with hyperuricemia in the subclinical hypothyroid population. However, it is unknown whether the association exists in the euthyroid population. This study aimed to explore the association of impaired sensitivity to thyroid hormones (assessed by the thyroid feedback quantile-based index [TFQI], parametric thyroid feedback quantile-based index [PTFQI], thyrotrophic thyroxine resistance index [TT4RI] and thyroid-stimulating hormone index [TSHI]) with hyperuricemia and quantify the mediating effect of body mass index BMI in the euthyroid population. Methods This cross-sectional study enrolled Chinese adults aged ≥ 20 years who participated in the Beijing Health Management Cohort (2008–2019). Adjusted logistic regression models were used to explore the association between indices of sensitivity to thyroid hormones and hyperuricemia. Odds ratios [OR] and absolute risk differences [ARD] were calculated. Mediation analyses were performed to estimate direct and indirect effects through BMI. Results Of 30,857 participants, 19,031 (61.7%) were male; the mean (SD) age was 47.3 (13.3) years; and 6,515 (21.1%) had hyperuricemia. After adjusting for confounders, individuals in the highest group of thyroid hormone sensitivity indices were associated with an increased prevalence of hyperuricemia compared with the lowest group (TFQI: OR = 1.18, 95% CI 1.04–1.35; PTFQI: OR = 1.20, 95% CI 1.05–1.36; TT4RI: OR = 1.17, 95% CI 1.08–1.27; TSHI: OR = 1.12, 95% CI 1.04–1.21). BMI significantly mediated 32.35%, 32.29%, 39.63%, and 37.68% of the associations of TFQI, PTFQI, TT4RI and TSHI with hyperuricemia, respectively. Conclusions Our research revealed that BMI mediated the association between impaired sensitivity to thyroid hormones and hyperuricemia in the euthyroid population. These findings could provide useful evidence for understanding the interaction between impaired sensitivity to thyroid hormone and hyperuricemia in euthyroid individuals and suggest the clinical implications of weight control in terms of impaired thyroid hormones sensitivity.

The acceleration of aging is a risk factor for numerous diseases, and diet has been identified as an especially effective anti-aging method. Currently, research on the relationship between dietary nutrient intake and accelerated aging remains limited, with existing studies focusing on the intake of a small number of individual dietary nutrients. Comprehensive research on the single and mixed anti-aging effects of dietary nutrients has not been conducted. This study aimed to comprehensively explore the effects of numerous dietary nutrient intakes, both singly and in combination, on the acceleration of aging. Data for this study were extracted from the 2015–2018 National Health and Nutrition Examination Surveys (NHANES). The acceleration of aging was measured by phenotypic age acceleration. Linear regression (linear), restricted cubic spline (RCS) (nonlinear), and weighted quantile sum (WQS) (mixed effect) models were used to explore the association between dietary nutrient intake and accelerated aging. A total of 4692 participants aged ≥ 20 were included in this study. In fully adjusted models, intakes of 16 nutrients were negatively associated with accelerated aging (protein, vitamin E, vitamin A, beta-carotene, vitamin B1, vitamin B2, vitamin B6, vitamin K, phosphorus, magnesium, iron, zinc, copper, potassium, dietary fiber, and alcohol). Intakes of total sugars, vitamin C, vitamin K, caffeine, and alcohol showed significant nonlinear associations with accelerated aging. Additionally, mixed dietary nutrient intakes were negatively associated with accelerated aging. Single dietary nutrients as well as mixed nutrient intake may mitigate accelerated aging. Moderately increasing the intake of specific dietary nutrients and maintaining dietary balance may be key strategies to prevent accelerated aging.

During the infusion process of a glass-fiber-reinforced thermosetting composite hose, the viscosity of its resin matrix undergoes temporal variations. Consequently, if the impact of resin viscosity changes over time on the internal resin fluidity is not considered during the infusion process, this may result in the incomplete impregnation of the hose, characterized by the presence of numerous voids. This phenomenon adversely affects the quality of the pipe’s curing and forming process. Therefore, based on the characteristic variations in resin viscosity, this paper considers the changes in fluidity caused by the resin’s temporal evolution within the material. We establish a finite element simulation model to calculate and analyze the overall infusion effects of resin viscosity changes during the hose infusion process. Furthermore, based on the predicted analysis, a variable parameter infusion strategy is proposed to increase resin impregnation in the hose, thereby reducing internal void content and subsequently improving the quality of material curing and forming.

With the increasing availability of satellite monitoring data, the demand for storage and computational resources for updating the results of monitoring the surface subsidence in a mining area continues to rise. Sequential adjustment (SA) models are considered effective for rapidly updating time series interferometry synthetic aperture radar (TS-InSAR) measurements. However, the accuracy of surface subsidence values estimated through traditional sequential adjustment is highly sensitive to abnormal observations or prior information on anomalies. Moreover, the surface subsidence associated with mining exhibits nonlinear and large gradient characteristics, making general InSAR methods challenging for obtaining reliable monitoring results. In this study, we employ the phase unwrapping network (PUNet) to obtain unwrapped values of differential interferograms. To mitigate the impact of abnormal errors in the near real-time small baseline subset InSAR (SBAS-InSAR) sequential updating process in mining areas, a robust sequential adjustment method based on M-estimation is proposed to estimate the temporal deformation parameters by using the equivalent weight model. Using a coal backfilling mining face in Shanxi, China, as the study area and the Sentinel-1 SAR dataset, we comprehensively evaluate the performance of unwrapping methods and subsidence time series estimation techniques and evaluate the effect of filling mining on surface subsidence control. The results are validated using leveling measurements within the study area. The relative error of the proposed method is less than 5%, which can meet the requirements of monitoring the surface subsidence in mining areas. The method proposed in this study not only enhances computational efficiency but also addresses the issue of underestimation encountered by InSAR methods in mining area applications. Furthermore, it also mitigates unwrapping phase anomalies on the monitoring results.

Geogenic arsenic (As)-rich groundwater poses a significant environmental challenge worldwide, yet our understanding of the interplay between dissolved organic matter (DOM) transformation and arsenic mobilization during microbial sulfate reduction remains limited. This study involved microcosm experiments using As-rich aquifer sediments from the Singe Tsangpo River basin (STR) and Jianghan Plain (JHP), respectively. The findings revealed that microbial sulfate reduction remarkably increased arsenic mobilization in both STR and JHP sediments compared to that in unamended sediments. Moreover, the mobilization of As during microbial sulfate reduction coincided with increases in the fluorescence intensity of two humic-like substances, C2 and C3 (R = 0.87/0.87 and R = 0.73/0.66 in the STR and JHP sediments, respectively; p < 0.05), suggesting competitive desorption between DOM and As during incubation. Moreover, the transformations in the DOM molecular characteristics showed significant increases in CHOS molecular and low-O/C-value molecular intensities corresponding to the enhancement of microbial sulfate reduction and the possible occurrence of methanogenesis processes, which suggests a substantial bioproduction contribution to DOM components that is conducive to As mobilization during the microbial sulfate reduction. The present results thus provide new insights into the co-evolution between As mobilization and DOM transformations in alluvial aquifer systems under strong microbial sulfate reduction conditions.

Numerous single-image dehazing algorithms have been developed, employing a spectrum of techniques ranging from intricate physical computations to state-of-the-art deep-learning methodologies. However, conventional deep-learning approaches, particularly those based on standard convolutional neural networks (CNNs), often result in the persistence of residual fog patches when applied to images featuring high fog concentration or heterogeneous fog distribution. In response to this challenge, we propose an innovative solution known as the multi-feature fusion image dehazing network (MFID-Net). This approach employs an end-to-end methodology to directly capture the mapping relationship between hazy and fog-free images. Central to our approach is the introduction of a novel multi-feature fusion (MF) module, strategically designed to address channel and pixel characteristics in regions with uneven or high fog concentrations. Notably, this module achieves effective haze reduction while minimizing computational resources, thereby mitigating the issue of residual fog patches. Experimental results underscore the superior performance of our algorithm compared to similar dehazing methods, as evidenced by higher scores in structural similarity (SSIM), peak signal-to-noise ratio (PSNR), and computational velocity. Moreover, MFID-Net exhibits significant advancements in restoring details within expansive monochromatic areas, such as skies and white walls.

To improve the performance of sprinklers that work under low pressure, a new type of automatic rotating sprinkler equipped with different water dispersion devices was designed and the jet diffusion angle and hydraulic performance of the new and original sprinklers were compared using experimental methods. The results indicate that, when the working pressure is below 200 kPa, the jet diffusion angle of the new sprinkler with the water dispersion device B is the largest. Although the pattern radius of the new sprinkler is slightly lower than that of the original, it is more effective in improving the combined coefficient of uniformity (CU) at lower working pressures. Specifically, when the working pressure was 100 kPa, the new sprinkler with the water dispersion device B, which has a square inner hole with a side length of 6.2 mm, improved the CU by 54.89% on average. Remarkably, the CU of the new sprinkler with water dispersion device B at a working pressure of 100 kPa was 6.65% higher than that of the original sprinkler at a working pressure of 200 kPa. The research results provide valuable insights into the design of sprinklers to improve the performance of low-pressure sprinkler irrigation systems. HIGHLIGHTS A novel automatic rotating sprinkler (NARS) with a water dispersion device has been successfully developed.; The lower the working pressure, the more pronounced the effect of the NARS on improving the overlapped water application uniformity.; The NARS with water dispersion device B ensures that the water application rates within a local area are consistent.;

Abstract Introduction Clinical practice of dentistry entails the use of indirect vision using a dental mirror. The Mirrosistant is a device that helps dental students become proficient with use of indirect vision mirror operation. This study aimed to explore the role of the Mirrosistant on students’ performance with the virtual simulation dental training system. Materials and methods A total of 72 dental students were equally assigned to the Control group and the Experimental group. Subsequently, Mirrosistant was used to conduct a series of mirror training exercises in the Experimental group. The training consisted of tracing the edge and filling in the blank of the prescribed shape, as well as preparing the specified figure on raw eggs using indirect vision via Mirrosistant. Next, both groups were examined using the SIMODONT system, a virtual reality dental trainer, for mirror operation. In addition, a five-point Likert scale questionnaire was used to assess student feedback by using Mirrosistant. Results The mirror operation examination conducted by the SIMODONT system revealed that mirror training using Mirrosistant had statistically improved students’ performances (score: 80.42 ± 6.43 vs. 69.89 ± 15.98, P = 0.0005) and shorten their performance time of mirror operation (time of seconds: 243.28 ± 132.83 vs. 328.53 ± 111.89, P = 0.0013). Furthermore, the questionnaire survey indicated that the participants had positive attitudes toward the mirror training using Mirrosistant. Most students believed that the mirror training device could improve their perceptions of direction and distance, as well as their sensations of dental operation and dental fulcrum. Conclusion Mirror training using Mirrosistant can enhance dental students’ mirror perceptual and operational skills on virtual simulation dental training system.

Abstract Background Effective risk prediction models are lacking for personalized endoscopic screening of gastric cancer (GC). We aimed to develop, validate, and evaluate a questionnaire-based GC risk assessment tool for risk prediction and stratification in the Chinese population. Methods In this three-stage multicenter study, we first selected eligible variables by Cox regression models and constructed a GC risk score (GCRS) based on regression coefficients in 416,343 subjects (aged 40–75 years) from the China Kadoorie Biobank (CKB, development cohort). In the same age range, we validated the GCRS effectiveness in 13,982 subjects from another independent Changzhou cohort (validation cohort) as well as in 5348 subjects from an endoscopy screening program in Yangzhou. Finally, we categorized participants into low (bottom 20%), intermediate (20–80%), and high risk (top 20%) groups by the GCRS distribution in the development cohort. Results The GCRS using 11 questionnaire-based variables demonstrated a Harrell’s C-index of 0.754 (95% CI, 0.745–0.762) and 0.736 (95% CI, 0.710–0.761) in the two cohorts, respectively. In the validation cohort, the 10-year risk was 0.34%, 1.05%, and 4.32% for individuals with a low (≤ 13.6), intermediate (13.7~30.6), and high (≥ 30.7) GCRS, respectively. In the endoscopic screening program, the detection rate of GC varied from 0.00% in low-GCRS individuals, 0.27% with intermediate GCRS, to 2.59% with high GCRS. A proportion of 81.6% of all GC cases was identified from the high-GCRS group, which represented 28.9% of all the screened participants. Conclusions The GCRS can be an effective risk assessment tool for tailored endoscopic screening of GC in China. Risk Evaluation for Stomach Cancer by Yourself (RESCUE), an online tool was developed to aid the use of GCRS.

Abstract Background Pre-existing metabolic diseases may predispose individuals to particulate matter (PM)-induced adverse health effects. However, the differences in susceptibility of various metabolic diseases to PM-induced lung injury and their underlying mechanisms have yet to be fully elucidated. Results Type 1 diabetes (T1D) murine models were constructed by streptozotocin injection, while diet-induced obesity (DIO) models were generated by feeding 45% high-fat diet 6 weeks prior to and throughout the experiment. Mice were subjected to real-ambient PM exposure in Shijiazhuang City, China for 4 weeks at a mean PM2.5 concentration of 95.77 µg/m3. Lung and systemic injury were assessed, and the underlying mechanisms were explored through transcriptomics analysis. Compared with normal diet (ND)-fed mice, T1D mice exhibited severe hyperglycemia with a blood glucose of 350 mg/dL, while DIO mice displayed moderate obesity and marked dyslipidemia with a slightly elevated blood glucose of 180 mg/dL. T1D and DIO mice were susceptible to PM-induced lung injury, manifested by inflammatory changes such as interstitial neutrophil infiltration and alveolar septal thickening. Notably, the acute lung injury scores of T1D and DIO mice were higher by 79.57% and 48.47%, respectively, than that of ND-fed mice. Lung transcriptome analysis revealed that increased susceptibility to PM exposure was associated with perturbations in multiple pathways including glucose and lipid metabolism, inflammatory responses, oxidative stress, cellular senescence, and tissue remodeling. Functional experiments confirmed that changes in biomarkers of macrophage (F4/80), lipid peroxidation (4-HNE), cellular senescence (SA-β-gal), and airway repair (CCSP) were most pronounced in the lungs of PM-exposed T1D mice. Furthermore, pathways associated with xenobiotic metabolism showed metabolic state- and tissue-specific perturbation patterns. Upon PM exposure, activation of nuclear receptor (NR) pathways and inhibition of the glutathione (GSH)-mediated detoxification pathway were evident in the lungs of T1D mice, and a significant upregulation of NR pathways was present in the livers of T1D mice. Conclusions These differences might contribute to differential susceptibility to PM exposure between T1D and DIO mice. These findings provide new insights into the health risk assessment of PM exposure in populations with metabolic diseases.

This study aimed to explore the differences in raw Angelica Sinensis (RAS), wine washing AS (WAS), and wine stir-frying AS (WSAS). The results showed there were differences among the three AS in color and aroma, and 34 aroma compounds were identified. The content determination results revealed the ferulic acid and Z-ligustilide levels of RAS decreased after processing, and those in WAS were higher than in WSAS. Furthermore, 85 representative common components and 37 unique components were tentatively identified in three AS. Finally, the free radical scavenging assay results indicated the antioxidant capacity of RAS was reduced after processing, and the antioxidant capacity of WAS was better than WSAS. Collectively, the RAS undergoes significant changes in color, aromas, components, and antioxidant ability after processing, and the different processing methods also result in significant differences between WAS and WSAS.

Summary: Large biosynthetic gene cluster (BGC) cloning is important for discovering natural product-based drugs and remains challenging in high GC content microorganisms (e.g., Actinobacteria). Here, we present an in vitro CRISPR-Cas12a-mediated protocol for direct cloning of large DNA fragments. We describe steps for crRNA design and preparation, genomic DNA isolation, and CRISPR-Cas12a cleavage and capture plasmid construction and linearization. On a global scale, drought, salinity, extreme temperature, and other abiotic stressors severely limit the quality and yield of crops. Therefore, it is crucial to clarify the adaptation strategies of plants to harsh environments. Chloroplasts are important environmental sensors in plant cells. For plants to thrive in different habitats, chloroplast homeostasis must be strictly regulated, which is necessary to maintain efficient plant photosynthesis and other metabolic reactions under stressful environments. To maintain normal chloroplast physiology, two important biological processes are needed: the import and degradation of chloroplast proteins. The orderly import of chloroplast proteins and the timely degradation of damaged chloroplast components play a key role in adapting plants to their environment. In this review, we briefly describe the mechanism of chloroplast TOC-TIC protein transport. The importance and recent progress of chloroplast protein turnover, retrograde signaling, and chloroplast protein degradation under stress are summarized. Furthermore, the potential of targeted regulation of chloroplast homeostasis is emphasized to improve plant adaptation to environmental stresses.

Abstract Background Malignant pleural mesothelioma (MPM) is a rare and highly malignant thoracic tumor. Although alternative splicing (AS) is associated with tumor prognosis, the prognostic significance of AS in MPM is unknown. Methods Transcriptomic data, clinical information, and splicing percentage values for MPM were obtained from The Cancer Genome Atlas (TCGA) and TCGA SpliceSeq databases. Least absolute shrinkage and selection operator (LASSO) regression and multivariate Cox analyses were performed to establish a model affecting the prognosis of MPM. Survival and ROC analyses were used to test the effects of the prognostic model. LASSO/multivariate Cox analysis was used to construct the MPM prognostic splicing factor (SF) model. The SF–AS interaction network was analyzed using Spearman correlation and visualized using Cytoscape. The association between the MPM prognostic SF model and drug sensitivity to chemotherapeutic agents such as cisplatin was analyzed using pRRophetic.R. Results The LASSO/multivariate Cox analysis identified 41 AS events and 2 SFs that were mostly associated with survival. Nine prognostic prediction models (i.e., seven types of AS model, total AS model, and SF model) were developed. An MPM prognostic SF–AS regulatory network was subsequently constructed with decreased drug sensitivity in the SF model high‐risk group (p = 0.025). Conclusion This study provides the first comprehensive analysis of the prognostic value of AS events and SFs in MPM. The SF–AS regulatory network established in this study and our drug sensitivity analysis using the SF model may provide novel targets for pharmacological studies of MPM.
