Mid-infrared laser absorption spectroscopy utilizing a high-finesse optical cavity enables high precision trace analysis of gas molecules. In particular, optical detection of radiocarbon (14C) based on cavity ringdown spectroscopy using a quantum cascade laser (QCL) is gaining attention as an alternative to accelerator mass spectrometry. This paper reports a compact-packaged narrow-linewidth QCL system utilizing resonant optical feedback from an external V-shaped cavity. Based on frequency noise analysis, the derived laser linewidth is 44 kHz for 100 μs integration time with the capability to perform seamless frequency scanning around 10 GHz. We installed this laser system within a table-top cavity ringdown spectrometer for 14CO2. A single-shot detection limit of 1.2 × 10−9 cm−1 Hz−1/2 leading to a detectable abundance evaluated from a noise analysis of 0.2 in fraction modern 14C for a 10-s averaging time was achieved. This capability of rapid analysis for 14CO2 is suitable for various applications requiring trace 14C analysis.

ATP2B1 is a gene associated with hypertension. We reported previously that mice lacking ATP2B1 in vascular smooth muscle cells (VSMC ATP2B1 KO mice) exhibited high blood pressure and increased intracellular calcium concentration. The present study was designed to investigate whether lack of the ATP2B1 gene causes a higher response to calcium channel blockers (CCBs) than to other types of anti-hypertensive drugs. Both VSMC ATP2B1 KO and control mice were administered anti-hypertensive drugs while monitoring blood pressure shifts. We also examined the association of nitric oxide synthase (NOS) activity in those mice to investigate whether another mechanism of hypertension existed. VSMC ATP2B1 KO mice exhibited significantly greater anti-hypertensive effects with a single injection of nicardipine, but the effects of an angiotensin II receptor blocker (ARB), an α-blocker and amlodipine on blood pressure were all similar to control mice. However, long-term treatment with amlodipine, but not an ARB, significantly decreased the blood pressure of KO mice compared with control mice. Both mRNA and protein expression levels of the L-type calcium channel were significantly upregulated in KO VSMCs. There were no alterations in neural NOS protein expression of VSMCs or in urinary NO production between the two groups. VSMC ATP2B1 KO mice had a higher response to CCBs for blood pressure-lowering effects than other anti-hypertensive drugs. These results mean that increased intracellular calcium concentration in VSMCs due to lack of ATP2B1 and subsequent activation of L-type calcium channels mainly affects blood pressure and suggests increased susceptibility to CCBs in this type of hypertension.

A linewidth reduction of a distributed feedback quantum cascade laser (DFB-QCL) based on optical feedback for a mid-infrared (MIR) cavity ring down spectroscopy (CRDS) 14C spectrometer is presented. A cat-eye reflector as well as a path-length enhancement by a Herriott cell were employed for a compact optical setup. The laser linewidth was evaluated by monitoring the beat frequency between the DFB-QCL and a MIR optical frequency comb (OFC). The linewidth reduction by optical feedback was clearly observed although slow frequency drifts caused by environmental changes were still visible. A low-bandwidth beat-note lock to the OFC was conducted for long-term stabilization as well as for precise frequency scanning and thereby 14C measurement by MIR CRDS with optical feedback was demonstrated successfully.

In the 'Millennium Genome Project', we identified ATP2B1 as a gene responsible for hypertension through single-nucleotide polymorphism analysis. The ATP2B1 gene encodes the plasma membrane calcium ATPase isoform 1, which contributes to the maintenance of intracellular calcium homeostasis by removing calcium ions.Since ATP2B1 knockout mice are reported to be embryo-lethal, we generated systemic heterozygous ATP2B1 null (ATP2B1(+/-)) mice, and evaluated the implication of ATP2B1 in blood pressure.ATP2B1(+/-) mice revealed significantly higher SBP as measured by a radiotelemetric method. Phenylephrine-induced vasoconstriction was significantly increased in vascular rings from ATP2B1(+/-) mice, and the difference in this contraction disappeared in the presence of a nitric oxide synthase (NOS) inhibitor. Vasorelaxation to acetylcholine was significantly attenuated in vascular rings from ATP2B1(+/-) mice. In addition, cultured endothelial cells of ATP2B1(+/-) mice showed that the phosphorylation (Ser-1177) level of endothelial NOS protein was significantly lower, and nitric oxide production in endothelial cells and aorta was lower compared with those in control mice. In contrast, neural NOS expression in vascular smooth muscle cells from ATP2B1(+/-) mice and control mice were not significantly different.These results suggest that decreased ATP2B1 gene expression is associated with impaired endothelial NOS activity and nitric oxide production, and the ATP2B1 gene plays a crucial role in the regulation of blood pressure.

Debris flows form deposits when they reach an alluvial fan until they eventually stop. However, houses located in the alluvial fan might affect the debris flow flooding and deposition processes. Few previous studies have considered the effects of houses on debris flow flooding and deposition. This study conducted model experiments and numerical simulations using the Kanako2D debris flow simulator to determine the influence of houses on debris flow flooding and deposition. The model experiments showed that when houses are present, the debris flow spreads widely in the cross direction immediately upstream of the houses, especially when the flow discharge is large or the grain size is small. Houses located in the alluvial fan also influence the deposition area. The presence of houses led to flooding and deposition damage in some places and reduced the damage in others. The simulation also demonstrated the influence of houses. Both the model experiment and the simulation showed that houses change the flooding and deposition areas.

We had a deluge in July 2012 in the northern Kyushu district with intense rainfall of 800mm and 108mm/hr. This intensity yielded countless traces of debris flow and slope failures that induced tremendous damage and causalities in the area. Hence, several field investigations and reconnaissance tasks were conducted by the society to delve into this sediment-related disaster. The various results and the information obtained through this investigation were reported as the primary report, mentioning the damage, geologic-geomorphologic features and hydraulic characteristics of the debris flows, vegetation effects, and the efficiency of Sabo facilities in other scenes of this disaster.

Blood pressure is regulated by extrinsic factors including noradrenaline, the sympathetic neurotransmitter that controls cardiovascular functions through adrenergic receptors. However, the fine-tuning system of noradrenaline signaling is relatively unknown. We here show that l-3,4-dihydroxyphenylalanine (L-DOPA), a precursor of catecholamines, sensitizes the vascular adrenergic receptor alpha1 (ADRA1) through activation of L-DOPA receptor GPR143. In WT mice, intravenous infusion of the ADRA1 agonist phenylephrine induced a transient elevation of blood pressure. This response was attenuated in Gpr143 gene-deficient (Gpr143-/y) mice. Specific knockout of Gpr143 in vascular smooth muscle cells (VSMCs) also showed a similar phenotype, indicating that L-DOPA directly modulates ADRA1 signaling in the VSMCs. L-DOPA at nanomolar concentrations alone produced no effect on the VSMCs, but it enhanced phenylephrine-induced vasoconstriction and intracellular Ca2+ responses. Phenylephrine also augmented the phosphorylation of extracellular signal-regulated kinases in cultured VSMCs from WT but not Gpr143-/y mice. In WT mice, blood pressure increased during the transition from light-rest to dark-active phases. This elevation was not observed in Gpr143-/y mice. Taken together, our findings provide evidence for L-DOPA/GPR143 signaling that exerts precursor control of sympathetic neurotransmission through sensitizing vascular ADRA1.

Although the association between the Ser9Gly polymorphism of the dopamine D3 receptor gene (DRD3) and schizophrenia has been investigated by many research groups, it is not known whether the Ser9Gly polymorphism alone or a variation in linkage disequilibrium may effect susceptibility to schizophrenia. We searched the 5' region of the DRD3 gene and found three novel polymorphisms: -712G/C, -205A/G, and Ala38Thr. The Ala38Thr polymorphism is located in the first transmembrane region and is conserved in the monkey, mouse, and rat. Case-control comparisons in 153 Japanese schizophrenia patients and 122 Japanese controls did not suggest an association between Ala38Thr and schizophrenia. However, there was a marginally significant association between the Ser9 allele of the Ser9Gly polymorphisms and schizophrenia (P = 0.02). Furthermore, there was a highly significant association between haplotypes of the -712G/C, -205A/G, and Ser9Gly polymorphisms and schizophrenia (P = 0.0007, corrected P = 0.007). These positive findings were replicated in an additional 99 Japanese schizophrenia patients and 132 controls (P = 0.04 and 0.0004, respectively). The most allelic differences of the Ser9Gly polymorphism between patient and control groups arose from the chromosome carrying specific alleles of the other three polymorphisms. This study indicates unknown variant(s) in linkage disequilibrium with the DRD3 haplotypes associated with schizophrenia.

Polymorphism in the human dopamine D4 receptor gene (DRD4) exon III has been associated in some but not all studies of the human personality trait of Novelty Seeking. We searched for polymorphisms in the 5′ region of DRD4 and identified six polymorphisms as follows: −1217G Ins/Del, −809G/A, −616C/G, −603T Ins/Del, −602(G)8–9, and −521C/T. Associations between these polymorphisms and personality traits measured by the Temperament and Character Inventory (TCI) were investigated in 86 healthy Japanese volunteers. The −521C/T polymorphism was significantly associated with Novelty Seeking (P = 0.0001). Subjects with the C/C genotype exhibited the highest Novelty Seeking scores and those with the T/T genotype exhibited the lowest. A transient expression method revealed that the T variant of the C-521T polymorphism reduces transcriptional efficiency. The present study suggests a contribution of dopamine D4 receptor availability to individual differences in Novelty Seeking behavior.

The human dopamine D4 receptor gene (DRD4) is an important candidate gene for schizophrenia. We identified a novel -521C>T polymorphism in the 5'-promoter region of DRD4. A transient expression method revealed that the T allele of this polymorphism reduces the transcriptional efficiency by 40% compared with the C allele. This polymorphism is of interest because of reported elevation of D4-like sites and DRD4 mRNA in the postmortem schizophrenic brain. The C allele frequency was significantly higher in 252 Japanese schizophrenics (0.48) than in 269 Japanese controls (0.41) (p = 0. 02) [odds ratio = 1.35 (95% confidence interval 1.05 - 1.72)]. Although the association is weak and should be considered tentative until other studies replicate it, this polymorphism provides a tool with the potential to examine whether DRD4 is related to susceptibility to and neuroleptic response in schizophrenia.

We report a high-resolution microscope system for imaging ultracold ytterbium atoms trapped in a two-dimensional optical lattice. By using the ultraviolet strong transition combined with a solid immersion lens and high-resolution optics, our system resolved individual sites in an optical lattice with a 544-nm spacing. Without any cooling mechanism during the imaging process, the deep potential required to contain the atoms was realized using a combination of a shallow ground-state and a deep excited-state potentials. A procedure to estimate S parameters and far-field radiation power from structural parameters of coupled-resonant wireless power transfer in kHz range is discussed. Helical antenna is modeled by series connection of loop antennas. All combinations of loop antennas are taken into account for calculation of self and mutual inductance. Stray capacitance between neighboring loops and ohmic loss are also regarded. Numerical demonstration shows that the proposed procedure has calculation accuracy as same as method of moment (MoM) calculation, when wire length is much smaller than the wavelength. Calculation time for the proposed procedure was 0.5 seconds whereas MoM requires 200 minutes.

A procedure to calculate parameters of an equivalent circuit of the magnetic-coupled resonant wireless power transfer is discussed. Equivalent circuit parameters are calculated from the mechanical and material parameters of the wireless power transfer structure. Both direct-fed type and indirect-fed type structure are considered. MoM calculation shows that the equivalent circuit has capability to calculate S parameters and radiation power correctly.

An equivalent circuit of wireless power transmission is discussed. We demonstrate that an input impedance of the wireless power transmission is expressed by a simple equivalent circuit. The equivalent circuit spells out how the resonant frequency is determined. The parameters in the equivalent circuit is found by using genetic algorithm so that the input impedance calculated by method of moment (MoM) agreed with one calculated by the equivalent circuit. The equivalent circuit expression will provide analytical approach of consideration on power transmission efficiency.

Background Cyclosporine and prednisolone combination therapy has been used in the treatment of minimal change nephrotic syndrome (MCNS). However, few studies have evaluated the efficacy of cyclosporine combined with intravenous methylprednisolone pulse therapy (MPT) as a first-line treatment for new-onset MCNS. We conducted a retrospective clinical study to evaluate the efficacy and safety of cyclosporine combined with MPT and oral prednisolone for new-onset MCNS in adults. Methods Forty-six adult patients with biopsy-proven MCNS were analyzed retrospectively. This study included three groups. Group 1 (n = 17) was treated with intravenous MPT (0.5 or 1.0 g/day for 3 days) followed by oral cyclosporine (2–3 mg/kg/day) and prednisolone (30 mg/day). Group 2 (n = 15) was treated with intravenous MPT followed by oral prednisolone (0.4–0.8 mg/kg/day). Group 3 (n = 14) was treated with oral prednisolone (0.6–1.0 mg/kg/day) alone. Background Tolvaptan, a vasopressin V2 receptor blocker, has a diuretic effect for patients with heart failure. However, there were a few data concerning the effects of tolvaptan in patients with chronic kidney disease (CKD). Methods We retrospectively analyzed 21 patients with chronic heart failure and CKD. Tolvaptan was co-administered with other diuretics in-use, every day. We compared clinical parameters before and after the treatments with tolvaptan. Furthermore, we examined the correlations between baseline data and the change of body weight. Results Tolvaptan decreased the body weight and increased the urine volume (p = 0.001). The urine osmolality significantly decreased throughout the study period. Urinary Na/Cr ratio and FENa changed significantly after 4 h, and more remarkable after 8 h (p = 0.003, both). Serum creatinine increased slightly after 1 week of treatment (p = 0.012). The alteration of body weight within the study period correlated negatively with the baseline urine osmolality (r = −0.479, p = 0.038), the baseline urine volume (r = −0.48, p = 0.028), and the baseline inferior vena cava diameter (IVCD) (r = −0.622, p = 0.017). Hyponatremia was improved to the normal value, and the augmentations of the sodium concentration were negatively associated with the basal sodium levels (p = 0.01, r = −0.546). Conclusions Tolvaptan is effective in increasing diuresis and improved hyponatremia, even in patients with CKD. The baseline urine osmolality, urine volume, and IVCD may be useful predictors for diuretic effects of tolvaptan.
