Background While extensive research has provided a wealth of information on psoriasis in general, there remains a critical gap in understanding the unique characteristics of psoriasis in special body areas, such as the scalp, nails, palms, and genitals.Objective To investigate the characterization and treatment of psoriasis patients in special body areas.Methods The study was a retrospective analysis of patients with psoriasis enrolled in the Psoriasis Standardized Diagnosis and Treatment Center Project between January 2020 and September 2021.Results The study encompassed 346 patients, 81% of them had psoriasis in at least two special body areas, with the nails as the most common area. Patients with genital psoriasis reported higher Dermatology Life Quality Index (DLQI) scores. A higher propensity for scalp and palmoplantar psoriasis was noted in patients with genital psoriasis. The proportion of patients treated with biologics rose, as the number of specific areas involved increased.Conclusions Patients with genital psoriasis are more likely to have scalp and palmoplantar psoriasis. This study highlights the significant escalation in the proportion of biologics when the involvement of special body areas was ≥2.

Permanent magnet-assisted synchronous reluctance motors (PMA-SynRMs) are widely used in various fields due to their significant advantages, including strong torque output, high efficiency, excellent speed regulation, and low cost. The PMA-SynRM with asymmetric-rotor structure has a weaker anti-demagnetization performance than the conventional PMA-SynRM due to its multi-layer and thin permanent magnets construction. According to the finite element (FEM) simulation analysis, the anti-demagnetization performance of the asymmetric-rotor PMA-SynRM can be improved by adding bypass magnetic bridges on the ribs of the flux barriers and by changing the positions of the permanent magnets. The rotor structure of the proposed model is globally optimized by combining the two methods. Anti-demagnetization performance is improved as much as possible under the premise of ensuring the torque performance of the basic model. After multi-objective optimization, there is almost no difference between the optimized model and the basic model in terms of no-load air-gap flux density, no-load Back-electromotive force (EMF), and average torque. The maximum demagnetization rate of the optimized model is reduced by 81.44% compared with the basic model, and the anti-demagnetization performance is significantly improved. At the same time, the torque ripple is also reduced by 44.14%, which is obviously reduced. Compared with the basic model, the optimized model has better stability and reliability.

Psoriasis is a complex, chronic autoimmune disorder predominantly affecting the skin. Accumulating evidence underscores the critical role of localized cellular inflammation in the development and persistence of psoriatic skin lesions, involving cell types such as keratinocytes, mesenchymal cells, and Schwann cells. However, the underlying mechanisms remain largely unexplored. Long non-coding RNAs (lncRNAs), known to regulate gene expression across various cellular processes, have been particularly implicated in immune regulation. We utilized our neural-network learning pipeline to integrate 106,675 cells from healthy human skin and 79,887 cells from psoriatic human skin. This formed the most extensive cell transcriptomic atlas of human psoriatic skin to date. The robustness of our reclassified cell-types, representing full-layer zonation in human skin, was affirmed through neural-network learning-based cross-validation. We then developed a publicly available website to present this integrated dataset. We carried out analysis for differentially expressed lncRNAs, co-regulated gene patterns, and GO-bioprocess enrichment, enabling us to pinpoint lncRNAs that modulate localized cellular inflammation in psoriasis at the single-cell level. Subsequent experimental validation with skin cell lines and primary cells from psoriatic skin confirmed these lncRNAs’ functional role in localized cellular inflammation. Our study provides a comprehensive cell transcriptomic atlas of full-layer human skin in both healthy and psoriatic conditions, unveiling a new regulatory mechanism that governs localized cellular inflammation in psoriasis and highlights the therapeutic potential of lncRNAs in this disease’s management.

The effects of electrolyzing water-cooled green tea with negative redox potential on the quality of tea soup was studied by comparing the bottled natural water-cooled green tea and conventional hot bubble green tea, in order to provide a new choice for the processing water of cold bubble green tea with high content of active ingredient and strong antioxidant activity. Electrolyzing drinking water by adjusting the electrolysis time to generate electrolytic water samples 1 and 2 with different negative redox potentials. The basic physicochemical indexes, main active ingredient, antioxidant activity, sensory quality and aroma components of electrolyzed water with negative redox potential, bottled natural water-cooled green tea and conventional hot bubble green tea were determined and compared by UV spectrophotometry, high performance liquid chromatography, headspace solid-phase microextraction and gas chromatography-mass spectrometry. The dq current transformation method can be used to achieve the drive control of the maglev permanent magnet synchronous linear motor (MPMSLM). Specifically, the control of the suspension force Fz can be achieved by controlling the d-axis current, and the control of the thrust force Fx can be achieved by controlling the q-axis current. However, the direct use of the dq current transformation method on traditional MPMSLM structures produces an additional rotation torque around the y-axis. In order to reduce this additional torque, a new MPMSLM structure is proposed in this work. First, the characteristics of additional torque through finite element analysis are analyzed. Second, the Halbach permanent magnet array and coil size are optimized, and the topology of the MPMSLM coil is designed to reduce the additional torque. The decoupling performance and current of the proposed MPMSLM are experimentally verified through open-loop experiments using finite element simulation software. Finally, the decoupling algorithm of the generalized inverse matrix is used to achieve the decoupling between forces Fx and Fz and torque Ty and the d-axis and q-axis currents in each drive unit. Based on this, a three-degree-of-freedom closed-loop control system of the MPMSLM is designed. The LabVIEW 2018 software is used for the simulation analysis of the three-degree-of-freedom MPMSLM motion control system, and the results show that the proposed motor structure has superior closed-loop control performance.

Abstract Psoriasis is the most common skin disease in adults. Current experimental and clinical evidences suggested the infiltrating immune cells could target local skin cells and thus induce psoriatic phenotype. However, recent studies indicated the existence of a potential feedback signaling loop from local resident skin cells to infiltrating immune cells. Here, we deconstructed the full-thickness human skins of both healthy donors and patients with psoriasis vulgaris at single cell transcriptional level, and further built a neural-network classifier to evaluate the evolutional conservation of skin cell types between mouse and human. Last, we systematically evaluated the intrinsic and intercellular molecular alterations of each cell type between healthy and psoriatic skin. Cross-checking with psoriasis susceptibility gene loci, cell-type based differential expression, and ligand-receptor communication revealed that the resident psoriatic skin cells including mesenchymal and epidermis cell types, which specifically harbored the target genes of psoriasis susceptibility loci, intensively evoked the expression of major histocompatibility complex (MHC) genes, upregulated interferon (INF), tumor necrosis factor (TNF) signalling and increased cytokine gene expression for primarily aiming the neighboring dendritic cells in psoriasis. The comprehensive exploration and pathological observation of psoriasis patient biopsies proposed an uncovered immunoregulatory axis from skin local resident cells to immune cells, thus provided a novel insight for psoriasis treatment. In addition, we published a user-friendly website to exhibit the transcriptional change of each cell type between healthy and psoriatic human skin.

To solve the artifact problem in fused images and the lack of enough generalization under different scenarios of existing fusion algorithms, the paper proposes an image fusion algorithm based on improved RGF and visual saliency map to realize fusion for infrared and visible light images and a multimode medical image. Firstly, the paper uses RGF (rolling guidance filter) and Gaussian filter to decompose the image into the base layer, interlayer, and detail layer by a different scale. Secondly, the paper obtains a visual weight map by the calculation of the source image and uses the guided filter to better guide the base layer fusion. Then, it realizes the interlayer fusion through maximum local variance and realizes the detail layer fusion through the maximum absolute value of the pixel. Finally, it obtains the fused image through weight fusion. The experiment demonstrates that the proposed method shows better comprehensive performance and obtains better results in fusion for infrared and visible light images and medical images compared to the contrast method.

Abstract Background Glioblastoma multiforme (GBM) is the most malignant central nervous system tumor. Alkylating agent, temozolomide (TMZ), is currently the first-line chemotherapeutic agent for GBM. However, the sensitivity of GBM cells to TMZ is affected by many factors. And, several clinic trials, including co-administration of TMZ with other drugs, have failed in successful treatment of GBM. We have previously reported that Netrin-4 (NTN4), a laminin-like axon guidance protein, plays a protective role in GBM cell senescence upon TMZ-triggered DNA damage. However, the master regulator of NTN4 needs further elucidation. Epidermal growth factor/Epidermal growth factor receptor (EGF/EGFR) can modulate the expression of various extracellular matrix related molecules, and prevent DNA damage in GBM cells. In this study, we investigated the relationship between EGF/EGFR signaling and NTN4, and explored their effect on therapeutic efficacy in GBM cells upon TMZ treatment. Methods Co-expression analysis were performed by using the RNA sequencing data from NIH 934 cell lines and from single cell RNA sequencing data of GBM tumor. The co-expressing genes were used for GO enrichment and signaling pathway enrichment. mRNA expression of the target genes were quantified by qPCR, and cell senescence were investigated by Senescence-Associated Beta-Galactosidase Staining. Protein phosphorylation were observed and analyzed by immunoblotting. The RNA sequencing data and clinical information of TMZ treated patients were extracted from TCGA-glioblastoma project, and then used for Kaplan-Meier survival analysis. Results Analysis of RNA sequencing data revealed a potential co-expression relationship between NTN4 and EGFR. GO enrichment of EGFR-correlated genes indicated that EGFR regulates GBM cells in a manner similar to that in central nervous system development and neural cell differentiation. Pathway analysis suggested that EGFR and its related genes contribute to cell adhesion, extracellular matrix (ECM) organization and caspase related signaling. We also show that EGF stimulates NTN4 expression in GBM cells and cooperates with NTN4 to attenuate GBM cell senescence induced by DNA damage, possibly via AKT and ERK. Clinical analysis showed that co-expression of EGFR and NTN4 significantly predicts poor survival in TMZ-treated GBM patients. Conclusions This study indicates that EGF/EGFR regulates and cooperates with NTN4 in DNA damage resistance in GBM. Therefore, our findings provide a potential therapeutic target for GBM.
