Production of valuable compounds including biofuels and pharmaceutical precursors derived from microalgae has garnered significant interest. Stable production of algal biomass is essential to make the microalgal industry commercially feasible. However, one of the largest issues is severe biological contamination by predators grazing the algal biomass, resulting in the crash of outdoor cultures. In the present study, we propose a novel engineering strategy for microalgae to cope with predators. The overexpression of plant chlorophyllase (CLH) in a microalga resulted in the enhancement of resistance to the predator. This result supported our hypothesis that CLH promotes chlorophyll breakdown in the chloroplasts of the microalgae when they are digested by the predator, generating the phototoxic catabolite chlorophyllide that damages the predator. To the best of our knowledge, this is the first study to establish predator-resistant microalgae by enhancing the CLH activity.

International audience; PFS (Prime Focus Spectrograph), a next generation facility instrument on the Subaru telescope, is now being tested on the telescope. The instrument is equipped with very wide (1.3 degrees in diameter) field of view on the Subaru's prime focus, high multiplexity by 2394 reconfigurable fibers, and wide waveband spectrograph that covers from 380nm to 1260nm simultaneously in one exposure. Currently engineering observations are ongoing with Prime Focus Instrument (PFI), Metrology Camera System (MCS), the first spectrpgraph module (SM1) with visible cameras and the first fiber cable providing optical link between PFI and SM1. Among the rest of the hardware, the second fiber cable has been already installed on the telescope and in the dome building since April 2022, and the two others were also delivered in June 2022. The integration and test of next SMs including near-infrared cameras are ongoing for timely deliveries. The progress in the software development is also worth noting. The instrument control software delivered with the subsystems is being well integrated with its system-level layer, the telescope system, observation planning software and associated databases. The data reduction pipelines are also rapidly progressing especially since sky spectra started being taken in early 2021 using Subaru Nigh Sky Spectrograph (SuNSS), and more recently using PFI during the engineering observations. In parallel to these instrumentation activities, the PFS science team in the collaboration is timely formulating a plan of large-sky survey observation to be proposed and conducted as a Subaru Strategic Program (SSP) from 2024. In this article, we report these recent progresses, ongoing developments and future perspectives of the PFS instrumentation.

Exploiting the Subaru prime focus, 2394 reconfigurable fibers will be distributed in the 1.3 degree-diameter field of view. The spectrograph system has been designed with 3 arms of blue, red, and near-infrared cameras to simultaneously deliver spectra from 380nm to 1260nm in one exposure. The instrumentation has been conducted by the international collaboration managed by the project office hosted by Kavli IPMU. The team is actively integrating and testing the hardware and software of the subsystems some of which such as Metrology Camera System, the first Spectrograph Module, and the first on-telescope fiber cable have been delivered to the Subaru telescope observatory at the summit of Maunakea since 2018. The development is progressing in order to start on-sky engineering observation in 2021, and science operation in 2023. In parallel, the collaboration is trying to timely develop a plan of large-sky survey observation to be proposed and conducted in the framework of Subaru Strategic Program (SSP). This article gives an overview of the recent progress, current status and future perspectives of the instrumentation and scientific operation.

The expansion of protein sequence databases has enabled us to design artificial proteins by sequence-based design methods, such as full-consensus design (FCD) and ancestral-sequence reconstruction (ASR). Artificial proteins with enhanced activity levels compared with native ones can potentially be generated by such methods, but successful design is rare because preparing a sequence library by curating the database and selecting a method is difficult. Utilizing a curated library prepared by reducing conservation energies, we successfully designed two artificial l-threonine 3-dehydrogenases (SDR-TDH) with higher activity levels than native SDR-TDH, FcTDH-N1, and AncTDH, using FCD and ASR, respectively. Microalgal harvesting and dewatering are the main bottlenecks that need to be overcome to tap the potential of microalgae for production of valuable compounds. Water surface-floating microalgae form robust biofilms, float on the water surface along with gas bubbles entrapped under the biofilms, and have great potential to overcome these bottlenecks. However, little is known about the molecular mechanisms involved in the water surface-floating phenotype. In the present study, we analysed the genome sequence of a water surface-floating microalga Chlorococcum sp. FFG039, with a next generation sequencing technique to elucidate the underlying mechanisms. Comparative genomics study with Chlorococcum sp. FFG039 and other non-floating green microalgae revealed some of the unique gene families belonging to this floating microalga, which may be involved in biofilm formation. Furthermore, genetic transformation of this microalga was achieved with an electroporation method. The genome information and transformation techniques presented in this study will be useful to obtain molecular insights into the water surface-floating phenotype of Chlorococcum sp. FFG039.

Exploiting the Subaru prime focus, 2394 reconfigurable fibers will be distributed over the 1.3 deg field of view. The spectrograph has been designed with 3 arms of blue, red, and near-infrared cameras to simultaneously observe spectra from 380nm to 1260nm in one exposure at a resolution of ~ 1.6-2.7Å. An international collaboration is developing this instrument under the initiative of Kavli IPMU. The project recently started undertaking the commissioning process of a subsystem at the Subaru Telescope side, with the integration and test processes of the other subsystems ongoing in parallel. We are aiming to start engineering night-sky operations in 2019, and observations for scientific use in 2021. This article gives an overview of the instrument, current project status and future paths forward.

The reaction of aromatic or benzylic disulfides with 2.5 equiv of Selectfluor™ in acetonitrile/water (10:1) at room temperature efficiently produced the corresponding thiosulfonates. Conversely, the reaction of disulfides with 6.5 equiv of Selectfluor™ or thiosulfonates with 4.5 equiv of Selectfluor™ in refluxing acetonitrile/water (10:1) provided sulfonyl fluorides in high yields. Accufluor™ and FP-T300™ are also effective in preparing sulfonyl fluorides from disulfides under the similar reaction conditions. Sulfonyl chlorides or sulfonyl bromides were effectively obtained from the reaction of disulfides with 6 equiv of either N-chlorosuccinimide or N-bromosuccinimide in acetonitrile/water (10:1) at room temperature. Some other electrophilic chlorinating or brominating reagents are also able to be used instead of N-chlorosuccinimide or N-bromosuccinimide for the syntheses of sulfonyl halides from disulfides. These reactions of disulfides with electrophilic halogenating reagents are convenient methods to prepare thiosulfonates and sulfonyl halides.

Water surface-floating microalgae have great potential for biofuel applications due to the ease of the harvesting process, which is one of the most problematic steps in conventional microalgal biofuel production. We have collected promising water surface-floating microalgae and characterized their capacity for biomass and lipid production. In this study, we performed chemical mutagenesis of two water surface-floating microalgae to elevate productivity. Floating microalgal strains AVFF007 and FFG039 (tentatively identified as Botryosphaerella sp. and Chlorococcum sp., respectively) were exposed to ethyl methane sulfonate (EMS) or 1-methyl-3-nitro-1-nitrosoguanidine (MNNG), and pale green mutants (PMs) were obtained. The most promising FFG039 PM formed robust biofilms on the surface of the culture medium, similar to those formed by wild type strains, and it exhibited 1.7-fold and 1.9-fold higher biomass and lipid productivities than those of the wild type. This study indicates that the chemical mutation strategy improves the lipid productivity of water surface-floating microalgae without inhibiting biofilm formation and floating ability.

我々はスポーツ活動時の腰痛発生予防を目的に高校サッカー選手に体幹筋トレーニングを継続して行っている。体幹機能検査にはKraus-Weber test変法大阪市大方式(以下、KW)とSide-Bridge test(以下、SB)を用い、体幹筋機能と運動時腰痛発生の経時的変化を入学年度別に調査し、検討を加えたので報告する。結果は2年以上の継続した指導によって平成19年度入学の選手はSBの点数向上に伴い運動時腰痛保有者は減少する傾向となった。20年度入学の選手はKW、SBともに点数の変化がなく運動時腰痛保有者は増加した。21年度入学の選手はKW、SBともに点数は向上し、運動時腰痛保有者は減少する傾向にあった。学年別に検討したところ、KWとSBの経時的な変化と運動時腰痛保有者の変化の傾向は異なるが、KWとSBの点数に反映される体幹筋機能を向上させることが腰痛保有者の減少や予防に関与する可能性が示唆された。, We have been providing trunk muscle training for senior high school soccer players on a continuing basis for the purpose of preventing low back pain in sports activities. We report the results of our investigation and discussion on time-dependent change by school year in relation between trunk muscle function and occurrence of low back pain during exercise which are based on Kraus-Weber tests(KW) modified by the Osaka-City-University system and Side-Bridge tests(SB) as means to examine trunk muscle function. Our findings indicated a decreasing tendency of players with low back pain during exercise among students enrolled in 2007 along with improved SB scores attributable to more than 2 years of continuous coaching. Students enrolled in 2008 did not show change in KW or SB scores and the players with low back pain during exercise increased. Students enrolled in 2009 had a tendency to improve both in KW and SB scores and to decrease in the number of players with low back pain during exercise. Our examination of the results by school year suggested the potential association between improved trunk muscle function reflected in KW and SB scores, and reduction and prevention of players with low back pain despite the difference in the tendencies of time-dependent change in KW and SB scores and in the number of players with low back pain during exercise.

Hyper Suprime-Cam (HSC) is an 870 Mega pixel prime focus camera for the 8.2 m Subaru telescope. The wide field corrector delivers sharp image of 0.25 arc-sec FWHM in r-band over the entire 1.5 degree (in diameter) field of view. The collimation of the camera with respect to the optical axis of the primary mirror is realized by hexapod actuators whose mechanical accuracy is few microns. As a result, we expect to have seeing limited image most of the time. Expected median seeing is 0.67 arc-sec FWHM in i-band. The sensor is a p-ch fully depleted CCD of 200 micron thickness (2048 x 4096 15 μm square pixel) and we employ 116 of them to pave the 50 cm focal plane. Minimum interval between exposures is roughly 30 seconds including reading out arrays, transferring data to the control computer and saving them to the hard drive. HSC uniquely features the combination of large primary mirror, wide field of view, sharp image and high sensitivity especially in red. This enables accurate shape measurement of faint galaxies which is critical for planned weak lensing survey to probe the nature of dark energy. The system is being assembled now and will see the first light in August 2012.

N-Ethylmaleimide-sensitive factor (NSF) and its homologues play a central role in vesicular trafficking in eukaryotic cells. We have identified a NSF homologue in Plasmodium falciparum (PfNSF). The reported PfNSF gene sequence (GenBank accession number CAB10575) indicated that PfNSF comprises 783 amino acids with a calculated molecular weight of 89,133. The overall identities of its gene and amino acid sequences with those of rat NSF are 50.9 and 48.8%, respectively. Reverse transcription-polymerase chain reaction analysis and Northern blotting with total P. falciparum RNA indicated expression of the PfNSF gene. Polyclonal antibodies against a conserved region of NSF specifically recognized an 89-kDa polypeptide in the parasite cells. After homogenization of the parasite cells, approximately 90% of an 89-kDa polypeptide is associated with particulate fraction, suggesting membrane-bound nature of PfNSF. PfNSF was present within both the parasite cells and the vesicular structure outside of the parasite cells. The export of PfNSF outside of the parasite cells appears to occur at the early trophozoite stage and to terminate at the merozoite stage. The export of PfNSF is inhibited by brefeldin A, with 9 microM causing 50% inhibition. Immunoelectromicroscopy indicated that intracellular PfNSF was associated with organelles such as food vacuoles and that extracellular PfNSF was associated with vesicular structures in the erythrocyte cytoplasm. These results indicate that PfNSF expressed in the malaria parasite is exported to the extracellular space and then localized in intraerythrocytic vesicles in a brefeldin A-sensitive manner. It is suggested that a vesicular transport mechanism is involved in protein export targeted to erythrocyte membranes during intraerythrocytic development of the malaria parasite.

The antimalarial activity of the O-acylated bruceolide derivative, 3,15-di-O-acetylbruceolide, was evaluated against Plasmodium berghei in vivo. The concentration of 3,15-di-O-acetylbruceolide required for 50% suppression (ED50) of P. berghei in mice was 0.46 +/- 0.06 mg/kg/day, whereas bruceolide was only half as effective as 3,15-di-O-acetylbruceolide. Two antimalarial drugs used clinically, chloroquine and artemisinin, demonstrated only low activity corresponding to 1/4 and 1/12 of the ED50 value of 3,15-di-O-acetylbruceolide, respectively. These results may be helpful in the design of better chemotherapeutic bruceolides against falciparum malaria.

The effects of dietary potassium on the hemodynamics and plasma norepinephrine (NE) kinetics were studied in 10 patients with borderline hypertension. Potassium supplement (96 mEq daily for 5-7 days) induced a significant (p less than 0.05) fall in blood pressure and a slight decrease in cardiac output. Both urine volume and urinary sodium excretion increased significantly (p less than 0.05) for a first few days following the potassium supplement. The baseline values of the half-time of the rapid NE removal from plasma was significantly delayed in the hypertensive patients (1.05 +/- 0.06 min, p less than 0.05) when compared with those (0.88 +/- 0.04) in normal controls. Potassium supplement induced a significant rise in both plasma NE levels and NE outflow rate (p less than 0.01) in the hypertensive patients, while their half-times were significantly shortened (0.89 +/- 0.07 min, p less than 0.01). The pressor responsiveness to exogenously infused NE tended to diminish during the potassium supplement. These findings indicate that a high potassium intake might accelerate the slowed neuronal NE uptake in the hypertensive patients, while a potassium-induced fall in blood pressure might exert a baroreflex stimulation of NE release. As a net result, an increased NE outflow into the circulation has been confirmed. It is likely that a natriuresis-induced volume contraction might be a predominant factor responsible for the early reduction of blood pressure during the high potassium intake.

The effects of epinephrine (E) on plasma norepinephrine (NE) concentrations and cardiovascular responses before and after beta-blockade were studied in 36 patients with essential hypertension and 10 age-matched normal controls. The resting plasma NE and E were significantly higher in the borderline hypertensives (251 +/- 21 (SE) pg/ml, p less than 0.005; 57 +/- 5, p less than 0.05) than in normal controls (127 +/- 18; 38 +/- 7). An immediate rise in plasma E and NE was observed after one bolus intravenous administration of glucagon (1 mg). Then, plasma E fell rapidly to the baseline, whereas plasma NE declined much slowly. Propranolol did not affect the plasma E response to glucagon, but eliminated a prolonged rise of plasma NE. Exogenous E infusion (1.25-1.50 micrograms/min) for 30 min caused a definite rise in both plasma NE and blood pressures and lasted more than 30 min after termination of the infusion. Propranolol did not alter the time course of plasma E, but again inhibited a prolonged rise in both plasma NE and blood pressures. These findings support the view that plasma E can act physiologically as a sustained stimulator to the presynaptic beta-adrenoceptors, leading to an enhancement of NE release and a rise in blood pressure in man.

悪性褐色細胞腫はきわめてまれな疾患であるが,広範な遠隔転移を伴う場合,長期間にわたつて対症療法が必要となる.本稿では, 37才の男性例で骨,肝,リンパ節などに広範な転移をきたし,経過中にイレウス症状が持続したため,延べ10カ月間にわたりαおよびβ遮断薬の静脈内投与を必要とした症例を報告した.これらの薬物の投与量は1日に最大phentolamineが3500mg, propranololが80mgにも達し治療に難渋した.本例に合供したイレウス症状は過剰な血中カテコラミンによる腸管麻痺に起因するものと考えられ, phentolamineが他のα遮断薬や蠕動促進薬に比べて著効を示した.本薬の長期大量投与にもかかわらず,治療期間中重篤な副作用は認められなかつた.

The concentrations of unconjugated plasma dopamine (PDA) were studied in patients with various types of hypertension. Catecholamines were extracted from plasma specimens (1.0-3.0 ml) through an Amberlite CG50 (Li+-form) microcolumn and eluted by a magnesium sulfate - ethanol solution. The elute was then desalinated and deproteinized by the ethanol-treated precipitation procedure and dried in a vacuum oven at 25 degrees C. A fraction of catecholamines was assayed with the modified procedures of the COMT-mediated radio-enzymatic method. This assay system was sensitive enough to permit an accurate measurement of PDA as low as 6.0 pg per ml of plasma without any detectable contamination of the conjugated dopamine. The resting levels of PDA were 10.1 +/- 1.0 pg/ml (mean +/- SEM), 9.5 +/- 1.0 and 13.7 +/- 0.6 in patients with borderline hypertension (BH, n = 25), essential hypertension (EH, n = 22) and renovascular hypertension (RVH, n = 8), respectively. The values in EH patients were significantly smaller than those in age-matched normal controls (13.0 +/- 1.4, n = 14, p less than 0.05). Remarkably increased PDA values were observed in patients with pheochromocytoma (76.5 +/- 25.4, n = 9, p less than 0.01). Significantly raised PDA values were also found in patients with primary aldosteronism (PA, 27.8 +/- 9.0, n = 6, p less than 0.05), while their plasma norepinephrine levels (PNE, 169 +/- 39 pg/ml) tended to be lower than those of normal controls (206 +/- 20), showing an apparent dissociation between the values of PDA and PNE. Upright posture for 15 minutes induced a significant rise in PDA (p less than 0.05) in all subjects except PA patients. The postural changes of PDA, however, were invariably smaller than those of PNE (p less than 0.05). The resting values of PDA in normal, BH and EH patients showed a significant negative correlation with their mean arterial pressures (r = -0.301, n = 61, p less than 0.05) and a positive correlation with those of PNE (r = 0.381, p less than 0.01). There was no correlation between PDA and age in any group studied. These findings indicate that PDA might not be only a precursor fraction of neurotransmitters released from the sympathetic nervous system but could also represent a physiological function of the dopaminergic regulatory system. The varied but distinctive features of PDA status in various types of hypertension suggest the possibility that the peripheral dopaminergic mechanisms play an inherent role in the pathogenesis of hypertension.

The effects of sodium depletion on plasma norepinephrine (NE) kinetics were studied in 14 patients with essential hypertension using the steady state NE infusion technique on the theoretical basis of a two-compartmental open model. The calculated half time of rapid removal phase (t 1/2 alpha) of plasma NE was significantly delayed in the hypertensives (1.20 +/- 0.22 min, mean +/- SD) than in normal subjects (0.89 +/- 0.14) (p less than 0.01). The total clearance rates were comparable between the 2 groups (47 +/- 7 vs 48 +/- 11 ml/kg/min). The rates of the endogenous NE outflow into the circulation (RE-NE) were not significantly different between normal (5.29 +/- 1.68 ng/kg/min) and hypertensives (7.69 +/- 5.42), although 5 hypertensive patients (36%) showed an increased RE-NE above the normal range. The prolonged half time (t 1/2 alpha) in the hypertensives tended to be shortened after sodium depletion induced by either low sodium diet or diuretics (1.04 +/- 0.22, p less than 0.001). Both plasma NE and RE-NE were significantly increased after sodium depletion. A close correlation was observed between plasma NE and RE-NE (r = 0.907, p less than 0.001) in these subjects. These results suggest the slowed neuronal uptake of NE at the sympathetic nerve terminals in some patients with essential hypertension, occurring in part as a sodium-mediated consequence.
