Fast tool servo and slow slide servo-based diamond turning are very important and widely used machining methods for freeform surfaces. In diamond turning of freeform surfaces, the research on tool path generation and machined surface topography prediction is generally for diamond turning processes using zero rake angle tools. But diamond turning with non-zero rake angle tools is also of great importance, for which very little research has been done on tool path planning and surface topography modeling. Therefore, based on coordinate transformations, this paper develops a new tool path generation method and discusses the tool geometry parameter selection for diamond turning utilizing non-zero rake angle tools. In addition, a new surface topography prediction model based on the actual position and orientation of the tool cutting edge is constructed for the machined surface. The machining experiments of a typical freeform surface are conducted based on the calculated tool paths and necessary critical tool geometry parameters. The measured results validate the developed tool path generation and tool geometry selection methods, and the constructed surface topography model.

Sex pheromones are deemed to play a significant role in sexual communication of most insects. Although many sex pheromone components in mirid bugs have been identified, the roles of odorant receptors in sex pheromone perception in Adelphocoris spp. (Hemiptera: Miridae) remain unknown so far. Here, AlinOR33, a candidate sex pheromone receptor in Adelphocoris lineolatus was functionally characterized. Phylogenetic analysis showed that AlinOR33 clustered with the sex pheromone receptor AlucOR4 fromApolygus lucorum. Quantitative real-time PCR measurement revealed that the expression of AlinOR33 increased gradually from nymph to adult stage and reached its peak in the antennae of 3-day-old mated male bugs. The subsequent in situ hybridization demonstrated that AlinOR33 was mainly expressed in sensilla trichoid on the antennae of A. lineolatus. In the two-electrode voltage clamp recordings, AlinOR33/AlinOrco was specifically tuned to four sex pheromone components including butyl butyrate, hexyl hexanoate, trans-2-hexenyl butyrate and hexyl butyrate, and especially most sensitive to the major component trans-2-hexenyl butyrate. After dsAlinOR33 injection, the electroantennogram responses of males to four sex pheromone components were reduced significantly (∼50%). Compared to control bugs, dsAlinOR33-injected male bugs almost lost behavioral preference for trans-2-hexenyl butyrate. Furthermore, the wingbeat frequency of dsAlinOR33-injected male bugs notably declined. Therefore, we conclude that as a candidate sex pheromone receptor, AlinOR33 plays essential roles in the sexual behavior of A. lineolatus.

Studying the chemical composition and characteristic differences of soil dissolved organic matter (DOM) is significant for understanding the mechanism of Cd immobilization by soil amendments. Soil amendments have been widely applied to contaminated farmlands to reduce the accumulation of heavy metals in crops, but the spectral characteristics of DOM in soils under amendment regulation have rarely been studied. Typical Cd-contaminated paddy soil from South China was collected, three categories of amendments (organic-based, inorganic-based, and lime-based, a total of 11 types) were applied, rice planting pot trials were done, and the effects of different amendments on soil DOM were investigated. The spectral characteristics of rhizosphere soil DOM under the regulation of different amendments were comparatively analyzed using UV-Vis spectroscopy, 3D fluorescence spectroscopy, and parallel factor analysis (PARAFAC). The results showed that the lime-based amendments increased the soil pH and promoted the dissolution of organic matter from the soil solid phase, resulting in a significant increase in soil DOM content. Compared with that of the control, organic-based amendments increased the relative molecular weight and recent autochthonous origin contribution of soil DOM, inorganic-based amendments increased the aromatic content and hydrophobic components of soil DOM, and lime-based amendments increased the chromogenic components and the degree of humification of soil DOM. Four fluorescence components, C1 (255/465), C2 (325/400), C3 (275/390), and C4 (240/460), were identified using PARAFAC and verified with the OpenFluor database, all of which were humic-like. Two types of spectra corroborated with each other, indicating that soil DOM dominated by humus-like matter originated from terrestrial source input. The correlation analysis showed that the fluorescent component C4 of soil DOM could be used to predict Cd accumulation in brown rice in the red limestone soil-rice system. These results will provide a reference for the selection of the appropriate soil amendments.

The Lunggar iron deposit belongs to the Bangong-Nujiang metallogenic belt and is located in central Lhasa on the Tibetan Plateau. In the Lunggar deposit, iron mineralization formed in the skarnization contact zone between the Early Cretaceous granodiorite and the late Permian Xiala Formation limestone. In this study, we achieved detailed zircon U-Pb-Hf isotopes and mineral chemistry for the Early Cretaceous granodiorite. Zircon U-Pb dating results indicate that the Early Cretaceous granodiorite emplaced at ca. 119 Ma. Based on the trace elements in zircons and the mineral chemical composition of amphibole and biotite, the Early Cretaceous granodiorite was believed to form under condition of high temperature (>700 °C), low pressure (100–400 MPa), and relatively high oxygen fugacity (lg/O2)(−13.6 to −13.9) and H2O content (4%–8%). Zircon trace elements, Hf isotope and biotite chemistry collectively reveal that significant juvenile mantle-derived magmas contributed to the source of the granodiorite. The relatively high log/O2 and shallow magma chamber are beneficial for skarn iron mineralization, implying remarkable potential for further prospecting in the Lunggar iron deposit.

To assess the effects of bed-sharing experiences in infancy on sleep patterns and sleep problems at 2 years of age.A total of 1564 children from an ongoing Shanghai Maternal-Child Pairs Cohort were included. Bed-sharing experiences were collected when children were 2, 6, and 24 months old via caregiver-completed questionnaires (whether caregivers shared a bed with children during the night), and children's bed-sharing experiences were classified as follows: no bed-sharing, early-only bed-sharing, late-onset bed-sharing, and persistent bed-sharing. Sleep outcomes at month 24 were assessed using the Brief Infant Sleep Questionnaire. Sleep patterns and problems were compared among the 4 types of bed-sharing experiences.Of the 1564 infants, 10.10% had no bed-sharing, 18.35% had early-only, 27.94% had late-onset, and 43.61% had persistent bed-sharing. Compared with children with no bed-sharing, children with late-onset and persistent bed-sharing had shorter nighttime sleep durations and longer daytime sleep durations (P .05) and were more likely to snore (aOR 1.87 [95% CI 1.25-2.79]; aOR 1.68 [95% CI 1.14-2.47]) and have sleep onset difficulty (aOR 2.06 [95% CI 1.37-3.09]; aOR 2.07 [95% CI 1.41-3.05]). However, caregivers of infants in the late-onset and persistent bed-sharing groups perceived less problematic sleep (aOR 0.38 [95% CI 0.26-0.56] and aOR 0.40 [95% CI 0.28-0.58]).Bed-sharing is a common experience among Chinese children. Although bed-sharing may reduce caregivers' perception of children's problematic sleep, late-onset or persistent bed-sharing in infancy is associated with sleep problems at 2 years of age.

Under the spirit of the quality criteria of talent cultivating in Engineering Education Professional Certification (EEPC) and the nature of Engineering Graphics course, the objective of this course was adjusted from the traditional knowledge imparting to abilities cultivating. A series of participatory sections were designed in the process of Engineering Graphics course. Basically, students were divided into groups of about five of each, and typical topics were designed and assigned to individual students or study groups, and the group members discussed and completed the tasks. Making solids practice was designed as making real objects according to the dimensions students gave, which is used to deepen the understanding of the relationship between knowledge and real application, and train the practical ability of students. Representation scheme practice was designed to cultivate the ability of using knowledge that the students learned to solve major related complex problems. Retrieving extension materials practice was designed to train the ability of students to retrieve, select and induct needed materials. Creative modeling section was designed to develop innovative spirit of students by virtually constructing complex models.

To explore the effects of prenatal exposure to polybrominated diphenyl ethers (PBDEs) on placental size and birth outcomes.Based on the perspective Wenzhou Birth Cohort, this nested case-control study included 101 fetal growth restriction (FGR) and 101 healthy newborns. Maternal serum samples were collected during the third trimester and measured for PBDEs by gas chromatography tandem mass spectrometry. The basic information of mother-newborn pairs was collected from questionnaires, whereas the placental size and birth outcomes of newborns were obtained from hospital records.A total of 19 brominated diphenyle ether (BDE) congeners were detected in maternal serum samples. Higher concentrations of BDE-207, -208, -209, and ∑A negative association was found between PBDE levels in maternal serum and placental size and birth outcomes. Prenatal PBDE exposure may be associated with elevated risk of the incidence of FGR birth.

The authors in this paper, by using SEM model and other methods, analyze the mechanism of how the characteristics of enterprises affect the innovation of taxation policy from the qualitative and quantitative aspects of the questionnaire survey from 1000 different tax-type enterprises in Guangdong Free Trade Zone (taking Nansha for example). Five theoretical research hypotheses were tested after exploring. The research shows that through the modeling and calculation of structural equations, the current taxpayer enterprises in the Guangdong (Nansha) Free Trade Zone are increasing their demands for tax incentive innovation, propaganda channel innovation, knowledge counseling innovation and service optimization initiatives. However, from the point of view from the intensity of the appeal (standardized regression coefficient), the general taxpayer enterprises are significantly higher than the small-scale taxpayer ones. These new studies have found that it will provide a basis for deepening tax incentives, innovation and collection, and taxation services to propose tax policy recommendations to promote the construction of China’s free trade zone.

Based on the total and available concentrations of heavy metal elements in agricultural soil, and the concentrations of heavy metals in rice collected from Daye city, Hubei province, and the thresholds of available concentrations of heavy metals were derived by species sensitivity distribution (SSD). The over-standard rates of Cd and Cu in the soil were 90.7% and 42.6%, respectively, which indicates that agricultural soil in Daye city exhibits signs of Cd and Cu pollution. The rates of Cd and Pb were 50.9% and 89.8% in brown rice samples, respectively, which exceeds the safety standards of agricultural products. Understory vegetation hosts high biodiversity and plays a critical role in the ecosystem processes of boreal forests. However, the drivers of understory plant diversity in this high-latitude ecosystem remain uncertain. To investigate the influences of forest type and latitude on understory beta diversity at different scales, we quantified the species composition of Vaccinium uliginosum Linnaeus communities under broadleaf and coniferous forests at two latitudes at the quadrat (2 &times, 2 m) and plot (10 &times, 10 m) scales in the Greater Xing&rsquo, an Mountains, NE China. At the quadrat scale, species alpha diversity of V. uliginosum communities was higher in broadleaf forests than that in coniferous forests at both latitudes. The differences in species beta diversity (the S&oslash, rensen&rsquo, s dissimilarity) in two forest types depended on the latitude: beta diversity in broadleaf forests was higher than that in coniferous forests at the higher latitude, while beta diversity in coniferous forests was higher at the lower latitude. At the plot scale, alpha and beta diversity of V. uliginosum communities decreased from broadleaf forests to coniferous forests at the higher latitude, and they did not show significant differences between forest types at the lower latitude. These results indicate the interactive effects of forest type and latitude on beta diversity of understory vegetation. Moreover, the influences of forest type and latitude on species alpha and beta diversity were different across the two spatial scales, suggesting that the assembly mechanisms underlying species diversity may be different at different scales. Understanding the maintenance of understory vegetation diversity will benefit the conservation and management of boreal forests.

The presence of patients with diverse pathologies in hospitals results in an environment that can be rich in various microorganisms including respiratory and enteric viruses, leading to outbreaks in hospitals or spillover infections to the community. All hospital patients are at risk of nosocomial viral infections, but vulnerable groups such as older adults, children and immuno-compromised/-suppressed patients are at particular risk of severe outcomes including prolonged hospitalization or death. These pathogens could transmit through direct or indirect physical contact, droplets or aerosols, with increasing evidence suggesting the importance of aerosol transmission in nosocomial infections of respiratory and enteric viruses. Factors affecting the propensity to transmit and the severity of disease transmitted via the aerosol route include the biological characteristics affecting infectivity of the viruses and susceptibility of the host, the physical properties of aerosol particles, and the environmental stresses that alter these properties such as temperature and humidity. Non-specific systematic and individual-based interventions designed to mitigate the aerosol route are available although empirical evidence of their effectiveness in controlling transmission of respiratory and enteric viruses in healthcare settings are sparse. The relative importance of aerosol transmission in healthcare setting is still an on-going debate, with particular challenge being the recovery of infectious viral bioaerosols from real-life settings and the difficulty in delineating transmission events that may also be a result of other modes of transmission. For the prevention and control of nosocomial infections via the aerosol route, more research is needed on identifying settings, medical procedures or equipment that may be associated with an increased risk of aerosol transmission, including defining which procedures are aerosol-generating; and on the effectiveness of systematic interventions on aerosol transmission of respiratory and enteric viruses in healthcare settings., Highlights • Respiratory viruses have been recovered in the air in hospitals despite existing systematic and personal level interventions. • Studies on the relative contribution of different routes of transmission for respiratory and enteric viruses are lacking. • Physical filtration efficiencies may not directly translate to effectiveness in mitigating virus transmission. • More effectiveness studies with biological measures (e.g. viral load and infection events) as outcomes are needed.

Acute lung injury (ALI) is a common complication following intestinal ischemia/reperfusion (II/R) injury and contributes to the associated high mortality rate. However, the underlying mechanism is poorly understood and treatments are limited. RNA interference (RNAi) has been demonstrated to provide a promising disease treatment strategy both in vitro and in vivo. Therefore, the present study aimed to test whether blocking the proinflammatory cytokine IL‑6 by RNAi may protect the lungs from remote organ injury following II/R, and to investigate the potential underlying mechanisms. A total of 176 adult healthy male Sprague‑Dawley rats were randomly divided into sham, II/R, negative‑control and IL‑6‑short hairpin (sh)RNA groups. The rats underwent II/R injury with occlusion of the superior mesenteric artery and coeliac artery to induce ischemia for 40 min, and were subsequently reperfused for 0‑48 h. The negative‑control group received a control lentiviral vector containing scrambled or non‑specific sequences, and the IL‑6‑shRNA groups were administered with a vector containing an IL‑6 shRNA sequence to affect RNAi‑mediated knockdown of IL‑6. ALI severity was determined by lung edema (lung wet/dry ratio) and histological analysis (lung injury scores). IL‑6 localization, and mRNA and protein expression levels, were detected by immunofluorescence, reverse transcription‑quantitative polymerase chain reaction and western blot analysis, respectively. IL‑10 expression induced by IL‑6 knockdown in lung tissues was additionally detected. IL‑6 RNAi was revealed to significantly reduce the expression of IL‑6, which was associated with upregulated IL‑10 expression in lung tissues. Consequently, the severities of ALI and edema induced by II/R were substantially improved. In conclusion, the present study demonstrated that IL‑6 RNAi may protect the lung from ALI induced by II/R, and that this protective role may be associated with upregulation of IL‑10. These findings may contribute to the development of an IL‑6‑RNAi‑based therapeutic strategy for the treatment of II/R‑induced ALI.

To explore the optimum conditions for the extraction of anthocyanins and flavonols from bog bilberry (Vaccinium uliginosum L.) marc on a single-factor experimental basis, a response surface methodology was adopted for this intensive study. The extraction procedure was carried out in a Waring blender and followed an ultrasonic bath, and the natural antioxidant carnosic acid was added to inhibit oxidation. The optimum extraction conditions were as follows: a volume fraction of ethanol of 70%, an antioxidant content of 0.02% (the mass of sample) carnosic acid, a liquid&ndash, solid ratio of 16 mL/g, a homogenization time of 3 min, a reaction temperature of 55 &deg, C, an ultrasound irradiation frequency of 80 kHz, an ultrasound irradiation power of 200 W, and an ultrasound irradiation time of 40 min. Satisfactory yields of anthocyanins (13.95 &plusmn, 0.37 mg/g) and flavonols (3.51 &plusmn, 0.16 mg/g) were obtained. The experimental results showed that the carnosic acid played an effective antioxidant role in the extraction process of anthocyanins and flavonols with a green and safety guarantee.

Coxsackievirus A16 (CV-A16) is one of the viruses that is most frequently associated with hand-foot-and-mouth disease (HFMD). Previous studies have shown that CV-A16 infections are mostly self-limiting, but in recent years, it has been gradually found that CV-A16 infections can also induce neurological complications and eventually cause death in children with HFMD. Moreover, no curative drugs or preventative vaccines have been developed for CV-A16 infection. Therefore, it is particularly important to investigate the mechanism of CV-A16 infection-induced neuropathy. In the current study, transcriptome sequencing technology was used to identify changes in the transcriptome of SH-SY5Y cells infected with CV-A16, which might hide the mechanism of CV-A16-induced neuropathology. The transcriptome profiling showed that 82,406,974, 108,652,260 and 97,753,565 clean reads were obtained in the Control, CV-A16-12 h and CV-A16-24 h groups, respectively. And it was further detected that a total of 136 and 161 differentially expressed genes in CV-A16-12 h and CV-A16-24 h groups, respectively, when compared with Control group. Then, to explore the mechanism of CV-A16 infection, we focused on the common differentially expressed genes at different time points of CV-A16 infection and found that there were 34 differentially expressed genes based on which clustering analysis and functional category enrichment analysis were performed. The results indicated that changes in oxidation levels were particularly evident in the GO term analysis, while only the “Gonadotropin-releasing hormone receptor pathway” was enriched in the KEGG pathway analysis, which might be closely related to the neurotoxicity caused by CV-A16 infection. Meanwhile, the ID2 closely related to nervous system has been demonstrated to be increased during CV-A16 infection. Additionally, the data on differentially expressed non-protein-coding genes of different types within the transcriptome sequencing results were analyzed, and it was speculated that these dysregulated non-protein-coding genes played a pivotal role in CV-A16 infection. Ultimately, qRT-PCR was utilized to validate the transcriptome sequencing findings, and the results of qRT-PCR were in agreement with the transcriptome sequencing data. In conclusion, transcriptome profiling was carried out to analyze response of SH-SY5Y cells to CV-A16 infection. And our findings provide important information to elucidate the possible molecular mechanisms which were linked to the neuropathogenesis of CV-A16 infection.

We investigated roles of PI3K-AKT-mTOR pathway in recovery from general anesthesia. Sprague-Dawley rats divided into five groups: saline+artificial cerebrospinal fluid (ACSF; Group A), ketamine+ACSF (Group B), ketamine+IGF-1 (Group C), ketamine+PI3K inhibitor (Group D), and PI3K/Akt agonists (Group E). Proportion of δ waves on ECoGs was recorded. Rats were tested for duration of loss of righting reflex (LORR), ataxic period and behavior in Morris water maze. mRNA and protein expression of members of PI3K-AKT-mTOR pathway were measured by RT-qPCR and Western blots. Histopathologic changes in hippocampal tissues observed by HE staining. We found that the proportion of δ waves decreased in Group C, while increased in Group D compared with Group B; the durations of LORR and ataxic period were shorter in Group C, but longer in Group D. In Morris water maze, escape latency (EL) and duration and frequency of staying on platform was shorter in Group C and longer in Group D than in Group B. Group A exhibited low expression of proteins in PI3K-AKT-mTOR pathway, while p-AKT, p-mTOR and p-P70S6K expression increased in cerebral cortex, brain stem, and thalamus in Group C. By contrast, expression of those proteins was lower in Group D than Group B. Those proteins expressions were higher in Group E than in Group A. HE staining showed that anesthesia may induce cell apoptosis in rat hippocampal CA1 areas, and PI3K/Akt agonists could inhibit apoptosis. Our results suggest that activation of PI3K-AKT-mTOR pathway may promote recovery from general anesthesia and enhance spatial learning and memory.

Objective To investigate the effects of lasR/rhlR gene on Foxp3, TGF-β1 and IL-10 of lung tissue in rat tracheal intubation model with biofilm infection of Pseudomonas aeruginosa (Ps. aer) wild strain (PAO1) and quorum sensing (QS) deficient strain (ΔlasRΔrhlR). Methods Twenty-one SD rats were randomly assigned into 3 groups (7 each): ΔlasRΔrhlR-treated group, PAO1-treated group and sterile control group. Biofilms (BF) were cultured in vitro, and the BF coated tube (infected respectively with Ps. aer PAO1 strain, ΔlasRΔrhlR strain, or with asepsis) was inserted into the trachea to establish the rat model. The rats were sacrificed on the 7th day after intubation. Colony count of lung tissue homogenate (cfu) and lung HE staining were performed, and IL-10 content in bronchoalveolar lavage fluid (BALF), TGF-β1 in lung tissue, and the expression of Foxp3 mRNA in lung cells were determined. Intestinal ischemia and reperfusion (II/R) injury often triggers severe injury in remote organs, with the lungs being considered the main target. Excessive elevation of proinflammatory cytokines is a major contributor in the occurrence and development of II/R‑induced acute lung injury (ALI). Therefore, the present study aimed to investigate whether blocking tumor necrosis factor‑α (TNF‑α) expression could protect the lungs from injury following II/R, and to explore the possible underlying mechanism involving interleukin‑10 (IL‑10). Briefly, II/R was induced in rats by 40 min occlusion of the superior mesenteric artery and celiac artery, followed by 8, 16 or 24 h of reperfusion. Subsequently, lentiviral vectors containing TNF‑α short hairpin (sh)RNA were injected into the right lung tissues, in order to induce TNF‑α knockdown. The severity of ALI was determined according to lung injury scores and lung edema (lung wet/dry weight ratio). The expression levels of TNF‑α were analyzed by quantitative polymerase chain reaction (qPCR), western blotting and immunofluorescence (IF) staining. IL‑10 expression, in response to TNF‑α knockdown, was detected in lung tissues by qPCR and IF. The results detected marked inflammatory responses, and increased levels of lung wet/dry weight ratio and TNF‑α expression, in the lungs of II/R rats. Conversely, treatment with TNF‑α shRNA significantly alleviated the severity of ALI and upregulated the expression levels of IL‑10 in lung tissues. These findings suggested that TNF‑α RNA interference may exert a protective effect on II/R‑induced ALI via the upregulation of IL‑10. Therefore, TNF‑α knockdown may be considered a potential strategy for the prevention or treatment of ALI induced by II/R in future clinical trials.

Diamondback moth, Plutella xylostella L., is a very important pest of cruciferous vegetables causing excessive economic losses worldwide. Bioactivities of halo-, diazo-, and cyclopropane acetates of P. xylostella sex pheromone have been evaluated using electrophysiology and enzyme inhibition assays.A total of 23 sex pheromone analogs of P. xylostella were designed and synthesized and the result shows that (11Z)-hexadec-11-en-1-yl 2,2,2-trifluoroacetate, (11Z)-hexadec-11-en-1-yl 2,2,3,3,3-pentafluoropropanoate, and (11Z)-hexadec-11-en-1-yl trifluoromethanesulfonate elicited potential inhibitory effects at all doses tested in the electrophysiology and enzyme inhibition assays. Interference of locating the sex pheromone source was found strongest when these three analogs were mixed with the sex pheromone at a 10:1 ratio. In addition, field test showed that the rate of mating disruption was over 90% when (11Z)-hexadec-11-en-1-yl 2,2,2-trifluoroacetate or (11Z)-hexadec-11-en-1-yl 2,2,3,3,3-pentafluoropropanoate was mixed with the sex pheromone at a 10:1 ratio.Two sex pheromone antagonists were screen out by electrophysiology, enzyme inhibition assays, wind tunnel and field tests. We believe that these antagonists could be used to establish a novel eco-friendly measure to control P. xylostella and provide evidence for clarifying the specific functions and molecular mechanisms of sex pheromone antagonists. © 2018 Society of Chemical Industry.

To evaluate whether mono (2-ethylhexyl) phthalate (MEHP) affects genomic DNA methylation and the methylation status of some specific genes such as patched gene (PTCH) and smoothened gene (SMO) in LNCaP cells.LNCaP cells were treated with MEHP (0, 1, 5, 10, and 25 μmol/L) for 3 days. An ELISA assay was preformed to detect genomic methylation, including 5-methylcytosine (5-mC) and 5-hydroxymethylcytosine (5-hmC) content. A pyrosequencing assay was applied to assess DNA methylation in PTCH and SMO gene promoters. The correlation between DNA methylation and gene expression was assessed.The proportion of cytosines with 5-mC methylation in LNCaP cells was significantly decreased by MEHP (1, 5, 10, and 25 μmol/L) in a dose-dependent manner (P0.01). For genes in the Hedgehog pathway, there was no significant MEHP concentration-dependent difference in the DNA methylation of PTCH and SMO.MEHP might affect the progression of prostate cancer through its effect on global DNA methylation.

Background Recruit training sites are places with a high incidence of respiratory infectious diseases. Effective surveillance for acute respiratory infectious diseases in a recruit training site is an important way to prevent disease outbreaks. Methods Eight hundred recruits (722 males and 78 females) enlisted in autumn 2015 received a background survey within 24 h of settlement at the recruit training site, including their general personal information, vaccination history, mental status and clinical symptoms. Then, nasopharyngeal swabs of these recruits were collected to detect common respiratory pathogens [influenza virus type A, influenza virus type B, adenovirus (Adv), human respiratory syncytial virus, human bocavirus and human metapneumovirus] by PCR. In addition, fasting venous blood was collected in the morning for adenovirus IgG antibody detection. During the three months of training, the recruits were monitored for symptoms of respiratory infection, and nasopharyngeal swabs were collected from those with an axillary temperature ≥38 °C and other respiratory symptoms within 4 h of symptom onset. Samples were further examined by PCR. Results Among the 795 effective nasopharyngeal swab samples collected during survey, two cases of group C type 1 adenovirus were identified by PCR. During the 3 months of training, fever and respiratory symptoms occurred in 39 recruits (incidence rate of 4.9%) and 5 cases of adenovirus were detected (positive rate of 12.8%). Genotyping showed 3 cases of type 4 adenovirus and 2 of type 3 adenovirus. No type 7, 14 or 55 adenovirus was detected. The Adv-IgG positive rate of recruits was 48.2%. Among the 5 Adv positive cases with fever and respiratory symptoms, 4 were Adv-IgG positive. Conclusion The pathogen carrier rate in recruits was low, and only group C adenovirus, which causes mild infection in humans, was detected. No respiratory outbreak was observed at the recruit training site, and sporadic cases were mainly caused by type 3 and type 4 adenoviruses.

Spinal cord injury (SCI) often results in motor disability concomitant with limit neuroplasticity; the underlying mechanism, however, is still unclear. This study established spinal cord transection rats model (T10), then performed cDNA microarray analysis and found that vimentin located in astrocytes was increased significantly in scar tissues after transection. To understand the role of vimentin and it's mechanism of regulation, RNA interference and luciferase assay were used. Vimentin knockdown in the scar tissues showed a significant improvement on locomotor function in hindlimbs, while vimentin overexpression exhibited an opposite effect. In vitro, vimentin downregulation or overexpression can effectively inhibit or increase astrogliosis, respectively. Moreover, by using biological informatics technology, we predicted that vimentin may be as the target of micRNA138 (miR-138), and confirmed that miR-138 could regulate vimentin by luciferase activity assay. The present results not only validated the exact role of vimentin in transected spinal cord, but also exhibited a novel regulation mechanism, in which miR-138 may regulate vimentin to promote neuroplasticity. It, therefore, provides a novel target for gene drug discovery based on miRNA-138 or vimentin for the treatment of SCI in the future clinic trial.

Mutants with overexpression of α-acetolactate synthase (ALS), α-acetolactate decarboxylase, and acetoin reductase (AR), either individually or in combination, were constructed to improve 2,3-butanediol (2,3-BD) production in Klebsiella pneumoniae. The recombinant strains were characterized in terms of the enzyme activity, 2,3-BD yield, and expression levels. The recombinant K. pneumoniae strain (KG-rs) that overexpressed both ALS and AR showed an improved 2,3-BD yield. When cultured in the media with five different carbon sources (glucose, galactose, fructose, sucrose, and lactose), the mutant exhibited higher 2,3-BD productivity and production than the parental strain in all the tested carbon sources except for lactose. The 2,3-BD production of KG-rs in a batch fermentation with glucose as the carbon source was 12% higher than that of the parental strain.

Acute lung injury (ALI), induced by intestinal ischemia reperfusion (II/R) injury, is characterized by pulmonary edema and inflammation. Aquaporin 4 (AQP4), has been pointed out recently involving in edema development. Previous studies have shown that p38 mitogen activated protein kinase (MAPK) activation resulted in lung inflammation, while p38 MAPK inhibitor can alleviate the pathology injury of lung tissue. However, the regulated mechanism of p38 MAPK in ALI induced by II/R is unclear. In this study, we established II/R rats' model by clamping the superior mesenteric artery (SMA) and coeliac artery (CA) for 40min and subsequent reperfusion for 16h, 24h, 48h. Subsequently, SB239063, a specific inhibitor of the activity of p38 MAPK, was injected (10mg/kg) intraperitoneally 60min before the operation. The severity of ALI was determined by histology analysis (HE staining and ALI scoring) and lung edema (lung wet/dry weight ratio) assessment. Western blot (WB) was applied to detect the expression level of AQP4 and phosphorylated (P)-p38 MAPK, and the localization of AQP4 was detected by immunofluorescent staining (IF). We found that AQP4 could express in the lung tissue. II/R could significantly induce lung injury, confirmed by lung injury scores and lung wet/dry weight ratios. The level of P-p38 MAPK and AQP4 were largely up-regulated in lung tissues. Moreover, inhibition of p38 MAPK activity could effectively down-regulate AQP4 expression and diminish the severity of II/R-induced ALI. These novel findings suggest that inhibition of p38 MAPK function should be a potential strategy for the prevention or treatment of ALI, by targeting AQP4 in future clinic trial.

In this paper, secondary variable is selected with this mechanism analysis and information processing technology of choice, the establishment of the ball fast and accurate output of neural network soft sensor model can not only contribute to estimate the steady-state mill and ball mill on the dynamic process can effectively contribute to an online estimate, the soft-sensor model has a strong possibility. At the same time，it provides a reference for the soft measurement of power plant other parameters. This modeling method has portability.

To observe the in vivo effects of oxysophoridine on hepatocellular carcinoma in mice and to study the related mechanisms.C57BL mice were inoculated with mouse hepatoma H22 cells subcutaneously, then divided into 5 groups (14 per group), and treated with oxysophoridine (50, 100, or 150 mg/kg) or cisplatin (4 mg/kg) for 10 days. Inhibitory rate of tumor, body weight gain, and influence indices on internal organs (liver, spleen and thymus) were evaluated. The differentially expressed genes between the oxysophoridine-treated group, and the control group were analyzed using cDNA microarray and quantitative real-time PCR (qRT-PCR) experiments.Compared with the tumor weight of the control group (2.75±0.66 g), oxysophoridine significantly suppressed hepatocellular carcinoma growth in mice (P0.01), with 0.82±0.36 g, 0.57±0.22 g, and 1.22±0.67 g for the tumor weight in the low, moderate, and high dose treatment group, respectively. The moderate dose led to the highest inhibitory rate, 79.3%. Observation of body weight gain and influence on three organs showed that compared with cisplatin, oxysophoridine produced fewer side effects in vivo. cDNA microarray and qRT-PCR showed that the most significant differentially expressed genes in the tumor samples of oxysophoridine-treated mice were mostly involved in regulating apoptosis, with the Tnfrsf11b (osteoprotegerin) gene being the most significantly affected.Oxysophoridine was a promising compound for developing drugs against hepatocellular carcinoma, and its anti-hepatoma effect was probably related to osteoprotegerin activation.

Background. Quorum sensing (QS) systems play an important role in modulating biofilm formation. Recent studies have found that the QS molecules had complex effects on the host immune systems. In addition, regulatory T cells (Tregs), known as important negative regulators in the immune system, have been found upregulated in multiple chronic infections. Therefore, the QS systems were hypothesized to be involved in modulating Tregs in biofilm-associated infections.Object. To explore the effects of QS systems on Tregs in catheter-relatedPseudomonas aeruginosabiofilm infection rat models.Method. Catheter-relatedPseudomonas aeruginosabiofilm infection rat models were established; the bacterial clearance rates, total cell counts in bronchoalveolar lavage (BAL) fluid, pathological changes of lungs, and the levels of Foxp3, TGF-β1, and IL-10 in PAO1 strain group were examined and compared with the QS-mutant ΔlasRΔrhlRandΔlasIΔrhlIgroups.Results. In PAO1 group, the bacterial clearance rates were lower, total cell counts were higher, pathological changes were severer, and the levels of Foxp3, TGF-β1, and IL-10 were significantly higher compared with QS-mutant groups(p<0.05). No significant difference was observed between the two QS-mutant groups(p>0.05).Conclusion. QS systems can trigger host immune system, accompanied with the upregulation of Tregs.
