To improve the standard screening, diagnosis, and treatment of hypertension in patients in China; realize the standardization of clinical practice of hypertension; and improve the prevention and control level of hypertension in China, it is both important and necessary to develop a clinical practice guideline for hypertension according to a recognized methodology. Jointly sponsored by the National Center for Cardiovascular Diseases, Chinese Medical Doctor Association, Hypertension Committee of the Chinese Medical Doctor Association, Chinese Society of Cardiology, and Hypertension Committee of Cross-Straits Medicine Exchange Association, the “Chinese Clinical Practice Guidelines of Hypertension” was proposed. Research Unit of Evidence-Based Evaluation and Guidelines, Chinese Academy of Medical Sciences, Guideline and Standards Research Centre of Chinese Medical Association Publishing House, Lanzhou University Institute of Health Data Science, and Lanzhou University GRADE Center will provide methodological support for the guidelines.

Objective: To explore the associations between dietary zinc intake and cardiovascular diseases (CVDs), including congestive heart failure (CHF), coronary heart disease (CHD), angina, heart attack, and cerebrovascular accident (CVA), this study was performed. Setting: Data from the National Health and Nutrition Examination Survey (2005–2018) were used in this study. Dietary zinc intake was stratified into quartiles. Restricted cubic splines were constructed to assess nonlinear associations and identify cut-off values based on the type of nonlinearity. Binary logistic regressions were performed using the cut-offs. Results: Positive associations were detected between the second, third, and fourth quantiles of dietary zinc intake and decreased risks of overall CVDs (Q2: OR = 0.83, 95 % CI = 0.72–0.96; Q3: OR = 0.83, 95 % CI = 0.71–0.96; Q4: OR = 0.79, 95 % CI = 0.67–0.93). The second, third, and fourth quantiles were significantly associated with decreased risks of various CVDs (all P 0.05). Abstract Here we presented an electrophysiological dataset collected from layer V of the primary motor cortex (M1) and the corresponding behavior dataset from normal and hemi-parkinson rats over 5 consecutive weeks. The electrophysiological dataset was constituted by the raw wideband signal, neuronal spikes, and local field potential (LFP) signal. The open-field test was done and recorded to evaluate the behavior variation of rats among the entire experimental cycle. We conducted technical validation of this dataset through sorting the spike data to form action potential waveforms and analyzing the spectral power of LFP data, then based on these findings a closed-loop DBS protocol was developed by the oscillation activity response of M1 LFP signal. Additionally, this protocol was applied to the hemi-parkinson rat for five consecutive days while simultaneously recording the electrophysiological data. This dataset is currently the only publicly available dataset that includes longitudinal closed-loop DBS recordings, which can be utilized to investigate variations of neuronal activity within the M1 following long-term closed-loop DBS, and explore additional reliable biomarkers.

Human enteroviruses (EVs) represent a global public health concern due to their association with a range of serious pediatric illnesses. Despite the high morbidity and mortality exerted by EVs, no broad-spectrum antivirals are currently available. Herein, we presented evidence that doxycycline can inhibit in vitro replication of various neurotropic EVs, including enterovirus A71 (EV-A71), enterovirus D68 (EV-D68), and coxsackievirus (CV)-A6, in a dose-dependent manner. Further investigations indicated that the drug primarily acted at the post-entry stage of virus infection in vitro, with inhibitory effects reaching up to 89 % for EV-A71 when administered two hours post-infection. These findings provide valuable insights for the development of antiviral drugs against EV infections.

Over the past decade, advancements in high-throughput sequencing technologies have led to a qualitative leap in our understanding of the role of the microbiota in human diseases, particularly in oncology. Despite the low biomass of the intratumoral microbiota, it remains a crucial component of the tumor immune microenvironment, displaying significant heterogeneity across different tumor tissues and individual patients. Although immunotherapy has emerged a major strategy for treating tumors, patient responses to these treatments vary widely. Increasing evidence suggests that interactions between the intratumoral microbiota and the immune system can modulate host tumor immune responses, thereby influencing the effectiveness of immunotherapy. Therefore, it is critical to gain a deep understanding of how the intratumoral microbiota shapes and regulates the tumor immune microenvironment. Here, we summarize the latest advancements on the role of the intratumoral microbiota in cancer immunity, exploring the potential mechanisms through which immune functions are influenced by intratumoral microbiota within and outside the gut barrier. We also discuss the impact of the intratumoral microbiota on the response to cancer immunotherapy and its clinical applications, highlighting future research directions and challenges in this field. We anticipate that the valuable insights into the interactions between cancer immunity and the intratumoral microbiota provided in this review will foster the development of microbiota-based tumor therapies.

Accurate prediction of significant wave height is crucial for ocean engineering. Traditional time series prediction models fail to achieve satisfactory results due to the non-stationarity of significant wave height. Decomposition algorithms are adopted to address the problem of non-stationarity, but the traditional direct decomposition method exists information leakage. In this study, a hybrid VMD-LSTM-rolling model is proposed for non-stationary wave height prediction. In this model, time series are generated by a rolling method, after which each time series is decomposed, trained and predicted, then the predictions of each time series are combined to generate the final prediction of significant wave height. The performance of the LSTM model, the VMD-LSTM-direct model and the VMD-LSTM-rolling model are compared in terms of multi-step prediction. It is found that the error of the VMD-LSTM-direct model and the VMD-LSTM-rolling model is lower than that of the LSTM model. Due to the decomposition of the testing set, the VMD-LSTM-direct model has a slightly higher accuracy than the VMD-LSTM-rolling model. However, given the issue of information leakage, the accuracy of the VMD-LSTM-direct model is considered false. Thus, it has been proved that the VMD-LSTM-rolling model exhibits superiority in predicting significant wave height and can be applied in practice.

Through a blend of conventional and novel pre-processing methods, this module significantly improves the accuracy of existing toxic text detection tools, addressing the challenge of content that is deliberately altered to evade standard detection methods. • Integration with Existing Systems: “Specialis Revelio” is designed to augment popular toxic text classifiers, enhancing their ability to detect and filter toxic content more effectively. • Innovative Pre-processing Methods: The module combines traditional pre-processing steps like lowercasing and stemming with advanced strategies, including the handling of adversarial examples and typo correction, to reveal concealed toxicity. • Validation through Comparative Study: Its effectiveness was validated via a comparative analysis against widely used APIs, demonstrating a marked improvement in the detection of various toxic text indicators.

Previous epidemiologic studies have shown a close association between type 1 diabetes mellitus (T1DM), hyperlipidemia, and inflammatory bowel disease (IBD), but the causal relationship has not been established. In the current study the causal relationships between T1DM and hyperlipidemia with IBD were assessed using Mendelian randomization (MR) analysis. A two-sample MR study was conducted utilizing accessible genome-wide association study data from public sources with the selection of suitable instrumental variables adhering to the principles of MR analysis. The primary technique utilized was the inverse variance weighted method, complemented by additional methods, such as MR-Egger regression, weighted median, simple mode, weighted mode, and the MR pleiotropy residual sum and outlier approach. Genetically determined T1DM had no causal relationship with IBD or IBD subtypes based on MR analysis. These findings were consistent across all supplementary methods used. In addition, genetically determined hyperlipidemia had no causal relationship with IBD or IBD subtypes, even after increasing the number of instrumental variables used. Our study supports the notion that there is no causal relationship between T1DM and IBD, as well as hyperlipidemia and IBD, which contradicts most observational studies.

Abstract Nocturnal hypertension is highly prevalent among Chinese and Asian populations, which is mainly attributed to high salt intake and high salt sensitivity. Nocturnal hypertension increases the risk of cardiovascular and all‐cause mortality, independent of daytime blood pressure (BP). However, it can usually be detected by 24‐h ambulatory BP monitoring, rather than routine office or home BP measurement, thus is often underdiagnosed in clinical practice. Currently, no specific guidance is available for the management of nocturnal hypertension in China or worldwide. Experts from the Chinese Hypertension League summarized the epidemiologic and pathophysiologic characteristics and clinical phenotype of nocturnal hypertension and provided consensus recommendations on optimal management of nocturnal hypertension, with the goal of maximally reducing the cardiovascular disease risks. In this consensus document, 24‐h ABPM is recommended for screening and diagnosis of nocturnal hypertension, especially in the elderly, patients with diabetes, chronic kidney diseases, obstructive sleep apnea and other conditions prone to high nocturnal BP. Lifestyle modifications including salt intake restriction, exercise, weight loss, sleep improvement, and mental stress relief are recommended. Long‐acting antihypertensive medications are preferred for nocturnal and 24‐h BP control. Some newly developed agents, renal denervation, and other device‐based therapy on nocturnal BP reduction are evaluated.

Accurate prediction of significant wave height is of great reference value for wave energy generation. However, due to the non-linearity and non-stationarity of significant wave height, traditional algorithms face difficulties in achieving satisfactory prediction results. In this study, a hybrid CEEMDAN-VMD-TimesNet model is proposed for non-stationary significant wave height prediction. Based on the significant wave height in the South Sea of China, the performance of the SVM model, the GRU model, the LSTM model, the TimesNet model, the CEEMDAN-TimesNet model and the CEEMDAN-VMD-TimesNet model are compared in terms of multi-step prediction. It is found that the prediction accuracy of the TimesNet model is higher than that of the SVM model, the GRU model and the LSTM model. The non-stationarity of significant wave height is reduced by CEEMDAN decomposition. Thus, the CEEMDAN-TimesNet model performs better than the TimesNet model in predicting significant wave height. The prediction accuracy of the CEEMDAN-VMD-TimesNet model is further improved by employing VMD for the secondary decomposition of components with high and moderate complexity. Additionally, the CEEMDAN-VMD-TimesNet model can accurately predict trends and extreme values of significant wave height with minimal phase shifts even during typhoon periods. The results demonstrate that the CEEMDAN-VMD-TimesNet model exhibits superiority in predicting significant wave height.

Abstract Background This study aimed to identify an initial screening tool for congenital ectopia lentis (CEL) by comparing ocular biological parameters in children with myopia. Methods A retrospective case-control study was conducted at one tertiary referral centre, from October 2020 to June 2022. Axial length (AL), corneal curvature (CC), refractive astigmatism (RA), corneal astigmatism (CA), internal astigmatism (IA), the difference between the axis of RA and CA [AXIS(RA-CA)], white-to-white corneal diameter (WTW), and axial length-corneal radius ratio (AL/CR) were compared in 28 eyes of CEL patients, and 60 eyes of myopic patients matched for age and refraction. The spherical equivalent of each eye was < -3.00 D. Area under the curve (AUC) of the receiver operating characteristic curves were calculated. Abstract Anoikis resistance, a notable factor in osteosarcoma, plays a significant role in tumor invasion and metastasis. This study seeks to identify a distinct gene signature that is specifically associated with the anoikis subcluster in osteosarcoma. Clinical, single-cell, and transcriptional data from TARGET and GEO datasets were used to develop a gene signature for osteosarcoma based on the anoikis subcluster. Univariate Cox and LASSO regression analyses were employed. The signature's predictive value was evaluated using time-dependent ROC and Kaplan–Meier analyses. Functional enrichment analyses and drug sensitivity analyses were conducted. Validation of three modular genes was performed using RT-qPCR and Western blotting. Signature (ZNF583, CGNL1, CXCL13) was developed to predict overall survival in osteosarcoma patients, targeting the anoikis subcluster. The signature demonstrated good performance in external validation. Stratification based on the signature revealed significantly different prognoses. The signature was an independent prognostic factor. The low-risk group showed enhanced immune cell infiltration and improved immune function. Drug sensitivity analysis indicated efficacy of chemotherapy agents. Prognostic nomograms incorporating the signature provided greater predictive accuracy and clinical utility. Signatures related to the anoikis subcluster play a significant role in osteosarcoma progression. Incorporating these findings into clinical decision-making can improve osteosarcoma treatment and patient outcomes.

Abstract Objective To study the development of microalbuminuria (MAU) in essential hypertension (EHT), we investigated the association of MAU with central blood pressure (CBP), direct renin concentration (DRC), plasma aldosterone (PA), and uric acid (UA). Method We determined 24 h-urinary albumin excretion (24 h-UAE) in patients with EHT who were hospitalized at TEDA International Cardiovascular Hospital from June 2020 to May 2022. We defined MAU as 24 h-UAE in the range of 30 mg/24 h to 300 mg/24 h. Univariate and multivariate analyses were conducted to determine the associations of MAU with CBP, DRC, PA, and UA in EHT, considering demographic and clinical information. We also plotted receiver operating characteristic curves (ROCs) for predicting MAU using these results. Results More than a quarter of patients (26.5%, 107/404, 95% CI: 22.2–31.1%) were diagnosed with MAU in EHT. A higher body mass index (BMI), longer duration of hypertension, and higher severity were associated with MAU. Also, nearly 10% more creatinine levels were recorded in the MAU group than in the control group (69.5 ± 18.7 µmol/L vs. 64.8 ± 12.5 µmol/L, P = 0.004). Abstract Cuproptosis, caused by excessively high copper concentrations, is urgently exploited as a potential cancer therapeutic. However, the mechanisms underlying the initiation, propagation, and ultimate execution of cuproptosis in tumors remain unknown. Here, we show that copper content is significantly elevated in gastric cancer (GC), especially in malignant tumors. Screening reveals that METTL16, an atypical methyltransferase, is a critical mediator of cuproptosis through the m6A modification on FDX1 mRNA. Furthermore, copper stress promotes METTL16 lactylation at site K229 followed by cuproptosis. The process of METTL16 lactylation is inhibited by SIRT2. Elevated METTL16 lactylation significantly improves the therapeutic efficacy of the copper ionophore– elesclomol. Combining elesclomol with AGK2, a SIRT2-specific inhibitor, induce cuproptosis in gastric tumors in vitro and in vivo. These results reveal the significance of non-histone protein METTL16 lactylation on cuproptosis in tumors. Given the high copper and lactate concentrations in GC, cuproptosis induction becomes a promising therapeutic strategy for GC.

BackgroundLeft ventricular (LV) diastolic dysfunction is an independent predictor of future cardiovascular events. Early detection of patients with LV diastolic dysfunction can improve clinical outcomes through active management. However, the assessment of diastolic function is very complicated, and there are currently lack of effective biomarkers to assess the risk of LV diastolic dysfunction. Connective tissue growth factor (CTGF) plays a significant role in cardiac remodeling and dysfunction. We aimed to investigate the associations between plasma CTGF level and the risk of LV diastolic dysfunction in this study and judge its effectiveness in diagnosing LV diastolic dysfunction.MethodsA total of 169 patients with overt hyperthyroidism were included. LV diastolic function was evaluated and the subjects were divided into normal LV diastolic function group and LV diastolic dysfunction group. Routine clinical medical data, biochemical data, thyroid related parameters and echocardiographic parameters were recorded for analysis.ResultsCompared with normal LV diastolic function group, the LV diastolic dysfunction group had higher age and BMI, as well as lower heart rate, lower serum albumin, lower eGFR, higher serum TgAb and BNP level, and the incidences of hypertension were also higher (all P

The reactivation of colluvial landslides in reservoir banks poses a serious threat to the safety of hydropower projects and nearby towns. This study aims to research the morphological evolution of this type of landslides under the action of reservoir water and the impact of morphological evolution on landslide stability. The study focused on the Yulinerzu landslide, a large reactivated colluvial landslide in the Xiluodu Reservoir, China. Field surveys were conducted to analyze the geological structure of the landslide. In situ monitoring and surveys were used to obtain the deformation characteristics and morphological evolution of the landslide. A combined seepage-slope stability analysis was conducted to reveal the deformation mechanism. The results show that the reactivation of the Yulinerzu landslide is dominated by reservoir water fluctuations rather than rainfall. The underlying geological condition of the colluvial landslide is its hydrogeological structure, which causes the landslide to deform in a step-like manner during reservoir operation. With the accumulation of displacement and morphology evolution, the landslide displayed self-stabilizing characteristics. Therefore, in the stability analysis and risk assessment of large deformation landslides, it is essential to take into account not only the hydraulic effects of reservoir fluctuation but also the evolution of landslide morphology.

Background Preeclampsia (PE) mainly occurs in pregnant women and is hereditary. Several genome-wide association studies (GWAS) on Caucasian samples have reported some gene loci that are associated with preeclampsia. However, these studies have not reached consistent conclusions. No previous GWAS has examined preeclampsia in the Chinese Han population. Method This study aimed to identify common genetic variations associated with preeclampsia in the Chinese Han population through two-stage case‒control studies. The discovery cohort included 92 patients with severe preeclampsia and 187 healthy controls. The validation cohort included 52 patients with preeclampsia and 104 controls. A genome-wide association study was performed to identify putative preeclampsia genes in the discovery cohort, with validation in the validation cohort. Results In the discovery cohort, GWAS demonstrated that 19 single-nucleotide polymorphisms (SNPs) were associated with preeclampsia (P 0.05). Furthermore, rs13210237 and rs13176432 were related to preeclampsia in the adjusted regression model (P < 0.000). Conclusion In this study of two independent cohorts, we found that rs13210237 and rs13176432 might be novel preeclampsia-susceptible genetic factors in the Han population in China. However, there was no association between the onset of preeclampsia and these genotypes.

Heterogeneous thermal-catalytic CO2 hydrogenation to alcohols using renewable energy is a highly attractive approach for recycling greenhouse gases into high-value chemicals and fuels, thereby reducing the dependence on fossil fuels, while simultaneously mitigating the CO2 emission and environmental problems. Currently, great advances have been made on the heterogeneous catalysts, but an in-depth and more comprehensive understanding to further promote this reaction process is still lacking. Herein, we highlight the thermodynamic and kinetic analysis of CO2 hydrogenation reaction firstly. Then, various reaction pathways for CO2 hydrogenation to methanol and higher alcohols (C2+ alcohols) have been discussed in detail, respectively, by combining the experimental studies and density functional theory calculations. On this basis, the key factors influencing the reaction performance, such as metal dispersion, support modification, promoter addition and their structural optimization, are summarized on the metal-based and metal-oxide-based catalysts. In addition, the catalytic performance of CO2 hydrogenation to alcohols and the relationship between structure and properties are mainly summarized and analyzed in the past five years. To conclude, the current challenges and potential strategies in catalyst design, structural characterization and reaction mechanisms are presented for CO2 hydrogenation to alcohols.

Abstract Objective To estimate the relationship among uric acid (UA), 24-h microalbumin (24 h-MAU) and estimated glomerular filtration rate (eGFR) in hypertensive patients. BackgroundBrain gray matter alterations in patients with trigeminal neuralgia (TN) have been detected in prior neuroimaging studies, but the results are heterogeneous. The current study conducted coordinate-based meta-analyses across neuroimaging studies, aiming to find the pattern of brain anatomic and functional alterations in patients with TN.MethodsWe performed a systematic literature search of PubMed, Embase, and Web of Science to identify relevant publications. A multimodal meta-analysis for whole-brain voxel-based morphometry (VBM) studies and functional imaging studies in TN was performed using anisotropic effect size-based signed differential mapping.ResultsThe meta-analysis comprised 10 VBM studies with 398 TN patients and 275 healthy controls, and 13 functional magnetic resonance imaging studies with 307 TN patients and 264 healthy controls. The multimodal meta-analysis showed conjoint structural and functional brain alterations in the right fusiform gyrus and inferior temporal gyrus, bilateral thalamus, left superior temporal gyrus, left insula, and inferior frontal gyrus. The unimodal meta-analysis showed decreased gray matter volume alone in the left putamen, left postcentral gyrus, and right amygdala as well as only functional abnormalities in the left cerebellum, bilateral precuneus, and left middle temporal gyrus.ConclusionThis meta-analysis revealed overlapping anatomic and functional gray matter abnormalities in patients with TN, which may help provide new insights into the neuropathology and potential treatment biomarkers of TN.

The specific air jet of a diffuser is formed by the complex internal structure, which affects the outlet airflow distribution of the diffuser directly and the indoor environment distribution indirectly. If the diffusers are developed based on their actual geometry structure and their boundary conditions are set as their inlet flowrate, the simulated indoor temperature distribution will be more accurate. However, it is noted that many problems may arise, such as model complexity, many grid cells, and slow convergence of calculations. Therefore, this paper focuses on a simplified method for four-way square diffusers in a computational fluid dynamics (CFD) simulation of indoor non-uniform temperature distribution. Firstly, the airflow distribution is simulated on the outlet air supply cross-section of the diffuser. Then, according to the outflow characteristics of the diffuser, the diffuser model is simplified and simulated in an experimental room. Finally, the temperature distribution at the 1.2 m height plane is obtained from CFD simulation and compared with the experimental results. The results show that the 68-point air supply opening model can well simulate the effects of the outlet airflow distribution of the diffuser, and the simulated indoor temperature distribution meets the experiment results well. The deviations for three scenarios are between −7.4~1.7% and the average deviation is −3.0%, while the root mean square error of temperature for three scenarios is 0.7 °C, 0.7 °C, and 1.0 °C, respectively. The results also demonstrate the mutual influence of the airflow from different diffusers and the indoor non-uniform temperature distribution under the action of multiple diffusers. The proposed method can contribute to balancing the model complexity and the accuracy in CFD simulation, especially for multiple diffusers in the room.

Abstract Background The causal association between educational attainment (EA) and stroke remains unclear. Hence, a novel multivariable Mendelian randomization (MVMR) approach was applied to solve this issue. Methods The single nucleotide polymorphisms (SNPs) from a recent genome-wide association study (GWAS) on years of schooling served as instruments. Univariable mendelian randomization (MR) and MVMR analyses were performed to detect the relationship between genetically predicted EA and the stroke risk. In the MVMR, cigarette consumption, alcohol consumption, body mass index (BMI), intelligence, and hypertension were adjusted. The summary statistics for stroke from the MEGASTROKE consortium included 446,696 participants (40,585 cases of stroke and 34,217 cases of ischemic stroke), most of whom were of European descent. Results In the univariable MR, genetically predicated EA could decrease the risks of total stroke (OR = 0.66, 95% CI 0.61–0.72, P = 2.70 × 10–23), ischemic stroke (OR = 0.67, 95% CI 0.61–0.73, P = 2.58 × 10–18), large artery atherosclerosis (OR = 0.51, 95% CI 0.40–0.64, P = 1.80 × 10–8), small vessel stroke (OR = 0.60, 95% CI 0.49–0.73, P = 5.59 × 10–7), and cardioembolic stroke (OR = 0.81, 95% CI 0.68–0.96, P = 1.46 × 10–2) using the inverse-variance weighted (IVW) estimator. Higher EA might be negatively correlated with the odds of total stroke (OR = 0.62, 95% CI 0.50–0.77, P = 1.44 × 10–5), ischemic stroke (OR = 0.63, 95% CI 0.50–0.80, P = 1.41 × 10–4), and cardioembolic stroke (OR = 0.59, 95% CI 0.39–0.90, P = 0.01), but was not significant in large artery atherosclerosis (OR = 0.65, 95% CI 0.37–1.15, P = 0.14) and small vessel stroke (OR = 0.68, 95% CI 0.41–1.13, P = 0.14) after controlling other exposures. Conclusions We found that genetically predicated higher EA decreased the risks of total stroke, ischemic stroke, and cardioembolic stroke, independent of smoking, alcohol consumption, BMI, intelligence, and hypertension.

BackgroundPancreatic ductal adenocarcinoma (PDAC) is one of the most fatal malignancies worldwide, mostly as a result of the absence of early detection and specific treatment solutions. Consequently, identifying mutational profiles and molecular biomarkers is essential for increasing the viability of precision therapy for pancreatic cancer.MethodsWe collected blood and tumor tissue samples from 47 Chinese pancreatic cancer patients and used whole-exome sequencing (WES) to evaluate the genetic landscape.ResultsOur results showed the most frequently somatic alteration genes were KRAS (74.5%), TP53(51.1%), SMAD4 (17%), ARID1A (12.8%), CDKN2A (12.8%), TENM4 (10.6%), TTN (8.5%), RNF43(8.5%), FLG (8.5%) and GAS6 (6.4%) in Chinese PDAC patients. We also found that three deleterious germline mutations (ATM c.4852C>T/p. R1618*, WRN c.1105C>T/p. R369*, PALB2 c.2760dupA/p. Q921Tfs*7) and two novel fusions (BRCA1-RPRML, MIR943 (intergenic)-FGFR3). When compared to the Cancer Genome Atlas (TCGA) database, there is a greater mutation frequency of TENM4 (10.6% vs. 1.6%, p = 0.01), GAS6(6.4% vs. 0.5%, p = 0.035), MMP17(6.4% vs. 0.5%, p = 0.035), ITM2B (6.4% vs. 0.5%, p = 0.035) and USP7 (6.4% vs. 0.5%, p= 0.035) as well as a reduced mutation frequency of SMAD4 (17.0% vs. 31.5%, p = 0.075) and CDKN2A (12.8% vs. 47.3%, p < 0.001) were observed in the Chinese cohort. Among the 41 individuals examined for programmed cell death ligand 1(PD-L1) expression, 15 (36.6%) had positive PD-L1 expression. The median tumor mutational burden (TMB) was found to be 12muts (range, 0124). The TMB index was higher in patients with mutant-type KRAS MUT/TP53 MUT (p < 0.001), CDKN2A (p = 0.547), or SMAD4 (p = 0.064) compared to patients with wild-type KRAS/TP53, CDKN2A, or SMAD4.ConclusionsWe exhibited real-world genetic traits and new alterations in Chinese individuals with cancer of the pancreas, which might have interesting implications for future individualized therapy and medication development.

ABSTRACT: Objectives: The aims of this study were to investigate the characteristics of Klebsiella pneumoniae meningitis and the impact of convergence of carbapenem resistance and hypervirulence on patient mortality.Methods: Antimicrobial resistance and virulence-related genes were investigated in 25 K. pneumoniae strains causing meningitis. Clinical data for 25 patients from February 2009 to February 2019 were evaluated. Multilocus sequence typing (MLST), serotyping, analysis of mobile genetic elements and pulsed-field gel electrophoresis (PFGE) were performed. GraphPad Prism was used for statistical analysis.Results: The mortality rate of patients with K. pneumoniae meningitis was 30.0%. Significant differences were observed between non-survivor and survivor groups regarding mechanical ventilation, peripheral deep vein catheter insertion and GCS score, but not sex, age or meningeal integrity destruction. Multidrug resistance was observed in 21 isolates. Rates of detection for each virulence gene ranged from 4.0% for wzy-K1 to 100.0% for entB. Detection rates of carbapenem-resistant K. pneumoniae (CRKP), hypervirulent K. pneumoniae (HvKP) and hypervirulent carbapenem-resistant K. pneumoniae (Hv-CRKP) were 68.0%, 68.0% and 48.0%, respectively. In total, 16 clusters and 19 clones were identified among the 25 isolates. Mortality rates differed significantly between the non-Hv-CRKP (1/11) versus Hv-CRKP groups (5/9), but were comparable in the carbapenem-susceptible K. pneumoniae versus CRKP groups and the classical K. pneumoniae versus HvKP groups.Conclusions: Klebsiella pneumoniae meningitis is associated with high mortality. Klebsiella pneumoniae-induced meningitis has highly divergent origins. Convergence of carbapenem resistance and hypervirulence leads to high mortality in patients with K. pneumoniae meningitis, which is of great clinical concern.

Abstract Heart failure is the terminal stage of many cardiac diseases, in which β1-adrenoceptor (β1-AR) autoantibody (β1-AA) has a causative role. By continuously activating β1-AR, β1-AA can induce cytotoxicity, leading to cardiomyocyte apoptosis and heart dysfunction. However, the mechanism underlying the persistent activation of β1-AR by β1-AA is not fully understood. Receptor endocytosis has a critical role in terminating signals over time. β2-adrenoceptor (β2-AR) is involved in the regulation of β1-AR signaling. This research aimed to clarify the mechanism of the β1-AA-induced sustained activation of β1-AR and explore the role of the β2-AR/Gi-signaling pathway in this process. The beating frequency of neonatal rat cardiomyocytes, cyclic adenosine monophosphate content, and intracellular Ca2+ levels were examined to detect the activation of β1-AA. Total internal reflection fluorescence microscopy was used to detect the endocytosis of β1-AR. ICI118551 was used to assess β2-AR/Gi function in β1-AR sustained activation induced by β1-AA in vitro and in vivo. Monoclonal β1-AA derived from a mouse hybridoma could continuously activate β1-AR. β1-AA-restricted β1-AR endocytosis, which was reversed by overexpressing the endocytosis scaffold protein β-arrestin1/2, resulting in the cessation of β1-AR signaling. β2-AR could promote β1-AR endocytosis, as demonstrated by overexpressing/interfering with β2-AR in HL-1 cells, whereas β1-AA inhibited the binding of β2-AR to β1-AR, as determined by surface plasmon resonance. ICI118551 biasedly activated the β2-AR/Gi/G protein-coupled receptor kinase 2 (GRK2) pathway, leading to the arrest of limited endocytosis and continuous activation of β1-AR by β1-AA in vitro. In vivo, ICI118551 treatment attenuated myocardial fiber rupture and left ventricular dysfunction in β1-AA-positive mice. This study showed that β1-AA continuously activated β1-AR by inhibiting receptor endocytosis. Biased activation of the β2-AR/Gi/GRK2 signaling pathway could promote β1-AR endocytosis restricted by β1-AA, terminate signal transduction, and alleviate heart damage.

ObjectiveThis study seeks to investigate the relationship between Obstructive Sleep Apnea-Hypopnea Syndrome (OSAHS) and hearing impairment by meta-analysis.MethodsCochrane Library, PubMed, Embase, Web of Science and other databases are searched from their establishment to July 1st, 2022. Literature on the relationship between OSAHS and hearing loss is collected, and two researchers independently perform screening, data extraction and quality evaluation on the included literature. Meta-analysis is performed using RevMan 5.4.1 software. According to the heterogeneity between studies, a random-effects model or fixed-effects model is used for meta-analysis.ResultsA total of 10 articles are included, with 7,867 subjects, 1,832 in the OSAHS group and 6,035 in the control group. The meta-analysis shows that the incidence of hearing impairment in the OSAHS group is higher than in the control group (OR = 1.38; 95% CI 1.18–1.62, Z = 4.09, P < 0.001), and the average hearing threshold of OSAHS patients is higher than that of the control group (MD = 5.89; 95% CI 1.87–9.91, Z = 2.87, P = 0.004). After stratifying the included studies according to hearing frequency, the meta-analysis shows that the OSAHS group has a higher threshold of 0.25, and the response amplitudes at frequencies 2, 4, 6, and 8 kHz are all higher than those of the control group.ConclusionCompared with the control group, the OSAHS group has a higher incidence of hearing loss, mainly high-frequency hearing loss. Thus, OSAHS is closely associated with and a risk factor for hearing loss.

BackgroundRecent studies have shown that the neutrophil-to-lymphocyte ratio (NLR) has gradually been identified as a more reliable marker of inflammation, with predictive value for the development of many diseases. However, its association with left ventricular (LV) diastolic dysfunction in overt hyperthyroid patients is unclear. Here, we aimed to explore the relationship between NLR and LV diastolic dysfunction in overt hyperthyroid patients.MethodsFor this study, we retrospected the consecutive medical files of 350 overt hyperthyroid patients. Their medical data and laboratory findings were recorded. According to the presence or absence of LV diastolic dysfunction, the patients with overt hyperthyroidism were divided into two groups. One group with LV diastolic dysfunction included 104 patients and another group with non-LV diastolic dysfunction included 246 patients. The NLR values between the two groups were compared, and the relationship between NLR levels and the prevalence of LV diastolic dysfunction was also explored.ResultsThe NLR value in LV diastolic dysfunction group in the overt hyperthyroid subjects was significantly higher than that in non-LV diastolic dysfunction group [1.100 (0.907-1.580) vs 1.000 (0.761-1.405), P=0.016]. The prevalence of LV diastolic dysfunction in Low- (NLR1.287) NLR level groups were 20.9%, 32.5% and 35.7% respectively. Moreover, increased NLR is associated with increased prevalence of LV diastolic dysfunction, and after adjustment for potential associated factors, NLR remained significantly associated with LV diastolic dysfunction. (OR = 11.753, 95%CI = 1.938-71.267, P = 0.007).ConclusionsOur findings demonstrated that the NLR was associated with LV diastolic dysfunction in the overt hyperthyroid patients, and the prevalence of LV diastolic dysfunction may be positively correlated with NLR levels.

Glioma, one of the most common malignant tumors in the nervous system, is characterized by limited treatment, high mortality and poor prognosis. Numerous studies have shown that lncRNAs play an important role in the onset and progression of glioma by acting on various classical signaling pathways of tumors through signaling, trapping, guiding, scaffolding and other functions. LncRNAs contribute to the malignant progression of glioma via proliferation, apoptosis, epithelial-mesenchymal transformation, chemotherapy resistance, ferroptosis and other biological traits. In this paper, relevant lncRNA signaling pathways involved in glioma progression were systematically evaluated, with emphasis placed on the specific molecular mechanism of lncRNAs in the process of ferroptosis, in order to provide a theoretical basis for the application of lncRNAs in the anticancer treatment of glioma.

Abstract Disease progression prediction and therapeutic drug target discovery for Coronavirus disease 2019 (COVID-19) are particularly important, as there is still no effective strategy for severe COVID-19 patient treatment. Herein, we performed multi-platform omics analysis of serial plasma and urine samples collected from patients during the course of COVID-19. Integrative analyses of these omics data revealed several potential therapeutic targets, such as ANXA1 and CLEC3B. Molecular changes in plasma indicated dysregulation of macrophage and suppression of T cell functions in severe patients compared to those in non-severe patients. Further, we chose 25 important molecular signatures as potential biomarkers for the prediction of disease severity. The prediction power was validated using corresponding urine samples and plasma samples from new COVID-19 patient cohort, with AUC reached to 0.904 and 0.988, respectively. In conclusion, our omics data proposed not only potential therapeutic targets, but also biomarkers for understanding the pathogenesis of severe COVID-19.

In this work, the contribution of different types of carbon blacks (N115, N330, N550, N660) and their primary and secondary thermally cracked recovered carbon blacks to the mechanical properties of NR composites was evaluated. The thermally cracked recovered carbon blacks were prepared by cracking the rubber composites at 500 °C and de-hybridizing them at 900 °C. The characterization of the thermally cracked recovered carbon blacks by scanning electron microscopy, Raman spectroscopy, and X-ray photoelectron spectroscopy showed that carbon blacks after primary and secondary thermal cracking recovery were more prone to aggregation and exhibited a higher degree of carbon defects. The number and type of functional groups on the surface of these carbon blacks were significantly reduced. For NR composites with pristine samples added, the mechanical properties and the bound rubber content tests showed that the mechanical properties of the NR composites became weaker with the increase in carbon black particle size. The bound rubber content also decreased with increased carbon black particle size. The mechanical properties of the NR composites reinforced with carbon black recovered by primary and secondary thermal cracking would therefore decrease. The results of AFM and DSC tests further confirmed the decreasing trend of bound rubber. The present work demonstrates the effect of bound rubber content variation on the mechanical properties of rubber, demonstrates the morphology of bound rubber more visually, and provides new insights into the reinforcement theory of CB.

Type 2 diabetes (T2D) is one of the most escalating global metabolic diseases, which is highly associated with insulin resistance (IR) and risk of combination with nonalcoholic fatty liver disease (NAFLD). Previous studies suggest that soluble klotho (sKL) could serve as a circulating hormone to mediate energy metabolism, but the detailed mechanism is poorly understood. In this study, we generated T2D models of wild-type (WT), sKL heterozygous (KL+/−), and sKL transgenic (TgKL) mice continuously fed a high-fat diet (HFD) and constructed L02 cell lines that stably overexpress sKL to investigate the effect of sKL on hepatic glucose and lipid metabolism. Surprisingly, we discovered that sKL deficiency resulted in exacerbated diabetic phenotypes and hepatic glucolipid metabolism disorders in HFD-fed KL+/− diabetic mice (KL+/− DM), whereas TgKL diabetic mice (TgKL DM) exhibited ameliorated diabetic phenotypes and decreased IR. Mechanistic studies in vitro and in vivo demonstrated that sKL could inhibit the PI3K/AKT/mTORC1 signaling to upregulate peroxisome proliferator-activated receptor α (PPARα) expression by directly interacting with type 1 insulin-like growth factor receptor (IGF1R) in HFD-fed T2D mice. Thus, sKL could improve hepatic glucolipid homeostasis to ameliorate diabetic phenotypes and lipid accumulation and may function as a potential therapeutic target for the treatment of T2D and reduce the risk of NAFLD.

Abstract Background Increasing numbers of studies have demonstrated that circulating tumor cells (CTCs) undergo a phenotypic change termed epithelial‐mesenchymal transition (EMT), and researchers have proposed that EMT might provide CTCs with increased potential to survive in the different microenvironments encountered during metastasis through various ways, such as by increasing cell survival and early colonization. However, the exact role of EMT in CTCs remains unclear. Methods In this study, we identified CTCs of 41 patients with gastric cancer using Cyttel‐CTC and im‐FISH (immune‐fluorescence in situ hybridization) methods, and tested the expression of EMT markers and ULBP1 (a major member of the NKG2D—natural killer [NK] group 2 member D—ligand family) on CTCs. Moreover, we investigated the relationship between the expression of EMT markers and ULBP1 on CTCs and gastric cancer cell lines. Results Our results showed that the CTCs of gastric cancer patients exhibited three EMT marker subtypes, and that the expression of ULBP1 was significantly lower on mesenchymal phenotypic CTCs (M+CTCs) than on epithelial phenotypic CTCs (E+CTCs). EMT induced by TGF‐β in vitro produced a similar phenomenon, and we therefore proposed that EMT might be involved in the immune evasion of CTCs from NK cells by altering the expression of ULBP1. Conclusions Our study indicated that EMT might play a vital role in the immune invasion of CTCs by regulating the expression of ULBP1 on CTCs. These findings could provide potential strategies for targeting the immune evasion capacity of CTCs.

Nitrogen doped carbon species (NC) was introduced into Fe based catalyst and applied into propane non-oxidative dehydrogenation. With characterizations of XRD, TEM and XPS, it was deduced that the NC modification induces the electrons transferred from NC species to Fe species, leading to electron-rich Fe species. Consequently, propane conversion and propene selectivity were increased by suppressing the by-products formation. It was also confirmed that such a modification strategy could be also extended to ones with different supports, iron precursors, and NC precursors.

The channel attention mechanism is widely used in deep learning. However, the existing channel attention mechanism directly performs the global average pooling and then full connection for all channels, which causes the local information to be ignored and the feature information cannot be reasonably assigned with the proper weights. This paper proposed a local channel attention module, based on the channel attention. This module focuses on the local information of the feature image, obtains the weight of each regional channel through convolution, and then integrates the information, so that the regional information can be fully utilized. Moreover, the local channel attention module is combined with the residual module, and the local channel attention residual network LSERNet is constructed to detect the abnormal state of the blast furnace tuyere image. With sufficient experiments on the collected datasets of the blast furnace tuyere, the results show that the proposed method can efficiently extract the feature information, and the recognition accuracy of the LSERNet model reached 98.59%. Further, our model achieved the highest accuracy, compared with SE-ResNet50, ResNet50, LSE-ResNeXt, SE-ResNeXt, and ResNeXt models.

Phosphate-solubilizing bacteria (PSB) can alleviate available phosphorus (AP)-deficiency without causing environmental pollution like chemical phosphate fertilizers. However, the research and application of PSB on the barren rocky soil is very rare. We screened six PSB from sweetpotato rhizosphere rocky soil. Among them, Ochrobactrum haematophilum FP12 showed the highest P-solubilizing ability of 1,085.00 mg/L at 7 days, which was higher than that of the most reported PSB. The assembled genome of PSB FP12 was 4.92 Mb with P-solubilizing and plant growth-promoting genes. In an AP-deficient environment, according to transcriptome and metabolomics analysis, PSB FP12 upregulated genes involved in gluconic acid synthesis and the tricarboxylic acid cycle, and increased the concentration of gluconic acid and malic acid, which would result in the enhanced P-solubilizing ability. Moreover, a series of experiments in the laboratory and field confirmed the efficient role of the screened PSB on significantly increasing AP in the barren rocky soil and promoting sweetpotato yield. So, in this study, we screened highly efficient PSB, especially suitable for the barren rocky soil, and explored the P-solubilizing mechanism. The research will reduce the demand for chemical phosphate fertilizers and promote the environment-friendly agricultural development.

Dickkopf-2 (DKK2) is an antagonist of canonical Wnt signaling, which is involved in various biological processes of development, such as epidermal appendage formation and eye development. To identify underlying effects of DKK2 during embryonic development, we generated a DKK2 homozygous knockout human embryonic stem cell (hESC) line through the CRISPR/Cas9 genome-editing technology. This cell line, which maintains normal stem cell morphology and stably expresses pluripotent markers, could provide an ideal platform for exploring the role of DKK2 in embryonic development. In addition, Zeocin selection combined with tiny clone picking might be a highly efficient way to generate gene-knockout hESC lines.

Recent studies combining neuroimaging with machine learning methods successfully infer an individual’s brain age, and its discrepancy with the chronological age is used to identify age-related diseases. However, which brain networks play decisive roles in brain age prediction and the underlying biological basis of brain age remain unknown. To answer these questions, we estimated an individual’s brain age in the Southwest University Adult Lifespan Dataset (N = 492) from the gray matter volumes (GMV) derived from T1-weighted MRI scans by means of Gaussian process regression. Computational lesion analysis was performed to determine the importance of each brain network in brain age prediction. Then, we identified brain age-related genes by using prior brain-wide gene expression data, followed by gene enrichment analysis using Metascape. As a result, the prediction model successfully inferred an individual’s brain age and the computational lesion prediction results identified the central executive network as a vital network in brain age prediction (Steiger’s Z = 2.114, p = 0.035). In addition, the brain age-related genes were enriched in Gene Ontology (GO) processes/Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways grouped into numbers of clusters, such as regulation of iron transmembrane transport, synaptic signaling, synapse organization, retrograde endocannabinoid signaling (e.g., dopaminergic synapse), behavior (e.g., memory and associative learning), neurotransmitter secretion, and dendrite development. In all, these results reveal that the GMV of the central executive network played a vital role in predicting brain age and bridged the gap between transcriptome and neuroimaging promoting an integrative understanding of the pathophysiology of brain age.

The efficient and clean conversion of light alkanes is a research hotspot in the petrochemical industry, and the development of effective and eco-friendly non-noble metal-based catalysts is a key factor in this field. Among them, gallium is a metal component with good catalytic performance, which has been extensively used for light alkanes conversion. Herein, we critically summarize recent developments in the preparation of gallium-based catalysts and their applications in the catalytic conversion of light alkanes. First, we briefly describe the different routes of light alkane conversion. Following that, the remarkable preparation methods for gallium-based catalysts are discussed, with their state-of-the-art application in light alkane conversion. It should be noticed that the directional preparation of specific Ga species, strengthening metal-support interactions to anchor Ga species, and the application of new kinds of methods for Ga-based catalysts preparation are at the leading edge. Finally, the review provides some current limitations and future perspectives for the development of gallium-based catalysts. Recently, different kinds of Ga species were reported to be active in alkane conversion, and how to separate them with advanced in situ and ex situ characterizations is still a problem that needs to be solved. We believe that this review can provide base information for the preparation and application of Ga-based catalysts in the current stage. With these summarizations, this review can inspire new research directions of gallium-based catalysts in the catalysis conversion of light alkanes with ameliorated performances.

ABSTRACTSARS-CoV-2 caused a major outbreak of severe pneumonia (COVID-19) in humans. Viral RNA was detected in multiple organs in COVID-19 patients. However, infectious SARS-CoV-2 was only isolated from respiratory specimens. Here, infectious SARS-CoV-2 was successfully isolated from urine of a COVID-19 patient. The virus isolated could infect new susceptible cells and was recognized by its’ own patient sera. Appropriate precautions should be taken to avoid transmission from urine.

Summary: Non-oxidative propane dehydrogenation (PDH) is an attractive reaction from both an industrial and a scientific viewpoint because it allows direct large-scale production of propene and fundamental analysis of C-H activation respectively. The main challenges are related to achieving high activity, selectivity, and on-stream stability of environment-friendly and cost-efficient catalysts without non-noble metals. Here, we describe an approach for the preparation of supported ultrasmall ZnO nanoparticles (2–4 nm, ZnO NPs) for high-temperature applications. The approach consists of encapsulation of NPs into a nitrogen-doped carbon (NC) layer in situ grown from zeolitic imidazolate framework-8 on a Silicalite-1 support. The NC layer was established to control the size of ZnO NPs and to hinder their loss to a large extent at high temperatures. The designed catalysts exhibited high activity, selectivity, and on-stream stability in PDH. In the steelmaking industry, the state of the blast furnace tuyere is an important basis for obtaining the internal information of the blast furnace. Traditional detections mainly rely on manual experience judgment, which is a time-consuming and tiring procedure for a human. In order to improve the efficiency of the detection, this paper is devoted to applying artificial intelligence methods to blast furnace anomaly detection. However, because of the low imaging degree of the abnormal state monitoring of the furnace mouth, the difference in the abnormal category is inconspicuous, and it is difficulty to extract the features with the existing intelligent models. To solve these problems, a novel and stable method is proposed in this paper to classify the image recognition of the abnormal state of the tuyere into one category; this is a new architecture that combines multiple technologies. For the fine-grained image classification task, an improved abnormal state recognition algorithm of the blast furnace tuyere based on the channel attention residual mechanism is proposed. In the model, the dataset is augmented by rotating it at random angles to balance the amount of data in each category; then, the residual module is used to integrate high- and low-order feature information and optimize the network; then, the multi-layer channel attention module is added based on the channel attention residual mechanism, and it obtains the optimal parameter combination of the model through k-fold cross-validation. Moreover, the number of channels was reduced by half after channel fusion, which could effectively reduce the model parameters and model complexity. It is shown in our experiments that the proposed method has an accuracy rate of 97.10% in identifying the abnormal state of the tuyere in our collection of blast furnace tuyere datasets. In order to test the performance of the proposed method, some existing models, such as SERNet, ResNeXt, and repVGG, are involved for comparison, and the proposed method has a better classification effect in comparison to them.

Photoelectrocatalytic (PEC) oxidation is an advanced technology that combines photocatalytic oxidation (PC) and electrolytic oxidation (EC). PEC activity can be greatly enhanced by the PC and EC synergy effect. In this work, novel copolymerized g-C3N4 (denoted as CNx)/TiO2 core-shell nanowire arrays were prepared by chemical vapor deposition. CNx were deposited on the surface of TiO2 nanowire arrays using organic monomer 4,5-dicyanidazole and dicyandiamide as copolymerization precursor. TiO2 nanowire arrays provide a direct and fast electron transfer path, while CNx is a visible light responsive material. After CNx deposition, the light response range of TiO2 is broadened to 600 nm. The deposition of CNx shell effectively improves the PC efficiency and PEC efficiency of TiO2. Under visible light irradiation and 1 V bias potential, the rate constant k of PEC degradation of CNx/TiO2 core-shell nanowire arrays is 0.0069 min−1, which is 72% higher than that of pure TiO2 nanowires. The built-in electric field formed in the interface between TiO2 core and CNx shell would effectively promote photogenerated charge separation and PEC activity.
