We conducted two-year seroprevalence surveys of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) antibodies among outpatients and healthcare workers (HCWs) at Ehime University Hospital. Data were collected for outpatients and HCWs in June 2020 (1st survey), December 2020 (2nd survey), July 2021 (3rd survey), and December 2021 (4th survey), focusing on demographics, occupation, and the seroprevalence of anti-SARS-CoV-2 antibodies. Blood samples were obtained from randomly selected outpatients who visited our hospital for medical care and HCWs undergoing regular medical checks with opt-out informed consent. SARS-CoV-2 antibody positivity was evaluated using two laboratory-based quantitative tests. The total number of participants enrolled was 6,369 (1st survey: 1,000 outpatients and 743 HCWs, 2nd survey: 1,000 outpatients and 407 HCWs, 3rd survey: 1,000 outpatients and 804 HCWs, 4th survey: 1,000 outpatients and 415 HCWs). The prevalence of SARS-CoV-2 antibodies among outpatients and HCWs was 0-0.1% and 0-0.124% during the research period, respectively, and changed little over time. These findings suggest that the magnitude of COVID-19 infection during the pandemic among outpatients and HCWs in this rural hospital might have been small.

O objetivo do trabalho foi determinar o pHi, a capacidade tamponante do citosol na ausência de CO2/HCO3- (βi) de neurônios dos gânglios das raízes dorsais e investigar a expressão de trocadores Na+/H+ e sua função na regulação do pHi. O pHi foi estimado com o indicador fluorescente BCECF. A operação do trocador era quantificada na recuperação da acidose intracelular induzida experimentalmente. Na ausência do tampão CO2/HCO3- a taxa de alcalinização (k) deve-se, por hipótese, ao transporte de H+ pelo trocador. A hipótese foi confirmada pela ação de agentes farmacológicos, e.g., amiloride. Em soluções tamponadas por CO2/HCO3- as células tem pHi de 7,24 e, em soluções tamponadas com HEPES, 7,04. A βi foi de 8,17 mM/pH. As células se recuperam da acidose com k médio de 0,0138 s-1. O efeito inibitório do amiloride em concentração de 1 mM deve-se ao fato dos fenótipos celulares expressarem diferentes isoformas do trocador. Segundo RT-PCR, todas as 5 isoformas do trocador são expressas e a quantidade de RNAm, avaliada por qPCR, é maior para a NHE1, seguida de NHE5. The objective here was to determine intracellular pH (pHi), cytosolic buffering power in CO2/HCO3- free medium (βi) of neurons from dorsal root ganglia and to investigate the functional expression of the Na+/H+ exchangers in the regulation of pHi. pHi was estimated with fluorescence indicator BCECF. Exchanger operation was quantified during recovery from intracellular acidification induced experimentally. In CO2/HCO3- free medium the alkalinization rate (k) is due, hypothetically, H+ extrusion by the exchanger. This assumption was confirmed by action of pharmacologic agents, e.g., amiloride. In medium buffered with CO2/HCO3- cells have pHi of 7.24 and, in medium buffered with HEPES, 7.04. βi calculated was 8.17 mM/pH. Cells recovery from acidosis with mean k of 0.0138 s-1. Inhibitory effect of amiloride in 1 mM concentration is due to cellular phenotypes expressing different Na+/H+ exchanger isoforms. According to RT-PCR, all the five exchanger isoforms are expressed and mRNA quantity, evaluated by qPCR, is greater to NHE1, followed by NHE5.

This study aims to clarify the features of the clear zone of odontoclasts on shedding teeth of a teleost fish, Chinook salmon, Oncorhynchus tshawytscha (Walbaum), using a light microscope to determine the orientation between a cell body and a resorptive lacuna, followed by transmission electron microscopy. Ultrathin sections of LR White embedded material were incubated in rabbit anti-actin polyclonal antibody and then were incubated with 15 nm gold-conjugated goat anti-rabbit IgG. The clear zones of odontoclasts showed a variable structure with electron-dense structures on sections, but distinct clear zones were not always seen on odontoclasts. In odontoclasts sectioned in the direction perpendicularly to the surface of a resorptive lacuna, some cells showed a wide clear zone, but two types of clear zones were usually observed: a part composed of some cytoplasmic processes and one composed of several complicatedly interwoven processes. Gold particles were localized on the clear zones, especially in electron-dense structures; very few gold particles were detected in ruffled borders. These results show that the clear zone of odontoclasts in Chinook salmon contains actin. Our results suggest that the clear zone of an odontoclast in Chinook salmon is not always a wide annular structure.

Predatory behavior of the lizardEumeces okadae upon 40 kinds of potential prey was observed under laboratory conditions. Prey items eaten byE. okadae were those having no particular defense mechanisms. Invertebrates with chemical repellants (phenol, alkaloids and cyanide), dense and long hairs around the body, and with lethal or poisonous bites were rejected. Sensory modalities for prey discrimination varied between the well eaten (preferred) and completely rejected (undesirable) prey. Almost all tasty prey were attacked on sight, while undesirable ones were rejected at various stages using visual and chemical senses. Despite the keen chemical prey discrimination ability inEumeces, this study demonstrated thatE. okadae did not always use chemical cues for prey discrimination. Fast predatory attacks immediately after visual discrimination are directed toward preferred and probably agile prey.

Laboratory experiments were conducted to examine whether the lizardEumeces okadae can visually discriminate between ground beetles with different chemical defenses and color patterns.Harpalus spp., which are uniformly black and emit formic acid, andDischissus mirandus, which has four bright yellow spots on a jet black back and emits metacresol, were tested. TheHarpalus beetles were attacked immediately after presentation, whereasD. mirandus was ignored on sight. By artificially paintingD. mirandus all black, a significantly higher frequency of tongue licks (close chemical examination of prey) was clicited than that by normally colored control beetles, indicating that the conspicuous coloration ofD. mirandus was functionally and visually warning to the lizards.

Laboratory experiments were conducted to test the hypothesis that widelyforaging generalist lizard,Eumeces okadae, visually discriminates palatable queen ants from unpalatable worker ants. The workers ofCamponotus japonicus andLasius niger were rejected on sight, while the queens of both species were eaten with little prior chemical examination by tongue flicks or licks. Comparison of the lizards' responses towards the workers and wingless queens of similar size indicated that neither body size nor presence or absence of wings accounted for difference in responses toward the 2 ant castes. The lizards probably discriminated different ant castes by body proportions.

The effects of mechanical stress release on osteoclastogenesis may be as important as those of mechanical stress application. However, the direct effects of mechanical stress on the behavior of osteoclasts has not been thoroughly investigated and there is limited information on the results of the release from mechanical stress. In this study, the effects of mechanical stress application and its release on osteoclast differentiation were examined. The number of tartrate-resistant acid phosphatase (TRAP)-positive multinucleated osteoclasts derived from RAW264.7 cells were measured and the expression of osteoclast differentiation genes, which was altered in response to the release from mechanical stress according to the Flexercell tension system was evaluated by real-time PCR. Osteoclast differentiation and fusion were suppressed by mechanical stress application and were rapidly induced after mechanical stress release. The mRNA expression of the osteoclast specific genes, TRAP, matrix metalloproteinase-9 (MMP-9), cathepsin-K (cath-k), calcitonin receptor (CTR), ATPase H+ transporting vacuolar proton pump member I (ATP6i), chloride channel-7 (ClC7) and dendritic cell-specific transmembrane protein (DC-STAMP) was decreased with mechanical stress application, and increased up to 48 h after the release from it. These alterations in gene mRNA expression were associated with the number of osteoclasts and large osteoclasts. Inducible nitric oxide synthetase (iNOS) mRNA was increased with mechanical stress and decreased after its release. Nitric oxide (NO) production was increased with mechanical stress. Nuclear factor of activated T cells cytoplasmic (NFATc) family mRNAs were not altered with mechanical stress, but were up-regulated up to 48 h after the release from it. These findings indicate that the suppression of osteoclast differentiation and fusion induced by mechanical stress is the result of NO increase via iNOS, and that the promotion of osteoclast differentiation and fusion after the release from mechanical stress is related to the NFATc family genes, whose expression remained constant during mechanical stress but was up-regulated after the release from mechanical stress.

This study was designed to establish the apoptosis of odontoclasts during physiological root resorption of human deciduous teeth. Deciduous teeth were fixed, decalcified, and embedded in paraffin for immunohistochemical (IHC) observations and in Epon for transmission electron microscopy (TEM). Apoptotic cells were identified by terminal deoxynucleotidyl transferase (TdT)-mediated dUTP-digoxigenin nick-end labeling (TUNEL), and then tartrate-resistant acid phosphatase (TRAP) activity was determined on the same sections. Epon-embedded specimens were sectioned serially into 0.5-microm semithin sections; some of these sections were re-embedded in Epon, sectioned into 0.1-microm ultrathin sections, and observed by TEM. IHC revealed that the nuclei of TRAP-positive odontoclasts on the dentine were generally TUNEL-negative. Around these odontoclasts, a few TRAP-positive structures were present together with TUNEL-positive structures, e.g., a TRAP-positive structure with one TUNEL-positive nucleus, a TRAP-positive structure with one TUNEL-positive nucleus plus one or two TUNEL-negative nuclei, or a TRAP-positive structure with no nucleus. By TEM, some odontoclasts showed nuclear fragments including compacted chromatin. The results suggest that, during apoptosis, odontoclasts fragment into variously sized cellular parts including three or fewer nuclei.

Odontoclasts resorbing teeth are multinucleated cells. Previously, the authors have investigated the distribution of number of nuclei per human odontoclast and showed that the mean number of nuclei per cell is 5.3, the median is 4, and 93.8% of cells have 10 or fewer nuclei. Teleost odontoclasts have features similar to those of mammals; however, the distribution of number of nuclei per cell remains unknown. The present study aimed to examine the distribution of number of nuclei per odontoclast in a teleost fish, Chinook salmon, Oncorhynchus tshawytscha (Walbaum), and to clarify the difference of number of nuclei in odontoclasts between Chinook salmon and humans. The maxillae and mandibles of Chinook salmon were fixed, decalcified, and embedded in Epon 812. Specimens were serially sectioned into 0.5-microm semithin sections and examined by light microscopy. Cells possessing a brush border adjacent to a resorptive lacuna were identified as odontoclasts, and 246 odontoclasts were investigated to determine the distribution of nuclei per cell. The mean number of nuclei per cell was 21.8 and the median was 17; only 24.4% of odontoclasts had 10 or fewer nuclei, and 95.5% had 50 or fewer nuclei. These results suggest that the range for the number of nuclei per odontoclast in Chinook salmon is greater than that in humans.
