The three-dimensional (3D) retinal organoids (ROs) derived from human induced pluripotent stem cells (hiPSCs), mimicking the growth and development of the human retina, is a promising model for investigating inherited retinal diseases in vitro. However, the efficient generation of homogenous ROs remains a challenge. Here we introduce a novel polydimethylsiloxane (PDMS) microwell platform containing 62 V-bottom micro-cavities for the ROs differentiation from hiPSCs. The uniform adherent 3D ROs could spontaneously form using neural retina (NR) induction. Our results showed that the complex of NR (expressing VSX2), ciliary margin (CM) (expressing RDH10), and retinal pigment epithelium (RPE) (expressing ZO-1, MITF, and RPE65) developed in the PDMS microwell after the differentiation. It is important to note that ROs in PDMS microwell platforms not only enable one-stop assembly but also maintain homogeneity and mature differentiation over a period of more than 25 weeks without the use of BMP4 and Matrigel. Retinal ganglion cells (expressing BRN3a), amacrine cells (expressing AP2a), horizontal cells (expressing PROX1 and AP2α), photoreceptor cells for cone (expressing S-opsin and L/M-opsin) and rod (expressing Rod opsin), bipolar cells (expressing VSX2 and PKCα), and Müller glial cells (expressing GS and Sox9) gradually emerged. Furthermore, we replaced fetal bovine serum with human platelet lysate and established a xeno-free culture workflow that facilitates clinical application. Thus, our PDMS microwell platform for one-stop assembly and long-term culture of ROs using a xeno-free workflow is favorable for retinal disease modeling, drug screening, and manufacturing ROs for clinical translation.

Per- and polyfluoroalkyl substances (PFASs) levels in Indo-Pacific finless porpoises (Neophocaena phocaenoides) in the Pearl River Estuary (PRE), near the most economically developed region in China, have not been characterized. We measured the hepatic concentrations of twelve PFASs, including nine perfluoroalkyl carboxylic acids (PFCAs) and three perfluoroalkane sulfonic acids (PFSAs) in the finless porpoises (n = 21) collected from the PRE between 2007 and 2020. The average level of PFSAs was more than 2-times higher than that of PFCAs. The order of six dominant PFASs was perfluorooctane sulfonate (PFOS) perfluoroundecanoic acid (PFUdA) perfluorodecanoic acid (PFDA) perfluorotridecanoic acid (PFTrDA) perfluorononanoic acid (PFNA) perfluorododecanoic acid (PFDoDA). The levels of Hepatic PFOS of 29% samples exceeded the no observable adverse effect level (NOAEL) values. The concentration of PFASs in males was significant higher than in females. PFASs levels were significantly negatively correlated with body length in males and positively correlated in females. PFASs levels in the PRE finless porpoises were lower than in humpback dolphins possibly due to different foraging habitat toward the coast and the consumption of less fish. PFCAs levels in finless porpoises from the western PRE were higher compared to Hong Kong, possibly due to the high-intensity sources of terrestrial anthropogenic pollutants. Significant increasing spatiotemporal trends of PFSAs, PFCAs and PFASs were found in finless porpoises from 2007 to 2020, suggesting a continuously increased risk of PFASs exposure for PRE cetaceans in the last decade.

Epithelial-mesenchymal transition (EMT) of retinal pigment epithelium (RPE) cells is critically involved in the occurrence of subretinal fibrosis. This study aimed to investigate the role of enhancer of zeste homolog 2 (EZH2) in EMT of human primary RPE cells and the underlying mechanisms of the anti-fibrotic effect of EZH2 suppression. Primary cultures of human RPE cells were treated with TGF-β1 for EMT induction. EZH2 was silenced by siRNA to assess the expression levels of epithelial and fibrotic markers using qRT-PCR, Western blot, and immunofluorescence staining assay. Furthermore, the cellular migration, proliferation and barrier function of RPE cells were evaluated. RNA-sequencing analyses were performed to investigate the underlying mechanisms of EZH2 inhibition. Herein, EZH2 silencing up-regulated epithelial marker ZO-1 and downregulated fibrotic ones including α-SMA, fibronectin, and collagen 1, alleviating EMT induced by TGF-β1 in RPE cells. Moreover, silencing EZH2 inhibited cellular migration and proliferation, but didn't affect cell apoptosis. Additionally, EZH2 suppression contributed to improved barrier functions after TGF-β1 stimulation. The results from RNA sequencing suggested that the anti-fibrotic effect of EZH2 inhibition was associated with the MAPK signaling pathway, cytokine-cytokine receptor interaction, and the TGF-beta signaling pathway. Our findings provide evidence that the suppression of EZH2 might reverse EMT and maintain the functions of RPE cells. EZH2 could be a potential therapeutic avenue for subretinal fibrosis.

In the search for a more reasonable yet effective treatment method, this study explores the application of nanometer magnetic particles and prednisolone combined with rituximab in children with nephrotic syndrome (NS). According to the diagnostic criteria of children with hormone sensitivity and frequent recurrence of primary NS from the nephrology group of the Chinese Academy of Pediatrics, 40 children with NS diagnosed in our hospital from January 2018 to October 2019 were selected. Nanomagnetic α-Fe2O3 particles were prepared by chemical reaction synthesis. Before and after treatment, urine protein, oral hormone dosage, changes in peripheral blood CD19 + B lymphocyte levels, changes in plasma immunoglobulin levels, and patient general improvement were observed, along with the recurrence of renal disease after treatment. Immunoglobulin was measured by the immunoturbidimetric method, and the endpoint of the follow-up was the first relapse after administration. This study shows that the use of nanomagnetic particles and prednisolone in combination with rituximab in the treatment of pediatric NS has a significant effect, improving the renal function indicators of children and effectively controlling the condition of children while maintaining a higher level of safety.

Litopenaeus vannamei, as an important marine aquaculture species, has attracted more and more attentions in past several years. More recently people got its genome fine mapping, which unveiled a gene treasure. In this study, we have identified a novel trypsin-like protein which came from previous WSSV-infected shrimp plasma iTRAQ data. This protein is a 39 kDa protein with 363 amino acids. It contains a conserved trypsin-domain and could be strongly induced with WSSV infection. Interestingly, knockdown of this protein made shrimps vulnerable to WSSV infection. Further exploration unveiled that this fragility was probably due to the fact that knockdown of this protein could cause shrimp hemocytes apoptosis, which indicated that this protein played key roles in preventing shrimp hemocytes from apoptosis. To further explore how LvTLAP protected shrimp hemocytes from apoptosis, GST pull down assay was applied to screen LvTLAP interacting protein in shrimp plasma. L. vannamei growth and transformation-dependent-like protein (LvGTD-like protein) was identified as a LvTLAP interacting protein, which played proapoptotic roles in cells. Thus, a possible explanation for LvTLAP anti-apoptosis activity was that this protein could block LvGTD-like protein proapoptotic activity to protect shrimp hemocytes from death. In general, our study has uncovered a novel WSSV responsive shrimp plasma protein, which played key roles in shrimp hemocytes anti-apoptosis and shrimp against WSSV infection.

Diabetic retinopathy (DR) is one of the most common and severe microvascular complications of diabetes, and the leading cause of preventable blindness in working-aged people. Here, we generated an induced pluripotent stem (iPS) cell line using blood-derived cells from a patient with DR. Peripheral blood mononuclear cells (PBMCs) were reprogrammed with Sendai virus.

Background: Among severely immunosuppressed patients resistant or refractory cytomegalovirus (CMV) retinitis is not uncommon, and is potentially blinding in the affected eye. Immunotherapy using CMV-specific cytotoxic T lymphocytes (CTLs) is an emerging and promising alternative for the control of resistant or refractory CMV retinitis. However, highly purified CMV-CTLs are still not available in the majority of medical institutions in China because the generation of CMV-CTLs is labor intensive, time-consuming and expensive.Purpose: This letter describes the clinical responses to a simplified CMV-CTL immunotherapy aimed at controlling drug-resistant CMV retinitis in two immunodeficient patients.Conclusions: Our cases suggest that the simplified CMV-CTL immunotherapy offers a highly efficient and low-cost solution to drug-resistant CMV retinitis. This immunotherapy strategy targeting drug-resistant CMV retinitis is likely to be cost-effective in the context of a specific patient population.

With the dramatic increase in anthropogenic threats to the Pearl River Estuary (PRE), the population size of the Indo-Pacific humpback dolphins (Sousa chinensis) has significantly decreased over the past decade. To understand the impact and potential risks of intense human activities on these dolphins, factors related to the mortality of humpback dolphins in the PRE were investigated by a detailed examination of 343 dolphin specimens stranded during 2003-2017. There was a significant (p 0.01) increasing trend for humpback dolphin stranding, reflecting the accelerating rate of the population decline. A large proportion of strandings (35.88%) were neonates. A low recruitment rate implies slow population growth, and hence, limited capacity to resist anthropogenic stress. The most commonly diagnosed causes of death were vessel collision and net entanglement. The concentrations of trace metals, polychlorinated biphenyl (PCB) congeners, dichlorodiphenyltrichloroethane, polycyclic aromatic hydrocarbons, and most of per- and polyfluoroalkyl substances (PFASs) in the dolphin samples were greater than those previously reported in cetaceans globally. Furthermore, Cu, PCB77, PCB169, PCB81, PCB37, and PFASs (excluding PFBA, PFPeA, PFHxA, PFHxDA, and PFODA) were the major pollutants accumulated in neonates. 67% of PCB, 78% of Cu, and 100% of perfluorooctane sulfonate concentrations in the neonates exceeded the threshold for toxicological effects in marine mammals, suggesting that these compounds could be important factors contributing to the low survival rate of calves in this area. This study revealed that vessel transportation, fishing activities, and pollutant bioaccumulation are the three major causes of humpback dolphin mortality in the PRE. These results highlight the need for more efforts to restrict anthropogenic activities, especially vessel traffic, the catching of these marine animals and fishing, and pollutant discharge, in order to prevent vulnerable species from continuous population decline and further extinction.

Background Retinopathy of prematurity (ROP) is a vascular proliferative disorder of the developing retina and a significant cause of childhood blindness around the world. The incidence of ROP is affected by many factors, and the incidence rate varies from country to country. The purpose of this study is to report the incidence and risk factors of ROP in neonatal intensive care unit (NICU) of Guangzhou First People’s Hospital in China. Methods A retrospective review was performed on 436 premature infants who were consecutive ROP screened in the NICU of Guangzhou First People’s Hospital from March 2013 to October 2017. The single-factor analysis and the logistic multivariate regression analysis were used to detect risk factors of ROP. Results Total 436 premature infants were consecutive ROP screened, 138 (31.65%) were found ROP, and 61(13.99%) were treated. The single-factor analysis revealed that the incidence of ROP was associated with multiple births, gestational age, birth weight, mechanical ventilation, intravascular hemolysis, the number of operations and blood culture results. The logistic multivariate regression analysis revealed that gestational age; birth weight, mechanical ventilation, minimum SaO2 and daily weight gain were independent risk factors for ROP onset. Forty-nine patients underwent retinal laser photocoagulation with recurrence 20 patients. Twelve patients underwent anti-VEGF drug (Ranibizumab) via intraocular injection with 5 patients of recurrence. Conclusions The incidence of ROP in NICU of Guangzhou China will match those in middle-income countries, but higher than high-income countries. Anti-VEGF drugs could be preferred as a good treatment method for zone 1 ROP and aggressive posterior ROP.

Exogenous application of plant-growth promoting substances in combination with chelators is a common way to enhance the phytoextraction of heavy metals. A pot experiment was used to explore the influences of indole-3-acetic acid (IAA)/gibberellin (GA3) alone or together with oxalic acid (OA) on the growth, physiological response, and nutrient contents of hyperaccumulator Sedum alfredii Hance, and cadmium (Cd) and lead (Pb) phytoextraction efficiency. The results showed that a foliar spray of IAA/GA3 alone or together with OA increased plant growth. The largest shoot biomass with increase by 29.7% was produced by the 50 μmol L−1 IAA combined with 2.5 mmol kg−1 OA (50I+2.5OA) treatment as compared with the control treatment (CK). The presence of IAA and GA3 enhanced the chlorophyll a, carotenoid, and potassium contents in leaves and decreased the malondialdehyde content. The Cd content in leaf and the translocation factor (TFshoot) value from 50I+2.5OA treatment was increased by 4.29% and 21.4%, and the Pb content in stem and shoot, and the TFshoot of Pb after applying 50 μmol L-1 GA3 combined with 2.5 mmol kg−1 OA was enhanced by 32.5%, 13.4%, and 57.6%, compared with CK, respectively. The optimal Cd and Pb phytoextraction efficiency occurred from 50I+2.5OA treatment with increase by 82.4% and 79.3% as compared with CK, respectively. Therefore, the results showed that a combined application of 50 μmol L−1 IAA and 2.5 mmol kg−1 OA could effectively enhance S. alfredii Hance phytoremediation of Cd and Pb co-contaminated soil.

The screening and cultivation of salt-tolerant crops are becoming more and more important owing to the constant increase in the saline soil area worldwide. Asparagus (A. officinalis L.) is a highly nutritious vegetable crop and widely consumed globally for a long time; however, little research has been done on asparagus. In this study, the salt tolerance of 95 asparagus germplasm accessions was evaluated at three growth stages (germination, seedling, and adult stages) under both salt-stressed and control conditions. Results showed that the growth parameters of most germplasm accessions were obviously inhibited by salt stress. The mean value of the seed germination rate at the germination stage decreased by half under salt-stressed conditions, the mean salt-injury index at the seedling stage reached 57.68%, and the fresh weight of the aboveground part (FWA) and the dry weight of the aboveground part (DWA) decreased the most among the traits determined at the adult stage by more than 60%. Our study screened out 30, 19, and 18 tolerant germplasm accessions (including highly salt-tolerant and salt-tolerant germplasm accessions) at the germination stage, seedling stage, and adult stage, respectively. Among them, two germplasm accessions (Ji08-2 and Jx1502) were simultaneously identified to be tolerant in all three growth stages, while other germplasm accessions were tolerant only at one or two stages. Thus, the salt tolerance of asparagus has periodic characteristics and changes throughout the lifecycle, and the identification of salt tolerance at all the main growth stages facilitates adequate assessment and application of tolerant germplasm accessions.

Agricultural soils contaminated with cadmium (Cd) pose a risk to receiving surface water via drainage or runoff. A 90-day laboratory incubation experiment was conducted to investigate the release characteristics and transformation of Cd from contaminated paddy soil amended with agrochemical (NPK fertilizer) and lime (L) under water management regimes of continuous flooding (F) and drying-wetting cycles (DW). The result showed that the dissolved Cd concentrations in overlying water of the fertilizer treatment under flooding (NPK+F) and drying-wetting (NPK+DW) reached up to 81.0 μg/L and 276 μg/L, and were much higher than that from the corresponding controls without NPK fertilizer addition at the end of experiment. The Cd concentration showed significantly negative correlation with overlying water pH, but positive correlation with soil redox potential and concentrations of dissolved total nitrogen, sulfate and manganese in overlying water (P 0.05), indicating that drying-wetting cycles and N fertilizer addition may enhance soil Cd release. The Cd concentrations in overlying water from all treatments except NPK+L+F treatment exceeded the Cd threshold limit of Chinese Environmental Quality Standards for Surface Water (10 μg/L Grade V) and poses potential risk to surface water quality. Meanwhile, the proportion of Cd in the acid-soluble fraction from all incubated soil except NPK+L+F treatment increased compared to before incubation. The results indicated that continuous flooding was a reasonable water management candidate coupled with lime addition for immobilizing soil Cd.

Knowledge on adsorption behaviour of vanadium (V) on natural clay minerals is critical to understand V mobility in soil and aqueous systems. In this study, the characteristics and mechanisms of V(V) adsorption on two abundant soil clay minerals, namely, natural kaolinite and montmorillonite, as well as the effects of solution pH and ionic strength on the adsorption process were investigated. The results showed that the kinetics of V(V) adsorption on kaolinite and montmorillonite conformed to pseudo second-order, and reached equilibrium within 240 min and 360 min respectively. The adsorption isotherms of V(V) on kaolinite were better fit with the Freundlich model, while the Langmuir model was a better fit for montmorillonite. The Langmuir model indicated that natural montmorillonite and kaolinite have low affinity for V(V) anions with maximum adsorption capacity of 0.98 mg·g−1 and 0.78 mg·g−1, respectively. The higher capacity was found at pH 4–10 due to the mass formation of deprotonated hydroxyl groups for V adsorption. The V adsorption capacity on montmorillonite increased with the increasing ionic strength of NaNO3. The results of pH and ionic-strength effect showed that surface complexation and electrostatic interaction may be the V(V) adsorption mechanisms.

The plasma effects of energetic electron bombardment on component materials severely limit the output pulsed energies of high-power microwave (HPM) sources. This paper proposes a feasible method for choosing materials for beam collectors in HPM devices. First, electron energy deposition in metal materials is theoretically and numerically investigated. Considering the energy threshold resulting in material ablation, the resilience of several metals to intense relativistic electron beam (IREB) bombardment is compared. This resilience is mainly determined by the material density and melting point. Titanium shows good resilience compared with stainless steel, copper, molybdenum, and tungsten. With an incident 780 keV, 9.5-kA IREB, the maximum deposition energy in titanium just slightly exceeds its ablation threshold. Thus, the theoretical results indicate that titanium is a promising material for application in HPM devices. The experimental results validate the theoretical analyses. Compared with the conventional stainless steel collector, the titanium collector has better stability and lifetime. We intend to investigate more materials in future studies.

Denitrification is important for nitrogen balance in agricultural ecosystems. It is well established that the abundance and structure of denitrifier are strongly influenced by environmental factors. However, knowledge of how nirS- and nirK-harboring microbial communities vary in paddy soils of different climatic regions is lacking, along with how these microbes are associated with denitrification potential. In this study, forty paddy soils from tropical, sub-tropical, warm-temperate, and mid-temperate climate zones were collected. The results showed that soils from the sub-tropical zone had the highest denitrification rates. The abundances of nirS and nirK had a strong negative association with C/N ratio and clearly varied among the four climate zones. Variation in nirS- and nirK-containing communities existed across sampling sites from each climate zone in terms of α- and β-diversity. Soil pH and climate factors significantly affected community diversity. Network analysis revealed that different climate regions had similar keystone taxa like Azospira, and Achromobacter, which were significantly related to denitrification rates. Structural equation modeling indicated that the differences in denitrifying enzyme activity among the four climate zones were mostly explained by climate factors, soil pH, and nirS biodiversity. Specifically, the biodiversity of nirS was more important than that of nirK in regulating potential denitrification activity in paddy soil, suggesting that nirS-type denitrifiers may have high activity under anaerobic conditions. Our results allow a deeper insight into the relative contribution of nirS- and nirK- containing communities to the soil denitrification activity in paddy soils across climate zones. This study highlighted the need of manipulation experiment to explain how denitrifier biodiversity affect potential denitrification activity in the future.

Soil salinity is one of the most serious abiotic stresses that limit agricultural productivity and the distribution of crops worldwide. Arbuscular mycorrhizal fungi (AMF) can improve the salt tolerance of host plants. A systematic study on plant responses to AMF under salinity stress may provide insights into the acquired salt tolerance. Here, we performed transcriptome sequencing using the Oxford Nanopore Technologies (ONT) MinION platform for asparagus (Asparagus officinalis L.) roots with 4 treatments: non-inoculated plants under optimal growth conditions (NI), inoculated plants under optimal growth conditions (AMF), non-inoculated plants under salinity stress conditions (NI + S), and inoculated plants under salinity stress conditions (AMF + S). A total of 6124 differentially expressed transcripts (DETs) were determined. Based on Venn diagram analysis, 391 DETs were specially regulated by AMF under salinity stress rather than under optimal growth conditions, which may be linked to the salinity stress adaptation mediated by AMF. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses revealed that these transcripts mainly involved the regulation of ROS-scavenging capacity, water and nutrient status, and cell wall synthesis and modification. Furthermore, 23 differentially expressed transcription factors (TFs) were identified in the NI + S and AMF + S comparison, which are known to be associated with plant abiotic and biotic stress responses and regulation. Moreover, the identification of alternative splicing (AS) events indicated a more comprehensive response to AMF at the post-transcriptional level, which mainly occurs in genes involved in cell wall-related processes, transport, regulation of transcription, and response to abiotic stimulus. These results provide important insights into the salt tolerance induced by AMF and a solid basis for the future enhancement of plant tolerance to salinity stress.

Persimmon (Diospyros kaki Thunb.) has been widely cultivated in China but the majority of its germplasm accessions are pollination constant and astringent (PCA) type with serious issues of synonyms and homonyms. In order to identify the germplasm resources, genetic diversity and relationship among 133 accessions originating from China, Japan and America were elucidated based on SSR (simple sequence repeat) markers. A total of 158 alleles and 610 genotypes were obtained from 17 loci, with a mean value of 9.29 alleles and 35.88 genotypes per locus, respectively. The analysis of genetic variation revealed high SSR polymorphism both in Chinese and Japanese cultivars. Genetic diversity of varieties in Qinling Mountain was highest and Dabieshan Mountain was lowest. The majority of Chinese PCA genotypes were classified into a mixing group without agreement with their geographic origins in the cluster and principal coordinate analysis. All the genotypes of D. kaki were grouped distantly from the related species which were distinguished completely by 17 SSR markers. A few Chinese PCA cultivars of synonyms or homonyms such as “Niuxinshi”, “Damopan” and “Shuishi” were efficiently identified. The results indicated that Chinese PCNA varieties were closely related with Chinese PCA cultivars and revealed relatively distant relationships with Japanese PCNA types.

Diospyros is the largest genus in Ebenaceae, comprising more than 500 species with remarkable economic value, especially Diospyros kaki Thunb., which has traditionally been an important food resource in China, Korea, and Japan. Complete chloroplast (cp) genomes from D. kaki, D. lotus L., D. oleifera Cheng., D. glaucifolia Metc., and Diospyros ‘Jinzaoshi’ were sequenced using Illumina sequencing technology. This is the first cp genome reported in Ebenaceae. The cp genome sequences of Diospyros ranged from 157,300 to 157,784 bp in length, presenting a typical quadripartite structure with two inverted repeats each separated by one large and one small single-copy region. For each cp genome, 134 genes were annotated, including 80 protein-coding, 31 tRNA, and 4 rRNA unique genes. In all, 179 repeats and 283 single sequence repeats were identified. Four hypervariable regions, namely, intergenic region of trnQ_rps16, trnV_ndhC, and psbD_trnT, and intron of ndhA, were identified in the Diospyros genomes. Phylogenetic analyses based on the whole cp genome, protein-coding, and intergenic and intron sequences indicated that D. oleifera is closely related to D. kaki and could be used as a model plant for future research on D. kaki; to our knowledge, this is proposed for the first time. Further, these analyses together with two large deletions (301 and 140 bp) in the cp genome of D. ‘Jinzaoshi’, support its placement as a new species in Diospyros. Both maximum parsimony and likelihood analyses for 19 taxa indicated the basal position of Ericales in asterids and suggested that Ebenaceae is monophyletic in Ericales.

In this paper, the effect of the gases absorbed on the surface of the RBWO high-frequency structure and the background gases inside the system on the pulse duration of high power microwave is analyzed. To simulate the plasmas originating from the ionized gases, a PIC model is studied. Simulation results show that the microwave power declines and the pulse-duration decreases with plasma filled in the RBWO. Based on the simulation results, a method of baking for suppressing gas desorption is presented. The preliminary experimental results show that the pulse duration can be increased to 31ns by suppressing the gas desorption, while for the same microwave power, the pulse duration is only 19ns without treatment for the RBWO.

Study on the thermal process in repetitively operated electron beam collectors of relativistic backward wave oscillators is proposed. Using Monte Carlo method, the energy deposition of the beam was calculated. With energy deposition as the internal heat generation, numerical model of thermal process in collectors was estimated in finite element method. Temperature development was obtained and compared to the results of the experiment, which was performed with thermocouples. The heating process is adiabatic in some extent for the very short pulse, but the cooling is quite time-consuming on the contrary. Thus, temperature rises in sharp waveforms and can be extremely high, which is quite different from previous studies. Differences between simulation and experiment are discussed, including cooling boundary condition simplification of the numerical model and some drawbacks of the measuring system.
