Abstract Retained fetal membranes (RFM) is an important reproductive disease in dairy cows, caused by maternal and fetal placental tissue adhesion. The main collagen in maternal and fetal placenta tissues is collagen type IV (COL-IV) and its breakdown is the key to placental expulsion. Focal adhesion kinase (FAK) has been shown to regulate the hydrolysis of Col-IV by affecting the activity of MMP-2 and MMP-9 activity, but the regulation of the mechanisms involved in placenta expulsion in dairy cows after postpartum are still unclear. The aim of this study was to investigate the pathogenic mechanism of RFM by studying the relationship between the FAK signaling pathway and COL-IV regulation. Maternal placental tissues were collected from six healthy and six cows with RFM of similar age, parity, body condition and milk yield at 12 h postpartum. In vitro experiments were performed on bovine endometrial epithelial cells from three groups including a FAK inhibitor group, a FAK activator group and a control group without FAK inhibitor and activator. The abundance of molecules involved in the FAK signaling pathway and COL-IV was detected by immunohistochemistry, quantitative real-time polymerase chain reaction (qRT-PCR) and western blot. The immunohistochemical results showed that the key molecules of FAK signaling pathway FAK, Src, MMP-2 and MMP-9 and Col-IV were expressed in placental tissues. IntroductionAt present, the incidence of adolescent depression is increasing each year, having a wide and profound impact on adolescents. This study investigated the mood state of mid-to-late adolescents and young adults and analyzed related factors; clarified the incidence of depression, suicide, and self-injurious thoughts/behaviors in this population; and conducted relevant analysis of related factors of depression and anxiety.MethodsStudy subjects were students aged 14–25 years, from three high schools and one university in Liaoning Province. Study subjects were evaluated using several questionnaires that combined online and offline methods. Specifically, the Self-rating Depression Scale (SDS), Self-rating Anxiety Scale (SAS), Simplified Coping Style Questionnaire (SCSQ), Child Depression Inventory (CDI), the Chinese version of the Spence Child Anxiety Scale (SCAS), and a general questionnaire were utilized. Single-factor ANOVA, t-test, Chi-square, and multiple regression analysis were used to analyze the data.ResultsThe results showed that, among the 14–17-year-old subjects, the incidence of depression was 336 (14.7%), the incidence of anxiety was 763 (33.5%). Among the 18–25-year-old subjects, the incidence of depression was 34 (8.6%), the incidence of anxiety was 7 (1.8%). In the general questionnaire, 2081 (77.8%) individuals were depressed, 689 (25.8%) had thoughts of self-injury, and 323 (12.1%) had self-injurious behaviors. Among the 14–17-year-old subjects, 1789 (78.46%) were depressed, 689 (30.22%) had self-injury thoughts, and 319 (1.71%) had self-injurious behaviors. Among the 18–25-year-old subjects, 292 (73.92%) were depressed, but 4 (1.01%) had self-injurious behaviors. The incidence of depression and anxiety in adolescents is high, presenting with a certain risk of self-injury. However, age is an important factor in the occurrence of depression and anxiety; among the 18–25-year-old subjects, the incidence of depression (8.6% vs. 4.7%) and anxiety (1.8% vs. 33.5%) was lower than that among the 14–17-year-old population. Through comparative analysis, adolescents aged 14–17 remained at high risk of depression and anxiety.DiscussionIn the analysis of risk factors for depression and anxiety, relationships with classmates, teachers, and parents were reported as important influencing factors of emotional state. Further, a good coping style was found to be protective against depression and anxiety.

Diabetic cardiomyopathy (DCM) is a major complication of diabetes and is characterized by left ventricular dysfunction. Currently, there is a lack of effective treatments for DCM. Ubiquitin-specific protease 7 (USP7) plays a key role in various diseases. However, whether USP7 is involved in DCM has not been established. In this study, we demonstrated that USP7 was upregulated in diabetic mouse hearts and NMCMs co-treated with HG+PA or H9c2 cells treated with PA. Abnormalities in diabetic heart morphology and function were reversed by USP7 silencing through conditional gene knockout or chemical inhibition. Proteomic analysis coupled with biochemical validation confirmed that PCG1β was one of the direct protein substrates of USP7 and aggravated myocardial damage through coactivation of the PPARα signaling pathway. USP7 silencing restored the expression of fatty acid metabolism-related proteins and restored mitochondrial homeostasis by inhibiting mitochondrial fission and promoting fusion events. Similar effects were also observed in vitro. Our data demonstrated that USP7 promoted cardiometabolic metabolism disorders and mitochondrial homeostasis dysfunction via stabilizing PCG1β and suggested that silencing USP7 may be a therapeutic strategy for DCM.

The accurate prediction of building energy consumption is a crucial prerequisite for demand response (DR) and energy efficiency management of buildings. Nevertheless, the thermal inertia and probability distribution characteristics of energy consumption are frequently ignored by traditional prediction methods. This paper proposes a building energy consumption prediction method based on Bayesian regression and thermal inertia correction. The thermal inertia correction model is established by introducing an equivalent temperature variable to characterize the influence of thermal inertia on temperature. The equivalent temperature is described as a linear function of the actual temperature, and the key parameters of the function are optimized through genetic algorithm (GA). Using historical energy usage, temperature, and date type as inputs and future building energy comsuption as output, a Bayesian regression prediction model is established. Through Bayesian inference, combined with prior information on building energy usage data, the posterior probability distribution of building energy usage is inferred, thereby achieving accurate forecast of building energy consumption. The case study is conducted using energy consumption data from a commercial building in Nanjing. The results of the case study indicate that the proposed thermal inertia correction method is effective in narrowing the distribution of temperature data from a range of 24.5°C to 36.5°C to a more concentrated range of 26.5°C to 34°C, thereby facilitating a more focused and advantageous data distribution for predictions. Upon applying the thermal inertia correction method, the relative errors of the Radial Basis Function (RBF) and Deep Belief Network (DBN) decreases by 2.0% and 3.1% respectively, reaching 10.9% and 7.0% correspondingly. Moreover, with the utilization of Bayesian regression, the relative error further decreases to 4.4%. Notably, the Bayesian regression method not only achieves reduced errors but also provides probability distribution, demonstrating superiority over traditional methods.

Traumatic brain injury (TBI) is a complex neurological disorder that often results in long-term disabilities, cognitive impairments, and emotional disturbances. Despite significant advancements in understanding the pathophysiology of TBI, effective treatments remain limited. In recent years, exosomal non-coding RNAs (ncRNAs) have emerged as potential players in TBI pathogenesis and as novel diagnostic and therapeutic targets. Exosomal ncRNAs are small RNA molecules that are secreted by cells and transported to distant sites, where they can modulate gene expression and cell signaling pathways. They have been shown to play important roles in various aspects of TBI, such as neuroinflammation, blood-brain barrier dysfunction, and neuronal apoptosis. The ability of exosomal ncRNAs to cross the blood-brain barrier and reach the brain parenchyma makes them attractive candidates for non-invasive biomarkers and drug delivery systems. However, significant challenges still need to be addressed before exosomal ncRNAs can be translated into clinical practice, including standardization of isolation and quantification methods, validation of their diagnostic and prognostic value, and optimization of their therapeutic efficacy and safety. This review aims to summarize the current knowledge regarding the role of exosomal ncRNAs in TBI, including their biogenesis, function, and potential applications in diagnosis, prognosis, and treatment. We also discuss the challenges and future perspectives of using exosomal ncRNAs as clinical tools for TBI management.

Atrial fibrillation (AF) is a common cardiac arrhythmia that often occurs in patients with structural heart disease and is a significant cause of morbidity and mortality in clinical settings. AF is typically associated with significant changes of both the structure of the atria and the cardiac conduction system. AF can result in reduced heart function, heart failure, and various other complications. Current drug therapy for AF patients is often ineffective and may have adverse effects. Radiofrequency ablation is more effective than traditional drug therapy, but this invasive procedure carries potential risks and may lead to postoperative recurrence, limiting the clinical benefits to some extent. Therefore, in-depth research into the molecular mechanisms of AF and exploration of new treatment strategies based on research findings are prerequisites for improving the treatment of AF and the associated cardiac conditions. Long noncoding RNAs (lncRNAs) are a new class of noncoding RNA (ncRNAs) with a length exceeding 200 nt, which regulate gene expression at multiple levels. Increasing evidence suggests that lncRNAs participate in many pathological processes of AF initiation, development, and maintenance, such as structural remodeling, electrical remodeling, renin-angiotensin system anomalies, and intracellular calcium deregulation s. LncRNAs that play key roles in structural and electrical remodeling may become molecular markers and targets for AF diagnosis and treatment, respectively, while lncRNAs critical to autonomic nervous system remodeling may bring new insights into the prognosis and recurrence of AF. This review article provides a synopsis on the up-to-date research findings relevant to the roles of lncRNAs in AF.

Abstract Arsenic trioxide is a first-line treatment drug for acute promyelocytic leukemia, which is also effective for other kinds of leukemia. Its side effects, however, limit its clinical application, especially for patients with complex leukemia symptoms. Combination therapy can effectively alleviate these problems. This review summarizes the research progress on the combination of arsenic trioxide with anticancer drugs, vitamin and vitamin analogs, plant products, and other kinds of drugs in the treatment of leukemia. Additionally, the new progress in arsenic trioxide-induced cardiotoxicity was summarized. This review aims to provide new insights for the rational clinical application of arsenic trioxide.

Forty wheat samples of ten wheat varieties harvested from optimal or late sowings in 2019 and 2020 were evaluated for nutrient composition. This included crude protein (CP), starch, amino acids, minerals, phytate-phosphorus (phytate-P) and non-starch polysaccharides (NSPs). The objective was to investigate the impact of high temperature on wheat grain quality as a feedstuff for broiler chickens. Growth performance and economic impact of such changes were predicted by the Emmans, Fisher and Gous broiler growth model. On average, 2019 was 1 °C hotter than 2020 during the growing season (Narrabri, NSW 2390, Australia). The wheat harvested in 2019 had higher concentrations of CP, phytate-P, total P and calcium. In 2019, late sowing increased average protein concentrations from 166.6 to 190.2 g/kg, decreased starch concentration from 726 to 708 g/kg and increased total NSPs from 693 to 73.9 g/kg. Unlike the 2019 harvest, the late sowing in 2020 had no impact on CP concentrations in almost all wheat varieties. The 2019 varieties had higher concentrations of 16 assessed amino acids (p < 0.001) compared to the 2020 harvest. The largest difference was in lysine (19.2%), and the smallest difference was in proline (11.1%). It was predicted that broiler diets formulated from 2019 wheat varieties would have better efficiency of feed conversion with an advantage of 2.53% (1.539 versus 1.579) than 2020 varieties to 35 days post-hatch. This would translate to a cost saving of approximately AUD 16.45 per tonne of feed, much of which would represent additional profit.

Ecological stoichiometry (ES) is an important index that reflects the balance of various elements in ecological processes. Therefore, it is of great significance to understand the soil nutrient cycle to clarify the environmental control of soil carbon (C), nitrogen (N), phosphorus (P), and potassium (K). In this study, we analyzed the spatial distribution of soil C, N, P, and K contents and the C:N:P:K stoichiometric characteristics of 0–20 cm and 20–40 cm of farmland and grassland in four agro-pastoral areas in Inner Mongolia. Spearman correlation was used to analyze the effects of environmental factors on the soil C:N:P:K stoichiometric relationship. The results showed that there was no fixed Redfield ratio for the soil stoichiometric relationship of farmland and grassland in Inner Mongolia, and the values were 15:2:1:9 to 145:10:1:26 and 25:1:1:29 to 228:15:1:65, respectively. The stoichiometric relationships between farmland and grassland were consistent with the law of geographical and spatial heterogeneity. The ratios of C:N, C:P, C:K, N:P, and N:K showed an N distribution from west to east, while the ratio of P:K showed a V distribution. The stoichiometric relationships in grassland soil were mainly affected by soil organic carbon and total nitrogen content, while those in farmland were mainly affected by total nitrogen and total phosphorus content. The annual mean precipitation has a significant effect on stoichiometric relationships in farmland, while the annual mean temperature has a more significant effect on grassland. In conclusion, the spatial distribution difference in the soil stoichiometric relationship in the agro-pastoral ecotone of Inner Mongolia was more significant than the difference in the land use pattern. The influences of annual mean temperature and annual mean precipitation on soil ecological stoichiometry were in accordance with the geographical spatial similarity law. Compared with grassland, the stoichiometric relationship of farmland soil was greatly affected by fertilization, and farmland in this region was mainly limited by carbon and nitrogen. Thus, field management should be carried out according to local conditions. This study is of great significance as it promotes the rational utilization of land resources and the sustainable development of agriculture.

Abstract Background/Aims Arsenic trioxide (ATO) is the first-line therapeutic drug for acute promyelocytic leukemia. However, the cardiotoxicity of ATO limits its clinical application. This study aims to explore the long noncoding RNA (lncRNA) involved molecular mechanism in ATO-induced cardiotoxicity and to identify available prevention strategies. Methods ATO was administered to mice or primary cultured mouse cardiomyocytes. Small interfering RNA targeting lncRNA Kcnq1ot1 (si-Kcnq1ot1) was used to knockdown lncRNA Kcnq1ot1. MiR-34a-5p mimic and antisense morpholino oligonucleotide targeting miR-34a-5p (AMO-34a-5p) were used to upregulate and downregulate the expression of miR-34a-5p, respectively. TUNEL staining was conducted to detect cell DNA damage. Flow cytometry assay was used to detect cell apoptosis. Western blot was conducted to detect Bcl-2, Bax and Sirt1 protein expression. Real-time PCR was used to detect lncRNA Kcnq1ot1, miR-34a-5p, and Sirt1 mRNA expression. Dual-luciferase reporter assay was performed to validate the predicted binding site. Results ATO induced apoptosis in cardiomyocytes both in vivo and in vitro. Simultaneously, the expression of lncRNA Kcnq1ot1 and Sirt1 was downregulated, and miR-34a-5p was upregulated. MiR-34a-5p has binding sites with lncRNA Kcnq1ot1 and Sirt1. Knockdown of lncRNA Kcnq1ot1 induced apoptosis of cardiomyocytes, with increased miR-34a-5p and decreased Sirt1 expression. Inhibition of miR-34a-5p attenuated si-Kcnq1ot1-induced apoptosis in cardiomyocytes. Therefore, the lncRNA Kcnq1ot1/miR-34a-5p/Sirt1 signaling pathway is involved in ATO-induced cardiotoxicity. Propranolol alleviated ATO-induced apoptosis in cardiomyocytes both in vivo and in vitro, which was related to the lncRNA Kcnq1ot1/miR-34a-5p/Sirt1 signaling pathway. Conclusion The lncRNA Kcnq1ot1/miR-34a-5p/Sirt1 pathway is involved in ATO-induced cardiotoxicity. Propranolol can attenuate ATO-induced cardiotoxicity at least partially through the lncRNA Kcnq1ot1/miR-34a-5p/Sirt1 pathway. Combined administration with propranolol may be a new strategy for alleviating the cardiotoxicity of ATO.

Postpartum inactive ovaries (IOs) in dairy cows reduce the economic returns of the dairy industry. It is related to energy metabolism disorder, inflammatory response and oxidative stress. The objective of this study was to investigate the association of liver function index (LFI) and serum cytokines at 21 days postpartum with IO and to predict the risk of IO in dairy cows. The blood of 60 parturient cows was collected through caudal root vein at 3, 21, 28 and 55 days postpartum. Ultrasonography was performed at 50 and 55 days postpartum to determine follicular development. With the median LFI as the standard, it was divided into high LFI (n = 30) and low LFI group (n = 30). A cohort study was used to analyse the risk of LFI to IO and t-test was used to compare the blood biochemical indicators of different LFI groups. Then, 12 cows (oestrus = 6 and IO = 6) were slaughtered 55 days postpartum. The differences of LFI, cytokines and biochemical indexes were compared, and data were analysed by t-test, Spearman’s correlation analysis, binary logistic regression analysis and receiver operating characteristic analysis. The results show that growth and development of follicles of low LFI dairy cows were impaired, the risk of IO increased by 2.67 times. Cows with lower LFI had energy metabolism disorders, increased inflammation and oxidative stress and decreased ability to resist oxidative stress at 21 days postpartum. LFI and serum MDA, IL-6, FGF21 and ANGPTL at 21 days postpartum can predict IO in dairy cows.Highlights The risk of inactive ovaries (IOs) in cows with low liver function index (LFI) will increase by 2.67 times, serum MDA, IL-6, FGF21 and ANGPTL8 at 21 days postpartum can predict IOs in dairy cows at 50–55 days postpartum. A high predictive risk of IOs in dairy cows occurred when the LFI was less than −3.18, MDA more than 3.62 mmol/L, IL-6 more than 23.68 ng/L, FGF21 less than 812.41 ng/L and ANGPTL8 less than 695.79 ng/L at 21 days postpartum.

Abstract Diabetic cardiomyopathy (DCM) is a serious cardiac complication of diabetes that currently lacks specific treatment. The cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) signaling pathway has been suggested to contribute to the pathogenesis of cardiovascular diseases. However, whether cGAS-STING is involved in the development of DCM has not been established. Our study aimed to determine the role of cGAS-STING in the initiation of nucleotide-binding oligomerization domain-like receptor pyrin domain containing 3 (NLRP3) inflammasome-induced cardiac pyroptosis and chronic inflammation during the pathogenesis of DCM. C57BL/6J mice were preinjected with adeno-associated virus 9 (AAV9) intravenously via the tail vein to specifically knock down myocardial STING. After four weeks, mice with myocardium-specific knockdown of STING received injections of streptozotocin (STZ; 50 mg/kg) and a high-fat diet to induce diabetes. Measurements included echocardiography, immunohistochemical analyses, wheat germ agglutinin (WGA) staining, and western blotting. Here, we showed that the cGAS-STING signaling pathway was activated in diabetic hearts, which was indicated by the increased phosphorylation of TANK-binding kinase 1 (TBK1) and interferon (IFN) regulatory factor 3 (IRF3), leading to the activation of the NLRP3 inflammasome in the hearts of diabetic mice and proinflammatory cytokine release into serum. Moreover, STING knockdown via adeno-associated virus-9 (AAV9) in diabetic mouse heart alleviated cardiac pyroptosis and the inflammatory response, prevented diabetes-induced hypertrophy, and restored cardiac function. Mechanistically, we showed that palmitic acid (PA)-induced lipotoxicity impairs mitochondrial homeostasis, producing excessive mitochondrial reactive oxygen species (mtROS), which results in oxidative damage to mitochondrial DNA (mtDNA) and its release into the cytoplasm while switching on cGAS-STING-mediated pyroptosis in cardiomyocytes, thereby worsening the progression of diabetic cardiomyopathy. Our study demonstrated that activation of the cGAS-STING pathway caused by mitochondrial oxidative damage and mtDNA escape induced by free fatty acids promoted pyroptosis and proinflammatory responses in cardiomyocytes in a NLRP3 inflammasome-dependent manner, thus promoting myocardial hypertrophy during the progression of DCM.

Abstract Background Left displacement of the third gastric compartment (LDC3) in alpacas is an extremely rare condition and has not been reported thus far. Therefore, we describe the clinical diagnosis and treatment of LDC3 in an alpaca. Case presentation A 2-year-old brown female alpaca (Vicugna pacos) was presented to evaluate a 3-day history of abdominal distension causing loss of both appetite and thirst, along with oliguria and low to no defecation. Clinical examination, X-ray examination, surgical exploration, and determination of gastric pH (pH ~ 2.35) confirmed that LDC3 resulted in abdominal distension. The gastric wall of the displaced third gastric compartment was incised for the expulsion of pneumatosis, and a medical-grade silicone tube was inserted into the incision to remove the effusion by siphoning. Surgical treatment proved to effectively alleviate the abdominal distension caused by LDC3 without apparent side effects. Conclusions To our knowledge, this case is the first known report of LDC3 in an alpaca in China. A similar condition, left displaced abomasum, has previously been described in cattle and sheep.

Vitamin E, a potent antioxidant, is a necessary and complex micronutrient for cows. During the transition period, vitamin E deficiency (VED) is among the highest prevalent micronutrient deficits in dairy cows. It may eventually result in oxidative stress and immunological malfunction, and it increases the risk of peripartum disorders. At present, detailed data on blood metabolites in VED cows are limited. Consequently, the purpose of this research was to examine the alterations in the serum metabolic profile of VED cows throughout the early postpartum period. Using comprehensive 1H nuclear magnetic resonance (1H NMR), the alterations in serum metabolic activities of VED cows were analyzed. In total, 28 multiparous Holstein cows were assigned according to serum α-tocopherol (α-Toc) concentrations into normal (α-Toc ≥ 4 μg/mL, n = 14) and VED (α-Toc < 3 μg/mL, n = 14) groups at 21 days postpartum, and their blood samples were collected for biochemical and 1H NMR analyses. A t-test on independent samples as well as multivariate statistics were used to assess the findings. In comparison with normal cows, VED cows showed significantly worse body condition scores, milk yield, and dry matter intake (p < 0.05). Significantly higher levels of serum non-esterified fatty acids, aspartate aminotransferase, low-density lipoprotein, and malonaldehyde were found in VED-affected cows, as well as lesser concentrations of serum albumin, high-density lipoprotein, and total antioxidant capacity in comparison with normal cows (p < 0.01), while other vitamins and minerals concentrations showed no distinction between the groups (p > 0.05). Furthermore, 24 upregulated serum metabolites were identified under VED conditions. The metabolomics pathway analysis of these metabolites demonstrated that a global metabolic response to VED in cows was represented by changes in 11 metabolic pathways, comprising energy, carbohydrate, and amino acid metabolism. From these results, we conclude that VED cows were more likely to experience a negative energy balance characterized by alterations of common systemic metabolic processes and develop oxidative stress, inflammation, and ultimately liver injury. This study provides the first evidence of metabolic changes in cows with VED.

The resource utilization of cyanide tailings has significant environmental and economic benefits. The efficient recovery of gold from low-grade cyanide tailings containing 1.71 g/t Au was performed by a chlorination roasting–flotation process. The effects of roasting temperature, calcium chloride, internal coke, external coke, copper sulfide concentrate, and kaolin on the recovery rate of concentrate, gold grade, and sorting efficiency were investigated. The optimized process parameters were as follows: 16 wt% calcium chloride dihydrate, 6 wt% internal carbon, 1 wt% external carbon, 9 wt% copper sulfide concentrate, 2 wt% kaolin, and roasting temperature of 730 °C. The sorting rate, gold grade, and recovery rate of gold concentrate can reach 88.48%, 33.46 g/t, and 76.7%, respectively, and the gold grade of tailings was as low as 0.17 g/t. In the matte phase, gold can be enriched in the form of gold grains. Therefore, through chlorination roasting, the trapped gold can be released and deposited on the surface of the matte phase due to the transformation from hematite to magnetite. The gold-deposited metal sulfide can be effectively recycled through flotation. These results have potential guiding significance for the efficient recovery of gold from cyanide tailings.

Microwave-roasting as an efficient method for extracting copper form chalcopyrite has attracted increasing attention. In this work, the microwave roasted chalcopyrite was pressure leached in 1.0 mol/L sulfuric acid solution at the temperature range of 110–150 °C, and the reasons why microwave-roasting improved the leaching kinetics were further studied by the electrochemical characterizations. Results show that microwave-roasting had a positive effect on better mineral leachability. Under the optimal conditions, the final copper extraction was 90.1% and the iron extraction was 87.5%. The leaching process of the microwave roasted chalcopyrite was controlled by chemical reaction, and the calculated activation energy was 41.7 kJ/mol. The electrochemical study results indicated that microwave-roasting had a significant effect on both the anodic dissolution kinetics and the cathodic reactions kinetics, and the charge transfer resistance of chalcopyrite oxidation and the resistance of the passive film were both lower than that of the un-roasted sample. X-ray diffraction (XRD), and scanning electron microscope (SEM) results suggested that microwave-roasting effectively activate chalcopyrite through modifying the surface structure. Besides the enlargement of the effective reaction surface, the differential scanning calorimetry-thermogravimetric analysis (DSC-TGA) results revealed that the increasing of active sites and/or the internal stress can increase the strain energy and instability of chalcopyrite, which plays an important role in improving chalcopyrite leaching kinetics. The main product of pressure leaching was elemental sulfur that mixed with the copper-rich sulfide, causing the passivation of chalcopyrite.

Delimiting radiation protection distance of uranium mining and metallurgy facilities is an important radiation protection approach to control the effective public dose caused by radon and radon progeny. Ventilation shafts are the main radon release paths of underground uranium mines. It is of great importance to research the diffusion regularities and influence range of the radon around the ventilation shaft. In this study, long-term and short-term radon accumulation monitoring approaches were adapted for onsite investigation. More than 520 sets of radon concentration were acquired. The survey results effectively revealed the distribution regularities of the radon concentration around the ventilation shaft with time, space, and working conditions. These results provide data for radiation protection in uranium mines. In addition, a radiation protection distance delimiting way was proposed for the investigated facility through effective public dose assessment.

Pressure oxidation leaching behavior of chalcopyrite in sulfuric acid solution from 110 °C to 150 °C were investigated by in-situ electrochemical methods. Leaching experiments under saturated vapor pressure conditions were used to simulate the anoxic environment that may be encountered in industrial applications. Scanning electron microscope and X-ray photoelectron spectroscopy were used to characterize the morphology and the chemical status of chalcopyrite surface. Results show that the copper extraction was increased with the increase of leaching temperature. Under the optimal leaching conditions under saturated vapor pressure, the copper and iron extraction are 8.3% and 29.8%, respectively. When the temperature increased from 110 °C to 150 °C, the self-corrosion potential and electrochemical reaction resistance firstly increased and then decreased. In contrast, the resistance of the passive film was always increased with the increase of temperature. The electrochemical study results indicated that the increase in temperature affected the oxidation of chalcopyrite by altering the kinetics of the cathodic reaction and the anodic passivation. Both the self-corrosion current density (icorr) and rate constant were affected by the reduction of Fe(III). The XPS results show that elemental sulfur and H3O(Fe3(SO4)2(OH)6) were the main leaching solid products. The formation of H3O(Fe3(SO4)2(OH)6) not only caused a decrease in cathodic reaction kinetics, but also increased the resistance of mass transfer process. Due to the faster release of iron, copper-rich sulphides were formed, which mixed with the elemental sulfur and/or H3O(Fe3(SO4)2(OH)6) led to coverage of the chalcopyrite surface.

Background/Aims: Arsenic trioxide (ATO) is an effective anti-cancer drug. Nonetheless, it possesses cardiotoxic effects which limit its clinical application. The present study aims to elucidate the molecular basis of ATO-induced cardiotoxicity through using whole transcriptome analysis. Methods: The whole transcriptome in ATO-treated mice myocardium was analyzed using RNA sequencing technique. These results were confirmed by real-time PCR. The lncRNA-mRNA and circRNA-mRNA co-expression networks were constructed. Finally, a circRNA-lncRNA co-regulated competing endogenous RNA (ceRNA) network was constructed. GO and KEGG pathway analyses were performed. The expression levels of Txnip and Spp1 in ATO-treated neonatal mouse cardiomyocytes were validated by real-time PCR. Results: A total of 113 mRNAs, 159 lncRNAs, 35 miRNAs, and 94 circRNAs were differentially expressed in ATO-treated mice myocardium. A lncRNA-circRNA co-regulation network was constructed. Function annotation revealed that aberrantly expressed genes may be enriched in the ‘Wnt signaling pathway’, ‘Hippo signaling pathway’, ‘Notch signaling pathway’, etc. Finally, the expression levels of Txnip and Spp1 were validated in ATO-treated cardiomyocytes, which was in accordance with the RNA-sequencing results. Conclusion: ATO altered coding and noncoding RNA profiles in myocardium of mice. The ATO-related lncRNA-circRNA co-regulation network was constructed. Genes in the co-regulation network are likely to play important roles in the cardiotoxicity of ATO. This study provides new insights into the prevention and treatment of ATO-induced cardiotoxicity.

Myocardial infarction (MI) is a myocardial injury caused by coronary thrombosis or persistent ischemia and hypoxia. Due to its high morbidity and mortality, a safer and more effective treatment strategy is urgently needed. Daming capsule (DMC), a hypolipidemic drug, reportedly exerts cardioprotective effects in clinical and basic research, although its protective mechanism remains unknown. To investigate the mechanism underlying DMC-mediated improvement of cardiac function post-MI, C57/BL6 mice subjected to coronary artery ligation were administered DMC for 4 weeks. Our data demonstrated that DMC significantly improved cardiac structure and function compared to the saline group. Moreover, DMC inhibited inflammatory response and oxidative stress and improved mitochondrial structure and function in MI mice and hypoxia-stressed cardiomyocytes. Next, our research proved that DMC increased the expression of mitophagy receptor NLRX1. Interestingly, with the administration of DMC and siNLRX1, NLRX1 expression, mitochondria and lysosome colocalization, and mitochondrial membrane potential decreased, while mitochondrial ROS accumulation increased, suggesting that DMC promoted mitophagy to improve mitochondrial function via NLRX1 regulation. Further analysis showed that DMC activated the SIRT1/AMPK signaling pathway in vivo and in vitro. Our data showed that SIRT1 knockdown downregulated NLRX1 expression, leading to structural damage and functional impairment in mitochondria, as well as increased oxidative stress, inflammatory response, and decreased cardiac function in MI mice. Collectively, our findings reveal that DMC improves cardiac function post-MI by increasing mitophagy and inhibiting oxidative stress and inflammotory response in cardiomyocytes through the SIRT1/AMPK signaling pathway.

Negative energy balance (NEB) is an important risk factor for the dairy cow’s anoestrus. The purpose of the experiment is to screen out differentially expressed proteins (DEPs) in the follicular fluid (FF) of cows with anoestrus caused by NEB and oestrus from positive energy balance (PEB). NEB and PEB holstein cows were selected 14–21 days postpartum according to the serum concentrations of β-hydroxybutyric acid (BHB) and glucose (Glu). Based on the follicle diameter at 55–60 days postpartum, the two groups were assigned into the anoestrus group (NEB-A; n = 6; follicular diameter < 8 mm) and oestrus group (PEB-E; n = 6; follicular diameter: 15–20 mm). Their FF was aspirated in vivo to screen out DEPs, and analysed by bioinformatics. Five DEPs were verified by western blotting (WB). The study identified 367 proteins. Compared with PEB-E, NEB-A expressed 135 downregulated DEPs and 37 upregulated DEPs. RBP4 (retinol-binding protein 4), adiponectin B, SOD (superoxide dismutase), IGF2 (insulin-like growth factor 2), and CRP (C-reactive protein) were verified by WB, their may have confirmed that cellular lipid metabolism, oxidative stress, and cellular immune function are related to follicular development and anoestrus.Highlights 367 proteins identified in the Follicle fluid of dairy cows 55–60 days postpartum. In the Negative energy balance-anoestrus group, 172 differentially expressed proteins were identified—135 upregulated DEPs and 37 downregulated DEPs. Confirmed that postpartum Negative energy balance may affect cows’ oestrus behaviour or performance during early lactation by lipid metabolism, the IGF regulation system, and immune and complement systems.

Nuclear facilities are widely used in fields such as national defense, industry, scientific research, and medicine, which play a huge role in military and civilian use. However, in the process of widespread application of nuclear technology, uranium and its compounds with high carcinogenic and biologically toxic cause a lot of environmental problems, such as pollutions of water, atmosphere, soil, or ecosystem. Bioensors with sensitivity and specificity for the detection of uranium are highly demand. Nucleic acid enzymes (DNAzyme) with merits of high sensitivity and selectivity for targets as excellent molecular recognition elements are commonly used for uranium sensor development. In this perspective review, we summarize DNAzyme-based biosensors for the quantitative detection of uranyl ions by integrating with diverse signal outputting strategies, such as fluorescent, colorimetry, surface-enhanced Raman scattering, and electrochemistry. Different design methods, limit of detection, and practical applications are fully discussed. Finally, the challenges, potential solutions, and future prospects of such DNAzyme-based sensors are also presented.

Diabetic cardiomyopathy (DCM) is a major cardiovascular complication of diabetes mellitus (DM), and cardiac fibrosis is a characteristic pathological manifestation of DCM. DCM can be exacerbated by pyroptosis, and pyroptosis is a potential target of microRNAs (miRNAs). miR-135b is involved in delaying the progression of numerous cardiovascular diseases, Nonetheless, the role of miR-135b in diabetic cardiac fibrosis is unclear. Ranolazine is a piperazine derivative and is effective for the treatment of cardiovascular disease. The purpose of the study was to elucidate the mechanism of action of ranolazine against diabetic cardiac fibrosis and to investigate the role of miR-135b in this process. Functional and structural changes in the rat heart were examined by echocardiography, hematoxylin-eosin (H&E) and Masson staining. Immunohistochemistry was used to assess the expression of caspase-1, interleukin-1β (IL-1β), gasdermin D (GSDMD), transforming growth factor-β1 (TGF-β1), collagen I and collagen III in the rat left ventricle. Western blot and immunofluorescence were used to detect the protein expression of caspase-1, IL-1β, GSDMD, TGF-β1, collagen I and collagen III proteins, and the mRNA levels were determined using fluorescent quantitative PCR. Ranolazine reduced pyroptosis and inhibited collagen deposition, improving cardiac function in rats. Ranolazine increased miR-135b expression in high glucose-treated cardiac fibroblasts, and miR-135b directly bound to caspase-1. Interference with miR-135b reduced the effects of ranolazine on pyroptosis and collagen deposition. Ranolazine treatment of diabetic cardiac fibrosis inhibited pyroptosis and collagen deposition by upregulating miR-135b. Our study provides a solid theoretical basis for understanding the pathogenesis of diabetic cardiac fibrosis and the clinical use of ranolazine in the treatment of DCM.

Aiming at the problem that it is difficult to ensure the trajectory tracking accuracy and driving stability of the distributed driven driverless formula racing car under high-speed tracking conditions, a lateral and longitudinal coordinated control strategy is proposed. Based on the adaptive model predictive control theory, the lateral motion controller is designed, and the prediction time domain of the controller is changed in real time according to the change of vehicle speed. Based on the sliding mode variable structure control theory, a longitudinal motion controller is designed to accurately track the desired vehicle speed. Considering the coupling between the lateral and longitudinal controls, the lateral controller inputs the longitudinal speed and displacement of the vehicle, using the feedback mechanism to update the prediction model in real time, the longitudinal controller takes the front wheel angle as the input, the driving torque is redistributed through the differential drive control, and the lateral and longitudinal coordinated control is carried out to improve the trajectory tracking accuracy and driving stability. The typical working conditions are selected for co-simulation test verification. The results show that the lateral and longitudinal coordinated control strategy can effectively improve the vehicle trajectory tracking control accuracy and driving stability.

The traditional A∗ algorithm, applied to the motion planning of autonomous vehicles, easily causes high computational costs and excessive turning points generated in the planning path. In addition, the vehicle cannot track the path due to the unsmooth inflection point. To overcome these potential limitations, an improved A∗ algorithm-based motion planning algorithm and a tracking control strategy based on model predictive control theory were proposed in this work. The method of expanding the search neighborhood is adopted to improve the planning efficiency of A∗ algorithm. The artificial potential field method is also incorporated into the proposed A∗ algorithm. The resultant force generated by each potential field is further introduced into the evaluation function of A∗ algorithm to plan the driving path, which could be suitable for autonomous vehicles. The sharp nodes in the path are smoothed by cubic quasi-uniform B-spline curve. The tracking control strategy is designed based on model predictive control theory to realize the accurate tracking of the planned path. Typical obstacle avoidance conditions were selected for co-simulation test verification. The experimental results show that the proposed motion planning algorithm and tracking control strategy can effectively plan the obstacle avoidance path and accurately track the path in different environments.

In this work, a mineralogical study and thermal decomposition analysis of jarosite residue sample from a zinc plant were carried out. The mineralogical analysis showed that the jarosite residue is mainly composed of four components, including jarosites, sulfates–hydroxides, sulfides and spinel. The distribution of relevant metallic elements in these components was characterized using a sequential extraction process. The X-ray diffraction results showed that AFe3(OH)6(SO4)2 and ZnFe2O4 are the main components of the jarosite residue sample. The thermal decomposition mechanisms of ammoniojarosite and sodium jarosite were studied by TG-DSC (Thermogravimetric analysis–differential scanning calorimetry). The thermal decomposition process and the corresponding mechanism of the jarosite residue were analyzed. The jarosite thermal decomposition process includes the removal of crystal water, dihydroxylation and deammoniation, desulfonation of component jarosite and desulfonation of component zinc sulfate.

Collectively migrating tumor cells have been recently implicated in enhanced metastasis of epithelial malignancies. In oral squamous cell carcinoma (OSCC), αv integrin is a crucial mediator of multicellular clustering and collective movement in vitro; however, its contribution to metastatic spread remains to be addressed. According to the emerging therapeutic concept, dissociation of tumor clusters into single cells could significantly suppress metastasis-seeding ability of carcinomas. This study aimed to investigate the anti-OSCC potential of novel endostatin-derived polypeptide PEP06 as a cluster-dissociating therapeutic agent in vitro. Firstly, we found marked enrichment of αv integrin in collectively invading multicellular clusters in human OSCCs. Our study revealed that metastatic progression of OSCC was associated with augmented immunostaining of αv integrin in cancerous lesions. Following PEP06 treatment, cell clustering on fibronectin, migration, multicellular aggregation, anchorage-independent survival and colony formation of OSCC were significantly inhibited. Moreover, PEP06 suppressed αv integrin/FAK/Src signaling in OSCC cells. PEP06-induced loss of active Src and E-cadherin from cell–cell contacts contributed to diminished collective migration of OSCC in vitro. Overall, these results suggest that PEP06 polypeptide 30 inhibiting αv integrin/FAK/Src signaling and disrupting E-cadherin-based intercellular junctions possesses anti-metastatic potential in OSCC by acting as a cluster-dissociating therapeutic agent. Inactive ovaries (IO) and ovarian (follicular or luteal) cysts (FC or LC) are two common ovarian diseases leading to infertility in dairy cattle. Both disorders are associated with altered metabolites and hormones. There are currently no known effective biomarkers that can be used for early diagnosis of ovarian diseases. The purpose of this study was to identify the plasma biomarkers of ovarian diseases in Holstein dairy cows that facilitate an early diagnosis of the diseases and control its progression. The experiment was performed from 3 weeks postpartum and last for 7 weeks. Seventy-six multiparous Holstein cows (mean age, 4.36 years; weight, 635.63 kg) were divided into healthy control group (HC, n = 22), FC group (n = 18), LC group (n = 18) and IO group (n = 18) by rectal palpation or ultrasonography during the last 2 weeks before trial end. Blood was collected via tail vein for measurement of plasma energy metabolites, liver function indicators, minerals, and hormones at 3 and 8 weeks postpartum. Data were analyzed by Mann-Whitney U, Kruskal-Wallis, Spearman correlation, binary logistic regression analysis and receiver operating characteristic analysis, where applicable. At 8 weeks postpartum, FC cows had a more severe body condition score loss and these had greater levels of non-esterified fatty acids (NEFA) and estradiol, and lesser levels of alanine aminotransferase (ALT), progesterone and insulin-like growth factor 1 (IGF-1) levels than HC cows (P < 0.05). LC cows had a lower milk yield, higher NEFA and progesterone levels, and lower calcium, phosphorus and magnesium levels than HC cows (P < 0.05). IO cows had a lower body condition score, higher NEFA levels, and lower ALT, calcium, phosphorus, magnesium, estradiol, progesterone and IGF-1 levels than HC cows (P < 0.05). At 3 weeks postpartum, cows with ovarian diseases had greater (P < 0.05) concentrations of NEFA, and lesser concentrations of ALT, calcium, phosphorus and IGF-1 than HC cows. Diabetic cardiomyopathy (DCM) is a vital cause of fatalities in diabetic patients. The programmed death of cardiomyocytes and inflammation critically contribute to cardiac hypertrophy and fibrosis in DCM. Furthermore, circular RNA (circRNA) is a key regulator of various diseases. However, the role of circRNAs in DCM remains to be elucidated. Our previous study found that pyroptosis was markedly activated in the cardiomyocytes subjected to high-glucose conditions, and miR-214-3p regulated the expression of caspase-1. The aim of this study was to elucidate whether circRNA is involved in DCM pyroptosis via the miR-214-3p/caspase-1 pathway. Herein, we identified that hsa_circ_0076631, named caspase-1-associated circRNA (CACR), was increased both in high-glucose-treated cardiomyocytes and in the serum of diabetic patients. CACR also sponged an endogenous miR-214-3p to sequester and inhibit its expression. CACR knockdown in cardiomyocytes counteracted high-glucose-induced caspase-1 activation. Conversely, miR-214-3p knockdown partially abolished the beneficial effects of CACR silencing on pyroptosis in cardiomyocytes. Therefore, this study elucidated that CACR might be a novel therapeutic target via the CACR/miR-214-3p/caspase-1 pathway in DCM. Diabetic cardiomyopathy (DCM) is one of the major causes of death in diabetic patients. Its pathogenesis involves inflammation and fibrosis that damages the heart tissue and impairs cardiac function. Interleukin (IL)-17, a pro-inflammatory cytokine that plays an important role in a variety of chronic inflammatory processes can serve as an attractive therapeutic target. Anthocyanin, a water-soluble natural pigment, possesses impressive anti-inflammatory activity. However, its role in DCM is unclear. Hence, we investigated the protective effect of anthocyanin on the cardiovascular complications of diabetes using a mouse type 1 diabetes mellitus model induced by streptozotocin. Cardiac function and structural alterations in diabetic mice were tested by echocardiography, hematoxylin and eosin staining, and Masson trichrome staining. Immunohistochemistry was performed to evaluate the distribution and deposition of IL-17 and collagen I and III from the left ventricular tissues of diabetic mice. Cell viability was measured using the methyl thiazolyl tetrazolium assay. Protein levels of IL-17, tumor necrosis factor α, IL-1β, and IL-6 were determined using enzyme-linked immunosorbent assay. IL-17 and collagen I and III were detected by western blotting and immunofluorescence, and their mRNA levels were quantified using quantitative reverse transcription PCR. We observed that anthocyanin lowered blood glucose, improved cardiac function, and alleviated inflammation and fibrosis in the heart tissue of diabetic mice. Meanwhile, anthocyanin reduced the expression of IL-17 in high-glucose-treated cardiac fibroblasts and exhibited an anti-inflammatory effect. Deposition of collagen I and III was also decreased by anthocyanin, suggesting that anthocyanin contributes to alleviating myocardial fibrosis. In summary, anthocyanin could protect cardiac function and inhibit IL-17-related inflammation and fibrosis, which indicates its therapeutic potential in the treatment of diabetes mellitus-related complications.

Inactive ovaries (IOs) affect the estrus cycle and timed artificial insemination (TAI) efficiency in dairy cows during early lactation. The objective of the experiment was to determine metabolic changes in the serum and milk whey of dairy cows with IO and estrus. Twenty-eight healthy postpartum Holstein cows in similar age, milk production, and body condition were selected at 30 days postpartum for tracking to 70 days postpartum, and estrus performance was recorded through Afi Farm® software. The ovarian status and follicular diameter of dairy cows were examined by an experienced breeder through B-ultrasound and rectal examination. Fourteen normal estrus cows were allocated to control group A and 14 cows with IO to group B, all at 30–70 days postpartum. The serum and milk whey in the two groups of cows at 70 days postpartum were used for non-targeted nuclear magnetic resonance (1H-NMR) analysis to measure the different metabolites of cows with IO. In group B compared with group A at 70 days postpartum, there was an increase in the milk whey of six different metabolites including succinate, creatine phosphate, glycine, myo-inositol, glycolate, and orotate and a decrease in the milk whey of seven metabolites, including alanine, creatinine, o-phosphorylcholine, lactose, taurine, galactose, and glucose-1-phosphate. There was an increase in the serum of group B cows of four differential metabolites, including 3-hydroxybutyrate, acetate, glutamine, and glycine and a decrease in the serum of nine differential metabolites, including alanine, succinate, citrate, creatinine, o-phosphocholine, glucose, myo-inositol, tyrosine, and histidine compared with group A. Group B cows with IO had decreased glucose metabolism and impaired tricarboxylic acid cycle, increased lipid mobilization, and abnormal amino acid metabolism. The study provides a potential prevention strategy for IO in dairy cows in future.

Emodin is a promising anti-cancer reagent. To improve the physicochemical and anti-cancer property, we modified its structure and get a derivative called emodin succinyl ester (ESE). Here, we investigated the effect of ESE on the suppression of hepatocellular carcinoma (HCC) and the underlying mechanism. Our results showed that ESE strongly inhibited HCC cell proliferation and migration in vitro. Further study revealed that ESE treatment decreased transcription level and protein expression of androgen receptor (AR) and enhancer of zeste homolog 2 (EZH2), two key factors interacting to promote aggressive HCC development. Conversely, overexpression of AR attenuated the inhibitory effect of ESE on EZH2 expression, and vice versa. Importantly, overexpression of AR or EZH2 could counteract ESE-suppressed cell proliferation and migration. The association of ESE-targeted AR and EZH2 with the suppression of tumorigenicity was further confirmed in xenograft and diethylnitrosamine (DEN)-induced HCC mouse models. These findings validate the therapeutic effect of ESE on HCC aggression by targeting the interaction of AR and EZH2, suggesting ESE may be a potent drug in the clinical treatment of HCC.

The incidence and mortality of cardiovascular disease in diabetic patients are 2-3 times higher than those in non-diabetic patients. Abnormal function of the L-type calcium channel in myocardial tissue might result in multiple cardiac disorders such as a prolonged QT interval. Therefore, QT prolongation is an independent risk factor of cardiovascular disease in patients with diabetes mellitus. Metformin, a hypoglycemic agent, is widely known to effectively reduce the occurrence of macrovascular diseases. The aim of the present study was to evaluate the effect of metformin on prolonged QT interval and to explore potential ionic mechanisms induced by diabetes. Diabetic mouse models were established with streptozotocin and an electrocardiogram was used to monitor the QT interval after 4 weeks of metformin treatment in each group. Action potential duration (APD) and L-type calcium current (ICa-L) were detected by patch-clamp in isolated mice ventricular cardiomyocytes and neonatal cardiomyocytes of mice. The expression levels of CACNA1C mRNA and Cav1.2 were measured by real-time PCR, western blot and immunofluorescence. A shortened QT interval was observed after 4 weeks of metformin treatment in diabetic mice. Patch-clamp results revealed that both APD and ICa-L were shortened in mouse cardiomyocytes. Furthermore, the expression levels of CACNA1C mRNA and Cav1.2 were decreased in the metformin group. The same results were also obtained in cultured neonatal mice cardiomyocytes. Overall, these results verify that metformin could shorten a prolonged QT interval by inhibiting the calcium current, suggesting that metformin may play a role in the electrophysiology underlying diabetic cardiopathy.

Background/aims: Acute myocardial infarction (AMI) is a major clinical manifestation of ischemic heart disease and represents a significant cause of morbidity and mortality. However, key regulators in the pathogenesis of ischemic heart disease remain controversial. The present study was designed to investigate the involvement of miR-200a and its related mechanism in AMI. Methods: Left coronary artery (LCA) ligation was conducted to induce an AMI mouse model. The infarct size was measured by TTC staining. H2O2 was used to induce an AMI model in vitro. miR-200a mimics, anti-miR-200a antisense oligodeoxyribonucleotides (AMO-200a), as well as corresponding negative controls were transfected into cardiomyocytes to observe the effect of miR-200a. Flow cytometry was used to detect cell apoptosis. Real-time PCR, immunofluorescence and western blot assays were used to evaluate gene expression at RNA or protein levels, respectively. Results: Apoptosis was activated in AMI models. The expression of miR-200a was upregulated both in the peri-infarcted region of mice myocardium and H2O2-treated cardiomyocytes. The co-administration of AMO-200a decreased the number of apoptosis cells and altered the expression of apoptosis related proteins. Interestingly, bioinformatics analysis results revealed that miR-200a could bind to the 3′-untranslated regions (3’-UTR) of Fus mRNA. In addition, the expression of Fus was downregulated in the AMI mouse models and in H2O2-treated cardiomyocytes. The alteration of miR-200a negatively regulated Fus expression in cardiomyocytes. Also, the protective effect of AMO-200a was observed through its regulation of Fus. Conclusion: MiR-200a-dependent apoptosis signaling pathway plays an important role in the pathogenesis of AMI injury and could be an exciting potential therapeutic target.

Type 2 diabetes is a common metabolic disorder related to insulin resistance, or deficiency of insulin secretion, caused by decreased insulin sensitivity and destruction of islet structure and function. As the second human genome, the microbiota has been observed to have a growing relationship with diabetes in recent years. Microbiota imbalance has been hypothesized to be involved in the regulation of energy metabolism and the inflammatory immune response in diabetes. The present study aimed to investigate whether fecal microbiota transplantation (FMT) could alleviate the symptoms associated with type 2 diabetes. To this end, a type 2 diabetes mouse model was first established through the consumption of a high-fat diet combined with streptozotocin (100 mg/kg), and FMT was used to rebuild the gut microbiota of diabetic mice. Fasting blood glucose, oral glucose tolerance tests, and HbA1c levels were monitored, while the hypoglycemic effects of FMT were also observed. Insulin levels were tested by ELISA and related indexes such as HOMA-IR, HOMA-IS, and HOMA-β were calculated. We found that insulin resistance and pancreatic islet β-cells were improved after FMT treatment. Meanwhile, the markers of inflammation in the pancreatic tissue were detected by ELISA and immunohistochemistry, which indicated that inflammatory response decreased following FMT treatment. Furthermore, flow cytometry and western blot results revealed that FMT inhibited the β-cell apoptosis. Here, the effect of FMT on hypoglycemia in type 2 diabetes was addressed by improving insulin resistance and repairing impaired islets, thereby providing a potential treatment strategy for type 2 diabetes.

Inactive ovaries (IO) accounts for 50% of ovarian disease in postpartum dairy cows, which seriously affects their reproductive efficiency. To investigate the metabolic changes in the serum and follicular fluid of dairy cows with IO during lactation, six estrus (E) cows and six IO cows at 50 to 55 days in milk were selected based on B ultrasonic detection and clinical manifestations. The differential metabolites in serum and follicular fluid between the E cows and IO cows were identified by ultra-high-pressure liquid chromatography–quadrupole time-of-flight mass spectrometry, combined with multidimensional statistical methods. The results showed that dairy cows with IO were in a subclinical ketosis status where beta-hydroxybutyrate (BHB) exceeded 1.20 mmol/L, 14 differential metabolites in the serum of IO cows included 10 increased metabolites and 4 decreased metabolites, and 14 differential metabolites in the follicular fluid of IO cows included 8 increased metabolites and 6 decreased metabolites. These differential metabolites mainly involved nine metabolic pathways. The common enrichment pathway of different metabolites in serum and follicular fluid were glycerophospholipid metabolism and pentose and glucuronate interconversions. In conclusion, there were significant differences in the differential metabolites and enrichment pathways between serum and follicular fluid of IO cows, implying that there were complex changes in blood metabolism and local follicular metabolism of IO cows, whose interactions need further investigation.

Our previous studies found that mitochondrial uncouplers CCCP and niclosamide inhibited artery constriction and the mechanism involved AMPK activation in vascular smooth muscle cells. BAM15 is a novel type of mitochondrial uncoupler. The aim of the present study is to identify the vasoactivity of BAM15 and characterize the BAM15-induced AMPK activation in vascular smooth muscle cells (A10 cells). BAM15 relaxed phenylephrine (PE)-induced constricted rat mesenteric arteries with intact and denuded endothelium. Pretreatment with BAM15 inhibited PE-induced constriction of rat mesenteric arteries with intact and denuded endothelium. BAM15, CCCP, and niclosamide had the comparable IC50 value of vasorelaxation in PE-induced constriction of rat mesenteric arteries. BAM15 was less cytotoxic in A10 cells compared with CCCP and niclosamide. BAM15 depolarized mitochondrial membrane potential, induced mitochondrial fission, increased mitochondrial ROS production, and increased mitochondrial oxygen consumption rate in A10 cells. BAM15 potently activated AMPK in A10 cells and the efficacy of BAM15 was stronger than that of CCCP, niclosamide, and AMPK positive activators metformin and AICAR. In conclusion, BAM15 activates AMPK in vascular smooth muscle cells with higher potency than that of CCCP, niclosamide and the known AMPK activators metformin and AICAR. Background/Aims: Diabetic cardiomyopathy (DCM) is a common complication of diabetes and can cause heart failure, arrhythmia and sudden death. The pathogenesis of DCM includes altered metabolism, mitochondrial dysfunction, oxidative stress, inflammation, cell death and extracellular matrix remodeling. Recently, pyroptosis, a type of programmed cell death related to inflammation, was proven to be activated in DCM. However, the molecular mechanisms underlying pyroptosis in DCM remain elusive. The long non-coding RNA (lncRNA) Kcnq1ot1 participates in many cardiovascular diseases. This study aims to clarify whether Kcnq1ot1 affects cardiac pyroptosis in DCM. Methods: AC16 cells and primary cardiomyocytes were incubated with 5.5 and 50 mmol/L glucose. Diabetic mice were induced with streptozotocin (STZ). Kcnq1ot1 was silenced both in vitro and in vivo. qRT-PCR was used to detect the expression level of Kcnq1ot1. Immunofluorescence, qRT-PCR and western blot analyses were used to detect the degree of pyroptosis. Echocardiography, hematoxylin and eosin staining, and Masson’s trichrome staining were used to detect the cardiac function and morphology in mice. Cell death and function were detected using TUNEL staining, immunofluorescence staining and Ca2+ measurements. Results: The expression of Kcnq1ot1 was increased in patients with diabetes, high glucose-induced cardiomyocytes and diabetic mouse cardiac tissue. Silencing Kcnq1ot1 alleviated pyroptosis by targeting miR-214-3p and caspase-1. Furthermore, silencing Kcnq1ot1 reduced cell death, cytoskeletal structure abnormalities and calcium overload in vitro and improved cardiac function and morphology in vivo. Conclusion: Kcnq1ot1 is overexpressed in DCM, and silencing Kcnq1ot1 inhibits pyroptosis by influencing miR-214-3p and caspase-1 expression. We clarified for the first time that Kcnq1ot1 could be a new therapeutic target for DCM.

Abstract The human immunodeficiency virus (HIV) is a major global public health issue. HIV-related cardiovascular disease remains a leading cause of morbidity and mortality in HIV positive patients. HIV Tat is a regulatory protein encoded by tat gene of HIV-1, which not only promotes the transcription of HIV, but it is also involved in the pathogenesis of HIV-related complications. This review is aimed at summarizing the current understanding of Tat in HIV-related cardiovascular diseases.

Background: As the milk production of dairy cows increases, the reproductive capacity gradually declines. Ovarian quiescence has become one of the concerns of the dairy industry. Objective: To explore the different plasma metabolite levels between estrus and anestrus in energy balanced (EB) dairy cows. Methods: Ten estrous and 10 anestrus EB Holstein cows in early lactation were selected for the study. 1H nuclear magnetic resonance technology was used to detect plasma metabolites and screen different plasma metabolites between anestrous and estrous cows at 60–90 days postpartum using multivariate statistical analysis. Results: Within an elective waiting period of 60−90 days postpartum mean plasma concentration of total estrogens was significantly higher in estrus cows as compared to anestrus cows (71.2 ± 26.0 and 42.4 ± 16.7 pg/mL, respectively). Seven plasma metabolites (isoleucine, leucine, valine, alanine, arginine, choline and phosphatecholine) demonstrated significant decreases in estrous dairy cows relative to anestrous subjects. The main pathway was leucine, isoleucine and valine biosynthesis. Conclusion: Anestrus in dairy cows is accompanied by alterations in amino acid, glucose and lipid metabolism based on 1H NMR analysis.

Background/Aims: Arsenic trioxide (ATO) is a known anti-acute promyelocytic leukemia (APL) reagent, whose clinical applications are limited by its serious cardiac toxicity and fatal adverse effects, such as sudden cardiac death resulting from long QT syndrome (LQTS). The mechanisms of cardiac arrhythmia due to ATO exposure still need to be elucidated. Long non-coding RNAs (lncRNAs) are emerging as major regulators of various pathophysiological processes. This study aimed to explore the involvement of lncRNAs in ATO-induced LQTS in vivo and in vitro. Methods: For in vivo experiments, mice were administered ATO through the tail vein. For in vitro experiments, ATO was added to the culture medium of primary cultured neonatal mouse cardiomyocytes. To evaluate the effect of lncRNA Kcnq1ot1, siRNA and lentivirus-shRNA were synthesized to knockdown lncRNA Kcnq1ot1. Results: After ATO treatment, the Kcnq1ot1 and Kcnq1 expression levels were down regulated. lncRNA Kcnq1ot1 knockdown prolonged the action potential duration (APD) in vitro and exerted LQTS in vivo. Correspondingly, Kcnq1 expression was decreased after silencing lncRNA Kcnq1ot1. However, the knockdown of Kcnq1 exerted no effect on lncRNA Kcnq1ot1 expression. Conclusions: To our knowledge, this report is the first to demonstrate that lncRNA Kcnq1ot1 downregulation is responsible for QT interval prolongation induced by ATO at least partially by repressing Kcnq1 expression. lncRNA Kcnq1ot1 has important pathophysiological functions in the heart and could become a novel antiarrhythmic target.

Objective Hypocalcemia is an important metabolic disease of dairy cows during the transition period, although the effect of hypocalcemia on biological function in dairy cows remains unknown. Methods In this study, proteomic, mass spectrum, bioinformatics and western blotting were employed to identify differentially expressed proteins related to serum Ca concentration. Serum samples from dairy cows were collected at three time points: 3rd days before calving (day −3), the day of calving (day 0), and 3rd days after calving (day +3). According to the Ca concentration on day 0, a total of 27 dairy cows were assigned to one of three groups (clinical, subclinical, and healthy). Samples collected on day −3 were used for discovery of differentially expressed proteins, which were separated and identified via proteomic analysis and mass spectrometry. Bioinformatics analysis was performed to determine the function of the identified proteins (gene ontology and pathway analysis). The differentially expressed proteins were verified by western blot analysis. Results There were 57 differential spots separated and eight different proteins were identified. Vitamin D-binding protein precursor (group-specific component, GC), alpha-2-macroglobulin (A2M) protein, and apolipoprotein A-IV were related to hypocalcemia by bioinformatics analysis. Due to its specific expression (up-regulated in clinical hypocalcemia and down-regulated in subclinical hypocalcemia), A2M was selected for validation. The results were consistent with those of proteomic analysis. Conclusion A2M was as an early detection index for distinguishing clinical and subclinical hypocalcemia. The possible pathogenesis of clinical hypocalcemia caused by GC and apolipoprotein A-IV was speculated. The down-regulated expression of GC was a probable cause of the decrease in calcium concentration.

Hepatocellular carcinoma (HCC) is an extremely fatal malignancy. Intestinal microRNAs, which can be detected in fecal samples in humans may be involved in the pathological process of HCC. Therefore, screening for functional intestinal microRNAs in fecal samples and investigating their potential roles in the molecular progression of HCC are necessary. Quantitative real-time PCR (qRT-PCR) has been widely used in microRNA expression studies. However, few genes have been reported as reference genes for intestinal microRNAs in fecal samples. In order to obtain a more accurately analyzed intestinal microRNAs expression, we first searched for reliable reference genes for intestinal microRNAs expression normalization during qRT-PCR, using three software packages (GeNorm, NormFinder, and Bestkeeper). Next we screened and predicted the target genes of the differentially intestinal microRNAs of control and HCC mice through quantitative RT-PCR or miRtarBase. Finally, we also analyzed the mRNA targets for enrichment of Gene Ontology (GO) terms and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways using the DAVID Bioinformatic Resources database. This study has successfully screened relatively suitable reference genes and we have discovered that the differential intestinal microRNAs play significant roles in the development of HCC. The top reference genes identified in this study could provide a theoretical foundation for the reasonable selection of a suitable reference gene. Furthermore, the detection of intestinal microRNAs expression may serve as a promising therapeutic target for the diagnosis and treatment of HCC.

With the rapid development of materials technology, fluorescent gold nanoclusters (AuNCs) are emerging as novel functional materials for diagnostic applications including the detection of biomarkers and bioimaging due to the advantages of their ultra-small size, tunable emissions, size-dependent fluorescence and excellent biocompatibility. In this review, we introduced the synthetic methods, and physical and chemical properties of AuNCs. Subsequently, we described the AuNCs-based design strategies for the detection of biomarkers including small molecules, DNA and proteins. The applications of AuNCs for tumor imaging in vitro and in vivo were also presented. Finally, we discussed the challenges and potential solutions of AuNCs-based nanosensors.

The development of biosensors with high sensitivity and low-detection limits provides a new direction for medical and personal care. Graphene and graphene derivatives have been used to prepare various types of biosensors due to their excellent sensing performance (e.g., high specific surface area, extraordinary electronic properties, electron transport capabilities and ultrahigh flexibility). This perspective review focuses on graphene-based biosensors for quantitative detection of cancer-related biomarkers such as DNA, miRNA, small molecules and proteins by integrating with different signal outputting approaches including fluorescent, electrochemistry, surface plasmon resonance, surface enhanced Raman scattering, etc. The article also discussed their challenges and potential solutions along with future prospects.

BACKGROUND:Vitamin D could be a mediator in the association between tuberculosis (TB) and diabetes mellitus (DM). A large scale multi-center study confirmed that TB patients with DM had significantly lower serum vitamin D level compared with those without DM and reported that DM was a strong independent risk factor for vitamin D deficiency. OBJECTIVES:This study was undertaken to determine amongst patients with both TB and DM living in different economically defined areas in China: i) their baseline characteristics, ii) their vitamin D status and iii) whether certain baseline characteristics were associated with vitamin D deficiency. METHODS:In DM-TB patients consecutively attending seven clinics or hospitals, we measured 25 hydroxycholecalciferol at the time of registration using electrochemiluminescence in a COBASE 601 Roche analyser by chemiluminescence immunoassay. Data analysis was performed using chi square test and multivariate logistic regression. RESULTS:There were 178 DM-TB patients that included 50 from economically well-developed areas, 103 from better-off areas and 25 from a poverty area. Median vitamin D levels in well-developed, better-off and poverty areas were 11.5ng/ml, 12.2ng/ml and 11.5ng/ml respectively. Amongst all patients, 149 (84%) had vitamin D deficiency-91 (51%) with vitamin D deficiency (10-19.9 ng/ml) and 58 (33%) with severe deficiency (< 10 ng/ml). There was a significantly higher proportion with vitamin D deficiency in the poverty area. The adjusted odds of vitamin D deficiency (25-(OH)D3
