2. Earthworm species and density in semi-natural grasslands on rice paddy levees in Japanese satoyama

Abstract

Earthworms contribute to the sustainability of food webs in the semi-natural grasslands of levees at paddy margins, which are typical components of satoyama, the traditional agricultural landscapes of Japan. Thus far, few studies have focused on earthworm fauna of paddy levees in satoyama. In this study, we investigated earthworm fauna and regional and monthly changes in earthworm density. We found at least 11 species of earthworms living within levees on Sado Island, central Japan. Two endogeic species, Amynthas corticis (Megascolecidae) and Eisenia japonica (Lumbricidae), were dominant in terms of number of adult individuals; these two species appeared in all study regions. We also estimated an average of ~57.4 individuals/m2 for all stages of earthworms in levees, suggesting that rice paddy levees have relatively-high earthworm density. However, such tendencies differed depending on the region. In addition, monthly changes in density were observed in the topsoil of the levees. Our results imply that differences in earthworm assemblages amongst regions and months may influence the availability of food resources for various animals inhabiting satoyama.

Abstract

Many studies have reported the complete mitochondrial genome sequences of Chinese Megascolecidae earthworms, however, there have been no reports on sequences originating from Japanese Megascolecidae earthworms. In this study, we determined complete mitochondrial genome sequences of two Japanese earthworms belonging to the Pheretima complex within the Megascolecidae family. Metaphire hilgendorfi is one of the most common earthworms in Japan and Amynthas yunoshimensis was found to be morphologically similar to M. hilgendorfi. The complete mitochondrial genomes of M. hilgendorfi (15,186 bp; LC573968) and A. yunoshimensis (15,109 bp; LC573969) contained typical 13 protein coding genes, 22 tRNA genes, and 2 rRNA genes. The phylogenetic analysis revealed that these two species were sister species. Therefore, our findings will further contribute to phylogenetic and genetic diversity analyses of Megascolecidae earthworms.

Abstract

The distribution and species assembly of Japanese Protura collected from forest soils were examined using published databases and statistical analysis. We used records from 3110 sites where 71 taxa were found. The species richness of Protura ranged from one to 16 species, and TWINSPAN analysis of regional populations indicated that the northern and southern regions could be separated into distinct groups. Three major species assemblages were identified by cluster analysis from points containing more than six species. Three groups reflected historical migration from northern and western linkages to the Asian continent. The northern assemblage showed a negative correlation to winter minimum temperature and the other two assemblages exhibited relationships to precipitation and temperature. Vegetation was not responsible for proturan distribution. These results suggest that the history of Protura invasion explains the biogeography of these soil-based, small arthropods and also that climate change will induce a shift in the distribution of species irrespective of changes in vegetation type.

Abstract

The invasion of jumping worms, a small group of pheretimoid earthworm species from Asia, has increasingly become an ecological, environmental and conservation issue in forest ecosystems and urban-suburban landscapes around the world. Their presence is often noticed due to their high abundance, distinctive “jumping” behavior, and prominent granular casts on the soil surface. Although they are known to affect soil carbon dynamics and nutrient availability, no single paper has summarized their profound impacts on soil biodiversity, plant community, and animals of all trophic groups that rely on soil and the leaf litter layer for habitat, food, and shelter. In this study, we summarize the biology, invasion, and ecological impacts of invasive jumping worms across North America. We highlight potential impacts of this second wave of earthworm invasion, contrast them with the preceding European earthworm invasion in temperate forests in North America, and identify annual life cycle, reproductive and cocoon survival strategies, casting behavior and co-invasion dynamics as the key factors that contribute to their successful invasion and distinct ecological impacts. We then suggest potential management and control strategies for practitioners and policy makers, underscore the importance of coordinated community science projects in tracking the spread, and identify knowledge gaps that need to be addressed to understand and control the invasion.

Abstract

Agricultural practices often lead to a decrease in soil carbon (C), which often begins shortly after a natural system is converted to arable land. Conservation agriculture practices, such as no-till farming and weed mulching (NWM), may help maintain soil C. Increases in plant biomass through fertilization can also increase soil C. However, there is limited knowledge of the initial C dynamics following land conversion followed by conservation agriculture and fertilizer application, and of the soil biological and physical processes associated with these dynamics. Here, we studied the effects of two tillage practices (conventional tillage and NWM), fertilizer, and their interaction on earthworm abundance, soil aggregates, and soil C at two years after land conversion from grassland to agricultural land. We conducted a 2 × 2 factorial experiment (tillage/NWM and fertilized/not fertilized) in an experimental field in central Japan. We found that soil bulk densities and C concentrations at 0–5 cm depth were lower and higher, respectively, in the NWM plots than in the tillage plots at two years after the start of land conversion. After two years, earthworms were found only in the NWM plots. The percentage of macroaggregates (> 2 mm) was higher, and larger amounts of C were stored, in macroaggregates in NWM plots than in tillage plots. Amounts of crop biomass, roots, and plant residues were larger in the fertilized NWM plots. In the long term, such organic matter could accumulate in the form of macroaggregates, thereby further increasing soil C stock. Overall, we found that the soil biological and physical parameters that potentially control soil C responded to agricultural practices within a short period. The maintenance of biological activities by NWM and increased organic matter input by crop residues through fertilization can enhance soil C accumulation, which could have significant consequences for sustainable crop yields.

Abstract

The magnitude of the change in tree growth caused by climate warming can be highly dependent on biological context, including factors such as the presence of associated decomposers and their behaviors. We test the hypothesis that 1) the presence of earthworms modifies the response of soil inorganic N and tree growth to warming via the change of cast production; and furthermore 2) the type of soil as food for earthworm differentiate the earthworm response and its effects on the plant-soil system. We conducted a microcosm experiment rearing a geophageous earthworm species (Aporrectodea rosea) and birch (Betula ermanii) seedlings with two types of soil (sedimentary soil and serpentine soil) under two temperature regimes (ambient and ambient +3.3 °C). While the warming increased the cast production by the earthworms in the serpentine soil, it did not influence cast production in the sedimentary soil. The net nitrogen mineralization rate and the net inorganic N production showed similar dynamics to cast production under the warmer conditions, although the net nitrification rate did not. A significant increase in tree height from warming was observed only in the serpentine soil with earthworms, and the tree height was positively correlated with inorganic N production. The context-dependent change of soil N dynamics to warming could be caused by differential feeding activity of earthworm, and it results in the modification of the growth characteristics of birch seedlings.

Abstract

The radiological risks to the Tohoku hynobiid salamanders (class Amphibia), Hynobius lichenatus due to the Fukushima Dai-ichi Nuclear Power Plant accident were assessed in Fukushima Prefecture, including evacuation areas. Aquatic egg clutches (n = 1 for each sampling date and site; n = 4 in total), overwintering larvae (n = 1-5 for each sampling date and site; n = 17 in total), and terrestrial juveniles or adults (n = 1 or 3 for each sampling date and site; n = 12 in total) of H. lichenatus were collected from the end of April 2011 to April 2013. Environmental media such as litter (n = 1-5 for each sampling date and site; n = 30 in total), soil (n = 1-8 for each sampling date and site; n = 31 in total), water (n = 1 for each sampling date and site; n = 17 in total), and sediment (n = 1 for each sampling date and site; n = 17 in total) were also collected. Activity concentrations of (134)Cs + (137)Cs were 1.9-2800, 0.13-320, and 0.51-220 kBq (dry kg) (-1) in the litter, soil, and sediment samples, respectively, and were 0.31-220 and0.29-40 kBq (wet kg)(-1) in the adult and larval salamanders, respectively. External and internal absorbed dose rates to H. lichenatus were calculated from these activity concentration data, using the ERICA Assessment Tool methodology. External dose rates were also measured in situ with glass dosimeters. There was agreement within a factor of 2 between the calculated and measured external dose rates. In the most severely contaminated habitat of this salamander, a northern part of Abukuma Mountains, the highest total dose rates were estimated to be 50 and 15 μGy h(-1) for the adults and overwintering larvae, respectively. Growth and survival of H. lichenatus was not affected at a dose rate of up to 490 μGy h(-1) in the previous laboratory chronic gamma-irradiation experiment, and thus growth and survival of this salamander would not be affected, even in the most severely contaminated habitat in Fukushima Prefecture. However, further studies of the adult salamanders may be required in order to examine whether the most severe radioactive contamination has any effects on sensitive endpoints, since the estimated highest dose rate to the adults exceeded some of the guidance dose rates proposed by various organisations and programmes for the protection of amphibians, which range from 4 to 400 μGy h(-1). Conversely, at one site in Nakadori, a moderately contaminated region in Fukushima Prefecture, the dose rate to the adult salamanders in spring of 2012 was estimated to be 0.2 μGy h(-1). Estimated dose rates to the overwintering larvae in spring of 2012 were 1 and 0.2 μGy h(-1) at one site in Nakadori, and in Aizu, a less contaminated region in Fukushima Prefecture, respectively. These results suggest that there is a low risk that H. lichenatus will be affected by radioactive contamination in these districts, though further studies on dose rate estimation are required for definitive risk characterisation.

12. Changes in water stable aggregate and soil carbon accumulation in a no-tillage with weed mulch management site after conversion from conventional management practices

Abstract

Soil carbon (C) is crucial for maintaining soil functions, and it increases after conversion of an agricultural field from conventional tillage management to no-tillage management due to decreasing human-induced soil disturbance and the modification of soil structure through ecosystem engineers such as earthworms. To improve soils and prevent degradation, understanding the effects of no-tillage management over time in changing water stable aggregates (WSA) and soil C is important. We investigated the changes in WSA and soil C at a site in Akame, Mie, Japan, operating a no-tillage with weed mulch management (NWM) system over a chronosequence from 0 to 17 years after conversion from conventional tillage practices (NWM for 0, 5, 10, 15, and 17 years). We measured weed aboveground biomass, litter accumulation, and root and earthworm density and biomass, and analyzed the WSA and C of bulk soil and each WSA size fraction. Weed aboveground biomass increased with site age, while litter accumulation, root biomass (soil depth of 0–4 cm), and earthworm density and biomass did not appear to be related to site age. Endogeic earthworm density and biomass tended to increase at year 5 of NWM compared to year 0. The WSA > 2 mm and soil C stock in WSA of > 2 mm increased over time under NWM at a soil depth of 0–15 cm, while the soil C stock of 0.25–1-mm WSA decreased at soil depths of 0–5 cm. The total soil C accumulation rate was 60 g C m − 2 yr − 1 at a soil depth of 0–25 cm over the NWM chronosequence. Therefore, our results indicated that by adopting NWM, C inputs to the soil from weed aboveground biomass, as well as increases in the WSA of > 2 mm, might be responsible for soil C sequestration.

Abstract

In arthropods, both diapause duration and ability to produce eggs in early adult life (early fecundity) are important life-history traits for successful settling in a new habitat. In herbivorous spider mites (Acari: Tetranychidae), inbreeding frequently occurs because new colonies are established by only one or a few females. In the present study, we investigated the impact of inbreeding on the phenotypic variance in diapause duration and early fecundity in the Kanzawa spider mite, Tetranychus kanzawai Kishida. Diapause duration was compared between the inbreeding treatment (strongly inbred strains) and the control (strains immediately taken from the stock culture) under winter-mimicking laboratory conditions. The variance in diapause duration was smaller in the inbreeding treatment than in the control, though the magnitude of decrease of variance was less than expected. On the other hand, early fecundity did not show any reduction of variance. The present results revealed that inbreeding reduces phenotypic variation, as expected in theory.

Abstract

The comparative morphology and anatomy of the leaves of the rheophytic Rhododendron ripense and the closely related inland species Rhododendron macrosepalum were examined. The leaf of R. ripense is thinner than that of R. macrosepalum, with leaf length to width ratios (leaf index) of 2.92 and 1.91, respectively. Moreover, the leaf of R. ripense consists of fewer cells than the leaf of R. macrosepalum, suggesting stenophyllization of R. ripense caused by the decreased number of cells. In addition, leaf thickness and the number of stomata per leaf of R. ripense were significantly greater than those of R. macrosepalum, but the density of the short glandular pilose hairs on the leaf of R. ripense was lower. The observed morphological differences between the two species may be explained by certain aspects of the riparian environment, such as high irradiation and frequent flooding after heavy rainfall, to which R. ripense is exposed.

Abstract

Arisaema tosaense Makino develops either 1 or 2 leaves during both the male and female phases. To clarify the relationship between growth stage (sexual stage) and leaf number in this species, we conducted morphological analysis using A. tosaense, A. japonicum Bulme, A. sikokianum French. & Sav., and A. ringens (Thunb.) Schott. We found that increase in size of pseudostem of these Arisaema species is correlated to changes in the growth stage from sterile to male and from male to female phases. In particular, 1-leaf individuals of A. tosaense appeared at the beginning of both male and female phases, suggesting that they contribute to the earlier appearance of the fertile phase from sterile stages

Abstract

Pilophorus typicus (Distant) (Heteroptera: Miridae) is a predatory bug occurring in East, Southeast, and South Asia. Because the active stages of P. typicus prey on various agricultural pest insects and mites, this species is a candidate insect as an indigenous natural enemy for use in biological control programs. However, the mass releasing of introduced natural enemies into agricultural fields may incur the risk of affecting the genetic integrity of species through hybridization with a local population. To clarify the genetic characteristics of the Japanese populations of P. typicus two portions of the mitochondrial DNA, the cytochrome oxidase subunit I (COI) (534 bp) and the cytochrome B (cytB) (217 bp) genes, were sequenced for 64 individuals collected from 55 localities in a wide range of Japan. Totals of 18 and 10 haplotypes were identified for the COI and cytB sequences, respectively (25 haplotypes over regions). Phylogenetic analysis using the maximum likelihood method revealed the existence of two genetically distinct groups in P. typicus in Japan. These groups were distributed in different geographic ranges: one occurred mainly from the Pacific coastal areas of the Kii Peninsula, the Shikoku Island, and the Ryukyu Islands; whereas the other occurred from the northern Kyushu district to the Kanto and Hokuriku districts of mainland Japan. However, both haplotypes were found in a single locality of the southern coast of the Shikoku Island. COI phylogeny incorporating other Pilophorus species revealed that these groups were only recently differentiated. Therefore, use of a certain population of P. typicus across its distribution range should be done with caution because genetic hybridization may occur.

Morphologically putative introgressive hybrids of Arisaema sikokianum Franch. et Sav. and A. tosaense Makino were newly found in Kochi and Tokushima Prefectures in Japan. All the individuals have the same morphological characteristics as A. tosaense excluding a purple spathe. Molecular analysis using PCR-RFLP of internal transcribed spacer (ITS) in nuclear DNA (nrDNA) indicates that these putative introgressive hybrids have the same pattern as A. tosaense. Moreover, the sequences of chloroplast DNA (cpDNA) of the putative introgressive hybrids were identical to A. sikokianum from Kochi Prefecture and A. tosaense from Tokushima Prefecture. The results suggest that the plants are introgressive hybrids of A. sikokianum and A. tosaense and that they have highly exchanged genes with A. tosaense.

Abstract

Although earthworms are hermaphroditic animals with biparental sexual reproduction, some parthenogenetic species have been found. Evolutionary trends in parthenogenetic earthworms revealed a reduction in the reproductive organs. To clarify the phylogenetic relationships of parthenogenetic earthworms with different degree of degraded reproductive organs, we conducted a morphological analysis of the reproductive organs and molecular phylogenetic analyses of Amynthas vittatus which usually degraded a part of reproductive organs. Morphological analysis revealed that almost all individuals collected around Mt. Aobayama, Sendai city of northeastern Japan, possessed male pores, while individuals collected from areas located across Hirose River did not. Phylogenetic analysis using mitochondrial DNA sequences of 48 individuals representing 20 populations indicated that almost all individuals collected around Mt. Aobayama belonged to a different lineage from the other populations collected around Sendai, and that almost all individuals collected from across Japan belonged to the latter lineage. We suggest that the difference in the male pore possession rate was caused by histories of each population, but the A. vittatus population found on Mt. Aobayama belongs to a different lineage as compared to the other Japanese populations and not the primitive population. Thus, the parthenogenetic earthworm A. vittatus has undergone at least two morphological evolutionary processes.

To describe the phylogeographic structures of Asparagus schoberioides Kunth (Asparagaceae) in Japan, we investigated its nucleotide sequence variations with respect to its geographic distribution pattern. Sequencing of the internal transcribed spacer (ITS) 1 region in 29 samples of A. schoberioides revealed 20 polymorphic nucleotide sites. As a result, the 29 samples of A. schoberioides fell into 15 distinct haplotypes and phylogenetic analyses revealed these haplotypes fell into two major clades, Clade 1 and Clade 2. The haplotypes of Clade 1 were distributed chiefly along the Pacific Ocean side of Japan, while those of Clade 2 occurred mainly along the Japan Sea side. This result suggests that A. schoberioides has migrated via two routes in Japan.

To determine the eVolutionary history of Asparagus cochinchinensis (Asparagaceae), we investigated the geographic pattern of its nucleotide sequence variations in Japan, Taiwan, South Korea and China. We found 21 polymorphic nucleotide sites by sequencing the internal transcribed spacer (ITS) region, which gave rise to a total of 15 haplotypes, labeled A to O. The A-type was found only in inland China (Guizhou and Sichuan), and the other haplotypes in China extended to several lineages; therefore, A. cochinchinensis may have its origin in the interior of China. The I-type has large distribution area and it also experienced a quick expansion in relative recent years. Haplotype differentiation was observed between the eastern and western side of the Central Mountain Ridge in Taiwan. Two lineages were found in Japan, one in the Yaeyama Islands and the other in remaining areas of Japan, implying that A. cochinchinensis independently colonized in Japan at least twice.

A morphologically intermediate plant between Arisaema sikokianum Franch. et Sav. and A. serratum (Thunb.) Schott has been newly found in Kochi Prefecture, Shikoku, Japan. The putative hybrid has the intermediate morphological characteristics of the parental species. Molecular analysis using PCR-RFLP of internal transcribed spacer (ITS) in nuclear DNA (nrDNA) indicates that the putative hybrid has a combined pattern of the two putative parent species. Moreover, the sequence result of chloroplast DNA (cpDNA) of the putative hybrid was identical to that of A. sikokianum. These results suggest that the putative hybrid is a hybrid between A. sikokianum and A. serratum and that it was formed by interactive gene exchanging via pollens from A. serratum to A. sikokianum. It is the first record of a hybrid between A. sikokianum and A. serratum.

To clarify the historical phylogeography of a Japanese earthworm, nucleotide sequence variations of Metaphaire sieboldi were investigated with respect to its geographic distribution. Sequencing of mitochondrial DNA (mtDNA), including the cytochrome oxidase subunit I (COI) region and the 16S rDNA region, of 72 samples including 4 outgroup taxa, and phylogenetic analyses using neighbour-joining (NJ) and Bayesian inference (BI) methods indicated that M. sieboldi forms a monophyletic group. This monophyletic group was divided into seven subgroups; most subgroups comprised individuals from several areas, except for southern Shikoku and southern Kyushu. The phylogenetic tree demonstrated that the Shikoku and Kinki populations were paraphyletic at the basal node, and individuals from Kyushu and western Shikoku were positioned at the advanced group. Estimated phylogeographic events based on mtDNA analyses include (1) M. sieboldi originated in Shikoku and/or Kinki, (2) individuals from Chugoku and Kyushu have a common ancestor from Shikoku, and (3) individuals from western Shikoku originated from Kyushu progenitors (about 0.4–0.5 mya) after separation by rising sea levels.

Abstract

Pilophorus typicus (Heteroptera: Miridae) is a candidate insect for biological control against agricultural pests such as aphids, whiteflies, and spider mites. This species has two genotypes in Japan, i.e., the northern and southern types, but it is difficult to differentiate them because of few morphological characters separating them. Artificial migration across the distribution range of each genotype may incur ecological and genetic risks. To conduct a prompt identification of the two genotypes, we developed the method of PCR-RFLP using the cytochrome oxidase subunit I (COI) region on mitochondrial DNA. The PCR product of the southern type yielded two DNA bands by the action of the restriction enzyme of Psi I, whereas that of the northern type gave only a single band on electrophoresis. Moreover, the PCR-RFLP using samples from many localities revealed that the restriction site of Psi I is well conserved within each type. These results suggest that the PCR-RFLP of the COI region provides the accurate and easy discrimination of the two genotypes of P. typicus.

Abstract

Information from phylogenetic trees implies zygomorphic flowers have evolved independently from their actinomorphic ancestors several times. There are various types of zygomorphic flowers in the legume subfamilies Caesalpinioideae and Papilionoideae. To understand the relationship between the cell number and size in the petals of zygomorphic flowers, the comparative morphology and anatomy of flower petals were examined in Cercis chinensis and Caesalpinia decapetala var. japonica in Caesalpinioideae, and Lespedeza formosa subsp. velutina and Pueraria lobata in Papilionoideae. Increased petal size is correlated to a higher number and/or size of the cells of the petal in Caesalpinioideae. The characteristics flower morphology of the Papilionoideae has evolved by increased cell numbers, although the cell size was the smallest. Papilionoideae has characteristic papilionoid flowers consisting of five specialised petals. These characteristic morphologies were generated from the complicated changing pattern of cells in petals in the course of legume diversification.

A comparative study of leaf morphology of Aster hispidus and its coastal variety, A. hispidus var. insularis revealed that the latter has larger and thicker leaves than the former. The anatomical analysis indicated that the cell number of the leaf was lower but the cell size was larger in A. hispidus var. insularis. The reduction of cell number in the leaves of A. hispidus var. insularis was compensated by an increase in cell volume while the expansion of cells could be considered an adaptive strategy to coastal environments that allowed saving of water in the plant body. Salinity and drought seemed to have played a crucial role in the ecotypic differentiation.

To investigate whether coelomic fluid secreted by earthworms can be a noninvasive source of DNA, we amplified and sequenced DNA extracted from the coelomic fluid and muscle tissue of eight worms. The sequences obtained using DNA extracted from both sources were identical. All cytochrome c oxidase subunit I (COI) mitochondrial DNA sequences, including those retrieved from GenBank, formed a monophyletic group of Metaphire sieboldi. The results indicate that we successfully extracted total DNA from coelomic fluid secreted by earthworm.

Abstract

Earthworms are an important soil taxon as ecosystem engineers, providing a variety of crucial ecosystem functions and services. Little is known about their diversity and distribution at large spatial scales, despite the availability of considerable amounts of local-scale data. Earthworm diversity data, obtained from the primary literature or provided directly by authors, were collated with information on site locations, including coordinates, habitat cover, and soil properties. Datasets were required, at a minimum, to include abundance or biomass of earthworms at a site. Where possible, site-level species lists were included, as well as the abundance and biomass of individual species and ecological groups. This global dataset contains 10,840 sites, with 184 species, from 60 countries and all continents except Antarctica. The data were obtained from 182 published articles, published between 1973 and 2017, and 17 unpublished datasets.
