Abstract Pluripotent stem cells can differentiate into distinct cell types but the intracellular pathways controlling cell fate choice are not well understood. The social amoeba Dictyostelium discoideum is a simplified system to study choice preference as proliferating amoebae enter a developmental cycle upon starvation and differentiate into two major cell types, stalk and spores, organised in a multicellular fruiting body. Factors such as acidic vesicle pH predispose amoebae to one fate. Here we show that the mechanistic target of rapamycin complex 1 (mTORC1) pathway has a role in cell fate bias in Dictyostelium. Inhibiting the mTORC1 pathway activity by disruption of Rheb (activator Ras homolog enriched in brain), or treatment with the mTORC1 inhibitor rapamycin prior to development, biases cells to a spore cell fate. Conversely activation of the pathway favours stalk cell differentiation. The Set1 histone methyltransferase, responsible for histone H3 lysine4 methylation, in Dictyostelium cells regulates transcription at the onset of development. Disruption of Set1 leads to high mTORC1 pathway activity and stalk cell predisposition. The ability of the mTORC1 pathway to regulate cell fate bias of cells undergoing differentiation offers a potential target to increase the efficiency of stem cell differentiation into a particular cell type.

Abstract Purpose The objective of this study is to assess the impact of smoking on stroke prevalence and to delineate the relationship between smoking-related factors and the risk of stroke, incorporating an analysis of demographic variations influencing this association. Methods Our analysis encompassed 9,176 participants, evaluating clinical attributes alongside smoking-related characteristics such as duration of cigarette consumption, and levels of nicotine, tar, and carbon monoxide. We employed weighted univariate logistic regression and restricted cubic splines to examine the association between smoking indicators and stroke risk, complemented by subgroup analyses for demographic differentiation. Abstract Sowing date and soil fertility are very important factors in the overwintering and production performance of alfalfa (Medicago sativa L.), yet there’s a knowledge gap in knowledge on how late-seeded alfalfa responds to phosphorus (P) fertilization. A field study was conducted in Inner Mongolia from 2020 to 2022 using a split-plot design. The main plots consisted of five sowing dates (31 July, 8, 16, and 24 August, and 1 September), while the subplots involved five P application rates (0, 40, 70, 100, and 130 kg P2O5 ha−1). Throughout the growing seasons, the overwintering rate, root traits, forage yield, and yield components were measured. The results revealed a consistent decrease in overwintering ability and productivity with the delayed sowing. This reduction in overwintering rate was mainly due to diminished root traits, while the decrease in forage yield was largely associated with a reduction in plants per square meter. However, P fertilizer application to late-seeded alfalfa demonstrated potential in enhancing the diameter of both the crown and taproot, thus strengthening the root system and improving the overwintering rate, the rate of increase ranges from 11.6 to 49%. This adjustment could also improve the shoots per square meter and mass per shoot, increasing by 9.4–31.3% and 15.0–27.1% respectively in 2 years, which can offset the decline in forage yield caused by late sowing and might even increase the forage yield. Regression and path analysis indicated that alfalfa forage yield is primarily affected by mass per shoot rather than shoots per square meter. This study recommended that the sowing of alfalfa in similar regions of Inner Mongolia should not be later than mid-August. Moreover, applying P fertilizer (P2O5) at 70.6–85.9 kg ha−1 can enhance the forage yield and persistence of late-seeded alfalfa. Therefore, appropriate late sowing combined with the application of P fertilizer can be used as an efficient cultivation strategy for alfalfa cultivation after a short-season crop harvest in arid and cold regions.

Extreme rainfall is often accompanied by mass geological disasters, which seriously endangers the safety of people 's lives and property in prone areas and affects the healthy development of the economy and society. Summarizing and analyzing the time-space distribution characteristics of geological disasters due to extreme rainfall and the effectiveness of early warning is of great significance for improving the comprehensive defense ability against geological disasters. Taking the sudden geological disasters caused by “23•7” heavy rainfall in 2023 as the research object, based on the refined precipitation data from the Beijing sudden geological disaster monitoring and early warning system, the time-space distribution characteristics of “23•7” heavy rainfall and the development and distribution characteristics of geological disasters were analyzed, and the early warning effect of geological disasters was discussed. The results show that the“23•7”heavy rainfall has the characteristics such as a large total amount, strong rainfall, long duration and wide range, and the disasters due to extreme rainfall have the characteristics of group occurrence. The multi-dimensional early warning of geological disaster classification has achieved remarkable results and has achieved the goal of zero casualties due to geological disasters under extreme weather conditions. The research results can provide a reference for actively preventing and scientifically responding to extreme rainfall geological disasters.

The tensile-shear interactive damage (TSID) model is a novel and powerful constitutive model for rock-like materials. This study proposes a methodology to calibrate the TSID model parameters to simulate sandstone. The basic parameters of sandstone are determined through a series of static and dynamic tests, including uniaxial compression, Brazilian disc, triaxial compression under varying confining pressures, hydrostatic compression, and dynamic compression and tensile tests with a split Hopkinson pressure bar. Based on the sandstone test results from this study and previous research, a step-by-step procedure for parameter calibration is outlined, which accounts for the categories of the strength surface, equation of state (EOS), strain rate effect, and damage. The calibrated parameters are verified through numerical tests that correspond to the experimental loading conditions. Consistency between numerical results and experimental data indicates the precision and reliability of the calibrated parameters. The methodology presented in this study is scientifically sound, straightforward, and essential for improving the TSID model. Furthermore, it has the potential to contribute to other rock constitutive models, particularly new user-defined models.

Commitment schemes are cryptographic schemes that can be applied to zero-knowledge proof construction and blockchain construction. Recently, lattice-based cryptography has been intensively investigated due to the promising potential in quantum cryptography. Accordingly, commitment schemes based on lattice assumptions have been studied for practical applications. Notably, applications often require committing an arbitrary message with low communication costs, so commitment schemes must be satisfied with fewer length restrictions and fewer extensions to the messages. Several studies have been conducted to achieve the problem, including the study published by Baum et al. in 2018. However, the output length of their scheme is large in relation to the input length. We design a length-extension-free commitment scheme $\mathsf { Com_{MWM}}$ in which the length of the message string is large relative to the length of the commitment string, improving on the commitment scheme of Baum et al. Furthermore, we prove that the hiding and binding properties of $\mathsf { Com_{MWM}}$ are based on the hardness of the decisional search knapsack problem and extended search knapsack problems, respectively. Finally, we evaluate the computation costs of generating commitment value between ours and Baum et al.’s commitment scheme.

Evapotranspiration (ET) serves as a pivotal metric for studying ecohydrological processes. Its dynamics are intricately linked to the interplay among water balance, energy balance, and land use. However, the precise estimation of ET at a regional scale, coupled with a comprehensive understanding of its response to changes in water, heat, and vegetation factors, presents an ongoing challenge. In this study, we assessed the necessity to refine soil evaporation simulations in arid and semiarid regions. Subsequently, we introduced the random forest algorithm to optimize the soil evaporation module of the Priestley-Taylor Jet Propulsion Laboratory (PT-JPL) model to enable a more accurate prediction of ET. Then, we quantitatively explored the correlation between ET and pertinent factors, such as soil moisture, solar radiation, and the normalized difference vegetation index (NDVI) in the Inner Mongolia section of the Yellow River Basin (IMSYRB), which is a typical arid and semiarid region. The findings underscore that soil evaporation is the predominant constituent of ET in the IMSYRB. Moreover, soil moisture, as opposed to relative humidity, better represent the near-surface moisture conditions within the study area. The refined model (PT-JPL-RF) exhibited enhanced simulation performance at both individual stations and in the overall region for ET. Furthermore, the application of the PT-JPL-RF model for scrutinizing the spatiotemporal ET variations in the study area revealed an average ET of 250 mm over the past four decades. Since 1982, a fluctuating upward trend of 0.99 mm/year has been observed. Spatially, ET exhibits an eastward distribution, with elevated values in the eastern sectors compared to those in the western regions, particularly in the Big Black River Basin (296 mm). Vegetation was the primary factor influencing ET variations in the IMSYRB, contributing to an annual increase by 1.58 mm. These findings demonstrate the benefits of integrating machine learning algorithms with physical models and provide a practical example for accurately simulating and predicting ET across diverse regions.

Managing water resources amidst the pressures of climate change and human activities is a significant challenge, especially in regions experiencing shrinking lakes, deteriorating water quality, and ecological degradation. This review focuses on achieving integrated river basin management by learning from the governance experiences of typical watersheds globally, using the Hulun Lake Basin as a case study. Hulun Lake, China’s fifth-largest lake, experienced severe ecological problems from 2000 to 2009 but saw improvements after comprehensive management efforts from 2012 onward. This review systematically explores methods to address water resource, environment, and ecological challenges through the lenses of data acquisition, mechanism identification, model simulation, and regulation and management. Drawing lessons from successful basins such as the Rhine, Ganges, Mississippi, and Murray–Darling, the review proposes key goals for comprehensive management, including establishing extensive monitoring networks, developing predictive models, and creating contingency plans for routine and emergency management. Leveraging advanced technologies like satellite imagery and IoT sensors, alongside continuous improvement mechanisms, will ensure the sustainable use and protection of river basins. This review provides a detailed roadmap for achieving comprehensive watershed management in Hulun Lake, summarizing effective strategies and outcomes from data acquisition to regulation, thus serving as a model for similar regions globally.

Traditional lightweight foam concrete typically introduces a large number of voids into the concrete using surfactants to reduce density. However, in 3D printing, the instability of lightweight foam concrete can affect the workability of the slurry. Additionally, the lower strength of foam with more pores also reduces its mechanical performance. This study found that by replacing sand with expanded perlite in 3D-printed foam concrete, the stability of the foam is improved, enhancing the workability of the mixture and increasing the constructability of printed concrete. Furthermore, analyses of mechanical properties, porosity, and pore size distribution showed that at the same dry density, foam concrete with a higher expanded perlite replacement ratio exhibited higher compressive strength, with a maximum strength increase of up to 39%. Moreover, the introduction of expanded perlite optimized the pore distribution of the foam concrete, resulting in a more uniform material structure. The 3D-printed expanded perlite–foam concrete (3DPFC) prepared in this study provides new insights for the preparation of lightweight 3D-printed concrete, which is of significant importance for the sustainable development of the construction industry.

BackgroundIncreasing numbers of people are suffering from sleep disorders. The gut microbiota of these individuals differs significantly. However, no reports are available on the causal associations between specific gut microbiota and sleep disorders.MethodsData on gut genera were obtained from the MiBioGen consortium. Twenty-four cohorts with 18,340 individuals of European origin were included. Sleep disorder data, which included 216,454 European individuals, were retrieved from the FinnGen Biobank. Subsequently, two-sample Mendelian randomization was performed to analyze associations between sleep disorders and specific components of the gut microbiota.ResultsInverse variance weighting (IVW) revealed a negative correlation between Coprobacter and sleep disorders (OR = 0.797, 95% CI = 0.66–0.96, and p = 0.016), a positive correlation between Lachnospiraceae and sleep disorders (OR = 1.429, 95% CI = 1.03–1.98, and p = 0.032), a negative association between Oscillospira and sleep disorders (OR = 0.745, 95% CI = 0.56–0.98, and p = 0.038), and a negative association between Peptococcus and sleep disorders (OR = 0.858, 95% CI = 0.74–0.99, p = 0.039).ConclusionA significant causal relationship was found between four specific gut microbiota and sleep disorders. One family, Lachnospiraceae, was observed to increase the risk of sleep disorders, while three genera, namely, Coprobacter, Oscillospira, and Peptococcus, could reduce the risk of sleep disorders. However, further investigations are needed to confirm the specific mechanisms by which the gut microbiota affects sleep.

A feeding study lasting 30 days was carried out to determine the effects of dietary Oryzanol (Ory) on the survival, growth, antioxidant capacity, peptic enzymes, as well as lipid metabolism of Larimichthys crocea larvae (11.87 ± 0.59 mg) using four different Ory concentrations in microfeed formulations (0, 20, 40, and 80 mg/kg), all preserving isolipidic (18.25% crude lipid) and isonitrogenous (52.08% crude protein) conditions. Results revealed that larvae given 40 and 80 mg/kg Ory revealed markedly higher survival rates; in particular, the 80 mg/kg Ory larvae exhibited a considerably higher specific growth rate than the control. Furthermore, the 80 mg/kg dietary Ory resulted in increased peptic enzyme activity, indicating heightened digestive capabilities of larvae. Meanwhile, Ory supplementation at 20, 40, and 80 mg/kg also increased antioxidant enzyme activities and reduced malondialdehyde levels, indicating an enhanced antioxidant capacity. Moreover, the incorporation of 20 and 40 mg/kg Ory demonstrated efficacy in enhancing the lipid metabolism of the larvae. This was evident in the reduction of triglyceride and total cholesterol levels in the larval visceral mass, attributed to the downregulation of genes that participate in lipid synthesis, and the upregulation of genes associated with lipid catabolism. Overall, the study suggests that the addition of Ory (ranging from 40 to 80 mg/kg) significantly improves both survival and developmental aspects, possibly mediated by enhanced digestion and antioxidative effects, alongside the induction of lipid metabolism in Larimichthys crocea larvae.

BackgroundPatients with DN (diabetic nephropathy) show remarkable variations in their gut microbiota composition. However, to date, no study has shown whether a causal relationship exists between gut microbiota composition and DN.MethodsHere, we performed a two-sample Mendelian randomization (MR) investigation for identifying causal associations of gut microbiota with DN. Gut microbiota genetic data were gathered from the recent genome-wide association study pooled data of the MiBioGen consortium, which included 24 cohorts and 18,340 individuals.ResultsIVW(Inverse variance weighting) revealed that Verrucomicrobia [odds ratio (OR) = 1.390; 95% confidence interval (CI) = 1.10–1.75; p = 0.005], Peptostreptococcaceae (OR = 1.284; 95% CI = 1.03–1.59; p = 0.012), Verrucomicrobiaceae (OR = 1.390; 95% CI = 1.10–1.75; p = 0.005), Akkermansia (OR = 1.390; 95% CI = 1.10–1.75; p = 0.005), Butyricimonas (OR = 1.261; 95% CI = 1.02–1.55; p = 0.031), Catenibacterium (OR = 1.278; 95% CI = 1.02–1.59; p = 0.030).ConclusionTwo-sample MR analysis identified 12 microbial taxa in gut microbiota (one of which is yet to be officially named) that showed significant causal associations with DN; 8 of these taxa significantly increased the risk of DN, while the remaining 4 taxa (including the one without an official name) reduced the risk of DN. The precise mechanisms influencing the interactions of gut microbiota with DN occurrence remain unclear; hence, additional investigations should be conducted to clarify these mechanisms.

This study investigated the effects of ultrasound–assisted immersion freezing (UIF) at different power rates (0, 200, 400, and 600 W) on the changes in beef quality and flavor after braising. The results demonstrated that UIF treatment at 400 W significantly reduced the juice loss (cooking loss decreased from 49.04% to 39.74%) and fat oxidation (TBARS value decreased from 0.32 mg/kg to 0.20 mg/kg) of braised beef. In addition, the tenderness (hardness value decreased from 5601.50 g to 2849.46 g) and color stability of braised beef were improved after UIF treatment. The flavor characteristics of braised beef were characterized using an electronic nose and an electronic tongue. The PCA analysis data showed that the cumulative contribution rates of the first and second principal components were 85% and 93.2%, respectively, with the first principal component accounting for a higher proportion. The UIF–400 W group had the highest concentration for the first principal component, and the differentiation was not significant compared to the control group. The total amino acid values of different power UIF treatment groups were improved compared to the AF treatment group, indicating that UIF can effectively reduce the losses caused by freezing. The results demonstrate that ultrasound–assisted freezing treatment is beneficial in enhancing the tenderness and flavor attributes of beef after braising, providing new insights into the processing of meat products with desirable quality characteristics.

China’s northern Hulun Lake is a significant body of water internationally. The issue of eutrophication has gained prominence in recent years. The achievement of precise chlorophyll-a (Chl-a) monitoring is crucial for safeguarding Hulun Lake’s ecosystem. The machine learning-based remote sensing inversion method has been shown to be effective in capturing the intricate relationship between independent and dependent variables; however, it lacks a priori knowledge and is limited by the quality of remote sensing data sources. The relationship between independent and dependent variables can be more accurately simulated with the use of suitable auxiliary variables. Therefore, three machine learning models—random forest (RF), adaptive boosting (AdaBoost), and extreme gradient boosting (XGBoost)—were established in this study using meteorological observation parameters as auxiliary variables combined with Sentinel-2 satellite image remote sensing band combinations as independent variables and measured Chl-a data as dependent variables. The estimation effects before and after the fusion of meteorological ground observation data were compared, and the best model was used to estimate the spatial–temporal variation trend of Chl-a in the regional water body. The results show that (1) the addition of meteorological parameters as auxiliary variables improved the precision of the three machine models; the decision coefficient (R2) rose by 7.25%, 5.71%, and 7.20%, respectively, to 0.76, 0.66, and 0.73. (2) The concentration of Chl-a in the lake region was projected from June to October 2019 to October 2021 using the RF optimal estimating model of meteorological fusion. The northeast, southwest, and south of the lake were where the comparatively high concentration values of Chl-a were located, whereas the lake’s center had a generally low concentration of the substance. Chromatically, Chl-a typically peaked in August after initially increasing and then declining. (3) The three rivers that feed into the river have varying levels of water pollution, with chemical oxygen demand (COD) and total nitrogen (TN) pollution being the most severe. This is what primarily caused the higher levels of Chl-a in the northeast, southwest, and south. This study is crucial for the preservation and restoration of Hulun Lake’s natural ecosystem and offers some technical support for the monitoring of the lake’s concentration of Chl-a.

The identification and analysis of the deposit slope stability is the focus of geological disaster prevention and control. Taking dense-fine grained slopes and loose gravel slopes as examples, a series of flume tests under rainfall infiltration were carried out. The effects of density, material composition, slope angle and vegetation coverage on slope stability and rainfall threshold were systematically analyzed. The results show that under rainfall infiltration, the dense-fine grained slope with uniform material composition and high density was more stable, resulting in a higher rainfall threshold and a lower catastrophability. The slope with non-uniformmaterial composition and loose gravel was more likely to fail than the slope with dense fine particles. The influence of the stone content on the stability of the slope is greater than that of the slope gradient. The critical rainfall of landslides increases first and then decreases with an increasing vegetation coverage; that is, when rainfall infiltration causes the slope soil to be supersaturated, the higher vegetation coverage will trigger the whole deformation of slope and vegetation due to the strong development of vegetation roots, which increases the catastrophability of the slope.The research results can provide a theoretical basis for the instability mechanism and stability evaluation of deposit slopes.

Background: Aspiration Pneumonia (AP) is a leading cause of death in patients with Acute Ischemic Stroke (AIS). Early detection, diagnosis and effective prevention measures are crucial for improving patient prognosis. However, there is a lack of research predicting AP occurrence after AIS. This study aimed to identify risk factors and develop a nomogram model to determine the probability of developing AP after AIS. Method: A total of 3258 AIS patients admitted to Jinshan Hospital of Fudan University between January 1, 2016, and August 20, 2022, were included. Among them, 307 patients were diagnosed with AP (AP group), while 2951 patients formed the control group (NAP group). Univariate and multivariate logistic regression analyses were conducted to identify relevant risk factors for AP after AIS. Abstract Wind power input to oceanic near‐inertial oscillations (NIOs) plays a crucial role in sustaining the global ocean conveyor belt. However, the impact of tropical cyclones (TCs) on wind power input to NIOs, despite being the most vigorous atmospheric dynamics capable of exciting NIOs, is often overlooked in global estimations due to their transient nature and a lack of observations. Utilizing hourly wind and ocean current records, we quantified the wind power on NIOs induced by TCs from 1990 to 2019. Our findings reveal that the wind power on NIOs due to TCs is estimated to be between 0.028 and 0.065 TW, which accounts for a significant proportion, that is, 8%–17%, of that over the globe. This study highlights the importance of incorporating the wind power induced by TCs when estimating the global wind power on NIOs, as its impact is non‐negligible. Our findings contribute to a better understanding of the global energy balance by improving the estimation of wind power on NIOs.

With the widespread use of large artificial intelligence (AI) models such as ChatGPT, AI-generated content (AIGC) has garnered increasing attention and is leading a paradigm shift in content creation and knowledge representation. AIGC uses generative large AI algorithms to assist or replace humans in creating massive, high-quality, and human-like content at a faster pace and lower cost, based on user-provided prompts. Despite the recent significant progress in AIGC, security, privacy, ethical, and legal challenges still need to be addressed. This paper presents an in-depth survey of working principles, security and privacy threats, state-of-the-art solutions, and future challenges of the AIGC paradigm. Specifically, we first explore the enabling technologies, general architecture of AIGC, and discuss its working modes and key characteristics. Then, we investigate the taxonomy of security and privacy threats to AIGC and highlight the ethical and societal implications of GPT and AIGC technologies. Furthermore, we review the state-of-the-art AIGC watermarking approaches for regulatable AIGC paradigms regarding the AIGC model and its produced content. Finally, we identify future challenges and open research directions related to AIGC.

Abstract An unprecedented warm sea surface temperature (SST) anomaly event, namely, the Blob, occurred in the northeast Pacific during the winter (October–January) of 2013/2014, causing substantial economic and ecological impacts. Here, we explore the driving forces of the Blob from both atmospheric and oceanic perspectives and show that the Blob primarily resulted from weak wintertime cooling due to the reduced air-sea heat flux transfer from the ocean to the atmosphere and the reduced horizontal advection of cold water in the upper ocean. Both mechanisms were attributed to an anomalous high-pressure system over the study region. Specifically, the anomalous air-sea heat flux, which was dominated by turbulent heat flux anomalies, was mainly induced by the increased air temperature (i.e., with a contribution of approximately 70%) and the weakened wind speed associated with the high-pressure system. The reduced horizontal heat advection was mainly due to the weakened winds acting on the ocean temperature meridional gradient. Using a regional ocean numerical model with different experimental runs, we evaluated the contributions of air temperature and wind drivers to the Blob at both the surface and subsurface of the ocean. The Blob was absent when the model was forced by climatology-air-temperature. Both the SST and integrated ocean heat content (OHC, 0–150 m) decreased, and the mixed layer depth (MLD) was deeper than that in the control run forced by real atmospheric conditions. In the climatology-winds experiment, obvious warm anomalies still existed, which were similar to but weaker than the control run. The SST (OHC) and MLD values in the climatology-winds run were between those of the climatology-air-temperature run and the control run. Compared to former studies that attribute the formation of the Blob to an anomalous air-sea heat flux and horizontal advection mainly induced by reduced winds, our study demonstrates that anomalous warm air temperatures played a more important role in its formation.

Abstract The security of lattice‐based cryptosystems is generally based on the hardness of the Shortest Vector Problem (SVP). The original enumeration (ENUM) algorithm solving SVP runs in exponential time due to the exhaustive search, which is used as a subroutine for the block Korkin–Zolotarev (BKZ) algorithm. It is a critical issue to reduce the computational complexity of ENUM. In this paper, first, we improve the reordering method proposed by Wang et al. in ACISP 2018. We call our proposed method DPR, which permutates the projected dual lattice vectors by decreasing norms. Preliminary experimental results show that the proposed reordering methods can reduce the ENUM complexity compared to the predecessor; for instance, DPR reduces around 32.8% on average in 45‐dimensional lattices. Moreover, the authors’ simulation shows that the higher the lattice dimension, the more DPR can reduce the ENUM complexity. In addition, we study a condition for deciding when the reordering method shall be executed or not. Finally, we improve the BKZ algorithm with DPR methods and the proposed condition.

Reservoir operation rule curves are crucial for managing water supply and power generation in reservoirs. As the number of objectives and management requirements increase, there is a growing demand for optimized operation rule curves. The objective of this study is to explore the most effective forms of reservoir operation rule curves, focusing on the case of the Nierji Reservoir and considering the dual objectives of water supply and power generation. The parameter–simulation–optimization framework, specifically employing the NSGA-II algorithm, was used to analyze and compare two basic forms of operation rule curves: the shared type and independent type. The impact of these curves on water supply potential and multi-objective optimization results with various water demand scenarios was assessed. The analysis revealed that the choice of operation rule curve form can influence the maximum water supply potential of the reservoir to some extent. The independent type operation rule curve was significantly more effective in enhancing the water supply potential for industrial and domestic users, resulting in a notable increase of 3.5 × 108 m3. Additionally, it also proved beneficial for environmental users, with an increase of 1 × 108 m3. Conversely, the shared type operation rule curve demonstrated similar functionality to the independent type curve with fewer decision variables, particularly when the water demand was relatively low. In scenarios with high water demand, the independent type curve outperformed the shared type curve by generating 6549 superior, non-dominated solutions for multi-objective optimization, specifically focused on maximizing reservoir operation benefits. In conclusion, selecting the appropriate form of reservoir operation rule curve is crucial to balance different reservoir functional objectives and achieve optimal results. Further research could focus on quantifying the specific benefits and trade-offs associated with each type of curve in order to provide more robust evidence for the advantages of a complex reservoir system.

Objective: The goal of this pilot study was to investigate the effects of exosomes derived from synovial fluid-derived cells (SFDCs) cultured under normoxic conditions in a two-dimensional (2D) monolayer or encapsulated within a three-dimensional (3D) matrix for chondrogenic differentiation in vitro and cartilage defect repair in vivo. Design: Synovial fluid samples were obtained from three patients, and SFDCs were isolated and expanded either in a 2D monolayer culture or seeded within a transglutaminase cross-linked gelatin (Col-Tgel) to create a 3D gel culture. Exosomes derived from each environment were isolated and characterized. Then, their effects on cartilage-cell proliferation and chondrogenic differentiation were assessed using an in vitro organoid model, and their potential for enhancing cartilage repair was evaluated using a rat cartilage defect model. Results: SFDCs obtained from different donors reached a state of senescence after four passages in 2D culture. However, transferring these cells to a 3D culture environment mitigated the senescence and improved cell viability. The 3D-cultured exosomes exhibited enhanced potency in promoting chondrogenic differentiation, as evidenced by the increased expression of chondrogenic genes and greater deposition of cartilage-specific extracellular matrix. Furthermore, the 3D-cultured exosomes demonstrated superior effectiveness in enhancing cartilage repair and exhibited better healing properties compared to exosomes derived from a 2D culture. Conclusions: The optimized 3D culture provided a more favorable environment for the proliferation of human synovial cells and the secretion of exosomes compared to the 2D culture. The 3D-cultured exosomes exhibited greater potential for promoting chondrogenic gene expression in vitro and demonstrated improved healing properties in repairing cartilage defects compared to exosomes derived from the 2D culture.

Tropical cyclones (TCs) exert a significant influence on the upper ocean, leading to sea surface temperature (SST) changes on a global scale. However, TC-induced SST responses exhibit considerable variability in the northern Indian Ocean (NIO), and the general understanding of these responses remains limited. This paper investigates the SST changes caused by 96 TCs over an 18-year period in the NIO. Through a composite analysis utilizing satellite SST data, a comprehensive study is conducted to examine the relationship between TC characteristics, including wind speed and translation speed, and the associated SST changes. The overall findings reveal that within a radius of 300 km from the TC center, SST decreases were observed at 1702 (86%) locations, with an average SST response to TC of −0.46 °C and a maximum decrease of −2.07 °C. The most significant reduction in SST typically occurred two days after the passage of TCs, followed by a gradual recovery period exceeding 15 days for the SSTs to return to their initial values. Consistent with findings in other ocean basins, stronger and slower-moving TCs induced more substantial cooling effects. Conversely, at 279 (14%) locations, particularly associated with TCs of weaker intensities, SST increases were observed following the TC passage. Notably, 140 of these locations were situated at low latitudes, specifically between 8° N and 15° N. This study provides a quantitative analysis of the comprehensive SST response to TCs in the NIO.

Blast stress waves profoundly impact engineering structures, exciting and affecting the rupture process in brittle construction materials. A novel numerical model was introduced to investigate the initiation and propagation of cracks subjected to blast stress waves within the borehole-crack configuration. Twelve models were established with different crack lengths to simulate sandstone samples. The influence of crack length on crack initiation and propagation was investigated using those models. The linear equation of state was used to express the relationship between the pressure and density of the material. The major principal stress failure criterion was used to evaluate the failure of elements. A triangular pressure curve was adopted to produce the blast stress wave. The results indicated that the pre-crack length critically influenced the crack initiation and propagation mechanism by analyzing the stress history at the crack tip, crack propagation velocity, and distance. The inducement of a P-wave and S-wave is paramount in models with a short pre-crack. For long pre-crack models, Rayleigh waves significantly contribute to crack propagation.

A 30-day feeding trial was designed to evaluate the effect of supplemental fulvic acid (FA) on survival, growth performance, digestive ability and immunity of large yellow croaker (Larimichthys crocea) larvae (initial body weight 11.33 ± 0.57 mg). Four isonitrogenous and isolipids diets containing 0.00%, 0.01%, 0.02% and 0.04% FA were formulated, respectively. Results showed that the supplementation of 0.04% FA significantly improved survival rate of large yellow croaker larvae. Meanwhile, supplemental FA significantly increased final body weight and specific growth rate. Based on the specific growth rate, the optimal supplementation was 0.0135% FA. Larvae fed the diet with 0.01% FA had significantly higher villus height than the control. The supplementation of 0.01%–0.02% FA significantly increased the muscular thickness of intestine. Moreover, supplementation of FA significantly increased mRNA expression of intestinal epithelial proliferation and barrier genes (pcna, zo-1 and zo-2). Diets supplemented with 0.02%–0.04% FA significantly increased the activity of trypsin in the intestinal segment, while 0.01%–0.02% FA significantly increased the activity of trypsin in the pancreatic segment. Compared with the control, supplementation of FA remarkably increased activities of alkaline phosphatase and leucine aminopeptidase in the brush border membrane of intestine. Larvae fed the diet with 0.01% FA significantly increased activities of lysozyme and total nitric oxide synthase. Furthermore, the supplementation of 0.01% to 0.02% FA significantly decreased the mRNA expression of pro-inflammatory cytokines (tnf-α and il-6). Concurrently, supplemental FA significantly increased anti-inflammatory cytokine (il-10) mRNA expression level. In conclusion, this study indicated that the supplementation of FA could improve the survival rate and growth performance of larvae by promoting intestinal development, digestive enzymes activities and innate immunity.

IntroductionAlfalfa (Medicago sativa L.) is a highly nutritious leguminous forage that plays an essential role in animal husbandry. In the middle and high latitudes of the northern hemisphere, there are problems with its low rates of overwintering and production. The application of phosphate (P) is an important measure to improve the cold resistance and production of alfalfa, but little is known about the mechanism of P in improving the cold resistance of alfalfa.MethodsThis study integrated the transcriptome and metabolome to explain the mechanism of alfalfa in response to low-temperature stress under two applications of P (50 and 200 mg kg-1) and a control of none applied.ResultsThe application of P fertilizer improved the root structure and increased the content of soluble sugar and soluble protein in the root crown. In addition, there were 49 differentially expressed genes (DEGs) with 23 upregulated and 24 metabolites with 12 upregulated when 50 mg kg-1 of P was applied. In contrast, there were 224 DEGs with 173 upregulated and 12 metabolites with 6 upregulated in the plants treated with 200 mg kg-1 of P compared with the Control Check (CK). These genes and metabolites were significantly enriched in the biosynthesis of other secondary metabolites and the metabolic pathways of carbohydrates and amino acids. The integration of the transcriptome and metabolome indicated that P affected the biosynthesis of N-acetyl-L-phenylalanine, L-serine, lactose, and isocitrate during the period of increasing cold. It could also affect the expression of related genes that regulate cold tolerance in alfalfa.DiscussionOur findings could contribute to a deeper understanding of the mechanism that alfalfa uses to tolerate cold and lay a theoretical foundation for breeding alfalfa that is highly efficient at utilizing phosphorus.

Coastal eutrophication and hypoxia remain a persistent environmental crisis despite the great efforts to reduce nutrient loading and mitigate associated environmental damages. Symptoms of this crisis have appeared to spread rapidly, reaching developing countries in Asia with emergences in Southern America and Africa. The pace of changes and the underlying drivers remain not so clear. To address the gap, we review the up-to-date status and mechanisms of eutrophication and hypoxia in global coastal oceans, upon which we examine the trajectories of changes over the 40 years or longer in six model coastal systems with varying socio-economic development statuses and different levels and histories of eutrophication. Although these coastal systems share common features of eutrophication, site-specific characteristics are also substantial, depending on the regional environmental setting and level of social-economic development along with policy implementation and management. Nevertheless, ecosystem recovery generally needs greater reduction in pressures compared to that initiated degradation and becomes less feasible to achieve past norms with a longer time anthropogenic pressures on the ecosystems. While the qualitative causality between drivers and consequences is well established, quantitative attribution of these drivers to eutrophication and hypoxia remains difficult especially when we consider the social economic drivers because the changes in coastal ecosystems are subject to multiple influences and the cause–effect relationship is often non-linear. Such relationships are further complicated by climate changes that have been accelerating over the past few decades. The knowledge gaps that limit our quantitative and mechanistic understanding of the human-coastal ocean nexus are identified, which is essential for science-based policy making. Recognizing lessons from past management practices, we advocate for a better, more efficient indexing system of coastal eutrophication and an advanced regional earth system modeling framework with optimal modules of human dimensions to facilitate the development and evaluation of effective policy and restoration actions.

Eddies are ubiquitous mesoscale dynamics in oceans worldwide that prominently influence the upper ocean. Specifically, the presence of preexisting eddies can modulate oceanic responses to passing typhoons, for which the underlying mechanism requires further investigation. In 2013, Typhoon Soulik traveled over 2,000 km in the western North Pacific, encountering multiple eddies, offering a suitable opportunity for investigating the impact of eddies on typhoon-induced upper ocean responses. A coupled ocean-atmosphere model was developed for this event, and the simulations were validated with multiple observations. The surface cold wake after Typhoon Soulik was successfully reproduced by the coupled model, showing a multicore structure, which was due to the preexisting mesoscale cyclonic eddies along the typhoon path. An experimental case excluding eddies was also tested in the numerical model. The comparison between the control and experimental cases revealed that sea surface cooling was enhanced by eddies via the vertical mixing and horizontal advection of eddy-related currents. In particular, vertical mixing enhanced sea surface cooling beneath the typhoon center due to the stronger vertical stratification within the preexisting cyclonic oceanic eddies. The eddy-related current with a horizontal temperature gradient contributed to advection, enhancing sea surface cooling at the cooling center and leading to a rightward-shifted pattern of enhanced sea surface cooling. This study enriches the understanding of the complex upper ocean dynamics under the combined effects of typhoons and eddies.

The ecologically important marine phytoplankton group coccolithophores have a global distribution. The impacts of ocean acidification on the cosmopolitan species Emiliania huxleyi have received much attention and have been intensively studied. However, the species-specific responses of coccolithophores and how these responses will be regulated by other environmental drivers are still largely unknown. To examine the interactive effects of irradiance and ocean acidification on the physiology of the coastal coccolithophore species Pleurochrysis carterae, we carried out a semi-continuous incubation experiment under a range of irradiances (50, 200, 500, 800 μmol photons m−2 s−1) at two CO2 concentration conditions of 400 and 800 ppm. The results suggest that the saturation irradiance for the growth rate was higher at an elevated CO2 concentration. Ocean acidification weakened the particulate organic carbon (POC) production of Pleurochrysis carterae and the inhibition rate was decreased with increasing irradiance, indicating that ocean acidification may affect the tolerating capacity of photosynthesis to higher irradiance. Our results further provide new insight into the species-specific responses of coccolithophores to the projected ocean acidification under different irradiance scenarios in the changing marine environment.

In this study, the effect of chickpea dietary fiber (CDF) concentration (0%, 0.4%, 0.8%, 1.2%, 1.6%, and 2.0%) on emulsion gel properties of myofibrillar protein (MP) was investigated. It was found that the emulsifying activity index (EAI) and emulsifying stability index (ESI) of MP increased with the increasing content of CDF. Moreover, the water- and fat-binding capacity (WFB), gel strength, storage modulus (G’), and loss modulus (G”) of MP emulsion gel also increased with increasing content of CDF. When the concentration of CDF was 2%, the most significant improvement was observed for EAI, breaking force, and WFB (p < 0.05); the three-dimensional gel network structure of the MP emulsion gel was denser and the pore diameter was smaller. The T21 relaxation time of emulsion gel decreased while the PT21 increased significantly with the increasing content of CDF, suggesting that the emulsion gel with CDF had a better three-dimension network. The addition of CDF led to an increased content of β-sheet and reactive sulfhydryl and increased surface hydrophobicity of MP, thus improving the gel properties of the MP emulsion gel. In conclusion, the addition of CDF improved the functional properties and facilitated the gelation of the MP emulsion, indicating that CDF has the potential to improve the quality of emulsified meat products.

BackgroundBig data useful for epidemiological research can be obtained by integrating data corresponding to individuals between databases managed by different institutions. Privacy information must be protected while performing efficient, high-level data matching. ObjectivePrivacy-preserving distributed data integration (PDDI) enables data matching between multiple databases without moving privacy information; however, its actual implementation requires matching security, accuracy, and performance. Moreover, identifying the optimal data item in the absence of a unique matching key is necessary. We aimed to conduct a basic matching experiment using a model to assess the accuracy of cancer screening. MethodsTo experiment with actual data, we created a data set mimicking the cancer screening and registration data in Japan and conducted a matching experiment using a PDDI system between geographically distant institutions. Errors similar to those found empirically in data sets recorded in Japanese were artificially introduced into the data set. The matching-key error rate of the data common to both data sets was set sufficiently higher than expected in the actual database: 85.0% and 59.0% for the data simulating colorectal and breast cancers, respectively. Various combinations of name, gender, date of birth, and address were used for the matching key. To evaluate the matching accuracy, the matching sensitivity and specificity were calculated based on the number of cancer-screening data points, and the effect of matching accuracy on the sensitivity and specificity of cancer screening was estimated based on the obtained values. To evaluate the performance, we measured central processing unit use, memory use, and network traffic. ResultsFor combinations with a specificity ≥99% and high sensitivity, the date of birth and first name were used in the data simulating colorectal cancer, and the matching sensitivity and specificity were 55.00% and 99.85%, respectively. In the data simulating breast cancer, the date of birth and family name were used, and the matching sensitivity and specificity were 88.71% and 99.98%, respectively. Assuming the sensitivity and specificity of cancer screening at 90%, the apparent values decreased to 74.90% and 89.93%, respectively. A trial calculation was performed using a combination with the same data set and 100% specificity. When the matching sensitivity was 82.26%, the apparent screening sensitivity was maintained at 90%, and the screening specificity decreased to 89.89%. For 214 data points, the execution time was 82 minutes and 26 seconds without parallelization and 11 minutes and 38 seconds with parallelization; 19.33% of the calculation time was for the data-holding institutions. Memory use was 3.4 GB for the PDDI server and 2.7 GB for the data-holding institutions. ConclusionsWe demonstrated the rudimentary feasibility of introducing a PDDI system for cancer-screening accuracy assessment. We plan to conduct matching experiments based on actual data and compare them with the existing methods.

BackgroundConventional transradial access (TRA) has been the preferred access for coronary intervention. Recently, distal radial access (DRA) is introduced as an alternative choice to reduce radial artery occlusion (RAO) risk. The study sought to assess the impact of DRA on early RAO using Doppler ultrasound in patients with ST-segment elevation myocardial infarction (STEMI) who underwent primary percutaneous coronary intervention (PCI).MethodsThis is a prospective, single-center, open-label randomized clinical trial in which patients with indications for primary PCI from January 2022 to September 2022 were assigned to DRA or TRA group with 100 cases in each group. The primary endpoint was the incidence of forearm RAO, evaluated by Doppler ultrasound before discharge.ResultsThe rate of access success was comparable between the DRA and TRA groups (98.0 vs. 94.0%, P = 0.279). Compared with the TRA group, longer puncture time was observed in the DRA group [2.4 (1.7–4.2) min vs. 1.7 (1.4–2.3) min; P < 0.001] whereas the door-to-wire time was not delayed in primary PCI [71 (54–88) min vs. 64 (56–82) min, P = 0.103]. Shorter hemostasis time was required in the DRA group [3.1 (2.7–3.3) h vs. 6.2 (5.9–6.4) h; P < 0.001]. Significant reduction of the incidence of forearm RAO was observed in the DRA group (2.0 vs. 9.0%, P = 0.030). Local hematomas ≤ 5 cm was similar in both groups (4.0 vs. 6.0%, P = 0.516), while those > 5 cm were significantly more frequent in the TRA group (0 vs. 6.0%, P = 0.029).ConclusionDistal radial access is associated with a comparable lower incidence of forearm RAO, shorter hemostasis time, and lower rate of vascular complications compared to TRA in primary PCI.Systematic review registration[https://www.chictr.org.cn], identifier [ChiCTR2200061841].

Epitranscriptomics has emerged as another level of epigenetic regulation similar to DNA and histone modifications. N6-methyladenosine (m6A) is one of the most prevalent and abundant posttranscriptional modifications, widely distributed in many biological species. The level of N6-methyladenosine RNA methylation is dynamically and reversibly regulated by distinct effectors including methyltransferases, demethylases, histone modification and metabolites. In addition, N6-methyladenosine RNA methylation is involved in multiple RNA metabolism pathways, such as splicing, localization, translation efficiency, stability and degradation, ultimately affecting various pathological processes, especially the oncogenic and tumor-suppressing activities. Recent studies also reveal that N6-methyladenosine modification exerts the function in immune cells and tumor immunity. In this review, we mainly focus on the regulatory mechanisms of N6-methyladenosine RNA methylation, the techniques for detecting N6-methyladenosine methylation, the role of N6-methyladenosine modification in cancer and other diseases, and the potential clinical applications.

BackgroundThe effect of a single transradial guiding catheter (STGC) for culprit vessel percutaneous coronary intervention (PCI) first on door-to-balloon (D2B) time remains unclear.Materials and methodsBetween February 2017 and July 2019, 560 patients with ST-elevation myocardial infarction (STEMI) were randomized into either the STGC group (n = 280) or the control group (n = 280) according to direct culprit vessel PCI with a STGC. In the STGC group, a dedicated transraidal guiding catheter (6F either MAC3.5 or JL3.5) was used for the treatment of electrocardiogram (ECG)-guided culprit vessel first and later contralateral angiography. In the control group, a universal diagnostic catheter (5F Tiger II) was used for complete coronary angiography, followed by guiding catheter selection for culprit vessel PCI. The primary endpoint was D2B time, and the secondary endpoint included catheterization laboratory door-to-balloon (C2B), procedural, fluoroscopy times, and major adverse cardiac events (MACE) at 30 days.ResultsThe median D2B time was significantly shorter in the STGC group compared to the control group (53.9 vs. 58.4 min; p = 0.003). The C2B, procedural, and fluoroscopy times were also shorter in the STGC group (C2B: 17.3 vs. 24.5 min, p < 0.001; procedural: 45.2 vs. 49.0 min, p = 0.012; and fluoroscopy: 9.7 vs. 11.3 min, p = 0.025). More patients achieved the goal of D2B time within 90 min (93.9% vs. 87.1%, p = 0.006) and 60 min (61.4% vs. 51.1%, p = 0.013) in the STGC group. Radial artery perforation (RAP) was significantly reduced in the STGC group compared with the control group (0.7% vs. 3.2%, P = 0.033). MACE at 30 days was similar (2.5% vs. 4.6%, P = 0.172) between the two groups.ConclusionECG-guided immediate intervention on culprit vessel with a STGC can reduce D2B, C2B, procedural, and fluoroscopy times (ECG-guided Immediate Primary PCI for Culprit Vessel to Reduce Door to Device Time; NCT03272451).

The Kuroshio Extension (KE) region is one of the most energetic regions in the global ocean where prominent mesoscale dynamics persistently occur. The spatial distribution and temporal evolution of the sea surface temperature (SST) gradient and mesoscale eddies in the KE are investigated. The SST gradient can be applied for identifying the fronts, and the SST gradient within two times the radii of the eddies is composited to quantify the impact of eddies on frontal activities. Depressed SST gradients are identified for eddies with both polarities, but prominent spatial variance in the SST gradient reveals that a large SST gradient is located to the north of anticyclones and along the south periphery for cyclones. The eddies are further separated into two groups depending on their location relative to the main path of the KE, as the background fields to the north and south of the KE are largely different. The spatial pattern, e.g., monopole and dipole features, and temporal variation in the SST gradient are fully studied over the lifespans of eddies. The results show that most eddies can significantly weaken the internal SST gradient and induce the horizontal redistribution of the SST gradient in surrounding regions. Cyclonic eddies north of the KE elevate the fronts along the periphery of eddies. The temporal variability in the SST gradient is prominent and largely varies for each group of eddies. This study offers quantitative analyses of the spatial and temporal relationships between eddies and fronts that are important for understanding the mesoscale dynamics in the world’s oceans.

Changes in the molecular composition of dissolved organic matter (DOM) and its light-absorbing component (CDOM) along the river–coastal ocean continuum are crucial for better understanding the source and fate of DOM in coastal oceans. Both optical (absorbance and fluorescence) techniques and ultrahigh-resolution mass spectrometry have been widely used to trace DOM cycling. However, to the best of our knowledge, the linkage between these two techniques is rarely investigated along the river–coastal ocean continuum. In this regard, bulk characterization, optical techniques, and ultrahigh-resolution mass spectrometry [Fourier transform ion cyclotron resonance mass spectrometer (FT-ICR MS)] were applied to determine the DOM chemistry along a spatial transect from the Yangtze River Estuary (YRE) to the East China Sea. The results showed that DOM in the YRE was mainly controlled by the mixing of freshwater and seawater. Besides terrestrial input, multiple sources (tributary input from the Huangpu River, wastewater input, and sediment resuspension and subsequent release at the turbidity maximum zone) have been identified. In addition, the linkages between CDOM and thousands of formulas were developed based on Spearman’s rank correlations between optical parameters and FT-ICR MS peaks. The linkages showed that the molecular groups associated with the optical parameters generally agreed with conventional biogeochemical interpretations. Nevertheless, each technique has its unique advantage and weakness in interpreting DOM composition. Therefore, the combination of the untargeted FT-ICR MS approach and optical techniques could be valuable for studying the DOM sources and transformation in large river estuarine systems.

Alfalfa (Medicago sativa L.) is an important forage for the development of herbivorous animal husbandry, which is widely planted in the cold climate areas of northern China, where there is low overwintering and forage yield in production, and fertilization is a vital production strategy. A field study was conducted to determine the response of alfalfa’s winter survival rate, yield, and yield components to different gradients of phosphate (P) fertilizer (0, 45, 90, and 135 kg P2O5 ha−1) in two types of low-phosphorus alkaline soils. The results show that the winter survival rates and forage yield increased at first and then decreased with the increase of P application; the greater winter survival rates were achieved at 45~90 kg ha−1 of P fertilizer applied, while the greater forage yield were achieved at 90 kg ha−1 in the two sites, and the suitable P fertilizer application rates for greater winter survival were less than those for greater forage yield; plant height and shoot mass made a greater contribution to forage yield than other yield components. To pursue greater forage yield, the suitable P fertilization rates for aeolian sandy soil and silt loam soil are 108.1 and 78.3 kg ha−1, respectively, based on the regression equations. Therefore, in cold winter and soil P deficiency areas, applying P fertilizer can be used as an effective strategy to improve alfalfa’s forage yield and persistence, and the most appropriate amount of fertilizer should be determined according to climate and soil conditions.

The area fluctuation of lakes directly affects the stability of the surrounding ecological environment. Research on the area evolution of lakes and the driving factors affecting it plays an important role in sustainable water resource management. In this study, Hulun Lake, located in the Hulunbuir grassland, was taken as the research object. Based on remote sensing images of the Hulun Lake area from 1986 to 2020, MNDWI interpretation was used to obtain the change law of lake surface area over a long time frame. Combined with natural factors and anthropogenic factors, Pearson correlation analysis and principal component analysis were used to analyze the driving force. The results showed that (1) in the past 35 years, the water surface area of Hulun Lake has decreased significantly. The dynamic change in water area could be divided into four stages. The areas with dramatic changes in water area are distributed mainly in the northeast and south of Hulun Lake. (2) In terms of natural factors, the meteorological factors based on evaporation and relative humidity, the runoff of rivers entering the lake, and the vegetation with medium-high coverage and medium-low coverage had significant effects. In terms of anthropogenic factors, the population had the most significant impact. The artificial water diversion project had different degrees of influence on the response of the Hulun Lake area change to natural factors. (3) Anthropogenic factors were the main driving force causing the rapid change in the Hulun Lake area from 2000 to 2016, explaining 48% of the change in the Hulun Lake area. These research results can provide a scientific basis for the development and utilization of water resources and sustainable development in the Hulun Lake area.

Evapotranspiration (ET) plays an important role in transferring water and converting energy in the land–atmosphere system. Accurately estimating ET is crucial for understanding global climate change, ecological environmental problems, the water cycle, and hydrological processes. Machine learning (ML) algorithms have been considered as a promising method for estimating ET in recent years. However, due to the limitations associated with the spatial–temporal resolution of the flux tower data commonly used as the target set in ML algorithms, the ability of ML to discover the inherent laws within the data is reduced. In this study, a hybrid framework was established to simulate ET in data-deficient areas. ET simulation results of a coupled model comprising the Budyko function and complementary principle (BC2021) were used as the target set of the random forest model, instead of using the flux station observation data. By combining meteorological and hydrological data, the monthly ET of the Inner Mongolia section of the Yellow River Basin (IMSYRB) was simulated from 1982 to 2020, and good results were obtained (R2 = 0.94, MAE = 3.82 mm/mon, RMSE = 5.07 mm/mon). Furthermore, the temporal and spatial variations in ET and the influencing factors were analysed. In the past 40 years, annual ET in the IMSYRB ranged between 241.38 mm and 326.37 mm, showing a fluctuating growth trend (slope = 0.80 mm/yr), and the summer ET accounted for the highest proportion in the year. Spatially, ET in the IMSYRB showed a regular distribution of high ET in the eastern region and low ET in the western area. The high ET value areas gradually expanded from east to west over time, and the area increased continuously, with the largest increase observed in the 1980s. Temperature, precipitation, and normalized difference vegetation index (NDVI) were found to be the most important factors affecting ET in the region and play a positive role in promoting ET changes. These results provide an excellent example of long-term and large-scale accurate ET simulations in an area with sparse flux stations.

Background: Lung cancer has the highest morbidity and mortality rate among types of malignant tumors, and as such, research into prolonging the survival time of patients is vital. The emergence of immune checkpoint inhibitors (ICIs) has greatly improved the survival of patients with non-small cell lung cancer (NSCLC), however, the lack of effective biomarkers to predict the prognosis of immunotherapy has made it difficult to maximize the benefits. T cell receptor (TCR) is one of the most important components for recognizing tumor cells, and with this study we aim to clarify the relationship between TCR coexpression and the prognosis of NSCLC patients receiving immunotherapy.Methods: Univariate COX regression, logistics regression, and KM survival analysis were used to evaluate the relationship between TCR coexpression and the prognosis of immunotherapy. Additionally, CIBERSORT, Gene Set Enrichment Analysis (GSEA), and single-sample GSEA (ssGSEA) algorithms were used to evaluate the tumor immune microenvironment (TIME) of NSCLC patients.Results: Univariate Cox regression analysis showed that the TCR coexpression signature can be used as a clinical prognostic indicator for NSCLC patients receiving immunotherapy (p = 0.0205). In addition, those in the NSCLC group with a high TCR coexpression signature had significantly improved progression-free survival (PFS) (p = 0.014). In the ICI treatment cohort (GSE35640). In addition, there was a high infiltration of CD8+T cells, activated memory CD4+T cells, and M1 macrophages in the TIME of those with a high TCR coexpression signature. The results of pathway enrichment analysis showed that patients with a high TCR coexpression signature had significantly activated signal pathways such as lymphocyte proliferation and activation, chemokine binding, and inflammatory cytokine production. Also, we found that patients with a high TCR coexpression signature had an elevated T cell inflammation gene expression profile (GEP).Conclusion: We show that the TCR coexpression signature may be useful as a new biomarker for the prognosis of NSCLC patients undergoing immunotherapy, with high signatures indicating better treatment response. Additionally, we found that patients with a high TCR coexpression signature had tumor immune microenvironments with beneficial anti-tumor characteristics.

Seventeen years of satellite observational data are used to describe the variability in sea surface temperature (SST) fronts and associated features, e.g., frontal intensity and probability, in the northwestern Pacific Ocean. Compared with the SST gradient and frontal probability, the frontal intensity is less impacted by background noise in satellite observations and precisely represents the variability in frontal patterns. The seasonal variability in frontal intensity is prominent, and the corresponding seasonality varies spatially. Fronts are more common during winter in the northern region around the Oyashio Current but are most common during spring in the Kuroshio Current and to the south of the Kuroshio Extension. The meridional migration of fronts is associated with the seasonal movement of the North Pacific subtropical gyre. Though overall averaged frontal intensity along the Kuroshio Extension is most prominent in the northwestern Pacific Ocean, the associated variability in fronts is highly complex. The current study reveals that seasonality itself cannot capture the whole picture of frontal features because eddy-induced intraseasonal variability and basin-scale signal-induced interannual variability can modulate frontal dynamics. In particular, the monthly frontal intensity in regions where the seasonal cycle is not significant depends on the Pacific Decadal Oscillation and North Pacific Gyre Oscillation. Furthermore, the oscillation of the Kuroshio Extension and associated mesoscale eddies can impact its intraseasonal variability. The comprehensive analysis of frontal intensity in the Kuroshio Extension is helpful for delineating regional dynamics and has the potential to improve our understanding of controls on marine primary production.

The lateral advection of sinking particles is a well-known phenomenon in the South China Sea (SCS) and has a significant impact on the estimation of the efficiency of the biological carbon pump. However, little is known about the sources and pathways of sinking particles. Here, we present benthic and freshwater diatom fluxes and relative abundances collected by a sediment trap deployed at a water depth of 1,000 m and more than 500 m above the seafloor in the northwestern SCS, indicating that laterally transported resuspended sediment accounts for a significant part of the particle flux to the deep sea. A Lagrangian particle tracking model (LPTM) revealed that the resuspended particles likely originated from the neighboring continental slope, approximately 12–145 km to the west of the study site. Sediment trap observations and the LPTM together indicated that the impact of resuspended sediment occurred mainly in the deep water, and especially strong sediment resuspension was related to summer monsoon-induced coastal upwelling. The results suggest that particle resuspension has an important impact on the biological pump as well as on paleoenvironmental reconstructions of the SCS.

A 30-day feeding trial was conducted to determine the effects of dietary glycyrrhizin (GL) on survival, growth performance, expression of feeding-related genes, activities of digestive enzymes, antioxidant capacity, and expression of inflammatory factors of large yellow croaker larvae with an initial weight of 3.78±0.27 mg. Four 53.80% crude protein and 16.40% crude lipid diets were formulated with supplementation of 0%, 0.005%, 0.01%, and 0.02% GL, respectively. Food-grade high internal-phase Pickering emulsions (HIPPEs) stabilized by solid or colloidal particles with different advantages have attracted extensive attention nowadays. However, looking for new appropriate particle stabilizers is the common practice for HIPPEs preparation. It is crucial to find a new strategy for the development of functional HIPPEs with controllable properties. In this study, a high concentration of xanthan gum/lysozyme nanoparticles (XG/Ly NPs) was used for the preparation of HIPPEs for the first time. Visual observations, creaming index (CI), microstructure, and rheology tests were carried out to investigate the potential of XG/Ly NPs as HIPPEs stabilizers. Results indicated that XG/Ly NPs could stabilize oil droplets in the concentration range of 0.5–4% (w/v). The HIPPEs with a minimal particle concentration of 1% exhibited remarkable physical stability. Rheological measurements showed that a high stability of solid-like HIPPEs was successfully obtained with a higher concentration of XG/Ly NPs. Overall, the HIPPEs stabilized by different concentrations of XG/Ly NPs exhibited excellent rheological and structural properties, which might provide a feasible strategy for the development of functional emulsion systems with controllable structures.

Abstract Background Two-pore channels (TPCs) release Ca2+ from acidic intracellular stores and are implicated in a number of diseases, but their role in development is unclear. The social amoeba Dictyostelium discoideum proliferates as single cells that aggregate to form a multicellular organism on starvation. Starvation is sensed by the mTORC1 complex which, like TPC proteins, is found on acidic vesicles. Here, we address the role of TPCs in development and under starvation. Results We report that disruption of the gene encoding the single Dictyostelium TPC protein, TPC2, leads to a delay in early development and prolonged growth in culture with delayed expression of early developmental genes, although a rapid starvation-induced increase in autophagy is still apparent. Ca2+ signals induced by extracellular cAMP are delayed in developing tpc2 − cells, and aggregation shows increased sensitivity to weak bases, consistent with reduced acidity of the vesicles. In mammalian cells, the mTORC1 protein kinase has been proposed to suppress TPC channel opening. Here, we show a reciprocal effect as tpc2 − cells show an increased level of phosphorylation of an mTORC1 substrate, 4E-BP1. mTORC1 inhibition reverses the prolonged growth and increases the efficiency of aggregation of tpc2 − cells. Conclusion TPC2 is required for efficient growth development transition in Dictyostelium and acts through modulation of mTORC1 activity revealing a novel mode of regulation.

Traffic sign detection is a very important part of intelligent assisted driving system. However, with the interference of various target sizes, geometric distortion, occlusion and motion blur, fast and accurate detection on large-size car camera image is extremely hard. To achieve both high efficient and accurate detection, we present a traffic sign detection method within a coarse-to-fine framework, which sequentially detects the targets in grid-level and image-level. We demonstrate that focusing first is a more effective detection strategy for small targets in wide detection space. We propose a target grid prediction network, which is a fully convolutional network for binary classification, to realize rapid coarse localization of the target and effectively guide the clipping and scaling of the target area. With the flexible potential target region extracting strategy, the detecting space can be significantly reduced. At the same time, the correctly extracted local areas for the targets can further facilitate the accurate detection of the subsequent traffic sign detector. In the experiments, our method achieves impressive performance in terms of both efficiency and accuracy. On the challenging Tsinghua-Tencent 100K (TT100K) dataset, our method achieves 20.9 FPS detection speed for 1600 × 1600 images and the F1-score of the proposed method for all-scale targets is 91.55.

Voice communication using an air-conduction microphone in noisy environments suffers from the degradation of speech audibility. Bone-conduction microphones (BCM) are robust against ambient noises but suffer from limited effective bandwidth due to their sensing mechanism. Although existing audio super-resolution algorithms can recover the high-frequency loss to achieve high-fidelity audio, they require considerably more computational resources than is available in low-power hearable devices. This paper proposes the first-ever real-time on-chip speech audio super-resolution system for BCM. To accomplish this, we built and compared a series of lightweight audio super-resolution deep-learning models. Among all these models, ATS-UNet was the most cost-efficient because the proposed novel Audio Temporal Shift Module (ATSM) reduces the network’s dimensionality while maintaining sufficient temporal features from speech audio. Then, we quantized and deployed the ATS-UNet to low-end ARM micro-controller units for a real-time embedded prototype. The evaluation results show that our system achieved real-time inference speed on Cortex-M7 and higher quality compared with the baseline audio super-resolution method. Finally, we conducted a user study with ten experts and ten amateur listeners to evaluate our method’s effectiveness to human ears. Both groups perceived a significantly higher speech quality with our method when compared to the solutions with the original BCM or air-conduction microphone with cutting-edge noise-reduction algorithms.

The Taklamakan Desert and the Gobi Desert in East Asia constitute the second-largest sources of dust in the world. In particular, dust originating from the Gobi Desert is more susceptible to long-range transport, with consequent impacts in downwind Asian countries and the Northwest Pacific region. Two intensive dust events (the 3·15 dust event and the 3·28 dust event) were experienced in North China in March 2021. The 3·15 dust process was rated as the most intensive dust process in China in the past 10 years. In this study, by using a combination of spaceborne remote sensing datasets from geostationary and polar-orbiting satellites, ground-based columnar observations of aerosol optical parameters, meteorological reanalysis data, and backward trajectory simulations of air masses, the transport pathways and the three-dimensional structure characteristics of dust aerosols during the transport of the two dust events in March 2021 were cross-validated. The results of the study indicated that the two dust events were induced by the Mongolian cyclone. Due to the different configurations of the ground meteorological system conditions, a backflow process occurred during the 3·15 dust event transmission process. After passing over North China and the Bohai Sea, the direction of transport of the dust plume was reversed. The wind deflected from northwest to northeast, and the dust reached the eastern coastal areas of China and was finally deposited on land. The 3·28 dust event exhibited aerosol stratification in the transport path, the higher pure dust layer reached up to 9 km height, and the lower layer underwent aerosol mixing and became a polluted dust aerosol. This study implies that the investigation of dust aerosol transport and the deposition processes, the impact on the ocean, and the impact of marine aerosols on land also needs to be taken into consideration; the integration of advanced satellites and ground-based remote sensing data, the meteorological reanalysis data and the backward trajectories simulation, which complemented and verified each other, can enhance the ability to delineate the transport pathways and the three-dimensional structural characteristics of dust events.

The subarctic Pacific is one of the major high-nitrate, low-chlorophyll (HNLC) regions where marine productivity is greatly limited by the supply of iron (Fe) in the region. There is a distinct seasonal difference in the chlorophyll concentrations of the east and west sides of the subarctic Pacific because of the differences in their driving mechanisms. In the western subarctic Pacific, two chlorophyll concentration peaks occur: the peak in spring and early summer is dominated by diatoms, while the peak in late summer and autumn is dominated by small phytoplankton. In the eastern subarctic Pacific, a single chlorophyll concentration peak occurs in late summer, while small phytoplankton dominate throughout the year. In this study, two one-dimensional (1D) physical–biological models with Fe cycles were applied to Ocean Station K2 (Stn. K2) in the western subarctic Pacific and Ocean Station Papa (Stn. Papa) in the eastern subarctic Pacific. These models were used to study the role of Fe limitation in regulating the seasonal differences in phytoplankton populations by reproducing the seasonal variability in ocean properties in each region. The results were reasonably comparable with observational data, i.e., cruise and Biogeochemical-Argo data, showing that the difference in bioavailable Fe (BFe) between Stn. K2 and Stn. Papa played a dominant role in controlling the respective seasonal variabilities of diatom and small phytoplankton growth. At Stn. Papa, there was less BFe, and the Fe limitation of diatom growth was two times as strong as that at Stn. K2; however, the difference in the Fe limitation of small phytoplankton growth between these two regions was relatively small. At Stn. K2, the decrease in BFe during summer reduced the growth rate of diatoms, which led to a rapid reduction in diatom biomass. Simultaneously, the decrease in BFe had little impact on small phytoplankton growth, which helped maintain the relatively high small phytoplankton biomass until autumn. The experiments that stimulated a further increase in atmospheric Fe deposition also showed that the responses of phytoplankton primary production in the eastern subarctic Pacific were stronger than those in the western subarctic Pacific but contributed little to primary production, as the Fe limitation of phytoplankton growth was replaced by macronutrient limitation.
