Abstract Background Chronic kidney disease (CKD) is the third most common cause of death after cancer and heart disease. The continuous treatment of children with CKD was greatly challenged during the COVID-19 pandemic, which significantly impacted the CKD children’s prognosis and the caregivers’ psychological status. However, the influence mechanism of socioeconomic status, medical delay duration, traffic pressure, and online consultation duration on caregivers’ hope and psychological resilience still lacks relevant evidence. Methods This study investigated the general social information, hope, and psychological resilience of 247 caregivers with CKD in 13 provinces of China in March 2020. Factor analysis and an exploratory Structural Equation Model ( SEM ) were used to find the best-fit model, and Bootstrapping was used to calculate the 95% CI of indirect effects. Results The factor analysis obtained four common factors, namely, socioeconomic status (annual family income, education, and career stability), medical accessibility (online consultation duration, medical delay duration, and traffic pressure), hope (positive attitude, positive action, and intimate relationship) and psychological resilience (optimism, tenacity, and strength), with the cumulative contribution rate of 65.34%. Bootstrapping obtains the socioeconomic status β = 0.30 (95% CI [0.14, 0.47], P = 0.002), medical accessibility β = 0.31 (95% CI [0.12, 0.47], P = 0.002), and hope β = 0.40 (95% CI [0.27, 0.52], P = 0.002) has a direct impact on psychological resilience of CKD children caregivers, followed by medical accessibility β = 0.20 (95% CI [0.10, 0.38], P = 0.001) and hope β = 0.23 (95% CI [0.16, 0.32], P = 0.001) plays a mediating role between socioeconomic status and psychological resilience. The indirect impact effect β = 0.35 (95% CI [0.25, 0.50], P = 0.001) is greater than the direct impact effect β = 0.30 (95% CI [0.14, 0.47], P = 0.002). Conclusions Sufficient attention should still be given to children with immunodeficiency after the COVID-19 pandemic, such as CKD, to avoid infection of deadly. Secondly, the government should vigorously develop Primary medical institutions to ensure efficient treatment of severe patients in tertiary hospitals; Finally, the professional literacy of medical workers in remote diagnosis and treatment should be improved to enhance the country’s emergency response capacity for similar major public events and the requirements for normalised epidemic prevention and control.

Abstract Colorectal cancer (CRC) stands as a major contributor to cancer-related fatalities within China. There is an urgent need to identify accurate biomarkers for recurrence predicting in CRC. Reduced representation bisulfite sequencing was used to perform a comparative analysis of methylation profiles in tissue samples from 30 recurrence to 30 non-recurrence patients with CRC. Least absolute shrinkage and selection operator method was performed to select the differential methylation regions (DMRs) and built a DNA methylation classifier for predicting recurrence. Based on the identified top DMRs, a methylation classifier was built and consisted of eight hypermethylated DMRs in CRC. The DNA methylation classifier showed high accuracy for predicting recurrence with an area under the receiver operator characteristic curve of 0.825 (95% CI 0.680–0.970). The Kaplan–Meier survival analysis demonstrated that CRC patients with high methylation risk score, evaluated by the DNA methylation classifier, had poorer survival than low risk score (Hazard Ratio 4.349; 95% CI 1.783–10.61, P = 0.002). And only CRC patients with low methylation risk score could acquire benefit from adjuvant therapy. The DNA methylation classifier has been proved as crucial biomarkers for predicting recurrence and exhibited promising prognostic value after curative surgery in patients with CRC.

In unstructured environments, dense grape fruit growth and the presence of occlusion cause difficult recognition problems, which will seriously affect the performance of grape picking robots. To address these problems, this study improves the YOLOX-Tiny model and proposes a new grape detection model, YOLOX-RA, which can quickly and accurately identify densely growing and occluded grape bunches. The proposed YOLOX-RA model uses a 3 × 3 convolutional layer with a step size of 2 to replace the focal layer to reduce the computational burden. The CBS layer in the ResBlock_Body module of the second, third, and fourth layers of the backbone layer is removed, and the CSPLayer module is replaced by the ResBlock-M module to speed up the detection. An auxiliary network (AlNet) with the remaining network blocks was added after the ResBlock-M module to improve the detection accuracy. Two depth-separable convolutions (DSC) are used in the neck module layer to replace the normal convolution to reduce the computational cost. We evaluated the detection performance of SSD, YOLOv4 SSD, YOLOv4-Tiny, YOLO-Grape, YOLOv5-X, YOLOX-Tiny, and YOLOX-RA on a grape test set. The results show that the YOLOX-RA model has the best detection performance, achieving 88.75 % mAP, a recognition speed of 84.88 FPS, and model size of 17.53 MB. It can accurately detect densely grown and shaded grape bunches, which can effectively improve the performance of the grape picking robot.

Abstract Auricularia heimuer, the third most frequently cultivated edible mushroom species worldwide, has high medicinal value. However, a shortage of molecular marker hinders the efficiency and accuracy of genetic breeding efforts for A. heimuer. High-throughput transcriptome sequencing data are essential for gene discovery and molecular markers development. This study aimed to clarify the distribution of SSR loci across the A. heimuer transcriptome and to develop highly informative EST-SSR markers. These tools can be used for phylogenetic analysis, functional gene mining, and molecular marker-assisted breeding of A. heimuer. This study used Illumina high-throughput sequencing technology to obtain A. heimuer transcriptome data. The results revealed 37,538 unigenes in the A. heimuer transcriptome. Of these unigenes, 24,777 (66.01%) were annotated via comparison with the COG, Pfam, and NR databases. Overall, 2510 SSRs were identified from the unigenes, including 6 types of SSRs. The most abundant type of repeats were trinucleotides (1425, 56.77%), followed by mononucleotides (391, 15.58%) and dinucleotides (456, 18.17%). Primer pairs for 102 SSR loci were randomly designed for validity confirmation and polymorphism identification; this process yielded 53 polymorphic EST-SSR markers. Finally, 13 pairs of highly polymorphic EST-SSR primers were used to analyze the genetic diversity and population structure of 52 wild A. heimuer germplasms, revealing that the 52 germplasms could be divided into three categories. These results indicated that SSR loci were abundant in types, numbers, and frequencies, providing a potential basis for germplasm resource identification, genetic diversity analysis, and molecular marker-assisted breeding of A. heimuer.

Abstract A 71‐year‐old male developed anastomotic tumor bleeding after subtotal gastrectomy (Billroth II), and the surgery was refused due to coronary atherosclerotic heart disease. Endoscopic sclerotherapy (ES) in the treatment of esophagogastric variceal bleeding has been commonly reported, but few in non‐variceal upper gastrointestinal bleeding. As far as we know, endoscopic sclerotherapy for malignant tumor hemorrhage has not been reported. Here a valuable case is presented: endoscopic sclerotherapy was performed on the anastomotic tumor bleeding, which is an effective try in a particular period.

Early mutation identification guides patients with colorectal cancer (CRC) toward targeted therapies. In the present study, 414 patients with CRC were enrolled, and amplicon-based targeted next-generation sequencing (NGS) was then performed to detect genomic alterations within the 73 cancer-related genes in the OncoAim panel. The overall mutation rate was 91.5 % (379/414). Gene mutations were detected in 38/73 genes tested. The most frequently mutated genes were TP53 (60.9 %), KRAS (46.6 %), APC (30.4 %), PIK3CA (15.9 %), FBXW7 (8.2 %), SMAD4 (6.8 %), BRAF (6.5 %), and NRAS (3.9 %). Compared with the wild type, TP53 mutations were associated with low microsatellite instability/microsatellite stability (MSI-L/MSS) (P = 0.007), tumor location (P = 0.043), and histological grade (P = 0.0009); KRAS mutations were associated with female gender (P = 0.026), distant metastasis (P = 0.023), TNM stage (P = 0.013), and histological grade (P = 0.004); APC mutations were associated with patients

Abstract Although Dynamic Movement Primitives (DMP) is an effective tool for robotic arm trajectory generalisation, the application of DMP in the 3C (Computer, Communication, Consumer Electronics) industry still faces challenges such as low precision and high‐time consumption. To address this problem, we propose a novel Cauchy DMP framework. The main improvements and advantages of Cauchy DMP, compared to the original DMP, are (1) since the Cauchy distribution has a simpler model and wider shape, using the Cauchy distribution instead of the Gaussian distribution in the original DMP reduces the complexity of the algorithm and saves time. (2) Singular Value Decomposition (SVD) can effectively model the error. To reduce the interference of the rounding and human error on the trajectory, SVD can be used to obtain the weight of each basis function. The proposed Cauchy DMP framework combines the above two points and is validated on a real UR5 robotic arm. The results show that Cauchy DMP retains the learnability of the original DMP and has the advantages of short time consumption and low error rate.

Objective To investigate the correlations between multigene alterations and clinicopathological features in papillary thyroid carcinoma (PTC) samples. Methods In this retrospective study, 111 cytological specimens of thyroid nodules, including 74 PTC samples and 37 benign samples, were analyzed using a 22-gene mutation assay employing next-generation sequencing. Abstract Background Hemorrhagic fever with renal syndrome (HFRS) caused by Orthohantavirus (OHV) and scrub typhus (ST) caused by Orientia tsutsugamushi (OT) are two infectious diseases prevalent in southwest China. Rodents are the natural host and the main source of the two diseases. OT infection to humans is usually resulted from bite of an infective chigger mite on rodents, and OHV is transmitted through contact or inhalation of aerosols and secretions from infected rodent. The use of antibiotics and hormones is crucial for infectious diseases, although the clinical manifestations are not obvious and a definitive diagnosis becomes more difficult in the presence of these drugs. Clinically, fever is the first symptom of these two diseases, and most of them are accompanied by common symptoms such as chills and headaches. The clinical symptoms of these two diseases are very similar and therefore it is not easy to make a differential diagnosis. Case presentation In this case, a 44-year-old male famer with pulmonary tuberculosis and a history of working in coal transportation was admitted to the hospital because of respiratory symptoms accompanied by fever, headache, and skin rashes on his body. Biochemical and urinalysis revealed the hepatic and renal injury. The subsequent molecular testing confirmed he suffered from HFRS and scrub typhus simultaneously that the serological and clinical diagnosis could not identify the cause of infection before. Such case has not been reported in Yunnan Province before. Conclusion The clinical diagnosis should be combined with serological and nucleic acid testing approaches for differential diagnosis in areas where HFRS and ST are endemic.

Geographical differences are conspicuous in lung cancer, and the distinct molecular features of lung tumor between Western patients and Asian patients have been demonstrated. However, the etiology of non-small-cell lung cancer (NSCLC) and the distribution of associated molecular aberrations in China have not been fully elucidated. The mutational profiles of 12 lung cancer-related genes were investigated in 85 patients from eastern China and 88 patients from southern China who had been histologically confirmed NSCLC. Overall, 93.6% (162/173) of tumor samples harbored at least one somatic alteration. The most frequently mutated genes were TP53 (56.1%), EGFR (50.3%), and KRAS (14.5%). We found that EGFR mutated much more frequently (60.0% vs 40.9%, P = 0.012) and TP53 mutations had significantly lower incidence (47.1% vs 64.8%, P = 0.019) in eastern cohort than that in southern cohort. Mutational signature analysis revealed a region-related mutagenesis mechanism characterized by a high prevalence of C to T transitions mainly occurring at CpG dinucleotides in southern patients. This study reveals the difference in the mutational features between NSCLC patients in eastern and southern China. The distinct patterns of gene mutation could provide clues for the mechanism of carcinogenesis of lung cancer, offering opportunities to stratify patients into optimal treatment plans based on genomic profiles.

The farming–pastoral zone in northern China is one of the most ecologically sensitive areas globally, having experienced extensive cropland abandonment and land reclamation over decades, primarily influenced by policy adjustment and global warming. However, the spatiotemporal patterns and suitability of long-term cropland change remain poorly understood. Using the annual China land cover dataset (CLCD), we provide a cropland abandonment and land reclamation mapping approach based on actual land use processes (rather than land cover conditions) to investigate spatiotemporal features of abandonment and reclamation and evaluate the rationality. Our findings show that: (1) Returning farmland to forest and grassland has been a clear trend in the study area over the past 30 years. Specifically, cropland use has undergone three phases of change, i.e., cropland contraction and expansion alternately (before 2000), followed by substantial abandonment (after 2000), and low-intensity reclamation (after 2010). (2) In the last decade, the intensity of the abandonment of cropland with high and moderate suitability is low. The rate of abandonment decreased, while the intensity of land reclamation was relatively high. The rate of the reclamation increased, and the spatial distribution of cropland tended to be reasonable. Our study emphasizes the importance of monitoring actual cropland changes based on land use processes, and this method can be effectively extended to regional or global long-term cropland monitoring.

The genus Hepacivirus contains single-stranded positive-sense RNA viruses belonging to the family Flaviviridae, which comprises 14 species. These 14 hepaciviruses have been found in different mammals, such as primates, dogs, bats, and rodents. To date, Hepacivirus has not been reported in the shrew genus of Crocidura. To study the prevalence and genetic evolution of Hepacivirus in small mammals in Yunnan Province, China, molecular detection of Hepacivirus in small mammals from Yunnan Province during 2016 and 2017 was performed using reverse-transcription polymerase chain reaction (RT-PCR). Our results showed that the overall infection rate of Hepacivirus in small mammals was 0.12% (2/1602), and the host animal was the Southeast Asian shrew (Crocidura fuliginosa) (12.5%, 2/16). Quantitative real-time PCR showed that Hepacivirus had the highest viral RNA copy number in the liver. Phylogenetic analysis revealed that the hepaciviruses obtained in this study does not belong to any designated species of hepaciviruses and forms an independent clade. To conclude, a novel hepacivirus was identified for the first time in C. fuliginosa specimens from Yunnan Province, China. This study expands the host range and viral diversity of hepaciviruses.

Abstract Background Data on the genus Sarcocystis in insectivores are limited. The Asian gray shrew Crocidura attenuata is one of the most common species of the insectivore family Soricidae in South Asia and Southeast Asia. To our knowledge, species of Sarcocystis have never been recorded previously in this host. Methods Tissues were obtained from 42 Asian gray shrews caught in 2017 and 2018 in China. Sarcocysts were observed using light microscopy (LM) and transmission electron microscopy (TEM). To describe the parasite life cycle, muscle tissues of the host infected with sarcocysts were force-fed to two beauty rat snakes Elaphe taeniura. Individual sarcocysts from different Asian gray shrews, and oocysts/sporocysts isolated from the small intestines and feces of the experimental snakes, were selected for DNA extraction, and seven genetic markers, namely, two nuclear loci [18S ribosomal DNA (18S rDNA) and internal transcribed spacer region 1 (ITS1)], three mitochondrial genes [cytochrome oxidase subunit 1 (cox1), cox3 and cytochrome b], and two apicoplast genes (RNA polymerase beta subunit and caseinolytic protease C), were amplified, sequenced and analyzed. Results Sarcocysts were found in 17 of the 42 (40.5%) Asian gray shrews. Under LM, the microscopic sarcocysts showed saw- or tooth-like protrusions measuring 3.3–4.5 μm. Ultrastructurally, the sarcocyst wall contained numerous lancet- or leaf-like villous protrusions, similar to those described for type 9h of the common cyst wall classification. The experimental beauty rat snakes shed oocysts/sporocysts measuring 11.9–16.7 × 9.2–10.6 μm with a prepatent period of 10–11 days. Comparison of the newly obtained sequences with those previously deposited in GenBank revealed that those of 18S rDNA and cox1 were most similar to those of Sarcocystis scandentiborneensis recorded in the tree shrews Tupaia minor and Tupaia tana (i.e., 97.6–98.3% and 100% identity, respectively). Phylogenetic analysis based on 18S rDNA or ITS1 sequences placed this parasite close to Sarcocystis spp. that utilize small animals as intermediate hosts and snakes as the known or presumed definitive host. On the basis of morphological and molecular characteristics and host specificity, the parasite was proposed as a new species, named Sarcocystis attenuati. Conclusions Sarcocysts were recorded in Asian gray shrews, to our knowledge for the first time. Based on morphological and molecular characterization, a new species of parasite is proposed: Sarcocystis attenuati. According to the LM and TEM results, S. attenuati sarcocysts are distinct from those of Sarcocystis spp. in other insectivores and those of S. scandentiborneensis in tree shrews. The 18S rDNA or cox1 sequences of Sarcocystis attenuati shared high similarity with those of Sarcocystis scandentiborneensis, Sarcocystis zuoi, Sarcocystis cf. zuoi in the Malayan field rat, and Sarcocystis sp. in the greater white-toothed shrew. Therefore, we suggest that more research on the relationships of these closely related taxa should be undertaken in the future. Graphical abstract

Hemorrhagic fever with renal syndrome (HFRS) is a zoonotic disease transmitted by several rodent species. We obtained clinical data of HFRS patients from the medical records of the People’s Hospital of Xiangyun County in Dali Prefecture from July 2019 to August 2021. We collected epidemiological data of HFRS patients through interviews and investigated host animals using the night clip or night cage method. We systematically performed epidemiological analyses of patients and host animals. The differences in the presence of rodent activity at home (χ2 = 8.75, p = 0.031 < 0.05), of rodent-proof equipment in the food (χ2 = 9.19, p = 0.025 < 0.05), and of rodents or rodent excrement in the workplace (χ2 = 10.35, p = 0.014 < 0.05) were statistically different in the four clinical types, including mild, medium, severe, and critical HFRS-associated diseases. Furthermore, we conducted molecular detection of orthohantavirus in host animals. The total orthohantavirus infection rate of rodents was 2.72% (9/331); the specific infection rate of specific animal species was 6.10% (5/82) for the Apodemus chevrieri, 100% (1/1) for the Rattus nitidus, 3.77% (2/53) for the Rattus norvegicus, and 12.50% (1/8) for the Crocidura dracula. In this study, a total of 21 strains of orthohantavirus were detected in patients and rodents. The 12 orthohantavirus strains from patients showed a closer relationship with Seoul orthohantavirus (SEOOV) L0199, DLR2, and GZRn60 strains; the six orthohantavirus strains from Rattus norvegicus and Apodemus chevrieri were closely related to SEOOV GZRn60 strain. One strain (XYRn163) from Rattus norvegicus and one strain (XYR.nitidus97) from Rattus nitidus were closely related to SEOOV DLR2 strain; the orthohantavirus strain from Crocidura dracula was closely related to the Luxi orthohantavirus (LUXV) LX309 strain. In conclusion, patients with HFRS in Xuangyun County of Dali Prefecture are predominantly affected by SEOOV, with multiple genotypes of orthohantavirus in host animals, and, most importantly, these orthohantavirus strains constantly demonstrated zoonotic risk in humans.

Grassland gross primary productivity (GPP) is an important part of global terrestrial carbon flux, and its accurate simulation and future prediction play an important role in understanding the ecosystem carbon cycle. Machine learning has potential in large-scale GPP prediction, but its application accuracy and impact factors still need further research. This paper takes the Mongolian Plateau as the research area. Six machine learning methods (multilayer perception, random forest, Adaboost, gradient boosting decision tree, XGBoost, LightGBM) were trained using remote sensing data (MODIS GPP) and 14 impact factor data and carried out the prediction of grassland GPP. Then, using flux observation data (positions of flux stations) and remote sensing data (positions of non-flux stations) as reference data, detailed accuracy evaluation and comprehensive trade-offs are carried out on the results, and key factors affecting prediction performance are further explored. The results show that: (1) The prediction results of the six methods are highly consistent with the change tendency of the reference data, demonstrating the applicability of machine learning in GPP prediction. (2) LightGBM has the best overall performance, with small absolute error (mean absolute error less than 1.3), low degree of deviation (root mean square error less than 3.2), strong model reliability (relative percentage difference more than 5.9), and a high degree of fit with reference data (regression determination coefficient more than 0.97), and the prediction results are closest to the reference data (mean bias is only −0.034). (3) Enhanced vegetation index, normalized difference vegetation index, precipitation, land use/land cover, maximum air temperature, potential evapotranspiration, and evapotranspiration are significantly higher than other factors as determining factors, and the total contribution ratio to the prediction accuracy exceeds 95%. They are the main factors influencing GPP prediction. This study can provide a reference for the application of machine learning in GPP prediction and also support the research of large-scale GPP prediction.

Individuals' health status is an essential indicator of the overall strength of a country. Existing literature has studied the determinants of individuals' health, but there is no direct evidence to date on the impact of mobile payment on health. To supplement relevant research and provide insightful policy suggestions to families, government and societies, based on data of 32,058 observations from the 2017 China Household Finance Survey, we estimate the effects of mobile payment on physical health using ordinary least squares and two-stage least squares strategy. This paper provides direct evidence that mobile payment has a positive impact on citizens' physical health. Heterogeneity analysis shows that mobile payment has a more profound impact on the health of citizens who are rural and less educated. Finally, further findings in this paper suggest that commercial insurance and leisure consumption are the mechanism through which the link between mobile payment and individuals' health operates.

The transition zone between the Mu Us Sandy Land and the Loess Plateau is considered an ecologically fragile area. However, significant changes in land use have occurred in the past few decades due to changes in land policies and the implementation of major national ecological projects. Despite this, there is still a lack of clear investigation into the impact of these changes on the landscape structure and ecological health of the area. This study utilizes high-resolution annual land use data from China, along with multi-index models and algorithms, to comprehensively analyze regional land use changes, landscape patterns, and the ecological environment’s quality. Through a comprehensive analysis of various factors, including changes in quantity, transformation in land types, spatial dynamics, landscape structure, and ecological quality, we aim to provide a better understanding of the complex interactions between land use and ecological systems in this area. The research results indicate that: (1) Since 2000, 9057.4 km2 of land in the study area has undergone changes. The grassland area has the largest increase, the forest area has the fastest growth rate, while cropland and barren land have decreased to varying degrees, and impervious surface has slightly expanded. (2) The movement trajectory of the center of gravity for different land types is closely related to human activities such as land development and utilization, as well as ecological restoration. Land changes have resulted in an escalation of landscape fragmentation, a reduction in landscape diversity, and a decline in the uniform distribution of different types. (3) Ecological land is the key to improving the ecological environment. The increase in ecological land area in the study area has led to an improvement in the quality of the ecological environment. The net contribution rate of land change to ecological improvement reaches 1.99%. The analysis methods and perspectives used in this study can be applied to other similar studies. The study’s findings enhance the understanding of how land and vegetation changes affect the ecological environment in this crucial area. They are of great significance in guiding the development and utilization of land resources and the implementation of ecological environment projects.

Beauty tea with special flavor can be affected by the degree of leafhopper puncturing. The present research adopted widely targeted metabolomics to analyze the characteristic metabolites of fresh tea leaves and beauty tea with different degrees of leafhopper puncturing. Low-puncturing beauty tea (LPBT) exhibited a superior quality. Altogether, 95 and 65 differential metabolites, including tea polyphenols, saccharides, and lipids, were identified from fresh leaves and beauty tea, respectively. The partial least squares regression (PLSR) analysis results showed that isomaltulose, theaflavic acid, and ellagic acid, may be the characteristic metabolites that form the different taste outlines of beauty tea. Based on odor activity values (OAVs) and partial least squares discriminant analysis (PLS-DA), dihydrolinalool and cis-linalool oxide were identified as characteristic volatile components, which may be essential for the formation of the different aroma characteristic of beauty tea. The results provide a theoretical basis for selecting raw materials, performing quality research, and developing beauty tea industrially.

This paper examines the effect of the image of destinations on the wage income of resident households, and the corresponding income inequality, from a novel perspective. This work uses China’s excellent tourism city image program, which is an urban planning policy implemented by the central government across cities to enhance the image of the city destination in the minds of tourists, and then promote tourist motivation and local tourism development to assess the effect on household wage income and its distribution. Results show that the program significantly increases household wage income by increasing employment opportunities, promoting business and population agglomeration, and improving urban infrastructure. Additionally, the image of the city destination promotion causes an increase in income inequality among households.

Using the triaxial shear or compressive strength as a single index of the resistance of frozen soils to failure does not always meet frozen soil engineering requirements for the comprehensive evaluation of the resistance. In this study, triaxial compression experiments were carried out on undisturbed ice-rich frozen clay samples with various levels of water content under different confining pressures to study the characteristics of the failure strain energy density of the samples. The results indicate that as the confining pressure increased, the failure strain energy density first increased and then decreased. The failure strain energy density reached a maximum at a critical confining pressure of 2.00 MPa for 13.25–25.76% water content and 1.00 MPa for 26.02–45.82% water content. The failure strain energy density increased as the water content increased at low confining pressures (0.05–0.50 MPa) but then declined slightly at intermediate confining pressures (1.00–2.00 MPa). At a high confined pressure of 3.00 MPa, the failure strain energy density decreased overall as the water content increased. There were similarities and differences between the change characteristics of the compressive strength and the failure strain energy density. The failure strain energy density can be used as a supplementary reference index of the resistance of frozen soils to damage. The variation characteristics of the failure strain energy density of undisturbed frozen clay are essentially consistent with those of remolded frozen sandy soils. However, there are also clear differences between the characteristics of the failure strain energy density of these two types of frozen soil.

To carry out engineering construction in a permafrost area, it is of great importance to grasp the spatial distribution of permafrost and its hydrothermal characteristics within the scope of engineering activities. Compared with the continuous permafrost area, the permafrost distribution in discontinuous permafrost area is complicated due to existence of island talik, which causes highly poor continuity and uniformity of ground hydrothermal conditions and brings great difficulty and uncertainty to the engineering geological investigation. In the present study, the spatial distribution of permafrost and the freeze-thaw process of shallow ground at the I-stage terrace of a river on the Qinghai-Tibet Plateau were studied based on field investigation including drilling, electrical resistivity tomography, and borehole temperature measurement. The results show that the relict permafrost, patchy permafrost, and island talik are distributed in the limited range of the study area. Among them, the buried depth of relict permafrost layer is in the range of 7 ~ 14 m, while the patchy permafrost layer is in the range of 3.0 ~ 17.0 m depth, and the mean annual ground temperature of both is close to -0.1°C, which belongs to extremely high-temperature permafrost. The seasonal freezing/thawing depths of shallow ground within the relict permafrost, patchy permafrost, and island talik range from 2.5 to 3.0 m. The freezing period of the seasonal freezing layer is 5.5 months, while the thawing period of the seasonal thawing layer is 6 months. The complex plane and spatial distribution of permafrost in the field are closely related to local factors such as topography, geomorphology, groundwater, and lithology of shallow ground. It is believed that the formation of the island talik in the study area is controlled by solar radiation and precipitation infiltration.

This paper aims to investigate the impact of environmental regulations on trade patterns, in terms of trade in environmental goods listed in Asia-Pacific Economic Cooperation (APEC) and Organization for Economic Co-operation and Development (OECD). Environmental goods are defined here as those that enable sustainable growth and reduce pollution from human activity. For this reason, issues on environmental goods can be put at the forefront to analyze the linkage between environmental regulations and trade flows in the perspective of international institutions. Within the framework of a gravity model, panel data on 112 exporter countries and 53 importer countries is used at a bilateral level over the period of 1989–2013 to gain an understanding of this trade-environmental regulation nexus. This paper proxies the environmental policies by employing the z-score, which includes three comprehensive environmental indicators. The findings indicate that the strict environmental policies impede the trade, and this effect is greater in terms of the environmental goods listed in APEC compared to the ones listed in OECD. Finally, although these stringent environmental regulations reduce the trade flows, they can help to foster the demand for environmentally-friendly products.

Due to the little variation in defect points, tile block defect detection typically detects subtle defects in large-format images, allowing defective characteristics to be displayed regionally. Traditional convolutional neural network architectures that extract regional features take into account the connection between regional features simply, resulting in the presence of region-specific bias, which makes tile block defect detection still a challenging task. To address this challenge, this paper divides feature information into patches that can represent different regional features. Additionally, the relationship between different patches and tile block defects is studied; as a result, this paper proposes a new attention mechanism called the Cross Patch Attention Module (CPAM). Since the regional performance of patches is consistent with the tile block defect characteristics, CPAM can distinguish various regional features by patches. Then, in order to create reliable one-dimensional patch information, CPAM provides a method to connect patches linearly in two spatial directions. This takes into account the correlation of adjacent patches in various spatial directions. Finally, by extracting the regional characteristics of patches, CPAM can successfully assist the model in distinguishing the importance of different patches. The experimental results demonstrate that CPAM has excellent performance for tile block defect detection, and plugging CPAM into different end-to-end models can have a good gain effect, which can effectively and stably help the model to complete the task of tile block defect detection.

To improve the adaptive clamping performance of traditional single-tendon-driven underactuated grippers for grasping multiple categories of objects, a novel dual-tendon-driven underactuated gripper is proposed in this paper. First, two independent tendons with different winding paths are designed in the gripper to realize the changeable resultant moment of the end knuckle rotating joint and the movement sequences of gripper knuckles driven by different tendons are analysed too. Then, some kinematic analysis and dynamical simulations are carried out to verify the validation of the knuckle structure and dual-tendon winding path design. At last, a prototype of the novel gripper is manufactured and some grasping experiments are carried out on multiple categories of objects, with different sizes and shapes. The experimental results show that all the objects can be clamped tightly. Compared with the traditional single-tendon-driven gripper, the novel one can achieve a more flexible grasping operation and a larger end clamping force, which are more suitable for the adaptive grasping requirements of robotic automatic sorting.

When performing robotic automatic sorting and assembly operations of multi-category hardware, there are some problems with the existing convolutional neural network visual recognition algorithms, such as large computing power consumption, low recognition efficiency, and a high rate of missed detection and false detection. A novel efficient convolutional neural algorithm for multi-category aliasing hardware recognition is proposed in this paper. On the basis of SSD, the novel algorithm uses Resnet-50 instead of VGG16 as the backbone feature extraction network, and it integrates ECA-Net and Improved Spatial Attention Block (ISAB): two attention mechanisms to improve the ability of learning and extract target features. Then, we pass the weighted features to extra feature layers to build an improved SSD algorithm. At last, in order to compare the performance difference between the novel algorithm and the existing algorithms, three kinds of hardware with different sizes are chosen to constitute an aliasing scene that can simulate an industrial site, and some comparative experiments have been completed finally. The experimental results show that the novel algorithm has an mAP of 98.20% and FPS of 78, which are better than Faster R-CNN, YOLOv4, YOLOXs, EfficientDet-D1, and original SSD in terms of comprehensive performance. The novel algorithm proposed in this paper can improve the efficiency of robotic sorting and assembly of multi-category hardware.

To evaluate social support and loneliness as well as their association among caregivers of children with chronic kidney disease (CKD) from China during the coronavirus disease 2019 (COVID-19) pandemic. We collected data for caregivers of children with CKD and caregivers of healthy children and matched the two groups using propensity score matching (PSM). We compared the differences in social support and loneliness between the two groups after matching and analyzed the relationship between social support and loneliness in the observation group. Before PSM, we analyzed the data for 247 caregivers of children with CKD and 315 caregivers of healthy children from 13 provinces. After PSM, the two groups each included 202 caregivers. The social support score of caregivers of children with CKD was lower than that of caregivers of healthy children (P < 0.002), while the loneliness score was higher for caregivers of children with CKD than for caregivers of healthy children (P < 0.008). The social support score was negatively correlated with the loneliness score (r = −0.598, P < 0.001). Caregivers of children with CKD experienced less social support and greater loneliness than caregivers of healthy children during the COVID-19 pandemic. Therefore, greater attention should be paid to providing social support for caregivers of CKD children and to improving the ability of these caregivers to cope with loneliness.

Paenibacillus triticisoli BJ-18, a N2-fixing bacterium, is able to promote plant growth, but the secondary metabolites that may play a role in promoting plant growth have never been characterized. In this study, untargeted metabolomics profiling of P. triticisoli BJ-18 indicated the existence of 101 known compounds, including N2-acetyl ornithine, which is the precursor of siderophores, plant growth regulators such as trehalose 6-phosphate, betaine and trigonelline, and other bioactive molecules such as oxymatrine, diosmetin, luotonin A, (-)-caryophyllene oxide and tetrahydrocurcumin. In addition, six compounds were also isolated from P. triticisoli BJ-18 using a combination of silica gel chromatography, sephadex LH-20, octadecyl silane (ODS), and high-performance liquid chromatography (HPLC). The compound structures were further analyzed by Nuclear Magnetic Resonance (NMR), Mass Spectrometry (MS), and Electronic Circular Dichroism (ECD). The six compounds included three classical siderophore fusarinines identified as deshydroxylferritriacetylfusigen, desferritriacetylfusigen, and triacetylfusigen, and three indolic acids identified as paenibacillic acid A, 3-indoleacetic acid (IAA), and 3-indolepropionic acid (IPA). Both deshydroxylferritriacetylfusigen and paenibacillic acid A have new structures. Fusarinines, which normally occur in fungi, were isolated from bacterium for the first time in this study. Both siderophores (compounds 1 and 2) showed antimicrobial activity against Escherichia coli, Staphylococcus aureus and Bacillus subtilis, but did not show obvious inhibitory activity against yeast Candida albicans, whereas triacetylfusigen (compound 3) showed no antibiosis activity against these test microorganisms. Paenibacillic acid A, IAA, and IPA were shown to promote the growth of plant shoots and roots, and paenibacillic acid A also showed antimicrobial activity against S. aureus. Our study demonstrates that siderophores and indolic acids may play an important role in plant growth promotion by P. triticisoli BJ-18.

Abstract Background Rabies is an important but underestimated threat to public health, with most cases reported in Asia. Since 2000, a new epidemic wave of rabies has emerged in Yunnan Province, southwestern China, which borders three countries in Southeast Asia. Method We estimated gene-specific evolutionary rates for rabies virus using available data in GenBank, then used this information to calibrate the timescale of rabies virus (RABV) spread in Asia. We used 452 publicly available geo-referenced complete nucleoprotein (N) gene sequences, including 52 RABV sequences that were recently generated from samples collected in Yunnan between 2008 and 2012. Results The RABV N gene evolutionary rate was estimated to be 1.88 × 10−4 (1.37–2.41 × 10−4, 95% Bayesian credible interval, BCI) substitutions per site per year. Phylogenetic reconstructions show that the currently circulating RABV lineages in Yunnan result from at least seven independent introductions (95% BCI: 6–9 introductions) and represent each of the three main Asian RABV lineages, SEA-1, -2 and -3. We find that Yunnan is a sink location for the domestic spread of RABV and connects RABV epidemics in North China, South China, and Southeast Asia. Cross-border spread from southeast Asia (SEA) into South China, and intermixing of the North and South China epidemics is also well supported. The influx of RABV into Yunnan from SEA was not well-supported, likely due to the poor sampling of SEA RABV diversity. We found evidence for a lineage displacement of the Yunnan SEA-2 and -3 lineages by Yunnan SEA-1 strains, and considered whether this could be attributed to fitness differences. Conclusion Overall, our study contributes to a better understanding of the spread of RABV that could facilitate future rabies virus control and prevention efforts.

Globally, mosquitoes are known to be competent vectors to various arboviruses that cause serious and debilitating diseases to humans and animals. Conversely, mosquitoes harbor a wide array of insect specific viruses (ISVs) that are generally neglected. Extensive characterization of these ISVs is important in understanding their persistence infection effect on host behavior and arbovirus transmission. Herein, we report first time isolation of Tanay virus (TANAV) isolate YN15_103_01 in Anopheles sinensis mosquitoes from Yunnan Province, China. Phylogenetically, the isolate’s nucleotide identity had more than 14.47% variance compared to previous TANAV isolates, and it clustered into an independent branch within the genus Sandewavirus in the newly proposed taxon Negevirus. TANAV growth and high titers was attained in Aag2 cells (107 PFU/mL) but with no CPE observed up to 7 days.p.i. compared to C6/36 cells that exhibited extensive CPE at 48 h.p.i. with titers of 107 PFU/mL. Contrarywise, the viral isolate did not replicate in vertebrate cell lines. Electron microscopy analyses showed that its final maturation process takes place in the cell cytoplasm. Notably, the predicted viral proteins were verified to be corresponding to the obtained SDS-PAGE protein bands. Our findings advance forth new and vital knowledge important in understanding insect specific viruses, especially TANAV.

Infection by Zika virus (ZIKV) is linked to microcephaly and other neurological disorders, posing a significant health threat. Innate immunity is the first line of defense against invading pathogens, but relatively little is understood regarding host intrinsic mechanisms that guard against ZIKV. Here, we show that host tripartite motif-containing protein 56 (TRIM56) poses a barrier to ZIKV infection in cells of neural, epithelial and fibroblast origins. Overexpression of TRIM56, but not an E3 ligase-dead mutant or one lacking a short C-terminal portion, inhibited ZIKV RNA replication. Conversely, depletion of TRIM56 increased viral RNA levels. Although the C-terminal region of TRIM56 bears sequence homology to NHL repeat of TRIM-NHL proteins that regulate miRNA activity, knockout of Dicer, which abolishes production of miRNAs, had no demonstrable effect on ZIKV restriction imposed by TRIM56. Rather, we found that TRIM56 is an RNA-binding protein that associates with ZIKV RNA in infected cells. Moreover, a recombinant TRIM56 fragment comprising the C-terminal 392 residues captured ZIKV RNA in cell-free reactions, indicative of direct interaction. Remarkably, deletion of a short C-terminal tail portion abrogated the TRIM56-ZIKV RNA interaction, concomitant with a loss in antiviral activity. Altogether, our study reveals TRIM56 is an RNA binding protein that acts as a ZIKV restriction factor and provides new insights into the antiviral mechanism by which this E3 ligase tackles flavivirus infections.

Multivesicular bodies (MVBs) are critical intermediates in the trafficking of ubiquitinated endocytosed surface proteins to the lysosome/vacuole for destruction. Recognizing and packaging ubiquitin modified cargoes to the MVB pathway require ESCRT (Endosomal sorting complexes required for transport) machinery, which consists of four core subcomplexes, ESCRT-0, ESCRT-I, ESCRT-II, and ESCRT-III. Fusarium graminearum is an important plant pathogen that causes head blight of major cereal crops. Our previous results showed that ESCRT-0 is essential for fungal development and pathogenicity in Fusarium graminearum. We then, in this study, systemically studied the protein-protein interactions within F. graminearum ESCRT-I, -II or -III complex, as well as between ESCRT-0 and ESCRT-I, ESCRT-I and ESCRT-II, and ESCRT-II and ESCRT-III complexes and found that loss of any ESCRT component resulted in abnormal function in endocytosis. In addition, ESCRT deletion mutants displayed severe defects in growth, deoxynivalenol (DON) production, virulence, sexual, and asexual reproduction. Importantly genetic complementation with corresponding ESCRT genes fully rescued all these defective phenotypes, indicating the essential role of ESCRT machinery in fungal development and plant infection in F. graminearum. Taken together, the protein-protein interactome and biological functions of the ESCRT machinery is first profoundly characterized in F. graminearum, providing a foundation for further exploration of ESCRT machinery in filamentous fungi.

BACKGROUND:Mammarenaviruses are associated with human hemorrhagic fever diseases in Africa and America. Recently, a rodent mammarenavirus, Wēnzhōu virus (WENV) and related viruses, have been reported in China, Cambodia, and Thailand. Moreover, in Cambodia, these viruses were suspected to be associated with human disease. In China, Yunnan Province is famous for its abundant animal and plant diversity and is adjacent to several South-eastern Asia countries. Therefore, it is necessary to know whether WENV-related viruses, or other mammarenaviruses, are prevalent in this province. METHODOLOGY/PRINCIPAL FINDINGS:Small mammals were trapped, euthanized, and sampled. Mammarenavirus RNA was detected using a nested reverse transcription polymerase chain reaction (RT-PCR) and quantified by real-time RT-PCR. A total of 1040 small mammals belonging to 13 genera and 26 species were trapped in Yunnan Province. WENV-related mammarenaviruses were detected in 41 rodent liver samples, mainly in brown rats (Rattus norvegicus) and oriental house rats (R. tanezumi).Viral nucleocapsid protein was detected in liver sections by indirect immunofluorescence assay. Full-length-genomes were amplified by RT-PCR and used for phylogenetic analysis with the MEGA package. Recombination analysis was performed using the SimPlot and Recombination Detection Program. CONCLUSIONS/SIGNIFICANCE:WENV related viruses circulated in small mammals in Yunnan Province. Whole genome sequence analysis of five selected viral strains showed that these viruses are closely related to WENVs discovered in Asia and form an independent branch in the phylogenetic tree in the WENV clade. Paying attention to investigate the influence of these viruses to public health is essential in the epidemic regions.

Despite ongoing efforts to control transmission, rabies prevention remains a challenge in many developing countries, especially in rural areas of China where re-emerging rabies is under-reported due to a lack of sustained animal surveillance. By taking advantage of detailed genomic and epidemiological data for the re-emerging rabies outbreak in Yunnan Province, China, collected between 1999 and 2015, we reconstruct the demographic and dispersal history of domestic dog rabies virus (RABV) as well as the dynamics of dog-to-dog and dog-to-human transmission. Phylogeographic analyses reveal a lower diffusion coefficient than previously estimated for dog RABV dissemination in northern Africa. Furthermore, epidemiological analyses reveal transmission rates between dogs, as well as between dogs and humans, lower than estimates for Africa. Finally, we show that reconstructed epidemic history of RABV among dogs and the dynamics of rabid dogs are consistent with the recorded human rabies cases. This work illustrates the benefits of combining phylogeographic and epidemic modelling approaches for uncovering the spatiotemporal dynamics of zoonotic diseases, with both approaches providing estimates of key epidemiological parameters.

The activation of interferon (IFN)-regulatory factor-3 (IRF3), characterized by phosphorylation and nuclear translocation of the latent transcription factor, is central to initiating innate antiviral responses. Whereas much has been learned about the upstream pathways and signaling mechanisms leading to IRF3 activation, how activated IRF3 operates in the nucleus to control transcription of IFNs remains obscure. Here we identify EAP30 (a.k.a, SNF8/VPS22), an endosomal sorting complex required for transport (ESCRT)-II subunit, as an essential factor controlling IRF3-dependent antiviral defense. Depletion of EAP30, but not other ESCRT-II subunits, compromised IRF3-dependent induction of type I and III IFNs, IFN-stimulated genes (ISGs) and chemokines by double-stranded RNA or viruses. EAP30, however, was dispensable for the induction of inflammatory mediators of strict NF-κB target. Significantly, knockdown of EAP30 also impaired the establishment of an antiviral state against vesicular stomatitis virus and hepatitis C virus, which are of distinct viral families. Mechanistically, EAP30 was not required for IRF3 activation but rather acted at a downstream step. Specifically, a fraction of EAP30 localized within the nucleus, where it formed a complex with IRF3 and its transcriptional co-activator, CREB-binding protein (CBP), in a virus-inducible manner. These interactions promoted IRF3 binding to target gene promoters such as IFN-β, IFN-λ1 and ISG56. Together, our data describe an unappreciated role for EAP30 in IRF3-dependent innate antiviral response in the nucleus.

Land cover and its dynamic information is the basis for characterizing surface conditions, supporting land resource management and optimization, and assessing the impacts of climate change and human activities. In land cover information extraction, the traditional convolutional neural network (CNN) method has several problems, such as the inability to be applied to multispectral and hyperspectral satellite imagery, the weak generalization ability of the model and the difficulty of automating the construction of a training database. To solve these problems, this study proposes a new type of deep convolutional neural network based on Landsat-8 Operational Land Imager (OLI) imagery. The network integrates cascaded cross-channel parametric pooling and average pooling layer, applies a hierarchical sampling strategy to realize the automatic construction of the training dataset, determines the technical scheme of model-related parameters, and finally performs the automatic classification of remote sensing images. This study used the new type of deep convolutional neural network to extract land cover information from Qinhuangdao City, Hebei Province, and compared the experimental results with those obtained by traditional methods. The results show that: (1) The proposed deep convolutional neural network (DCNN) model can automatically construct the training dataset and classify images. This model performs the classification of multispectral and hyperspectral satellite images using deep neural networks, which improves the generalization ability of the model and simplifies the application of the model. (2) The proposed DCNN model provides the best classification results in the Qinhuangdao area. The overall accuracy of the land cover data obtained is 82.0%, and the kappa coefficient is 0.76. The overall accuracy is improved by 5% and 14% compared to the support vector machine method and the maximum likelihood classification method, respectively.

Rocky desertification is an important type of ecological degradation in southwest of China. The author uses the web crawler technology and obtained 9345 journal papers related to rocky desertification from 1950s to 2016 in China. The authors also constructed a technological process to extract research hot regions on rocky desertification and then a spatial distribution map of research hot regions on rocky desertification was formed. Finally, the authors compared the spatial distribution using the sensitivity map of rocky desertification to find the differences between two maps. The study shows that: (1) rocky desertification research hot regions in China are mainly distributed in Guizhou, Yunnan and Guangxi, especially in Bijie, Liupanshui, Guiyang, Anshun, Qianxinan Autonomous Prefecture, QianNan Autonomous Prefecture, Qiandongnan Autonomous Prefecture in Guizhou Province, Hechi, Baise, Nanning, Guilin in Guangxi Zhuang Autonomous Region and Zhaotong in Yunnan Province. (2) The research hot regions on rocky desertification have good spatial consistency with the sensitivity regions of rocky desertification. At the prefecture level, the overlap rate of the two regions reached 85%. Because of the influence of topography, vegetation coverage, population distribution, traffic accessibility and other factors, there were regions that consisted of combinations of high sensitivity but low research popularity regarding rocky desertification; these sites included Qionglai Mountain-Liangshan Area of Sichuan, Wushan-Shennongjia Area of Hubei, Hengduan Mountain Area of western Yunnan and Dupangling Area of southern Hunan. (3) The research hot regions and sensitive regions cannot be matched completely in time, space and concept. Therefore, we can use their spatial distribution differences to improve the pertinence of planning, governance and study of rocky desertification.

We provide here a comparative genome analysis of 31 strains within the genus Paenibacillus including 11 new genomic sequences of N2-fixing strains. The heterogeneity of the 31 genomes (15 N2-fixing and 16 non-N2-fixing Paenibacillus strains) was reflected in the large size of the shell genome, which makes up approximately 65.2% of the genes in pan genome. Large numbers of transposable elements might be related to the heterogeneity. We discovered that a minimal and compact nif cluster comprising nine genes nifB, nifH, nifD, nifK, nifE, nifN, nifX, hesA and nifV encoding Mo-nitrogenase is conserved in the 15 N2-fixing strains. The nif cluster is under control of a σ(70)-depedent promoter and possesses a GlnR/TnrA-binding site in the promoter. Suf system encoding [Fe-S] cluster is highly conserved in N2-fixing and non-N2-fixing strains. Furthermore, we demonstrate that the nif cluster enabled Escherichia coli JM109 to fix nitrogen. Phylogeny of the concatenated NifHDK sequences indicates that Paenibacillus and Frankia are sister groups. Phylogeny of the concatenated 275 single-copy core genes suggests that the ancestral Paenibacillus did not fix nitrogen. The N2-fixing Paenibacillus strains were generated by acquiring the nif cluster via horizontal gene transfer (HGT) from a source related to Frankia. During the history of evolution, the nif cluster was lost, producing some non-N2-fixing strains, and vnf encoding V-nitrogenase or anf encoding Fe-nitrogenase was acquired, causing further diversification of some strains. In addition, some N2-fixing strains have additional nif and nif-like genes which may result from gene duplications. The evolution of nitrogen fixation in Paenibacillus involves a mix of gain, loss, HGT and duplication of nif/anf/vnf genes. This study not only reveals the organization and distribution of nitrogen fixation genes in Paenibacillus, but also provides insight into the complex evolutionary history of nitrogen fixation.

BackgroundNatural resistance associated macrophage protein 1 (NRAMP1), encoded by the SLC11A1 gene, has been described to regulate macrophage activation and be associated with infectious and autoimmune diseases. The relation between SLC11A1 polymorphisms and tuberculosis susceptibility has been studied in different populations.MethodsWe systematically reviewed published studies on SLC11A1 polymorphisms and tuberculosis susceptibility until September 15, 2010 and quantitatively summarized associations of the most widely studied polymorphisms using meta-analysis.ResultsIn total, 36 eligible articles were included in this review. In Meta-analysis, significant associations were observed between tuberculosis risk and widely studied SLC11A1 polymorphisms with summarized odds ratio of 1.35 (95%CI, 1.17-1.54), 1.25 (95% CI, 1.04-1.50), 1.23 (95% CI, 1.04-1.44), 1.31 (95%CI, 1.08-1.59) for 3' UTR, D543N, INT4, and 5' (GT)n, respectively. Heterogeneity between studies was not pronounced, and the associations did not remarkably vary in the stratified analysis with respect to study population and study base.ConclusionsThe association between SLC11A1 polymorphisms and tuberculosis susceptibility observed in our analyses supports the hypothesis that NRAMP1 might play an important role in the host defense to the development of tuberculosis.
